快速脱水抑制葡萄干制过程中膜脂过氧化及褐变   总被引:1,自引:1,他引:1  
为探索脱水速度对无核白葡萄干制过程中膜脂过氧化作用及褐变的影响,以新疆无核白葡萄为试验材料,经过葡萄促干剂处理后,采用快速脱水和缓慢脱水2种处理,质量每减轻10%进行取样,测定脱水过程中果实干基含水率、干燥速率、褐变度、细胞膜透性、丙二醛(malonaldehyde,MDA)含量、脂氧合酶(lipoxygenase,LOX)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、过氧化物酶(peroxidase,POD)、多酚氧化酶(polyphenol oxidase,PPO)活性以及总酚含量的变化。结果表明:与缓慢脱水相比,快速脱水处理显著(P0.05)降低了无核白葡萄褐变度的上升,减少MDA生成量以及膜透性的增加,抑制LOX活性的升高,保持较高的活性氧清除酶SOD、CAT及POD活性及较高的总酚含量,且使PPO活性保持在一个较低的水平。因此认为,快速脱水可以有效地抑制无核白葡萄的膜脂过氧化作用对细胞膜的破坏,保持细胞的完整性,且PPO活性较低,从而减少无核白葡萄脱水褐变的发生。研究结果为快速脱水在无核白葡萄干制中的应用提供参考。  相似文献

Chlozolinate (Serinal) is a dicarboximide fungicide used in southern European countries principally on grapes. Maximum residue levels have not yet been set by FAO/WHO and are under evaluation in the EU. Field trials have been carried out in Greece on two varieties of table grapes (Cardinal and Victoria) during two consecutive years to assess residues remaining after application according to good agricultural practice. Analysis using a multiresidue method with gas chromatography (ECD) showed that the parent compound decays with a first-order rate constant of 0.057 +/- 0.011 day(-)(1) and that residues had fallen below the proposed MRL of 5 mg/kg in all samples by 21 days postapplication (the proposed PHI). The contribution of the main metabolite, S1, to the total residue is generally <20%. Washing removes a substantial amount (up to 80%) of chlozolinate, which appears to be nonsystemic on grapes, thus reducing real consumer exposure to this pesticide.  相似文献

Dissipation of the fungicide spiroxamine in grapes of two vine varieties, Roditis and Cabernet Sauvignon, exposed to field treatments was evaluated. Vines of a grape vineyard located in central Greece were sprayed once or twice with a commercial formulation of the fungicide at 30 g a.i./hL. Residues in grapes, must, and wine were determined by gas chromatography/IT-MS after extraction with cyclohexane-dichloromethane (9:1), with a limit of quantitation 0.02 mg/kg in grapes and 0.012 mg/kg in wine. Under field conditions, spiroxamine dissipation on grapes was faster during the first 2 weeks and then slower to the sixth week. About 7 days after application, half of the initial spiroxamine concentration remained on the grapes; the respective proportion at 42 days was about 10%. At 14 and 35 days, residues were lower than 0.44 and 0.22 mg/kg, respectively, values below the maximum residue levels set by the European Union (1 mg/kg). Spiroxamine residues transferred from grapes into the must and through the vinification process into the wine were also studied. Mean transfer factors of 0.26 and 0.55 were found from grapes into wine for the wines obtained without maceration and with maceration, respectively. Residues in wine, prepared from grapes with a spiroxamine content of 0.11-0.20 mg/kg, varied from <0.026 to 0.09 mg/kg. Spiroxamine diastereomer B was found to dissipate slower than diastereomer A in the field as well as during the vinification process.  相似文献

Quinoxyfen is a new fungicide that belongs to the family of the quinolines, recently introduced to control powdery mildew (Uncinula necator). In this paper the fate of quinoxyfen residues from vine to wine and in their processing products was studied. After the last of four applications at the recommended rate, 0.38 mg/kg of residue was found on the grapes, which is under the legal limit fixed in Italy (0.5 mg/kg). The degradation rate was according to a pseudo-first-order kinetics (r = 0.964) and the half-life was 7.24 days. Vinification was carried out with and without maceration. During the vinifications without maceration <50% of the residues passed from the grapes to the musts. Separation of the lees (8%) from the must by centrifugation caused no detectable residues in centrifuged must. At the end of fermentation with and without maceration no quinoxyfen residues were determinable in the wine. No effect on the alcoholic or malolactic fermentation was observed even in the presence of higher quinoxyfen concentrations than those found in the grapes at harvest time. During fermentation, the yeasts partially degraded the pesticides and completly adsorbed them. Bacteria, on the other hand, do not have any degradative effect on the pesticides. The raisins obtained by sun-drying did not contain any residues, whereas those obtained by oven-drying show the same amount of residues as in the fresh grapes. During the sun-drying process the fruit weight decreased by a factor of 4; the decrease in the oven-drying was equivalent. Samples of dregs and liquid lees, fortified with high levels of quinoxyfen. were double-distilled. The first dregs distillate, with an alcohol content of 32.1%, did not show any residues, whereas the first lees distillate, with an alcohol content of 34.5%, showed 7% of the initial residues. After the second lees distillation, the obtained product showed an alcoholic content of 81.2% and no residues of quinoxyfen (<0.01 mg/kg).  相似文献

Grapes and raisins provide phenolic antioxidants, which contribute to their potential health benefits. The objectives of this study were to compare the antioxidant capacity and phenolic content of green Thompson seedless grapes (the most common variety of grapes consumed in the United States), sun-dried raisins, and golden raisins (both produced from Thompson seedless grapes) and to observe the effects of their consumption over 4 weeks in 15 healthy human males with a cross-over design. The oxygen radical absorbance capacity (ORAC) (positive statistical significance for grapes after 2 weeks and golden raisins after 3 weeks), serum oxidation (positive statistical significance for golden raisin lag time after 4 weeks), total phenolics (no significant effects), and C-reactive protein (no significant effects) were monitored. Immediately postconsumption, there were some significant nonpositive changes. It is hypothesized that these negative results may be explained by postprandial oxidation, a known effect after carbohydrate consumption. Golden raisins had the highest antioxidant capacity and phenolic content. The consumption of a serving of grapes or raisins each day, in addition to a typical diet, may not be sufficient to overcome postprandial oxidation when consumed with other high carbohydrate foods but may have beneficial antioxidant effects over time.  相似文献

Dissipation of benzoylurea insecticide teflubenzuron in grapes exposed to field treatments was evaluated. Vines of a white grape vineyard located in central Greece were sprayed twice, at a 28-day interval, with a commercial formulation of the insecticide at 12 g of active ingredient/100 L. Residues were determined by HPLC diode array detection, after ethyl acetate extraction and cleanup on silica phase cartridges, with a detection limit of 0.005 mg/kg. Under field conditions teflubenzuron residues in grapes were found to be very stable with no significant reduction for the whole experimental period of 49 days. However, residues were slightly lower than the maximum residue limits set by individual countries. The fate of teflubenzuron during the vinification process was also studied. Residues were found to be transferred completely into the must but, due to their high affinity for the suspended matter, were removed by approximately 98%; thus, very low concentrations were detected in the produced wine. Among various clarifying agents studied, charcoal was the only one found to be effective for removing teflubenzuron residues from wine.  相似文献

Residues of ethylenethiourea (ETU) in grapes and wine were determined by capillary gas chromatography and paper chromatography, without a cleanup step, and after derivatization to S-benzyl-ETU. The detection limit was 0.0002 mg/kg for flame ionization detection, 0.008 mg/kg for paper chromatography with photodensitometric evaluation of the detected spot. Results were compared with a generally used GC method specifying electron capture detection of trifluoroacetylated S-benzyl-ETU. The recoveries of ETU in grapes and wine at different concentration levels were determined. ETU residues were determined in treated grapes but no residues were detected in wine.  相似文献

In a 2 year experiment, residues in field-grown strawberries were investigated from the fungicides fenhexamid, pyrimethanil, tolylfluanid, and kresoxim-methyl resulting from different strategies, as regards the dose, number, and time of fungicide applications. Kresoxim-methyl was only used the first year and in full or no dose to control powdery mildew. In the first year, the highest concentrations analyzed were 0.66 mg kg(-1) for pyrimethanil and 0.63 mg kg(-1) for fenhexamid resulting from the use of recommended dose rates and a preharvest interval (PHI) of 10 days, thus not exceeding the Danish maximum residue limit (MRL) of 1 mg kg(-1). Tolylfluanid was used no later than 21 days before harvest, which left residue contents in the berries of 0.48 mg kg(-1), a value well below the MRL of 5 mg kg(-1). In the second year, fungicide residues found in the samples were generally lower, 0.39 and 0.03 mg kg(-1) for pyrimethanil and fenhexamid, respectively. No residues of kresoxim-methyl were found in any of the samples from the field trials, indicating that kresoxim-methyl residues had declined to a level well below the detection limit within the 28 day period between the last application and the harvest.  相似文献

Pesticide residues in grapes, wine, and their processing products   总被引:10,自引:0,他引:10  
In this review the results obtained in the 1990s from research on the behavior of pesticide residues on grapes, from treatment to harvest, and their fate in drying, wine-making, and alcoholic beverage processing are reported. The fungicide residues on grapes (cyproconazole, hexaconazole, kresoxim-methyl, myclobutanil, penconazole, tetraconazole, and triadimenol), the application rates of which were of a few tens of grams per hectare, were very low after treatment and were not detectable at harvest. Pyrimethanil residues were constant up to harvest, whereas fluazinam, cyprodinil, mepanipyrim, azoxystrobin, and fludioxonil showed different disappearance rates (t(1/2) = 4.3, 12, 12.8, 15.2, and 24 days, respectively). The decay rate of the organophosphorus insecticides was very fast with t(1/2) ranging between 0.97 and 3.84 days. The drying process determined a fruit concentration of 4 times. Despite this, the residue levels of benalaxyl, phosalone, metalaxyl, and procymidone on sun-dried grapes equalled those on the fresh grape, whereas they were higher for iprodione (1.6 times) and lower for vinclozolin and dimethoate (one-third and one-fifth, respectively). In the oven-drying process, benalaxyl, metalaxyl, and vinclozolin showed the same residue value in the fresh and dried fruit, whereas iprodione and procymidone resides were lower in raisins than in the fresh fruit. The wine-making process begins with the pressing of grapes. From this moment onward, because the pesticide on the grape surface comes into contact with the must, it is in a biphasic system, made up of a liquid phase (the must) and a solid phase (cake and lees), and will be apportioned between the two phases. The new fungicides have shown no effect on alcoholic or malolactic fermentation. In some cases the presence of pesticides has also stimulated the yeasts, especially Kloeckera apiculata, to produce more alcohol. After fermentation, pesticide residues in wine were always smaller than those on the grapes and in the must, except for those pesticides that did not have a preferential partition between liquid and solid phase (azoxystrobin, dimethoate, and pyrimethanil) and were present in wine at the same concentration as on the grapes. In some cases (mepanipyrim, fluazinam, and chlorpyrifos) no detectable residues were found in the wines at the end of fermentation. From a comparison of residues in wine obtained by vinification with and without skins, it can be seen that their values were generally not different. Among the clarifying substances commonly used in wine (bentonite, charcoal, gelatin, polyvinylpolypyrrolidone, potassium caseinate, and colloidal silicon dioxide), charcoal allowed the complete elimination of most pesticides, especially at low levels, whereas the other clarifying substances were ineffective. Wine and its byproducts (cake and lees) are used in the industry to produce alcohol and alcoholic beverages. Fenthion, quinalphos, and vinclozolin pass into the distillate from the lees only if present at very high concentrations, but with a very low transfer percantage (2, 1, and 0.1%, respectively). No residue passed from the cake into the distillate, whereas fenthion and vinclozolin pass from the wine, but only at low transfer percentages (13 and 5%, respectively).  相似文献

Dissipation of residues of benzoylurea insecticides teflubenzuron (TFB) and triflumuron (TFM) under field conditions was evaluated on a pear orchard in Greece. Residues were determined by UV-HPLC analysis, with a detection limit of 0.030 mg/kg for both pesticides. TFB residues in pears were found to persist for 2 weeks and decline thereafter with 48% of the initial deposit remaining 42 days after the last application. TFM residues were found to decline following first-order kinetics and with a half-life of 39(+/-7) days. Residues of both pesticides found in pears collected at harvest maturity were lower than the maximum residue limits (MRLs) set by individual countries. Dissipation of TFB and TFM in cold-stored pears was also evaluated. TFB residues were very persistent for the whole storage period, whereas TFM residues did not dissipate for 6 weeks and then showed a constant decline; 7% of the initial concentration remained at the end of the storage period of 29 weeks.  相似文献

Raisins are dried grapes that are popular shelf-stable snacks. Three commercially important types of raisins were studied: sun-dried (natural), artificially dried (dipped), and sulfur dioxide-treated (golden) raisins. Dietary fiber composition was analyzed by AACC method 32-25. Polysaccharides were hydrolyzed, and the resulting sugars were analyzed by colorimetric and gas chomatographic methods. Fructans were measured with a colorimetric kit assay. Total dietary fiber values agreed with published values, with pectins and neutral polysaccharides of mannose and glucose residues predominating. Dipped raisins had over 8% fructans. No fructans were found in fresh grapes. Raisin types varied in their ability to bind bile acids in vitro. Coarsely chopped raisins bound more bile than did finely chopped or whole raisins.  相似文献

Concerns that raisins may be contaminated by fumonisins stem from the persistent occurrence of Aspergillus niger spores on raisins and the recent discovery of fumonisin production by A. niger on grapes, which leads to the widespread occurrence of fumonisin B(2) in wine. This study presents an LC-MS/MS survey of fumonisins in retail raisins. In 10 of 21 brands collected in Denmark, Germany, and The Netherlands, fumonisins B(2) and B(4) were detected at levels up to 13 and 1.3 μg/kg, respectively. Only fumonisin B(2) has been detected in wine, so the presence of fumonisin B(4) in raisins suggests that the fumonisins are produced mainly during the drying process concomitant with the decreasing water activity. Analysis of multiple packages from one manufacturer showed a 3-fold package-to-package variation, suggesting that a few raisins per package are contaminated.  相似文献

Maximum residue limits (MRLs) have been established by the European Union when tylosin is used therapeutically. They are fixed at 200 microg/kg for eggs. A highly sensitive and selective quantitative liquid chromatography electrospray ionization tandem mass spectrometry (LC/ESI/MS/MS) method suitable for monitoring tylosin residues in eggs to determine its depletion kinetics was developed and validated. For sample pretreatment all samples were liquid-liquid extracted with citrate buffer (pH 5.0) and acetonitrile. Liquid chromatographic separation was carried out on a reversed phase C18 column employing a 0.5% formic acid/acetonitrile gradient system. The tylosin recovery in eggs at a concentration range from 1.0-400 microg/kg was >82% with relative standard deviations between 1.5 and 11.0%. In two experimental studies administrating tylosin via feed (final dosage: 1.5 g/kg) or drinking water (final dosage: 0.5 g/L), no residues above the MRL were found during and after treatment. Moreover, all samples were well below the actual MRL of 200 microg/kg. Therefore, our residue data suggest that a withholding period for eggs is not required when laying hens are treated with tylosin in recommended dosages via feed or drinking water. The broad spectrum, systemic fungicides tebuconazole and propiconazole are used to control rust in boronia (Boronia megastigma Nees). Gas chromatography combined with either a benchtop quadrupole mass spectrometer or a high-resolution mass spectrometer allowed for the monitoring of both pesticides in boronia leaves, flowers, and concrete. Field trials were established at two sites to determine the rate of dissipation of tebuconazole and propiconazole in boronia. At site 1, two application rates of 125 and 250 g active ingredient/hectare (ai/ha) tebuconazole were employed. Treatments were repeated 17 days later. At harvest, 286 days after the final application, tebuconazole was detected at levels of 0.06 +/- 0.05 and 0.5 +/- 0.1 [mg/kg +/- standard error, on a dry matter basis (DMB)] in the leaves collected from plots treated with 125 and 250 g ai/ha of tebuconazole, respectively. The oil produced from the flowers collected at the final harvest had residues of tebuconazole at levels of 0.06 +/- 0.03 and 0.10 +/- 0.08 mg/kg for the 125 and 250 g ai/ha application rates, respectively. Two repeat applications of 125 g ai/ha propiconazole were also used at site 1. Residues of propiconazole were detected at 0.09 +/- 0.03 mg/kg (DMB) 286 days after the final application. At site 2, treatments of 125 g ai/ha of tebuconazole were applied twice. At harvest, 279 days after the final application of tebuconazole, residues were recorded at 0.30 +/- 0.09 mg/kg in the leaves (DMB) while the oil produced had 0.20 +/- 0.07 mg/kg.  相似文献

The behavior in field and the transfer from grapes to wine during winemaking of iprovalicarb, indoxacarb, and boscalid was studied. The residue levels found in grapes were far below the MRLs set for grapes in EU, accounting at harvest time 0.81, 0.43, and 4.23 mg/kg for iprovalicarb, indoxacarb, and boscalid, respectively. The residue levels in the samples treated with boscalid may have residual problems (due to an accumulation effect) if repeated field treatments will be performed. Winemaking experiments showed a complete transfer of all pesticide from grapes to the must, while in wine the residues were low or negligible due to the adsorbing effect of lees and pomace. The clarification experiments showed a good removal of pesticide residues from the wine media, for all pesticides. The GC-ITMS method showed good performance with adequate recoveries ranging from 75 to 115%, and good method limits of quantitation (LOQs) and of determination (LODs) far below MRLs.  相似文献

Residue levels and degradation rates of procymidone residues were studied in green beans grown in a greenhouse. Experiments were planned to also assess the influence of planting density on the behavior of procymidone residues on this type of crop. The study was carried out in four random blocks considering three sub-blocks of different planting densities into each block. Plants were sprayed with Sumisclex 50 WP (1077.5 g of ai/ha) 52 days after the transplantation, and sampling was carried out daily during two different periods of 6 and 5 consecutive days, respectively, around the two harvest days (days 12 and 28 after the treatment). Residue levels of procymidone were determined by using the Luke extraction method and GC-NPD. The average residue levels of procymidone in the overall planting (mean of 12 determinations) were below 2 mg/kg (European maximum residue limit) for all the sampling days, obtaining values of 1.01 +/- 0.55 and 0.37 +/- 0.10 mg/kg, respectively, at the two harvest days. The decline behavior of procymidone residues in the overall plantation and in each block could be described as a pseudo-first-order reaction, obtaining half-life values (t(1/2)) of 10-11 days in all cases. The calculated residue level at the preharvest time (5 days) in the overall plantation was 1.7 mg/kg, but this value in the blocks depended on the block position along the greenhouse and ranged from 2.3 to 0.9 mg/kg. In this work, additional data on the residual behavior of the fungicide pyrazophos in green beans were also obtained.  相似文献

The results of residue determinations of the growth promotors carbadox, tylosin, and virginiamycin in kidney, liver, and muscle from pigs in feeding experiments are described as well as the analytical methods used. Residues of the carbadox metabolite quinoxaline-2-carboxylic acid were found in liver from pigs fed 20 mg/kg in the diet with a withdrawal time of 30 days. No residues were detected in muscle with zero withdrawal time. The limit of determination was 0.01 mg/kg for both tissues. No residues of virginiamycin and tylosin were found in pigs fed 50 and 40 mg/kg, respectively, in the diet, even with zero withdrawal time. Residues of tylosin of 0.06 mg/kg and below were detected in liver and kidney from pigs fed 200 or 400 mg/kg and slaughtered within 3 h after the last feeding.  相似文献

Two fungicides (cyprodinil and fludioxonil) have recently been used in southeast Spain to control disease in lettuce and grape. Gas chromatography with a nitrogen-phosphorus detector (GC-NPD) was used to study the disappearance of these compounds from crops under field conditions and during refrigeration. Residual values 21 days after application were below the maximum residue limit (MRL = 0.05 mg kg(-1)) established by Spanish law in the field experiment for both compounds. However, with the exception of fludioxonil in lettuce, residues were above the MRL in the refrigerated farm produce for both fungicides. The half-lives were 3-6 times greater under refrigeration.  相似文献

采用液质联用仪比较分析了3个不同种植区域(江苏南京、广西南宁和湖南长沙)露地和大棚两种种植条件下黄瓜和土壤中甲基硫菌灵及其代谢物多菌灵的残留动态,同时对黄瓜中的最终残留量进行了比较分析。施药后,甲基硫菌灵在黄瓜和土壤中均能很快转化为多菌灵[施药后1 d甲基硫菌灵未检出(〈0.01 mg·kg-1)],多菌灵在露地黄瓜和土壤中的原始沉积量均低于大棚。3个试验点露地黄瓜中的半衰期分别为2.3、1.4 d和1.4 d,在大棚黄瓜中的半衰期分别为2.6、1.7 d和2.0 d。在3个试验点露地土壤中的半衰期分别为1.6、1.7 d和2.3 d,在大棚土壤中的半衰期分别为2.3、2.0 d和2.3 d。最终残留试验在最后一次施药后1 d采样时,大棚、露地黄瓜中的甲基硫菌灵均未检出(〈0.01 mg·kg-1),多菌灵在3个试验点露地黄瓜中的最终残留量为0.014~0.162 mg·kg-1,而在3个试验点大棚黄瓜中的最终残留量为0.121~0.561 mg·kg-1。参照我国所制定的黄瓜中多菌灵的MRL(0.5 mg·kg-1),露地种植方式下所有处理黄瓜中甲基硫菌灵代谢物多菌灵的最终残留量均符合国家标准的规定,但大棚种植方式下其残留量有超标的风险。  相似文献

Chlorpropham, a herbicide and sprout suppressant, is used on stored potatoes to prolong the storage period without deterioration of produce quality. Data for residue concentrations on an individual tuber basis are required by WHO for the estimation of the variability factor. In this study, the levels of chlorpropham in individual tubers and in composite samples were determined. The distribution of chlorpropham between the peel and the tuber flesh was examined, and the fate during the cooking process (washing, boiling, frying) was studied. The concentrations in individual tubers ranged from 1.8 to 7.6 mg/kg 10 days postapplication (mean 3.8 mg/kg, RSD 39%), from 0.7 to 4.0 mg/kg 28 days postapplication (mean 2.9 mg/kg, RSD 28%), and from 0.8 to 3.8 mg/kg 65 days postapplication (mean 2.2 mg/kg, RSD 48%). The calculated residues in composite samples 10 days postapplication ranged from 4.3 to 6.1 mg/kg (mean 4.9 mg/kg, RSD 20%). Those in samples taken 28 days postapplication ranged from 3.1 to 4.2 mg/kg (mean 3.8 mg/kg, RSD 15%). The concentrations determined in composite samples of whole tubers 65 days postapplication ranged between 2.6 and 3.2 (mean 2.9 mg/kg, RSD 11%). Peeling removed 91-98% of the total residue; washing reduced residues by 33-47%. Detectable residues were found in boiled potatoes and the boiling water, and in french fries and the frying oil. Monitoring data on commercial prefried frozen french fries are reported.  相似文献
