The selection of meat quality has received considerable focus in chicken breeding. This study was aimed at investigating the effect of CAPN1 gene on meat quality traits in chicken populations. Primer pairs for 3 UTR in CAPN1 were designed from database of chicken genomic sequence. Polymorphisms were detected using PCR-SSCP and DNA sequencing. A mutation at position 9 950 nt (G/A, locus A) was found among individuals in each population. The allele and genotype frequencies significantly differed among eight lines with higher frequencies of allele A2 and genotype AIA2 (P〈0.01). The least square analysis showed that there was significant difference (P 〈 0.05) in muscle fiber density and some carcass traits among genotypes and that the breast muscle fiber density (BFD) of birds ofAIA1 genotype was significantly higher (P〈0.05) than that of birds ofA2A2 genotype. It was concluded that the CAPN1 gene was the major gene affecting the muscle fiber traits of chicken or was linked with the major gene. These results were useful for studying the molecular mechanism that influences meat traits and were used as the base of molecular-assisted selection to meat quality traits. So, this site may be a potential marker affecting the muscle traits of chickens.  相似文献

This study was carried out at the Genetic Engineering Lab, College of Agriculture, University of Basrah. DNA was extracted from samples containing meat of commercial products in the Basrah markets. The products subjected to analysis using polymerase chain reaction (PCR) with species specific repeat (SSR) and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) techniques based on the sequence of the mitochondrial DNA cytochrome b gene (mtDNA cyt b gene). Then digestion with the Alu l restriction enzyme to establish a differential diagnosis detects and discriminates between meat species and adulteration in the products. SSR primers were applied, has been detected amplification of the encoded gene product, generated 221 bp in some imported minced and canned meat samples. The results show that SSR analysis produced a pattern that allowed a direct identification of horse and donkey meats in some imported minced and canned meat samples (Hana, Monde and Bavaria). The amplified 359 bp gene of mtDNA cyt b gene from samples in different product was cut using Alu 1 restriction enzyme resulting in restriction fragment length polymorphism (RFLP). Alu 1 was used to distinguish between the animals meat that belong to the family or one species. The digestion of the PCR product showed differences between products. Where the fragment length generated were 74, 76 and 189 bp. It belonged to horse meat. The fraud was detected in Hana, Monde and Bavaria products available in Basrah markets showing the presence of horse meat in these products that labeled as beef meats 100%. This revealed mtDNA cyt b gene as highly conserved and consequently a good molecular marker for diagnostic studies. Thus, this technique can be applied to food authentication for the identification of different species of animals in food products.  相似文献

The purpose of this study is to explore the influence of co-suppressing tomato ACC oxidase Ⅰ on the expression of fruit ripening-related and pathogenesis-related protein genes, and on the biosynthesis of endogenous ethylene and storage ability of fruits. Specific fragments of several fruit ripening-related and pathogenesis-related protein genes from tomato (Lycopersicon esculentum) were cloned, such as the l-aminocyclopropane-1-carboxylic acid oxidase 1 gene (LeAC01), 1- aminocyclopropane-l-carboxylic acid oxidase 3 gene (LeAC03), EIN3-binding F-box 1 gene (LeEBF1), pathogenesis-related protein 1 gene (LePR1), pathogenesis-related protein 5 gene (LePR5), and pathogenesis-related protein osmotin precursor gene (LeNP24) by PCR or RT-PCR. Then these specific DNA fragments were used as probes to hybridize with the total RNAs extracted from the wild type tomato Ailsa Craig (AC++) and the LeAC01 co-suppression tomatoes (V1187 and T4B), respectively. At the same time, ethylene production measurement and storage experiment of tomato fruits were carded out. The hybridization results indicated that the expression of fruit ripening-related genes such as LeACO3 and LeEBF1, and pathogenesis-related protein genes such as LePR1, LePR5, and LeNP24, were reduced sharply, and the ethylene production in the fruits, wounded leaves decreased and the storage time of ripening fruits was prolonged, when the expression of LeACO1 gene in the transgenic tomato was suppressed. In the co-suppression tomatoes, the expression of fruit ripening-related and pathogenesis-related protein genes were restrained at different degrees, the biosynthesis of endogenous ethylene decreased and the storage ability of tomato fruits increased.  相似文献

Dietary amino acids imbalance will result in stunted broiler performance and deteriorated meat quality, which are involved in various biochemical cycles in vivo. In this study, the effects of dietary methionine on meat quality and methylation of myostatin exon 1 were investigated. Drip loss of the broilers fed with diet of high methionine levels (0.2%) increased from (6.3 ± 0.1)% (control group) to (10.1 ± 1.0)%, and the muscle shearing force increased from (22.8 ± 1.9) N (control group) to (26.3 ±2.3) N. Moreover, many CpG sites were found at the myostatin exon 1 region (nucleotides 2 360-2 540 bp). To further understand the regulation of broiler myostatin expression, the methylation status of broiler myostatin exon 1 and its mRNA expression were analyzed. At the myostatin exon 1 region where CG enriches (nucleotides 2 360-2 540 bp), the percentages of methylation were 46 and 84% in low Met and high Met content groups after 55-d feeding, respectively. In skeletal muscle tissues, the exon 1 hypermethylation status of myostatin gene was found to be negatively correlated with the gene expression. These results suggested that methylation of this gene is a dynamic process, which plays a dominant role in regulating gene expression for development of individuals.  相似文献

The plant hormone abscisic acid (ABA) regulates many important physiological and developmental processes in plants. The objective of this study was to clone the ABA 8′-hydroxylase gene in common wheat. In the present study, we used the eDNA sequence of barley HvCYP707A1 gene (GenBank accession no. AB239299) as a probe for BLAST search against the common wheat (Triticum aestivum L.) EST database in GenBank. All wheat ESTs sharing high similarity with the reference gene were subjected to contig assembly. Primers were designed based on the constructed contigs to clone the wheat CYP707A1 gene, designated as TaCYP707A1. The genomic DNA sequence of TaCYPTO7A1 gene comprised five exons and four introns, with a size of 2225 bp. The corresponding cDNA sequence of TaCYP707A1 was 1737 bp, containing an open reading frame (ORF) of 1431 bp, a 42-bp 5′-untranslated region (UTR) and a 264-bp 3′UTR, with 94.9% of identical sequences to HvCYP707A1 gene (AB239299). The neighbor joining tree indicated that the deduced amino acid sequences of TaCYP707A1 gene was highly similar to those of barley and rice. The TaCYP707A1 gene was located on chromosome 6BL using a set of Chinese Spring nullisomic-tetrasomic lines and ditelosomic line 6BS. These results will be of high importance in understanding of molecular mechanism of ABA catabolism.  相似文献

The promoter region (BCSP666) of β-conglycinin α-subunit gene from the genomic DNA of soybean Jilin 43 was isolated by PCR method. Sequencing analysis showed that the cloned fragment BCSP666 had the similar structure to the soybean seed-specific promoter β-conglycinin α'-subunit gene promoter and β-conglycinin β-subunit gene promoter, and it also contains many motifs that contribute to the seed-specific promoter activity. Based on this sequencing analysis, we deduced that promoter fragment BCSP666 had the seed-sepecific promoter activity. And then we constructed the seedspecific expression vector pBMI666 with the promoter fragment BCSP666 and △6-fatty acid desaturase gene from Mortierella isabellina. The △6-fatty acid desaturase is the rate-limiting enzyme of the desaturation of linoleic acid in the production of a human essential fatty acid, γ-linolenic acid(GLA). The production of γ-linolenic acid(GLA) was observed in soybean callus cells, which were transformed with this vector. This confirmed the activity of the activity fragment BCSP666.  相似文献

The aim of this research was to evaluate the dietary supplementation 1% (w/w) of oregano and sage dried leaves on performances and meat quality of broiler rabbits. A feeding trial, which lasted 48 d, was carried out on 105 male Bianca Italiana rabbits randomly divided in seven groups and fed ad libitum. At the end of the trial ten animals per group were slaughtered and samples of dorsal muscle were taken in order to perform laboratory analysis. Mortality rate did not statistically differ between groups. Growth performances of animals fed diets supplemented with aromatic plants were higher (P〈0.05) than those of animals of control group, whereas carcass parameters were not affected by treatments excepting for the slaughter weight that showed the same trend as growth performances. Meat quality traits, oxidative lipid stability and fatty acid proifle were not inlfuenced by aromatic plant supplementation. In conclusion, oregano and sage in form of dried leaves can be used in rabbit without adverse effects on performance, carcass characteristics and meat quality traits.  相似文献

采用胶原酶消化法分离猪皮下前脂肪细胞,以含50 nmol/L胰岛素、100 nmol/L地塞米松、0.25 mmol/L 3-异丁基-1-甲基黄嘌呤、100 nmol/L罗格列酮的分化培养液对前脂肪细胞进行诱导分化,借助实时定量RT-PCR方法检测了PPARγ、DECR1和ECHS1 3基因的表达模式,结果发现:PPARγ、DECR1和ECHS1 3基因均于诱导分化后48 h达到表达高峰,此时的表达量分别是诱导前的93.0,5.4和7.8倍;PPARγ同DECR1和ECHS1两基因表达量变化趋势间的相关系数分别为0.92和0.97,DECR1同ECHS1的相关系数为0.94,均达到极显著相关(P<0.01)。实验结果表明:前脂肪细胞在分化过程中细胞内脂肪酸生物合成和脂肪酸β-氧化同时发生,以维持细胞的稳态。  相似文献

PHKG1基因g.8283 C>A位点是影响猪肉糖原酵解潜能及系水力的因果突变位点。为评估该位点是否对其他生产性状有影响,采用RFLP技术检测该基因位点在153头杜洛克猪群体中的多态性,并分析不同基因型对体质量、日增重、背膘厚、眼肌面积及瘦肉率的影响。结果表明:PHKG1 g.8283 C>A位点CC、AC、AA 3种基因型频率分布分别为0.320、0.536、0.144,优势等位基因C的频率为0.588,劣势等位基因A的频率为0.412;PHKG1不同基因型对体质量、日增重、背膘厚、眼肌面积、瘦肉率均无显著影响(P>0.05)。据此,为提高肉质性状,在兼顾体型外貌及性能测定的前提下,可尽快提高优势等位基因C的频率。  相似文献

MyoD基因家族能促使肌肉转化,影响着肉质品质.为了研究MyoD1与肉质性状间的关系,采用PCR-SSCP技术分析杜长大三元杂交猪MyoD1基因第1外显子的单核苷酸多态性.为了将第1外显子(长630 bp)全部扩出,故将第1对引物设在MyoD1基因的5'UTR上.结果显示:MyoD1基因5'UTR的119 bp处发现G>A的突变,第1外显子387 bp处发现C>T突变,经检测这2个突变均为AA和AB基因型,2个变异位点在群体中都是AA型为优势基因型,且都处于Hardy Weinberg平衡状态.在5'UTR上的突变对猪肌肉的肌内脂肪和pH的影响差异显著(P<0.05),而在第1外显子上突变对本试验所研究的肉质性状影响均不显著(P>0.05).  相似文献

旨在研究反转录转座子1基因(retrotransposon-like-1,RTL1)潜在的变异与猪胴体和肉质性状的相关性。测序发现在猪RTL1基因编码区1 209 bp处存在G/A突变,利用PCR-Fnu4H I-RFLP方法在9个猪种中进行了基因分型,并且在360头大白猪×梅山猪F2代群体中进行性状关联分析。结果表明,该位点与皮率、内脂率、6-7腰椎间膘厚、胸腰椎间膘厚、背最长肌pH、股二头肌肌肉色值呈显著相关(P0.05),与骨率、肩部膘厚、臀部膘厚、平均背膘厚、背最长肌失水率、背最长肌系水力、背最长肌肌肉色值呈极显著相关(P0.01)。  相似文献

以长×大白猪为对照,对野莱(长白山野猪×莱芜猪)F1代猪的肉质特性进行了系统的研究。结果表明:野莱F1代猪肉的肉色、大理石纹、失水率、滴水损失、熟肉率、剪切值和肌内脂肪含量均显著(P<0.05)或极显著(P<0.01)优于对照;野莱F1代猪肉的饱和脂肪酸含量极显著低于对照,而多不饱和脂肪酸含量尤其是人体必需脂肪酸含量极显著高于对照;野莱F1代猪肉的肌纤维比对照细,肌小节比对照长(P<0.01);除脯氨酸外,野莱F1代猪肉的其他17种氨基酸含量、氨基酸总量、必需+半必需氨基酸含量、鲜味氨基酸含量及肌苷酸含量均显著或极显著高于对照。因此,野莱F1代猪肉的品质和营养价值均比长×大白猪好。  相似文献
