AIM: To investigate the role of anti-miR-21 oligonucleotide (AMO) in the anti-leukemic activity of decitabine (DCA) in vitro.METHODS: AMO and scramble oligonucleotide (SCR) were constructed and transfected into HL-60 cells. The miR-21 expression was analyzed by real-time PCR to identify the transfection efficiency. The cells were treated with DCA at gradient concentrations (0.5, 2.0 and 4.0 μmol/L) for 48 h. The mRNA expression of human period circadian protein 3 (hPer3) was detected by real-time PCR. The early apoptotic rates were determined by flow cytometry with Annexin V/PI staining. Mean fluorescence intensities (MFI) of CD117 and CD11b were also measured by flow cytometry. RESULTS: The miR-21 relative expression level in AMO group was significantly lower than that in blank group and SCR group (P<0.01). IC50 of DCA in AMO group was significantly lower than that in blank group and SCR group (P<0.01).With the same concentration of DCA, the early apoptotic rate, the mRNA expression of hPer3 and the MFI of CD11b in AMO group were significantly higher than those in blank group and SCR group (P<0.01). The MFI of CD117 in AMO group were significantly lower than those in blank group and SCR group (P<0.01). CONCLUSION: Activation of hPer3 expression plays an important role in enhanced anti-leukemic activity of decitabine by AMO in vitro.  相似文献

甘蓝自交不亲和决定因子的体外表达和相互作用的检测   总被引:5,自引:3,他引:2  
 S位点受体激酶(SRK)和S位点富含半胱氨酸蛋白/S位点蛋白11(SCR/SP11)分别为甘蓝自交不亲和(self-incompatibility, SI)信号传导的雌雄决定因子。为了深入研究两者的作用机理和进行人工调控,本研究以结球甘蓝ZQ为材料,利用pET·NusA融合蛋白表达系统,将包含SRK胞外域和跨膜域的mSRK蛋白和SCR蛋白在大肠杆菌BL21中融合表达,经SDS-PAGE电泳检测表达出融合蛋白大小分别约为116 kD和74 kD。进一步将两融合蛋白进行体外相互作用的检测,结果表明mSRK与SCR蛋白能够相互结合形成稳定的复合体,这为下一步实现SRK-SCR复合体聚合与解离的人为调控提供了技术平台。  相似文献

采用PCR法,从水仙(Narcissus tazetta var. chinensis)鳞茎主芽中分离克隆到1个温度诱导脂质运载蛋白基因NtTIL。该基因含有567 bp开放阅读框,编码188个氨基酸。该基因属于lipocalin-2 superfamily基因家族;推导的氨基酸序列含有SCR1、SCR2和SCR3 3个植物lipocalin的典型结构域,与小麦、拟南芥等植物的同源性大于75%;编码的蛋白为稳定酸性亲水蛋白,不含有信号肽,进行N末端朝外由外到内的跨膜运动。qRT-PCR结果分析表明,水仙生长发育过程中鳞茎膨大期NtTIL表达量较低;休眠期表达量先升高后降低。推测NtTIL对水仙鳞茎主芽休眠具有一定调控作用。  相似文献

AIM:To investigate the role of tiny antisense nucleic acid against miR-155 (tiny antimiR-155, t-antimiR-155) in multiple myeloma cells. METHODS:According to the seed sequence of miR-155, t-antimiR-155 was designed and synthesized. t-antimiR-155 was transfected by LipofectamineTM 2000 into RPMI-8266 cells. The cells were divided into t-antimiR-155 group, scrambled control (SCR) group and blank control group. The growth-inhibitory potencies were measured by MTT assay. The ability of cell colony formation was detected by cell colony formation assay. The cell apoptosis was assessed by flow cytometry with annexin V/PI double staining. RESULTS:The best concentration and time were 0.4 μmol/L and 48 h, respectively. The cell colony forming experiment showed that the circumstances of forming cell community in t-antimiR-155 group was weaker than that in SCR group, and the colony formation inhibitory rate of former was significant higher than the latter. Compared with SCR group, the cell apoptosis in t-antimiR-155 group significantly increased. CONCLUSION:The t-antimiR-155 inhibits the progression of multiple myeloma cells by interfering with miR-155. miR-155 may serve as a potential target in gene therapy for treating multiple myeloma.  相似文献

eSRK(S位点受体激酶胞外域)、SCR/SP11(S位点富含半胱氨酸蛋白-S位点蛋白11)和THL1(类硫氧还蛋白)是甘蓝自交不亲和性信号传导过程重要的3个元件。利用PCR技术获得两种甘蓝SCR2(class-Ⅱ)、eSRK2(class-Ⅱ)、THL1、SRKJ(class-Ⅰ,SRK6激酶域)和eSRK28(class-Ⅰ)的编码序列,并分别构建以pGBKT7为载体的SCR2、THL1的重组诱饵质粒和以pGADT7为载体的eSRK2、eSRK28、SRKJ的重组猎物质粒。利用酵母双杂交系统验证eSRK2-SCR2、eSRK28-SCR2、SRKJ-THL1的相互作用。重组质粒在宿主细胞中未产生毒性及自激活作用,表明重组表达载体构建成功;组合Y2HGold〔pGBKT7-SCR2〕×Y187〔pGADT7-eSRK2〕在三缺平板(SD/-Trp-Leu-His/x-α-gal/AbA,TDO/x/A)上生长出蓝色克隆,激活了报告基因AUR1-C、MEL1、HIS3;组合Y2HGold〔pGBKT7-SCR2〕×Y187〔pGADT7-eSRK28〕能够在二缺平板(SD/-Trp-Leu)上生长,但在三缺平板上不生长,表明不同单倍型SRK-SCR可能不发生相互作用;组合Y187〔pGADT7-SRKJ〕×Y2HGold〔pGBKT7-THL1〕能够在四缺平板上生长,激活了4种报告基因(AUR1-C、MEL1、HIS3、ADE2)。相同的单倍型SRK与SCR之间能够相互作用,不同的单倍型之间不会发生相互作用;酵母中报告基因表达的数目和类型的差异反映了class-Ⅱ型内部的SCR-SRK和class-Ⅰ的SCR-SRK互作强度,Ⅰ型高于Ⅱ型;THL与SRK之间存在较高的互作强度。这为Ⅰ型和Ⅱ型不同的自交不亲和性表型所依赖的3个起始信号传导元件之间的识别程度差异提供了直接证据和新内容。  相似文献

 为了深入研究S位点受体激酶(SRK)和S位点富含半胱氨酸蛋白(SCR)甘蓝自交不亲和(self-incompatibility,SI)信号传导的雌雄决定因子的相互作用机制,以自交不亲和性高的结球甘蓝为材料,采用酵母双杂交体系,构建pGADT7-eSRKs和pGBKT7-SCRs重组载体,并分别转化到Y187和Y2HGold酵母中,通过SD/-Ade-His-Trp-Leu平板上菌落的形成,PCR以及x-α-gal显色反应鉴定转化到酵母中的两个重组质粒相互作用。结果表明,SRK S域的SRK1及SRK4分别与SCR2相互作用,且初步显示其相互作用区域为SRK第1个外显子的第16 ~ 421 bp的片段和SCR第1 876 ~ 2 068 bp的片段。这既为SRK-SCR相互作用提供了具体证据,也为甘蓝自交不亲和性分子机理的深入研究提供了新内容。  相似文献

Electronic dimming of high-intensity discharge lamps offers control of photosynthetic photon flux (PPF) but is often characterized as causing significant spectral changes. Growth chambers with 400-W metal halide (MH) and high-pressure sodium (HPS) lamps were equipped with a dimmer system using silicon-controlled rectifiers (SCR) as high-speed switches. Phase control operation turned the line power off for some period of the alternating current cycle. At full power, the electrical input to HPS and MH lamps was 480 W (root mean squared) and could be decreased to 267 W and 428 W, respectively, before the arc was extinguished. Concomitant with this decrease in input power, PPF decreased by 60% in HPS and 50% in MH. The HPS lamp has characteristic spectral peaks at 589 and 595 nm. As power to the HPS lamps was decreased, the 589-nm peak remained constant while the 595-nm peak decreased, equaling the 589-nm peak at 345-W input, and the 589-nm peak was almost absent at 270-W input. The MH lamp has a broader spectral output but also has a peak at 589 nm and another smaller peak at 545 nm. As input power to the MH lamps decreased, the peak at 589 diminished to equal the 545-nm peak. As input power approached 428 W, the 589-nm peak shifted to 570 nm. While the spectrum changed as input power was decreased in the MH and HPS lamps, the phytochrome equilibrium ratio (Pfr : Ptot) remains unchanged for both lamp types.  相似文献

 通过自交不亲和甘蓝‘A1’自花授粉1 h 和未授粉柱头蛋白质表达谱的对比,鉴定出1 个受 自花授粉诱导上调表达的蛋白,通过PCR 技术获取了其编码序列,命名为BosiPA1 蛋白,其基因全长为 3 730 bp,具有1 个1 092 bp 的完整编码框(KF314579)。BosiPA1 由6 个外显子、5 个内含子组成,编码 363 个氨基酸。序列分析表明BosiPA1 蛋白具有典型的basic helix-loop-helix(bHLH)功能域,在SCR、SRK、 Exo70A1 基因的ATG 上游启动子序列中存在可以被bHLH 功能区识别并结合的E-box(CANNTG)序列。 在分子进化上甘蓝BosiPA1与AtbHLH128 距离最近,与AtHEC1/2/3 和TgGBOF-1 处在同一个分支。RT-PCR 检测表明甘蓝BosiPA1 基因在花瓣、萼片、花粉、柱头和叶片中均有表达;荧光定量PCR 分析表明BosiPA1 基因在自花授粉10 min、30 min、1 h 的柱头中呈逐渐上升趋势。上述结果说明BosiPA1 与bHLH 的进化 分支较早,可能是一个参与甘蓝多器官发育的自交不亲和相关的转录因子。  相似文献

AIM: To investigate the protective effect of recombinant SCR15-18 domain of human complement receptor type 1 (CR1-SCR-15-18) on intestinal ischemia and reperfusion in a rat model. METHODS: Sprague-Dawley rats were randomly divided into 3 groups: sham operation(SO) group, ischemia and reperfusion (I/R) group and CR1-SCR15-18 treatment group. The superior mesenteric artery of the rats was clamped for 30 min followed by 60 min of reperfusion. PBS alone or CR1-SCR15-18 protein (30 mg/kg) in PBS was intravenously administered 5 min before reperfusion. Intestinal vascular permeability, myeloperoxidase (MPO), malondialdehyde (MDA) and superoxide dismutase (SOD) were measured. The histopathological changes of intestinal mucosa were examined by HE staining and complement 3 was detected by immunohistochemical analysis. RESULTS: Compared with SO group, the vascular permeability, the activity of MPO and the content of MDA in I/R group were significantly increased, and the activity of SOD was decreased. HE staining demonstrated that I/R induced severe intestinal histological damages and the increased amount of complement 3 and its derivates were deposited in the necrosis area. Compared with I/R group, the vascular permeability, the activity of MPO and the content of MDA were decreased and the activity of SOD was significantly increased in CR1-SCR15-18 treatment group. CR1-SCR15-18 also significantly attenuated intestinal histological injury, and reduced the deposition of complement 3 and its derivates in the necrosis zone. CONCLUSION: sCR1-SCR15-18 protein exerts a protective effect against intestinal I/R injury in rats, possibly by inhibiting the activation of complement.  相似文献

ObjectivesMany studies have found that natural environments benefit human health and wellbeing, but few have measured restorativeness of specific landscape components, especially in Chinese settings. Because the rapid urbanization of China is accompanied by increasing predomination of hardscape components in cities, the restorative quality of urban green space is a crucial issue. This study explored the stress recovery effects of different videotaped scenes, using six urban parks and one urban roadway scene. Potentially restorative urban park scenes were controlled for nature-based vs. hardscape components, presence/absence of people, and level of openness.MethodsSubjects were Chinese university students (N = 140) aged 18–24. After completing an oral exam as a stressor, an equal number of males and females were randomly assigned to watch one of the seven videotaped scenes during a stress recovery stage, while data were collected on changes in stress and attentional levels. Physiological responses were measured by Electrocardiography (ECG) and Skin Conductance Response (SCR). Psychological responses were measured by the state (short-term) version of the State-Trait Anxiety Inventory (STAI-S), the Digit Span Backwards (DSB) test, and the Perceived Restorativeness Scale (PRS).ResultsIn a Chinese sample, this study confirmed previous findings that nature-based urban park scenes relieved stress and restored attentional levels, while viewing an urban roadway increased negative feelings. Overall perceived restorativeness was significantly higher in two scenes depicting a Lawn without people and a Small Lake, compared with a paved Plaza with or without people, confirming previous findings that restorativeness is associated with predominance of nature-based landscape components. This study also confirmed previous findings that outdoor scenes without people were more restorative than scenes depicting people.DiscussionThis study found different levels of restorativeness associated with different landscape features, and helped confirm that nature-based components are more likely to reduce stress than hardscape components, using Chinese urban scenes with a Chinese population. Findings can be used in future planning and design of urban spaces in China, emphasizing the value of parks and green spaces in relevant contexts.  相似文献

用CTAB法提取三七景天基因组DNA,根据已报道的辣椒WRKY的转录因子保守区域设计引物,对影响PCR扩增反应的主要因素进行初步研究,建立景天基于转录因子的分子标记技术体系。在20μl的反应体系中,dNTPs0.4mmol/L,DNA模板30ng。扩增程序为:94℃预变4min,反应前7个循环在94℃30s、60℃30s、72℃1min条件下进行,随后5个循环在94℃30s、60℃30s、72℃45s条件下进行,随后13个循环在94℃30s、48℃30s、72℃1min、94℃30s、40℃30s、72℃1min条件下进行,最后72℃延伸10min。  相似文献

应用PCR扩增技术, 根据目的S基因与GenBank中已知S 基因同源性100%原理鉴定了我国育成的12个梨品种的S基因型, 分别是: ‘红酥脆’为S4S36 , ‘新梨7号’为S28Sd , ‘金水3号’为S5S29 , ‘八月酥’为S3S16 , ‘金水1号’为S3S29 , ‘龙泉酥’为S3S22 , ‘雪花’为S4S16 , ‘雪芳’为S4S16 ,‘雅青’为S4S34 , ‘新雅’为S4S34 , ‘德胜香’为S3S29 , ‘富源黄’为S16S33。通过对S基因的DNA序列分析, 确认S16、S31为同一S基因。本研究结果丰富了我国梨品种的S基因型信息, 为田间合理配置授粉品种提供了依据。  相似文献

2010年采用实地调查的方法调查了辽宁省苹果矮化栽培状况。结果表明:辽宁省苹果矮化栽培面积仅占全省苹果栽培总面积的2%左右,主要存在的问题为中间砧长短不一,栽植深浅不同;树形不标准,管理技术不规范;修剪不当;果园地下管理较差;苗木质量极差,价格高;贪产不合理负担等。据此,提出了规范育苗、标准化建园、整形修剪、土肥水管理等技术措施。  相似文献

对8个甜瓜品种的14个主要性状进行了配合力综合分析。结果表明,炮台红品种是选育晚熟、产量高、品质中等、大果型品种的优良亲本,皇后是选育品质好、产量高、中熟品种的优良亲本,93-13是选育早熟、丰产、外观美丽品种的优良亲本;该结果为甜瓜杂交育种的亲本选择提供了依据。  相似文献

A commercial seed lot of Portulaca grandiflora was sampled and the variation for several flower traits was analysed. A genotypic component of variance was estimated for the traits petal length, petal width, and petal number. The heritability for petal number was high (64 · 3), for petal width was intermediate (31 · 3), and for petal length was low (12–3). The environmental variance was partitioned into three components, but more than 90% of it was unaccountable. A significant positive correlation between petal length and petal width was found. A covariance analysis was used to test for genetic variance for petal length and petal width after the effects of covariance had been removed. No relationship was found between total corolla area and flower production. Thus, selection for larger flowers is not expected to reduce the number of flowers produced. Selection responses are predicted for different selection differentials and the assumptions are discussed. The results suggest that selection for increased petal number will be very effective.  相似文献

利用一套大白菜双单倍体(DH)群体和包含287个分子标记的遗传图谱,运用复合区间作图法对叶长、无柄叶长、有柄叶长、叶宽、叶柄长、叶柄宽6个叶部形态学性状进行QTL定位及遗传效应的分析。结果表明:在4个连锁群上共检测到12个QTL,分别为叶长相关的3个,无柄叶长相关的2个,有柄叶长相关的2个,叶宽相关的1个,叶柄长相关的2个,叶柄宽相关的2个。各位点的遗传贡献率介于7.5 %~18.9 %之间。其中,叶长与叶柄长的QTL存在共定位,叶宽与叶柄宽的QTL也存在共定位。  相似文献
