A comparison of rates of germination and appressorium formation by an isolate of Colletotrichum gloeosporioides on mango leaves, fruit surfaces and cellophane membranes showed that behaviour was broadly similar on all three substrates. Frequency of appressorium formation was slightly higher on cellophane membranes, and both hyaline and melanized appressoria were formed. Only melanized appressoria were formed on mango surfaces. In vitro experiments on membranes showed comparative differences in physiological behaviour with temperature for two Philippine isolates of C. gloeosporioides . The most stimulatory temperature for production of appressoria differed in isolates I-2 and I-4 (25 and 20°C, respectively). At 30°C more appressoria became melanized than at lower temperatures, but the frequency of formation of penetration pegs was highest at 25°C. Conidia of C. gloeosporioides germinated on cellophane membranes at relative humidities as low as 95%, but the percentage of conidia germinating and forming appressoria increased as the RH approached 100%. Approximately 18% of conidia of C. gloeosporioides I-2 held at 62 and 86% RH for 4 weeks retained viability, and some were capable of forming appressoria when placed at 100% RH. Carpropamid (WIN™, KTU 3616) provides good control of leaf and panicle blast by ‘one-shot’ nursery-box treatment. It inhibits melanin biosynthesis in appressorial cells of Pyricularia oryzae, making them hyaline. Penetration by infection hyphae from the hyaline appressoria into rice epidermal cells is substantially hindered. In addition, the spread of rice blast spores from primary lesions to the other parts of the plant leading to secondary infection is largely prevented when the plants are treated with carpropamid by spray or water surface application. Secondary infection was simulated in a glass chamber fitted with an ultrasonic humidifier. On treated plants, many blast spores formed in the lesions, but the number of air spora that were dispersed from the lesions decreased significantly. A similar suppression of the spore liberation was observed in vitro when lesions on rice leaf segments, or discs from Pyricularia cultures on oatmeal agar were treated with the chemical. Spores from treated lesions or from the cultures on oatmeal agar amended with the chemical germinated normally and produced well-melanized appressoria on cellophane membranes. In addition, the spores proved to be fully pathogenic towards rice seedlings, producing normal disease symptoms. These results strongly suggest that carpropamid reduces the secondary infection of rice by Pyricularia by specifically hindering spore liberation. © 1999 Society of Chemical Industry  相似文献

Ulvan is an algal polysaccharide known for its ability to induce resistance to plant diseases such as the Glomerella leaf spot of apple caused by Colletotrichum gloeosporiodes. This study was aimed at investigating microscopically, in tests in vitro and in vivo, whether ulvan interferes in the development of pre-infective structures of C. gloeosporioides. Conidial germination and appressoria formation were monitored hourly on agar and cellophane, and at 48 h on water- and ulvan-treated susceptible as well as resistant apple leaves. Amendment of agar with ulvan (10 mg ml?1) enhanced the germination and resulted in longer germ tubes at 7 h of incubation. On cellophane it significantly delayed appressoria formation up to 8 h, but later after 14 h increased the number of appressoria per conidium. Spraying of susceptible leaves with ulvan 6 days before inoculation decreased disease severity by 50%. This was associated with inhibition of appressoria formation and stimulus in growth of germ tubes, without interfering with conidial germination, when compared with both water-treated control and resistant plants. Appressorium formation occurred preferentially on anticlinal walls of epidermal cells and its location was not influenced by host resistance or by ulvan treatment. This study suggests a new mode of action for ulvan interfering with appressorium formation that could protect apple plants against C. gloeosporioides infection.  相似文献

 Reactive oxygen species (ROS) generation was examined in the interaction of Alternaria alternata Japanese pear pathotype and host plants using three methods: nitro blue tetrazolium (NBT) method for microscopic detection of O2 , diaminobenzidine (DAB) methods for microscopic detection of H2O2, and cerium chloride methods for ultrastructural detection of H2O2. ROS generation was detected by NBT and DAB methods at appressoria on leaves of susceptible cultivars and heat-shocked leaves of resistant cultivars but not in leaves of resistant cultivars. Ultrastructural detection by the cerium chloride method identified ROS generation at cell walls of appressoria and penetration pegs in susceptible, resistant leaves and heat-shocked leaves. These differences in the ultrastructural and microscopic data in resistant areas were due to the restriction of ROS generation in limited areas, the side facing the plant surface, of appressoria and penetration pegs. Therefore, ROS generation was apparently induced regardless of the resistance or susceptibility of the cultivar with the difference being in the volumes generated. After evaluating the pathological role of ROS generation in fungal structures, such generation was found to be associated with early penetration of cell walls in pear plants. Additionally, ROS generation in plants was also found in degrading pectin layers near infected hyphae and in plasma membrane modification sites in susceptible leaves but not in resistant leaves. ROS generation in susceptible leaves might be accompanied with plasma membrane damage, although the role of ROS generation in the pectin layers is not clear. 为明确苹果炭疽叶枯病病原菌围小丛壳Glomerella cingulata的侵染致病特征,在分离获得该病原菌的基础上,采用形态学观察、ITS序列分析和致病性测定对其进行了鉴定,并利用光学和扫描电子显微镜对病原菌在嘎啦苹果叶片上的侵染过程进行了研究.结果表明,在陕西咸阳地区分离获得的9株病原菌均为围小丛壳G.cingulata.25 ℃下接种9 h后,分生孢子中间产生隔膜,双胞化,并萌发产生芽管和附着胞;24 h后分生孢子的2个细胞均可萌发并形成芽管及附着胞,部分芽管顶端可产生次级分生孢子;48 h后次级分生孢子萌发形成附着胞;72 h后,附着胞下形成的侵染钉可直接入侵寄主,在表皮细胞内形成初生菌丝和次生菌丝,此时叶片表面已出现褐色斑点.接种7 d后叶片病斑处出现分生孢子盘和子囊壳.表明陕西省近年出现的苹果炭疽叶枯病病原菌为围小丛壳G.cingulata,该病菌在嘎啦叶片上的一些特殊侵染行为可能是导致该病害易在短时间内暴发的重要原因.  相似文献

Spores and infection structures such as germ tubes and appressoria of Magnaporthe oryzae, the fungus causing blast disease of wheat, produced an extracellular matrix (ECM) on the surfaces of host leaves during fungal differentiation. The chemical components and function of the ECM were studied to understand the pathological roles using two immunological techniques and ECM-digesting enzymes. The ECM was characterized by fibrous and amorphous materials, located in the spaces between fungal cell walls and plant cuticles. Immunohistochemical and immunoelectron microscopy suggested that ECM includes components positively reacted with antibodies of four animal cell adhesion factors (collagen VI, vitronectin, fibronectin and laminin) and an animal integrin α3. ECM, incubated on a cellulose membrane, was rapidly digested by matrix metalloproteinases (collagenase and gelatinase B), resulting in the detachment of most infection structures from membrane surfaces. Both ultrastructural observation and immunological responses showed that more ECM was located at the appressoria than at the spores and germ tubes. This result suggested that appressoria needed a powerful adhesion force for aggressive action of penetration pegs into plant cuticles. An erratum to this article can be found at  相似文献

This study investigates the infection process of Phoma koolunga on field pea (Pisum sativum) stems and leaves using different susceptible and resistant pea genotypes for each tissue, viz. 05P778‐BSR‐701 (resistant) and 06P830‐(F5)‐BSR‐5 (susceptible) for stems and ATC 866 (resistant) and ATC 5347 (susceptible) for leaves. On both resistant and susceptible genotypes, light and scanning electron microscopy showed P. koolunga conidia infect stem and leaf tissues directly via appressoria or stomatal penetration, but with more infections involving formation of appressoria on stems than on leaves. On leaves of the resistant genotype, at 72 h post‐inoculation, P. koolunga penetrated more frequently via stomata (5.2 conidia per 36 893 μm2) than by formation of appressoria (1.8 conidia); yet no such difference was observed on stems of the resistant genotype. In contrast, at the same time point, the number of conidia infecting the susceptible genotype by formation of appressoria on either stems (135 conidia) or leaves (11.3 conidia) was significantly greater than via stomata (8 and 7.3 conidia, stems and leaves, respectively). Mean germ tube length of germinating P. koolunga conidia on both stems (29.8 μm) and leaves (32.9 μm) of the resistant genotype was less than on the susceptible genotype (40.5 and 63.7 μm, stem and leaves, respectively). In addition, there were differences in the number of germ tubes emerging from conidia on resistant and susceptible genotypes. These are the first insights into the nature of leaf and stem resistance mechanisms operating in field pea against P. koolunga.  相似文献

The rice blast fungus Magnaporthe grisea differentiates appressoria, which are required to attack its rice plant host. Clone A26, tentatively named LPL1, was previously found to be homologous to the known lysophospholipase genes from our subtractive cDNA library. The LPL1 protein had a consensus motif (GxSxG) and a catalytic triad (S, D, H) of esterases in the deduced amino acid sequence, and the protein expressed in Escherichia coli had lysophospholipase activity. To clarify the functions and possible roles of LPL1, the gene was disrupted by targeted gene replacement. The ΔLPL1 mutants formed fewer appressoria on the hydrophobic surface of GelBond film, and the appressoria had reduced turgor pressure and penetration into cells of the leaf sheath. The ΔLPL1 mutants and wild-type differentiated normal appressoria on other artificial substrata such as polycarbonate plate and on rice leaf sheath. Cytological analysis of the appressoria indicated that ΔLPL1 mutants had a delay in the disappearance of lipid droplets. These findings imply that LPL1, phospholipid metabolism, or both are involved in glycerol biosynthesis and accumulation to generate turgor pressure in the appressorium. LPL1 was, however, dispensable for full pathogenicity, suggesting that other complementary pathways or similar genes related to phospholipid metabolism probably function in M. grisea.  相似文献

Diplocarpon mali, the causal agent of Marssonina leaf blotch of apple, causes severe defoliation during the growing season. Little information is available on the mode of infection and the infection process. In this study, the infection strategies of D. mali in apple leaves were investigated using fluorescence and electron microscopy. Conidia attached to leaf surface apparently by mucilage and germinated on both sides of leaves 6 h post-inoculation (hpi). The pathogen penetrated the cuticle by infection pegs formed either in germ tubes or appressoria in 6 hpi, and then formed haustoria in host epidermal and mesophyll cells accompanied by extension of subcuticular and intercellular hyphae. Five days post-inoculation (dpi), the intracellular hyphae were observed. At the same time, the subcuticular hyphal strands (SHS) were produced as a means for fast expansion and reproduction. About 7 dpi, acervuli formed on inoculated leaves. This was the first observation that D. mali formed haustoria and SHS as infection strategies. Our results suggest that D. mali may behave like a hemibiotroph, which can use both biotrophic and necrotrophic strategies to establish infections on apple leaves.  相似文献

In growth cabinet experiments, the common phyllosphere yeastsSporobolomyces roseus andCryptococcus laurentii var.flavescens were sprayed as a mixture (11) onto the fourth leaves of maize plants (Zea mays) two-three days prior to inoculation withColletotrichum graminicola. In four experiments the average yeast population of the treated leaves at the time of pathogen inoculation varied between 5× 104 and 8× 105 cells cm–2 leaf, whereas on the untreated leaves the yeast population varied from <103 to 104 cells cm–2 leaf. The yeasts reduced lesion density and necrosis fromC. graminicola infection by approximately 50%. Contrary to findings with other necrotrophic pathogens, conidial germination, superficial mycelial growth and appressorium formation were not affected. Colletotrichum lagenarium is the casual agent of anthracnose disease of melons. Light and scanning electron microscopy were used to observe the infection process of C. lagenarium on the leaves of two melon cultivars differing in susceptibility. On both cultivars conidia began germinating 12 h after inoculation (hai), forming appressoria directly or at the tips of germ-tubes. By 48 hai appressoria had melanised and direct penetration of host tissue had begun. On the susceptible cultivar, infection vesicles formed within 72 hai and developed thick, knotted primary hyphae within epidermal cells. By 96 hai C. lagenarium produced highly branched secondary hyphae that invaded underlying mesophyll cells. After 96 hai, light brown lesions appeared on the leaves, coincident with cell necrosis and invasion by secondary hyphae. While appressoria formed more quickly on the resistant cultivar, fewer germinated to develop biotrophic primary or invasive necrotrophic secondary hyphae than on the susceptible cultivar. These results confirm that C. lagenarium is a hemibiotrophic pathogen, and that resistance in melons restricts colonisation by inhibiting the development of necrotrophic secondary hyphae.  相似文献

The leaf alcohols cis -3-hexen-1-ol and trans -2-hexen-1-ol have been previously shown to induce the wheat stem rust (Puccinia graminis f. sp. tritici) to differentiate appressoria in a complex axenic culture medium. In this paper, the possible role of these chemicals in appressorium induction during infection has been analysed further. The inductive potential of ethylene and three cutin monomers were also tested because they have been found to induce appressoria in other fungi. Both hexenols were found to be inductive in aqueous solution in the absence of media constituents. 0.5–1 m trans -2-hexen-1-ol was the most inductive resulting in a maximum of 51% appressorium differentiation. trans -2-hexen-1-ol was also shown to be inductive in vapour form. In aqueous solution, trans -2-hexen-1-ol acted synergistically with topographical signals by increasing the rate of appressorium induction. Combining the two signals also increased the total number of appressoria differentiated (88%). The other chemicals tested were non-inductive although a biochemical domain rich in exposed cutin was localized around stomatal apertures of wheat leaves. The characteristic bipolar morphology of appressoria formed over stomata was found to be determined by topographical signals. Overall, our data support a role for both chemical signals (hexenols or their analogues) and topographical signals being involved in appressorium induction by wheat stem rust.  相似文献

Anthracnose fruit rot of blueberries caused by Colletotrichum acutatum is a serious problem in humid blueberry‐growing regions of North America. In order to develop a disease prediction model, environmental factors that affect mycelial growth, conidial germination, appressorium formation and fruit infection by C. acutatum were investigated. Variables included temperature, wetness duration, wetness interruption and relative humidity. The optimal temperature for mycelial growth was 26°C, and little or no growth was observed at 5 and 35°C. The development of melanized appressoria was studied on Parafilm‐covered glass slides and infection was evaluated in immature and mature blueberry fruits. In all three assays, the optimal temperature for infection was identified as 25°C, and infections increased up to a wetness duration of 48 h. Three‐dimensional Gaussian equations were used to assess the effect of temperature and wetness duration on the development of melanized appressoria (R2 = 0·89) on Parafilm‐covered glass slides and on infection incidence in immature (R2 = 0·86) and mature (R2 = 0·90) blueberry fruits. Interrupted wetness periods of different durations were investigated and models were fitted to the response of melanized appressoria (R2 = 0·95) and infection incidence in immature (R2 = 0·90) and mature (R2 = 0·78) blueberry fruits. Additionally, the development of melanized appressoria and fruit infection incidence were modelled in relation to relative humidity (R2 = 0·99 and 0·97, respectively). Three comprehensive equations were then developed that incorporate the aforementioned variables. The results lay the groundwork for a disease prediction model for anthracnose fruit rot in blueberries.  相似文献

One of the components of partial resistance of barley to leaf rust,Puccinia hordei, is a reduced infectibility. It was investigated whether this low infectibility may rest on a hampered appressorium formation of the leaf rust fungus. The appressorium formation on the primary leaves of 11 barley genotypes with an intermediate-to-low infectibility was compared with that on the highly infectible L94. The number of stomata per cm2 leaf area occupied by appressoria ofP. hordei was determined per genotype by means of fluorescence microscopy. No cosistent differences could be detected, indicating that the mechanisms causing a low infectibility of partially resistant barley seedlings act at a phase later than the formation of the appressoria. On the non-host wheat not fewer appressoria were formed than on L94, but no appressoria were found on a lettuce genotype. The latter probably lacks the stimuli that enable the fungus to find stomata.Samenvatting Eén van de componenten van partiële resistentie van gerst tegen dwergroest,Puccinia hordei, is een verminderde infectiedichtheid. Het mechanisme, dat hieraan ten grondslag ligt, is onbekend. Een experiment werd uitgevoerd om na te gaan of bij partieel resistente rassen een verminderde appressoriumvorming optreedt. Na inoculatie in een inoculatietoren en een zorgvuldig uitgevoerde incubatie werd het aantal huidmondjes per cm2 bladoppervlak bepaald dat bezet was door appressoria vanP. hordei. De elf weinig vatbare gerstlijnen uit deze studie bleken niet reproduceerbaar te verschillen van de zeer vatbare gerstlijn L94 in de mate van appressoriumbezetting. Dit wijst erop dat infectiedichtheidsverschillen t.g.v. partiële resistentie veroorzaakt worden door mechanismen die werken na de appressoriumvorming. In een tweede experiment werd aangetoond dat zelfs op de niet-waardsoort tarwe, waaropP. hordei geen symptomen veroorzaakt, niet minder appressoria worden gevormd dan op L94. Op een sla-genotype trad echter geen appressoriumvorming op. Op deze laatste niet-waardsoort ontbreken waarschijnlijk de stimuli die de schimmel in staat stellen huidmondjes te vinden.  相似文献

为研究稻瘟病菌Magnaporthe oryzae不同菌株间的相互作用,选择与单抗性基因系水稻IRBL5-M (携带抗性基因Pi5)表现为亲和性的菌株HN52与非亲和性的菌株HN119为研究对象,将其单独或混合接种到单抗性基因系水稻IRBL5-M中,并通过荧光显微镜观察接种后水稻叶鞘的发病情况及病斑面积,测定接种后水稻内相关抗性基因OsWRKY45、OsNPR1、OsPR10、OsMAPK2的表达量以及活性氧的变化。结果显示,相较于单独接种亲和性菌株,混合接种后单抗性基因系水稻IRBL5-M病斑发病面积减少;混合接种中亲和性菌株HN52菌丝侵染能力降低,侵染菌丝细胞间扩展率显著降低73.13%;同时单抗性基因系水稻IRBL5-M中OsWRKY45、OsNPR1、OsPR10OsMAPK2抗性基因表达量显著增加,水稻叶片中活性氧含量增加,表明在菌株混合侵染过程中,非亲和性菌株可通过激发水稻的抗性反应来降低亲和性菌株对水稻的侵染程度。  相似文献

When the influence of host species, inoculum density, temperature, leaf wetness duration, and leaf position on the incidence of gentian brown leaf spot caused by Mycochaetophora gentianae, was examined, the fungus severely infected all seven Gentiana triflora cultivars, but failed to infect two cultivars of G. scabra and an interspecific hybrid cultivar. Inoculum density correlated closely with disease incidence, and a minimum of 102 conidia/mL was enough to cause infection. In an analysis of variance, temperature and leaf wetness duration had a significant effect upon disease incidence, which increased with higher temperature (15–25°C) and longer duration of leaf wetness (36–72 h). No disease developed at temperatures lower than 10°C or when leaf wetness lasted <24 h. At 48-h leaf wetness, disease incidence was 0, 28, 77, and 85% at 10, 15, 20, and 25°C, respectively. Middle and lower leaves on the plant were more susceptible than upper leaves. In microscopic observations of inoculated leaves, >50% of conidia germinated at temperatures >15°C after 24-h leaf wetness. More appressoria formed at higher temperatures (15–25°C) with extended duration of leaf wetness (24–72 h). At 48-h leaf wetness, appressorium formation was 0, 8, 26, and 73% at 10, 15, 20, and 25°C, respectively. These results suggest that temperature and leaf wetness duration were important factors for infection of gentian leaves.  相似文献

The mode of action of soluble silicon against strawberry powdery mildew (Sphaerotheca aphanis var. aphanis) was investigated in four experiments. First, silicon-treated leaves from plants grown with silicate (Si+) and control leaves were excised, inoculated with conidia, and subsequent germination and formation of appressoria in a petri dish was assessed after 24 h. The germination rate was 49.7% on Si+ leaves, and was 67.2% on control leaves (t-test, P < 0.01). Second, we soaked cellulose membranes in various solvents and then placed the membranes on 4% water agar, dusted the membranes with conidia, and examined after 12 h. No difference was apparent between any treatment and the control (distilled water). Third, strawberries growing hydroponically with additional silicon in the medium were inoculated with conidia, and leaves were observed with a scanning electron microscope 1–2 days after inoculation. Germ tubes and secondary hyphae were shorter and had fewer branches on Si+ leaves than on the control. Moreover, penetration appeared to be inhibited. Fourth, the cuticle was separated from leaves from plants grown as in the third experiment, placed on water agar, and dusted with conidia. Germination of conidia, observed with a light microscope, on Si+ leaves was suppressed markedly to 40%–60% of that of the control. These results suggested that soluble silicon induced physiological changes in the cuticle layer after absorption by the plant. In addition, soluble silicate reduced germination of conidia, formation of appressoria, and possibly the penetration of powdery mildew.  相似文献

Glomerella leaf spot (GLS), characterized by black necrotic spots and severe defoliation, is a destructive foliar disease of apple. Widely grown cultivars such as Gala and Golden Delicious are highly susceptible to GLS. Currently, the infection biology of the causal pathogen, Colletotrichum fructicola, on apple leaves is unclear. In the present study, the penetration and colonization processes of C. fructicola were characterized on apple (cv. Gala) leaves using light and transmission electron microscopy. C. fructicola conidia produced germ tubes 4 hours post-inoculation (hpi) and appressoria at 8 hpi. In melanized appressoria, funnel-shaped appressorial cones formed around the penetration pore. At 12 hpi, C. fructicola produced secondary conidia. After penetration, C. fructicola began to develop infection vesicles at 36 hpi. At 48 hpi, the primary hyphae of C. fructicola were produced from infection vesicles within host epidermal cells; the host epidermal cell plasma membrane remained intact, indicating a biotrophic association. Subsequently, secondary hyphae penetrated epidermal cells and destroyed cell components, initiating necrotrophic colonization. C. fructicola also produced biotrophic subcuticular infection vesicles and hyphae. Together, these results demonstrate that C. fructicola forms special infection structures and colonizes apple leaves in a hemibiotrophic manner, involving intracellular as well as subcuticular colonization strategies. Detailed characterization of the infection process of C. fructicola on apple leaves will assist in the development of disease management strategies and provide a foundation for studies of the molecular mechanism of the C. fructicola–apple leaf interaction.  相似文献

Cochliobolus miyabeanus forms a specialized infection structure, an appressorium, to infect rice. Contacting a hard surface induces appressorium formation in C. miyabeanus, while the hydrophobicity of the substratum does not affect this morphogenic infection event. To determine whether the calcium/calmodulin-dependent signaling system is involved in prepenetration morphogenesis in C. miyabeanus, the effects of a calcium chelator (ethylene glycol tetraacetic acid; EGTA), phospholipase C inhibitor (neomycin), intracellular calcium channel blocker (TMB-8), calmodulin antagonists (chlorpromazine, phenoxybenzamine, and W-7), and calcineurin inhibitor (cyclosporin A) on morphogenesis and infection were examined. Addition of Ca2+ and the calcium ionophore A23187 did not affect conidial germination, while the number of appressoria decreased with higher concentrations. EGTA inhibited conidial germination and appressorium formation. The calcium channel blocker did not affect appressorium formation at any concentration; however, calmodulin antagonists and the calcineurin inhibitor specifically reduced appressorium formation at the micromolar level. One of the calmodulin antagonists, W-7, also inhibited accumulation of mRNA of the calmodulin gene within germinating conidia and/or appressorium-forming germ tubes. Thus, biochemical processes controlled by the calcium/calmodulin signaling system seem to be involved in the induction of prepenetration morphogenesis on rice.  相似文献
