Meat samples from diaphragm pillars were randomly taken from 3,048 pigs of 52 Bavarian herds after slaughtery. Meat-juice was collected and tested for salmonella antibodies in an indirect ELISA. The number of samples was calculated according to the annual production of slaughter pigs of a farm outlined in the "Leitlinien für ein Programm zur Reduzierung des Eintrags von Salmonellen durch Schlachtschweine in die Fleischgewinnung" from February 05th, 1998 (< 100 slaughter pigs: 45 samples, 100-200 slaughter pigs: 50 samples, > 200 slaughter pigs: 60 samples per year). Salmonella antibodies were detected in 48 carcasses (1.6%) of 12 farms (23.1%). However, 33 (68.8%) of these carcasses were originated from a single farm which had to be classified into category III (prevalence of > 40% in the samples). No bacteria could be isolated from this farm in a follow up examination. The 51 other farms (98%) were classified into category I (prevalence of < 20% in the samples). Farms with in/out-management showed a higher degree of reagents (2.1T%) than farms with continuous stabling (0.8T%). In a pig experimentally immunized with LPS-antigen preparations of Salmonella typhimurium it was shown that antibodies induced were nearly at the same level in all meat samples and even in selected organs (liver, kidney, parotis, mesenteric lymph nodes).

Methicillin resistant Staphylococcus aureus (MRSA) colonization has recently been identified in pigs and people that work with pigs, raising concerns about the role of pigs as reservoirs of MRSA for human infection. The objectives of this study were to evaluate the prevalence of MRSA colonization in pigs and pig farmers in Ontario, Canada and to characterize MRSA strains. Nasal and rectal swabs were collected from 285 pigs from three different age groups from 20 pig farms. Nasal swabs were collected from farm personnel and a brief questionnaire was also administered. The prevalence of MRSA colonization in farms was 45% (9/20) whereas the prevalence in pigs was 24.9% (71/285). There was no difference in MRSA colonization between age groups. The prevalence of MRSA colonization in pig farmers was 20% (5/25). There was a correlation between the presence of MRSA in pigs and humans on farms (P value=0.001). The results of spa typing revealed the predominant strain in pigs and humans was eGenomics spa type 539 (Ridom t034, clonal complex 398) which accounted for 59.2% of isolates and has been reported in pigs in Europe. A common human epidemic clone, CMRSA-2 (USA100, clonal complex 5) was also found in both pigs and pig personnel. Indistinguishable strains were found in pigs and pig personnel on all five farms with a colonized human. This study demonstrates that MRSA is common in pigs in Ontario, Canada, and provides further support to concerns about transmission of MRSA between pigs and humans.

Clostridium difficile is the leading cause of nosocomial diarrhea in humans and a major cause of enteritis in neonatal piglets, foals and calves. The aim of this longitudinal study was to determine and compare the prevalence, antimicrobial susceptibility, and toxinotype profiles of C. difficile isolated from pigs and their environment in the indoor conventional and outdoor antimicrobial free (ABF) production systems. Ten conventional and eight ABF cohorts of 35 pigs each and their environment were sampled at different stages of production at farm and slaughter. C. difficile prevalence in pigs was highest at the farrowing stage in both conventional (34%, 120/350) and ABF (23%, 56/244) systems, and decreased with age. This reduction in C. difficile prevalence in pigs at later stages of production mirrored the decreased prevalence in the farm environment. At slaughter, C. difficile was isolated at a low frequency from the carcasses and processing environment in both production systems. All but three isolates were resistant to ciprofloxacin (99%, 505/508), while 1.0% (5/508) and 6.0% (23/508) of isolates exhibited resistance to tetracycline and erythromycin, respectively. Toxinotype V (tcdA(+)tcdB(+)) was the predominant strain identified in both systems (conventional: 94%, 376/401; ABF: 82%, 88/107), while the rest were toxinotype XIII (tcdA(+)tcdB(+)). To conclude, we isolated antimicrobial resistant C. difficile regardless of antimicrobial use on the farm. Based on the phenotypic and genotypic similarity of C. difficile isolated in this study, we conclude that the unique production practices employed in conventional and ABF production systems have no impact on the pathogen population.

The aim of this study was to determine the phenotypic and genotypic diversity of multiple Campylobacter isolates (n = 3 per sample) present within individual (heterogeneity) pig faecal and carcass samples at farm and slaughter, respectively. We isolated 1459 Campylobacter coli (1110 on farm and 349 from slaughter) from 908 pigs and 757 carcasses and characterized them for their antimicrobial susceptibility profile to a panel of six antimicrobials using the agar dilution method. Overall, we detected a significantly higher Campylobacter prevalence at the farm (54.7%) than at slaughter (19%) level (P < 0.05). C. coli isolates were resistant most commonly to tetracycline (66.2%) and erythromycin (53.6%) while fluoroquinolone resistance was detected in isolates (n = 17) only from the farm level. Phenotypic diversity of C. coli isolates at the 4-fold minimum inhibitory concentration levels within the same sample was detected in 38.6% (n = 192) pigs and 40.2% (n = 58) carcass swabs with no significant difference between the two sources (P = 0.72). Phenotypic heterogeneity based on the antimicrobial resistance patterns was observed in 32.5% (n = 162) of the farm samples and in 30.5% (n = 44) carcass swabs at slaughter (P = 0.64). A subset of 40 isolates representing ten pigs and eight carcass samples (originating from separate pigs) were further genotyped by multi locus sequence typing. The observation of phenotypic diversity was replicated at the genotypic level, as it was highlighted by the 22 sequence types which represented the 40 isolates. In conclusion, we detected multiple C. coli subtypes from individual pig or carcass samples indicating unprecedented level of heterogeneity. Our study clearly signifies the importance of testing multiple colonies to make appropriate and valid conclusions in epidemiological-based studies.

We explored observed risk factors and drivers of infection possibly associated with African swine fever (ASF) epidemiology in Uganda. Representative sub-populations of pig farms and statistics were used in a case-control model. Indiscriminate disposal of pig viscera and waste materials after slaughter, including on open refuse dumps, farm-gate buyers collecting pigs and pig products from within a farm, and retention of survivor pigs were plausible risk factors. Wire mesh-protected windows in pig houses were found to be protective against ASF infection. Sighting engorged ticks on pigs, the presence of a lock for each pig pen and/or a gate at the farm entrance were significantly associated with infection/non-infection; possible explanations were offered. Strict adherence to planned within-farm and community-based biosecurity, and avoidance of identified risk factors is recommended to reduce infection. Training for small-scale and emerging farmers should involve multidimensional and multidisciplinary approaches to reduce human-related risky behaviours driving infection.

This study was conducted to determine the prevalence of livestock-associated methicillin-resistant Staphylococcus aureus (LA-MRSA) in pigs, farm workers, and the environment in northern Thailand, and to assess LA-MRSA isolate phenotypic characteristics. One hundred and four pig farms were randomly selected from the 21,152 in Chiang Mai and Lamphun provinces in 2012. Nasal and skin swab samples were collected from pigs and farm workers. Environmental swabs (pig stable floor, faucet, and feeder) were also collected. MRSA was identified by conventional bacterial culture technique, with results confirmed by multiplex PCR and multi locus sequence typing (MLST). Herd prevalence of MRSA was 9.61% (10 of 104 farms). Among pigs, workers, and farm environments, prevalence was 0.68% (two of 292 samples), 2.53% (seven of 276 samples), and 1.28% (four of 312 samples), respectively. Thirteen MRSA isolates (seven from workers, four from environmental samples, and two from pigs) were identified as Staphylococcal chromosomal cassette mec IV sequences type 9. Antimicrobial sensitivity tests found 100% of the MRSA isolates resistant to clindamycin, oxytetracycline, and tetracycline, while 100% were susceptible to cloxacillin and vancomycin. All possessed a multidrug-resistant phenotype. This is the first evidence of an LA-MRSA interrelationship among pigs, workers, and the farm environment in Thailand.

The distribution and persistence of pathogenic, virF/lcrF-positive Yersinia pseudotuberculosis were investigated in pigs and in the pig house environment during rearing to determine possible contamination routes of early infections. Based on Y. pseudotuberculosis-positive tonsils of slaughter pigs in our previous study, Y. pseudotuberculosis-positive animals were traced back to the farms. Eight farms were visited from 6-10 months later, and a total of 155 pooled and six individual faecal samples from pigs and 116 pooled environmental samples were collected for analysis by different culture methods. Four of the eight farms were found to be Y. pseudotuberculosis-positive. All positive faecal samples were obtained from fattening pigs, with prevalence varying from 5% to 71% on positive farms. Sows, boars and suckling piglets were Y. pseudotuberculosis-negative on all farms. Most Y. pseudotuberculosis-positive farms (three of four) were on a one-site production system, which had a higher prevalence of Y. pseudotuberculosis (5-26%) among fattening pigs than the all-in, all-out system (1-5%). All Y. pseudotuberculosis isolates belonged to serotype O:3 and carried the virF/lcrF gene on the virulence plasmid. Biotypes 2 and 3 were involved, the latter in one isolate and not being previously reported in pigs. Altogether 53 isolates from 16 positive samples were characterized with pulsed-field gel electrophoresis (PFGE). Using SpeI, NotI and XbaI enzymes, four, three and two different PFGE patterns were obtained respectively. A total of nine different genotypes were identified when the profiles of the enzymes were combined. The most common genotypes were gIV, found on three, and gXII, found on two of the four Y. pseudotuberculosis-positive farms. The same genotypes previously detected in pig tonsils were present in pig faeces from the same farm, indicating that some Y. pseudotuberculosis strains can persist in the pig house environment.

We set up a matched case-control study of potential risk factors for clinical encephalomyocarditis virus (EMCV) in 58 pig farms in West Flanders (Belgium). In total, 29 farms experienced a clinical outbreak of EMCV confirmed by EMC virus isolation. Mortality was seen only among suckling piglets (18 case farms), in piglets and other age-groups (4 case farms), or only among fattening pigs (7 case farms). Five farms had reproductive problems among the sows. Control farms were matched geographically on farm size and farm type and were selected on the absence of clinical signs. A questionnaire on potential risk factors for EMCV was developed to collect data at both case and control farms. The exploration of the data used clusters of factors associated with clinical EMCV infection: (a) rodents, (b) general farm set up and (c) general hygiene. The multivariable relationships between clinical appearance of EMCV and potential risk factors were tested with conditional logistic regression. The final model on all farms contained presence of mice (OR=8.3) as a risk factor for clinical EMCV infection while the flow of manure up through the slatted floor (OR=0.11) and movement of manure between manure pits in the pig stable (OR=0.14) were protective.

Streptococcus suis type 2 infections in pigs in the Netherlands (Part two)   总被引:2,自引:0,他引:2
Since 1983 some pig breeding and fattening farms in the Netherlands have been faced with a considerable mortality in pigs due to Streptococcus suis type 2 infections. The most predominant clinical feature of S. suis type 2 infection is meningitis, although sudden deaths often occur. It was noted that some affected farms had imported breeding stock from the United Kingdom. Tonsils of slaughter pigs were collected from herds with and without a history of S. suis type 2 infections. Bacteriological examination was done by using an elective-selective medium. No significant difference was found in carrier rates of S. suis type 2 between clinically healthy and affected herds (38% vs. 45%). A cohort study was carried out by regular bacteriological examination of tonsil biopsies on a farm with a high incidence of streptococcal meningitis. Twenty-seven percent of the pigs were carriers of S. suis type 2 at nine weeks of age. Possible methods for disease control are discussed.

The world-wide increase of antimicrobial resistance in micro-organisms complicates medical treatment of infected humans. We did a risk-factor analysis for the prevalence of antimicrobial resistant Campylobacter coli on 64 Swiss pig finishing farms. Between May and November 2001, 20 faecal samples per farm were collected from the floor of pens holding finishing pigs shortly before slaughter. Samples were pooled and cultured for Campylobacter species. Isolated Campylobacter strains were tested for resistance against selected antimicrobials. Additionally, information on herd health and management aspects was available from another study. Because data quality on the history of antimicrobial use on the farms was poor, only non-antimicrobial risk factors could be analysed. Statistical analyses were performed for resistance against ciprofloxacin, erythromycin, streptomycin, tetracycline, and for multiple resistance, which was defined as resistance to three or more antimicrobials. Risk factors for these outcomes – corrected for dependency of samples at herd level – were analysed in five generalised estimation-equation models. Prevalence of antimicrobial resistance among Campylobacter isolates was ciprofloxacin 26.1%, erythromycin 19.2%, streptomycin 78.0%, tetracycline 9.4%, and multiple resistance 6.5%. Important risk factors contributing to the prevalence of resistant strains were shortened tails, lameness, skin lesions, feed without whey, and ad libitum feeding. Multiple resistance was more likely in farms which only partially used an all-in-all-out system (OR = 37), or a continuous-flow system (OR = 3) compared to a strict all-in-all-out animal-flow. Presence of lameness (OR = 25), ill-thrift (OR = 15), and scratches at the shoulder (OR = 5) in the herd also increased the odds for multiple resistance. This study showed that on finishing farms which maintained a good herd health status and optimal farm management, the prevalence of antimicrobial resistance was also more favourable.

OBJECTIVE: To enumerate the prevalence of Campylobacter isolates in the intestinal tract of market-weight swine raised in an integrated swine operation in Texas. SAMPLE POPULATION: Samples of cecal contents were collected from 595 pigs (mean body weight, 110 kg [242 lb]) at time of slaughter. Pigs were off-spring of Yorkshire-Landrace sows and Duroc or Hampshire boars. Pigs originated from 4 farrow-to-finish farms. PROCEDURE: During a 9-month period, visits were made to a slaughter plant to remove cecal contents from market-weight hogs. Samples were obtained from 50 pigs/visit from designated farms so that samples were obtained 3 times from pigs of each of 4 farms. Isolation of Campylobacter spp was accomplished by use of enrichment broth and restrictive media, using microaerophilic conditions. RESULTS: Campylobacter spp were isolated from 70 to 100% of the pigs, depending on the farm and the date the samples were collected. Campylobacter coli was isolated from 20 to 100% (mean, 60%) of samples, and C jejuni was isolated from 0 to 76% (mean, 31%) of samples. Campylobacter lari was isolated from 2 pigs. Concentrations of C coli or C jejuni ranged from 10(3) to 10(7) colony-forming units/g of cecal content. CONCLUSIONS AND CLINICAL RELEVANCE: Campylobacter coli generally is accepted as a common inhabitant of the intestinal tract of swine. However, analysis of results of this study suggests that a relatively high prevalence of C jejuni may be found in pigs raised on specific farms.

Toxoplasma gondii is the aetiological agent of the zoonotic disease toxoplasmosis and transmitted among other ways by chemically and physically untreated, that is, raw pork to humans. The detection of Toxoplasma gondii is impossible by currently practiced meat inspection, but serological tests can be used to detect Toxoplasma gondii antibodies in pig herds and can consequently be helpful to identify potentially contaminated pork. Therefore, appropriate serological tests are required. In this study, serum samples of 1368 naturally exposed slaughter pigs from 73 Austrian farms were collected. Serum samples of at least 16 slaughter pigs per farm were tested. The prevalence of Toxoplasma gondii antibodies in serum was measured by a commercial available modified agglutination test (MAT) and compared to three different commercial available enzyme‐linked immunosorbent assays (ELISA). The MAT detected 6.5%, ELISA I 6.7%, ELISA II 4.8% and ELISA III 4.3% of the pigs as Toxoplasma gondii antibody positive. The agreement, according to the kappa coefficient (κ), was substantial between the MAT and ELISA I (κ = 0.62), II (κ = 0.64) and III (κ = 0.67). A better agreement was determined between ELISA I and II (κ = 0.715), ELISA I and III (κ = 0.747) and ELISA II and III (κ = 0.865). At least one pig per farm was detected Toxoplasma gondii antibody positive in 17 (23.3%) farms by the MAT, 26 (35.6%) farms by ELISA I, 16 (21.9%) farms by ELISA II and 11 (15.1%) farms by ELISA III. Pig farms with a high number of Toxoplasma gondii antibody‐positive pigs or high antibody titres were identified by all of the four used serological tests. Concerning the occurrence of Toxoplasma gondii antibodies in Austrian pig farms, a monitoring and surveillance programme would be reasonable to find high‐risk farms.

The study set out to explore risk factors for Salmonella infection in pigs, based on seroprevalence amongst slaughtered pigs, using a large study population of holdings and a comprehensive list of farm characteristics. Farm data were collected from pig quality assurance schemes and supplemented by a postal questionnaire. These data were used with meat juice serology results from ongoing abattoir Salmonella surveillance, for a multivariable risk factor analysis, modelling the ELISA sample to positive ratio directly (ELISA ratio). The study population contained 566 farms, covering a geographically representative spread of farms within the United Kingdom, with a mean average of 224 sample results per holding over a 4-year period. The model highlighted that temporal factors (quarterly and yearly cycles) and monthly meteorological summaries for rainfall, sunshine and temperature were associated with Salmonella presence (P < 0.01). The ELISA ratio was found to be highest in autumn and lowest in spring and summer, whereas yearly averages showed a greater degree of variation than seasonal. Two feed variables (homemix and barley) were found to be protective factors, as was a conventional, rather than organic or freedom foods, farm enterprise type. The number of annual pig deliveries and dead stock collections, and the main cause of pig mortality on the farm were found to be associated with Salmonella infection. Scottish farms had a lower ELISA ratio than other regions, and an increased number of pig farms within a 10-km radius was associated with a higher ELISA ratio. The study demonstrated that the analysis of routinely collected data from surveillance and quality assurance schemes was cost-effective, with sufficient power to detect modest associations between Salmonella and exposure variables. The model results can be used to inform on-farm Salmonella control policies and could target-specific geographical regions and seasons to assist the efficiency of surveillance.

This study investigated potential risk factors for the presence of Salmonella on 89 Alberta swine-finishing farms with the use of a questionnaire. Salmonella status was regressed on each fixed effect in a logistic mixed regression model, with farm as the random effect. Eleven variables were significant at the 10% level: farm type, number of square feet per pen, number of pigs per pen, source of feed, ration type, dust control measures, cat presence, reported effective mouse-control measures, time required to be away from pigs before visiting the farm, precautions taken when entering or leaving the farm, and reported use of antimicrobials through water. Three factors remained significant at the 5% level in the multivariable analysis: farm type, ration type, and precautions taken when entering or leaving the farm. Finishing barns at multisite operations or individual grow-to-finish farms had a greater risk of the presence of Salmonella at a single visit than did finishing barns at farrow-to-finish farms. The use of pelleted and wet feed was associated with higher odds of the presence of Salmonella than was the use of meal feed. Farms that required their personnel or visitors to shower before entering and before leaving had increased odds of the presence of Salmonella compared with farms that provided boots and coveralls; no significant difference was observed between the latter category and farms that used boot disinfection. Further work is necessary to better understand the effectiveness of all-in/all-out pig management and disinfection practices in reducing the presence of Salmonella in swine and to evaluate the association with certain types of feed.

Following a series of H5N1 cases in chickens and birds in a few states in Malaysia, there was much interest in the influenza A viruses subtypes that circulate among the local pig populations. Pigs may act as a mixing vessel for avian and mammal influenza viruses, resulting in new reassorted viruses. This study investigated the presence of antibodies against influenza H1N1 and H3N2 viruses in pigs from Peninsular Malaysia using Herdcheck Swine Influenza H1N1 and H3N2 Antibody Test Kits. At the same time, the presence of influenza virus was examined from the nasal swabs of seropositive pigs by virus isolation and real time RT-PCR. The list of pig farms was obtained from the headquarters of the Department of Veterinary Services, Malaysia, and pig herds were selected randomly from six of 11 states in Peninsular Malaysia. A total of 727 serum and nasal swab samples were collected from 4- to 6-month-old pigs between May and August 2005. By ELISA, the seroprevalences of swine influenza H1N1 and H3N2 among pigs were 12.2% and 12.1% respectively. Seropositivity for either of the virus subtypes was detected in less than half of the 41 sampled farms (41.4%). Combination of both subtypes was detected in 4% of all pigs and in 22% of sampled farms. However, no virus or viral nucleic acid was detected from nasal samples. This study identified that the seropositivity of pigs to H1N1 and H3N2 based on ELISA was significantly associated with factors such as size of farm, importation or purchase of pigs, proximity of farm to other pig farms and the presence of mammalian pets within the farm.

In Europe, on-farm biosecurity measures, involving a strict all-in/all-out batch-management system and decontamination of the rearing rooms between consecutive batches, are recommended to control Salmonella infection in growing pigs. However, implementation of these measures is often relaxed under common farming conditions. Therefore, this study was conducted to assess the relative contributions of batch-management system and room decontamination efficacy on Salmonella seroprevalence for different growing rates and subsequent slaughter ages of pigs. Because the impact of these factors cannot be easily evaluated by an observational approach in commercial farms, a stochastic simulation model representing the population dynamics, herd management, and Salmonella infection within a farrow-to-finish pig herd was used. Realistic levels were set for each factor under study (3 for batch-management system and slaughter age; 4 for room decontamination) to generate 54 simulation scenarios. Salmonella shedding prevalence in groups of slaughter pigs was then compared. A sensitivity analysis was performed to rank the impacts of the 3 factors on output. Batch-management system had little effect. In contrast, room decontamination efficacy had the greatest impact on Salmonella prevalence in pigs at slaughter. A drop in decontamination efficacy from 100 to 50%, with a strict all-in/all-out batch-management system and for all slaughter ages tested, noticeably increased (P<0.001) the prevalence and almost doubled it for the reference slaughter age. Our results suggest that the control of Salmonella in pig herds should primarily focus on room decontamination efficacy. Provided that a good level of room decontamination is ensured, some flexibility in batch management, in terms of pig mixing, would be acceptable to limit the number of underweight pigs delivered to the slaughterhouse.
