Groups of sheep were dosed with vaccines containing Corynebacterium pseudotuberculosis toxoid combined in varying amounts with 5 clostridial antigens. Resistance of the sheep to infection with C pseudotuberculosis was tested at 1, 6 and 12 months after vaccination by infection with pus from ovine lymph glands actively infected with C pseudotuberculosis. The outcome was assessed 3 months after challenge by slaughter and inspection of the sheep for caseous lymphadenitis lesions. Protection was demonstrated by a significant reduction in the proportion of immunised sheep exhibiting lesions compared with control sheep, and by fewer abscesses in affected immunised sheep than in affected control sheep. A positive correlation was found between amount of C pseudotuberculosis toxoid administered and degree of protection obtained. Chromatographically-purified toxoid induced essentially the same protection, suggesting that anti-toxic immunity is the major factor in protection.  相似文献

Resistance of sheep to infection was tested 6 months after immunisation by inoculation with caseous lymphadenitis pus. The outcome was assessed 3 months later by slaughter and inspection of the sheep for lesions of caseous lymphadenitis. immunised sheep were adequately protected against infection as shown by a significant reduction in the number of sheep exhibiting lesions compared with control sheep, and by fewer abscesses in affected vaccinated sheep than in affected control sheep. Influence of selenium on antibody production in sheep   总被引:3,自引:0,他引:3  
Three experiments were carried out, using sheep fed a marginally low selenium diet, to study the effect of selenium supplementation on the antibody response to tetanus toxoid and on the serum IgG concentration. Six groups of three six-month-old lambs were fed a basal diet containing 0.13 mg Se kg-1 supplemented with either 0.1, 0.5 or 1.0 mg Se kg-1, as sodium selenite or as selenomethionine. These animals generally showed enhanced antibody response to tetanus toxoid, parainfluenza-3 virus and Corynebacterium pseudotuberculosis, and their total serum IgG concentrations were higher than in unsupplemented control animals although few responses were statistically significant. In two field studies significantly higher titres to tetanus toxoid were detected in ewes injected with 100 mg selenium as barium selenate, although no influence on serum IgG concentrations was detected. Lambs from selenium supplemented ewes had significantly higher titres to tetanus toxoid than lambs from ewes in the control group. Dietary vitamin E supplementation had a similar effect on the antibody response to tetanus toxoid in ewes, though no additive effect was seen when vitamin E was given together with selenium.  相似文献

Sheep were immunized three times with a vaccine composed of filtrate from a 36 h culture of Corynebacterium pseudotuberculosis and a block polymer adjuvant. Immunization resulted in the development of exotoxin-neutralizing antibody. This corresponded to the recognition of a 31.6 kDa protein on sequential immunoblots of ammonium sulfate-precipitated filtrate. In addition sera from vaccinated sheep recognized at least eight bacterial cellular antigens on immunoblots of ether-extracted C. pseudotuberculosis, including bands of 12, 25.1, 31.6, 36.3, 39.8, 63.1, 70, 75 or 79.4 kDa. Sera from these sheep altered the colony growth characteristics of C. pseudotuberculosis in vitro. These results indicate that immunization with soluble C. pseudotuberculosis in vitro. These results indicate that immunization with soluble C. pseudotuberculosis antigen preparations that have been used in toxoid vaccines induces antibody responses to numerous cellular antigens in addition to exotoxin and suggest that serologically mediated antibacterial effects could be an important component in the protection from disease that has been reported following immunization with C. pseudotuberculosis toxoids.  相似文献

Cattle and sheep (30 each) were vaccinated with a combined Clostridium perfringens type C and C perfringens type D toxoid. Vaccination and blood sample collections were made every 2 weeks over a period of 8 weeks. Increases in antitoxin titers occurred after the 2nd administration of the 2.0-ml combined product. Highest titers were recorded when the 2nd vaccinal dose was given 2 weeks after the first. This confirms reports that response to clostridial antigens is greater when the 2nd immunizing dose is delayed 2 to 6 weeks after the initial dose.  相似文献

Corynebacterium pseudotuberculosis is a Gram positive bacterium that produces caseous lymphadenitis in sheep and goats, and a granulomatous lymphadenitis in llamas and alpacas. To evaluate the immune potential of different doses of cell wall and toxin components of C. pseudotuberculosis from alpaca origin, 12 adult alpacas were allotted at random to four groups, and SC inoculated in the left flank with vaccines composed of low and high doses of bacterial crude antigens, cell wall: 250 and 500 microg/ml and toxin: 133 and 265 microg/ml, respectively. The vaccines were supplemented with 20 microg/ml of muramyl dipeptide as adjuvant. Three alpacas were sham inoculated with adjuvant as a control. After 3 weeks, immunized and naive alpacas were challenged intradermally in the right flank with 1 x 10(6) colony forming units (CFU) of C. pseudotuberculosis. The alpacas were sacrificed at days 28, 58 and 112 after inoculation, and the degree of protection induced by vaccines was demonstrated by the absence of abscesses and/or bacteria. The alpacas vaccinated with high dose of toxin, did not show abscesses. In contrast, the alpacas vaccinated with a low dose of toxin showed abscesses at the inoculation site, regional, and renal lymph nodes. The cell wall vaccinated alpacas showed a lesser degree of protection than the other groups with superficial and internal abscesses. The control alpacas had persistent fever and abscesses at the inoculation site, regional, and internal lymph nodes. In addition, a robust and early humoral response was observed in all vaccinated alpacas after challenge, lasting at least 3 months. The results suggest that the toxin of C. pseudotuberculosis is a very important antigen, inducing a dose dependant protective immunity against this bacterium in alpacas.  相似文献

In Alberta, caseous lymphadenitis (CLA) is one of the leading causes of lamb and mutton carcass condemnation. In this study, serologic results confirmed a high (50-94%) incidence of exposure to Corynebacterium pseudotuberculosis, the causative agent of CLA, in mature, unvaccinated sheep in southern Alberta. To assess the efficacy and impact of vaccination with 2 commercial (Glanvac-6 and Case-Vac) and 1 experimental (WC+ MDP-GDP) CLA vaccines, a series of 3 field trials in 3249 ewes and lambs was conducted in affected flocks from 1992-1996. Efficacy was assessed from the serological response to vaccination, prevalence and size of injection site reactions by treatment, and the incidence of CLA abscesses. Overall, agglutinating antibody titres to C. pseudotuberculosis in lambs vaccinated with WC+MDP-GDP and Case-Vac remained significantly elevated above nonvaccinated control lambs for the 12 mo period after the initial vaccination. Lambs vaccinated with the WC/MDP-GDP maintained higher titres (P < 0.06) than those vaccinated with Case-Vac for the period from 6 to 12 mo after vaccination. Agglutinating antibody titres for lambs vaccinated with Glanvac did not differ from those of controls at any point during the 12 mo period after vaccination. The number of injection site reactions was elevated in lambs vaccinated with Glanvac as compared to those vaccinated with WC+MDP-GDP but the size of injection site reactions did not significantly differ. Sheep vaccinated with WC+ MDP-GDP also had a reduced incidence of putative CLA abscesses, although confirmation of the presence of C. pseudotuberculosis was only successful in a small number of instances.  相似文献

Two groups of male, 9-week-old goats (5 goats/group) were vaccinated subcutaneously with formalized exotoxin of Corynebacterium pseudotuberculosis, with Freund's incomplete adjuvant. Each goat was given 2 vaccinations, 2 weeks apart. At each vaccination, each group 1 goat was given 0.5 ml of toxoid, and each group 2 goat was given 1 ml of toxoid. Twenty days after the 2nd vaccination, vaccinated goats and 5 nonvaccinated 12-week-old goats (controls) were inoculated intradermally (challenge exposed) with live C pseudotuberculosis, monitored for 13 weeks, and euthanatized. At necropsy, 5 of the 10 vaccinated goats did not have C pseudotuberculosis lesions, 3 had abscesses limited to the inoculation site and draining lymph node, and 2 had disseminated bacterial lesions. Of the 5 nonvaccinated controls, 4 had disseminated abscesses and 1 had a single abscess in an internal node. Serologically, 9 of the 10 vaccinated goats developed positive (greater than or equal to 1:8) antibody titers against the exotoxin within 1 week after inoculation; the 10th goat seroconverted 2 weeks after inoculation, whereas control goats required 3 weeks to develop a positive antibody response. Therefore, early during an infection with C pseudotuberculosis, antibodies against the exotoxin may protect a goat against spread of the organism. Two hundred Merino wether hoggets were used to examine the effect of Corynebacterium pseudotuberculosis infection (caseous lymphadenitis) on wool production and bodyweight. Sheep which were challenged with C. pseudotuberculosis (artificially infected) and not vaccinated against this disease produced 0.20 kg less clean wool than unchallenged controls during the following 12 months. The incidence of sheep with lesions in the group that was vaccinated prior to challenge was 55% lower than in unvaccinated challenged sheep but their wool production was not significantly different from either the controls or the unvaccinated challenged sheep. Vaccinated sheep were also heavier than unvaccinated sheep 12 months after challenge. These results indicate that caseous lymphadenitis infection may reduce wool production.  相似文献

Over 3 years, the immunogenic responses of various batches of multi-component clostridial vaccines in sheep, rabbits and guinea pigs were compared. Fully susceptible healthy sheep were found to be more suitable than rabbits or guinea pigs for testing the potency of multi-component clostridial vaccines containing Clostridium novyi type B, C. perfringens type D, C. septicum and C. tetani, and recommendations are made that sheep are the preferred species for testing the potency of clostridial vaccines.  相似文献

Food and Drug Administration regulations currently permit addition of .3 mg of Se per kilogram of diet for chickens, turkeys, ducks, swine, sheep, and cattle. However, field reports indicate that this level may not be adequate for ruminants in all situations. Because sodium selenite is the most common supplemental form and is known to be readily absorbed to particles or reduced to insoluble elemental Se or selenides in acid, anaerobic environments, studies were conducted with dairy cattle, sheep, and horses fed sodium selenate to determine whether Se from this source was more bioavailable than Se from sodium selenite. A 2-wk period of no Se supplementation was followed by 49 or 56 d of Se supplementation at .3 mg/kg of dietary DM. Serum Se concentrations and glutathione peroxidase (GSHPx) activities measured initially and periodically thereafter revealed no difference between Se forms in sheep and horses and only a small (P less than .05) advantage for selenate in supporting serum Se concentration in dairy cattle. Selenium concentrations in skeletal muscle and liver of sheep were not different between Se forms. Serum Se, but not GSHPx, increased with time, and .3 mg of supplemental Se per kilogram of dietary DM from either sodium selenate or sodium selenite supported normal serum Se concentrations in sheep, dairy cattle, and horses.  相似文献

OBJECTIVES: To determine the effect of location for administration of clostridial vaccines on behavior, growth performance, and health of calves at a feedlot, the relative risk of calves developing an injection-site reaction or being misdiagnosed as having bovine respiratory disease complex (BRDC), and the percentage of subcutaneous injection-site reactions that were detectable on carcasses after the hides were removed. ANIMAL: 170 newly arrived calves at a feedlot. PROCEDURE: Eating and drinking behaviors of calves during the initial 57 days after arrival were observed at a commercial feedlot, using an electronic monitoring system. Calves were assigned randomly to receive a clostridial vaccine (base of ear or neck). Data on reactions at the injection site were collected. RESULTS: Mean daily gain (MDG) for the initial 57 days did not differ significantly between treatments. Risk of being misdiagnosed as having BRDC was not associated with location for administration of vaccine. Calves vaccinated in the base of the ear were at higher risk of having an injection-site reaction at day 57 or at slaughter. Eighty-nine percent (95% confidence interval, 52 to 100%) of injection-site reactions in the neck could not be located on the carcasses after hides were removed. Calves vaccinated in the neck drank significantly fewer times per day during the first 57 days than calves vaccinated in the base of the ear. CONCLUSIONS AND CLINICAL RELEVANCE: Location for administration of a clostridial vaccine did not significantly affect health, growth performance, or eating behavior. Most subcutaneous injection-site reactions were not detectable after the hide was removed.  相似文献

The effect of an inactivated vaccine against C. pseudotuberculosis infection was tested on castrated male kids from a herd free from caseous lymphadenitis. The animals were divided into 3 groups with 8 animals in each. Group 1 was immunized with crude filtrated C. pseudotuberculosis toxoid and whole killed organisms, while Group 2 in addition was given levamisole. The kids were vaccinated twice at an interval of 4 weeks. Group 3 consisted of unvaccinated animals. All groups were challenged subcutaneously with live bacteria 4 weeks after the last vaccination. Unvaccinated animals showed the most severe course of illness after challenge. Development of abscesses in the regional lymph nodes (Inn. subiliaci) was significantly more common in unvaccinated than in vaccinated kids at necropsy 2 months after challenge. There was, however, no such difference between the vaccinated groups, and there was no difference between any of the groups as regards abscess formation at the inoculation site. In each of the 2 vaccinated groups, there was a titre rise following vaccination in the hemolysis inhibition test, whereas no such rise was seen in the bacterial agglutination test. The titre values in both tests increased significantly after challenge in all the groups, the increase being most rapid in the vaccinated animals. The present investigation indicates that development of caseous lesions in lymph nodes in goats, following subcutaneous inoculation with C. pseudotuberculosis, can be reduced by an inactivated vaccine containing whole organisms and crude toxin.  相似文献

The efficacy of a Corynebacterium pseudotuberculosis bacterin to protect sheep immunologically against development of caseous lymphadenitis was evaluated in controlled challenge-exposure experiments. Sixty-three mixed-breed, white-faced lambs were used. The lambs were 10 to 12 weeks old and were randomly assigned to 3 groups (21 lambs/group). Group 1 was vaccinated once, using 2 ml of a C pseudotuberculosis bacterin (given subcutaneously) in the right axillary region at the beginning of the study. Group 2 was vaccinated twice; the 1st vaccination was given at the same time that lambs in group 1 were vaccinated and the 2nd vaccination was given 4 weeks later. Group 3 (nonvaccinated controls) was given physiologic saline solution (2 ml, subcutaneously). Each lamb was challenge exposed (ie, given 2 ml of live Corynebacterium pseudotuberculosis inoculum [6 X 10(6) colony-forming units/ml], subcutaneously at 4 different sites) during the 20th week of the study. All lambs were killed and necropsied during week 33. The mean number of abscesses per lamb was 7 for group 1, 4 for group 2, and 32 for group 3. Significant differences in the size of the abscesses were not found between the groups. Results of the study indicated that the vaccine provided immunologic protection of lambs against challenge exposure to Corynebacterium pseudotuberculosis.  相似文献

Sheep were vaccinated with a killed Staphylococcus aureus vaccine (2 doses) which had been cultured in vitro (Group 1), a killed S. aureus vaccine (2 doses) cultured in vivo (Group 2) or a single dose of a live vaccine (Group 3). Other sheep were used as non-vaccinated controls. All sheep were challenged by intravenous injection of 2.6 x 10(11) washed, viable S. aureus organisms, the vaccinated animals being given the challenge inoculum at various intervals after vaccination. The control sheep survived for 29h (mean) after challenge. Animals given killed vaccines survived longer, (particularly Group 2) if challenged less than 40 days post-vaccination, compared with those challenged more than 40 days post-vaccination. Animals in Group 3 survived longer if challenged after 40 days post-vaccination than those in Groups 1 or 2. There were no significant differences between the treatment groups for numbers of S. aureus recovered from blood in the 3h period following challenge. Histological and bacteriological evidence showed that the kidneys were more severely affected by the challenge inoculum than heart, spleen, liver or lungs. The kidneys showed both toxigenic and lymphoreticular reactions and large numbers of staphylococci were recovered more reliably from kidneys than other organs.  相似文献

SUMMARY: :Five groups of 5 shorn and 5 unshorn caseous lymphadenitis (CLA)-free Merino wether weaners were each placed in feedlot pens with 6 Merino ewes, 2 or more of which had CLA lung lesions but no discharging superficial lesions. The sheep were kept together for 5 months.
Twenty-eight per cent of the shorn weaners and 20% of the unshorn weaners developed antibodies to Corynebacterium pseudotuberculosis. At slaughter, 8% of the shorn weaners and 12% of the unshorn weaners had CLA lesions in either lungs, lymph nodes or both. In the absence of contact with CLA-infected ewes, a control group of 5 shorn and 5 unshorn weaners failed to develop antibodies to C. pseudotuberculosis or CLA lesions in the same period. This showed that sheep with CLA abscesses in the lungs but no discharging superficial abscesses were a source of C. pseudotuberculosis infection to other sheep. Aust Vet J. 64: 261–263  相似文献

Two Friesian cows were used to attempt to produce colostrum containing a high concentration of clostridial antibodies which could be fed to newborn lambs in order to passively transfer immunity to diseases caused by clostridia. One cow was given a commercial multicomponent clostridial sheep vaccine in two successive pregnancies and the second cow in one pregnancy. The first cow produced a low concentration of epsilon antitoxin (Clostridium perfringens, type D) in its blood and colostrum after the first course of three injections of vaccine. A higher concentration was produced by cow 2 after a course of six injections and by cow 1 after a further course of four injections in its next pregnancy. Two hundred ml of colostrum from cow 1 (after the second course of vaccine) was given to 12 newborn colostrum-deprived lambs. All showed a high concentration of antitoxin 48 hours later. The lambs were actively immunised by injections of the same clostridial vaccine at three and nine weeks or six and 12 weeks old and all produced sufficient antitoxin to protect up to slaughter at 24 weeks. It is concluded that colostrum from cows vaccinated with sheep clostridial antigens can be fed to protect lambs passively.  相似文献

The immunogenicity of the Pasteurella multocida toxin (PMT) was studied in murine model systems. Mice were vaccinated with either formaldehyde treated pure PMT (pure toxoid) or formaldehyde treated crude extract of toxigenic P. multocida (crude toxoid). The corresponding mean anti-PMT titres, sero-conversion rates and survival rates after challenge with affinity purified PMT were compared. When assessed both by anti-PMT titres and seroconversion and challenge, pure toxoid was a more potent immunogen than crude toxoid. This greater immunogenic potency was unaffected by the addition of killed cell preparations of Bordetella bronchiseptica, non-toxigenic P. multocida and B. pertussis. Increasing anti-PMT titres and seroconversion rates were induced by increasing doses of formaldehyde treated PMT (fPMT) in the pure toxoid vaccines, but not in the vaccines containing crude toxoid. However, improved survival rates were observed for both types of vaccine, when the fPMT content was raised. Immunization of pregnant mice with vaccines containing fPMT induced protection of the offspring against challenge with PMT; the protection of the offspring corresponded to that of the mother.  相似文献

AIMS: To evaluate a multivalent leptospiral and clostridial vaccine for prevention of renal colonisation and urinary shedding in sheep, following experimental challenge with New Zealand strains of Leptospira borgpetersenii serovar Hardjo type Hardjobovis and L. interrogans serovar Pomona.

METHODS: Two separate but similarly designed studies were conducted. In both studies, Romney-cross lambs, aged 9–11 weeks, were randomly allocated to a vaccinated group and a control group. Vaccinated lambs each received two 1.5-mL S/C doses of a multivalent leptospiral and clostridial vaccine, 4 weeks apart, and animals in the control groups received the same dose of saline. Groups of 12 vaccinated and 12 control lambs were randomly selected in each study for challenge with serovars Hardjo or Pomona. Challenge was initiated 16 weeks following the second vaccination with three daily doses of live leptospires by intranasal and conjunctival routes. Following challenge, urine samples were collected weekly for 6 weeks, for dark field microscopy and leptospiral culture; 6 weeks after challenge the lambs were slaughtered and kidneys collected for leptospiral culture.

RESULTS: In lambs challenged with serovar Hardjo, 8/12 unvaccinated lambs had ≥1 urine or kidney sample that was positive for leptospires following culture, compared with 0/12 lambs in the vaccinated group (p=0.001). In lambs challenged with serovar Pomona, 9/12 unvaccinated lambs had ≥1 urine or kidney sample that was positive following culture, compared with 0/12 lambs in the vaccinated group (p<0.001). Prevention of renal colonisation and urinary shedding, expressed as the prevented fraction, was 100 (95% CI=61.7–100)% and 100 (95% CI=68.3–100)% against challenge with serovars Hardjo and Pomona, respectively, at 4 months after vaccination.

CONCLUSIONS AND CLINICAL RELEVANCE: Use of a multivalent leptospiral and clostridial vaccine demonstrated protection against challenge from New Zealand strains of serovars of Hardjo and Pomona 4 months after vaccination in lambs first vaccinated at 9–11 weeks of age. Further studies are required to assess the duration of immunity against challenge in sheep.  相似文献

A field trial to evaluate a whole cell vaccine for the prevention of caseous lymphadenitis (CLA) in sheep and goats was performed in one goat herd and one sheep flock over a period of three years. In goats, there was a nonstatistically significant trend for fewer cases of CLA in the vaccinated animals compared to the controls. In sheep, from six months to 36 months postinitial vaccination, the proportion of vaccinated sheep that developed CLA was significantly less (p less than 0.05) than in the control sheep. The antibody titers to Corynebacterium pseudotuberculosis as detected by microagglutination assay were significantly different (p less than 0.0001) at all times except at the initial vaccination. Swellings occurred at the vaccination site at an incidence level of 29.6% in goats and 34.1% in sheep. The vaccine appeared to be efficacious in reducing the proportion of sheep that developed CLA when challenged naturally in a field situation.  相似文献
