采用PstⅠ、pvuⅡ、HindⅢ、EcoRⅠ4种限制性内切酶,研究了20尾青海湖裸鲤线粒体DNA的限制性片段长度多态性。其检测出11个酶切位点,发现PstI和EcoRI两种酶切类型具有多态性。  相似文献

用SDS-ProteinaseK消化EDSV抗原-抗体复合物,苯酚-氯仿抽提DNA,得到了完整的EDSV基因组DNA,用限制性内切酶BalⅠ、BalⅡ、BamHⅠ、ClaⅠ、EcoRⅠ、HindⅢ、HpaⅠ、KpnⅠ、PstⅠ、PvuⅡ、SalⅠ、SmalⅠ、XbaⅠ酶切及EcoRⅠ BamHⅠ双酶切消化,对EDSV分离株Hd1的DNA进行了限制性内切酶酶切分析。结果表明,分离株Hd1与标准株AV127DNA各酶切片段数及各片段的相对分子量(kb)未见差异,说明分离株Hd1和标准株AV127属于同一基因型。将EcoRⅠ BamHⅠ双酶切得到的7个片段,与双酶切线性化的pUC19载体质粒连接,转化E.coliDH5α,克隆了其中的5个片段。  相似文献

用 1 2种识别 6碱基的限制性内切酶ApaⅠ、BamHⅠ、Bg1Ⅰ、DraⅠ、EcoRⅠ、EcoRⅤ、KpnⅠ、PvuⅡ、PstⅠ、SacⅠ、SmaⅠ和XhoⅠ研究了河北省 5个山羊品种和类群共计 61个个体mtDNA的RFLP。结果检测到 3 3个酶切位点 ,1 4种限制性态型 ,3种基因单倍型 ,其中EcoRⅠ和CalⅠ表现出多态。基因单倍型间平均遗传距离为 0 .0 0 3 9,群体平均遗传多态度π值为 0 .1 2 %,表明其遗传分化程度较低  相似文献

摘要:选用BamH Ⅰ、Kpn Ⅰ、Pst Ⅰ、EcoR Ⅰ、Xba Ⅰ、HindⅢ等6种限制性内切酶对EDS两病毒内蒙古地区分离株(N3、B4)和参考株(AV-127)病毒基因组DNA进行酶切分析,均产生4,5,9,4,2和10条DNA片段,而且N3、B4株与参考株病毒基因组DNA之间的酶切图形、片段大小也十分相似,从分子水平上证实N3、B4株是EDS两病毒,同时也说明限制性内切酶分析法是检测EDS76病毒的一种简便快速的方法。  相似文献

鹑源减蛋综合征病毒QAVc—94株的酶切图谱分析   总被引:2,自引:0,他引:2  
选HindⅢ、EcoRⅠ、PstⅠ、SphⅠ、BamHⅠ、BamHⅠ和BglⅠ6种限制酶,对QAVc-94株和AV-127国际标准株的DNA进行酶切图谱比较结果发现,QAVc-94株用上述6种酶酶切得到的片段数分别为10、3、12、4、6、9条,将各片段碱基数相加得出其基因组长34.7kb,略高于国内的报道,而与Zask等的结果相接近,从分子水平上将QAVc-94鉴定为一株EDSV。对照AV-127株的酶切结果与文献报道相符。QAVc-94株酶切图谱分析显示:HindⅢ和EcoRⅠ的酶谱与报道的其他EDSV分离株基本相似,HindⅢ的酶谱与国内研究最多的AA-2长春分离株一致,EcoRⅠ的酶谱结果与Zask等报道的B8/78株相同。而其他4种限制酶的酶切图谱与已报道的各地分离株相比,差异较大,如PstⅠ和BglⅠ多出2-5个识别位点。说明鹌鹑源EDSV已与鸡源、鸭源、鹅源EDSV有较大的变异,是一株血清型相同而基因型不同的鹌鹑减蛋综合征病毒。  相似文献

利用高渗缓冲液裂解感染鸭瘟病毒的鸭胚成纤维细胞,加入核酸酶消化细胞DNA,再抽提病毒DNA的方法提取鸭瘟病毒基因组,结果证明,这种方法可以最大限度地消除细胞DNA的污染,得到纯净的病毒基因组DNA.用10种限制性内切酶对鸭瘟病毒疫苗株和分离株的基因组进行酶切分析,酶切产物电泳结果表明,Sma Ⅰ、Sal Ⅰ、Pst Ⅰ、Not Ⅰ、Hind Ⅲ、EcoR Ⅴ和BamH Ⅰ 7种限制酶产生酶切图谱二者几乎完全一致,EcoR Ⅰ和Sac Ⅰ仅有个别条带不同,BglⅡ产生的条带迁移率差别较大,但条带数量相同.  相似文献

试验旨在对基因组DNA的制备进行研究,为细菌人工染色体(bacterial artificial chromosome,BAC)文库的构建奠定基础。以豁眼鹅全血为试验材料,提取高质量的基因组DNA,分别采用HindⅢ、EcoRⅠ和BamHⅠ3种限制性内切酶对所提基因组DNA进行部分酶切,并利用控制酶切时间、设置不同的HindⅢ酶切浓度梯度对基因组DNA进行部分酶切。结果表明,HindⅢ为最佳限制性内切酶,并得到了最佳酶切用量(40U/μL)。该方法所制备的基因组DNA质量较好,可用于BAC文库的构建。  相似文献

以从内蒙古区分离的2株传染性喉气管炎病毒(ILTV)株的DNA为模板,用设计好的1对引物对这2株毒株的gB基因进行了PCR扩增,并对扩增产物分别用BglⅡ、PstⅠ、HindⅢ限制性内切酶酶切分析。结果表明,2个分离株均能特异性地扩增出该病毒gB基因片段,片段大小完全一致,为2.6kb;扩增产物的酶切图谱与参考株完全一致。  相似文献

根据不同物种间同一基因核苷酸序列的保守性及相似性,设计特异性引物,利用聚合酶链式反应技术(PCR),扩增鸡心脏型脂肪酸结合蛋白(H—FABP)基因第二内含子序列;根据已经发表的鸡脂肪型脂肪酸结合蛋白(A-FABP)基因mRNA序列,设计特异性引物,扩增鸡A-FABP基因第三内含子序列。选用Hha Ⅰ、Hae Ⅲ、Hinf Ⅰ、BamH Ⅰ、EcoR Ⅰ、Pst Ⅰ、Xmn Ⅰ和Hind Ⅲ8种限制性内切酶对扩增产物分别进行单酶切,发现H—FABP基因第二内含子Hha Ⅰ PCR—RFLP及A-FABP基因第三内含子Hinf Ⅰ PCR—RFLP。  相似文献

从2型犬腺病毒的MDCK细胞培养物中提取线状双链病毒基因组DNA,利用限制性内切酶Pst Ⅰ、Bgl Ⅰ和Hind Ⅲ分别进行酶切,在1%琼脂糖凝胶上电泳,随后Southern印迹到尼龙膜上,利用Digoxin标记的2型犬腺病毒E_3区PCR产物与膜上DNA杂交,结果发现,E_3区探针分别与病毒DNA的PstIB片段、BglIAl片段和Hind ⅢA2、B2片段呈现阳性杂交反应。该结果为E_3区的克隆及犬腺病毒载体构建奠定了基础。  相似文献

: Some feed ingredients bind more acid in the stomach than others and for this reason may be best omitted from pig starter foods if gastric acidity is to be promoted. The objective of this study was to measure the acid-binding capacity (ABC) of ingredients commonly used in pig starter foods. Ingredients were categorised as follows: (i) milk products (n = 6), (ii) cereals (n = 10), (iii) root and pulp products (n = 5), (iv) vegetable proteins (n = 11), (v) meat and fish meal (n = 2), (vi) medication (n = 3), (vii) amino acids (n = 4), (viii) minerals (n = 16), (ix) acid salts (n = 4), (x) acids (n = 10). A 0.5 g sample of food was suspended in 50 ml distilled de-ionised water with continuous stirring. This suspension was titrated with 0.1 mol/L HCl or 0.1 mol/L NaOH so that approximately 10 additions of titrant was required to reach pH 3.0. The pH readings after each addition were recorded following equilibration for three minutes. ABC was calculated as the amount of acid in milliequivalents (meq) required to lower the pH of 1 kg food to (a) pH 4.0 (ABC-4) and (b) pH 3.0 (ABC-3). Categories of food had significantly different (P < 0.01) ABC values. Mean ABC-4 and ABC-3 values of the ten categories were: (i) 623 (s.d. 367.0) and 936 (s.d. 460.2), (ii) 142 (s.d. 79.2) and 324 (s.d. 146.4), (iii) 368 (s.d. 65.3) and 804 (s.d. 126.7), (iv) 381 (s.d. 186.1) and 746 (s.d. 227.0), (v) 749 (s.d. 211.6) and 1508 (s.d. 360.8), (vi) 120 (s.d. 95.6) and 261 (s.d. 163.2), (vii) 177 (s.d. 60.7) and 1078 (s.d. 359.0), (viii) 5064 (s.d. 5525.1) and 7051 (s.d. 5911.6), (ix) 5057 (s.d. 1336.6) and 8945 (s.d. 2654.1) and (x) -5883 (s.d. 4220.5) and -2591 (s.d. 2245.4) meq HCl per kg, respectively. Within category, ABC-3 and ABC- 4 values were highly correlated: R2 values of 0.80 and greater for food categories i, iv, v, vi, vii and viii. The correlation between predicted and observed ABC values of 34 mixed diets was 0.83 for ABC-4 and 0.71 for ABC-3. It was concluded that complete diets with low ABC values may be formulated through careful selection of ingredients. The final pH to which ABC is measured should matter little as ABC-3 and ABC-4 are highly correlated.  相似文献

为探讨伊氏锥虫对安锥赛抗药性的分子机理,以布氏锥虫对砷剂药物抗药性相关的TbTA1基因设计引物,以55℃、57℃、60℃、63℃和65℃不同退火温度,分别从伊氏锥虫敏感株的cDNA和基因组DNA中扩增出TbTA1基因的全长序列,但是从安锥赛抗药株伊氏锥虫的cDNA和基因组DNA中都未扩增出TbTA1基因,这表明基因TbTA1可能与伊氏锥虫安锥赛抗药性的产生有关。伊氏锥虫与布氏锥虫的,TbTA1基因序列比较,它们有10个碱基不同,即第88位的G(T.e)-A(T.b)、第144位的T(T.e)-C(T.b)、第224位的C(T.e)-T(T.b)、第471位的C(T.e)-T(T.b)、第472位的A(T.e)-G(T.b)、第549位的G(T.e)-T(T.b)、第1008位的C(T.e)-T(T.b)、第1033位的A(T.e)-G(T.b)、第1293位的A(T.e)-G(T.b)和第1384位的T(T.e)-C(T.b),其中有5个碱基所编码的氨基酸不同,即Val^30(T.e)Ile(T.b)、Ala^75(T.e)-Val(T.b)、Ile^158(T.e)-Val(T.b)、Thr^345(T.e)-Ala(T.b)和Ser^462(T.e)-Pro(T.b),这表明伊氏锥虫与布氏锥虫的TbTA1差异可能是它们的种间差异。  相似文献

An acid–pepsin digestion technique was used to examine portions of breast muscle and heart from raptors for encysted protozoans. Apicomplexan zoites were present in 52 (45.6%) of the 114 samples examined: 11 of 12 (91.7%) red-shouldered hawks (Buteo lineatus), 20 of 34 (58.8%) red-tailed hawks (Buteo jamaicensis), two of seven (28.6%) Cooper's hawks (Accipiter cooperi), three of four (75%) sharp-shinned hawks (Accipiter striatus), one (100%) Mississippi kites (Ictinia misisippiensis), one of two (50%) American kestrels (Falco sparverius), one bald eagle (Haliaeetus leucocephalus), one of two (50%) golden eagles (Aquila chrysaetos), one of three (33%) turkey vultures (Cathartes aura), two of three (66.7%) black vultures (Coragyps atratus), three of six (50%) great-horned owls (Bubo virginianus), five of 15 (33.3%) barred owls (Strix varia), and one of 12 (8.3%) screech owls (Asio otus). Encysted protozoans were not observed in digests of tissues from three broad-winged hawks (Buteo platypterus), four ospreys (Pandion haliaetus), and five barn owls (Tyto alba). Apicomplexan cysts resembling Sarcocystis species were observed in tissue sections of muscles from 28 (37.8%) of 74 raptors.  相似文献

将120只蛋仔鸡(3日龄)随机分为10组,以常规方法进行饲喂,把不同浓度的铅与Vc(Vc用量为0.2mg/g,铅用量为0.02mg/g,0.05mg/g,0.08mg/g,0.11mg/g)配成溶液,用小针管进行口腔灌服,饲养14日后空腹抽血,测定血液中丙二醛(MDA)含量及超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性的变化。结果表明:随着铅浓度的增加,血液中MDA含量升高,其中Pb(3)、Pb(4)组升高显著(P<0.05),Vc Pb组无显著性差异;SOD活性显著降低,Pb(4)组差异极显著(P<0.01),Pb(1)、Vc Pb(1)组差异不显著,其它组差异显著(P<0.05);POD活性在Pb(1)、Pb(2)组明显降低(P<0.05),Pb(3)组显著升高(P<0.05),Pb(4)组无变化(P>0.05),Vc Pb组差异显著;CAT活性逐渐降低,Pb(3)、Pb(4)、Vc pb(3)、Vc pb(4)组差异极显著(P<0.01),Pb(2)、Vc Pb(2)差异显著(P<0.05);Vc Pb组与加Pb组比较,Vc Pb(4)组上述指标差异均显著(P<0.05)。  相似文献

Serum samples from 840 animals were examined for Leptospira spp. antibodies by microscopic agglutination test (MAT) to assess the risk factors and the prevalence of leptospirosis among animal herds of Assam, Meghalaya, Mizoram the north eastern (NER) provinces. They were compared with Tamilnadu (TN) the southern province of India for the serovar and risk factor inconsistency. Serovar Ballum was reported to be prevalent (28.1 %) in Assam and Grippotyphosa (16.1 %) in Tamilnadu. The overall seropositivity observed was 36.8 %(206/560) from NER and 30.7 %(86/280) from TN. In this study, the higher seroprevalence was observed in pigs (42.6 %), cattle (39.8 %) and goats (26 %) in NER. Cattle (36.4 %) and goat (17.6 %) showed higher prevalence in TN. The presence of rodents in pig herds was found to be significant (P = 0.0088) in NER and it was for cattle in TN (P = 0.0063). We suggest that a program of rodent control should be included in the flock management practices aiming to reduce transmission of the leptospires.  相似文献

甘肃禾本科饲用植物资源丰富,据初步统计,共有74属301种,是构成甘肃天然草地植被的重要成分。本研究利用灰色关联分析法,对甘肃禾本科饲用植物11个属的7项营养指标进行了综合分析,选出了优质种类。综合评价结果表明,各属营养成分灰色关联度为臭草属(Melica)(0.6887)>隐子草属(Cleistogenes)(0.6605)>早熟禾属(Poa)(0.6271)>剪股颖属(Agrostis)(0.5889)>羊茅属(Festuca)(0.5762)>雀麦属(Bromus)(0.5616)>披碱草属(Elymus)(0.5508)>针茅属(Stipa)(0.5095)>异燕麦属(Helictotrichon)(0.5082)>芨芨草属(Ach-natherum)(0.4577)>鹅观草属(Roegneria)(0.4333)。  相似文献

对沧海生物公司生产的益生素产品“康源菌宝”中两种主要益生菌-乳酸杆菌和蜡样芽孢杆菌进行了16种抗菌药的药敏实验。结果表明,头孢呋新、四环素、庆大霉素、青霉素、复合磺胺对两种菌无任何抑制作用;其他抗菌药对其均有不同程度的抑制作用,链霉素、利福平、头孢三嗪、头孢哌酮抑制作用最小;卡那霉素对乳酸杆菌抑制作用最小,对蜡样芽孢杆菌抑制作用较小。  相似文献

Previous structural characterizations of marsupial milk oligosaccharides have been performed in the tammar wallaby, red kangaroo, koala, common brushtail possum and the eastern quoll. To clarify the homology and heterogeneity of milk oligosaccharides among marsupial species, which could provide information on their evolution, the oligosaccharides of wombat milk carbohydrate were characterized in this study. Neutral and acidic oligosaccharides were isolated from the carbohydrate fractions of two samples of milk of the common wombat and characterized by 1H‐nuclear magnetic resonance spectroscopy. The structures of six neutral saccharides were found to be Gal(β1‐4)Glc (lactose), Gal(β1‐3)Gal(β1‐4)Glc (3'‐galactosyllactose), Gal(β1‐3)Gal(β1‐3)Gal(β1‐4)Glc (3',3”‐digalactosyllactose), Gal(β1‐3)Gal(β1‐3)Gal(β1‐3)Gal(β1‐4)Glc, Gal(β1‐3)Gal(β1‐3)[Gal(β1‐4)GlcNAc(β1‐6)]Gal(β1‐4)Glc (galactosyl lacto‐N‐novopentaose I) and Gal(β1‐3)[Gal(β1‐4)GlcNAc(β1‐6)]Gal(β1‐3)[Gal(β1‐4)GlcNAc(β1‐6)]Gal(β1‐4)Glc (lacto‐N‐novooctaose), while those of six acidic saccharides were Neu5Ac(α2‐3)Gal(β1‐3)Gal(β1‐4)Glc. (sialyl 3'‐galactosyllactose), Neu5Ac(α2‐3)Gal(β1‐3)Gal(β1‐3)Gal(β1‐4)Glc (sialyl 3',3”‐digalactosyllactose), Neu5Ac(α2‐3)Gal(β1‐3)[Gal(β1‐4)GlcNAc(β1‐6)]Gal(β1‐4)Glc (sialyl lacto‐N‐novopentaose a), Gal(β1‐3)[Neu5Ac(α2‐3)Gal(β1‐4)GlcNAc(β1‐6)]Gal(β1‐4)Glc (sialyl lacto‐N‐novopentaose c), Neu5Ac(α2‐3)Gal(β1‐3)Gal(β1‐3)Gal(β1‐3)Gal(β1‐4)Glc,, Neu5Ac(α2‐3)Gal(β1‐3)Gal(β1‐3)[Gal(β1‐4)GlcNAc(β1‐6)]Gal(β1‐4)Glc and Gal(β1‐3)Gal(β1‐3)[Neu5Ac(α2‐3)Gal(β1‐4)GlcNAc(β1‐6)]Gal(β1‐4)Glc. In addition, small amounts of sulfated oligosaccharides but no oligosaccharides containing Neu5Gc or α(2–6) linked Neu5Ac were detected.  相似文献

将8只30日龄、体重(6.9±1.0)kg的小尾寒羊×无角陶赛特母羔羊随机分为2组(每组4只),以研究口服福尔马林对羔羊自由采食、消化代谢、屠宰性能和甲醛残留的影响。结果表明,在55~80、95~120和125~150日龄3个日龄段,对照组羔羊的干物质采食量分别为(567.3±27.5)、(752.4±9.01)和(1044.1±30.8)g/(d.头),口服福尔马林组分别增加22.02%(P0.01)、28.75%(P0.01)和41.79%(P0.01);对照组羔羊的氮保留分别为(12.3±0.8)、(13.6±0.2)和(18.0±0.6)g/(d.头),口服福尔马林组分别增加25.20%(P0.01)、41.91%(P0.01)和46.11%(P0.01);对照组羔羊的日增重分别为(157.1±11.7)、(246.4±24.4)和(250.0±8.3)g/(d.头),口服福尔马林组分别增加18.20%(P0.05)、23.21%(P0.05)和27.16%(P0.01);而两组间的日粮消化率无显著差异。240日龄对照组和口服福尔马林组羔羊的胴体重分别为(20.37±1.78)和(24.55±1.14)kg/头(P0.01),屠宰率分别为48.59%±1.08%和50.46%±1.02%(P0.05),瘦肉率分别为65.22%±1.57%和64.84%±1.11%(P0.05)。羔羊血液、肌肉、肾脏、肝脏、瘤胃组织和小肠组织中的甲醛含量对照组分别为(1.84±0.44)mg/L、(0.35±0.26)mg/kg、(1.10±0.67)mg/kg、(0.74±0.34)mg/kg、(0.91±0.72)mg/kg和(0.79±0.41)mg/kg,在试验组分别为(1.47±0.61)mg/L、(0.54±0.34)mg/kg、(1.65±0.71)mg/kg、(0.91±0.38)mg/kg、(2.03±0.71)mg/L和(0.85±0.42)mg/L。因此,口服福尔马林可增加羔羊的自由采食量而不影响消化率,提高羔羊对日粮的消化量和羔羊日增重;以上这种作用随羔羊的日龄增加而增强;口服福尔马林提高羔羊的胴体重、屠宰率,但不影响胴体的瘦肉率;长期口服福尔马林羔羊的组织器官中甲醛的含量低于2.0 mg/kg。  相似文献
