Cotton yield improvement is vital to fulfill rising global demands. The identification of major quantitative trait loci (QTL) for yield components was helpful in molecular marker-assisted selection (MAS) to improve cotton yield. We previously identified a densely populated QTL region for fiber qualities and yield components on chromosome D8 (Chro.D8) of Upland cotton from a (7235 × TM-1)RIL. In the present study, to fine-map yield component QTLs, we chose three overlapped recombinant inbred lines (RILs) with different intervals included the yield component QTLs, and backcrossed each line with TM-1 to develop three large sized mapping populations. Phenotypic data for yield components were collected in Nanjing (JES/NAU) and Xinjiang (BES/XJ) in 2006 and 2007. Three simple sequence repeat (SSR) genetic linkage maps on chro.D8 were constructed using 907 individuals in (7TR-133 × TM-1)F2 (Pop A), 670 in (7TR-132 × TM-1)F2 (Pop B), and 940 in (7TR-214 × TM-1)F2 (Pop C). Three stable QTLs for boll size, two for lint percentage and one for boll number per plant, were detected on chro.D8 following analysis of three RIL backcrossed F2/F2:3 progeny at JES/NAU and BES/XJ although their cultivation practices differ greatly between these two cotton-growing regions. One QTL for boll number per plant exhibited a phenotypic variance (PV) of 5.6–10.1%, three QTLs for boll size exhibited 15.0–35.5% PV and two lint percentage QTLs exhibited 10.9–19.3% PV. Negative correlation between lint yield and fiber strength was confirmed.  相似文献

A high-density molecular marker linkage map of cotton based entirely on polymerase chain reaction-based markers is useful for a marker-assisted breeding program. Four kinds of markers—simple sequence repeats (SSRs), sequence-related amplified polymorphism (SRAP), random amplified polymorphic DNA (RAPD), and retrotransposon-microsatellite amplified polymorphism (REMAP)—were used to assay an F2 population from a cross between “Handan208” (Gossypium hirsutum) and “Pima90” (Gossypium barbadense). Sixty-nine F2 plants were used for map construction using 834 SSRs, 437 SRAPs, 107 RAPDs, and 16 REMAPs. Linkage analysis revealed that 1,029 loci could be mapped to 26 linkage groups that extended for 5,472.3 cM, with an average distance between 2 loci of 5.32 cM. The corresponding 69 F2:3 families were grown, arranged in two replicates, and scored for eight phenotypes. Quantitative trait loci (QTL) analysis was performed by means of composite interval mapping using WinQtlCart ver 2.0. A total of 52 distinct QTLs were detected: 4 QTLs for lint index, 8 for seed index, 11 for lint yield, 4 for seed cotton yield, 9 for number of seed per boll, 3 for fiber strength, 5 for fiber length, and 8 for micronaire value. The present map and QTL analysis may provide a useful tool for breeders to transfer desirable traits from G. barbadense to the mainly cultivated species, G. hirsutum.  相似文献

The most widely grown tetraploid Gossypium hirsutum and G. barbadense differ greatly in yield potential and fiber quality and numerous quantitative trait loci (QTLs) have been reported. However, correspondence of QTLs between experiments and populations is poor due to limited number of markers, small population size and inaccurate phenotyping. The purpose of the present study was to map QTLs for yield, yield components and fiber quality traits using testcross progenies between a large interspecific F2 population and a commercial cotton cultivar as the tester. The results were compared to these from its F2 and F2:3 progenies. Of the 177 QTLs identified from the three populations, 65 fiber QTLs and 51 yield QTLs were unique with an average of 8–12 QTLs per traits. All the 26 chromosomes carried QTLs, but differed in the number of QTLs and the number of QTLs between fiber and yield QTLs. The congruence of QTLs identified across populations was higher (20–60 %) for traits with higher heritabilities including fiber quality, seed index and lint percentage, but lower (10–25 %) for lower heritability traits-seedcotton and lint yields. Major QTLs, QTL clusters for the same traits and QTL ‘hotspots’ for different traits were also identified. This research represents the first report using a testcross population in QTL mapping in interspecific cotton crosses and provides useful information for further comparative analysis and marker-assisted selection.  相似文献

Molecular markers associated with fiber development traits have the potential to play a key role in understanding of cotton fiber development. Seventeen SSRs out of 304 markers tested from MGHES (EST-SSR), JESPR (genomic SSR), and TMB (BAC-derived SSR) collections showed significant linkage associations (using a Kurskal-Wallis non-parametric test) with lint percentage QTL in a set of recombinant inbred cotton lines (RILs) segregating for lint percentage. The permutation test of these potential markers associated with lint percentage QTL(s) determined that 12 SSR markers have stable estimates, exceeding empirically chosen threshold significance values at or above α = 0.01. Interval mapping demonstrated that 9 SSRs with stable critical LOD threshold values at α = 0.01 have significant QTL effect. Multiple QTL-mapping (MQM) revealed that at least, two highly significant fiber development QTLs exist around regions TMB0471 and MGHES–31 (explained about 23–59% of the phenotypic variation of lint percentage) and around markers MGHES–31 and TMB0366 (accounted for 5.4–12.5% phenotypic variation of lint percentage). These markers, in particular fiber-specific EST-SSRs, might be the possible ‘candidate’ loci contributing for fiber development in cotton. BAC-derived SSRs associated with fiber trait are the possible markers that are useful for the identification of physical genomic contigs that contain fiber development genes. Several lint percentage trait associated SSR markers have been located to chromosomes 12, 18, 23, and 26 using deletion analysis in aneuploid chromosome substitution lines. Outcomes of the work may prove useful in understanding and revealing the molecular basis of the fiber development, and the utilization of these markers for development of superior cotton cultivars through marker-assisted selection (MAS) programs. Plant height (PHT), stem and leaf fresh weight (SLFW), juice weight (JW) and sugar content of stem (Brix) are important traits for biofuel production in sweet Sorghum. QTL analysis of PHT, SLFW, JW and Brix was conducted with composite interval mapping using F2 and F2:3 populations derived from the cross between grain Sorghum (Shihong137) × sweet Sorghum (L-Tian). Three QTLs controlling PHT were mapped on SBI-01, SBI-07 and SBI-09 under four different environments. These QTLs could explain 10.16 to 45.29% of the phenotypic variance. Two major effect QTLs on SBI-07 and SBI-09 were consistently detected under four environments. Eight QTLs controlling SLFW were mapped across three environments and accounted for 5.49–25.36% of the phenotypic variance. One major QTL on SBI-09 located between marker Sb5-206 and SbAGE03 was observed under three environments. Four QTLs controlling Brix were identified under two environments and accounted for 11.03–17.65% of the phenotypic variance. Six QTLs controlling JW were detected under two environments, and explained 6.63–23.56% of the phenotypic variance. QTLs for JW on SBI-07 and SBI-09 were consistent in two environments showing higher environmental stability. In addition, two chromosome regions on SBI-07 and SBI-09 were identified in our study having major effect on PHT, SFLW and JW. The results would be useful for the genetic improvement of sweet Sorghum to be used for biofuel production.  相似文献

We searched for QTLs involved in tolerance to barley yellow dwarf (BYD), a serious viral disease of small grain cereals in two wheat populations, Opata × Synthetic (ITMI)and Frontana × INIA66 (F × I), for which marker data had previously been generated. The populations were evaluated in replicated field trials under artificial inoculation with a BYDV-PAV-Mex isolate and under disease-free conditions. Disease symptoms (yellowing, dwarfism and biomass reduction) were visually recorded and agronomic traits (number of tillers, height, biomass, yield and thousand-kernel weight) were measured on five plants per plot. Phenotypic data on all evaluated traits showed normal distribution with high correlation between visual estimates and measured values. Heritabilities were mostly moderate to high in the 114 lines of the ITMI population, and from low to moderate in the 117 lines of the F × I population. QTL analyses were based on genetic maps containing 443 loci for the ITMI population and 317 loci for the F × I population. Using composite interval mapping, 22 QTLs in the ITMI population and seven in the F × I population were detected, explaining9.8–43.3% of total phenotypic variation (σ2 P)per agronomic trait in the first population, and 4.1–13.7% in the second. Individual QTLs explained less than 15.8%of σ2 P. In the F × I population a minor QTL explaining 7% of σ2 P for yellowing was detected on the short arm of 7D, linked to leaf tip necrosis, a morphological marker for linked genes Bdv1, Yr18 andLr34. A QTL consistently detected for several traits on 2D in the ITMI population and on the short arm of group 6 chromosome(6S) in F × I explained 10–15% of σ2 P. The large number of QTLs having mostly small effects and the continuous distribution of all evaluated traits confirmed the polygenic nature and complexity of BYD tolerance in wheat. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

‘Conrad’, a soybean cultivar tolerant to Phytophthora root rot (PRR), and ‘OX760-6-1’, a breeding line with low tolerance to PRR, were crossed. F2 derived recombinant inbred lines were advanced to F6 to generate a population through single-seed descent. This population was used to identify quantitative trait loci (QTLs) influencing PRR tolerance in ‘Conrad’. A total of 99 simple sequence repeat (SSR), or microsatellite, markers that were polymorphic and clearly segregated in the F6 mapping population were used for QTL detection. Based on the data of PRR in the field at two planting locations, Woodslee and Weaver, for the years 2000 and 2001, one putative QTL, designated as Qsatt414-596, was detected using MapMaker/QTL. Qsatt414-596 was flanked by two SSR markers from the linkage group MLG J, Satt414 and Satt596. Satt414 and Satt596 were also detected to be significantly (P < 0.005) associated with PRR using the SAS GLM procedure and were estimated to explain 13.7% and 21.5% of the total phenotypic variance, respectively.  相似文献

The indica variety Dular has a high level of resistance to rice stripe virus (RSV). We performed quantitative trait locus (QTL) analysis for RSV resistance using 226 F2 clonal lines at the seedling stage derived from a cross between the susceptible japonica variety Balilla and the resistant indica variety Dular with two evaluation criteria, infection rate (IR) and disease rating index (DRI). The experiments were performed in both 2004 and 2005. Based on IR, three putative QTLs were detected and had consistent locations in the 2 years, one QTL was detected in the RM7324–RM3586 interval on chromosome 3. The other two QTLs were linked and located in the RM287–RM209 and RM209–RM21 intervals on the long arm of chromosome 11, and accounted for 87.8–57.8% of the total phenotypic variation in both years. Based on DRI, three putative QTLs were also detected and had consistent locations in both years. One of them was located in the RM1124–SSR20 interval on the short arm of chromosome 11, while the other two linked QTLs had the same chromosomal locations on chromosome 11 as those detected by IR, and accounted for 55.7–42.9% of total phenotypic variation in both years. In comparison to the mapping results from previous studies, one of the two linked QTLs had a chromosomal location that was similar to Stv-b i , an important RSV resistance gene, while the other appeared to be a newly reported one.  相似文献

Leaf shape is an important factor affecting canopy structure, photosynthetic characteristics, and yield. Introducing okra leaf shape is considered an important strategy in cotton heterosis breeding. The objective of the study was to evaluate the heterosis in okra leaf cotton hybrid F1s, which were developed by crossing two sterile near isolines and three restorer near isolines. A normal leaf check hybrid F1, Zhongza 29, and four newly developed okra leaf oriented hybrid F1s, including the super-okra (severely cleft and narrowly lobed leaf), okra (deeply cleft and narrowly lobed leaf), semi-okra (intermediate leaf lobe), and sub-okra (large leaf lobe) hybrids were selected in this experiment. Okra leaf oriented cotton hybrids significantly reduced leaf area index (LAI) (< 0.0001), thus more light penetrated into the lower canopy. The LAIs for the super-okra and okra hybrids, however, were too small to supply adequate photosynthate, thus resulting in early maturity, short boll filling period, and low yield. As the LAI increases, the semi-okra and sub-okra hybrids showed certain heterosis. The semi-okra hybrid showed 7–12% heterosis over the check hybrid in lint percent, 6–11% in boll weight, 2–147% in canopy light intensity, 1–10% in net photosynthetic rate (Pn), and −3 to 3% in lint yield. As well, all tested okra leaf hybrids improved fiber quality such as micronaire compared with the normal leaf check hybrid. The results suggested that adopting okra leaf parental lines in cotton breeding provided potentials in increasing fiber quality with comparable lint yield.  相似文献

A genetic linkage map with 70 loci (55 SSR, 12 AFLP and 3 morphological loci) was constructed using 117 F2 plants obtained from a cross between two upland cotton cultivars Yumian 1 and T586, which have relatively high levels of DNA marker polymorphism and differ remarkably in fiber-related traits. The linkage map comprised of 20 linkage groups, covering 525 cM with an average distance of 7.5 cM between two markers, or approximately 11.8% of the recombination length of the cotton genome. The present genetic linkage map was used to identify and map the quantitative trait loci (QTLs) affecting lint percentage and fiber quality traits in 117 F2:3 family lines. Sixteen QTLs for lint percentage and fiber quality traits were identified in six linkage groups by multiple interval mapping: four QTLs for lint percentage, two QTLs for fiber 2.5% span length, three QTLs for fiber length uniformity, three QTLs for fiber strength, two QTLs for fiber elongation and two QTLs for micronaire reading. The QTL controlling fiber-related traits were mainly additive, and meanwhile including dominant and overdominant. Several QTLs affecting different fiber-related traits were detected within the same chromosome region, suggesting that genes controlling fiber traits may be linked or the result of pleiotropy.  相似文献

A genetic linkage map of chromosome 6 was constructed by using 270 recombinant inbred lines originated from an upland cotton cross (Yumian 1 × T586) F2 population. The genetic map included one morphological (T1) and 18 SSR loci, covering 96.2 cM with an average distance of 5.34 cM between two markers. Based on composite interval mapping (CIM), QTL(s) affecting lint percentage, fiber length, fiber length uniformity, fiber strength and spiny bollworm resistance (Earias spp.) were identified in the t1 locus region on chromosome 6. The allele(s) originating from T586 of QTLs controlling lint percentage increased the trait phenotypic value while the alleles originating from Yumian 1 of QTLs affecting fiber length, fiber length uniformity, fiber strength and spiny bollworm resistance increased the trait phenotypic value.  相似文献

Summary Quantitative trait loci (QTL) analysis for Al tolerance was performed in rice using a mapping population of 98 BC1F10 lines (backcross inbred lines: BILs), derived from a cross of Al-tolerant cultivar of rice (Oryza sativa L. cv. Nipponbare) and Al-sensitive cultivar (cv. Kasalath). Three characters related to Al tolerance, including root elongation under non-stress conditions (CRE), root elongation under Al stress (SRE) and the relative root elongation (RRE) under Al stress versus non-stress conditions, were evaluated for the BILs and the parents at seedling stage. A total of seven QTLs for the three traits were identified. Among them, three putative QTLs for CRE (qCRE-6, qCRE-8 and qCRE-9) were mapped on chromosomes 6, 8 and 9, respectively. One QTL for SRE (qSRE-4) was identified on chromosome 4. Three QTLs (qRRE-5, qRRE-9 and qRRE-10) for RRE were detected on chromosomes 5, 9, 10 and accounted for 9.7–11.8% of total phenotypic variation. Interestingly, the QTL qRRE-5 appears to be syntenic with the genomic region carrying a major Al tolerance gene on chromosome 6 of maize. Another QTL, qRRE-9, appears to be similar among different rice populations, while qRRE-10 is unique in the BIL population. The common QTLs for CRE and RRE indicate that candidate genes conferring Al tolerance in the rice chromosome 9 may be associated with root growth rates. The existence of QTLs for Al tolerance was confirmed in substitution lines for corresponding chromosomal segments. These results also provide the possibilities of enhancing Al tolerance in rice through using marker-assisted selection (MAS) and pyramiding QTLs.  相似文献

The objective of this study was to examine the relationship between heterosis and genetic distance in upland cotton (Gossypium hirsutum L.). Heterosis in the different environments was evaluated and the relationship between heterosis and genetic distance (GD) was determined based on SSR markers for yield quantitative trait loci (QTL). Yields of seed cotton and lint showed a linear relationship with mid‐parent heterosis (MPH) and better‐parent heterosis (BPH). The variation in heterosis for other traits and their correlation with GD may be due to environmental factors or the effort of QTLs tested in the present study may vary in different environments. The present findings provide a foundation for heterotic grouping of parental lines and breeding of new cotton hybrids with improved seed cotton yield. This study calls for more research with stable QTLs as well as advance molecular marker techniques may be used in predicting yield heterosis in a more precise and reliable manner.  相似文献

Root traits are key components of plant adaptation to drought environment. By using a 120 recombined inbred lines (RILs) rice population derived from a cross between IRAT109, a japonica upland rice cultivar and Yuefu, a japonica lowland rice cultivar, a complete genetic linkage map with 201 molecular markers covering 1,833.8 cM was constructed and quantitative trait loci (QTLs) associated with basal root thickness (BRT) were identified. A major QTL, conferring thicker BRT, located on chromosome 4, designated brt4, explained phenotypic variance of 20.6%, was selected as target QTL to study the effects of marker-assisted selection (MAS) using two early segregating populations derived from crosses between IRAT109 and two lowland rice cultivars. The results showed that the flanking markers of brt4 were genetically stable in populations with different genetic backgrounds. In the two populations under upland conditions, the difference between the means of BRT of plants carrying positive and negative favorable alleles at brt4 flanking markers loci was significant. Phenotypic effects of BRT QTL brt4 were 5.05–8.16%. When selected plants for two generations were planted at Beijing and Hainan locations under upland conditions, MAS effects for BRT QTL brt4 were 4.56–18.56% and 15.46–26.52% respectively. The means of BRT for the homozygous plants were greater than that of heterozygous plants. This major QTL might be useful for rice drought tolerance breeding. L. Liu and P. Mu are contributed equally to this work.  相似文献

Drought tolerance was investigated in ‘C306’, one of the most drought tolerant wheat cultivars bred in India in the 1960’s. An intervarietal mapping population of recombinant inbred lines of the cross ‘C306’ × ‘HUW206’ was evaluated for drought tolerance components, namely potential quantum efficiency of photosystem (PS) II (Fv/Fm), chlorophyll content (Chl), flag leaf temperature (Lt), and grain yield per plant (Gyp) under stress. Three independent experiments were conducted under well-watered and water-stressed conditions in greenhouses and growth chambers at Kansas State University (USA). Five hundred and sixty microsatellite markers covering the entire genome were screened for polymorphism between the parents. A QTL (QLt.ksu-1D) for Lt (low flag leaf temperature under stress) on the short arm of chromosome 1D between markers Xbarc271 and Xgwm337 at LOD 3.5 explained 37% of the phenotypic variation. A QTL for Fv/Fm (QF v /F m .ksu-3B) and Chl (QChl.ksu-3B) controlling quantum efficiency of PS II and chlorophyll content under stress were co-localized on chromosome 3B in the marker interval Xbarc68–Xbarc101 and explained 35–40% of the phenotypic variation for each trait. A QTL (QGyp.ksu-4A) for Gyp on chromosome 4A at a LOD value of 3.2 explained 16.3% of the phenotypic variation. Inconsistent QTLs were observed for Fv/Fm on chromosomes 3A, 6A, 2B, 4B, and 4D; for Chl on 3A, 6A, 2B and 4B; and for Lt on 1A, 3A 6A, 3B and 5B. The identified QTLs give a first glimpse of the genetics of drought tolerance in C306 and need to be validated in field experiments using the marker-phenotype linkages reported here.  相似文献

The improvement of cotton fiber quality has become more important because of changes in spinning technology. Stable quantitative trait loci (QTLs) for fiber quality will enable molecular marker-assisted selection to improve fiber quality of future cotton cultivars. A simple sequence repeat (SSR) genetic linkage map consisting of 156 loci covering 1,024.4 cM was constructed using a series of recombinant inbred lines (RIL) developed from an F2 population of an Upland cotton (Gossypium hirsutum L.) cross 7235 × TM-1. Phenotypic data were collected at Nanjing and Guanyun County in 2002 and 2003 for 5 fiber quality and 6 yield traits. We found 25 major QTLs (LOD ≥ 3.0) and 28 putative QTLs (2.0 < LOD < 3.0) for fiber quality and yield components in two or four environments independently. Among the 25 QTLs with LOD ≥ 3, we found 4 QTLs with large effects on fiber quality and 7 QTLs with large effects on yield components. The most important chromosome D8 in the present study was densely populated with markers and QTLs, in which 36 SSR loci within a chromosomal region of 72.7 cM and 9 QTLs for 8 traits were detected.  相似文献

Preharvest sprouting (PHS) is one of the most important factors affecting the cereal production worldwide, in regions characterized by rainfall and high humidity during harvest season. It is sometimes a problem in rapeseed (Brassica napus L.), especially in production of commercial F1 hybrids. To detect quantitative trait loci (QTL) controlling PHS, a F2 population consisting of 269 F2:3 lines was created from the cross between a PHS-tolerant line (117AB) and a PHS-susceptible line (7,605). A linkage map was constructed using 35 Simple Sequence Repeat markers and 242 Amplified Fragment Length Polymorphism markers. PHS was measured as a percentage of sprouted seeds on the mother plant, 7 days after physiological maturity. Five putative QTLs for PHS were detected and located on LG2 (N11) and LG7 (N3), respectively. Phenotypic variance explained by each QTL ranged from 4.11 to 50.78% and the five putative QTLs explained about 75.63% of the total phenotypic variance. A major QTL was identified on LG2 (N11) flanked by P3C4180 and C6C13160, which explained 50.78% of the total phenotypic variance. Meanwhile, we detected four significant epistatic interactions with a total contribution of 17.16% of the total phenotypic variance.  相似文献

Reduced leaf senescence (stay-green) has been demonstrated to improve tolerance of post-flowering moisture stress in grain sorghum. A number of quantitative trait loci (QTLs) associated with stay-green have been identified in sorghum, to facilitate transfer of this trait into adapted genetic backgrounds. This study reports initial evaluations, in both well watered and post-flowering stress environments, following partial introgression (BC2F3/BC1F4 generations) of four stable stay-green QTLs (StgB, Stg1, Stg3 and Stg4) from donor parent B35 to senescent variety R 16. The majority of the introgression lines had higher leaf chlorophyll levels at flowering (a distinctive trait of the donor parent) and a greater percentage green leaf area during the latter part of grain filling, than did R 16, indicating that the stay-green QTLs were expressed phenotypically in the R 16 background. None of the QTL introgression lines achieved the same level of stay-green as B35, however. Maintenance of a greater relative green leaf area during the latter half of grain filling was related to a greater relative grain yield in two of three post-flowering moisture deficit environments in which the materials were evaluated (r 2 = 0.34 in 2004–2005 and r 2 = 0.76 in 2005–2006), as was a direct measure of leaf chlorophyll in one of the post-flowering stress environments in which this was measured (r 2 = 0.42, P < 0.05). Thus the study provided useful evidence that the marker-assisted backcross transfer of stay-green QTLs from B35 into an adapted, but senescent background has the potential to enhance tolerance of post-flowering drought stress in sorghum.  相似文献
