正红菇(Russula vinosa)为出发菌株,对其液体发酵生产胞外多糖的培养基组成(碳源、氮源和无机盐)和培养条件(温度、pH值、发酵时间和装液量)对胞外多糖和菌丝体生物量的影响进行了研究。结果表明:液体发酵培养基的最佳碳源为蔗糖,质量浓度为40g/L;最佳氮源为酵母膏,质量浓度为9g/L;添加无机盐组分为KH2PO42g/L和MgSO41g/L;适宜的发酵条件为初始pH为5.5-6.5,发酵温度为28-30℃,250ml三角瓶装液量为50-60ml,发酵时间为5d。通过优化培养基和发酵条件,胞外多糖产量达到4.96 g/L和菌丝体生物量达到22.34 g/L。 相似文献

对高毒BT菌株——CF1263进行液体振荡培养和正交试验结果表明:依据碳源、氮源和无机盐对芽孢产量的影响,对液体培养基进行成分优化组合,在碳源和氮源中分别筛选出最佳因子,淀粉、蛋白胨、牛肉膏、麦麸、酵母粉,从无机盐中筛选出KH2PO4和MgSO4.7H2O。运用以上7个因素进行L18(37)因素3水平的正交试验,从而得出CF1263最优配方为1.5%淀粉、2%蛋白胨、0.2%牛肉膏、0.3%麦麸、2.0%酵母粉、0.1%KH2PO4和0.2%Mg-SO4.7H2O。 相似文献

为给天麻的共生蜜环菌(Armillaria mellea)菌株MC4筛选最佳的母种培养基和栽培种培养基,将菌株MC4接种到不同的培养基上,测定MC4在不同培养基上的生长速度、生物量,并观察培养特性,结果表明:在以马铃薯淀粉为碳源、以酵母粉为氮源、以KH2PO4为无机盐的母种培养基上,菌株MC4的生长速度及生物量显著较其它培养基上的高(P<0.05);在以栎木枝条和马铃薯汁制成的Ⅳ号栽培种培养基上,菌株MC4菌丝萌发所需时间较短,生长速度快,分枝茂密,且菌索粗壮,直径达0.45cm。天麻共生蜜环菌菌株MC4最佳的母种培养基应以马铃薯淀粉为碳源、酵母粉为氮源、KH2PO4为无机盐,而以栎木枝条和马铃薯汁配制的Ⅳ号培养基更适合作为MC4的栽培种培养基。 相似文献

桑树轮纹病病原菌的分离鉴定及其生物学特性研究 总被引:1,自引:0,他引:1
2012年6~8月,通过对桑树轮纹病病原菌分离、田间致病性测定、病原菌形态观察,将该病原菌鉴定为桑膝节霉,并对此病病原菌的生物学特性进行了研究。结果表明,不同培养基配方、温度、酸碱度、碳源、氮源、光照对桑树轮纹病病原菌菌丝的生长有一定影响,菌丝生长的最适宜培养基是理查培养基,最适温度是25℃,最适pH值是7.0,最适碳源为葡萄糖,最适氮源为蛋白胨,光照对菌丝生长无显著性差异,菌丝的致死温度和时间分别为85℃和10 min。对该病病原菌生物学特性研究为云南首次报道,对桑树轮纹病的防治具有一定的参考价值。 相似文献

[目的]对野生食用菌进行分子鉴定及生物学特性研究,为其进一步液体深层发酵培养和开发利用奠定基础。[方法]采用rDNA ITS序列分析方法对一野生菌进行分子鉴定,通过单因素和正交试验确定了该菌株的生物学特性。[结果]由新鲜子实体分离得到的纯菌株为Paxillus ammoniavirescens,其菌丝体生长的最适碳源和氮源分别为可溶性淀粉和蛋白胨,最适培养温度为25℃,最适pH为4.5。[结论]卷边网褶菌菌丝体液体培养的最佳配方为:可溶性淀粉20 g,蛋白胨2 g,KH_2PO_4 3.0 g,MgSO_4 4.5 g,水1 L。按此配方培养,可获得平均干质量为80 mg的菌丝体,明显高于其他配方。 相似文献

通过P.t.优良菌株9109在不同培养基上生长速度、培养特性及其生长规律的研究,筛选出生长最佳培养基作为基础培养基,进行C、N营养研究,试验结果表明:P.t.9109不仅能利用单糖、低聚糖和无机N等速效C、N源,而且能利用多糖、多元醇和有机N等长效C、N源。其菌丝生长最适C源是玉米粉和甘露糖;最适N源是蛋白胨和氯化铵。在无机N利用中,对硝态N和铵态N的利用无显著差异。应用L16(45)正交设计确定了P.t.9109菌丝生长的最佳营养配方,菌丝增殖高达860倍,不但提高菌丝生物产量,而且节省了原料,降低了成本,为改进P.t.菌剂生产配方和研制新型菌剂提供了最佳营养条件。 相似文献

This work reports results of mycelial yield of Trichoderma viride from a number of carbon and nitrogen sources and different C:N ratios. The ratios were in two series: first by increasing the carbon content (d-glucose as standard), second – by decreasing the nitrogen content (1-as-paragine as standard). The best mycelium production was found on d-xylose and glucose as C sources and KNO3 and 1-asparagine as N sources. Increase in dosage of both sources caused increase in yield but different changes in pH, according to how nutrients were utilised. 相似文献

对分别生长于大兴安岭北坡、南坡的2种灵芝(LZ01、LZ02)的生物学特性进行了研究,通过平板培养方式观测不同温度、pH值、氮源、碳源及不同碳氮比对2种菌种生长速度的影响。结果表明:2种灵芝菌丝适宜生长温度范围为24~28℃,其中LZ01生长于背阴面,最适生长温度在28℃左右;LZ02生长于向阳面,最适生长温度为24℃。2种灵芝在偏酸性环境下生长较好。2种灵芝菌种既能利用有机氮源,也能利用无机氮源,但是有机氮源对其生长较为有利。可溶性淀粉为LZ01适宜生长的碳源,葡萄糖为LZ02适宜生长的碳源,5种碳氮比中比值为40∶1时,菌丝生长速度最快。 相似文献

Effect of different nitrogen and carbon sources and their concentrations in liquid media on the mycelial growth of six different ectomycorrhizal (ECM) fungal species was studied. Differences were found in the utilization of the different nitrogen and carbon sources between the fungal species. All the species showed better mycelial growth in the medium containing ammonium as nitrogen source. Growth was low in all species in medium in which nitrogen was supplied in nitrate form. All the ECM isolates investigated showed reduced growth in the medium containing maleic acid as the carbon source. The effect of glucose and di-ammonium hydrogen orthophosphate concentrations on mycelia growth of all the six fungal species was studied with ranges for glucose of 2–40 g/l, and for di-ammonium hydrogen orthophosphate of 2–20 g/l. Cortinarius fulvoconicus and Cortinarius flexipes showed maximal mycelial growth at a glucose concentration greater than 20 g/l. Suillus luteus, Scleroderma citrinum, Laccaria laccata, and Tricholoma aurantium showed maximal growth at a glucose concentration of 20 g/l. All six species showed maximal mycelial growth at 5–10 g/l of di-ammonium hydrogen orthophosphate concentration and with increased concentration mycelial growth in all species decreased. 相似文献

为提高微生物油脂产量,以产油微生物深黄被孢霉为试验菌株,采用单因素试验设计,通过摇瓶培养,研究了产脂培养基中碳源、氮源种类及其浓度、接种量、初始pH值、无机盐离子对菌体生长和油脂积累的影响,确定了深黄被孢霉摇瓶发酵产油脂的优化培养条件为:葡萄糖100 g.L-1,酵母粉3.0 g.L-1,接种量为20%,pH值为5.0~6.0,硫酸镁(MgSO4.7H2O)0.5 g.L-1,磷酸二氢钾(KH2PO4)2.0 g.L-1。在优化培养条件下菌体生物量为20.62 g.L-1,油脂含量为43.02%,油脂产量为8.87 g.L-1。 相似文献

Isolates of Cylindrocladium quinqueseptatum (CQ), which cause leaf blight of Eucalyptus in Kerala, differed significantly in growth and cultural characters on nine different agar media. Malt extract agar and yeast malt agar and yeast malt agar were the best media for discerning differences among the CQ isolates. Of the 11 carbon (C) and 13 nitrogen (N) sources evaluated in liquid media for their differential utilization by the five selected CQ isolates, five each of C and N sources differentiated them from each other. In general, organic N sources supported better growth and micro-sclerotia production over inorganic, and hexose and disaccharide over trisaccharide and polysaccharide. 相似文献
