A systematic review and meta-analysis was conducted to identify study-level variables that could explain the variation in apparent Salmonella spp. prevalence estimates. Electronic and non-electronic literature searches from 1990 until 2005 were carried out to identify all studies related to the prevalence of subclinical Salmonella infection in swine. The searches were restricted to studies published in English, Spanish, and French. Clinical trials or any other study where an intervention was evaluated were excluded from this analysis. A template was designed to retrieve the most relevant variables and data abstraction was performed in duplicate. A total of 98 papers containing 82 animal-level and 156 farm-level studies were used in the analyses. The median farm-level and animal-level prevalences were 59% and 17%, respectively. Meta-regression analyses were carried out on both farm and animal-level data. Diagnostic procedure, sample size, and country where study was conducted were the three most important predictors in explaining the differences in Salmonella prevalences between studies. When compared to a farm with a apparent prevalence of 50% determined by the blood ELISA, prevalences based on culture of fecal samples were 39% lower and prevalences based on cecum and tissue cultures were 16% and 19% lower, respectively. Similar to farm-level models, animal-level models did not show any difference among serological tests and prevalence values based culture procedures were, on average, 9% lower than those from serological tests. Sample size was negatively associated with prevalence estimates. In conclusion, the methodology was useful for identifying and quantifying sources of variation in Salmonella apparent prevalence among studies and for establishing prevalence distributions that could be used as input parameters in risk assessment and decision models. The analysis provides some guidelines when interpreting and comparing apparent Salmonella prevalence results from studies using different study designs.  相似文献

We reviewed Bayesian approaches for animal-level and herd-level prevalence estimation based on cross-sectional sampling designs and demonstrated fitting of these models using the WinBUGS software. We considered estimation of infection prevalence based on use of a single diagnostic test applied to a single herd with binomial and hypergeometric sampling. We then considered multiple herds under binomial sampling with the primary goal of estimating the prevalence distribution and the proportion of infected herds. A new model is presented that can be used to estimate the herd-level prevalence in a region, including the posterior probability that all herds are non-infected. Using this model, inferences for the distribution of prevalences, mean prevalence in the region, and predicted prevalence of herds in the region (including the predicted probability of zero prevalence) are also available. In the models presented, both animal- and herd-level prevalences are modeled as mixture distributions to allow for zero infection prevalences. (If mixture models for the prevalences were not used, prevalence estimates might be artificially inflated, especially in herds and regions with low or zero prevalence.) Finally, we considered estimation of animal-level prevalence based on pooled samples.  相似文献

We review frequentist and Bayesian approaches for estimating animal-level disease prevalence using pooled samples obtained by simple random sampling. We determine the preferred approach for different prevalence scenarios and with varying knowledge about sensitivity and specificity values. When sensitivity and specificity are perfect or known, we can choose between the large-sample theory estimates and the one-to-one relationship exact estimates. When sensitivity and specificity are unknown, we must use large-sample theory estimates or Bayesian methodology (which gives exact estimates). However, when the large-sample theory produces a negative lower confidence limit, we must use one of the exact methods. We compare estimates from each approach using culture results from pools of 20 eggs from three flocks on a California ranch that were producing eggs that were contaminated with Salmonella enteritidis phage type 4.  相似文献

A 1-year study on the animal-level prevalence and concentration of Escherichia coli O157 in adult sheep at slaughter was performed, to collect qualitative and quantitative information on the diffusion of the pathogen in adult sheep from Italy. A total 533 samples were collected, with a similar distribution in the four seasons. For prevalence estimates, a simple random sampling technique was used. An immuno-magnetic separation technique was used for sample screening, with enumeration of the pathogen in positive samples, along with molecular and serological identification of isolates. An overall prevalence of 7.1% (38/ 533, 95% CI 4.9-9.3%) was observed for fully virulent E. coli O157. A wide interval of VTEC O157 per gram was observed (< 100 to 6 x 10(5) CFU g(-1)), with 28.9% (11/38) of positive samples > or = 1 x 10(3) CFU g(-1), set as the threshold for those animals defined 'active shedders' for the purpose of the study. Eight per cent (3/38) of animals shed > 1 x 10(4) g(-1) VTEC O157, which represents > 96% of the total VTEC O157 bacteria cultured from all animals tested. The prevalence estimate of active shedders was therefore 2.1% (95% CI 0.9-3.3%). Most (34/38, 89.5%) of the positive animals were found in summer (July-September). Prevalence and concentrations of virulent VTEC O157 obtained in this study contribute to the demonstration that adult sheep represent a relevant source of environmental contamination from virulent VTEC O157, as well as a source of VTEC O157 contamination for food of ovine origin (meat and dairy products), especially during warm months.  相似文献

Giardia duodenalis and Cryptosporidium spp. are intestinal protozoan parasites that infect a wide range of host species, including humans. Molecular characterization of these parasites has demonstrated that a number of genotypes and species are common to both humans and animals, and that zoonotic transmission may occur. Numerous studies have reported a high prevalence of G. duodenalis and Cryptosporidium spp. in cattle, particularly calves, and these animals are frequently associated with zoonotic transmission. In the present study, a total of 143 faecal samples from adults, heifers and calves were collected from two dairy cattle farms in eastern Ontario, Canada. The prevalence and molecular characteristics of G. duodenalis and Cryptosporidium spp. in these animals were determined in order to investigate the potential for transmission between cattle and humans in this region. Following DNA extractions from faecal samples, nested-PCR protocols were used to amplify fragments of the 16S rRNA gene and the heat-shock protein 70 (HSP-70) gene for determining the prevalence of G. duodenalis and Cryptosporidium spp. infections, respectively. Genotypes of G. duodenalis, and species of Cryptosporidium, were determined by means of DNA sequencing of amplicons, and subsequent sequence alignment. Cattle on both farms showed a high prevalence of G. duodenalis (42.0%) and Cryptosporidium spp. (27.3%). G. duodenalis infections were more prevalent in calves and heifers than in adults, and Cryptosporidium spp. infections were only observed in calves and heifers. The zoonotic genotype, G. duodenalis Assemblage B was isolated from 24.5% of the cattle tested, while G. duodenalis Assemblage E was found in 17.5% of the cattle tested. The overall prevalence of the zoonotic species Cryptosporidium parvum in the animals tested was found to be 21.7%, while only 1.4% were infected with C. bovis. These findings suggest that there is a potential risk of zoonotic and/or zooanthroponotic transmission of G. duodenalis and C. parvum infections between cattle and humans in eastern Ontario, likely by means of contaminated water or food, or through direct faecal-oral transmission in the case of farmers and veterinary staff.  相似文献

We used Monte Carlo simulation to estimate distributions for flock-level sensitivity of abattoir-based surveillance for ovine paratuberculosis as currently practised in New South Wales, Australia. Probability distributions were used as input variables for within-flock prevalence, years-infected and individual animal-level sensitivity and specificity of gross pathology as a screening test for the presence of paratuberculosis. Distributions used as inputs for the size of abattoir-slaughter groups were based on existing abattoir-surveillance data from NSW. Predicted flock-level sensitivity depended on within-flock prevalence and the number of animals examined and was sensitive to estimates of animal-level sensitivity and specificity. The median probability of detection of an infected flock based on the examination of one abattoir line was predicted not to exceed 0.95 unless the within-flock prevalence was ≥7%. If the within-flock prevalence was 2%, the probability distribution of flock-level sensitivity had a median of 0.73, with 80% of values lying between 0.55 and 0.84. Improvement in the flock-level sensitivity could be achieved by submitting more than three gross pathology-positive specimens per line, if available—but the degree of improvement depended on the number of sheep slaughtered (line size) and the within-flock prevalence. At 2% prevalence, a median flock-level sensitivity of 0.95 could be obtained in lines of >390 sheep if six gross pathology-positive specimens were submitted. We concluded that abattoir surveillance based on identification of gross pathology as a screening test is not a sensitive tool for detecting recently infected flocks or flocks which have a moderate or lower prevalence of infected animals. But—with relatively minor modifications of the protocol currently in use—it could become a key component of a surveillance programme which included additional testing strategies for small flocks.  相似文献

Our objectives were to estimate the seroprevalence of Brucella melitensis, and to identify some risk factors associated with goat seropositivity in Michoacan, Mexico. Blood samples were collected from 5114 animals from 79 herds. Sera were tested for antibodies against B. melitensis using the Rose Bengal plate test and the complement-fixation test. Information regarding the herds and each animal sampled were recorded through a personal interview at the farm. We used random-effects multivariable logistic regression to analyze our data. Fifty-six herds of the 79 tested had at least one seropositive animal. The animal-level true seroprevalence was 9.8% (CI = 8.8, 10.7). Animals in large herds (>34 animals), in herds with high stock density (>3.5 animals/m2) or animals >24 months old had higher odds of seropositivity (2.0, 1.7 and 1.8, respectively) than those in small herds, in herds with low stock density or animals ≤24 months old.  相似文献

We collected blood samples from 756 > or =2-year-old cattle in 54 herds in Yucatan, Mexico, and used all of those to determine the antibody seroprevalences (in an indirect enzyme-linked inmunosorbance assay) to bovine respiratory syncytial virus (BRSV) and risk factors for animal-level seropositivity. We used 728 of the same samples (from 52 of the same herds) to do the same for parainfluenza virus-3 (PIV3). Cattle were selected by two-stage cluster sampling. Herd-level and animal-level risk factors were obtained through a personal interview. We analyzed the data by using a random-effects multivariable logistic regression model for clustered observations. All herds had at least 3 (BRSV) or 5 (PIV3) seropositive animals. The animal-level true seroprevalences were: 90.8% (86.5, 95.2%) and 85.6% (80.9, 90.4%) for BRSV and PIV3, respectively. Animals in large herds and old animals had the highest odds of being seropositives to BRSV, and those risk factors plus animals born on the farm for PIV3 infection.  相似文献

Prevalences of Cryptosporidium spp. oocysts in cattle (n = 486) on five selected farms in Morogoro municipality and three species of herbivorous wildlife (n = 87) from Mikumi National Park, Morogoro, Tanzania, were determined using the modified Ziehl-Neelsen staining technique. Of 486 bovine faecal samples, 5.3% were positive for Cryptosporidium spp. The prevalence of Cryptosporidium was higher in calves less than 3 months of age compared to weaned calves and adults. Cryptosporidium spp. oocysts were detected in both diarrhoeic and non-diarrhoeic animals, but there was a significantly higher prevalence (P < 0.001) of oocyst shedding in diarrhoeic than in non-diarrhoeic animals. Of the 87 faecal specimens from the wildlife species, 36 were from the African buffaloes (Syncerus caffer), 25 from zebra (Equus zebra) and 26 from the wildebeest (Connochaetes gnou). Cryptosporidium spp. oocysts were detected in eight (22%) buffaloes, seven (28%) zebras and seven (27%) wildebeests. Confirmation of the diagnosis was performed using anti-Cryptosporidium monoclonal antibody specific for Cryptosporidium muris, Cryptosporidium parvum and Cryptosporidium baileyi (Pathasure Cryptosporidium test kit).  相似文献

A BVD control programme based on the identification and removal of persistently infected (PI) animals is being undertaken in an area in the Rome province, where BVD outbreaks had been previously detected. It involves 174 mainly dairy herds, from which blood samples of all bovines older than 1 year are obtained through the national brucellosis and leukosis eradication programme. Samples sufficient to detect the presence of seropositive animals at a prevalence of 5% or more are initially screened for antibodies against BVD virus (BVDV) using an immunoenzymatic assay. Upon identification of seroreagents additional blood samples are tested from the 6-12-month age category not included in the initial samples. Animals are considered immunotolerant if BVDV is demonstrated twice at a minimum 30-day interval. When no seropositive animals are detected during the first serological screening the herd is declared BVD-free if a second testing, preferably carried on the same animals previously tested, confirms the seronegative status of the herd. At present 147 farms have been tested, of which 63 (42.9%) are negative with respect to antibodies against BVDV. Of the 84 remaining herds in which one or more seropositives are detected, 13 are classified as recently infected. In eight of these recently infected herds, 22 PI animals have been identified.  相似文献

A cross-sectional study was conducted to identify risk factors for herd infection by Brucella spp. in dairy cattle in the suburbs of Asmara, Eritrea. Data were collected from 64 herds, randomly selected from a total of 99 herds with a minimum herd size of 9 cows. A questionnaire was used to gather data on management, hygiene and herd structure. Serum samples collected from all pregnant heifers, cows and bulls, were screened for Brucella infection by the Rose Bengal test (RBT), and all RBT-positive sera re-tested with the complement-fixation test (CFT) for confirmation. A seropositive herd was defined as one in which at least one animal tested positive in the CFT. There were 23 (36%) positive herds among the 64 studied. Both multiple logistic and multiple betabinomial regression modeling were used to analyze the data. Mixed-breed herds, compared to single (exotic)-breed herds, were found to be independently associated with increased herd seroprevalence (OR=5.2, 95% confidence interval 1.4–18.7) in the multiple logistic model with the herd infection status as the dependent variable. The importance of this variable was supported by the multiple betabinomial regression model (OR=3.3, 1.4–7.6) with animal-level prevalence within herd as the outcome variable. Both models also revealed the presence of a negative association between seropositivity and cattle stocking density.  相似文献

Although frequentist approaches to prevalence estimation are simple to apply, there are circumstances where it is difficult to satisfy assumptions of asymptotic normality and nonsensical point estimates (greater than 1 or less than 0) may result. This is particularly true when sample sizes are small, test prevalences are low and imperfect sensitivity and specificity of diagnostic tests need to be incorporated into calculations of true prevalence. Bayesian approaches offer several advantages including direct computation of range-respecting interval estimates (e.g. intervals between 0 and 1 for prevalence) without the requirement of transformations or large-sample approximations, direct probabilistic interpretation, and the flexibility to model in a straightforward manner the probability of zero prevalence. In this review, we present frequentist and Bayesian methods for animal- and herd-level true prevalence estimation based on individual and pooled samples. We provide statistical methods for detecting differences between population prevalence and frequentist methods for sample size and power calculations. All examples are motivated using Mycobacterium avium subspecies paratuberculosis infection and we provide WinBUGS code for all examples of Bayesian estimation.  相似文献

A cross-sectional study was carried out to assess the prevalence and circulation of bluetongue virus (BTV) in Spanish ibexes (Capra pyrenaica hispanica). A total of 770 sera samples, 380 blood samples and 34 spleen samples were collected between 2006 and 2009 in Andalusia (southern Spain), a region and time period with a wide circulation of BTV in livestock. Thirty-one out of 770 (4.0%; CI(95%): 2.6-5.4) sera samples analyzed by ELISA showed antibodies against BTV. Twenty-four out of 31 seropositive samples were tested against BTV serotypes 1, 4 and 8 by serum neutralization test (SNT). Neutralizing antibodies against BTV-1 and BTV-4 were detected in seven and ten animals, respectively, four of them showed neutralizing antibodies to both serotypes. The animals seropositive to BTV-4 were sampled between 2006 and 2008, while BTV-1 circulation was confirmed in ibexes sampled between 2007 and 2009. None of the ibexes presented neutralizing antibodies against BTV-8. Statistically significant differences were found among regions and years, which is in coincidence with what occurred in domestic ruminants. There were no statistically significant differences between sexes, age classes and habitats (captivity vs. free-living). BTV RNA was not found in any of the 380 blood samples analyzed. However, BTV-1 RNA was detected from spleen in one Spanish ibex from Málaga province in August 2008. This finding evidences the presence of BTV-1 in Spanish ibex in a municipality where BT outbreaks were not detected in domestic ruminants during that period. Results of the present study show that Spanish ibexes were exposed and responded serologically to both BTV-1 and BTV-4. The low seroprevalence obtained suggests that Spanish ibex is not a relevant species in the dissemination of BT. However, the detection of BTV-1 RNA and the presence of seropositive ibexes in areas where BT outbreaks were not detected in livestock, could not exclude a significant role in the epidemiology of BTV in certain areas.  相似文献

The present paper surveys the number of Salmonella isolations in animals and feedstuffs in Sweden during 1988–1992. It is the eighth in a series of reports published by the National Veterinary Institute (NVI) since 1949. During the period referred to, 602 outbreaks of Salmonella were reported in animals, both domestic and wild. Compared with the previous 5-year period there was a 20% reduction in the number of outbreaks (760). Fifty-six different serotypes were reported, 19 of which had never been isolated in any animal in Sweden previously. A temporary increase in the number of outbreaks in poultry was seen in 1991 following an extended sampling before slaughter of layers. A remarkably high prevalence (38%) of Salmonella was observed in snakes in the wild.In 1990, the end-point testing of feeds was replaced by an approach based on HACCP (Hazard Analysis Critical Control Point) principles for the monitoring of feed mills. Significantly higher number of Salmonella positive samples were found by using this technique compared with the previous analysis of finished feed.It is concluded that the adopted Salmonella control program has contributed to a reduced number of Salmonella outbreaks in animals in Sweden.  相似文献

Sera from 456 wild rabbits (Oryctolagus cuniculus) collected between 1992 and 2003 from five geographical regions of Spain were examined for antibodies to Toxoplasma gondii by the modified agglutination test. Antibodies to T. gondii were found in 65 (14.2%) wild rabbits. Prevalence of infection was significantly higher in samples collected from wild rabbits from Catalonia, northeast Spain (53.8%), where rabbits lived in forest, compared to other areas (Huelva and Cádiz, Andalucía, south Spain; Toledo, Castilla-La Mancha, central Spain; and Zaragoza, Aragón, northeast Spain) with more dry conditions, where prevalence ranged from 6.1 to 14.6%. No differences were observed on prevalence and age (young animals <7 months of age compared to older animals), sex, date of samples collection or season of samples collection. The results indicate that prevalence of T. gondii in some areas of Spain is high, and this finding could have environmental and/or public health implications if wild rabbits are to be used as a source of food.  相似文献

Directional freezing is based on a simple thermodynamic principle where ice crystals are precisely controlled through the sample by regulating the velocity of the sample movement through the predetermined temperature gradient. Directional freezing permits a precise and uniform cooling rate in both small and large volume samples. Directional freezing was used for slow and rapid freezing, as well as for vitrification of oocytes and embryos using the minimum drop size technique. Sperm samples from a wide range of domestic and wild animals were successfully cryopreserved using the directional freezing method. The method enabled, for the first time, successful freezing of a whole ovary and freeze-drying of mammalian cells followed by thawing and transplantation and rehydration, respectively.  相似文献

This study reports a prevalence and risk factor survey of brucellosis in small ruminants in Southern Zone of Tigray Region, Northern Ethiopia between October 2011 and April 2012 to determine the sero-prevalence of small-ruminant brucellosis and to identify associated risk factors for the occurrence of disease in small ruminants under extensive production system. Multistage random sampling was followed to select locations, flocks, and individual animals. Laboratory analysis of serum samples provided sero-prevalence estimates for flocks and geographic location. Information on risk factors at the individual and flock level was obtained by examination of individual animal and a questionnaire interview to flock owners. The overall individual animal-level sero-prevalence of brucellosis in small ruminants was 3.5 % and flock level sero-prevalence was 28.3 %, and the within-flock sero-prevalence was ranged from 0 % to 22.2 % based on the Complement Fixation Test. Multivariable logistic regression showed that the major risk factors for flock level sero-positivity were flock size and abortion history. This study showed that small-ruminant brucellosis is prevalent in the study area. Larger flock size and history of previous abortion in the flock were major risk factors identified for sero-positivity of small-ruminant brucellosis.  相似文献

Even with advancements in pre- and post-harvest food safety, Shiga toxin-producing Escherichia coli (STEC) still present challenges to human health. Since cattle are the primary reservoir for STEC, lowering the prevalence of this pathogen in farm animals may reduce STEC outbreaks in humans. However, because many of the factors that modulate the colonization and persistence of STEC in cattle remain unknown, reducing STEC in this host is challenging. In this study, we evaluated a cohort of beef cattle one to eleven years of age to determine the effect of animal age on the prevalence of STEC. During the first year of sample collection, heifers had significantly lower STEC prevalence than cows (37.5% vs. 70%). In the second year of sample collection, STEC prevalence peaked in cows that were two years of age and tended to decrease as animals became older. In addition, by studying a subset of the animals in both years, we observed an increase in STEC prevalence from 40.6% to 53.1% in heifers, whereas cows had a net decrease in STEC prevalence from 71.4% to 61.9%. The results from this study indicate that animal age is a significant factor that influences the prevalence of STEC in cattle. These findings have implications for the development of on-farm mitigation strategies by targeting animals with the highest risk of shedding; it could be possible to reduce pathogen transmission among cattle and prevent zoonotic or foodborne transmission to humans.  相似文献

Cross sectional serological surveys were conducted between March and December 2009 to determine the distribution of foot-and-mouth disease and also to validate the current passive surveillance system in Bhutan. A total of 1909 sera collected from cattle, goats, sheep, and pigs, from 485 herds in 106 villages, were tested using a foot-and-mouth disease non-structural protein 3ABC ELISA. The true prevalence at the animal-level for all species was 15% (95% CI: 13.5, 16.7) using the sensitivity (97.2%) and specificity (99.5%) for cattle. The true prevalence for cattle, goats, sheep and pigs were 17.6 (95% CI: 15.6, 19.5), 11.9% (95% CI: 5.6, 18.3), 11.9% (95% CI: 1.3, 25.1), and 1.9% (95% CI: 0.0, 3.8), respectively. The sub-districts that shared border with India had significantly (p=0.03) higher seroprevalence than the interior sub-districts. Villages located in the sub-tropical zone had significantly (p<0.0001) higher seroprevalence than those located at high altitude zones. Herds with known outbreaks of FMD were 3.6 times more likely (p<0.001) to be seropositive than those with no history of outbreaks of FMD. The study showed the usefulness of population-based serological surveys in detecting circulation of active infection in populations which were, until now, considered to be free of disease based on a passive surveillance system. The study also highlighted the benefits of conducting serological and questionnaire surveys, simultaneously, to ascertain the infection status of herds and animals. Some of the findings from this study could be considered for strengthening of the current FMD control program in Bhutan.  相似文献

Goat breeding in the Northeast region of Brazil plays an important socioeconomic role. However, there are significant losses caused by sanitary deficits and infectious diseases, particularly caseous lymphadenitis (CL). Although CL is considered endemic in Northeastern Brazil, a comprehensive and up-to-date study of this disease in goat herds in this region is necessary. The objective of this study was to determine the farm-level and animal-level seroprevalences for the disease and to identify the possible risk factors that characterize CL in the caprine species of five Northeastern’s states (Ceará, Piauí, Rio Grande do Norte, Paraíba, and Sergipe). A total of 2744 goat serum samples from 230 farms were collected between 2010 and 2012. The diagnosis of Corynebacterium pseudotuberculosis infection was performed using the indirect ELISA technique. Farm-level and animal-level seroprevalences were 87.8% and 30.3%, respectively, suggesting that C. pseudotuberculosis is widespread in goat herds of the Northeast region. The risk factors were as follows: absence of forage silage (odds ratio?=?5.39), not separating animals by sex (odds ratio?=?4.16) or by age (odds ratio?=?6.30), not replacing old goat breeders (odds ratio?=?7.80), and non-treatment of CL lumps prior to spontaneous rupture (odds ratio?=?10.34). This study supports the idea that caseous lymphadenitis is widely disseminated in goats from Northeastern Brazil and based on the risk factor analysis attention should be given to the need to establish adequate control measures, such as incision and early drainage of superficial abscesses, quarantine and elimination of affected animals, periodic inspection of the herd, non-introduction of infected animals, and early disposal of animals with recurrent CL.
