Fifteen cows with bovine spongiform encephalopathy (BSE) and 90 healthy cows were given xylazine intramuscularly at a dosage of 0.15 mg/kg bodyweight. The onset of sedation and of drooling was recorded, and the heart and respiratory rates and the systolic and diastolic blood pressure were measured every five minutes for 40 minutes. All the healthy cows but only five of the 15 cows with BSE became sedated, and the period between the administration of xylazine and the onset of sedation was twice as long in the cows with BSE than in the healthy cows (15.0 [7.5] and 7.6 [2.6] minutes). Throughout the observation period, the blood pressure of the cows with BSE was significantly higher than that of the healthy cows, and the blood pressure of the healthy cows, but not of the cows with BSE, decreased significantly towards the end of the observation period.  相似文献

During the preclinical phase of bovine spongiform encephalopathy (BSE), significantly increased concentrations of lactic acid were measured in the blood of infected dairy cows. Other plasma metabolites, including alanine, leucine, serine, and glutamic acid, also showed significantly altered concentrations in the preclinical BSE animals compared to a control group. This appears consistent with the exaggerated stress response observed in clinical BSE, and precedes the development of clinical signs and overt behavioural changes. A number of other plasma metabolites including other amino acids and components of the plasma fatty acid content showed no association with BSE status.  相似文献

Clinical signs associated with bovine spongiform encephalopathy (BSE) were studied in 1008 casualty slaughter cattle over 30 months of age to compare the results with the BSE status as determined by postmortem tests. The clinical BSE status was assessed using seven different criteria based on various publications. Only one (0.10%) out of 997 casualty slaughter cattle with a matching postmortem test result was positive for BSE. The BSE case was identified by only two case definitions tailored specifically to recumbent cases. The variety and often equivocal definition of clinical signs associated with BSE is reflected by the difference in the criteria that usually identified different animals as BSE suspects. The BSE status may be more difficult to assess in recumbent animals that do not allow a full clinical examination, and BSE may not be suspected if another disease is present that may mask signs of BSE.  相似文献

As part of a nutritional study lasting from six weeks before calving to 22 weeks of lactation, blood samples collected from 47 dairy cows maintained under well-defined conditions were analysed for a variety of metabolites and hormones. At various times after the completion of the study, six of the animals developed clinical signs of bovine spongiform encephalopathy (BSE), although when they were sampled it was not known that they were incubating the disease. The data were used to make comparisons between the animals that developed BSE and those that did not develop the disease and which had been maintained under the same conditions. The greatest differences between the animals incubating BSE and the control animals were observed at times of nutritional stress, at the start of lactation and when the intake of concentrate feeds was reduced at week 13 of lactation. In the animals that subsequently developed BSE, feed intakes were lower in early lactation; plasma beta-hydroxbutyrate concentrations were significantly higher (P<0.001) at weeks 3 and 5 of lactation; adjusted milk yields were lower until week 6 of lactation and milk fat concentrations were consistently lower. There was no effect on plasma glucose concentrations, although insulin concentrations were significantly lower in week 1 of lactation (2-27 v 2.50 microiu/ml) (P<0.05). The concentrations of plasma proteins and urea were unaffected by BSE incubation, apart from protein concentrations being significantly higher one week before calving, and the concentration of urea being significantly lower five weeks before calving. The plasma concentrations of somatotrophin, prolactin, oestradiol and progesterone were similar in the two groups of animals throughout the study. The differences observed indicate that the energy metabolism of dairy cows incubating BSE may be subtly altered before the onset of clinical signs of the disease.  相似文献

This study has described a method for generating the probability of a bovine spongiform encephalopathy (BSE) suspect being a true positive BSE case. A weighting equivalent to the clinical sign frequencies recorded in histopathologically confirmed BSE cases was assigned to 14 clinical observations and a clinical profile score for the case was generated by the summation of the weightings. This method was applied to 50 histopathologically confirmed (true positive cases) BSE suspects and 50 histopathologically unconfirmed (false positive cases) BSE suspects. The profile scores for the true positive BSE suspect cases were statistically significantly higher than the profile scores of the false positive BSE suspect cases (P = 0.0014) using a Mann-Whitney U non-parametric test. The mean profile score for the true positive cases was 944 and for the false positive scores was 879. Likelihood ratios for BSE suspects with different clinical profile scores were computed using different clinical profile score cut off point and ranges. A BSE suspect with profile score of 727 or above (the lowest cut off point) and 1037 or above (the highest cut off point) had likelihood ratios for being BSE of 1. 09 and 4.00 respectively. The likelihood ratios for a BSE suspect being a true positive BSE case in the profile score ranges 627-866, 867-966, 967-1036 and 1037-1067 were 0.35, 1.06, 1.30 and 4.0 respectively. Further investigations or a revisit may be justified in animals with a low probability of being BSE.  相似文献

The Prionics-Check PrioSTRIP is a rapid chromatographic immunoassay for bovine spongiform encephalopathy (BSE) approved by the European Union in 2004. In this study, the PrioSTRIP was used to analyse PrP(BSE) in 16 different brain areas of nine confirmed BSE cases. The levels of PrP(BSE) in the different brain areas were plotted to give the brain PrP(BSE) distribution curve (BPDC) and compared with the BPDC obtained previously by Western blotting and enzyme-linked immunosorbent assay (ELISA) methods on the same samples. The distribution of PrP(BSE) in different areas of the brain was similar, irrespective of the test applied, indicating that each test could be used for the characterisation of BSE cases.  相似文献

The clinical findings in 59 cows with bovine spongiform encephalopathy (BSE) were compared with those in 19 cattle that were submitted as BSE suspects but not confirmed by immunohistochemistry. Both groups were also compared with a control group of 20 healthy cows. Abnormalities in behaviour, temperament, mental status and activity, neurogenic disorders of gait and hyperreactivity to touch were frequently observed in the cattle with BSE. Not every animal with BSE displayed clinical signs in all these categories, and the severity of the signs was not always useful for differentiating them from the BSE suspects that were not confirmed by pathology. The neurological examination was better than passive observations for the clinical diagnosis of BSE. Tests of the animals' responses to sudden auditory, visual and tactile stimuli were very useful for distinguishing cases of BSE from unconfirmed BSE suspects if the cases did not display signs in all the categories.  相似文献

In France, implementation of systematic screening programs in 2000, as a complement to the mandatory reporting of animals with clinical signs of BSE (passive surveillance), revealed certain limitations of the mandatory system. Indeed, systematic screening showed that some BSE cases were not detected by the clinical surveillance system, implying considerable BSE case under-reporting throughout the epidemic. As the most likely explanation for variant Creutzfeldt-Jakob disease (vCJD) is exposure to the aetiologic agent of BSE, it is essential to reconstruct the French BSE epidemic pattern accounting for this under-reporting. We estimated age- and year-specific incidence rates of BSE by using a back-calculation method. This approach relies on the principle that the number of clinical BSE cases is the consequence of the number of BSE-infected animals after a known incubation time, defined as the time between infection and clinical onset. We generalized this model to take into account epidemiological characteristics of BSE, such as French cattle mortality, BSE case reporting probability, and age-dependent susceptibility and/or exposure to the BSE agent. We confirmed that the average BSE incubation period is five years and that the peak risk of bovine infection occurs between 6 and 12 months of age. The results also showed that the proportion of underreporting is the most influential parameter in the model, and that BSE was substantially underreported until rapid tests were introduced. Indeed, only 103 BSE cases were detected by passive surveillance up to June 2000, while we estimated that there was 301 200 (95% confidence interval (CI) [27 600-837 600]) cattle infected by the BSE agent. Despite uncertainty over the beginning of the epidemic, we showed that the French BSE epidemic in the late 1980s was completely undetected, and only the second wave, after 1990, was observed.  相似文献

The adequacy of a histopathological diagnosis of bovine spongiform encephalopathy (BSE) based exclusively on observations of neuroparenchymal vacuolation in three specific neuroanatomic nuclei was tested by using a standard coronal section of medulla oblongata cut at the obex. The agreement between the observations and the definitive histopathological diagnosis was assessed in each of 684 bovine brains - 563 confirmed cases of BSE, 20 with changes which did not diagnose BSE conclusively and 101 in which the lesions of BSE were not detected. When the assessment was confined to the solitary tract nucleus and the spinal tract nucleus of the trigeminal nerve a positive result was obtained in 99.6 per cent of confirmed cases of BSE and only 1 per cent of brains in which lesions of BSE were not detected gave a false positive result. Thus an initial examination of the single section, together with an examination of representative areas of the rest of the brain when no unequivocal lesion was found, provided a satisfactory method for the routine diagnosis of BSE.  相似文献

Bovine spongiform encephalopathy (BSE) is a transmissible fatal neurodegenerative disease in cattle with an average incubation time of five years. The first BSE case in an indigenous cow was detected in Germany in November 2000. This was almost eight years after the huge BSE epidemic in the United Kingdom had peaked, and several years after many EU member states had seen their first BSE case. In the 1990s, BSE had been diagnosed in six imported animals in Germany. However, after the implementation of an active surveillance programme using BSE rapid testing systems, 399 indigenous German BSE cases have been found up to the end of July 2006. The birth cohorts of 1995-1997 contribute to the vast majority of the first 250 German cases that were diagnosed between 2000 and 2003. However, the most recent German BSE cases belong primarily to the birth cohorts 1998-2000 which is indicative of a recycling of BSE infectivity at that time. Moreover, there were two BSE cases in cattle born in spring 2001, i.e. after the meat and bone meal feed ban had come into effect on 2nd December 2000. In this article, we describe the dynamics of the German BSE epidemic and compare these data with those of other countries that observed larger numbers of cases.  相似文献

By the term "meat" the consumer understands muscle meat, that is skeletal muscle. The german regulations, the "Leits?tze für Fleisch und Fleischerzeugnisse" as well as the Codex Alimentarius define meat in a much broader sense to include all edible parts of slaughtered or shot warm-blooded animals (skeletal muscles, fat, heart, gut, liver etc). Therefore, a differentiated procedure is required when the risk of BSE transmission by meat has to be estimated. This must be based on knowledge of occurrence, amount, and persistence of BSE agents in the organs of animals. The risk evaluation has to include further factors: differences between animal species, age of the animal at the time of slaughter, the possibility of contamination during the slaughtering and cutting process, and--as far as meat products are concerned--the processing technology. To date there are no indications for the existence of transmissible spongiform encephalopathy in pigs, poultry and fish, even though BSE has been transmitted to pigs experimentally by intracranial administration. Muscle meat of these species can be considered safe. Muscle meat of experimentally infected cattle in the preclinical stage and even of animals with clinically manifest BSE has not shown infectivity in homologous and autologous bioassays performed so far. This finding justifies the assumption that the risk of BSE exposure by the consumption of beef can be classified as extremely low. Nevertheless, the absence of a proof of infectivity can at present not be equated with absence of BSE agents. This is because of the limits of sensitivity of the bioassays and because muscle meat does contain non-muscle tissue such as connective tissue collagen, nerve and lymph tissue and blood vessels.  相似文献

For a long time, BSE was considered a problem of the UK exclusively. Even after the detection of BSE cases in countries outside the UK, the risk of having BSE was categorically denied by many other countries. Only after the introduction of active surveillance did several "BSE free" countries detect BSE in their country. However, before the detection of the first cases in several of these countries, a risk assessment (GBR) conducted by the Scientific Steering Committee of the EU showed that a risk could be present. There remain many countries with an unknown BSE risk. In order to minimise import risks from these countries, further risk assessment is needed to evaluate the real BSE distribution worldwide.  相似文献

The objective of this study was to model the expected numbers of cattle incubating bovine spongiform encephalopathy (BSE) and the numbers of clinical cases of BSE in the Swiss cattle population between 1984 and 2005. The results were compared with the observed number of clinical BSE cases and with the results of a culling and testing scheme on herdmates of cattle with BSE. The age distribution of the Swiss cattle population, the age-at-death distribution of the first 235 BSE cases and exposure information were used to calculate the expected number of infected cattle in each birth cohort and the resulting numbers of clinical cases and survivors incubating the disease for each year. The model which did not assume any under-reporting of cases fitted the observed epidemic curve of clinical cases reasonably well, and predicted that the Swiss BSE epidemic would come to an end between 2003 and 2005. The age of survivors incubating BSE is increasing. The higher than expected incidence of subclinical cases observed in animals from the culling scheme is most probably the result of the heterogeneous distribution of infected animals and affected herds in the population. The results of the model need to be taken into account when designing surveillance and testing schemes for BSE.  相似文献

Since 1996, bovine spongiform encephalopathy (BSE) in cattle has been linked to a new variant of Creutzfeldt-Jakob disease (vCJD), a fatal brain disease in man. This paper assessed the cost-effectiveness of BSE control strategies instituted by the European Commission. In a Monte Carlo simulation model, a non-intervention baseline scenario was compared to three intervention strategies: removal of specified risk materials from slaughter animals, post-mortem testing for BSE and the culling of feed and age cohorts of BSE cases. The food risk in the baseline scenario ranged from 16.98 lost life years in 2002 to 2.69 lost life years in 2005. Removing specified risk materials removal practices, post-mortem testing and post-mortem testing plus cohort culling reduced this risk with 93%, 82.7% and 83.1%. The estimated cost-effectiveness of all BSE measures in the Netherlands ranged from 4.3 million euros per life year saved in 2002 to 17.7 million euros in 2005. It was discussed that the cost-effectiveness of BSE control strategies will further deviate from regular health economics thresholds as BSE prevalence and incidence declines.  相似文献

The staining characteristics of gamonts of Hepatozoon canis in peripheral blood smears were evaluated. Three stains, Diff-Quik stain, Giemsa stain and a modification of the naphthol-ASD-chloroacetate esterase stain, were compared. The staining characteristics of the infected cell and the parasite were different in each stain used. The modified naphthol-ASD-chloroacetate esterase stain showed the most difference between the infected and noninfected cells, as well as a difference in staining characteristics of the parasite nucleus from those of the host nucleus. Although the parasite was identified with all stains, a definitive diagnosis was more easily obtained with the naphthol-ASD-chloroacetate esterase stain.  相似文献

In order to identify the management and feeding practices that might have contributed to the occurrence of bovine spongiform encephalopathy (BSE) in Bavaria, Germany, information from 110 dairy farms on which a case of BSE had been reported was compared with information derived from a questionnaire sent to approximately 10,000 Bavarian farms on which no case of BSE had been reported up to February 2003. Representative information was obtained from 4006 dairy farms. The results indicated that in comparison with these control farms a higher proportion of the BSE farms had also kept pigs or poultry, although the difference was not significant, and that a significantly higher proportion of the BSE farms had fed proprietary concentrates and/or milk replacers to their calves.  相似文献

Since the first detection of bovine spongiform encephalopathy (BSE) in homebred cattle in Germany on 26 November 2000, 382 cases have been confirmed until 30 September 2005. Thirty‐two of these cases were reported from the Federal State of Schleswig‐Holstein (SH). There are hypotheses on the routes of infection for German cattle, but only few efforts have been made to assess potential risk factors by epidemiological studies. The purpose of this study was to identify potential risk factors at the farm level for the occurrence of BSE in cattle in SH. By applying the method of indirect standardization, the prevalence of various structural and management parameters of BSE‐affected farms in SH was compared with the prevalence of these parameters in a standard population of cattle farms from SH. The data describing the standard population were obtained by a cross‐sectional study performed in SH in 2003. Data of the BSE case population were available from the central German BSE case database. A possible association of the occurrence of BSE with the feeding of milk replacers to calves was observed. There was a clear indication that the occurrence of BSE was associated with the presence of pigs and/or poultry on the farm. This finding suggests that cross‐contamination of feed or cross‐exposition may have occurred in SH. The results obtained using the indirect standardization analysis were validated by multiple logistic regression. This study indicates that the feedborne path has been the principal route of transmission for the BSE agent in SH.  相似文献
