香蕉叶鞘腐败病病原鉴定   总被引:6,自引:1,他引:5  
 香蕉叶鞘腐败病为近年来发生在广东、广西、海南等地蕉园的一种新病害。经病原菌分离及柯赫氏法则检测,证明引起该病的病原菌为一种细菌,根据致病性、寄主范围测定、菌体形态、培养特性、生理生化反应及16S rDNA序列分析将该病原细菌初步鉴定为成团泛菌(Pantoea agglomerans)。成团泛菌危害香蕉,引起叶鞘腐败病在国内尚属首次报道。  相似文献

 2013年12月,甘肃省白银市水川镇日光温室中的西葫芦发生了严重的根腐和茎基腐,部分棚室病株率达50%,从病根和病茎上分离得到拟漆斑菌属真菌3株,病株分出率27.3%。采用胚根和茎基部接种法测定了菌株FG-62对西葫芦的致病性:茎基部接种后27 d,植株开始出现凋萎;接种后40 d,两种接种法的西葫芦苗均呈现严重根腐和茎基腐症状,茎基部接种的西葫芦凋萎株率达30%;从病根和病茎上均可再分离出原接种菌。菌株FG-62在PDA平板上25℃培养14 d,产生大量墨绿色至黑色分生孢子座,分生孢子无色至淡榄黑色,单胞,杆状或腰鼓状,两端钝圆,大小为(7.04~9.15)μm ×(1.97~2.46)μm,聚集的分生孢子呈黑色。BLASTn分析结果显示,菌株FG-62(GenBank 登录号 MK252098)的rDNA-ITS序列与露湿拟漆斑菌Paramyrothecium roridum分离物E-469 (GenBank 登录号KY582183.1)和CBS 357.89(源自模式材料,GenBank 登录号NR_145077.1)的序列相似性分别达99.65%和96.83%。依据病原菌形态学和rDNA-ITS序列,将其鉴定为露湿拟漆斑菌P. roridum (Basionym:Myrothecium roridum)。这是露湿拟漆斑菌引起西葫芦根腐和茎基腐的首次报道。  相似文献

菜豆种子普通细菌性疫病菌检测   总被引:2,自引:0,他引:2  
 由Xanthomonas axonopodis pv. phaseoli和Xanthomonas fuscans subsp. fuscans引起的菜豆普通细菌性疫病是严重影响菜豆生产的限制因子之一,可造成严重的产量和品种损失。染菌种子是病原菌传播的主要途径。本研究对5个菜豆主要产区的60份菜豆种子样品进行普通细菌性疫病菌检测。在MT选择性培养基上有36份种子样品浸提液检测到目标病原菌, 种子样品带菌量为2.49×102~5.20×107 CFU/粒。选择36个分离物接种感病品种“英国红”植株,所有分离物均引起接种植株发病。特异PCR检测结果表明,有20个分离物为X. fuscans subsp. fuscans,16个分离物为X. axonopodis pv. phaseoli。试验结果表明,我国一些菜豆主产区商业种植和研究用种子多数污染普通细菌性疫病菌,建议建立无菌种子生产区和加强种子管理,以有效控制病害发生。  相似文献

1964—1966三年共分离采自严重黄萎病株下部的尚未开裂成铃种子11505粒,平均内部带菌率<0.026%。系统分离棉铃各部位,其带菌率的趋势为:铃柄>铃托>胎座。从已吐絮的成熟铃内种子内部带菌分析结果看:1964年共分离外部带菌率约为5%的种子1557粒,全部未长微菌核。1966年分离病铃内成熟种子4049粒,只有一粒在种壳上长出了微菌核,带菌率为0.025%。用浓胞子液在田间灌注接种初裂铃,依受病程度不同,种子内部带菌率为0.93—2.4%。青铃注射接种的内部带菌率为10.40—70.40%。新吐絮铃喷洒接种,虽保湿1—2天,种子内部未分离到病菌。说明在菌量大,营养及温、湿度条件非常适宜时,病菌也可自种壳侵入内部,使有相当高的带菌率。通过三年的试验,作者肯定在自然情况下黄萎病菌主要附着在棉籽短绒上。内部带菌率极低。在轻病或无病棉株上选收正常吐絮铃内种子,内部基本不带菌。  相似文献

从不同地区和不同海拔果园采集枇杷幼果,分离与测定INA细菌冰核活性,结果表明冰核细菌广泛分布在不同产区枇杷幼果,其带菌率与地理生态条件有关。高海拔果园幼果带菌率高达77.5%,且出现冰核活性强的分离菌,海拔愈低其带菌率相应下降。用2个菌株做人工模拟霜冻和果园接种试验,结果显示接种处理明显加剧冻害,多数情况严重度增50%以上。首次证明INA细菌是诱发枇杷幼果冻害的重要因素。  相似文献

2009年对甘肃省9个市的21个县、区玉米穗腐病、茎基腐病的发生危害情况进行了全面调查,并采集病害标样,进行病原分离。调查结果表明,玉米穗腐病、茎基腐病在全省各玉米种植区均普遍严重发生,病田率100%,病穗率和病株率平均为63.57%和34.47%。初步分离结果表明,全省玉米穗腐病、茎基腐病的主要致病菌为禾谷镰刀菌、黄色镰刀菌和轮枝镰孢菌,但在地区间的分布有明显差异。  相似文献

2020年,在由斯里兰卡入境的甘蔗种茎中发现多条发病蔗茎。通过分离培养获得菌株,对菌株进行形态学鉴定、序列比对、系统进化分析和致病性测定。结果表明,该菌株形态与暗色座腔孢菌Phaeocytostroma sacchari相符。用引物ITS1和ITS4进行核酸序列扩增,获得约536 bp的目的片段,所获菌株km-1序列与P. sacchari一致性为100%;经系统进化分析发现,菌株km-1与暗色座腔孢菌聚在同一个进化分支。致病性接种结果表明,接种后的甘蔗具茎腐病典型症状,内部组织变为橙红色,并再次分离出暗色座腔孢菌。根据形态学、分子鉴定和致病性测定结果,确证分离到的甘蔗茎腐病的病原菌是给南非和印度甘蔗种植国家带来严重损失的暗色座腔孢菌。  相似文献

 免疫分离法是一种新型的、高度灵敏和准确的种菌带菌检验法,它结合了利用专化性抗血清的选择法吸附作用和活细菌可形成菌落的两方面的优点,从种苗材料中分离到目标细菌,用纯菌测试它的回收率为50~70%,最低检出率为50~100个菌/ml,人工喷接种和病田收获的病种带菌率达87%,平均每粒自然病种上的细菌数在100个左右。免疫分离所得菌落经致病性测定,大多数为白叶枯菌。  相似文献

赤霉素GA4+7种子引发处理对玉米光合作用及产量的影响   总被引:1,自引:0,他引:1  
研究了外源赤霉素处理玉米种子对其光合特性的影响,可为通过外源激素调控玉米生长和种子加工技术开发提供依据。以玉米种子‘郑单958’为材料,设置10、60、120 mg/L 3个质量浓度的GA4+7[m(GA4):m(GA7)=40:60]进行浸种后回干的引发处理,以常规不处理的种子为对照,测定了大田试验玉米叶片光合参数、叶绿素荧光参数、叶面积及光合产物积累量及光合色素含量。结果表明:GA4+7种子引发处理可提高玉米叶片的净光合速率、气孔导度、胞间CO2浓度、单株叶面积、干物质积累量以及光合色素含量,同时可降低叶绿素a与叶绿素b比值,有效降低初始荧光(F0),增大最大荧光(Fm)、PSⅡ的光能转换效率(Fv/Fm)和实际量子效率(F/Fm'),提高百粒重和籽粒产量,其中以60 mg/L的GA4+7处理(G2)效果最明显。在三叶期、七叶期与拔节期,G2处理玉米的净光合速率分别比对照提高74.3%、42.6%和39.6%(差异显著,P0.05),单株叶面积分别提高30.9%、48.5%和24.1%(差异显著,P0.05),干物质积累量分别增加19.6%、46.9%和52.9%(差异显著,P0.05),总叶绿素含量分别提高21.5%、12.1%和11.1%(差异显著,P0.05),百粒重和籽粒产量显著高于对照,分别提高6.0%和9.7%(P0.05)。  相似文献

ABSTRACT The dynamics of the adherent and nonadherent populations of three bacterial species on maize leaves were examined to identify the extent to which bacteria adhere to leaves and the importance of this adhesion to leaf colonization. Pantoea agglomerans strain BRT98, Clavibacter michiganensis subsp. nebraskensis strain GH2390, and Pseudomonas syringae pv. syringae strain HS191R all rapidly adhered to maize leaves following inoculation, but differed in the percentage of cells that adhered to the leaves. Immediately following inoculation, the percentage of adherent cells was highest for the saprophyte P. agglomerans (8 to 10%) and was much lower for the pathogens C. michiganensis subsp. nebras-kensis and P. syringae pv. syringae (2 to 3 and <1%, respectively), although the results for P. syringae pv. syringae HS191R were based on only one experiment. In the 4 days following inoculation, the percentage of the P. agglomerans populations that adhered to the leaves increased to approximately 70%. Similarly, the percentage of C. michiganensis subsp. nebraskensis and P. syringae pv. syringae cells that resisted removal steadily increased in the days following inoculation, although these increases probably reflected both adherence and localization to endophytic sites. Based on differences in the percentage of cells adhering to several cuticular wax mutants of maize, the rapid adherence of C. michiganensis subsp. nebraskensis cells to maize leaves was influenced by the cuticular wax properties, while the rapid adherence of P. agglomerans was not. Finally, bacterial adherence to leaves was advantageous to P. agglomerans survival and growth on leaves based on the finding that the nonadherent populations of the P. agglomerans strain decreased significantly more than did the adherent populations in the 24 h following inoculation, and increased much less than did the adherent populations over the next 3 days. Similar results with the C. michiganensis subsp. nebraskensis and P. syringae pv. syringae strains indicate that bacterial adherence to leaves, bacterial movement to endophytic sites, or both were advantageous to the survival and growth of these strains on leaves.  相似文献

In an area of Nigeria where downy mildew of maize is present, histological assessment of maize seed revealed the presence of mycelium and oospores of Peronosclerospora sorghi in the kernels. Seed transmission of downy mildew of maize was demonstrated when grain purchased at local markets gave mean seedling infection rates of 12·3% (untreated seeds) and 10·0% (in metalaxyl-treated seeds) within 7 days of emergence, after storage in a desiccator for 30 days. When untreated seeds taken from nubbin ears of systemically infected plants from four states in southern Nigeria were planted at 9 days (17–22% moisture content) and 27 days (9–22% moisture content) after harvest, 20·0% infected seedlings resulted in both trials. Seeds from Borno state in northern Nigeria had 26·6% systemic seedling infection after 9 months of storage at 11% moisture content. When seeds harvested from maize plants inoculated with P. sorghi through silks were examined histologically, hyphae of P. sorghi were observed mostly in the scutellum of the embryo. Transmission of disease to seedlings was observed when the silk-inoculated seeds (9% moisture content) were planted in pots in a greenhouse; however, no disease transmission was observed when such seeds were planted in the field. The epidemiological significance of seed transmission is discussed with particular reference to survival of inoculum and development of epidemics. Also noteworthy is the overall significance of seed transmission in Nigeria, where the major source of seed is that saved by farmers from their grain crop, occasionally supplemented by seed bought from the local market.  相似文献

为研究低温胁迫下种子包衣对玉米种子出苗和幼苗生长的影响,以适宜温度(25℃/20℃,白天/晚上,下同)处理为对照,对不同剂量包衣的玉米种子进行低温胁迫。研究结果表明,戊唑醇和苯醚甲环唑种子包衣在适宜温度下对玉米种子出苗具有促进作用,能提高玉米幼苗植株的叶绿素含量,对玉米幼苗的株高和鲜重没有抑制作用。但在17℃/6℃、15℃/0℃、10℃/0℃低温胁迫下,戊唑醇和苯醚甲环唑均能抑制种子出苗和幼苗生长,加剧低温胁迫导致的膜脂质过氧化作用和对叶绿体的分解。但用苯醚甲环唑处理玉米种子比用戊唑醇更安全,前者能抑制低温胁迫导致的叶片细胞内电解质外渗,后者却会加剧电解质的外渗。比较玉米幼苗根系的呼吸作用也可以发现,苯醚甲环唑比戊唑醇更有利于玉米幼苗补偿和修复低温胁迫所致的伤害。  相似文献

为明确氯虫苯甲酰胺种子处理对玉米田地下害虫的防治效果,筛选出合适的田间用药剂量,通过室内盆栽接虫与田间接虫相结合综合评价了氯虫苯甲酰胺对玉米地下害虫沟金针虫、铜绿丽金龟以及小地老虎的综合防治效果。结果表明,盆栽播种后20 d,氯虫苯甲酰胺4 g/kg拌种处理对沟金针虫、铜绿丽金龟和小地老虎的盆栽防效分别为78.15%、80.26%和81.05%;田间播种后20 d,氯虫苯甲酰胺4 g/kg拌种对沟金针虫、铜绿丽金龟和小地老虎的田间接虫防效分别为83.87%、86.27%和88.37%;高剂量的氯虫苯甲酰胺对玉米田地下害虫的田间综合防效优于对照药剂吡虫啉和氟虫腈。氯虫苯甲酰胺拌种处理对玉米种子无不良影响,对出苗和生长有一定的促进作用,有明显的防虫增产作用,可推广应用于防治玉米田地下害虫。  相似文献

Rumex obtusifolius is believed to be a nitrophilous weedy species that spreads mainly by seed. However, the effects of nutrient availability on seed production never have been investigated. In this study, how the amount of seed production per plant, the chemical composition of the seeds, and their size and germination are affected by the supply of N, P, and K was investigated. A pot fertilizer experiment with 10 treatments (combinations of 0, 150 [N1], and 300 [N2] kg N ha?1, 0, 40 [P1], and 80 [P2] kg P ha?1, and 0 and 100 [K] kg K ha?1 applied twice per vegetation season) was carried out in Prague, Czech Republic, in 2008 and 2009. The seeds from plants that were cultivated since the spring were collected each August. The germination of the seeds was determined in long‐daylight conditions at a stable temperature of 20°C. The number of seeds per plant ranged from 0 in the P1 and P2 treatments and <200 in the control and K treatments to almost 9000 in the N2P2K treatment. The number of seeds per plant was positively correlated with the number of stems per plant and the number of seeds per stem. The relationship between the amount of seeds per plant and the individual seed weight was positive. The concentration of N, P, and K in the seeds was affected by the treatments. The weight of 1000 seeds ranged from 1.3 to 1.7 g. The seed length ranged from 2.11 to 2.34 mm and the seed width varied from 1.26 to 1.42 mm. With the exception of the control (with 94% germination), the amount of seed germination was >98% in all the other treatments in 2008. In 2009, the amount of seed germination was >95% in all the treatments, except N1 and N2, where 93% and 89% germination, respectively, was recorded. The germination rate was positively related to the P concentration in the seeds. The amount of time that was necessary for 50% seed germination ranged from 1.53 to 2.89 days and was not related to the N concentration in the seeds, but it was negatively related to the P and K concentrations. A balanced N, P, and K supply to the mother plant strongly affected the number of produced seeds and their chemical composition and germination ability.  相似文献

Plasmopara halstedii, the causal agent of downy mildew of sunflower, is an oomycete listed as a quarantine pathogen. This obligate parasite resides in a quiescent state in seeds of sunflower and can be spread from seed production areas to areas of crop production by international seed trade. To prevent the spread or the introduction of potentially new genotypes or fungicide-tolerant strains, an efficient method to detect P. halstedii in sunflower seed is required. This work reports the optimization of a real-time detection tool that targets the pathogen within sunflower seeds, and provides statistically validated data for that tool. The tool proved to be specific and inclusive, based on computer simulation and in vitro assessments, and could detect as few as 45 copies of target DNA. A fully optimized DNA extraction protocol was also developed starting from a sample of 1,000 sunflower seeds, and enabled the detection of <1 infected seed/1,000 seeds. To ensure reliability of the results, a set of controls was used systematically during the assays, including a plant-specific probe used in a duplex quantitative polymerase chain reaction that enabled the assessment of the quality of each DNA extract.  相似文献

Thirty-five maize seed samples from three geographic regions of Costa Rica were analysed for fumonisin content, germinability, and Fusarium verticillioides infection with and without surface disinfection. The concentration of fumonisins in the maize samples varied from 4 ng g−1 to 16 000 ng g−1 with an all over-all average of 2500 ng g−1. There was a significant difference in fumonisin content between samples from Alajuela and Guanacaste. Germination of the seed samples ranged from 12% to 98% with significant differences between regions. F. verticillioides infection was 12–98% and 13–97% for surface-disinfected and nondisinfected seeds, respectively. There was a significant negative correlation ( r  = -0.52) between fumonisin content and seed germination, but no significant correlation was found between fumonisin content and F. verticillioides infection, or between F. verticillioides infection and seed germination. Most of the high fumonisin seed samples had some mechanical or insect damage. Whether or not the fumonisins had a direct effect on germination was not further established. It is concluded that the large differences in fumonisin content of maize seeds within and between regions are primarily caused by differences in seed quality, genetic diversity of F. verticillioides strains in natural populations, climatic differences between regions, and varietal differences. Some of the fumonisin levels found in this study coincide with levels associated with risks to humans and animals in other countries.  相似文献

为明确苹果锈果类病毒在八棱海棠果实和种子中的分布特征、种传率以及药剂脱除效果,以带毒母株上的八棱海棠果实和种子为试材,运用RT-PCR技术分析了八棱海棠果实不同部位锈果类病毒的带毒率、实生后代的带毒情况以及氢氧化钠脱除病毒的效果。结果表明,八棱海棠果皮、果肉、种子以及种胚均不同程度携带苹果锈果类病毒,其带毒率分别为96.0%、96.0%、52.0%和4.0%;该病毒可经种子传递给后代,种传率为12.1%;经2%氢氧化钠溶液浸种10、15、20 min,种子的病毒检出率均为0,但后代实生苗的带毒率分别为2.5%、1.3%和0。表明苹果锈果类病毒可侵染种子不同部位并经种子传递给后代,氢氧化钠浸种是脱除该病毒的有效方法。  相似文献

成团泛菌引起的棉花烂铃病对棉花产量因子和品质的影响   总被引:5,自引:1,他引:4  
成团泛菌(Pantoea agglomerans)引起的棉花烂铃病主要造成棉铃吐絮不畅,棉纤维变色,籽粒干瘪。本试验对感染该病害的不同棉花品种的发病率、绒长、单铃重、衣分以及色泽等指标进行测定,用SAS软件分析,结果显示:所有棉花品种都有烂铃病的发生,平均发病率为20.6%,不同品种间发病有明显的差别。病害对单铃重、衣分、瘪籽率、绒长及色泽的影响主要体现在单铃重平均减轻27.6%、衣分平均降低5.3%和瘪籽率平均增加1.6倍、绒长平均缩短1.6mm及平均颜色加深2.0%。由成团泛菌引起的这种细菌病害可引起棉花减产高达10%~20%。  相似文献
