The biological activities of catfish LH-like (semi-purified: s200a and purified Qa) and FSH-like (semi-purified: s200b and purified: Qb) were compared in intact and hypophysectomized female catfish, Clarias batrachus, during preparatory and the pre-spawning periods on vitellogenesis and ovarian maintenance, as well as in vitro final maturation of oocytes, germinal vesicle breakdown (GVBD). During preparatory period, in intact catfish, semi-purified FSH-like induced complete vitellogenesis through the production of estradiol-17β (E2) and vitellogenin (Vg) accompanied by the formation of SIII yolky oocytes. On the other hand, semi-purified LH-like had induced the formation of only SII (characterized by the appearance of cortical alveoli in cytoplasm) oocytes, which indicates the initiation of vitellogenesis. In hypophysectomized female catfish, purified LH-like but not FSH-like induced the formation of SII oocytes in the ovaries. Treatment with semi-purified LH- and FSH-like at the dose level of 5 µg/fish/day for 7 days significantly maintained the yolky oocytes in gravid catfish after hypophysectomy with a significant reduction in plasma Vg, but not E2 levels, indicating some unknown GtH-induced factor doing the job. In in vitro oocytes culture, both LH- and FSH-like induced GVBD, but the response was significantly more with LH-like than FSH-like. All these findings revealed that both LH-like and FSH-like have overlapping physiological functions, but their responses differ depending on the physiological status of the catfish.  相似文献

Oocyte growth in most oviparous vertebrates including fish is due to the formation of yolk, and eggshell proteins (zona radiata proteins). Zonagenesis leads to the formation of zona radiata proteins in oocytes, which play an important role during oogenesis, whereas vitellogenesis leads to the formation of yolk in oocytes through a series of events during which the yolk precursor protein vitellogenin (Vg) is synthesized and secreted from liver into blood from where it is sequestered into the developing oocytes and thereafter proteolytically cleaved to form yolk proteins (YPs) and finally deposited in the ooplasm. Much research has been done in many fish species with respect to the number and nature of Vg and YPs and their probable functions during fish reproduction. Recent findings of multiplicity of Vg molecules in fishes reject the earlier view of a single-Vg model and have led scientists to explore the functions of individual Vg and their YP derivatives, lipovitellin, phosvitin, and β′-component. Two distinct types of Vg or Vg genes, containing or encoding the three YPs, have been detected in many teleosts. A third unusual, incomplete, phosvitin-poor Vg has been described recently in many fishes. In comparison to much of the information on vitellogenesis in many fishes very little is known for Indian fishes. In India research has been done in a few species such as the catfish, Heteropneustes fossilis and Clarias batrachus, the murrel, Channa punctatus and the Indian major carps, Labeo rohita and Cirrhinus mrigala. Immunological and biochemical analyses suggest the occurrence of multiple forms of Vg and their YP derivatives. The synthesis and incorporation of Vg are regulated by gonadotropin (GTH) and estradiol-17β (E2). A differential role between estrone (E1) and estriol (E3) has been demonstrated for Vg synthesis. Enzyme-linked immunosorbent assays (ELISAs) for Vg have been developed to measure plasma Vg. Finally the different roles of Vg1 (HAI) and Vg2 (HAII) on vitellogenesis have been demonstrated. However, more research remains to be carried out in other fish species with respect to the number and nature of Vg and YPs and their genes in order to describe their reproductive functions.  相似文献

Two forms of vitellogenin (Vg: Vg1 and Vg2) were purified from the plasma of estradiol-17β (E2)-treated Indian walking catfish, Clarias batrachus, by gel filtration and adsorption chromatography. Native Vg1 and Vg2 had apparent molecular masses of 375 and 450 kDa, respectively, and both Vgs resolved into two similar major bands (95 and 67 kDa) in SDS-PAGE under reducing condition. Polyclonal antisera raised against each form of Vg were absorbed with a combination of hypophysectomized male catfish serum proteins and alternate Vg to ensure specificity. Immunological analyses verified the presence of Vg1 and Vg2 in the plasma of female catfish. Homologous ELISAs were developed for Vg1 and Vg2 using their respective harvested antisera, which exhibited the detection limit of 100 ng ml?1 for Vg1 and 40 ng ml?1 for Vg2, and low level of cross-reactivity (not parallel to the standard) was found with alternate Vg in each assay. Treatment of male catfish with E2 induced both Vgs showing a proportionate ratio of Vg1 to Vg2 at 5.6:1. Plasma concentrations of both Vgs measured by ELISAs at different reproductive phases of field collected female catfish increased in accordance with the ovarian development, keeping the proportionate ratio of Vg1 to Vg2 at about 2:1 in fish undergoing vitellogenesis during prespawning period and 1:20 during spawning period, suggesting that Vg1 may be the major Vg to contribute in yolk formation, whereas Vg2, besides its role in yolk formation, may facilitate other physiological functions. The present study, thus, demonstrates the occurrence of two unequally synthesized Vgs in the catfish.  相似文献

A feeding trial with striped catfish, Pangasianodon hypophthalmus was performed to determine the effect of Yucca schidigera in practical diet on the growth, feed utilization, body composition, total ammonia–nitrogen (TAN) excretion and haematological parameters. A diet with fish meal as the main protein source without yucca extract was used as the control diet (Diet 1). Four diets were formulated with 0.075 (Diet 2), 0.1 (Diet 3) and 0.15 (Diet 4)% of yucca extract respectively. Fifteen fish per tank (initial mean weight 1.78 ± 0.05 g) were randomly allocated to 15 fibreglass tanks (80‐L) connected to a freshwater closed recirculation system (temperature 29.7 ± 1.0°C). The experimental diets were tested in triplicates for 12 weeks. The specific growth rate of fish fed Diet 4 was significantly higher when compared with fish fed Diet 1. The growth of fish fed diets Diet 2 and Diet 3 were not significantly different compared with fish fed the Diet 1. Striped catfish fed Diet 4 had significantly lower feed conversion ratio compared with fish fed Diet 1 and Diet 2 (< 0.05).The incorporation of high level Yucca schidigera extract in the diets reduced TAN compared with Diet 1. Dietary inclusion of Yucca extract levels did not significantly affect the biometric parameters or whole body proximate composition of the striped catfish (> 0.05). The PCV (%) in fish significantly increased with high levels of Yucca inclusion (Diet 4) compared with control diet. Fish fed Diet 4 showed significantly higher haemoglobin levels than Diet 1 (< 0.05). The results indicate that dietary inclusion of Yucca schidigera extract is promising as a feed additive that could improve growth performance and some haematological parameters and the best Yucca schidigera level was 0.15%.  相似文献

Channel catfish, Ictalurus punctatus (Rafinesque), populations in six lakes in northern Idaho, USA, were sampled to describe their population characteristics. During the summers of 2011 and 2012, 4864 channel catfish were sampled. Channel catfish populations had low to moderate catch rates, and length structure was dominated by fish <400 mm. Channel catfish were in good body condition. All populations were maintained by stocking age‐1 or age‐2 fish. Growth of fish reared in thermally enriched environments prior to stocking was fast compared to other North American channel catfish populations. After stocking, growth of channel catfish declined rapidly. Once stocked, cold water temperatures, prey resources and (or) genetic capabilities limited growth. Total annual mortality of age 2 and older channel catfish was generally <40%. Tag returns indicated that angler exploitation was low, varying from 0 to 43% among lakes. This research provides insight on factors regulating channel catfish population dynamics and highlights important considerations associated with their ecology and management.  相似文献

The aim of this research is to evaluate the effects of biofloc system application at different fish density on the nursery production performance and the robustness of African catfish Clarias gariepinus (Burchell) against Aeromonas hydrophila and salinity stress. An economic analysis was also performed to determine the most optimum fish density in biofloc‐based African catfish nursery production. African catfish with an average body weight of 0.96 ± 0.04 g and average body length of 4.20 ± 0.46 cm, respectively, were distributed in 12 units of circular plastic‐lined tanks (1,020 L). The experiment consisted of four treatments in triplicates, that is, a control without carbon source addition at a density of 4 fish/L, and three biofloc treatments at different densities, that is, BFT4 (4 fish/L), BFT6 (6 fish/L) and BFT8 (8 fish/L). Tapioca flour was used as the organic carbon source in biofloc systems and was added at an estimated C/N ratio of 10. Housing the fish in biofloc systems resulted in higher fish growth, more efficient feed utilization, higher fish robustness against A. hydrophila infection and salinity stress, as well as higher profitability of nursery production. Increasing the fish density resulted in higher mortality. However, higher number of fish produced and lower feed conversion ratio were observed in the treatments with higher density (6 and 8 fish/L). In conclusion, the application of biofloc technology at a density of 8 fish/L could be recommended to increase the production and profitability of African catfish nursery culture.  相似文献

This study was conducted to evaluate the immunostimulating activity and growth promoting of diets supplemented with different rations of soluble and insoluble linseed fibre to Rhamdia quelen under hypoxia‐induced acute stress. For this reason, soluble and insoluble fractions of linseed fibre were concentrated separately and combined into four ratios (1:0.5; 1:1; 1:2 and 1:4), which were added to the diets of silver catfish (6.43 ± 0.12 g) and evaluated in a biological assay, along with a control diet (without addition of linseed fibre). After being fed the experimental diets for 45 days, specimens of silver catfish were submitted to hypoxia‐induced acute stress. They were kept out of water for 60 s. Immediately afterwards, blood and cutaneous mucus were collected for subsequent determination of immunological indicators and stress. After stress, the fish were weighed and measured for performance parameter calculation. The experimental design was completely randomized with five treatments and four replications. The data underwent analysis of variance and the means were compared by Tukey's test (p < .05). The fish fed diets containing the 1:2 and 1:4 soluble:insoluble fibre ratios, showed higher total protein content, globulin, plasma alkaline phosphatase activity, and performance in addition to higher mucoprotein content in the cutaneous mucus of the fish. Regardless of their ratio in the diet, linseed fibre provided higher plasma levels of total immunoglobulins and reduction in cortisol levels. The 1:1, 1:2 and 1:4 diets led to higher levels of total immunoglobulins and alkaline phosphatase activity in cutaneous mucus. The results indicate that linseed fibre has a stress‐ reduction, immunostimulant and a growth promoter effect on silver catfish. The 1:2 or 1:4 soluble: insoluble fibre ratios provided greater stimulation of the target immunological indicators and performance.  相似文献

A feeding trial was conducted to evaluate the potential of dietary supplementation of autolysed brewer's yeast (AY) on African catfish. The catfish (22.5 ± 1.15 g/fish, 20 fish 33 L/tank) were fed with either of diets (390 g/kg crude protein, 140 g/kg lipid) supplemented with 0, 3, 6 or 10 g/kg AY (n = 3). After 49 days of feeding, the final body weight and metabolic growth rate of the catfish fed 3 g/kg AY (3‐AY) diet were higher than those fed the control diet (p < .05). The lowest level (p < .05) of alanine transaminase was detected in the blood of the catfish fed 3‐AY diet. The mid‐intestinal histology of the catfish revealed no significant difference (p > .05) in intestinal perimeter ratio. However, an elevated (p < .05) abundance of goblet cells and intraepithelial leucocytes were found in the intestine of catfish fed 3, 6 and 10 g/kg AY diets, with the highest level of abundance recorded in the mid‐intestine of the catfish fed 3‐AY diet. The results suggest that dietary 3 g/kg autolysed brewer's yeast supplementation improves growth performance of African catfish without deleterious effect on liver functionality and gut morphology.  相似文献

This study was conducted to determine effects of dietary Fe levels on growth performance, hepatic lipid metabolism and antioxidant response for juvenile yellow catfish Pelteobagrus fulvidraco. Yellow catfish were fed six isonitrogenous and isolipidic diets containing Fe levels of 16.20, 34.80, 54.50, 76.44, 100.42 and 118.25 mg/kg for 8 weeks. Weight gain (WG) and specific growth rate (SGR) increased with dietary Fe levels from 16.20 to 54.50 mg/kg diet and then plateaued over the level. Feed conversion rate (FCR) was highest and protein efficiency rate (PER) was lowest for fish fed the lowest Fe levels of diet. Fe contents in whole body and liver increased with increasing dietary Fe levels. Hepatic lipid content was lowest, but mRNA levels of carnitine palmitoyltransferase (CPT‐1) and peroxisome proliferator‐activated receptor α (PPARα) were highest for fish fed 54.50 mg Fe/kg diet. Fish fed adequate dietary Fe levels reduced hepatic malondialdehyde (MDA) level and increased activities of antioxidant enzymes Superoxide dismutase (SOD), Catalase (CAT) and GS. Based on the broken‐line regression analysis of WG against dietary Fe levels, optimal dietary Fe requirement for yellow catfish was 55.73 mg Fe/kg diets. Fe‐induced changes in MDA levels and antioxidant enzymatic activities paralleled with the change in hepatic lipid content, suggesting the potential relationship between oxidative stress and hepatic lipid accumulation in yellow catfish.  相似文献

A sixty‐day feeding trial was conducted to determine the ascorbic acid (AA) requirement for growth of striped catfish, Pangasianodon hypophthalmus juveniles. Seven iso‐nitrogenous and iso‐energetic (370 g protein per kg and 19.6 MJ/kg) purified diets were prepared with different levels of ascorbic acid such as control (0), T1 (17.5), T2 (35), T3 (70), T4 (175), T5 (350) and T6 (700) mg ascorbic acid (L‐ascorbyl‐2‐polyphosphate) equivalent per kg diet. Fish with a mean body weight of 3.2–3.4 g were stocked (fifteen fish per tank) in triplicates following a completely randomized design. Each group was fed to satiation twice a day for 60 days. Significant differences were observed in growth, survival, body composition and metabolic enzymes activities with different dietary ascorbic acid levels. Maximum weight gain, specific growth rate (SGR) and protein efficiency ratio (PER) were found in fishes fed with 35 mg AA per kg diet, supported by best feed conversion. Fish fed a diet containing vitamin C had the highest activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) compared to those fed with vitamin C‐depleted diets. In this study, based on using broken‐line regression analysis, the dietary vitamin C requirement for growth of P. hypophthalmus juveniles was estimated to be in the range of 46–76 mg AA per kg, depending on the criterion used, growth and liver storage. Our results will be helpful for the formulation of cost‐effective ascorbic acid incorporated diets for striped catfish, P. hypophthalmus.  相似文献

Stenohaline freshwater fish with narrow salinity tolerance are susceptible to saline stress from global climate change and anthropogenic activities. The present study elucidated that saline exposure during the sensitive window of preparatory phase of oocyte maturation significantly affected gonadosomatic index, ovarian histology and morphometric features of oocytes in a stenohaline freshwater catfish, Heteropneustes fossilis (Bloch, 1794) in a dose (2 ppt, 5 ppt)—and duration (8, 24 days)‐dependent manner. The gonads of H. fossilis show annual maturation cycle. Loss of integrity of ovigerous lamellae, disruption of ovarian stroma, disrupted oolemma, ooplasmic vacuolization, damaged germinal vesicles and altered morphometry of previtellogenic oocytes, such as chromatin‐nucleolus, early perinucleolar and late perinucleolar, elucidated consistent effects of saline exposure except at 8 days exposure to 2 ppt of saline. Increased salinity might have affected the transmembrane ion/water transport and disrupted the osmotic balance in ovary that eventually led to impairment in growth of ovary and oocyte maturation. The susceptibility of ovary to comparatively less concentrations of saline exposure might be due to sensitiveness of ovary/oocytes during the early phase of growth. Fluctuating salinity along with other stressors can affect metabolic and growth rates, fecundity and ultimately survival of fish.  相似文献

Survival rates of the larvae of sutchi catfish Pangasianodon hypophthalmus are reported to be three times higher under dim conditions (0.1 lx) than those under 100 lx. In this study, larval behaviour of sutchi catfish was examined under various light intensities (<0.01, 0.1, 1, 10 and 100 lx) using a CCD camera to understand why survival rates vary under different light intensities. Five‐day‐old larvae showed significantly higher swimming activity under <0.01, 0.1 and 1 lx than those under 10 and 100 lx. On the other hand, the larvae showed significantly higher aggressive behaviour under 10 and 100 lx; swimming larvae attacked resting individuals more frequently under 10 and 100 lx than those under 0 and 0.1 lx. Aggressive behaviour was considered to induce lesions, inflicted by the sharp teeth of attacking larvae, on larval skin surfaces. It is considered that the chemical substances would generate from injured skin surfaces then acted as stimuli, causing the cannibalistic behaviour in other fish around the injured fish. This study provided evidence that the observed higher survival rates depended on lower frequency of aggressive behaviour under dark or dim conditions. It is therefore recommended that larval rearing of sutchi catfish be conducted under dim (less than 1 lx) conditions.  相似文献

A feeding trial was performed to determine the effect of a commercial source of Yucca schidigera and Quillaja saponaria extracts (Nutrafito Plus®), in practical diets, on striped catfish Pangasianodon hypophthalmus growth, feed utilization, body composition, total ammonia‐nitrogen (TAN) excretion and haematological parameters. Four experimental diets were supplemented with Nutrafito Plus® at 0%, 0.01%, 0.02% and 0.03% (diets: control, N01, N02 and N03 respectively). Three replicate groups of striped catfish, with initial mean weight of 1.78 ± 0.05 g, were fed one of the four diets for 12 weeks. The specific growth rate (SGR) and final weight of fish fed diet N03 was significantly higher (P < 0.05) than fish fed the other dietary treatments. The growth performance of fish fed diets N01 and N02 were not significantly different compared to fish fed the control diet. Striped catfish fed diet N03 had improved feed conversion ratio (FCR), net protein utilization (NPU) and protein efficiency ratio (PER) than the control (< 0.05). The inclusion of the high dietary level of Yucca schidigera and Quillaja saponaria, diet N03, reduced TAN compared to all groups. Dietary inclusion of Yucca schidigera and Quillaja saponaria at all levels investigated did not affect the whole body proximate composition of the striped catfish (> 0.05). The packed cell volume (PCV) and haemoglobin level in fish fed diet N03 was significantly higher than in the fish fed the control diet. The present study demonstrates that dietary inclusion of Yucca schidigera and Quillaja saponaria induced positive effects on growth performance and haematological parameters and decreased TAN excretion in striped catfish.  相似文献

African catfish (Clarias gariepinus) has been growing in popularity as a culture species globally. Although considerable research has been carried out on this species’ feeding preferences, no research has been carried out on optimum water level for growth. This is particularly surprising given this species' benthic nature. This study assessed the effect of different combinations of water levels and types of feed on the growth performance of African catfish. The study employed a 3 × 2 factorial design with three ponds of different water levels (0.5, 1 and 1.5 m) and two types of feeds (floating and sinking). Twelve earthen ponds (1 × 2 m) were each stocked with 16 catfish fingerlings (mean weight ~100 g), and their growth was monitored for 12 weeks. The fish cultured in the shallowest water grew significantly faster than those cultured in the deepest ponds. At the same time, fish cultured in the shallowest ponds had the lowest feed intake rates and consequently the lowest feed conversion ratios. Catfish fed sinking pellets grew faster than those fed floating pellets although the difference was not significant. There was no significant interaction between pond water depth and feed type. The results of this study suggest that the growth and feeding efficiency of C. gariepinus can be optimized by culturing in fairly shallow ponds (0.5 m).  相似文献

Effects of various concentrations of Scutellaria baicalensis (SB) extract in diets on growth, body composition, serum chemistry and disease challenge test of far eastern catfish (Silurus asotus) were determined and compared with a commercially available immune enhancer. Eight experimental diets were prepared in triplicate: control (Con) diet without supplementation of SB and SB‐0.25, SB‐0.5, SB‐1, SB‐2, SB‐3 and SB‐5 diets containing SB at concentrations of 0.25, 0.5, 1, 2, 3 and 5%, respectively. In addition, 0.1% of a commercial immune enhancer product (CP) was also tested. No significant difference in weight gain of fish was found. Feed consumption, feed efficiency ratio and protein retention of fish were not affected by the experimental diets. At the end of the 8‐week feeding trial, 10 externally normal fish from each tank were infected by Vibrio anguillarum or Strepotococcus iniae. Cumulative mortality of fish fed the Con diet was higher than that of fish fed the all other diets in 10 and 25 days after V. anguillarum or S. iniae infection. Results of this study indicated that dietary inclusion of SB extract was effective in improving survival of eastern catfish after V. anguillarum and S. iniae infection, but the various concentrations of SB did not affect fish performance.  相似文献

Experimental diets were formulated containing six levels of linolenic acid (0%, 0.5%, 1.0%, 1.5%, 2.0% and 2.5%) and each diet was fed to darkbarbel catfish Pelteobagrus vachelli in triplicate for 84 days. Weight gain, specific growth rate and feed efficiency increased with the increasing levels of dietary linolenic acid, but were not significantly different among diets containing 1.0%, 1.5%, 2.0% and 2.5% linolenic acid. Red blood cell count and haemoglobin were significantly higher in fish fed the 1.0% linolenic acid than other diets. Lysozyme activity and immunoglobulin M content in fish fed 1.0% of linolenic acid were significantly higher compared with the other diets. Antibody titre in fish fed 1.0–2.5% linolenic acids was significantly higher than in fish fed 0 or 0.5% linolenic acid 14 days after Aeromonas hydrophila challenge. This study indicates that linolenic acid is essential to darkbarbel catfish. Two‐straight broken‐line analysis with SGR showed that a dietary level of 1.29% linolenic acid is the minimum required for adequate growth of darkbarbel catfish. Considering other measured indicators of fish performance, a level of 1.0–1.5% linolenic acid in the diet is recommended for the darkbarbel catfish.  相似文献

Hormone‐induced spawning of channel catfish held communally in tanks is a reliable method to produce channel catfish, Ictalurus punctatus ♀ × blue catfish, Ictalurus furcatus ♂, F1 hybrid catfish fry. However, mature catfish are crowded, and repeatedly handled during the process of induced ovulation. Repeated handling of gravid females is stressful and may impair ovulation, egg quality, and reproductive performance. Three trials were conducted to evaluate the effects of two methods of confining post‐hormone‐injected female channel catfish on stress response (cortisol concentrations) and reproductive performance: fish were either held individually while suspended in soft, nylon‐mesh bags or communally in a concrete tank. Percent of females ovulated to hormone treatment, relative fecundity, percent egg viability, and latency of channel catfish did not differ for fish in the two treatments. However, percent hatch and fry/kg of females was higher (P < 0.05) for fish held in bags that for fish held communally in tanks. Mean plasma cortisol response immediately prior to the first hormone injection (0 h) did not differ among fish groups in the two treatments. However, mean plasma cortisol concentrations were significantly lower (P < 0.05) for fish in the bag treatment at 16 and 36 h compared to fish held communally in tanks. Plasma estradiol levels (measure of oocyte maturation) were assessed at 0, 16, and 36 h after hormone injection; concentrations were (P < 0.05) higher at 16 h compared to 0 and 36 h; however, estradiol concentrations did not differ for fish held in the two treatments (P > 0.05). Suspending hormone‐injected broodfish individually in soft bags reduced stress response, improved egg hatching rate, and increased hybrid fry produced per kg weight of female broodfish. Using this simple technology, farmers can improve the efficiency of hatcheries producing hybrid catfish fry.  相似文献

Aeromonas hydrophila is emerging as one of the major concerns in catfish aquaculture in the Southeastern United States due to recent outbreaks of motile aeromonad septicaemia (MAS) caused by virulent clonal isolates. There is no effective vaccine currently available for the prevention of MAS. In this study, two virulence‐associated proteins of A. hydrophila, aerolysin and haemolysin, were heterologously expressed in Escherichia coli cells. Recombinant aerolysin (rArl) and haemolysin (rHly) were used to immunize catfish. Both rArl‐ and rHly‐induced humoral immune response as evidenced by immunoblotting and cell agglutination; immunized fish had significantly less mortality as compared to control fish upon challenge with virulent A. hydrophila. When a mixture of rArl and rHly was used to immunize the fish, significantly higher relative per cent survival (RPS) was obtained. Sustained RPS of 71–78% were observed at 2–5 weeks post immunization. The results of this study indicated that immunization against aerolysin and haemolysin had significant impact on the establishment of pathogenesis by A. hydrophila, suggesting that these two proteins could serve as general immunogens for future development of recombinant protein vaccines.  相似文献
