The changes in some factors of the innate immunity (phagocytosis, complement, lysozyme); haematological parameters-leukocytes, erythrocytes, differential white blood cell counts, haemoglobin, haematocrit and the serum concentrations of the microelements zinc and iron in six 2- to 3-months-old female piglets after the intravenous administration of lipopolysaccharide from Escherichia coli 0111:B4 were determined. It was found out that 1 h after the administration of lipopolysaccharide at the dosage rate of 10 microg/kg body weight resulted in a decrease in the phagocytic parameters, i.e. the phagocytic number and the index of phagocytic activity, which was followed by an increase in their values between post treatment hours 2 and 4. The leukocyte counts had decreased by hour 2 after the injection, but thereafter increased, and at post treatment hour 72, a leukocytosis was observed. The differential white blood cell counts were characterized by a shift to the left between hours 2 and 4 and a statistically significant increase in lymphocyte counts at hour 48 of the experiment. The serum zinc concentrations were increased an hour after the lipopolysaccharide application; after which their average values were lower. The haemolytic activities (CH50) of the classical and the alternative pathways of complement activation decreased. The haemolytic activity (CH50) for the classical pathway began to increase at hour 48 following the treatment. Significant changes were not observed in lysozyme activity, serum iron concentrations or the related haematological parameters (erythrocytes and haemoglobin).  相似文献

Analysis of canine and feline haemograms using the VetScan HMT analyser   总被引:2,自引:0,他引:2  
The VetScan HMT is an impedance counter haematology analyser which produces a full blood count and three-part white blood cell differential. The aim of this study was to compare the results generated by the analyser with those obtained by standard methods used routinely in the authors' laboratory. Blood samples from 68 dogs and 59 cats were run on the VetScan HMT analyser and also subjected to reference methods, and the results obtained were compared. Correlation coefficients (feline/canine) were: 0.97/0.99 for haematocrit (Hct), 0.98/0.99 for haemoglobin (Hb), 0.81/0.98 for total white blood cells (WBC), and 0.89/0.97 for granulocyte and 0.65/0.93 for platelet counts. Coefficients for lymphocyte counts were 0.25/0.28 and for monocyte counts were 0.12/0.79. In conclusion, the VetScan HMT performed well on canine samples, showing excellent correlation for canine Hct, Hb, RBC, WBC, granulocyte and platelet counts. For feline samples, although there was excellent correlation for Hct, Hb and RBC, the WBC and three-part white blood cell differential and platelet count should be interpreted with caution as they can be unreliable.  相似文献

We compared the effects of dipotassium ethylenediaminetetraacetic acid (EDTA) and lithium heparin on hematologic values of green iguanas (Iguana iguana). Thirty-two privately owned sibling iguanas had blood drawn, and the sample was divided into three components: an EDTA tube, a heparin tube, and a nonanticoagulated blood smear. A full reptilian complete blood count was performed on each anticoagulated sample, and white blood cell (WBC) and leukocyte differential counts were performed on the whole-blood smears. Heparin and EDTA samples differed significantly in absolute values of thrombocytes, WBC, heterophils, and monocytes. The EDTA had no significant effect on the packed cell volume or plasma protein values, and the white blood count and differential counts produced with EDTA were more similar to those of the nonanticoagulated blood smear than were the counts produced with heparin.  相似文献

Bronchoalveolar washings were harvested from the excised lungs of 68 normal sheep of 3 different age groups (A: birth to 8 weeks; B: 6 months to 2 years; C: older than 2 years). Cells and fluid obtained were examined quantitatively and functionally. Fewer cells were present in the lavage fluids of Group A sheep compared with those of Groups B and C. Macrophages were the predominant cell type (70-80%) in all sheep, with lymphocytes being second in number. The lower proportion of lymphocytes in young sheep (Group A) was attributable to lack of B-lymphocytes. As a proportion of the lymphocyte population T-cells were in the majority (60-80%) in all sheep. The proportion of null cells was higher in young sheep than in adults. Pulmonary lymphocytes from sheep of all ages responded poorly to stimulation by the non-specific mitogens phytohaemagglutinin, concanavalin A, pokeweed mitogen and lipopolysaccharide of Escherichia coli. The proportion of neutrophils was higher in sheep over 2 years old compared with younger animals. Eosinophils were present in all age groups but their proportion was greatly increased in animals over 2 years of age. Basophils were absent in young sheep and were present in only low numbers in the lungs of adult animals. In young sheep (Group A) IgG was the predominant immunoglobulin. With age, the percentage of IgG in lung fluid decreased while that of IgA increased so that IgA became the predominant immunoglobulin in older animals (Group C). In sheep of all ages IgM was present in negligible amounts. The highest value of complement (C3) occurred in adult sheep (Group B). The total white blood cell counts and differential blood counts of all the sheep were within accepted normal ranges. As in the lungs, there was an age-associated reduction in the proportions of blood T-lymphocytes and an increase in B-lymphocytes. In contrast to lung lymphocytes, those from the blood of sheep of all ages exhibited a wide range of responses to mitogens. The lowest stimulation indices were observed in the oldest sheep (Group C). The results provide background data against which assessment can be made of changes consequent upon infection with respiratory pathogens.  相似文献

Objective To examine whether sub-optimal temperature induced stress and immunosuppression in farmed saltwater crocodile (Crocodylus porosus) hatchlings. Design A clinico-pathological study. Animals A total of 140 hatchlings were used. Procedure Body weight and length, plasma corticosterone and immunoglobulin concentrations and total and differential white blood cell counts were measured in 140 hatchlings from five clutches divided between five water temperature treatment groups. Initially all groups were housed at 32°C for 10 weeks, then two groups (L, LC) were changed to low temperature (28°C) and two groups (H, HC) to high temperature (36°C), while one group (C) remained at 32°C. The LC and HC groups were maintained at these temperatures for 10 days, after which the water temperature of both groups was returned to 32°C. Blood samples were collected twice (at 6 and 9 weeks of age) before the initial temperature change, and at 10 days and 4 weeks after the initial temperature change (at 11.5 and 14 weeks of age). Results Except for an increase in plasma corticosterone in the HC group and a decrease in the L group when the temperature change was first introduced, changes in plasma corticosterone were not significant. There were no significant changes in immunoglobulin concentrations. There were, however, significant decreases in the total white cell and lymphocyte counts in the LC group after the temperature was decreased to 28°C, and an increase in these counts after water temperature was returned to 32°C. Clutch of origin had significant effects on body weight and length gains, and there were negative relationships between body weight and corticosterone concentrations and between body weight and immunoglobulin concentrations. Conclusions As haematological changes indicative of stress were not associated with significant changes in serum corticosterone, immunosuppression in young crocodiles may be independent of the hypothalamic-pituitary-adrenal cortical axis.  相似文献

试验对海拔 3 0 0 0m的藏南谷地林芝引入家兔进行了部分生理指标的对比分析。测定了呼吸频率、心率、生理体温、血清蛋白质含量和血红蛋白含量。统计分析表明 :所测定项目的结果在 3品种兔之间均无明显差异 (P >0 0 5) ;但血红蛋白电泳分析 :3品种兔均呈现出A、A2 ,A33个区带 ,发现A3带以青紫兰兔最低 ,与其他 2品种兔差异极显著 (P <0 0 1 ) ;加里福尼亚兔A3带最高 ,显著高于中国白兔 (P <0 0 5)及青紫兰兔 (P <0 0 1 )。血清蛋白质电泳分析表明 :高原家兔的血清白蛋白均明显高于文献报道的低海拔饲养的新西兰白兔 ,血清总蛋白及其球蛋白含量明显下降  相似文献

Pregnancy is characterized by transient changes in the maternal immune system, also evident at peripheral level. The present study analyzes the kinetics and possible factors affecting peripheral white blood cell populations throughout pregnancy in a herd of high-producing dairy cows chronically infected or not with Neospora caninum. We examined 54 pregnant parous cows: 29 Neospora-seronegative and 25 Neospora-seropositive cows. Blood samples were collected on Days 90, 120, 150, 180 and 210 of gestation. General Linear Model (GLM) repeated measures analysis of variance showed that the interaction Neospora-seropositivity × parity significantly affected total leukocyte, neutrophil and monocyte counts with lower levels of total leukocytes, lower neutrophil and higher monocyte counts recorded in primiparous Neospora-seropositive cows. In addition, N. caninum-seropositive cows had significantly increased monocyte counts on Day 180 of gestation compared to seronegative ones. Other factors significantly associated with changes in total and/or differential leukocyte profiles were period of pregnancy, season, twin pregnancy and milk production. In conclusion, a parity-associated effect of chronic N. caninum infection was observed on peripheral blood cell profiles in dairy cattle during gestation.  相似文献

Monitoring of changes in some blood laboratory parameters in 34 horses after ingesting freshly harvested maize that was to be used for ensiling, heavily contaminated with young Datura stramonium plants, is described. For a 7-day period the following parameters were monitored: haemoglobin content (HGB), red blood cell counts (RBC), white blood cell counts (WBC), haematocrit (HCT), mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), differential white cell counts (DWC), erythrocyte sedimentation rate (ESR), protein fractions, aspartate aminotransferase (AST), lactate dehydrogenase (LDH), total bilirubin (TB), direct bilirubin (DB), blood glucose (Glu), total protein (TP), globulin (Glob) and albumin (Alb). The intoxication was accompanied by erythrocytosis, leukocytosis, regenerative left shift neutrophilia, lymphopaenia, eosinopaenia, increased haematocrit values, low erythrocyte sedimentation rate, hyperglycaemia, bilirubinaemia, hypoproteinaemia and increased activity of AST and LDH. No changes occurred in the mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), albumin, globulin and globulin fractions (alpha 1, alpha 2, beta 1, beta 2 and gamma). The blood parameters returned to normal between post-intoxication days 2 and 5. The observed changes in clinical chemistry indices could be used in the diagnosis, differential diagnosis and prognosis of Jimson weed intoxication.  相似文献

The therapeutic activity of a combination of difluoromethylornithine (DFMO) with diminazene aceturate was investigated in mongrel dogs experimentally infected with Trypanosoma congolense. The criteria used in the assessment of the trypanocidal effect of the therapy include the examination of the blood for parasites, as well as clinical and haematological changes at intervals following treatment. Diminazene aceturate and DFMO alone and in combination produced intermittent aparasitaemia in the dogs. Although relapse infection occurred with all three treatment regimes, the drug combination gave the best result. The packed red cell volume, haemoglobin concentrations and red blood cell values decreased significantly following parasite inoculation but increased after treatment. The total leucocyte counts decreased in all the infected dogs but improved with treatment, and the differential leucocyte counts indicated neutropenia in all the infected animals prior to treatment.  相似文献

The present study was designed to evaluate the effects of tumour necrosis factor‐α (TNF‐α) on lactating bovine mammary functions such as milk protein secretion and the integrity of the milk‐blood barrier. The effect on the induction of the systemic inflammatory response was also examined using concentrations of serum haptoglobin (Hp), a major inflammatory acute‐phase protein, as an index. One hundred micrograms per mammary gland of recombinant bovine (rBo) TNF‐α or placebo saline was individually infused into a rear mammary gland of each of four lactating cows, and milk and blood samples were collected before and 4, 8, 24, 32, 48, 96 and 168 h after infusion. In the rBoTNF‐α‐infused gland, increases of somatic cell counts were observed at 4–48 h. Although concentrations of total milk protein were not changed, compositions of milk proteins varied following rBoTNF‐α infusion. Concentrations of caseins, α‐lactalbumin and β‐lactoglobulin were significantly decreased at 4 and 8 h. Lactoferrin concentrations were significantly increased at 4 h. Significant infiltrations of serum albumin, immunoglobulin G1 (IgG1) and IgG2 were observed at 4 and 8 h. Elevations of the serum concentration of Hp were detected at 8‐32 h, but were very small in comparison with those reported in inflammatory diseases. Changes in rectal temperature and white blood cell counts were not significant. These results show that single rBoTNF‐α infusion into the lactating mammary gland suppresses the lactogenic function of the gland and influences the function of the milk‐blood barrier, with little effect on the generalized inflammatory response.  相似文献

The effects of ovariectomy, ovariohysterectomy and hysterectomy on morphologically demonstrable characteristics of lymphoid organs, peripheral white blood cell counts and antibody production against sheep red blood cells (SRBC) were studied in female Lewis rats. Removal of ovaries induced enlarged thymus weight and cellularity. No differences were observed between the groups in spleen weight, while hysterectomy together with ovariectomy influenced relative uterus draining lymph node (UDLN) weight. The percent numbers of pan T, helper T lymphocytes, cytotoxic/suppressor T cells and IgG bearing cells (B lymphocytes) of all investigated organs showed only moderate changes caused by the surgical procedures. In contrast, removal of ovaries generated elevated total leukocyte and lymphocyte counts in peripheral blood. The changes in absolute blood lymphocyte counts were accompanied by similar variations in absolute T and B lymphocyte numbers. Ovariohysterectomy had slightly greater effects on these parameters than ovariectomy alone. In addition, ovaries and uterus had only moderate influences on systemic immune responses toward SRBC. In conclusion, the present results indicate that the removal of ovaries and uterus can modify morphological characteristics of lymphoid organs and peripheral blood but antibody production showed only moderate changes caused by the surgical procedures.  相似文献

Even though there is an abundance of information on the reference values of haematological parameters in adult rabbits, a little is known about the changes in haematology in newborn rabbits or during their postnatal development. Therefore, the aim of our study was to investigate changes in red blood cells (RBC), white blood cells (WBC) and differential leukocyte counts in SPF New Zealand White rabbits from the age of one day to 20 weeks. Significant age-related changes during the first four weeks of life were detected. These included an increase of RBC and WBC, reversal of the neutrophil/lymphocyte ratio and increase of total counts of eosinophils and basophils. From the age of six weeks of life, all of the studied haematological parameters were comparable to those of adult rabbits.  相似文献

This research investigated the effects of levamisole and Echinacea purpurea (EP), separately and together on the immune response of the rat as a laboratory model. In this experiment, 40 male Wistar rats were obtained and divided into four groups (n = 10). These groups received normal saline (10 mg/kg), EP (500 mg/kg), levamisole (2 mg/kg) and EP (500 mg/kg) with levamisole (2 mg/kg) as oral gavages every day for a period of 4 weeks, respectively. After obtaining blood samples (at the end of each week), haematocrit (HCT), differential and total white blood cell (WBC) counts, phagocyte activity (number), total protein, albumin and globulins levels of samples were obtained. The results of the study showed that the gamma globulin level, WBC, neutrophil and monocyte counts and phagocyte activity increased significantly in comparison with normal saline group during the study. According to the results, each of the mentioned agents had a stimulant effect on the immune system separately and together on rats. In the group that received EP and levamisole simultaneously, these effects were synergistically increased. These compounds, by increasing the mentioned factors, will probably affect the immune system.  相似文献

Helminth and tick burdens of N'Dama cattle kept in village herds were compared with parasite burdens in Zebu cattle introduced into the same herds. The animals were monitored regularly for tick and helminth burdens, and blood samples were examined for differential white cell counts. The Zebu were found to have significantly higher numbers of endo- and ectoparasites. The N'Dama had significantly higher levels of circulating eosinophils than the Zebu during peak parasite burdens.

No difference in antibody levels to Amblyomma variegatum infestation could be detected between the two breeds. It is suggested that observed differences in susceptibility to endo- and ectoparasites between the two breeds cannot be totally attributed to environmental naivety of the Zebu, but must be partially owing to enhanced innate resistance factors in the N'Dama.  相似文献

The red lionfish Pterois volitans is important not only in the aquarium trade but also as an invasive species in the western Atlantic. Introduced to waters off the southeastern coast of the United States, red lionfish have rapidly spread along much of the East Coast and throughout Bermuda, the Bahamas, and much of the Caribbean. Hematology and plasma biochemistry were evaluated in red lionfish captured from the offshore waters of North Carolina to establish baseline parameters for individual and population health assessment. Blood smears were evaluated for total and differential white blood cell counts, and routine clinical biochemical profiles were performed on plasma samples. To improve the interpretive value of routine plasma biochemistry profiles, tissue enzyme activities (alkaline phosphatase [ALP], alanine aminotransferase [ALT], aspartate aminotransferase [AST], gamma-glutamyl transferase [GGT], lactate dehydrogenase [LD], and creatine kinase [CK]) were analyzed from liver, kidney, skeletal muscle, gastrointestinal tract, and heart tissues from five fish. The hematological and plasma biochemical values were similar to those of other marine teleosts except that the estimated white blood cell counts were much lower than those routinely found in many species. The tissue enzyme activity findings suggest that plasma LD, CK, and AST offer clinical relevance in the assessment of red lionfish.  相似文献

为了解勺鸡、雉鸡和石鸡血液生理参数的正常值及其特点,为人工饲养繁殖以及健康状况监测提供参考依据,采用常规方法,对人工饲养的3种雉科鸟类(勺鸡、雉鸡和石鸡)红细胞数、白细胞总数、白细胞分类计数、血小板数、血红蛋白含量、红细胞沉降率、红细胞体积及红细胞比容8项血液生理指标进行了测定。结果表明,雉鸡的血小板明显高于勺鸡和石鸡(P<0.05);其它7项血液生理指标3种鸟之间差异不显著(P>0.05)。  相似文献

Fourteen calves were used to investigate the changes from birth to 83 days of age in the concentrations of serum albumin, alkaline phosphatase, beta-hydroxybutyrate, plasma cortisol, serum creatine kinase, creatinine, iron, plasma fibrinogen, serum gamma-glutamyl transferase, plasma glucose, haptoglobin, serum non-esterified fatty acids, total protein, transferrin, triglycerides, urea and gamma globulin; the haematological variables measured were: basophils, eosinophils, haematocrit, haemoglobin, lymphocytes, mean cell haemoglobin, mean cell haemoglobin concentration, mean cell volume, monocytes, band neutrophils, neutrophils, platelets, red blood cells and white blood cells. The changes are presented as a series of graphs and the values are discussed in relation to the published reference ranges for adult cattle. Two populations of calves were identified which gave rise to a bimodal distribution for some of the variables. Differences in haematocrit, haemoglobin and red blood cell counts were apparent at birth, with raised values for these measurements being associated with an increased white blood cell and neutrophil count between three and 27 days of age.  相似文献

A case of canine parvovirus type 2c (CPV-2c) infection in a 3-month-old feral kitten with a cerebral abscess and neurological disease is reported. The cat displayed ataxia and convulsions together with signs of gastroenteritis and profound alteration of the total and differential white blood cell counts. A parvovirus strain was detected by a TaqMan assay in the blood and faeces of the affected kitten, which was characterised as CPV by means of molecular assays but did not react with any of the CPV type-specific probes. By sequence and phylogenetic analyses of the VP2-protein gene, the CPV-2c strain displayed a non-coding mutation in the probe-binding region. Although the role of CPV-2c in this particular case is unclear, it is possible that it predisposed the kitten to the clinical signs seen. Continuous surveillance is needed to monitor future spreading of this CPV-2c mutant, and any associated clinical signs, in the dog and cat population.  相似文献

42 horses were examined. The physical signs with relation to circulatory insufficiency and the abdominal disease were registered following a two-phased examination procedure. Great prognostic value was found in the degree of circulatory insufficiency judged by pulse rate and character, filling of the jugular vein, skin temperature, colour of mucous membranes, capillary refill time, sweating, depression, skin turgor and degree of enophthalmus. In making a causal diagnosis the abdomen was examined for shape, tenderness, peristaltic sounds, gastric dilation by siphoning, abnormal rectal findings and macroscopic changes in peritoneal fluid. Greatest diagnostic difficulties were encountered in cases of intestinal atonia, acute enteritis and torsion of the colon. In selected (severe) cases laboratory tests were obtained. Blood samples were examined for packed cell volume, hemoglobin, red and white blood cell counts, differential white blood cell count, blood gases and acid-base status, lactate, serum total protein and albumin, plasma sodium, potassium, chloride, calcium, magnesium, inorganic phosphorus, glucose, creatinine, BUN, total bilirubin, ASAT, CK, BASP and GGT. Peritoneal fluid was examined for red blood and white cell counts, total protein, specific gravity, pH and lactate, and enzymes as in blood. Laboratory results generally confirmed the clinical signs of shock, and packed cell volume and blood lactate were regarded to be of greatest prognostic interest. Although the performed laboratory information, macroscopic evaluation was thought to reveal sufficient information in most cases. It was concluded that supervening shock is of decisive importance in severe forms of colic, and that a careful and repeated evaluation of the circulatory insufficiency often provides one with a tentative prognosis although the final diagnosis is not obtained. In spite of therapy fatal outcome was found in all seriously shocked horses.  相似文献
