To provide theoretical and applied references for biofortification of maize by increasing Zn and Fe concentration, the correlation and quantitative trait loci (QTL) of four mineral-related traits, i.e. zinc concentration of kernel (ZnK), zinc concentration of cob (ZnC), iron concentration of kernel (FeK) and iron concentration of cob (FeC) were studied for two sets of F2:3 populations derived from the crosses Mu6 × SDM (MuS) and Mo17 × SDM (MoS) under two different environments (CQ and YN). The parental lines were very different in Zn and Fe concentration of kernels and cobs. A large genetic variation and transgressive segregation of two F2:3 populations were observed for the four traits. The heritability of FeK was relatively lower (<0.6) than other three traits (>0.7). Analysis for each environment and joint analysis across two environments were used to identify QTL for each population. 16 and 15 QTL were identified in CQ and YN respectively via single environment analysis, some of which were identical in different environments and were also detected in joint analysis. The common regions for same trait at different environments were 3 and 5 in MuS and MoS respectively. Compared with the IBM2 2008 Neighbors Frame6, the distribution and effect of some QTL in two populations were highly consistent and many QTL on chromosome 2, 7 and 9 were detected in both populations. Moreover, several mineral QTL co-localized with each other for both populations such as the QTL for ZnK, ZnC, FeK and FeC on chromosome 2, QTL for Znk, FeK and FeC on chromosome 9 and QTL for ZnK and ZnC on chromosome 7, which probably were closely linked to each other, or were the same pleiotropic QTL.  相似文献

White mold (WM), caused by Sclerotinia sclerotiorum (Lib.) de Bary, is a widespread disease of dry and green bean (Phaseolus vulgaris L.) in North America. Gamete selection (GS) was effective to combine and pyramide resistant genes and quantitative trait loci (QTL) for common bacterial blight. Our objective was to determine the effectiveness of GS to introgress physiological resistance to white mold. Two inter-gene-pool double-cross populations were developed. Selection for WM resistance was practiced from F1 to F4. Thirteen selected F1:5 breeding lines of each population and their four parents were evaluated. Two separate inoculations were made on each plant 1 week apart using a cut-stem method. The WM reaction was scored at 16, 23, and 33 days post inoculation (DPI) using a scale from 1 (no disease) to 9 (severely diseased or dead). In F1, 52% of Pop I (USPT-WM-1/CORN 601//USPT-CBB-1/92BG-7) and 67% of Pop II (Chase/I9365-25//ABL 15/A 195) susceptible plants were discarded. In F4, only 1.2% of families from Pop I, and 0.9% for Pop II, survived the selection process. An average of 20.5% gain in WM resistance was obtained for both populations in F4. Four breeding lines of Pop I had significantly (P = 0.05) lower WM score (4.1–4.6) and four were equal (4.7–4.9) to the best WM-resistant parent 92BG-7 (4.9), while ten breeding lines of Pop II were equal (4.5–4.8) to the best WM-resistant parent A 195 (4.6). Two related segregating populations of Theobroma cacao L. were analysed for their resistance to Phytophthora palmivora. The first F1 population was obtained by crossing two susceptible cacao clones of Catongo (a highly homozygous genotype) and Pound 12(a highly heterozygous genotype) and the second population was obtained by backcrossing a single F1 tree with Catongo. The genetic maps obtained for each population were compared. The F1 map includes 162 loci and the backcross has 140 loci. The two maps, F1 and BC1, exhibit high co-linear loci organization covering respectively, 772 and 944 cM.Phytophthora resistance was assessed by measuring the size increase of a lesion at five (DL5)and ten days (DL10) after pod inoculation. Six different QTL were detected in the F1 and BC1 populations. One QTL was found in both populations, and appeared to be a major component of disease resistance, and explaining nearly 48% of the phenotypic variance in the F1 population. The absence of some yield QTL detection in the BC1 in comparison with the F1 population is due to the lack of transmission of the favouring alleles for these QTL from the single F1 tree used for the backcross. The phenotypic variance explained by the action of the quantitative trait alleles indicated that genetic factors of both major and minor effects were involved in the control of the character studied. QTL conferring increased resistance to Phytophthorawere identified in both susceptible parents, suggesting the presence of transgressive traits and the possibility of selection in cacao. Pleiotropic and epistatic effects for the QTL were also detected. Finally, the use of marker assisted selection (MAS) in cacao breeding programs is discussed. Four cycles of recurrent selection for FHB resistance were conducted in an intermating wheat breeding population using the dominant male-sterile gene ms 2 during 1987–1991.Five cycles of phenotypic mass selection for male-sterile plants were evaluated using the soil-surface inoculation method in Experiment I. Experiment II evaluated changes in FHB scores during five cycles of progeny selection for fertile plants using the single-floret inoculation method. In Experiment I, the average level of FHB response increased to MR level in C4, compared to MS level in C0. The numbers of infected spikelets and diseased kernels decreased 0.32 and 2.68 per cycle, respectively. In Experiment II, the average level of FHB response increased to R level in C4F1. The numbers of infected spikelets and diseased kernels decreased 0.93 and 4.58 per cycle, respectively. In both experiments, the largest selection gains were realized in the first cycle. The frequencies of R and MR individuals were increased significantly. The frequencies of individuals with FHB response equal and/or superior to Sumai 3 were increased to 5–8% in C4 and 25% in C4F1after the fourth cycle. Agronomic traits tended to be slightly improved in selected populations. Compared to 2% in C0, about 34% of lines superior in both FHB resistance and agronomic traits in C4F1 were selected to enter the conventional breeding program for further evaluation. Sixty three semidwarf lines superior in both FHB resistance and yield potential were selected from the F5 generations derived from C1F1 to C4F1. From them, two resistant cultivars with high-yielding potential were developed and commercialized in the Lower Yangtze Valley. Recurrent selection appears to be highly effective and feasible in shifting the average FHB response of the intermating population in the desirable direction, thereby enhancing the frequency of resistant individuals. Drought is one of the major factors limiting barley yields in many developing countries worldwide. The identification of molecular markers linked to genes controlling drought tolerance in barley is one way to improve breeding efficiency. In this study, we analyzed the quantitative trait loci (QTL) controlling chlorophyll content and chlorophyll fluorescence in 194 recombinant inbred lines (RILs) developed from the cross between the cultivar ‘Arta’ and Hordeum spontaneum 41-1. Five traits, chlorophyll content, and four chlorophyll fluorescence parameters, namely initial fluorescence (Fo), maximum fluorescence (Fm), variable fluorescence (Fv), and maximum quantum efficiency of PSII (Fv/Fm) which are related to the activity of the photosynthetic apparatus, were measured under well-watered and drought stress conditions at post-flowering stage. QTL analysis identified a total of nine and five genomic regions, under well-watered and drought stress conditions, respectively, that were significantly associated with the expression of the five target traits at post-flowering stage. No common QTL was detected except one for chlorophyll content, which was identified in both growth conditions, demonstrating that the genetic control of the expression of the traits related to photosynthesis differed under different water conditions. A QTL for Fv/Fm, which is related to the drought tolerance of photosynthesis was identified on chromosome 2H at 116 cM in the linkage map under drought stress. This QTL alone explained more than 15% of phenotypic variance of maximum quantum yield of PSII, and was also associated with the expression of four other traits. In addition, another QTL for Fv/Fm was also located on the same chromosome (2H) but at 135.7 cM explaining around 9% of the phenotypic variance under drought conditions. The result presented here suggest that two major loci, located on chromosome 2H, are involved in the development of functional chloroplast at post-flowering stage for drought tolerance of photosynthesis in barley under drought stress. If validated in other populations, chlorophyll fluorescence parameters could be used as selection criteria for drought tolerance.  相似文献

To further understand the nature of hybrid sterility between Oryza sativa and Oryza glaberrima, quantitative trait loci (QTL) controlling hybrid sterility between the two cultivated rice species were detected in BC1F1 and advanced backcross populations. A genetic map was constructed using the BC1F1 population derived from a cross between WAB450-16, an O. sativa cultivar, and CG14, an O. glaberrima cultivar. Seven main-effect QTLs for pollen and spikelet sterility were detected in the BC1F1. Forty-four sterility NILs (BC6F1) were developed via successive backcrosses using pollen sterility plants as female and WAB450-16 as the recurrent parent. Seven NILs, in which the target QTL regions were heterozygous while the other QTL regions as well as most of the reminder of the genome were homozygous for the WAB450-16 allele, were selected as the QTL identification materials. BC7F1 for the seven NILs showed a continuous variation in pollen and spikelet fertility. The four identified pollen sterility QTLs were located one each on chromosomes 1, 3, 7 and 7. Pollen sterility loci qSS-3 and qSS-7a were on chromosomes 3 and 7, respectively, which coincides with the previously identified S19, and S20, while loci qSS-1 and qSS-7b on chromosomes 1 and 7L appear distinct from all previously reported loci. An epistatic interaction controlling the hybrid sterility was detected between qSS-1 and qSS-7a.  相似文献

A population of 112 F1-derived doubled haploid lines was produced from a reciprocal cross of Brassica juncea. The parents differed for seed quality, seed color and many agronomic traits. A detailed RFLP linkage map of this population, comprising 316 loci, had been constructed, and was used to map quantitative trait loci (QTL) for seed yield and yield components, viz. siliqua length, number of seeds per siliqua, number of siliques per main raceme and 1000-seed weight. Stable and significant QTLs were identified for all these yield components except seed yield. For yield components, a selection index based on combined phenotypic and molecular data (QTL effects) could double up the efficiency of selection compared to the expected genetic advance by phenotypic selection. Selection indices for high seed yield, based on the phenotypic data of yield and yield components, could only improve the efficiency of selection by 4% of the genetic advance that can be expected from direct phenotypic selection for yield alone. Inclusion of molecular data together with the phenotypic data of yield components in the selection indices did not improve the efficiency of selection for higher seed yield. This is probably due to often negative relationships among the yield components. Most of the QTLs for yield components were compensating each other, probably due to linkage, pleiotropy or developmentally induced relationships among them. The breeding strategy for B. juncea and challenges to marker assisted selection are discussed.  相似文献

Xiangzamian 2 (XZM2) was the most widely cultivated cotton hybrid planted as F1 hybrids and as selfed F2 seeds in China before the release of transgenic Bt hybrids. By crossing two parents of XZM2, Gossypium hirsutum cv. Zhongmiansuo12 (ZMS12) and G. hirsutum acc. 8891, and through subsequent selfings, we obtained F8 and F9 populations of 180 recombinant inbred lines (RILs). A RIL population was cultivated in two cotton-growing regions in China for 2 years. The purpose of the present research was to detect quantitative trait loci (QTL) for fiber quality and provide information applicable to cotton breeding. A genetic map was constructed mainly using SSR markers. QTL controlling fiber quality traits were determined at the single-locus and two-locus levels, and genotype-by-environment interactions were analyzed. Among the main-effect QTL, a fiber length QTL qFL-D2-1 and a reflectance QTL qFR-D2-1 were simultaneously detected at two growing regions in 2 years, which suggested a high degree of stability in different environments, and might be of particular value for a marker-assisted selection (MAS) program. The results suggested that epistatic effects, as well as additive effects, of QTL play important roles in fiber quality in these RILs. In our research, the phenomenon of QTL clusters was detected in the cotton genome.  相似文献

A diversity arrays technology (DArT) map was constructed to identify quantitative trait loci (QTL) affecting seed colour, hairy leaf, seedling anthocyanin, leaf chlorosis and days to flowering in Brassica rapa using a F2 population from a cross between two parents with contrasting traits. Two genes with dominant epistatic interaction were responsible for seed colour. One major dominant gene controls the hairy leaf trait. Seedling anthocyanin was controlled by a major single dominant gene. The parents did not exhibit leaf chlorosis; however, 32% F2 plants showed leaf chlorosis in the population. A distorted segregation was observed for days to flowering in the F2 population. A linkage map was constructed with 376 DArT markers distributed over 12 linkage groups covering 579.7 cM. The DArT markers were assigned on different chromosomes of B. rapa using B. rapa genome sequences and DArT consensus map of B. napus. Two QTL (RSC1‐2 and RSC12‐56) located on chromosome A8 and chromosome A9 were identified for seed colour, which explained 19.4% and 18.2% of the phenotypic variation, respectively. The seed colour marker located in the ortholog to Arabidopsis thaliana Transparent Testa2 (AtTT2). Two QTL RLH6‐0 and RLH9‐16 were identified for hairy leaf, which explained 31.6% and 20.7% phenotypic variation, respectively. A single QTL (RSAn‐12‐157) on chromosome A7, which explained 12.8% of phenotypic variation was detected for seedling anthocyanin. The seedling anthocyanin marker is found within the A. thaliana Transparent Testa12 (AtTT12) ortholog. A QTL (RLC6‐04) for leaf chlorosis was identified, which explained 55.3% of phenotypic variation. QTL for hairy leaf and leaf chlorosis were located 0–4 cM apart on the same chromosome A1. A single QTL (RDF‐10‐0) for days to flowering was identified, which explained 21.4% phenotypic variation.  相似文献

Root traits are key components of plant adaptation to drought environment. By using a 120 recombined inbred lines (RILs) rice population derived from a cross between IRAT109, a japonica upland rice cultivar and Yuefu, a japonica lowland rice cultivar, a complete genetic linkage map with 201 molecular markers covering 1,833.8 cM was constructed and quantitative trait loci (QTLs) associated with basal root thickness (BRT) were identified. A major QTL, conferring thicker BRT, located on chromosome 4, designated brt4, explained phenotypic variance of 20.6%, was selected as target QTL to study the effects of marker-assisted selection (MAS) using two early segregating populations derived from crosses between IRAT109 and two lowland rice cultivars. The results showed that the flanking markers of brt4 were genetically stable in populations with different genetic backgrounds. In the two populations under upland conditions, the difference between the means of BRT of plants carrying positive and negative favorable alleles at brt4 flanking markers loci was significant. Phenotypic effects of BRT QTL brt4 were 5.05–8.16%. When selected plants for two generations were planted at Beijing and Hainan locations under upland conditions, MAS effects for BRT QTL brt4 were 4.56–18.56% and 15.46–26.52% respectively. The means of BRT for the homozygous plants were greater than that of heterozygous plants. This major QTL might be useful for rice drought tolerance breeding. L. Liu and P. Heterosis, or hybrid vigour, has been used to improve seed yield in several important crops for decades and it has potential applications in soybean. The discovery of over‐dominant quantitative trait loci (QTL) underlying yield‐related traits, such as seed weight, will facilitate hybrid soybean breeding via marker‐assisted selection. In this study, F2 and F2 : 3 populations derived from the crosses of ‘Jidou 12’ (Glycine max) × ‘ZYD2738’ (Glycine soja) and ‘Jidou 9’ (G. max) × ‘ZYD2738’ were used to identify over‐dominant QTL associated with seed weight. A total of seven QTL were identified. Among them, qSWT_13_1, mapped on chromosome 13 and linked with Satt114, showed an over‐dominant effect in two populations for two successive generations. This over‐dominant effect was further examined by six subpopulations derived from ‘Jidou12’ × ‘ZYD2738’. The seed weight for heterozygous individuals was 1.1‐ to 1.6‐fold higher than that of homozygous individuals among the six validation populations examined in different locations and years. Therefore, qSWT_13_1 may be a useful locus to improve the yield of hybrid soybean and to understand the molecular mechanism of heterosis in soybean.  相似文献

Chickpea shows a distinct domestication trajectory vis‐a‐vis pod dehiscence and growth cycle mediated by vernalization insensitivity compared with its companion Near Eastern legumes. Our objectives were: (i) to map the quantitative trait loci (QTLs) associated with vernalization response and seed free tryptophan in domesticated × wild chickpea progeny and (ii) estimate the genetic correlation between vernalization response and free tryptophan content. A domesticated × wild chickpea cross was used to document phenotypic segregation in both traits and to construct a skeletal genetic map for QTL detection. A number of vernalization response and seed free tryptophan content QTLs were documented in both F2 and F3 generations. No significant genetic correlation between these two traits was observed. Epistatic relationship between two free tryptophan loci was documented. It is evident that selection for high seed tryptophan is easier to accomplish relative to selection for vernalization insensitivity. This suggests that the two traits were selected independently in antiquity, thereby corroborating earlier claims for conscious selection processes associated with chickpea domestication.  相似文献

Despite the well-recognized importance of grain yield in high-oil maize (Zea mays L.) breeding and production, few studies have reported the application of QTL mapping of such traits. An inbred line of high-oil maize designated ‘GY220’ was crossed with two dent maize inbred lines to generate two connected F2:3 populations with 284 and 265 F2:3 families. Our main objective was to evaluate the influence of genetic background on QTL detection of grain yield traits through comparisons between the F2:3 populations. The field experiments were conducted during the spring in Luoyang and summer in Xuchang, Henan, China. Two genetic linkage maps were constructed with a genetic distance of 2111.7 and 2298.5 cM using 185 and 173 polymorphic SSR markers, respectively. In total, 18 and 15 QTL were detected for six grain yield traits in the two populations. Only one common QTL marker was shared between the two populations. A QTL cluster associated with five traits was identified at bin 1.05–1.06, including the shared QTL for 100GW, which demonstrated the largest effect (16.7%). Among the detected QTL, 12 digenic interactions were identified. Our results reflect the substantial influence of dent maize genetic background on QTL detection of grain yield traits.  相似文献

The objectives of this study were to validate the major quantitative trait locus (QTL) for scab resistance on the short arm of chromosome 3B in bread wheat and to isolate near‐isogenic lines for this QTL using marker‐assisted selection (MAS). Two resistant by susceptible populations, both using ‘Ning7840’ as the source of resistance, were developed to examine the effect of the 3BS QTL in different genetic backgrounds. Data for scab resistance and simple sequence repeat (SSR) markers linked to the resistance QTL were analyzed in the F2:3 lines of one population and in the F3:4 lines of the other. Markers linked to the major QTL on chromosome 3BS in the original mapping population (‘Ning7840’/‘Clark’) were closely associated with scab resistance in both validation populations. Marker‐assisted selection for the QTL with the SSR markers combined with phenotypic selection was more effective than selection based solely on phenotypic evaluation in early generations. Marker‐assisted selection of the major QTL during the seedling stage plus phenotypic selection after flowering effectively identified scab resistant lines in this experiment. Near‐isogenic lines for this 3BS QTL were isolated from the F6 generation of the cross ‘Ning7840’/‘IL89‐7978’ based on two flanking SSR markers, Xgwm389 and Xbarc147. Based on these results, MAS for the major scab resistance QTL can improve selection efficiency and may facilitate stacking of scab resistance genes from different sources.  相似文献

Modern rice varieties that ushered in the green revolution brought about dramatic increase in rice production worldwide but at the cost of genetic diversity at the farmers’ fields. The wild species germplasm can be used for broadening the genetic base and improving productivity. Mining of alleles at productivity QTL from related wild species under simultaneous backcrossing and evaluation, accompanied by molecular marker analysis has emerged as an effective plant breeding strategy for utilization of wild species germplasm. In the present study, a limited backcross strategy was used to introgress QTL associated with yield and yield components from Oryza rufipogon (acc. IRGC 105491) to cultivated rice, O. sativa cv IR64. A set of 12 BC2F6 progenies, selected from among more than 100 BC2F5 progenies were evaluated for yield and yield components. For plant height, days to 50% flowering and tillers/plant, the introgression lines did not show any significant change compared to the recurrent parent IR64. For yield, 9 of the 12 introgression lines showed significantly higher yield (19–38%) than the recurrent parent IR64. Four of these lines originating from a common lineage showed higher yield due to increase in grain weight and another three also from a common lineage showed yield increase due to increase in grain number per panicle. For analyzing the introgression at molecular level all the 12 lines were analyzed for 259 polymorphic SSR markers. Of the total 259 SSR markers analyzed, only 18 (7.0%) showed introgression from O. rufipogon for chromosomes 1, 2, 3, 5, 6 and 11. Graphical genotypes have been prepared for each line and association between the introgression regions and the traits that increased yield is reported. Based on marker trait association it appears that some of the QTL are stable across the environments and genetic backgrounds and can be exploited universally.  相似文献

Fusarium head blight (FHB), or head scab, is an economically important disease of wheat (Triticum aestivum L.). In developing FHB-resistant soft winter wheat cultivars, breeders have relied on phenotypic selection, marker assisted selection (MAS), or a combination of the two. The objectives of this study were to estimate heritability of resistance in a resistant × susceptible cross and to simulate selection in order to determine the optimal combination of phenotypic and genotypic selection. F2 derived lines from the cross of KY93C-1238-17-2 (high yielding, susceptible) × VA01W-476 (resistant line with two exotic quantitative trait loci (QTL) and additional resistance) were grown under artificial inoculation in scab nurseries at Lexington (2007 and 2008) and Princeton (2008), KY. Visual symptoms were estimated on a 1–3 scale; percentage Fusarium damaged kernels (FDK), and deoxynivalenol (DON) concentration were measured. VA01W-476 contributed resistance alleles at two major QTL: Fhb1 and a QTL on chromosome 2DL, QFhs.nau-2DL. In this genetic background, the effect of QFhs.nau-2DL was more pronounced than that of Fhb1: 55 vs. 25% DON reduction and 40 vs. 32% FDK reduction. Genotypic selection based on both QTL was equivalent to phenotypic selection of the most resistant 28% of the population for DON and the most resistant 24% of the population for FDK. We propose that an initial round of phenotypic selection at moderate selection intensity will enrich the population with major QTL resistance alleles while maintaining variation at minor scab resistance loci and for other traits in general. Genotyping can then be used to extract lines whose phenotypic worth has been demonstrated and which are homozygous for resistance alleles at the major QTL.  相似文献

Flooding is one of the major hazards of rice production for the rainfed lowland rice ecosystem, and tolerant cultivars are urgently needed to help protect farmers from submergence damage. A quick and efficient strategy was implemented to introgress SUB1, a major QTL for submergence tolerance, into a rainfed lowland mega variety BR11 of Bangladesh by only two backcrosses and one selfing generation. In marker-assisted backcrossing (MABC), one tightly-linked simple sequence repeat (SSR) and two gene-based markers, four flanking SSR and 116 background SSR markers were used for foreground, recombinant and background selection, respectively, in backcrosses between a SUB1 donor IR40931-33-1-3-2 and BR11. BR11-Sub1, identified in a BC2F2 plant, possessed BR11 type SSR alleles on all fragments analyzed except the SUB1 QTL. The introgression size in BR11-Sub1 was 800 Kb indicating approximately 99.8% identity to BR11. BR11-Sub1 along with other introgression lines showed submergence tolerance similar to the tolerant parent. Yield, yield-component parameters and grain physico-chemical properties showed successful recovery of the BR11 traits in BR11-Sub1, with yield potential ranging from 5.2 to 5.6 t/ha, not significantly different from the recurrent parent mega variety BR11. Producing a large number (~1000) of backcross F1 plants was considered essential to achieve recombination on both sides of the gene, limiting linkage drag with only two backcrosses. A large number of background markers ensured proper recovery of the recurrent parent genome in the BC2F2 generation. The study demonstrates a rapid and highly precise strategy to introgress a major QTL by BC2F2 generation into a modern rice variety using an unadapted donor. The variety can replace BR11 on more than 2 million of ha in Bangladesh and provide major increases in rice production.  相似文献

Summary Selection among microgametophytes usually exploits variation in pollen grain germination. Studies of variation in pollen grain size in common bean (Phaseolus vulgaris L.) suggested that selection for size might lead to changes in sporophytic traits. To determine whether microgametophytic selection based on size would affect pollen grain size in subsequent generations or sporophytic traits that were correlated with pollen grain size, pollen grains from three crosses were separated into two size categories by sieving and then used to pollinate cv. Diacol Calima. Selection resulted in changes in pollen grain diameter for pollen from F1, F2 and F3 plants for all crosses. In vitro germination indicated no differences between vigor of large and small grains, but extraction and sieving reduced germinability. F1 seed from two of the crosses with size-selected pollen varied in weight according to pollen grain size, but in subsequent generations, the effect disappeared. Both size categories of selected pollen resulted in F2 progeny with reduced numbers of seeds per pod as compared to controls, suggesting that the size selection process may have resulted in indirect selection for traits reducing seed set. The overall results suggested that genes determining pollen grain size in bean have little or no effect on sporophytic traits such as seed size and seed yield.  相似文献

Many important apple (Malus × domestica Borkh.) fruit quality traits are regulated by multiple genes, and more information about quantitative trait loci (QTLs) for these traits is required for marker-assisted selection. In this study, we constructed genetic linkage maps of the Japanese apple cultivars ‘Orin’ and ‘Akane’ using F1 seedlings derived from a cross between these cultivars. The ‘Orin’ map consisted of 251 loci covering 17 linkage groups (LGs; total length 1095.3 cM), and the ‘Akane’ map consisted of 291 loci covering 18 LGs (total length 1098.2 cM). We performed QTL analysis for 16 important traits, and found that four QTLs related to harvest time explained about 70% of genetic variation, and these will be useful for marker-assisted selection. The QTL for early harvest time in LG15 was located very close to the QTL for preharvest fruit drop. The QTL for skin color depth was located around the position of MYB1 in LG9, which suggested that alleles harbored by ‘Akane’ are regulating red color depth with different degrees of effect. We also analyzed soluble solids and sugar component contents, and found that a QTL for soluble solids content in LG16 could be explained by the amount of sorbitol and fructose.  相似文献

Phenotypic and genetic evaluation of morphological traits associated with herbage biomass production was undertaken in a perennial ryegrass (Lolium perenne L.) biparental F1 mapping population (n = 200) with parent plants from cultivars ‘Grasslands Impact’ and ‘Grasslands Samson’. Morphological traits measured on three clonal replicates of the parental genotypes and 200 F1 progeny in a glasshouse in two separate trials (autumn and spring) included: dry weight (DW), leaf elongation rate (LER), initial tiller number (TNs), final tiller number (TNe), site filling (Fs), tiller weight (TW), leaf lamina length, leaf tip and ligule appearance rates (ALf, ALg) and leaf elongation duration (LED). Principal component analysis of patterns of trait association identified negative correlation between TNs or TNe, and TW as the primary basis for morphological difference and indicated that either high LER or long LED could reduce TN. Plants with higher LER tended to have increased DW. Quantitative trait loci (QTL) were detected on all seven linkage groups (LG) of a perennial ryegrass linkage map for all but three traits. A total of 61 QTL were identified, many of which clustered at 15 shared genome locations. Significant genotype by environment effects were encountered, evidenced both by variation between experiments in genotype rankings and by a general lack of commonality for QTL for the same traits in the different experiments. Only five QTL, for ALf, ALg and TN, were conserved between autumn and spring trials. A QTL for TN and DW on LG6 is a strong candidate for application of MAS in future plant improvement work and was found to be co-linear with QTL for equivalent traits reported on chromosome 2 in rice.  相似文献
