Strain 213M0 was selected with productivity of a bacteriocin‐like inhibitory substance (BLIS) among 235 strains of lactic acid bacteria (LAB) isolated from Mongolian fermented milk ‘airag’. Strain 213M0 was species‐identified as Leuconostoc mesenteroides subsp. dextranicum by morphological observation, carbohydrate fermentation profiling and sequencing the 16S rRNA gene. Incubation temperature proper to produce the BLIS was 25°C rather than 30 and 37°C, and the production actively proceeded during the exponential growth phase of the producer cells. Antibacterial effect of BLIS 213M0 was limited to all nine strains of Listeria sp. bacteria and seven strains of LAB cocci among 53 tested strains, which corresponds to a typical feature of the class IIa pediocin‐like bacteriocins. BLIS 213M0 was not inactivated in every broad pH range solution (pH 2.0‐11.0), and was stable against storage at 25°C for 1 week and heating at 121°C for 15 min under pH 4.5. Peptide frame of BLIS 213M0 was confirmed by inactivation with some peptidases, and then its molecular weight was estimated to be 2.6‐3.0 kDa using an in situ activity assay following sodium dodecyl sulfate polyacrylamide gel electrophoresis. The estimated size was different from the other Leuconostoc bacteriocins already reported. These results suggest that BLIS 213M0 would be a novel listericidal bacteriocin.  相似文献

Gassericin A (GA) is a circular bacteriocin produced by Lactobacillus gasseri LA39. In this study, GA‐containing concentrate was prepared using a cross‐flow membrane filtration device (30 kDa cut‐off) from the culture supernatant of Lb. gasseri LA39 cultivated in a cheese whey‐based food‐grade medium. The bacteriocin activity titer in the concentrate was 16 times as high as that of the culture supernatant and was completely maintained through each incubation at 4°C for 3 months, 37°C for 2 months, 60°C for 5 h, and 100°C for 30 min. The GA‐containing concentrate was used with glycine powder to make custard creams, and then four representative strains of custard cream spoilage bacteria (Bacillus cereus, Lactococcus lactis subsp. lactis, Achromobacter denitrificans and Pseudomonas fluorescens) were individually inoculated at c. 103 colony forming units/g in the custard creams. Throughout 30 days of incubation at 30°C, all of the inoculated bacteria were completely inhibited by the combination of 5% (w/w) of the GA‐containing concentrate and 0.5% (w/w) glycine. This is the first highly practical application of GA to foods as a biopreservative, and the concentration method and the bacteriocin concentrate would contribute to biopreservation of several foods.  相似文献

Fourteen strains of Bdellovibrio‐like organisms were isolated from cultured fish ponds using Aeromonas hydrophila J‐1 as host, one of them formed large plaques after 48 h of incubation at 28°C on a double layer plate, designated as Bdellovibrio C‐1. The Bdellovibrio was confirmed by electron microscopy and PCR amplification of Bdellovibrio‐specific 16S rDNA. The optimum temperature for the growth of BdC‐1 was between 15–37°C and with optimal activity at temperatures of 25–30°C. The ability of BdC‐1 to lyse A. hydrophila was similar in the pH range of 6.5 to 8.5. It can lyse 23 Gram‐negative bacterial strains comprising three genera of fish pathogens and one strain of Escherichia coli but cannot lyse Gram‐positive bacteria such as Bacillus subtillis and Staphylococcus aureus. Immersion of fish in water containing different concentrations of BdC‐1 was used in protection against an experimental infection of A. hydrophila J‐1. Results show that the mortality of groups immersed with BdC‐1 was lower than the group without BdC‐1. These results suggest that it may be possible to use Bdellovibrio to control the disease caused by A. hydrophila.  相似文献

Anaerobic digestion is considered as a promising method to manage animal waste with antibiotic‐resistant bacteria. Current research was conducted to investigate the survival of multidrug‐resistant bacteria (MDRB) resistant to three groups of antibiotics: (i) cefazolin, neomycin, vancomycin, kanamycin (group 1); (ii) penicillin, oxytetracycline, ampicillin, streptomycin (group 2); and (iii) cefazolin, neomycin, vancomycin, kanamycin, penicillin, oxytetracycline, ampicillin, streptomycin (group 3), in anaerobic digestion of dairy manure and co‐digestion of dairy manure and waste milk at 37°C and 55°C for 22 days, respectively. The population densities of three groups of MDRB on peptone, tryptone, yeast and glucose agar plates incubated at 30°C for 7 days before and after digestion showed 100% destruction in both digestates at thermophilic temperature. Overall reduction of more than 90% of three groups of MDRB was observed in mesophilic digestion with no significant differences (P > 0.05) between manure and milk mixture. Co‐digestion of dairy manure and waste milk always produced significantly (P < 0.05) higher total gas and methane gas than digestion of manure alone at both temperatures. Gas production in each case was significantly (P < 0.05) higher in thermophilic digestion than in mesophilic digestion. The results demonstrate that thermophilic co‐digestion of dairy manure and waste milk offers more benefits in terms of the environment and economy.  相似文献

The purification and characterization of a bacteriocin produced by Leuconostoc mesenteroides strain 406 that was isolated from traditional Mongolian fermented mare's milk, airag, were carried out. Leuconostoc mesenteroides strain 406 was identified on the basis of its morphological and biochemical characteristics and carbohydrate fermentation profile and by API 50 CH kit and 16S ribosomal DNA analyses. The neutral‐pH cell‐free supernatant of this bacterium inhibited the growth of several lactic acid bacteria and food spoilage and pathogenic organisms, including Listeria monocytogenes and Clostridium botulinum. The bacteriocin was heat‐stable and not sensitive to acid and alkaline conditions, but was sensitive to several proteolytic enzymes such as pepsin, pronase E, proteinase K, trypsin, and α‐chymotrypsin, but not catalase. Optimum bacteriocin production (4000 activity units/mL) was achieved when the strain was cultured at 25°C for 24–36 h in Man Rogosa Sharpe medium. The bacteriocin was partially purified by ammonium sulfate precipitation (80% saturation), dialysis (cut‐off MW: 1000), and gel filtration chromatography. Sodium dodecyl sulfate‐polyacrylamide gel electrophoresis revealed that the bacteriocin had a molecular weight of approximately 3.3 kDa. To our knowledge, this is the first report of the isolation of a bacteriocin‐producing Leuconostoc strain from airag. An application to fermented milks would be desired.  相似文献

Hydrogen sulfide causes offensive odors. We attempted to isolate sulfur oxidizing bacteria (SOB) from cattle manure compost. A most probable number assay could not detect SOB by using a Cha medium which had been applied in order to isolate the SOB from active sewage. Cultivation using a Cha plate medium revealed 5.75 × 107 colony forming unit/g of bacteria. A single strain of SOB was isolated from a colony formed on the plates and identified as Halothiobacillus neapolitanus. This is the first report that H. neapolitanus has been found in cattle manure compost.  相似文献

To determine the effect of bacteria on the development of the ripening flavor of cured meat, pork loins were cured with pickles containing 8 or 16% sodium chloride at 0 and 8°C. The bacterial flora, total free amino acid content and total free fatty acid content in the cured loins, sensory properties of cooked pork loins and the relationships between the proteolytic and lipolytic activities of isolates and curing conditions were investigated. Desirable bacteria, including salt‐tolerant bacteria and lactic acid bacteria, were predominant under the condition of 8% sodium chloride in the pickles and a curing temperature of 8°C. The total free amino acid content and total free fatty acid content at a curing temperature of 8°C were higher than those at 0°C. Cooked pork loins cured in pickles containing 8% sodium chloride at a temperature of 8°C for more than 7 days were preferred in terms of color, flavor and taste. Before the curing procedure, gram‐negative bacteria (Vibrio, Acinetobacter, Pseudomonas and Enterobacteriaceae) were predominant in the pork loins. During the curing period, the numbers of viable gram‐positive bacteria (Micrococcus, Staphylococcus and Pediococcus) increased and the numbers of viable gram‐negative bacteria decreased. Strains of Micrococcus and Staphylococcus with proteolytic and/or lipolytic activities in cured meat also increased during the curing period and were more predominant in the pork loins cured in pickles containing 8% sodium chloride and at a curing temperature of 8°C than in pork loins cured in pickles containing 16% sodium chloride and at a curing temperature of 0°C. The actions of these bacteria were thought to be important factors affecting the flavor of the cured pork. Micrococcus and Staphylococcus with proteolytic activity might contribute to the development of the ripening flavor of ham. The results of the present study together with the results of further investigations on the relationships of the enzyme activities of Micrococcus and Staphylococcus in cured meat with a preferable flavor would be useful for establishing a novel effective method for using bacteria to produce ham of a high quality.  相似文献

To investigate the effects of inoculants and environmental temperature on fermentation quality and bacterial diversity of alfalfa silage, first‐cut alfalfa was ensiled with or without two screened lactic acid bacteria (LAB) strains, Lactobacillus plantarum, LP, and Lactobacillus casei, LC. Each treatment was divided into three parts and stored at 20°C, 30°C, 40°C, respectively. After 60 days ensiling, fermentation characteristics were measured and bacterial diversity was investigated by 16S ribosomal RNA gene sequencing using Illumina MiSeq platform. LP and LC decreased pH, coliform bacteria counts and increased lactic acid content at 20°C, and the two strains decreased pH, ammonia‐N concentration, coliform bacteria counts at 30°C. When the environmental temperature was 40°C, silage treated with LC showed lower LAB and coliform bacteria counts and higher lactic acid content than the untreated and LP treated silages. Butyric acid mainly appeared in silages stored at 40°C. The relative abundance of Lactobacillus in alfalfa silages stored at 20°C and 30°C was highest and increased after LP and LC were added. Garciella was another dominant genus in silages stored at 40°C. In conclusion, LP and LC improved fermentation quality of alfalfa silage by increasing Lactobacillus proportions at 20°C and 30°C; ensiling alfalfa at 40°C was difficult because of Garciella.  相似文献

Methane production from co‐digestion of dairy manure and waste milk, milk from cows treated with antibiotics for mastitis, was tested in a 2 × 4 factorial design. Four different waste milk percentages (w/w): 0% (SM), 10% (SMWM10), 20% (SMWM20) and 30% (SMWM30), were tested with two slurry percentages (w/w): 50% (A) and 25% (B) and the rest being manure at 55°C for 12 days in batch digesters. The results analyzed using a Gompertz model showed SMWM10 produced the highest methane production potential (Pm)/g volatile solids added followed by SM in both A and B. This Pm of SMWM10 in A and B was statistically non‐significant (P > 0.05). More than 96% of cefazolin‐resistant bacteria and 100% of multi‐drug‐resistant bacteria reductions were observed in all the treatments. Inclusion of waste milk at 10% in single stage digester enhances the methane production from dairy manure and could offer added benefit of waste milk treatment and disposal.  相似文献

Four lactic acid bacteria (LAB) strains isolated from straw silages on the Tibetan Plateau were characterized, and their effects on the fermentation quality of Italian ryegrass (Lolium multiflorum Lam.) at different temperatures (10°C, 15°C and 25°C) were studied. These LAB isolates were evaluated using the acids production ability test, morphological observation, Gram staining, physiological, biochemical and acid tolerance tests. All the isolates (M1, LM8, LO7 and LOG9) could grow at 5‐20°C, pH 3.5‐7.0 and NaCl (3.0%, 6.5%). Strains M1, LM8, LO7 and LOG9 were identified as Lactobacillus plantarum, L. coryniformis, Pediococcus pentosaceus and P. acidilactici, respectively, by sequencing 16S ribosomal DNA. The four isolates were added to Italian ryegrass for ensiling for 30 days at various temperatures. Compared with the corresponding control, inoculating with isolates M1, LM8 and LO7 could improve the silage quality of Italian ryegrass at low temperatures, indicated by significantly (< 0.05) higher lactic acid (LA) contents and ratios of lactic acid/acetic acid (LA/AA), and significantly (< 0.05) lower pH and ammonia nitrogen/total nitrogen (AN/TN). Compared with other isolates, LM8 performed better at 10°C and 15°C, indicated by the higher (< 0.05) LA content and ratio of LA/AA, and the lower (< 0.05) pH and AN/TN.  相似文献

In order to clarify the ensiling characteristics of stylo (Stylosanthes guianensis Swartz), the effects of wilting (no wilting, light wilting and heavy wilting) and storage temperatures (10°C, 20°C, 30°C and 40°C) on the fermentation quality and aerobic stability of stylo silage were investigated. Wilting had no significant influence on the contents of crude protein, ether extract and acid detergent fiber, and numbers of lactic acid bacteria, aerobic bacteria, yeasts and mold (P > 0.05). Heavy wilted material, wilted for 12 h, had higher neutral detergent fiber content and lower water‐soluble carbohydrate content than unwilted and light wilted materials (P < 0.05). Wilting and storage temperatures had significant effects on pH value, acetic acid, butyric acid and NH3‐N contents of stylo silage (P < 0.01 or P < 0.05). Wilting tended to reduce acetic acid and NH3‐N contents and improve the fermentation quality of stylo silage. In all the silages, no wilting silage ensiled at 30°C had the highest butyric acid content (P < 0.05). High temperature of 40°C markedly restricted the growth of lactic acid bacteria and aerobic bacteria in silage, irrespective of wilting. The wilted silage or silage stored at low temperature had poor aerobic stability.  相似文献

The efficacy of enterocoliticin, a phage tail‐like bacteriocin, as antimicrobial compound against infections with pathogenic Yersinia enterocolitica serotype O3 strains was assessed. In cell cultures, which were infected with the Y. enterocolitica strains 13 169 or 6471/76, bactericidal activity of enterocoliticin was found for bacteria adhering to the surface of eukaryotic cells, whereas bacteria, which had invaded the eukaryotic cells, were not accessible to the bacteriocin. The interaction of enterocoliticin with Y. enterocolitica was further examined in animals. Female BALB/c mice were experimentally infected with the two Y. enterocolitica strains and enterocoliticin was applied as antimicrobial compound by the oral route. Experimental variations concerning the infectious doses of the Y. enterocolitica strains and the time points of application of the bacteriocin were investigated. The increase of the Yersinia CFU titre in animals was retarded at time points shortly after the application of enterocoliticin indicating that the particles were effective on recently introduced Yersinia. The repeated application of enterocoliticin, however, did not prevent the colonization of the gastrointestinal tract by Yersinia.  相似文献

Gassericin A, a bacteriocin produced by Lactobacillus gasseri LA39, has a cyclic structure linking N‐ and C‐terminal amino acids. Gassericin A was expressed in Escherichia coli JM109 as a biotinylated fusion protein on the basis of the DNA sequence of mature bacteriocin. A positive clone accumulated the bacteriocin, with no activity, as a soluble fusion protein in the cytoplasm. After release of an N‐terminal tag with factor Xa protease, gassericin A was converted into an active peptide having N‐ and C‐termini. The total amount of purified bacteriocins (expressed and native) was 480 µg/L and 370 µg/L, respectively. However, the specific activity of expressed gassericin A was 15 AU/mg lower than that of native bacteriocin (2600 AU/mg). Although the actual Mr (molecular weight) of the expressed bacteriocin should be 5666, the peptide showed the same mobility (Mr 3800) in sodium dodecylsulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE) as native cyclic gassericin A, suggesting that the expressed peptide retains compact folding of the molecule similar to that of native gassericin A.  相似文献

Experiments carried out in a turkey processing plant showed that there was a fivefold increase in the number of psychrophilic bacteria present after holding eviscerated carcasses in static slush ice tanks for 24 hr. The predominant bacteria in the 24 hr chill tanks were strains of Flavobacterium, Cytophaga and Pseudomonas.

The carcasses were wrapped in heat‐shrunk‐oxygen‐impermeable film and after freezing packed in individual boxes and held at ‐20° C. The storage life at 1°, 10° and 20° C. was determined together with the time taken for the frozen carcasses to equilibrate to these temperatures. It was found that at 1° C. the carcasses kept for about 3 weeks, but at 10° C. they spoiled within 7 days and at 20° G. within 3 days.

An analysis of the spoilage flora showed that although Pseudomonas strains predominated on the turkeys stored at 1° C. fewer were isolated from turkeys stored at 10° C. and 20° C. At these two temperatures an organism identified as Enterobacter liquefaciens predominated together with atypical lactobacilli resembling unidentified strains previously described by Thornley and Sharpe (1959).  相似文献

A collection of 119 cultures of bacteria was assembled from eggs which had rotted on the premises of producers. All but 13 of the isolates were Gram‐negative. These were characterised in detail and the data used (i) to devise a determinative key that permits rot‐producing bacteria to be identified at generic level, and (it) to establish systems whereby identification at species level can be achieved with the minimum of tests. With the use of these schemes the following were recognised: Pseudomonas fluorescens, Pseudomonas maltophilia, Aeromonas liquefaciens, and members of Flavobacterium, Cytophaga, Proteus, Escherichia, Hafnia and Aerobacter. The literature dealing with the microbiology of rotten eggs was reviewed with the object of assessing the incidence of the organisms listed above in rotten eggs of commercial origin.  相似文献

Flavobacterium columnare and F. psychrophilum are major fish pathogens that cause diseases that may require antimicrobial therapy. Choice of appropriate treatment is dependent upon determining the antimicrobial susceptibility of isolates. Therefore we optimized methods for broth microdilution testing of F. columnare and F. psychrophilum to facilitate standardizing an antimicrobial susceptibility test. We developed adaptations to make reproducible broth inoculums and confirmed the proper incubation time and media composition. We tested the stability of potential quality-control bacteria and compared test results between different operators. Log phase occurred at 48 h for F. columnare and 72–96 h for F. psychrophilum, confirming the test should be incubated at 28°C for approximately 48 h and at 18°C for approximately 96 h, respectively. The most consistent susceptibility results were achieved with plain, 4-g/L, dilute Mueller–Hinton broth supplemented with dilute calcium and magnesium. Supplementing the broth with horse serum did not improve growth. The quality-control strains, Escherichia coli ATCC 25922 and Aeromonas salmonicida subsp. salmonicida ATCC 33658, yielded stable minimal inhibitory concentrations (MIC) against all seven antimicrobials tested after 30 passes at 28°C and 15 passes at 18°C. In comparison tests, most MICs of the isolates agreed 100% within one drug dilution for ampicillin, florfenicol, and oxytetracycline. The agreement was lower with the ormetoprim–sulfdimethoxine combination, but there was at least 75% agreement for all but one isolate. These experiments have provided methods to help standardize antimicrobial susceptibility testing of these nutritionally fastidious aquatic bacteria.

Changes in mastitis‐causing pathogens, pH and water content in composted manure solids (CMS) prepared from digested slurry were evaluated during turning at 2‐day intervals for 8 days (C1–C4). The numbers of streptococci, coagulase‐negative staphylococci and coliforms were 2.6 × 101, 1.7 × 102 and 1.0 × 101 colony‐forming units (cfu)/g in CMS (C4) (summer), and these counts were markedly lower (< 0.05) than those in CMS (C0 and C1). The bacterial counts ranged from 101 to 1.7 × 102 cfu/g in CMS (C4) (summer) and were within approved levels, <1 × 106 cfu/g, indicating a minimal mastitis risk. The temperatures in CMS (C1–C4) increased to 63°C–74°C in summer and 67°C–70°C in winter. The mean pH values in CMS (C0–C4) were 9.2 in summer and 8.7 in winter, and water contents ranged from 61.7% to 69.6% in summer and 73.2% to 66.2% in winter. The significant decrease of pathogenic bacteria in CMS appears to be closely related to temperature >63°C for 8 days, pH 8.7–9.2, and water content 62% to 73%. This study demonstrates that prepared CMS has value as a recycled material with the potential to alleviate udder health issues in dairy cows.  相似文献

This study evaluated the effects of hinokitiol (a natural antibacterial compound extracted from Thujopsis dolabrata var. hondai) and an organic acid mixture (citrate content 50%) on ruminal fermentation. Antibacterial properties were examined by measuring minimal inhibitory concentration. Hinokitiol at 1.56 µg/mL or an organic acid mixture at 1600 µg/mL inhibited Streptococcus bovis growth. The combination of 0.78 µg/mL hinokitiol and 200 µg/mL of an organic acid mixture also inhibited S. bovis growth. Both hinokitiol and the hinokitiol and an organic acid mixture combination showed strong antibacterial properties on Gram‐positive bacteria such as S. bovis, but relatively weak antibacterial activities on Gram‐negative bacteria such as Megasphaera elsdenii. Three ruminally cannulated heifers were fed a bloat‐producing diet containing barley, pelleted alfalfa meal, soybean meal and salt without long‐cut roughage to investigate the ruminal characteristics in vivo. Feeding to heifers a bloat‐producing diet containing 7.8 mg/kg hinokitiol and 0.2% of an organic acid mixture significantly decreased the increase in stable ingesta volume. Hinokitiol or an organic acid mixture did not affect ruminal volatile fatty acids, protozoa and bacteria. These results suggest that a combination of hinokitiol and an organic acid mixture might reduce frothy bloat in cattle fed high‐grain diets.  相似文献

The present experiment was performed to determine some functional properties of yak butter lipids such as lipid class composition; conjugated linoleic acid (CLA) composition, differential scanning calorimetric (DSC) analysis, tyrosinase inhibition activity and antioxidant property. Yak butter lipids composition contained 98% triacylglycerols, 0.9% free fatty acids, 0.32% free sterols and 0.27% phospholipids. The CLA content in yak butter was 2.5% and the major portion was of cis‐9 and trans‐11 (90%). The DSC analysis of yak butter lipids showed a similar path for transition temperature as cow butter lipids, although the enthalpy of yak lipid was higher (40.0 mJ/mg) compared to cow butter lipids (32.0 mJ/mg). Melting point of yak butter was observed at 41°C. Yak butter with lactic acid, NaCl, citric acid and ascorbic acid showed pronounced tyrosinase inhibition activity. Vegetable oils blended with yak butter have extended the oxidation induction time.  相似文献

To elucidate the characteristics of ear corn residue as a bulking agent, the composting process using this residue was compared with processes using three other conventional materials such as sawdust, wheat straw and rice husk, employing a bench‐scale composting reactor. As evaluated via biochemical oxygen demand (BOD), ear corn residue contains 3.3 and 2.0 times more easily digestible materials than sawdust and rice husk, respectively. In addition, mixing ear corn residue with manure resulted in reduced bulk density, which was the same as that of wheat straw and was 0.58 and 0.67 times lower than that of sawdust and a rice husk mixture, respectively. To evaluate temperature generation during the composting process, the maximum temperature and area under the temperature curve (AUCTEMP) were compared among the mixed composts of four bulking agents. Maximum temperature (54.3°C) as well as AUCTEMP (7310°C●h) of ear corn residue were significantly higher than those of sawdust and rice husk (P < 0.05), and they are similar to that of wheat straw mixed compost. Along with the value of AUCTEMP, the highest organic matter losses of 31.1% were observed in ear corn residue mixed compost, followed by wheat straw, saw dust and rice husk.  相似文献
