The transport of [(3)H]putrescine is demonstrated by autoradiography in the retino-tectal tract of Brachydanio rerio embryos. Transport of [(3)H]putrescine appears to be more rapid than that of tritium-labeled protein and is not inhibited by a colchicine effect on axonal neurotubules as is protein transport. The radioactivity transported to the brain is found, on electrophoresis, in the putrescine fraction.  相似文献

钾磷水平变化对辽杨中多胺含量有显著的影响。随着钾水平的降低,叶和根中腐胺含量大幅度增加,尤其是根中更为显著。它们的最高含量分别是同一时期对照的23倍和85倍。然而,磷水平的变化对叶和根中腐胺含量没有明显的影响。研究表明,钾对腐胺代谢有着灵敏的调节作用,腐胺含量可作为辽杨需钾的生化指标。缺钾和缺磷都使叶和根中精胺含量明显下降,叶中更为显著。在所有处理中,精胺含量与其相应部位的生物量积累有一定的相关性,表明精胺与生长之间可能存在着密切联系。  相似文献

The biological response modifier human beta-interferon had pronounced antigrowth effects on various histologic types of human brain tumor cells but no effects on a nontransformed cell line, MRC-5. The cultures of brain tumor cells showed severe alterations indicative of cell injury and death after exposure to beta-interferon for 2 to 6 days. Similar results were obtained with cells freshly explanted from human brain tumors. The results indicate that it may be possible to use fresh, explanted tumor tissue to identify patients who might benefit from therapy with beta-interferon.  相似文献

We report the development of a pseudorabies virus that can be used for retrograde tracing from selected neurons. This virus encodes a green fluorescent protein marker and replicates only in neurons that express the Cre recombinase and in neurons in synaptic contact with the originally infected cells. The virus was injected into the arcuate nucleus of mice that express Cre only in those neurons that express neuropeptide Y or the leptin receptor. Sectioning of the brains revealed that these neurons receive inputs from neurons in other regions of the hypothalamus, as well as the amygdala, cortex, and other brain regions. These data suggest that higher cortical centers modulate leptin signaling in the hypothalamus. This method of neural tracing may prove useful in studies of other complex neural circuits.  相似文献

Six structural homologs of spermidine and five of its precursor, putrescine, were studied for their ability to prevent cytostasis of cultured L1210 leukemia cells induced by alpha-difluoromethylornithine (DFMO), a specific inhibitor of putrescine biosynthesis. High-performance liquid chromatography and competition studies with spermidine indicated that the homologs, which vary in the length of the carbon chain separating the amines, penetrated the cells. The structural specificity of the spermidine carrier was defined. Three of the six spermidine homologs supported cell growth during a 48-hour incubation in the presence of DFMO, indicating that a two-carbon extension of spermidine structure was tolerated for biological function. Two of the five putrescine homologs supported growth after being converted by the cells to their respective spermidine homologs. The central nitrogen of spermidine appears to be essential for function since diamines of chain length comparable to that of spermidine did not prevent DFMO cytostasis. No more than 15 percent of the spermidine normally present in L1210 cells was required for cell proliferation in the presence of DFMO.  相似文献

将猿猴病毒抗原蛋白(SV40Tag)片段克隆进脑部特异表达载体pMM279中,通过显微注射法制作转基因小鼠。采用PCR和Southern blotting检测目的基因整合,并通过RT-PCR检测目的基因的表达。结果表明:共出生29只仔鼠,经PCR检测出3只阳性,Southern blotting检测出2只阳性。RT-PCR检测到SV40Tag仅在前脑部皮层和海马表达。成功获得的脑部特异表达SV40Tag转基因小鼠模型,为SV40Tag的致病机制及脑肿瘤的治疗等研究提供工具。  相似文献

Examination of human bladder, head and neck, and lung primary tumors revealed a high frequency of mitochondrial DNA (mtDNA) mutations. The majority of these somatic mutations were homoplasmic in nature, indicating that the mutant mtDNA became dominant in tumor cells. The mutated mtDNA was readily detectable in paired bodily fluids from each type of cancer and was 19 to 220 times as abundant as mutated nuclear p53 DNA. By virtue of their clonal nature and high copy number, mitochondrial mutations may provide a powerful molecular marker for noninvasive detection of cancer.  相似文献

运用实时荧光PCR定量检测了38例犬乳腺肿瘤病例(包括15例良性乳腺肿瘤和23例恶性乳腺肿瘤)、4例正常乳腺组织.结果表明:在犬恶性乳腺肿瘤组织中,VEGF-C表达量明显高于良性乳腺肿瘤和正常乳腺组织,两者差异极显著(P<0.001);在伴有淋巴结转移的犬乳腺癌组织中,VEGF-C基因表达量明显高于没有发现转移的乳腺癌组织,两者差异极显著(P<0.01),但VEGF-C的表达量与肿瘤的大小和发病动物年龄无关.由此表明,VEGF-C在犬乳腺癌组织中的表达量远高于其在正常乳腺和良性乳腺肿瘤组织中的表达,且与淋巴结是否发生转移相关,因此VEGF-C有可能作为乳腺癌转移、复发的预测指标之一.  相似文献

用达到电泳纯的大型溞(Daphniamagna)胆碱酯酶(cholinesterase,ChE)免疫BALB/c小鼠,得到效价达1∶8000的鼠抗大型溞ChE多克隆抗血清;通过优化抗原抗体反应条件,建立定量分析大型溞ChE含量的间接非竞争酶联免疫吸附法(indirectandnon‐competitiveenzyme‐linkedimmunosorbentassay).结果表明:此方法检测灵敏度为24ng爛mL-1;经免疫交叉反应鉴定,所获抗血清与5、10μg爛mL-1标准蛋白质和其他生物如花翅摇蚊、尖额溞、斑马鱼(脑、脑除外的整体)、家蚕、意大利蜜蜂、赤子爱胜蚯蚓、非洲爪蟾(蝌蚪)等来源的ChE交叉反应率分别为:<0畅25%、<0畅25%、 2畅13%、 10畅19%、3畅88%、 3畅56%、 2畅81%、 3畅93%、 5畅00%和2畅75%,具较高的特异性.说明用本试验得到的抗血清建立的间接非竞争酶联免疫吸附法可用于大型溞体内ChE含量的测定,以利于在农药暴露环境下大型溞体内ChE活性的精确检测.  相似文献

目的了解增殖细胞核抗原(PCNA)和CD44V6在喉癌组织中的表达。方法采用免疫组织化学S-P法检测46例喉癌组织中PCNA和CD44V6的表达,并与46例中的17例癌旁组织作对照分析。结果46例喉癌组织中PCNA和CD44V6的阳性表达率分别为60.9%、71.8%,与17例癌旁组织的PCNA、CD44V6阳性率比较,差异均有统计学意义(P<0.01)。不同性别、年龄、临床分型患者喉癌组织中的PCNA、CD44V6阳性表达率差异无统计学意义(P>0.05);但有或无淋巴结转移患者喉癌组织中的PCNA(CD44V6)阳性表达率差异有统计学意义(P<0.05),组织病理分级为Ⅰ级及Ⅲ级的PCNA阳性表达差异有统计学意义,而病理分级不同的喉癌组织的CD44V6阳性表达率差异无统计学意义(P>0.05)。结论PCNA和CD44V6在喉癌的发生发展中起一定的作用,有望作为判断喉癌恶性程度和预后的参考指标。  相似文献

alpha-Difluoromethylornithine (RMI 71,782), a specific irreversible inhibitor of the first step in polyamine biosynthesis, that is, the formation of putrescine from ornithine by ornithine decarboxylase, cures mice infected with a virulent, rodent-passaged strain of Trypanosoma brucei brucei. This parasite is closely related to the trypanosomes that cause human sleeping sickness. The drug, which is remarkably nontoxic, was effective when administered in drinking water or by intubation. The ability of the compound to inhibit ornithine decarboxylase in vitro was demonstrated by the reduced amounts of putrescine synthesized from tritiated ornithine in Trypanosoma brucei suspensions. These observations direct attention to polyamine metabolism as a target for chemotherapy of parasitic diseases.  相似文献

采用高效液相色谱法(HPLC)对天目铁木不同时期的雄花序3种内源多胺—腐胺(Put)、精胺(Spm)、亚精胺(Spd)进行测定分析。结果发现:(1)适宜的苯甲酰化反应条件为37℃、30 min。(2)将色谱条件设定为检测波长254 nm,流动相配比甲醇︰水为60:40,流速0.6 mL.min-1,柱温30℃,进样量5μL。在该条件下内源多胺出峰和分离效果最佳。(3)在采样期间,雄花序中Spd含量最高,Spm次之,Put最少,且3种内源多胺含量均呈先升高后降低趋势,变幅明显。采用优化的HPLC法可有效快速地测定天目铁木雄花序的内源腐胺、精胺和亚精胺,并初步揭示了雄花序发育过程中3种内源多胺的变化趋势。  相似文献

The tumor promoters 12-O-tetradecanoyl-phorbol-13-acetate and teleocidin markedly enhanced the transformation of C3H 10T1/2 mouse fibroblasts when these cells were transfected with the cloned human bladder cancer c-rasH oncogene. Transfection studies with the drug resistance marker gpt and time course studies indicate that this enhancement is not simply an effect on the process of DNA transfection. These findings, together with parallel studies with NIH 3T3 fibroblasts, also indicate that the competence of animal cells for DNA transfection is a function of the recipient cell line, the transfected marker, and the growth conditions. Our findings suggest that during multistage carcinogenesis tumor promoters may complement the function of activated cellular oncogenes.  相似文献

盐胁迫下刺槐幼苗的生长明显受抑,相对含水量(RWC)降低,质膜透性变大,超氧物歧化酶(SOD)和过氧化氢酶(CAT)活性变小,同时大量积累游离脯氨酸和腐胺含量。外源ABA能缓和盐胁迫对生长的抑制,减少质膜透性,提高RWC,SOD和CAT活性,脯氨酸量被继续增加,但腐胺量却被减少,表明腐胺量的变化与幼苗的水分状况和遭受胁迫的程度一致,腐胺积累的多少可作为植物遭受胁迫程度的生理指标。  相似文献

【目的】研究腐胺和脯氨酸对哺乳期仔猪空肠绒毛-隐窝轴上皮细胞的多胺(腐胺、亚精胺和精胺)代谢、Wnt信号通路关键基因的mRNA相对表达量的影响。【方法】选取18头0日龄的刚出生的三元杂交(长白×大白×杜洛克)仔猪,随机配对分成3组,每组6个重复,每个重复1头猪,对照组,腐胺组和脯氨酸组,分别灌喂等体积的生理盐水,5 mg·kg-1体重添加剂量的腐胺和25 mg·kg-1体重添加剂量的脯氨酸,到14日龄断奶,断奶后3 d屠宰,分离空肠绒毛-隐窝轴的3个不同分化程度(绒毛顶端,绒毛中段和隐窝)的细胞,分别为F1,F2,F3。高效液相色谱法测定F1,F2,F3中的多胺浓度,RT-PCR测定多胺代谢途径中的相关基因以及Wnt信号通路中关键基因的mRNA相对表达量。【结果】1、在绒毛顶端细胞F1中,与对照组相比,脯氨酸组的腐胺、亚精胺、精胺的浓度均显著增加(P<0.05),而腐胺组的多胺均无显著差异(P>0.05);在绒毛中段细胞F2中,除了腐胺的含量无显著差异外,脯氨酸组和腐胺组的亚精胺、精胺的浓度均显著高于对照组(P<0.05),且脯氨酸组的亚精胺和精胺的浓度均显著高于腐胺组(P<0.05);在隐窝底端细胞F3中,腐胺组和脯氨酸组的腐胺、亚精胺、精胺浓度与对照组的浓度差异均不显著(P>0.05)。2、鸟氨酸脱羧酶(ODC)基因在绒毛顶端上皮细胞F1中的mRNA表达量,腐胺组显著高于脯氨酸组和对照组(P<0.05);精氨酸酶(arginase)在F2中的mRNA表达量,脯氨酸组显著高于腐胺组和对照组(P<0.05);sFRP3在绒毛中段上皮细胞F2中的mRNA表达量,脯氨酸组的显著高于腐胺组(P>0.05);sFRP4在隐窝上皮细胞F3中的mRNA表达量,腐胺组显著高于对照组和脯氨酸组(P<0.05)。【结论】添加了外源腐胺和脯氨酸,促进了哺乳期仔猪的空肠绒毛-隐窝轴上皮细胞的分化细胞的多胺浓度及促进了多胺的代谢,而对未分化的隐窝细胞没显著影响,同时通过Wnt信号通路调控空肠绒毛-隐窝轴细胞的分化成熟。  相似文献

色泽是金针菇的重要性状之一,受一对等位基因的控制。以FL19(黄色)、8801(白色)及其后代(F1代、F2代)菌株和相应单核菌株为材料,采用分离群体分组分析(BulkedSegregantAnalysis,BSA)方法,对金针菇基因组DNA进行RAPD分析,通过对200个引物的筛选,获得了一个与白色性状基因紧密连锁的RAPD标记S3751800,并在亲本及其后代菌株进行了验证,具有较好的重复性和稳定性。可用于白色金针菇新品种的辅助选育。  相似文献

以水稻耐盐突变体sst为对照,对67个水稻亲本及突变体sst与水稻品种华占的F2群体进行耐盐性筛选,并结合分子标记结果,分析水稻苗期耐盐性基因SST分子标记ID27093、ID27101、ID27118、INDEL1和INDEL2的特异性和准确性.结果表明,分子标记ID27093、ID27101、ID27118和INDEL1的扩增多态性较低;而分子标记INDEL2的扩增多态性较高,为共显性标记,具有较高的特异性,能区分耐盐供体sst与85.07%的供试亲本,且筛选准确率高.因此,分子标记INDEL2标记可以直接用于耐盐性基因SST分子标记辅助育种.  相似文献

目的探讨乳腺肿瘤中基质金属蛋白酶(MMP)-13的表达及其与其他生物学指标和临床预后的关系。方法采用组织芯片及免疫组织化学SP法检测183例乳腺癌组织和30例乳腺良性疾病组织中MMP-13、ER、PR、HER2、MMP-2、MMP9、表皮生长因子受体(EGFR)、基质金属蛋白酶组织抑制因子TIMP-1、TIMP-2的表达并分析相互关系,同时分析MMP-13与肿瘤临床病理分期、淋巴结转移状态、组织学分级、肿瘤大小、年龄、月经状态、病理类型、雌孕激素受体状态以及临床预后等的关系。结果MMP-13的表达与TIMP-1、TIMP-2均呈负相关,与HER2的表达呈正相关,而与ER、PR、MMP-2、MMP-9、EGFR的表达水平不相关。MMP-13的表达与淋巴结转移和高组织学分级相关(均P〈0.01)。MMP-13低表达组10年生存率与高表达组10年生存率差异有统计学意义(98.4%vs68.3%,P〈0.01)。结论MMP-13蛋白与浸润性乳腺癌的侵袭和转移相关,可作为判断预后不良的指标。  相似文献

目的 分析血浆miR-200c的表达与肺癌患者临床病理特征的相关性,并探讨miR-200c能否作为肺癌早期诊断的潜在肿瘤标志物的可能性。方法 收集55例肺癌患者和53例健康体检者,采用实时荧光定量PCR检测受检者血浆miR-200c的表达情况,对检测结果进行统计学分析。结果 miR-200c在肺癌患者组血浆中表达水平均高于正常对照组,差异有统计学意义(P<0.01)。受试者工作特征曲线显示miR-200c能够将肺癌与正常对照组区分出来(AUC=0.676)。结论 血浆miR-200c的表达可能与肺癌的发生有关,提示miR-200c可作为特异性生物标志物对肺癌患者进行早期诊断的可能性。  相似文献
