The postnatal development and histochemistry of mucins of the lingual, preglottal and laryngeal glands in the quails were investigated by means of light microscopy using specific staining for complex carbohydrates. In this study, the tongues were taken from female and male quails from day 1 to day 60 after hatching. The salivary glands in quail's tongue comprised the lingual gland, with lateral and medial (paraentoglossal gland) portions that differ in morphology and histochemical staining, and the preglottal gland, with two lateral portions and one medial portion. The medial portion of the preglottal gland, which extended to the row of the laryngeal papillae on each side of the glottis, was described as the laryngeal gland. The salivary glands were present at hatching and their cells were functionally mature and secreted mucins. In quail of all ages, the histochemical reactions revealed that the cytoplasms of the secretory cells of the preglottal, laryngeal and paraentoglossal gland (medial portion of lingual gland) contained sialomucins and weakly sulphated epithelial mucins. Neutral mucins were absent in the paraentoglossal gland, while a small amount of neutral mucins was present in other glands. The mucins with vicinal diol groups, sialomucins and weakly sulphated epithelial mucins were mixed within the secretory cells of all the glands. All the histochemical reactions were restricted to the supranuclear regions of the secretory cells within the lateral portion of the lingual gland. In conclusion, the contents of mucins in the lingual, preglottal and laryngeal glands varied between different age groups, however, no differences in the glands' histochemistry between male and female quails were observed.  相似文献

The morphology of the gland cells in the glandular sac area of the Bactrian camel and the composition of secretory substances were examined by histochemical methods. It was found that the gland cells of the glandular sac area were of the same type and size as those of the cardiac glands. The composition of secretory substances from the glandular sac area was the same as that of secretory substances from the cardiac glands. Moreover, secretory substances from the gland cells of the glandular sac area contained a great deal of acid glycoconjugates, such as sialic acid, in addition to neutral saccharides (fucose, mannose, glucose, N-acetyl-D-glucosamin, galactose and N-acetyl-galactosamin). Furthermore, immunohistochemical examination showed that progastricsin was present in the gland cells of the glandular sac area and the cardiac gland. In this study, histological analysis suggested that the stomach of the Bactrian camel is a single cavity stomach, formed as a result of multiple differentiation and growth of cardiac glands through the process of evolution.  相似文献

The glycoconjugate content of major horse salivary glands was investigated by means of horseradish peroxidase-conjugated lectins. Qualitative differences were observed in the terminal sugar residues of secretory glycoproteins and glycoconjugates linked to the apical surface of excretory duct epithelial cells. Mucous acinar cells in mandibular and sublingual glands contained oligosaccharides with D-galactose, α- and β-N-acetylgalactosamine, N-acetylglucosamine and fucose residues, whereas mandibular, sublingual and parotid serous cells contained only oligosaccharides with terminal α- and β-N-acetylgalactosamine residues. The apical portion of striated and interlobular duct lining cells of mandibular and sublingual glands stained for α- and β-N-acetylgalactosamine and for N-acetylglucosamine. In parotid gland the cytoplasm of intercalated duct cells and the apical surface of striated duct epithelial cells stained for α-N-acetylgalactosamine.  相似文献

The chemical content of the secretion of the sheep lacrimal gland was analysed at the light and electron microscope levels by applying histochemical techniques and an ultrastructural histochemical method (periodic acid, thiocarbohydrazide and silver proteinate). Mucosubstance histochemistry demonstrated acidic glycoconjugates, mainly sulphated, in the mucous and seromucous glandular cells and in the apical portion of the cells lining the terminal ducts. Moreover, secretory granules, stained with PA-TCH-SP, showed a different localization of the reaction product. The presence of lysozyme was also found in the glandular serous cells. These histochemical studies demonstrate that the secretion of sheep lacrimal glands is mixed, having serous, mucous and seromucous components, and that an excellent correlation exists between the secretory granule substructure and glycoprotein localization.  相似文献

Lacrimal glands of 12 dogs free of ocular disease were examined to determine the normal structure of these glands. The glands consisted of tubuloacinar cells that ultrastructurally and histochemically were of a single type of secretory cell in the tubules and possibly 3 types of secretory cells in the acini. The tubular epithelium contained homogenous electron-dense granules that stained as neutral glycoconjugates (periodic acid-Schiff positive and Alcian blue and high iron diamine negative). The predominant acinar cells contained granules of lesser electron density than those of the tubules, and stained as sialomucin (Alcian blue [pH 2.5] and periodic acid-Schiff-positive, and high iron diamine-negative). A second type of acinar cell was in peripheral lobules that ultrastructurally and histochemically appeared like lipid granules (positive with oil red O and osmium tetroxide). Ultrastructurally, a third type of acinar granule was finely granular, electron-lucent, and frequently coalesced. It was not readily apparent whether the latter was an artifact, a stage in the maturation of the sialomucin granules, or a third type of acinar granule. Individual acinar cells usually had a predominance of 1 granule type, but greater than 1 granule type could be found in some cells. The basal surfaces of the acinar, tubular, and ductal cells were incompletely ensheathed by myoepithelial cells. Plasma cells, lymphocytes, mast cells, endothelial cells, fat cells, and Schwann cells composed the cellular elements of the interstitium. Lymphocytes, mast cells, and nerve endings also were found in the parenchyma.  相似文献

The aim of the study was to emphasize several specific functional aspects of the mammalian prostate gland. In this connection, the subcellular localization and characterization of glycoconjugates in the secretory epithelial cells were examined that line the prostate gland of the miniature pig, using cytochemistry, including lectin-gold methods. The results verified a diversified pattern of glycoconjugates in the structures of the secretory cells. The secretory granules and the free surface coat of the plasma membrane contained several sugar moieties such as alpha-D-Man, alpha-D-Glc, alpha-L-Fuc, beta-D-Gal, GalNAc, GlcNAc and NeuAc (sialic acid), whereas the Golgi apparatus exhibited GlcNAc and sialic acid residues. In addition, the cisterns of the endoplasmic reticulum showed alpha-D-Man and alpha-D-Glc moieties. It seems likely that the secretory glycoconjugates demonstrated contribute to favourable conditions for fertilization.  相似文献

The poll glands of the camel are tubuloalveolar glands. They consist of lobules separated by interlobular connective tissue. Adrenergic axons and blood vessels including fenestrated capillaries are present in the intralobular connective tissue in close proximity to the secretory cells. The prominent features of the secretory cell cytoplasm are many mitochondria, smooth endoplasmic reticulum, and many vesicles in various secretory stages.
It is concluded that the secretory cells have apocrine as well as merocrine modes of secretion. The glands may have the ability to uptake hormones and concentrate them. Additional work is needed before considering these glands as steroid-secreting glands.  相似文献

The secretory units and duct system of the echidna sublingual glands exhibit subtle architectural modifications to accommodate the viscous secretion produced by these glands. The glands are compound tubular glands, the secretory units of which are elongate with open lumina and consist only of mucous cells. Closely packed spindle-shaped myoepithelial cells invest the secretory units, but are absent around the ducts. The branched secretory tubules open into an abbreviated duct system characterized by wide lumina. Striated ducts normally associated with the second portion of the intralobular duct system are absent. The duct system shows the most obvious modification of general salivary gland architecture presumably to accommodate the viscous secretion propelled from the secretory units by surrounding myoepithelial cells.  相似文献

Histochemical staining techniques, which differentiate the main categories of carbohydrates, were applied to sections from different segments of the alimentary canal of the shi drum Umbrina cirrosa L. to study patterns of distribution of epithelial glycoconjugates. In the oesophagus, mucous cells contained sulphomucins, together with a small amount of sialomucins. Stomach epithelial cells secreted neutral and acidic glycoconjugates, while gastric glands only produced small quantities of sialomucins. Goblet cells showed the presence of sialo and sulphomucins in the pyloric caeca, whereas intestinal mucous cells secreted sulphated glycoconjugates. This work serves as a baseline for further studies on carbohydrate composition of the mucosa of the shi drum digestive system.  相似文献

Oesophageal and gastric glands of the anuran Kassina maculata Dumeril were studied using the electron microscope. The cells of the oesophageal glands contained abundant secretory granules, rough endoplasmic reticulum and mitochondria. The cells of the gastric glands were composed entirely of chief cells which contained abundant mitochondria, few secretory granules and rough endoplasmic reticulum. It is likely that in K. maculata the oesophageal glands produce more pepsinogen than the gastric glands.  相似文献

In the oesophagus, mucins, which originate from oesophageal submucosal glands, play an important role in the mucosal protection as a pre-epithelial barrier. In this study, the structure of cervical and thoracic parts of oesophagus of Japanese quail during the post-hatching period was compared, and the contents of carbohydrate and gastric mucin MUC5AC of the oesophageal glands in these parts were analysed at the light microscope levels by applying conventional histochemical and immunohistochemical methods. The oesophageal glands were present at hatching, located in the laminae propria. The numbers of glands were different in the cervical and thoracic parts, but the differences were found to be insignificant. The thoracic part has the oesophageal tonsils which are associated with the glands. Oesophageal tonsil was formed from day 5 after hatching. In quail of all ages, the secretory epithelium of glands contained neutral sialomucins and weakly sulphomucins. The cells in the neck region of secretory units contained sialomucins, while the cells of excretory ducts had strongly sulphomucins. Sialomucin containing cells in the secretory units increased with the advance of age and glandular development. But, in the secretory units, the sulphomucin content of glands was more in the thoracic part. The secretory epithelium of tonsil-associated glands contained mostly sulphomucins and a little sialomucin. From the hatching, MUC5AC mucin was detected in the cells of excretory ducts. Although the lymphoepithelium of the tonsil units exhibited negative reactions to all histochemical methods, it showed positive reaction to MUC5AC mucin antibody. In conclusion, the cervical and thoracic parts may be functionally different and the thoracic part of oesophagus was transformed into an immunological organ following day 5 after hatching.  相似文献

The dorsal lingual surfaces of adult owl (Strix uralensis) were examined by scanning electron microscopy. The length of the tongue was about 2 cm. The tip of the tongue of the owl was bifid. Three parts were distinguished in the tongue of the owl: the apex, the body and the root of the tongue. The conical region between the lingual apex and lingual root was a very wide area. There were thread‐shaped processes/cells of epithelium in the lingual apex. The small or large conical papillae were observed on the lingual body. The morphological differences between two types of secretory cells (clear and dark) from pig carpal glands were examined. The main difference is the presence in the dark cells of secretory granules of moderate electron density, made up of acidic and neutral glycoproteins. 1. The aim of the study was to examine the morphology of the tongue and the histochemical features of the lingual salivary glands in this species.

2. The tongue was elongated, terminating in a rather sharp, dagger-like apex. On the surface of the tongue and situated between the body and root of the tongue, two rows of conical papillae, the sharp apices of which pointed towards the posterior part of the tongue, were observed. The keratinised epithelium lining the dorsal surface lacked typical gustatory papillae. However, it was observed that taste buds were present in the epithelium of the lingual body and root. The tongue was supported by a structure composed of hyaline cartilage, the paraglossum, which extended from the lingual root to the apex. Simple branched tubular glands, which were encapsulated by connective tissue, were embedded within the submucosa in the body (anterior salivary glands) and root (posterior salivary glands) of the tongue. It was observed that the secretion of the lingual glands contained neutral mucins, proteoglycans containing carboxylic acid, weak and strong sulphated groups, N-acetylated sialomucins, but lacked glycogen.

3. It was demonstrated that, the general morphological features, papillary distribution of the tongue and the histological structure of the mucosa epithelium and the supportive elements displayed similarity to those of other domestic avian species. It was also determined that, in view of the particular feeding types, in the partridge, the presence of the papillary crest was not correlated with diet.  相似文献

Objective-To determine glycohistochemical characteristics of enzootic nasal tumors (ENTs) of sheep, compare results for ENT with those of histologically normal nasal mucosa of sheep, and identify the histologic origin of ENT. Sample-ENT and nasal mucosa samples obtained from cadavers of 5 adult Lacaune sheep with ENT and 5 Lacaune sheep unaffected by ENT, respectively. Procedures-Samples of ENT and nasal mucosa were collected from cadavers of sheep and sectioned. Conventional and lectin histochemical analyses were used to identify glycoconjugates in tissue sections on the basis of their principal chemical groups and principal terminal or internal oligosaccharidic glucidic residues, respectively. Results-ENTs had papillary and tubular portions. Cells in the papillary portion of ENTs had secretion and surface glycoconjugates, which included sulfated glycosaminoglycans and neutral and sialilated glycoproteins. Cells in the tubular portion of ENTs had surface glycoconjugates, which included neutral and sialilated glycoproteins. Both portions of ENTs had C(4)-acetylated sialoderivatives that were not detected in sections of histologically normal nasal mucosa. Conclusions and Clinical Relevance-The papillary portion of ENTs in sheep may originate from respiratory glands and goblet cells. The tubular portion of ENTs in sheep may originate from olfactory glands. Presence of C(4)-acetylated sialoderivatives in cells of ENTs could confer resistance against pathogens to those cells.  相似文献

Previously, the distribution of myoepithelial cells (mecs) in the salivary glands was studied by both immunohistochemistry, and transmission electron microscopy; however, little was elucidated concerning their morphological features, especially in goats. This study was performed to investigate the correlation between the cytoarchitecture of the mecs in goat major salivary glands (parotid, mandibular, and sublingual glands) and the nature of the saliva secretion. The cytoarchitectural features of the mecs were examined by scanning and transmission electron microscopy as well as immunohistochemically. The secretory endpieces in the parotid gland are of the pure serous type, but in both the mandibular and sublingual glands they are of the mixed type. In all studied glands, the intercalated ducts were covered by mecs which, unlike the large stellate cells that surrounded the secretory endpieces, were spindle-shaped with few cytoplasmic processes. Interestingly, the mecs were found to bulge on the basal surfaces of the serous acini and intercalated ducts in all glands and to be in close contact to the seromucous tubules surface in the mandibular and sublingual glands forming a continuous network around it. In conclusion, the differences in the degree of development of the mecs as well as the number of their cytoplasmic processes may be correlated with the nature of the secretion and the number of the secretory granules. Thus these observations may have some relevance in the diagnosis of atrophy and pathogenic conditions of these glands.  相似文献

Previous investigations failed to demonstrate mast cells in the alimentary tract and extraparietal glands of the ferret. It was decided therefore to test this and assess factors that may be of influence. Major salivary glands and tongues of mature ferrets, which had been fixed in formalin-calcium, were examined by means of light microscopical histochemistry. Staining of paraffin sections with techniques depending on basic dyes or esterolytic activity was carried out for conventional times with and without previous oxidation, hot acid hydrolysis, and trypsin and beta-glucuronidase digestion. Aldehyde fuchsin and high iron diamine consistently revealed the presence of few mast cells in interstitial stroma of salivary glands and lingual musculature, and in the lamina propria of lingual mucosa. Alcian blue at 0.5 M MgCl2 and safranin produced less consistent results, and even fewer metachromatic mast cells were detected. No staining of mast cells was obtained with the technique for naphthol AS-D chloroacetate esterase. Pretreatment did not increase the numbers and/or staining reactions of mast cells. The results refute the previous misconception and suggest that ferret is a species with a low incidence of mast cells largely expressing a connective-tissue phenotype.  相似文献

This work aimed to characterize the complex carbohydrates in the duodenum of fetal and adult fallow deer. The proximal parts of the duodenum were removed from the intestine of 12- and 16-week-old fetuses and of adult fallow deer. Sections were assessed by conventional carbohydrate histochemistry combined with glycosidase digestions and treatment with KOH. During development, there was an increase in the acid carboxylated components of the glycoconjugates in goblet cells, while the duodenal glands expressed high quantities of sulphated glycoconjugates at the beginning of development but, in the adult, mainly secreted neutral and carboxylated glycoconjugates containing sialic acid. Sulphated components, probably represented by chondroitin sulphate B-like and heparan sulphate-like glycosaminoglycans may play a role in the morphofunctional differentiation of the duodenum.  相似文献
