含有牛羊源性成分的动物源性饲料的使用,一直被认为是疯牛病(BSE)传播的主要途径;对反刍动物饲料中牛羊源性成分进行检测是防范疯牛病传播的有效措施之一;本文利用聚合酶链式反应(PCR)技术对反刍动物饲料中牛羊源性成分的DNA进行定性检测,该方法的检测限为0.1%,同时利用该方法对部分省区的863批反刍动物饲料样品进行了牛羊源性成分的筛查检测工作,取得了宝贵的技术资料。  相似文献

应用PCR法监测反刍动物饲料与动物源性饲料中的牛羊成分,是当前一项行之有效的检测方法,是防止疯牛病等烈性传染性疾病的发生与流行,保障市场上牛羊肉食品的质量安全,促进草食动物养殖进一步发展的重要措施。但是,由于各种检品的成分差异性较大,样品的均匀度不高,待检样品的取样量与检测值的相关性不强。为此,2008年和2009年,我们在对牛羊饲料与动物源性饲料中牛、羊成分采用PCR法进行检测的同时,进行了不同称量对检测值的影响和不同检测方法间的对比检测与研究。现将试验与检测的中的有关情况简要介绍如下。  相似文献

为了贯彻落实《动物源性饲料产品安全卫生管理办法》,加强对鱼粉等动物源性饲料和反刍动物饲料产品的监督管理,农业部组织在部分省(区、市)进行了牛羊源性成分的监督检测。一、检测结果本次监督检测共抽检鱼粉等动物源性饲料和反刍动物饲料样品4024批次,检出牛羊源性成分样品133批次,检出率为3.30%。鱼粉检出率为6.55%,其中进口鱼粉检出率为7.28%,国产鱼粉检出率为1.61%,产地不详的鱼粉检出率为12.5%。疯牛病疫区国家或产地不详的骨粉、肉粉、肉骨粉样品检出率为33.3%。反刍动物饲料样品检出率2.53%,其中配合饲料(含自配饲料)、精料补充饲料…  相似文献

[目的]检测反刍动物饲料和动物源性饲料中牛羊源成分。[方法]利用实时荧光定量PCR技术对反刍动物饲料产品(预混料、精料补充料、全价配合料等)和动物源性饲料产品(鱼粉、禽肉粉、猪肉粉、羽毛粉等)共计131批样品进行了牛羊源成分检测。[结果]所有样品中均未检出牛羊源性成分。[结论]该次抽检的反刍动物饲料产品和动物源性饲料产品中暂无牛羊源成分污染,但仍应加强对反刍动物饲料和动物源性饲料中牛羊源成分的监测。  相似文献

[目的]检测反刍动物饲料和动物源性饲料中牛羊源成分,掌握动物饲料安全情况。[方法]利用聚合酶链式反应法(PCR法)对反刍动物饲料产品(预混料、精料补充料、全价配合料等)和动物源性饲料产品(鱼粉、肉粉等)共计300个样品进行了牛羊源成分检测。[结果]牛羊源性成分检测合格率为99.33%。其中,反刍动物饲料产品260批次,牛羊源性成分合格率为99.23%;动物源性饲料产品40批次,均未检出牛羊源性成分。2个阳性样品均为牛源阳性。[结论]反刍动物饲料产品存在潜在的安全隐患。  相似文献

1986年英国发生疯牛病以来,我国采取一系列管控措施降低发生疯牛病的潜在风险,2003~2017年连续15年对饲料中牛羊源性成分(疯牛病传染因子)进行监测,监测结果持续降低,基本切断了疯牛病传染因子这个传播途径,发生疯牛病的风险很低,明确了今后防控的关键。  相似文献

SYBR Green荧光定量PCR法可对含牛羊源性成分饲料进行快速筛选。本文通过优化PCR条件,将原有的三步法简化为两步法,筛选时间由最初的116min减少至67min,并应用于170份饲料样品的检测。经过筛选,排除了166份阴性样品中的162份样品,占97.6%;被怀疑为阳性样品的8份样品中,有4份样品最终被确认为阳性样品。此方法的应用,将提高饲料中牛羊源成分检测的速度和灵敏度,加强了防止疯牛病传播的保障力度。  相似文献

目的:为了监测新疆反刍动物饲料和动物源性饲料产品中的牛羊源性成分,掌握动物饲料安全情况。方法:2006~2011年应用实时荧光PCR法对新疆15地(州、市)的4432批反刍动物饲料及动物源性饲料产品进行牛羊源性成分的抽样检测。结果:结果表明,2006年共有4批产品被检出含有牛羊源性成分,其涉及所有抽检产品类别和饲料生产、经营、使用3个环节;2007~2011年的抽检合格率均达100%。结论:说明新疆的反刍动物饲料和动物源性饲料的牛羊源性成分呈现下降趋势,反刍动物饲料逐步向安全生产及使用的方向发展。  相似文献

为了防止海绵状脑病疫区和痒病疫区反刍动物源性饲料进入我国 ,非常有必要对贸易往来中的肉骨粉品种来源进行鉴定检测。我们采用分子生物学方法从动物源性饲料中鉴别检测牛、羊特异性线粒体DNA的片段 ,并用限制性内切酶SspⅠ对PCR产物进行酶切鉴定及DNA序列测定。该方法的灵敏度可达 0 .12 5 % ,具有灵敏度高、快速和特异性强的优点。  相似文献

On 26 November 2000, the first autochthonous case of bovine spongiform encephalopathy (BSE) was detected in Germany. Since then, a total of 413 BSE cases have been confirmed, resulting in the culling and destruction of 17 313 heads of cattle. In view of the possible risks for human and animal health, Germany has adopted EU regulations along with some additional requirements concerning active surveillance and response measures after detecting a BSE‐positive animal. In this study, we used a stochastic model to estimate the costs incurred by the ensuing legislative amendments responding to BSE between November 2000 and December 2010. The total costs were estimated to range between 1847 and 2094 million Euros. They peaked in 2001 (about 394 million Euros) and declined since. About 54% of the costs (approximately 1000 million Euros) were incurred by the extension of the feed ban for animal protein to all farmed livestock. Active surveillance accounted for 21% (405 million Euros), the incineration of animal protein for 13% (249 million Euros) and the removal of specified risk material for 11% (225 million Euros). Only 1% of the costs was related to response measures after detecting a BSE‐positive animal, including indemnity payments for culled cattle and confiscated carcasses at the slaughterhouse.  相似文献

An assessment was made of the risks of bovine spongiform encephalopathy (BSE) occurring in Argentina. Most of the factors associated with the origin and development of the BSE epidemic in the UK are essentially absent. For example, Argentina's large sheep and cattle industries are based on low-cost production systems using grass. Concentrated feeds are not used for sheep, rarely for beef cattle and to a comparatively modest extent for dairy cows. Particularly important are the facts that scrapie (and BSE) has never been reported in Argentina—very small amounts of waste tissues from sheep are rendered to produce meat and bone meal (MBM)—and MBM is not used in concentrated feeds for cattle. We conclude that Argentina has an exceptionally low risk of BSE due to scrapie. There is a very small risk of BSE having been introduced via live animals imported from countries with BSE, but this could only give rise to isolated cases because MBM is not fed to cattle.

A surveillance programme has been carried out based largely on a histological examination of brains from three categories of old dairy cows: animals reported on the suspicion of having neurological disease; animals in poor condition at slaughter; healthy animals randomly selected in the abattoir. No evidence of transmissible spongiform encephalopathy was seen in several sections from each of a total of 1019 brains. We conclude that, for most practical purposes, Argentina may be considered to be free from BSE.  相似文献

我们选择了3对引物A,B,C.A可扩增大多数脊椎动物(哺乳类、鸟类、爬行类、两栖类和鱼类)细胞色素b基因上一个371 bp的区段,B可扩增位于牛mtDNAD-loop区段内的一个274 bp的区段,C可扩增羊mtDNA基因上一个199bp的区段.由于3对引物扩增的产物分别相差约100 bp左右,可通过琼脂糖凝胶电泳分离来并观察结果,建立了基于内对照的同时检测牛、羊源性成分的三重PCR方法.  相似文献

用异硫氰酸胍法提取鱼油、加入牛肉DNA的鱼油、加入山羊肉DNA的互油和牛油中的DNA。18S rDNA片段以及牛、羊源性成分的PCR扩增结果表明,建立的动物油脂DNA提取方法是可行的。用建立的DNA提取方法提取3份送检鱼油的DNA,牛、羊源性成分的PCR检测结果均为阴性。  相似文献

Since scrapie and bovine spongiform encephalopathy (BSE) in sheep are clinicopathologically indistinguishable, BSE in sheep may have been misdiagnosed as scrapie. Disease-specific prion protein (PrP(d)) patterns in archival tissues of 38 Irish ARQ/ARQ sheep diagnosed as scrapie-affected were compared to those in four Dutch BSE-challenged sheep. When medulla oblongata was immunolabelled with an antibody directed against amino acids 93-99 of ovine prion protein (ovPrP), intraneuronal PrP(d) was apparent in all 38 Irish sheep but was absent in BSE-challenged sheep. When lymphoid follicles were immunolabelled with antibodies directed against amino acids 93-106 of ovPrP, granule clusters of PrP(d) were seen in 34 of the 38 Irish sheep. Follicles of the remaining four archive sheep contained either no PrP(d) or single PrP(d) granules, similar to follicles from BSE-challenged sheep. Based on the medulla results, none of the archival cases had BSE-derived disease. The identification of some scrapie sheep with little or no intrafollicular PrP(d) suggests that this technique may be limited in discriminating between the two diseases.  相似文献
