A study was conducted to determine whether compensatory ovarian hypertrophy occurred in prepuberal heifers, and if so, could compensatory ovarian hypertrophy be prevented by injecting charcoal-extracted bovine follicular fluid (CFF). An additional objective was to determine the effect of CFF injections on follicle stimulating hormone (FSH) and luteinizing hormone (LH) concentrations following unilateral ovariectomy (ULO). Thirty-one prepuberal Angus, Hereford, and Angus X Hereford heifers were assigned by weight and breed to the following groups: Control-saline injections (C), unilateral ovariectomy-saline injections (ULO) and unilateral ovariectomy-CFF injections (ULO-CFF). In group C, a sham operation was performed on d 0 and both ovaries were removed on d 7. In groups ULO and ULO-CFF, either the right or left ovary was removed on d 0 and the remaining ovary was removed on d 7. From d 0 to 7, each heifer received an injection (iv, 7 ml/injection) of saline or CFF every 12 h. Jugular blood samples were collected immediately before each saline injection from d 0 to 7 and analyzed for FSH and LH. Unilateral ovariectomy increased ovarian (P less than .08) and follicular fluid (P less than .05) weights compared with groups C and ULO-CFF; however, lyophilized ovarian weight was similar (P greater than .05) for all groups. Injections of CFF blocked the increase in ovarian and follicular fluid weight following ULO. Duroc sows farrowed second litters in March and lactated 35 +/- 2 days. At 36 hr before weaning, electrocautery of follicles greater than or equal to 3 mm in diameter (n = 8) or sham surgery (n = 5) was performed to test the hypothesis that ablation of medium-sized follicles would prolong the duration of postweaning anestrus. Number of follicles and diameters at surgery were: 1.3 +/- .6 (greater than 5 mm diameter), 26 +/- 1 (3 to 5 mm) and greater than 20 (less than 3 mm). Blood samples were collected at 15 min intervals for 3 hr beginning at -12, 0, 12, 60 and 96 hr from weaning. Interval to estrus was 3.4 +/- .2 days in seven of eight cauterized sows and 3.6 +/- .6 days for sham-surgery sows. The remaining cauterized sow was anestrus at slaughter, 40 days after weaning. Number of corpora lutea and pregnancy rate were 15.8 +/- .6 and 92%, respectively, and were similar between sham-surgery and cauterized sows. Concentration of follicle stimulating hormone (FSH) at 12 hr before weaning was greater in sows subjected to electrocautery than for sham-surgery sows, but FSH values were similar at other sampling times. Concentrations of estradiol were similar at all times for both treatment groups. Luteinizing hormone (LH) was higher (P less than .05) at 60 hr in cauterized sows because of the onset of the preovulatory LH surge in one sow. We conclude that destruction of medium-sized ovarian follicles before weaning did not influence postweaning reproductive performance.  相似文献

In an effort to determine whether anestrus in swine is due to aberrant ovarian feedback control of gonadotropin release, this study contrasted the influence of ovariectomy on LH concentrations in serum of anestrous sows and in sows that returned to estrus following weaning. Blood samples were collected at 6-h intervals from 7 d prior to until 4 d after ovariectomy of 22 anestrous and 24 cyclic sows. Blood samples also were collected at 15-min intervals for 8 h at 2 d prior to and 2 d after ovariectomy. Sampling at 6-h intervals continued until 12 d after ovariectomy and additional 8-h windows of 15-min samples were taken at 7 and 12 d after ovariectomy of seven anestrous and nine diestrous sows. Mean LH concentrations and LH pulse frequencies were greater (P less than .05) 2 d after ovariectomy than 2 d prior to ovariectomy in both anestrous and diestrous sows. Mean pulse amplitude had increased by 2 d after ovariectomy in anestrous sows but did not change in cyclic sows. Baselines as determined from the mean of all LH measurements excluding pulses, remained the same in both anestrous and diestrous sows at 2 d after ovariectomy. Pulse frequency, pulse amplitude, and mean LH concentration were greater (P less than .05) in both anestrous and diestrous sows at 7 and 12 d after ovariectomy than at 2 d prior to and 2 d after ovariectomy. The purpose of this experiment was to determine the ovulation rate after treatment with human chorionic gonadotropin (hCG) in two groups of gilts characterized by different ovarian morphology: grape-type (GT; n = 11) and honeycomb-type (HT; n = 7). At 170 d of age (d 0), gilts were examined by laparoscopy and ovarian type was determined by the distribution of macroscopic follicles present on the ovarian surface. Five to ten minutes after surgery, each gilt received a single injection (i.m.) of 750 IU of hCG. At d 0, GT ovaries had a greater number of large follicles (greater than or equal to 6 mm) than HT ovaries (10.0 +/- .5 vs 2.6 +/- .3; P less than .05), whereas HT ovaries had more small follicles (1 to 3 mm; HT: 42.3 +/- .8 vs GT: 26.7 +/- .9; P less than .05) and total follicles (HT: 59.4 +/- 2.3 vs GT: 52.2 +/- 1.5; P less than .05), although numbers of medium follicles (4 to 5 mm) were similar (GT: 15.6 +/- .8 vs HT: 14.6 +/- 1.7; P greater than .10). Number of induced corpora lutea (CL) per ovary was greater (P less than .05) in gilts with GT ovaries (10.59 +/- 2.9 CL) than in gilts with HT ovaries (5.21 +/- .66 CL). The suppressive effects of exogenous 17 beta-estradiol (E2) on LH concentrations in sows that remained anestrus following weaning and in those that returned to estrus were evaluated. Four anestrous and four cyclic sows were treated subcutaneously with silastic implants containing E2 at 13 d after ovariectomy (d 0). Three anestrous and six cyclic sows received silastic implants without E2. Blood was collected at 6-h intervals from d -1 to d 12 and at 15-min intervals for 8 h on d -1, 2, 7 and 12. Sows were treated with 1 microgram GnRH/kg BW at the completion of each 8-h frequent sampling period. Blood was collected at intervals of 10 to 30 min for 3 h after GnRH treatment. Concentrations of E2 remained less than 5 pg/ml in sham-treated sows and were between 20 and 25 pg/ml in E2-treated females. Pulsatile LH concentrations was similar between anestrous and cyclic sows prior to implant treatment. Sham-treated anestrous sows had greater (P less than .05) pulse frequency and mean LH concentrations than E2-treated anestrous sows on d 2, 7 and 12. Differences in pulsatile LH concentrations between E2-treated and sham-treated cyclic sows were not detected. Pulse frequency was less (P less than .05) in E2-treated anestrous sows than in E2-treated cyclic sows on d 7 and 12. Peak LH concentrations were greater (P less than .05) in E2-treated cyclic sows than in E2-treated anestrous sows at each GnRH challenge. These results suggest that the hypothalamo-hypophyseal axis is more sensitive to the negative feedback effects of E2 in anestrous sows than in cyclic sows. In addition, chronic E2 treatment reduces pituitary responsiveness to GnRH to a greater extent in anestrous than in cyclic sows. Failure to return to estrus in swine may be due, at least in part, to an increased sensitivity of the hypothalamo-hypophyseal axis to the negative feedback effect of estradiol.  相似文献

Seven sows were placed into one of two environmental chambers at 22 C, 5 d prior to farrowing. On day 9 of lactation, one chamber was changed to 30 C (n = 4) and the other remained at 22 C (n = 3). On days 24 and 25, blood samples were collected every 15 min for 9 hr and 7 hr, respectively. On day 24, thyrotropin releasing hormone (TRH) and gonadotropin releasing hormone (GnRH) were injected iv at hour 8. On day 25 naloxone (NAL) was administered iv at hour 4 followed 2 hr later by iv injection of TRH and GnRH. Milk yield and litter weights were similar but backfat thickness (BF) was greater in 22 C sows (P less than .05) compared to 30 C sows. Luteinizing hormone (LH) pulse frequency was greater (P less than .003) and LH pulse amplitude was less (P less than .03) in 22 C sows. LH concentrations after GnRH were similar on day 24 but on day 25, LH concentrations after GnRH were greater (P less than .05) for 30 C sows. Prolactin (PRL) concentrations were similar on days 24 and 25 for both groups. However, PRL response to TRH was greater (P less than .05) on both days 24 and 25 in 30 C sows. Growth hormone (GH) concentrations, and the GH response to TRH, were greater (P less than .0001) in 30 C sows. Cortisol concentrations, and the response to NAL, were less (P less than .03) in 30 C sows. NAL failed to alter LH secretion but decreased (P less than .05) PRL secretion in both groups of sows. However, GH response to NAL was greater (P less than .05) in 30 C sows. Therefore, sows exposed to elevated ambient temperature during lactation exhibited altered endocrine function.  相似文献

Endocrine differences between European crossbred and Chinese Meishan females have been somewhat modest. Indwelling jugular cannulas were placed in Meishan (n = 7) and White composite (n = 6) multiparous sows before weaning, and blood was sampled from 4 h before to 240 h after removal of litters. Temporal changes in FSH, triiodothyronine (T3), and tetraiodothyronine (T4) after weaning differed between Meishan and White composite sows. Plasma cortisol concentrations were higher in Meishan sows than in White composite sows (P < 0.01), but there were no temporal differences between breeds after weaning. Other hormones monitored (prolactin, GH, IGF-I, and inhibin) were not different between breeds. In the second experiment, Meishan gilts (n = 7) and sows (n = 7) and White composite sows (n = 9) were cannulated during the luteal phase of the estrous cycle and sampled after treatments consisting of GnRH (15 and 150 ng/kg BW), ovariectomy, estradiol cypionate challenge after ovariectomy (10 microg/kg BW), and GnRH antagonist. In response to GnRH challenge, White composite sows had elevated (P < 0.05) concentrations of gonadotropins compared with Meishan. Cortisol concentrations were elevated in Meishan as compared with White composite females (P < 0.01) but unaffected by GnRH treatment. After ovariectomy, LH concentrations increased 3 h sooner in White composite than in Meishan females. After GnRH antagonist, declines in gonadotropins were comparable in both breeds, but LH increased in Meishan females by 20 h but not until after 54 h in White composite females. White composite females demonstrated only a short decline in FSH in response to the GnRH antagonist, but Meishan females had a prolonged decline in FSH concentrations. Consistently elevated cortisol concentrations in Meishan females may positively impact ovarian function; thus, the assumption that high plasma cortisol concentration as an index of stress that impairs reproductive function should be reevaluated in swine. There were few endocrine differences that would relate to or explain increased ovulation rates found in the Meishan breed; thus, other mechanisms must exist to explain the increase in ovarian function in the Meishan breed.  相似文献

The current experiment was carried out to determine whether exogenous GnRH treatment in primiparous, lactating sows undergoing feed restriction would improve reproductive performance after weaning. Sows were allocated to one of three treatments: AA sows (n = 8) were fed to appetite throughout a 28-d lactation, AR (n = 12) and AR + GnRH (n = 12) sows were fed as AA sows from farrowing to d 21 of lactation, and feed intake was reduced to 50% of the ad libitum intakes from d 22 to 28. The AR + GnRH sows received 800 ng of GnRH i.v. every 6 h from d 22 to 28 of lactation, and AA and AR sows received saline. Sow weight, backfat, and litter weight were recorded weekly. Within 2 d after farrowing, litter size was standardized to 8 to 10. At d 17 of lactation, an indwelling jugular catheter was surgically implanted in each sow. Blood samples were taken for characterization of plasma LH, FSH, insulin, IGF-I, and leptin by RIA at d 21 and before and after weaning on d 28 of lactation. After weaning, all sows were given ad libitum access to feed, checked for onset of standing estrus twice daily with mature vasectomized boars, and inseminated 12 and 24 h after onset of standing estrus with pooled semen from the same fertile boars (3 x 10(9) sperm/AI). After breeding, feed allowance was reduced to NRC (1988) requirements for gestation. At d 28 +/- 3 of gestation, sows were killed and ovulation rate and embryo survival were determined. Restricted sows lost more weight during lactation than AA sows (P < .02). During the period of feed restriction, plasma IGF-I and postprandial insulin and leptin in AR and AR + GnRH sows, and LH pulse frequency in AR sows, were lower than those in AA sows (P < .04). Associations (P < .004) between plasma insulin and leptin and between leptin and mean LH concentrations were established. The LH pulse frequency in AR + GnRH sows did not differ from that in AA sows before weaning. After weaning, maximum, mean, and minimum LH concentrations in the AA and AR sows, and FSH concentrations in AR sows, increased (P < .05) in response to weaning. Paradoxically, GnRH treatment in lactation seemed to suppress the expected LH and FSH responses to weaning. Ovulation rate and embryo survival were not different among the three groups. In conclusion, although exogenous GnRH therapy restored LH secretion in feed-restricted sows, it did not improve overall reproductive performance.  相似文献

We hypothesized that the LH response to GnRH would be greater as the interval from foaling increases, whereas the FSH response would decrease, and that corpus luteum function after the first ovulation would be similar to that after the second ovulation. At parturition, mares were assigned to receive GnRH (2 micrograms/kg) intravenously on 1) d 3 postpartum (n = 6); 2) d 6 postpartum (n = 6); 3) d 1 of first postpartum estrus (foal estrus) and again on d 1 of second postpartum estrus (n = 8). Blood was collected through an indwelling cannula at -2, -1 and 0 h relative to GnRH stimulation (basal concentrations) and at .25, .5, .75, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5 and 4.0 h post-GnRH. Samples were assayed for concentrations of LH and FSH. Basal concentrations of LH were lower (P less than .05) for mares given GnRH on d 3 postpartum than for mares on d 1 of foal estrus. A rise in concentrations of LH was noted within 30 min in all groups, but the response to GnRH on d 1 of the first estrus was less (P less than .05) than on d 1 of second postpartum estrus. As the interval from parturition increased, the amount of LH secreted in response to GnRH increased. This study was designed to test the effects of active immunization against estrogen and progesterone on patterns of luteinizing hormone (LH) and follicle stimulating hormone (FSH) secretion, ovarian characteristics and growth rate of heifers. Heifers were randomly assigned to four treatments: 1) control injection (n = 10); 2) ovariectomy (n = 9); 3) immunization against estrogen (anti-E, n = 10); and 4) immunization against estrogen and progesterone (anti-E+P4, n = 10). Three booster immunizations were administered at 1, 1.5 and 6 mo after primary immunization. Progesterone antibody binding was 40% (34 fmol at 1:600 final dilution) in the anti-E+P4 heifers, and estradiol-17 beta binding was 35% (30 fmol) and 60% (52 fmol at 1:100 final dilution) in the anti-E+P4 and anti-E heifers, respectively, after the final immunization. Anti-E+P4 heifers had more pulses of LH and higher basal concentrations of LH than anti-E or control heifers (P less than .05). Concentrations of LH in anti-E+P4 heifers did not increase to concentrations found in ovariectomized heifers (P less than .05). Immunization against steroids did not alter the secretion of FSH. The number of large follicles (greater than 15 mm diameter) in anti-E+P4 and anti-E heifers was greater than in control heifers (P less than .05). Ovarian weight was increased in anti-E+P4 heifers (P less than .05). Average daily gain was not different among groups (P greater than .05). It was concluded that active immunization against estrogen and progesterone in heifers increased LH secretion and stimulated ovarian function.  相似文献

This experiment was conducted to 1) determine whether chronic cystic-ovarian-diseased (CCOD) cows fail to respond to luteinizing hormone (LH) treatment because of a lack of adequate ovarian LH receptors and 2) determine the effect of follicle stimulating hormone (FSH) treatment on ovarian LH and FSH receptors in ovaries of CCOD cows. The CCOD cows were those that did not resume cyclic ovarian activity after repeated treatment with human chorionic gonadotropin (hCG) and(or) LH-releasing hormone (LHRH) and were considered chronic by veterinarians. Thirteen CCOD cows were purchased from producers; six of them were injected with 5 mg FSH twice daily for 3 or 5 d (TCCOD) and the remaining seven remained untreated. Seven control (noncystic) cows in the luteal phase of the estrous cycle were injected with Lutalyse approximately 48 to 50 h before slaughter so they would be in the follicular phase (FP) of the cycle at the time of slaughter. Analysis of serum and pituitaries showed no differences (P greater than .05) in mean concentrations of serum or pituitary LH and FSH or pituitary LHRH receptor concentration and affinity among FP, CCOD and TCCOD cows. Ovarian follicle wall concentrations of receptors for LH (3.2 +/- .6; 13.0 +/- 2.5; 22.4 +/- 5.1 fmol/mg protein) and FSH (10 +/- 2.6; 43 +/- 7.2; 29 +/- 6.7 fmol/mg protein) were lower (P less than .05) in CCOD cows compared with FP and TCCOD cows, respectively. Two experiments were conducted to determine the effects of pregnant mare's serum gonadotropin (PMSG) on follicular populations and ovulation rates in prepuberal gilts with grape-type (GT) and honeycomb-type (HT) ovaries. The follicular populations were determined at 170 d of age (d 0) and 19 d after PMSG (d 19). In Exp. 1, the mean number of macroscopic follicles of Classes 2 and 3 was greater (P less than .05) in GT (n = 11) than in HT (n = 32) ovaries at d 0, whereas the mean number of those of Class 1 was greater (P less than .05) in HT ovaries. At d 19, no difference was observed between the two ovarian types for any class of follicles. The PMSG-induced ovulation rates were comparable between the two ovarian types (8.3 vs 7.9, GT vs HT, respectively; P greater than .10). In Exp. 2, the microscopic follicular populations were determined on right and left ovaries removed, respectively, on d 0 and 19. At d 0, GT ovaries (n = 5) contained a greater number of Class 5 nonatretic (P less than .01) and atretic (P less than .02) follicles than did HT ovaries (n = 5), whereas at d 19 the mean number was not statistically different between the two ovarian types (P greater than .10). Five lighthorse mares were actively immunized against gonadotropin releasing hormone (GnRH) conjugated to bovine serum albumin (BSA) to study the involvement of GnRH in luteinizing hormone (LH) and follicle stimulating hormone (FSH) secretion following ovariectomy (OVX) and after administration of testosterone propionate (TP). Five mares immunized against BSA served as controls. Immunizations were started on November 1, and OVX was performed in June (d 1). All mares were treated with TP from d 50 to 59 after OVX. On the day of OVX, concentrations of LH were lower (P less than .05) in GnRH-immunized mares than in BSA-immunized mares and were generally nondetectable; FSH concentrations were reduced (P less than .05) by 50% in GnRH-immunized mares relative to BSA-immunized mares. In contrast to BSA-immunized mares, plasma concentrations of LH or FSH did not increase after OVX in GnRH-immunized mares. The LH response to GnRH analog (less than .1% cross-reactive with GnRH antibodies) on d 50 was reduced (P less than .05) by 97% in GnRH-immunized mares relative to BSA-immunized mares, whereas the FSH response was similar for both groups. The objective of this study was to determine if alterations in dietary intake of heifers can influence IGF-I concentrations in plasma and(or) follicular fluid (FFL), size of follicles, and steroid concentrations in FFL (as an indicator of steroidogenic capacity). Cyclic heifers [n = 23; mean +/- SE body weight (BW) = 373 +/- 7 kg] were individually fed for 10 weeks either: a) 1.8% of BW in dry matter (DM) per d (GAIN; n = 7), b) 1.1% of BW in DM per d (MAINT; n = 8) or c) 0.7% of BW in DM per d (LOSE; n = 8). After 10 wk of treatment, heifers were ovariectomized 36-40 hr after the second injection of prostaglandin F2 alpha analog (2 injections 11 d apart), and plasma and ovaries were collected. Heifers weighed 444 +/- 13,387 +/- 8 and 349 +/- 9 kg in the GAIN, MAINT and LOSE groups, respectively, at time of ovariectomy. Mean diameter of follicles greater than or equal to 10 mm was greater (P less than .05) for GAIN (15.6 mm) than for MAINT (11.0 mm) or LOSE (12.5 mm) heifers. Numbers of follicles and concentrations of IGF-I in plasma and FFL did not differ (P greater than .20) between LOSE, MAINT and GAIN heifers. Progesterone concentrations were greater (P less than .05) in small and medium follicles of GAIN than MAINT or LOSE heifers, but were unaffected by diet in large follicles. High concentrations of estrogens in the peripheral circulation during late gestation inhibit synthesis of LH and markedly reduce pituitary content of LH at the end of pregnancy in most domestic species. Because blood concentrations of estrogen peak shortly before mid-gestation in the mare and then gradually decrease until parturition, we hypothesized that pituitary content of LH may increase during late gestation. To test this hypothesis 10 horse mares were challenged with a maximally stimulatory dose (2 micrograms/kg) of GnRH on d 240 and 320 of gestation and d 3 after parturition. A separate group of four mares were treated with GnRH on d 2 or 3 estrus. Blood samples were collected at -2, -1, 0, .25, .5, .75, 1, 1.5, 2, 2.5, 3, 3.5, 4, 5, 6, 7 and 8 h relative to injection of GnRH and serum was analyzed for concentration of LH and FSH. Basal serum concentration and total quantity of LH released after GnRH stimulation (assessed by determining the area under the response curve) were not different on d 240 and 320 of gestation or on d 3 after parturition (12.5 +/- 3.5, 5.7 +/- 1.5 and 29.1 +/- 12.1 ng.min/ml, respectively) and were less (P less than .05) than on d 3 of estrus (311.0 +/- 54.0 ng.min/ml). There was little difference in the basal serum concentration of FSH at any of the time points examined. Two trials were conducted to examine reproductive function and feedlot performance by heifers after active immunization against GnRH. In trial 1, heifers were not immunized or were immunized with one of three doses of a GnRH-KLH (keyhole limpet hemocyanin) conjugate in Freund's complete adjuvant. Antibodies against GnRH were not detectable in non-immunized heifers (n = 9). However, antibodies against GnRH were noted in all immunized animals (n = 30) within 8 wk of primary immunization; anti-GnRH antibody concentrations were at a maximum 16 to 20 wk after immunization. This increased anti-GnRH titer was associated with a decreased serum concentration of progesterone. Ovarian and uterine weight and tissue concentrations of LH and GnRH receptor were reduced (P less than .05) by immunoneutralization of GnRH. Similarly, immunization against GnRH reduced (P less than .05) weight gain during feedlot confinement. In trial 2, feedlot performance after insertion of anabolic steroid implants (Synovex H) was evaluated in non-immunized heifers (n = 15), heifers actively immunized against GnRH-KLH (n = 15) or KLH alone (n = 15), or non-immunized heifers treated with melengestrol acetate (MGA; n = 15). Serum concentrations of progesterone were depressed in anti-GnRH and MGA-fed groups, but ovarian and uterine weights were depressed (P less than .05) only in heifers immunized against GnRH. Total weight gain and gain during the final 4 wk of confinement did not differ (P greater than .05) among groups with steroid implants. The GnRH-KLH conjugate is an effective immunogen in heifers, leading to suppression of reproductive activity. The depression of weight gain that attends development of anti-GnRH titers may be reversed by use of implants that contain anabolic steroids.  相似文献

Lactating sows were used to evaluate effects of morphine and suckling on secretion of LH and prolactin (PRL) and occurrence of estrus after weaning. In the first experiment, crossbred multiparous sows nursing 7.9 +/- .4 pigs per litter at 25.2 +/- .3 d of lactation were subjected to one of three treatments during the middle 8-h segment of a 24-h experimental period. Treatments were infusion (i.v.) of morphine (200 mg/h) with the litter present (n = 4) or transiently weaned (n = 4), or transient weaning of litters without morphine (n = 4). Transient weaning decreased (P less than .05) prolactin and increased (P less than .05) the frequency of LH pulses and average concentration of LH. Infusion of morphine caused transient hyperthermia and suppressed (P less than .05) LH release in two of four sows nursing litters and in four sows whose litters were absent. Infusion of morphine, in the presence or absence of litters, suppressed PRL during the middle and last 8-h segments. A second experiment was conducted to test the hypothesis that chronic administration of morphine delays onset of estrus after weaning. Primiparous Duroc sows were assigned at weaning (53 to 63 d postpartum) to receive morphine (n = 10) or saline (n = 11). Saline (1.5 ml) or morphine (75 mg) was administered s.c. three times a day for 5 d after weaning. The objective of this study was to determine effects of age and castration on follicle stimulating hormone (FSH) secretion in prepuberal heifers. In experiment 1, twelve heifers were bilaterally ovariectomized at 3, 6, or 9 months of age (n = 4/group). Blood was collected at 10 min intervals for 8 hr at 1 week before ovariectomy and 1 and 4 weeks after ovariectomy. Frequency, amplitude and duration of FSH pulses were calculated. Mean plasma concentration of FSH (ng/ml), and frequency (pulses/8 hr), amplitude (ng/ml), and duration (min/pulse) of FSH pulses were not altered by age. Mean concentration of FSH increased (P less than .01) from 1 week before to 1 week and 4 weeks after ovariectomy, respectively, in all age groups. Pulse frequency increased (P less than .05) from 1 week before ovariectomy to 4 weeks after ovariectomy in 3 month old heifers, from 1 week before to 4 weeks after ovariectomy in 6 month old heifers, and from 1 week before to 1 week and 4 weeks after ovariectomy in 9 month old heifers. In experiment 2, twelve heifers were bilaterally ovariectomized at 3, 6 or 9 weeks of age (n = 4/group). Our objective in this study was to determine endocrine responses and changes in ovarian structures after a single injection of a GnRH agonist in Holstein dairy heifers (n = 38). Heifers were inseminated and received (i.m.) either saline or 200 micrograms of fertirelin acetate once on d 11, 12, or 13 after estrus (d 0). Blood was collected at 15- to 30-min intervals for 6 h after the injection to determine concentrations of LH, FSH, estradiol (E), and progesterone (P) in serum and once daily for 8 to 12 d after the injection to determine concentrations of E and P. Pregnancy rates were 58% (11 of 19) in both treatment groups. Diameter of the corpus luteum and numbers and appearance of ovarian follicles were determined by real-time ultrasonography on d-1 through 5 after injection. No treatment-induced ovulations or changes in the number of ovarian follicles were observed after the injection of the GnRH agonist. More (P less than .05) of the largest follicles within heifers receiving fertirelin acetate showed changes in their appearance on at least the 1st d after injection (6 of 10 vs 1 of 9 control heifers). Yorkshire boars were used to evaluate the influence of duration of photoperiod and hemicastration on growth and testicular and endocrine functions. At 10 wk of age, 5 hemicastrate (HC) and 5 intact (I) boars were assigned to either 8 or 16 hr of light daily until 6 mo of age. Body weights were recorded biweekly throughout the experiment. Venous cannulae were placed in all boars at 6 mo of age, and serum was collected at 30 min intervals from 0800 to 2000 hr. Gonadotropin releasing hormone (GnRH) was infused at 2000 hr (50 micrograms) and at 2030 hr (250 micrograms), and samples of serum were collected until 2400 hr. The following day, all boars were castrated, and the weights and sperm content of the testes and epididymides were determined. At castration, all pigs were given implants containing testosterone. Two weeks later, pigs were again canulated, and serum was obtained at 15 min intervals for 2 hr. Growth of boars was not significantly affected by duration of photoperiod or number of testes. Duration of photoperiod did not affect weight or sperm content of testes or epididymides. Hemi-castrated boars had greater testicular (P less than .01) and capita-corpora (C-C) epididymal weights (P less than .05) and more testicular and C-C sperm (P less than .01) per testis. Neither average concentrations of luteinizing hormone (LH) nor number and amplitude of pulses of LH were affected by photoperiod treatment. However, HC boars had greater average concentrations of LH (P less than .05) than I boars (.71 +/- .05 vs .52 +/- .05 ng/ml). Hemicastrated boars in 16 hr light daily had greater concentrations of FSH in serum (P less than .05) than 8I, 8HC, and 16I boars. Intact and HC boars had similar concentrations of prolactin (PRL) and testosterone. Similarly, concentrations of PRL and testosterone were not affected by duration of photoperiod. Secretion of LH and testosterone after treatment with GnRH was not significantly affected by duration of photoperiod. In general, HC boars released more LH in response to GnRH treatment than I boars. Concentrations of LH were greater (P less than .05) in HC than I boars at .5, 1, 2, and 3 hr after GnRH and tended (P less than .10) to be elevated at 1.5, 2.5, 3.5 and 4 hr after GnRH.
