To elucidate the effects of seasonal temperature acclimatization on thermal gelation of grass carp myosin, myosins from fish in different seasons were prepared and investigated for the changes in dynamic viscoelastic parameters including storage modulus (G′), loss modulus (G″) and damping factor (tan δ) upon heating. Myosins from fish in spring and summer had a temperature region of 38–44°C for the first marked increase of G′ higher than that of myosins from fish in autumn and winter (28–33°C). The measurement temperature-dependent changes in dynamic viscoelastic parameters such as G″ and tan δ were also different among the four myosins. While gel formation was observed with the spring and summer myosins, apparently in two steps, three steps were found in the autumn myosin. Furthermore, the winter myosin exhibited more than three steps for gel formation. These differences in rheological properties among the four myosins were considered to be attributed to the differences in thermodynamic and structural properties of these myosins previously reported.  相似文献

ABSTRACT:   Enzymatic and structural properties of white croaker fast skeletal muscle myosin were determined and compared with those of walleye pollack counterpart. Ca2+-ATPase activity of white croaker myosin was decreased to approximately 70% of the original activity during 1 day of storage at 0°C and pH 7.0 in 0.5 M KCl and 0.1 mM dithiothreitol, whereas that of walleye pollack was decreased to approximately 20% under the same condition. The activation energy ( E a) for inactivation of white croaker myosin calculated by the Arrhenius plot for inactivation rate constant (KD) was 1.2-fold higher than that of walleye pollack. While Ca2+-ATPase showed a similar KCl-dependency for the two species, the maximal activity was observed at pH 6.2 and 6.3 for white croaker and walleye pollack, respectively. Actin-activated myosin Mg2+-ATPase activity of white croaker was approximately half that of walleye pollack at 0.05 M KCl and pH 7.0, although the two myosins showed a similar affinity to F-actin with K m of 1.7 and 1.4, respectively. Limited proteolysis with α-chymotrypsin cleaved heat-denatured white croaker myosin mainly at heavy meromyosin/light meromyosin (HMM/LMM) junction, whereas walleye pollack myosin was cleaved at several sites in LMM as well as at the HMM/LMM junction.  相似文献

Abstract. Cyprinid herpesvirus 1 (CHV) or Herpesvirus cyprini was virulent for carp, Cyprinus carpio L., fry following 1 h immersion in water at 20 °C. Cumulative mortality for carp fry was 86–97% in 2-week-old common carp, 20% in 4-week-old fancy carp, and 0% in both 8-week-old common and fancy carp. The virus did not produce mortality in fry of crucian carp, grass carp or other cyprinids. It was also oncogenic in carp, inducing papillomas to the extent of 55% among both common and fancy carp fry. The neoplasms appeared 5–6 months after carp had been exposed to the virus by immersion and recurred at an incidence of 83% in carp 7·5 months post-desquamation of the tumour. The CHV was reisolated from all moribund fish and from all survivors. It also induced papillomas at an incidence of 13% in adult mirror carp and at 10% in adult fancy carp 5 months after intraperitoneal inoculation of 105 TCID50 ml-1 fish. The virus was rcisolated only from the ncoplastic tissue and not from internal organs. The neoplasms were normally located on fin, skin or mandible, at the intraperitoneal inoculation site. Specific fluorescence for CHV antigen was frequently detected in the gills, liver, kidneys and intestine of 2-week-old fry from 3 to 21 days following challenge with CHV. It was found in greater concentrations in experimentally induced papillomata on 2-week-old carp fry survivors examined 24 weeks after challenge than in naturally occurring neoplasms.  相似文献

ABSTRACT:   Annual changes in the gonadosomatic index (GSI), oocyte diameter, gonadal histology and plasma estradiol-17β (E2) levels were investigated in female common Japanese conger Conger myriaster in captivity. Juveniles were caught in September 1999 and reared in seawater at temperatures ranging from 10–20°C for 3 years. All fish were immature when captured in September 1999. GSI and oocyte diameter increased from October 2000, peaked between summer and autumn 2001, and bottomed-out in winter 2001. Plasma E2 level also increased from October 2000, but remained high. The ovarian developmental stage was at the peri-nucleolus stage or the oil droplet stage until September 2000. Vitellogenesis started in October 2000 and oocytes progressed to the tertiary yolk globule stage by summer 2001. However, vitellogenic oocytes regressed in all females after autumn 2001. The neogenetic oocytes began to increase after November 2001 and ovarian development progressed in 2002 as they did in 2001, although maximum GSI in 2002 was half its 2001 value. These data indicate that ovarian development in the common Japanese conger has an annual periodicity, and that these congers may be able to spawn in multiple years under rearing condition.  相似文献

ABSTRACT:   Thermal tolerance was studied in a rainbow trout strain successively selected through high temperature breeding at 20–27°C since 1966 in Miyazaki Prefecture, Japan. The hatching rate and fry mortality at high temperatures were examined in the selected strain along with normal strains cultured at water temperature of 9–17°C. The hatching rate of embryos fertilized at either 10 or 14°C and subsequently subjected to high temperatures in the blastula or neurula stage of the selected strain, was marginally higher than that of the normal strain counterparts. The upper 50% lethal temperatures (LT50) for embryos in the early segmentation, blastula and neurula stages of the selected strain were also higher than those of the normal strain counterparts. Death temperatures and LT50 of fries acclimated to 20°C of the selected strain were significantly higher than those of the normal strains. However, no difference in the critical thermal maximum was detected between the different strains. These results suggest that the selected strain of rainbow trout established by selecting successively for many generations at high temperatures acquired a degree of thermal tolerance.  相似文献

Copepodite stage V Calanus finmarchicus were collected at locations on the continental shelf north of Scotland, in the Faroe–Shetland Channel and west of Ireland on six occasions covering winter, spring and summer from October 1993 to June 1995. Oxygen consumption by the overwintering and active spring/summer population of animals was determined at temperatures close to in situ temperatures. Laboratory measurements of oxygen consumption were also made at standardized temperatures (0°C, 5°C, 7°C and 12.5°C) to determine the sensitivity of animals to temperature change in the different seasons. Rates of oxygen consumption were very low (7–30 μmol O2 gC−1 h−1) at in situ temperatures during the winter and early spring and significantly higher (105–219 μmol O2 gC−1 h−1) for the active surface population in May and June. Animals collected from the overwintering population showed no significant response to changes in temperature. Due to the low respiration rates, the calculated rate of decrease in carbon content in diapausing copepodite stage CV was very low (approximately 0.250 μgC day−1). The respiration rates were used to construct a model to estimate survival of the animals with an initial carbon content equivalent to that expected of animals in October. The results showed that in order to survive during winter and have enough energy for moulting and migration to the surface in the spring, these animals have to live at temperatures close to 0°C and be in a diapause state.  相似文献

ABSTRACT:   The effects of temperature changes on the endogenous rhythm of oxygen consumption in the Japanese flounder Paralichthys olivaceus which were acclimated to 3, 14 or 20°C in advance were investigated. The oxygen consumption rate (OCR) of individuals (total length >28 cm) that were acclimated to 3°C, maintained a relatively low value of 6.9 ± 2.3 mL (mean ± SD) O2/kg ww (wet weight)/h till 5.8°C, which is considered a 'minimum' rate. Thereafter, the OCR gradually increased up to 18.7 mL O2/kg ww/h at the temperature of 10.6°C. The OCR of individuals acclimated to 14°C stabilized with 124.2 ± 24.1 mL O2/kg ww/h, when tested at the relatively constant temperature of 14.4 ± 0.17°C. Maximum entropy spectral analysis revealed that the peak OCR occurred at 23.9 h intervals, which could be a circadian rhythm. The mean OCR showed a peak at 06.00–08.00 hours. For the temperature above 25°C in which the fish were acclimated to 20°C, the rhythmic patterns of OCR were observed until the experimental temperature reached 26.4°C. For the temperature above 28.4°C, the metabolic activity rhythm of the OCR was dampened and then the OCR abruptly rose again. It is assumed that the oscillator of the endogenous rhythm lost the governing of normal metabolic activity in Japanese flounders above 28.4°C. These results suggest that the temperatures of around 5.8, 26.4 and 28.4°C are the 'lower limit temperature', 'upper incipient lethal temperature', and the 'critical thermal maximum', respectively, for the endogenous rhythm of oxygen consumption in the Japanese flounder.  相似文献

ABSTRACT: Temperature and salinity ranges in which Gymnodinium catenatum (Hiroshima Bay strain) showed specific growth rates higher than 0.2/day were approximately 20–30°C and 20–32. The specific growth rate (μ), expressed as a polynomial equation as functions of temperature ( T ; °C) and salinity ( S ) were μ = (−6.84 × 10−4 T 2 + 0.0354 T – 0.213) × (−1.03 × 10−3 S 2 + 0.0579 S – 0.548)/0.31; the maximum growth rate (0.31/day) was obtained at 25°C and 30. From a comparison with field data recording temperature, salinity and light intensity, this species may be expected to bloom from summer to autumn in Hiroshima Bay.  相似文献

ABSTRACT:   Biomass and production of Euphausia pacifica off south-eastern Hokkaido (41–43°N), Sanriku (38–41°N), and Joban (36–38°N) were investigated every two months from March 1997 to February 1998. High biomass was found in summer–autumn off south-eastern Hokkaido, and in the late winter–early summer off Sanriku and Joban. Annual mean biomass was 381, 314 and 258 mg C/m2 off south-eastern Hokkaido, Sanriku, and Joban, respectively. The total production (sum of flesh, moults and eggs) during the survey period off south-eastern Hokkaido (3829 mg C/m2) was comparable with that off Sanriku (3872 mg C/m2); both were much higher than that off Joban (2243 mg C/m2). Somatic production during the survey period contributed the highest proportion (51.5–70.9%) to the total production in each coastal area. The somatic production–biomass ratio in each coastal area (5.2–6.9) was an intermediate value among 25 populations of 16 euphausiid species previously reported. This effect resulted from the negative factor of stagnated growth of adults during summer–winter, coupled with positive factors including continuous occurrence of larvae which show high growth rate throughout the year off Sanriku and Joban, and the numerous larvae occurring in October off south-eastern Hokkaido.  相似文献

Carp undergo temperature acclimation of respiratory function by altering mitochondrial ATP synthase (FoF1-ATPase) both quantitatively and qualitatively (Itoi et al. 2003). To address such acclimation temperature-dependent changes of FoF1-ATPase activity, we investigated in this study the correlation between the fatty acid composition and FoF1-ATPase activity in fast muscle of thermally acclimated carp. The quantities of saturated fatty acids of mitochondria from carp acclimated to 10 °C were significantly lower than those of carp acclimated to 30 °C. While mono- and poly-unsaturated fatty acids tended to increase with cold acclimation of carp, the molar concentration of 16:0 aldehyde in mitochondria from the 10 °C-acclimated carp were less than those from the 30 °C-acclimated fish. The specific activities of FoF1-ATPase in the 10 °C- and 30 °C-acclimated fish mitochondria were calculated to be 167±22 and 56±10 nmol/min ⋅ mg mitochondrial protein, respectively, the difference being significant at P<0.005. Taken together, the increase in FoF1-ATPase activity in fast muscle mitochondria of carp after cold temperature acclimation may be closely related to the increase of unsaturated fatty acids in mitochondria. Malate dehydrogenase isozymes of grass carp,Ctenopharyngodon idella, were identified by mitochondrial preparation and thermal denaturation. The structural and kinetic characteristics of chromatographically separated thermostable and thermolabile cMDHs were different in (1) half-life at 42°C, 10 min and 24 h, respectively, (2) optimal substrate, oxaloacetate and malate, concentrations, and (3) the apparent Michaelis-Menten constants of NADH and oxaloacetate.Total MDH activity in white muscle of 11°C-acclimated fish was about twice that of the 30°C-acclimated group. In addition, the ratio of the thermostable to thermolabile cMDH activity in white muscle of 30°C-acclimated fish was significantly higher than that of 11°C-acclimated fish. These results suggest that temperature acclimation can induce temperature compensation in MDH activity and differential expression of thermostable and thermolabile cMDH isozymes in freshwater fish.  相似文献

Abstract. Electrophoretic analysis of six types of tissue (serum, erythrocyte, liver, kidney, muscle and eye) and seven different isoenzymes (LDH, MDH, Est, ADH, IDH, α-GPDH and SOD) from both healthy grass carp, Ctenopharyngodon idellus Cuvier & Valenciennes, and carp affected by haemorrhagic disease was performed. It was found that: (1) Enzymes from diseased grass carp showed the most drastic changes within each tissue. The absolute activity of LDH in serum from the diseased fish increased to approximately twice that of the healthy fish; whereas the relative activity of A4 and A3B1, the main components of A sub-base in both serum and erythrocytes, was accelerated. (2) The isoenzymic changes in the control groups, diseased fish, artificially infected fish and healthy fish from diseased ponds indicated that biochemical changes occurred in the tissues before pathological change.  相似文献

ABSTRACT:   The aim of this study was to establish effective procedures for chromosome manipulation in greenling Hexagrammos otakii Jordan et Starks, which has enormous aquacultural potential. To accomplish this, temperature-dependent measurements of the mitotic intervals (τ0) were carried out. The τ0 in this fish was determined by averaging the duration of the first and third embryonic divisions at temperatures ranging 5–25°C. At higher temperatures, eggs developed faster and underwent more identical development. For greenling, τ0 were 341.1 ± 3.60 min at 5°C, 275.5 ± 4.53 min at 10°C, 189.7 ± 6.93 min at 15°C, 99.2 ± 8.27 min at 20°C and 34.2 ± 8.74 min at 25°C. There were strong, negative correlations between the τ0 and water temperatures at all temperatures studied ( Y  = −79.3 X  + 425.3, R 2 = 0.9968, where Y is the mitotic interval and X is the temperature).  相似文献

ABSTRACT:     The acute toxicities of boron were examined for Japanese flounder Paralichthys olivaceus and red sea bream Pagrus major in terms of fish size and water temperature. Japanese flounder of 0.1–70.0 g and red sea bream of 0.6–20.3 g were exposed to different concentrations of boron for 96 h at 20°C under semi-static conditions. In both fish species, the median lethal concentration (LC50) for 96 h of boron increased linearly with increasing fish size, ranging from 108 to 252 mg B/L for the flounder, and from 97 to 172 mg B/L for the sea bream. The effect of water temperature on acute toxicity was examined for Japanese flounder of 0.6 and 1.5 g at 10, 15, 20 and 25°C, and for red sea bream of 0.6 and 2.4 g at 12, 15, 20 and 25°C. The toxicity of boron for the flounder increased linearly with increasing water temperature. The 96 h LC50 values ranged from 299 to 108 mg B/L for the 0.6 g flounder and from 350 to 113 mg B/L for the 1.5 g flounder. A similar trend was shown for the 2.4 g red sea bream; however, the relationship for the 0.6 g red sea bream was not significant.  相似文献

Abstract –  Habitat and diet of Arctic charr Salvelinus alpinus (L.) were studied by monthly sampling from late autumn to early summer in Linnévatn, Svalbard (78°3'N, 13°50'E). This is the first year-round study of a population of charr in the High Arctic, with samples being taken every 5–7 weeks. The ice cover lasted for more than 9 months, from mid-October to late July, with the greatest thickness in mid-May. Although most charr occupied the littoral zone during winter, the highest densities in April and October were found in the deeper areas (20 m) of the lake. The fish fed at all times of the year, but the number of stomachs with food and the stomach-filling indices were lowest during the darkest part of the season. The diet of smaller charr (<15 cm) varied strongly with season, showing a dominance of zooplankton in late autumn and chironomids in winter (larvae) and summer (pupae). The food choice was in accordance with the density of food items available. Larger fish (≥15 cm) were mostly cannibalistic during the entire year.  相似文献

Tank respirometry was used to study the interactive effects of protein:lipid level (55%:11% vs. 42%:16%; both diets isoenergetic) and temperature (11, 6 and 2 °C) on the magnitude and duration of specific dynamic action (SDA) in juvenile Atlantic cod and haddock. The protein:lipid level did not affect any measured variable. However, numerous temperature and species effects were observed. For example, although the maximum post-feeding oxygen consumption (30–50% above routine metabolic rate; RMR) and SDA duration (∼55–85 h; SDADUR) were not affected by temperature, SDADUR g−1 of food increased from 11 to 2 °C (from ∼3 to 12 h g food−1). While absolute SDA (mg O2) decreased by ∼60–65% in cod and ∼75% in haddock from 11 to 2 °C, due to a concomitant decrease in food consumption from ∼2.0% to 0.6% body mass, SDA comprised between 3.3% and 5.2% of the dietary energy content at all temperatures. Finally, RMR at 11 and 2 °C and SDADUR at 2 °C were 25–35% and 25% greater in cod, respectively, as compared with haddock. These results suggest that feeding reduced protein diets at low water temperatures is unlikely to improve the growth of these species.  相似文献

ABSTRACT: Heating temperatures of 30–40°C and KCl concentrations of 0.1–0.5 M altered the denaturation mode of carp myofibrils. In 0.1 M KCl medium, heating temperature affected the denaturation of rod more significantly than of subfragment-1 (S-1), and a slow decrease in solubility at 30°C was accompanied by a slow denaturation of rod. KCl concentration at heating altered the denaturation mode differently at 30°C and 40°C. Increased KCl concentrations for heating reduced the rod denaturation rate at 40°C, but it was increased at 30°C. At concentrations above 0.3*Τ*M KCl, the denaturation rate for rod became identical to that for S-1 at both temperatures. Upon heating of chymotryptic digest of myofibrils, S-1 denaturation was similarly detected as in intact myofibrils, whereas practically no rod denaturation was detected. Thus, it was concluded that myosin structure connecting S-1 and rod has an important role in the denaturation process.  相似文献

Abstract – There has been little investigation of the winter ecology of adult trout during winter, especially in regard to concealment behavior. We compared day vs night underwater counts of adult rainbow trout and brook trout from four streams. At water temperatures between 1°C and 9°C, daytime counts accounted for 44% and 16% of nighttime snorkeling counts for rainbow trout and brook trout adults, respectively. As winter progressed, nighttime counts declined more so for brook trout than rainbow trout, but the decline was not significant for either species. Nocturnalism of both species was higher in streams with colder water temperatures. We observed few fish within concealment structure; however, by electrofishing concealment habitat during the day, we captured 10 times more adult trout than we counted immediately beforehand by snorkeling. ABSTRACT:   Fast skeletal muscles of Japanese flounder Paralichthys olivaceus and red sea bream Pagrus major were examined for quantitative and qualitative changes of mitochondrial ATP synthase (FoF1-ATPase) in association with rearing temperatures. The specific activities of FoF1-ATPase from Japanese flounder reared at 10°C, 15°C and 25°C for 4 weeks were determined to be 81 ± 11, 74 ± 13 and 83 ± 11 nmol/min·mg mitochondrial protein, respectively. The corresponding activity from red sea bream reared at 8°C for 5 weeks was determined to be 65 ± 9 nmol/min·mg mitochondrial protein, which was higher than 33 ± 9 nmol/min·mg mitochondrial protein in fish reared at 23°C. The contents of α- and β-F1-ATPase in total mitochondrial proteins were not significantly different between fish reared at different temperatures for the two fish species. However, the contents of β-F1-ATPase in the total fast skeletal muscle extracts, prepared from Japanese flounder reared at 10°C, were 2.1- and 2.9-fold higher than those for fish reared at 15°C and 25°C, respectively. The corresponding content from red seabream reared at 8°C was 2.2-fold higher than that for fish reared at 23°C. Therefore, the changes in FoF1-ATPase depending on rearing temperatures were species-specific.  相似文献

为了解流溪河光倒刺鲃国家级水产种质资源保护区鱼类群落结构,分析不同河段、不同季节鱼类群落多样性的变化,于2017年12月-2018年10月对保护区上、中、下游的鱼类开展了每个季度一次的调查。调查结果显示,共采集鱼类57种,隶属于5目14科50属,其中鲤形目43种、鲈形目6种、鲇形目5种,合鳃鱼目2种,鳉形目1种;计算57种鱼类的相对重要性指数,结果显示,尼罗罗非鱼、越南■及鲤鱼为2017-2018年度优势种;Shannon-Wiener多样性指数、Margalef丰富度指数和Pielou均匀度指数计算结果显示,下游鱼类群落的多样性及均匀度最高,夏季鱼类群落的多样性、丰富度及均匀度最高;各监测点之间的种类相似性系数显示,保护区上中游、上下游之间鱼类种类组成为中等不相似,中下游种类组成为中等相似,春夏、春冬、夏冬及夏秋季节鱼类种类组成为中等不相似,春秋、秋冬季节为中等相似;ABC分析结果显示,春、夏及冬季鱼类群落处于稳定状态,秋季处于中度干扰状态。  相似文献
