对1株从不表现鸡新城疫临床症状,只引起产蛋下降的鸡群中分离的新城疫野毒株(PMV-C02)开展了研究。经测定,该毒株的颅内接种致病指数(ICPI)为1.85,静脉接种指数(IVPI)为2.42,确定为强毒型。对该病毒的融合蛋白型解基因的核酸序列进行了测定,其推测的F1/F2裂解位点附近的氨基酸序列为^112R-R-Q-K-R^116,这一序列附合典型鸡新城疫强毒^112P/K-R-Q-K/R-R^116,的普通规律,确定该毒株不是超强毒株。为进一步了解该病毒对鸡的非典型发病的原因,我们通过人工发病方法对不同ND抗体水平的产蛋鸡进行了攻毒试验。结果该毒株致使ND抗体阴性或抗体水平极低的蛋鸡(HI效价<=1:5)发病致死并呈现典型的ND病症和病理变化,低抗体水平鸡(HI效价1:10-1:40)呈现一过性产蛋停止,并在2周后恢复,未表现出典型的ND症状,高水平抗体鸡(HI效价1:80-1:320)则未见异常。从而推断抗体抑制为引起本病毒非典型ND发病的原因而非毒株变异引起。  相似文献

使用鸡新城疫病毒LaSota株和产蛋下降综合征病毒分离株HE03株制备鸡新城疫和产蛋下降综合征二联灭活氢氧化铝胶疫苗。把40只16周龄SPF蛋鸡分成A、B 组,分别是30只、10只。A组接种0.5 mL鸡新城疫和产蛋下降综合征二联灭活氢氧化铝胶疫苗,B组作为对照组,每隔14 d采集血清样本直到接种后第6周。至22周龄时,A组再次分成A1和A2两组。A2组与B组的鸡接受强致病性的产蛋下降综合征病毒攻击。结果表明,A组在不同时间采集的血清抗鸡产蛋下降综合征病毒的最高的抗体出现在接种后的第4周,抗鸡产蛋下降综合征病毒的HI抗体滴度从1∶128到1∶512不等,几何平均滴度(GMT)为1∶111;第4周时,抗鸡新城疫病毒的HI抗体滴度GMT为1∶134。该疫苗能保护鸡抵抗强致病性的产蛋下降综合征病毒和鸡新城疫病毒的感染,B组在攻毒感染后产蛋率显著下降,出现畸形蛋、软壳蛋和无壳蛋。  相似文献

以新城疫(ND)Ⅰ系苗对非典型ND污染场的产蛋鸡及一月雏鸡进行紧急预防接种,不仅安全可靠,产蛋鸡群的HI抗体水平很快由50%的鸡呈高滴度(1:1024)转变为60%的鸡呈正常免疫水平(1:64),且可保持100天以上,产卵率明显上升,死亡数显著减少。在此基础上,制定了科学的免疫程序,经一年的观察,再无ND发生。  相似文献

将新城疫病毒(NDV)Colon-30株和减蛋综合征病毒(EDS76AV)-127株分别接种于非免疫鸡胚和鸭胚,制备抗原液。抗原液经甲醛灭活后,与纯化的天然免疫增强剂蜂胶乳化,制成鸡新城疫(ND)-减蛋综合征(EDS76)蜂胶二联灭活疫苗,对疫苗的安全性和免疫原性进行了测定。对3-6周龄易感雏鸡以2倍使用剂量肌内注射,安全性良好;对1日龄雏鸡接种1个使用剂量进行急性毒性试验。15d生长曲线表明,对雏难 影响。对6周龄雏鸡注射1个使用剂量,每批疫苗免疫10只鸡,免疫后5-32d,鸡血清中EDS76血凝抑制(HI)抗体的几何平均滴度(GMT)为6.5-11.8log2,ND血凝抑制(HI)抗体的几何平均滴度(GMT)为4.8-9.8log2;对ND免疫攻毒试验表明,3批疫苗,每羽份含有的半数保护量(PD50)分别为≥127、≥131和≥127。  相似文献

为预防非典型鸡新城疫(CND),选母抗 HI 价1:16以上的2~10日龄雏鸡,以肌注 NDⅡ系苗800个免疫最进行首免后一个月,HI 价达1:16以上者达100%.此时用ND 中等毒力 MVK teswar 株Ⅰ系苗2000免疫量加强免疫,其 HI 价达1:16以上者,免疫后3、6、10个月均为100%,13个月为96.2%,16个月为100%.13及16个月免疫鸡攻 NDV 强毒(100个 MLD)可获全数保护.用接种13个月的免疫鸡与强毒对照鸡同居,不发生接触感染.以Ⅱ系苗800个免疫量首免Ⅰ系苗2000免疫量加强免疫共接种92.3万只鸡。未发现 CND;从中抽取102只免疫鸡做HI 试验,HI 价达1:16以上者为93.1%.可见,以Ⅱ系苗800个免疫首免,Ⅰ系苗2000个免疫量加强免疫的程序,能使生产鸡群在全程(16个月以上)保持较高的抗体效价,可防止 CND 的发生.  相似文献

新城疫单抗ELISA试剂盒监测免疫鸡群中新城疫强毒   总被引:1,自引:0,他引:1  
应用新城疫(ND)单抗ELISA试剂盒对2个免疫鸡群中ND强毒感染情况及个体感染强毒后排毒动态跟踪监测,并平等测定其HI抗体效价。共采集泄殖腔棉拭子和血液对应样品2317份。结果表明:HI效价2-14之间的个体均可检出强毒,其中HI效价在6以下和11以上的个体强毒检出率都较高。HI效价在6以上的个体仍角感染强毒,强毒感染导致HI抗体水平上升,且感染前低者上升速度较快,幅度亦较大;并且发现排毒个体的高水平抗体是强毒感染所致。个体感染强毒后排毒过程可长达3周,而且感染前抗体水平低者排毒时间相对较长。强毒一旦侵入鸡群便可在群内巡回传播,长期维持下来。  相似文献

给两组二次新城疫(ND)疫苗免疫的鸡群分别于40日龄、60日龄经胸肌注射强毒株F48E8(剂量为0.25mL),观察排毒和其HI抗体效价。结果表明:鸡在感染强毒后第2天开始排毒,在第3~6天和第11~14天出现两次排毒高峰,整个排毒过程可达3周,其中抗体水平较低者排毒时间相对较长,频率较高;强毒感染初期,个体HI抗体先下降1~3个滴度,然后上升,且感染前低者上升速度较快,幅度亦较大。  相似文献

对2007年3月~2008年5月泰安及周边地区鸡群新城疫抗体水平检测结果进行了统计分析,统计结果显示:抗体效价超过正常免疫水平且离散度较大,疑似新城疫野毒感染的鸡群较多.其中肉仔鸡(或麻鸡)群、育成蛋鸡群分别为12.3%、21.5%,而产蛋鸡群高达51.2%.采用接种鸡胚分离病毒和间接免疫荧光试验验证HI试验检测结果的判定,探讨鸡群的带毒状况.调查结果表明,目前鲁中地区各种鸡群的新城疫感染带毒状况较为严重,高比例带毒鸡群的存在是我国控制新城疫的最大难题.  相似文献

用不同方法测试了从新城疫患鸽分离到的新城疫病毒(NDV)野毒PB9601株的致病性。结果,该毒株对1日龄SPF雏鸡的脑内接种致病指数为2.00,对6周龄SPF鸡静脉接种致病指数为0.00;该毒株对6周龄左右鸽有很强的致病性,有的血清中已有一定量的抗鸽NDV抗体;经SPF鸡胚传13代的尿囊液病毒对鸽的半数致死量约为12个TCID50。由此认为,PB9601株NDV是对鸡致病性很弱但对鸽呈高度致病性的毒株,可作为国内鸽NDV强毒的参考株。  相似文献

The role of ducks in the transmission of Newcastle disease virus (NDV) to free-range village chicken was investigated experimentally. Newcastle disease (ND) seronegative ducklings reared in a pen were infected oronasally with velogenic NDV of intracerebral pathogenicity index (ICPI) 1.8 isolated from outbreaks in village chickens in Uganda. A first group of 3-week-old ND seronegative chicks was mixed with the ducks and they were kept together for 7 days. Both ducks and chicks were observed for ND clinical signs and any mortality, and they were bled and their sera were tested for ND antibodies by haemagglutination inhibition (HI) test. The chicks were removed, euthanized and examined for any ND lesions, while the ducks were transferred to a fresh pen and a second group of chicks was introduced and observed and treated as above. The ducks and the chicks tested positive for ND antibodies 7 days post infection and contact, respectively, but showed no clinical signs, post-mortem lesions or mortality. The mean ND antibody titre of the second group of chicks was lower than for the first group. This study has shown that although ducks can be infected with velogenic NDV, they do not show clinical signs but are able to transmit NDV to in-contact chicks. Further investigations are needed of the lack of clinical signs in the in-contact chicks and how long the ducks remain infective.  相似文献

应用电镜技术、血清学检查、生物学试验等方法,从未注射过疫苗的某自然发病鸡群分离出一株新城疫病毒。该分离株对10日龄鸡胚的平均致死时间为116小时,静脉接种致病指数为0.06,其毒力介于新城疫病毒的LaSota株与V4株之间。免疫原性试验结果表明,该分离株具有免疫后无不良反应、免疫后6天开始产生抗体、产生的抗体效价高、对新城疫强毒的攻击能100%保护等特点,是一株良好的新城疫候选疫苗株  相似文献

This study was designed to evaluate the synergistic effect of VA5 immunopotentiator on pigeon ND4416 inactivated vaccine.160 healthy pigeons were randomly divided into four groups,including ND4416 strain inactivated vaccine group (NDV group),ND4416 strain inactivated vaccine and immunopotentiator (VA5) mixed group (NDV+VA5 group),La Sota inactivated vaccine group (La Sota group) and normal saline as the control group (C group),to assess their vaccine efficacy against virulent pigeon NDV by serological analysis and animal testing.Pigeons sera were collected at different time points after immunization and measured the HI antibody titer of each group.The results showed that VA5 immunopotentiator significantly improved the serum antibody level of pigeons immunized with pigeon Newcastle disease inactivated vaccine (P<0.05).In addition,comparative test of spleen lymphocyte transformation was conducted at various time points after immunization.The results indicated that VA5 effectively stimulated the lymphocyte transformation of immune pigeon.Pigeons in each groups were challenged with ND4416 strain at the 30th,90th and 180th d after immunization.The results presented that the NDV+VA5 group had 100% protection rate and higher than La Sota group.The duration of immunization test showed that the antibody titer of NDV+VA5 group reached peak 11.20log2 at the 21st d,remained 7.50log2 at 180th d,and the protection rate remained 100% at 180th d.It indicated that VA5 immunopotentiator sustained the immune duration of pigeon NDV vaccine up to 180 d.Moreover,the in vitro detoxification test results suggested that VA5 immunopotentiator reduced the in vitro detoxification cycle of pigeons after challenge.Overall,this study suggested VA5 immunopotentiator could significantly improve the immune efficacy of pigeon Newcastle disease inactivated vaccine,which provided a basis for further enhancing the efficacy of Newcastle disease vaccine,as well increased experimental data for the application of immunopotentiators.  相似文献

试验旨在探究VA5免疫增强剂对鸽新城疫病毒(NDV)灭活疫苗(ND4416株)的免疫增效作用。选取160只健康鸽随机分为4组:ND4416株灭活疫苗组(NDV组)、ND4416株灭活疫苗与免疫增强剂(VA5)混合组(NDV+VA5组)、La Sota灭活疫苗组(La Sota组)及生理盐水空白对照组(C组),进行免疫效力及免疫持续期试验。采用血凝抑制试验检测各组鸽免疫后不同时间点的血清抗体效价,结果显示,VA5免疫增强剂能够显著提高鸽NDV灭活疫苗血清抗体水平(P<0.05)。脾脏淋巴细胞转化试验结果显示,VA5能够有效刺激免疫鸽的淋巴细胞转化。在免疫后第30、90、180天,各组鸽子采用ND4416株进行攻毒试验,结果显示,NDV+VA5组在3个时间点的攻毒保护率均为100%,均高于La Sota组。免疫持续期试验结果显示,NDV+VA5组血清抗体水平在免疫后第21天达峰值11.20log2,第180天仍能达到7.50log2,攻毒保护率达100%,表明VA5免疫增强剂可延长鸽新城疫疫苗的免疫效力持续期至180 d。攻毒后体外排毒检测结果表明,VA5免疫增强剂能够缩短攻毒后鸽的体外排毒周期,减少体外排毒。综上,VA5免疫增强剂能够显著提高鸽NDV灭活疫苗的免疫效果,为研制效果更好的鸽NDV疫苗提供了基础依据,同时也为免疫增强剂的应用增加了试验数据。  相似文献

为了分析免疫鸡群中新城疫强毒感染流行的原因,明确抗体效价与流行株感染排毒率之间的关系,本研究以LaSota为抗原制备新城疫灭活疫苗,并以0.02mL和0.4mL的量分别免疫3周龄的SPF鸡10只。免疫后7、14、21d分别测定免疫鸡血清中的抗体HI效价。免疫后21d以基因Ⅶd亚型新城疫流行株JS5/05进行攻毒,攻毒后每天观察试验鸡的临床症状,并于攻毒后3、5、7d采集试验鸡的喉气管与泄殖腔棉拭样品进行病毒分离,结果显示,免疫3周后0.02mL和0.4mL疫苗免疫组鸡的血清HI抗体平均效价分别为5.4log2和8.2log2;0.02mL免疫组在攻毒后的排毒率达到100%,且排毒时间较长,而0.4mL免疫组在攻毒后的排毒率明显降低,且排毒时间较短。上述结果表明新城疫抗体效价与流行株感染排毒率之间存在明显的负相关。  相似文献

1-day-old SPF chickens and commercial Jingfen chickens were vaccinated with IBD immune complex(IC) vaccine, NDV La Sota vaccine were inoculated simultaneously every one week and every two weeks.NDV La Sota immunization alone was as the control group.At the 2nd, 3rd, 4th and 5th week post inoculation, blood samples were taken and the ND HI antibody were tested.Experimental chickens were challenged with high virulent NDV at the 5th week post inoculation, the protective rate of each group was calculated.The results showed the ND HI antibody were not significant different in the combined immunization of IBD IC vaccine priming and NDV La Sota vaccine boost and NDV La Sota vaccine alone immunization (P>0.05).The results indicated that IBD IC vaccine has no immunosuppression on NDV La Sota vaccine in SPF chickens and commercial Jingfen chickens.  相似文献

鸡新城疫免疫鸡群强毒感染与HI抗体水平关系的研究   总被引:3,自引:1,他引:2  
应用RT-PCR方法对鸡新城疫4种不同免疫程序鸡群NDV强毒的感染进行监测。结果显示,鸡新城疫弱毒疫苗和油乳剂灭活疫苗同时接种的免疫鸡群NDV强毒的感染率最低。同时应用HA-HI试验对这4群鸡进行平行抽样检测其抗体效价,发现鸡群免疫水平整齐且平均HI效价在11 log2以上时,免疫鸡群基本不感染鸡NDV强毒。  相似文献
