针对水产品四环素残留的便捷化精准检测难题,该研究研制了一种四环素含量检测的便携式直读光电化学装置,并探究了装置用于鱼肉四环素残留检测的可行性。装置分别以LED和STC89C52单片机为激发光源和控制核心,利用设计的光源暗室避免外源光照对检测结果的影响。测试结果表明,LED激发光源的输出信号稳定,装置对于光电流信号的采集具有较好的准确性。进一步基于丝网印刷电极构建光电化学适配体传感器,采用所研制装置对信号进行采集与处理,最终以OLED显示器直接读取抗生素浓度信息。该方法对于四环素检测的线性范围为1×10-11~1×10-8mol/L,检测限为3×10-12 mol/L,具有较好的选择性,且对于鱼肉四环素残留检测的回收率为92%~94%,为水产品四环素的精准现场分析提供了新的方法与途径。  相似文献

Volatile phenols produced by Brettanomyces dekkera have been associate with off-flavors of wines. A versatile liquid chromatography-tandem mass spectrometry together with an HPLC-DAD-fluorescence methods were developed for the quantitation of two phenols, 4-ethylphenol (4EP) and 4-ethylguaiacol (4EG), in red and white wines. For LC-MS-MS analysis, fortified wines were directly injected after a dilution with methanol, and levels of phenols were measured by monitoring the multiple reaction (MRM) transitions of precursor ions mass charge (m/z) 121 --> 106 for 4EP and (m/z) 151 --> 136 for 4EG. Qualitative and quantitative confirmation data were acquired simultaneously by monitoring alternative MRM transitions following an external standard method. Calibration was linear over a working range of 10 and 5000 microg/L. Limit of determination (LOD) and limit of quantification (LOQ) were 10 and 50 microg/L for both 4EG and 4EP. HPLC analysis phenols were separated with a gradient system of acetonitrile-water and detected using a diode array detector (DAD) at 280 nm, and for the fluorescence analysis, excitation and emission wavelengths of 260 and 305 nm were used. Quantitative analysis of 4EP and 4EG was performed by the standard addition method to avoid matrix interferences. Calibration was linear over a concentration range from 10 to 5000 microg/L for HPLC-DAD, from 1 to 10,000 microg/L for 4EP, and from 10 to 10,000 for 4EG for fluorescence analysis. LOD and LOQ for the DAD analysis were 10 and 50 microg/L for both 4EG and 4EP. For fluorescence analysis, LOD and LOQ were 1 and 5 microg/L for 4EP and 10 and 50, respectively, for 4EG. The proposed methods can be easily used for the qualitative and quantitative determination of 4EP and 4EG in wines affected by microbial contamination with yeasts of the Brettanomyces genus.  相似文献

Bovine somatotropin (bST) and insulin-like growth factor-1 (IGF-1) are peptide hormones that are involved in the regulation of milk production in dairy cows. Because these hormones are present at extremely low concentration in fresh and processed bovine milk, a highly sensitive and specific electrochemiluminescent immunoassay (ECLIA) has been developed to better estimate the concentration of these hormones in milk. The assay employs an imager, a capture antibody bound to a carbon electrode, and a detection antibody coupled to a ruthenium label. In the presence of tripropylamine and an electric pulse, ruthenium generates light proportional to the amount of antigen bound, and the light is captured as signal by a charge-coupled device (CCD) camera. Using bovine milk as the starting matrix, 99.69% of bST and 104.79% of IGF-1 were recoverable. The limit of detection (LOD) was <5 pg/mL for bST and <1 pg/mL for IGF-1. The limit of quantification (LOQ) was <14 pg/mL for bST in milk and <2 pg/mL of IGF-1. The assay is highly specific and shows <0.2% cross-reactivity with other peptide hormones found in bovine milk such as insulin and IGF-2. These data indicate this new, ECLIA is highly sensitive and specific for estimating the concentration of bST or IGF-1 in milk.  相似文献

Very few methods for detecting residues of pesticides in food or agricultural samples have undergone rigorous colloborative study and possess official AOAC status. The Chemical Residue Laboratory has formalized a method validation scheme to use when incorporating or developing new, unofficial methods. These methods are validated by assessing certain performance parameters: scope, specificity, linear range, accuracy, precision, limit of detection (LOD), and limit of quantitation (LOQ). For accuracy and precision assessment, 12 replicate fortifications must yield recoveries within the range of 70-120% with a coefficient of variation (CV) that compares favorably to the Horwitz CV. LOD and LOQ are equivalent to 3 and 10 times, respectively, the background signal contributed by a sample matrix blank. This criterion that we use for LOD/LOQ is not universal. In fact, because of differing definitions, we have encountered difficulties in enforcing a tolerance by using a registrant's method. This paper also presents an example of our method validation scheme, using a recent method development project for detecting sulfamethazine in raw milk. The sulfamethazine project also revealed unanticipated personnel problems, underscoring the importance of the human factor in quality assurance.  相似文献

植物利用约400~700 nm波段的光驱动光合作用,但不同波长的光驱动效率不相同,而且随着植物类型及生长阶段的不同而变化。因此,准确获取被植物捕获并用于驱动光合作用的光辐射成为困扰科学家的难题。当前,光量子传感器被普遍接受并用于评价光合作用潜力,可测量400~700 nm波段的光量子通量密度或光量子通量,其光谱响应函数为直线。该文回顾了经典光合有效辐射(photosynthetically active radiation,PAR)定义的形成过程,介绍了PAR传感器的演化路径,讨论了PAR及其传感器的应用现状。由于测量对象及应用环境的多样化,PAR的定义仍然没有完全统一,且早期研究对光谱响应函数的度量不充分。随着当前人工光照明与植物生长发育相关研究的深入,发现植物光合作用吸收的光波长范围比400~700 nm要宽,不同的光谱能量分布(波长配比,能量配比)、光周期等对光合作用影响显著,并且很难将光辐射对光合作用的影响和光形态效应区分开,因此PAR的定义及其传感器的研发仍处于不断发展中。理想的PAR应该从植物光合作用的角度来定义,未来PAR传感器的光谱响应函数应与植物光合作用的能力曲线相一致,并能依据测量对象及应用需求而调整。与此相适应,未来PAR传感器应向用户可对光谱响应函数编程的方向发展。  相似文献

Spinosad is an insect control agent that is derived from a naturally occurring organism and is effective on a wide variety of crops, including citrus crops. A method is described for the determination of spinosad and its metabolites in citrus crops and orange processed commodities. The method determines residues of the active ingredients (spinosyns A and D) and three minor metabolites (spinosyn B, spinosyn K, and N-demethylspinosyn D). For dried orange pulp and orange oil, the method has a limit of quantitation (LOQ) of 0.02 microg/g and a limit of detection (LOD) of 0.006 microg/g. For all other sample matrices (whole fruit, edible fruit, juice, and peel), the method has an LOQ of 0.01 microg/g and an LOD of 0.003 microg/g. The analytes are extracted from the various sample types using appropriate solvents, and the extracts are purified by liquid-liquid partitioning and/or solid-phase extraction. All five analytes are determined simultaneously in the purified extracts by reversed-phase high-performance liquid chromatography with ultraviolet detection at 250 nm.  相似文献

针对小麦高速播种作业过程中高频排种种子流精准检测困难的问题,该研究设计了一套薄面光折射式小麦种子流多通道并行检测装置。基于将高通量变为低通量多通道并行同步检测的思路,设计了种子流分流结构。根据小麦种子物理特性,在已有传感原理的基础上,提出了一种“LED灯珠+窄缝”产生薄面光层,结合凸透镜折射原理扩大有效检测面积的方法,通过光路分析和窄缝尺寸分析确定了凸透镜焦距、薄面LED窄缝尺寸及传感元器件关键参数。利用多通道并行检测传感原理,设计了多路信号同步采集系统。为提升检测准确率,建立检测准确率-排种频率之间的关系,通过分析检测装置的误差规律,构建了准确率补偿模型。台架试验表明:排种器转速在80~180 r/min时,田间正常排种频率范围为52.10~321.55Hz,检测准确率均高于96.68%。田间播种试验表明:在2~9km/h的小麦播种机作业速度下,田间排种频率为67.65~323.95Hz,检测装置检测准确率高于95.28%。检测装置能够检测排种器的排种频率、各通道排种量、排种总量。正常田间小麦播种作业中机械振动、强光照和土壤粉尘对检测装置没有明显影响。该检测装置可为小麦高速播种作业中...  相似文献

A simple and rapid method based on microwave-assisted extraction (MAE) coupled to gas chromatographic analysis was developed for the analysis of triazine (atrazine, cyanazine, metribuzine, simazine and deethylatrazine, and deisopropylatrazine) and chloroacetanilide (acetochlor, alachlor, and metolachlor) herbicide residues in soils. Soil samples are processed by MAE for 5 min at 80 degrees C in the presence of acetonitrile (20 mL/sample). Mean recovery values of most solutes are >80% in the 10 to 500 microg/kg fortification range with respective RSDs (relative standard deviations) < 20%. The limits of quantification (LOQ) and limits of detection (LOD) are 10 and 1 to 5 microg/kg, respectively. The method was validated with two types of soils containing 1.5 and 3.0% organic matter content, respectively; no statistically significant differences were found between solute recovery values from the two types of soils. The solute mean recovery values from freshly spiked (24 h aging) and spiked samples stored refrigerated for one week before processed were also not statistically different. Residue levels determined in field weathered soils were higher when soils were processed by MAE than with a comparison method based on flask-shaking of soil suspensions overnight. 为研究不同LED红蓝光质比对水稻幼苗生长和生理特性的影响,以湘早籼45号为试验材料,通过在白光的基础上添加不同光质比例的红光(主波长662 nm)和蓝光(457 nm),设置T1(白光:蓝光=2:1)、T2(白光:蓝光=1:2)、T3(白光:红光=7:1)、T4(白光:红光=2:1)、T5(白光:红光=3:2)、T6(白光:红光=1:2)共6种不同光质配比,以白光为对照(CK),探究不同光质比对水稻幼苗形态指标和生理特性的影响。结果表明,增加红光比例提高了水稻幼苗株高、倒二叶叶长、叶面积、壮苗指数、地下部鲜重、根总长、根总数、根表面积、叶绿素含量和叶片过氧化氢酶(CAT)活性。增加蓝光比例抑制了水稻幼苗的株高、第一叶叶鞘长,但在蓝光作用下水稻幼苗茎基宽、倒二叶叶宽、叶面积、壮苗指数、幼苗生物量、根总长、根总数、根表面积、根体积、叶绿素含量增加。T2的茎基宽和壮苗指数最大,相比CK分别增加了23.1%和51.6%。T3和T4有利于生物量积累以及叶面积和叶片抗氧化酶活性的提高,相比CK,T3和T4叶面积分别提高46.9%和45.6%。综上,LED红光提高了水稻叶片叶面积和抗氧化酶活性,促进幼苗干物质积累和根系系统的发育,蓝光则提高了茎基宽和壮苗指数,对改善水稻幼苗形态和培育壮苗具有重要意义。本研究为优化水稻工厂化育秧光照参数提供了理论依据。  相似文献

为了对播种机播种质量进行监测,实现精量播种,该文采用红外检测技术,设计了一种对射式传感器,研制了配套播种质量监测系统,并进行了田间播种作业试验研究。该监测系统由多路传感器、监控终端、播种传动传感器组成,结构合理,安装方便,对播种过程出现的堵塞、漏播、缺种等现象,可发出声光报警信号,及时指示故障的位置。田间试验中监测数据与实际测量对比结果表明:该监测系统能够适应田间工作环境,播种质量监测准确率可达95%以上,能够解决人工难以实时监测播种质量的问题,可避免漏播断条现象发生,提高播种机的工作效率和播种质量。  相似文献

A method was developed for the quantification of alternariol and alternariol monomethyl ether on tangerines with and without symptoms of Alternaria brown spot disease. The method employs solid-phase extraction for cleanup, followed by high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS) for detection. This method was validated on flavedo (exocarp or epicarp, exterior yellow peel) and on albedo tissue (mesocarp, interior white peel). An excellent linearity over a range of 0.50-20.0 mg/kg was achieved, with r2 >or= 0.997. The limits of detection (LOD) and quantification (LOQ) were fewer than 0.13 and 0.50 microg/kg, respectively. 为探究LED蓝光处理对辣椒采后贮藏过程中品质的影响,采用强度为14 μmol·m-2·s-1的LED蓝光(470 nm),在温度25℃、相对湿度80%条件下,对采后新鲜绿辣椒和红辣椒分别进行蓝光照射、避光处理(对照组),测定贮藏过程中辣椒色泽、叶绿素含量、辣椒红素含量、辣椒碱含量、维生素C(Vc)含量和Ccs基因表达量等的变化。结果表明,与对照组相比,LED蓝光处理会加速绿辣椒退绿,使其叶绿素含量在第8天时降为初始值(0 d)的17%(P<0.01);LED蓝光处理可有效提高辣椒中辣椒红素的含量,红、绿辣椒中的辣椒红素含量在第8天时分别是对照组的1.60倍和5.59倍(P<0.01);LED蓝光处理可显著增加辣椒中辣椒碱含量,处理第8天时红、绿辣椒中的辣椒碱含量分别是对照组的1.53倍和1.85倍(P<0.01),Ccs基因的表达量分别为对照组的1.35倍和4.01倍(P<0.01)。此外,LED蓝光可促使辣椒中Vc含量显著升高,处理第8天时,红、绿辣椒中Vc含量分别为对照组的1.25倍和1.58倍(P<0.01)。综上,在采后贮藏过程中,LED蓝光照射可以加速绿辣椒退绿,并促使红、绿辣椒变红,显著提高辣椒红素、辣椒碱和Vc含量。本研究结果为LED蓝光在辣椒采后贮藏过程中品质保持,尤其是增加辣椒红素含量及改善辣椒色泽提供了新思路。  相似文献

联合收获机粮箱内稻谷含杂率传感器采样盒设计   总被引:2,自引:2,他引:0  
针对收割过程中通常只在开机时设置一次作业参数,不能经常停机观察籽粒的清洁度进而随时调节作业参数这一问题,需要研制一种能够自动获取联合收获机作业过程中含杂率数据的装置。但是,迄今为止,在收获过程中监测籽粒含杂率的研究还处于探索阶段。针对以上问题,结合联合收获机作业过程中谷物的流变特性,该文设计了一种谷物含杂率传感器,通过电磁铁(弹簧)控制挡板的伸出(复位),以使采样盒可视玻璃窗口前聚集(卸载)谷物,通过采样盒内CMOS相机获取数字图像,应用数字图像处理技术获得含杂率。依据图像直方图优化LED光源的照射及安装方法以获取高质量图像,分析了电磁铁提供的拉力和弹簧提供的推力、采样盒入口尺寸,采用全局阈值迭代算法,使用三个迭代步提取稻谷、茎秆和细柄连通域,计算每个连通域的像素,最终从形态上识别了谷物和杂质,同时测量稻谷千粒质量、茎秆面密度和细柄线密度,建立了计算谷物杂质质量的数学模型。结果表明:在采样盒内两侧各安装2个LED,间接照射视窗时获取的图像质量较好,有效避免了图像强度直方图中出现的峰值;额定拉力为60 N的电磁铁可以提供足够的拉力;采用线径为1 mm的弹簧能提供足够的推力,由于在收割过程中出现的短秸秆和细柄的长度在10 mm到30 mm之间,因此采样盒的设计高度为95.7 mm,宽度为76.5 mm,入口宽度为31.9 mm,视窗长度为57.1 mm,宽度为57.4 mm,如此稻谷、秸秆和细柄可以顺利流入采样盒,并且在视窗中可以看到约200粒谷粒。在不同含杂率下进行了籽粒含杂率传感器监测精度的台架试验,结果表明:该装置监测的籽粒含杂率与人工获得的籽粒含杂率具有一致的变化趋势,能够监测在0~2.88%范围内的籽粒含杂率。为了满足田间作业监测需求,同时设计了采样盒的防水防尘罩,为相机提供一个稳定抓取图像的工作环境。于2017年11月17日,在苏州九里湖进行了不同喂入量下含杂率监测试验,试验使用Yamma4LZ2.5联合收割机,结果表明,田间试验的相对误差在9.44%和19.67%之间。该研究可为田间收获过程中自动获取谷物含杂率提供参考。  相似文献

探讨全封闭饲养条件下,LED光源对肉鸡生产性能,养分表观消化率和鸡舍空气中NH3、CO2、粉尘浓度,以及空气微生物含量的影响。试验于2017年3月2日至3月30日在河南省某农业集团一大型肉鸡场进行。选用健康状况良好、体况一致的1日龄白羽肉鸡240只,采用单因子完全随机试验设计,分为2个处理组,一组采用暖白色光LED灯光源照明为试验组,另一组采用普通暖白色荧光灯光源照明为对照组。试验期间详细观察肉鸡生产情况,分别计算平均日采食量、平均日增质量、料质量比、发病率以及死淘率。于肉鸡8、15、22和28日龄当日全收粪法测定粗蛋白、粗脂肪、钙和磷的表观消化率。试验期间每天08:00,14:00和20:00,采用光化学方法分别在两栋鸡舍内测量NH3和CO2浓度,测量点分布依据五点法,测量高度为0.5 m。分别对8、15、22和28日龄的鸡舍,采用质量法测定粉尘含量,平板沉降法测定空气微生物含量。结果表明,与荧光灯相比,LED光源对8~22日龄肉鸡的生产性能、养分代谢及其生存环境质量没有不利影响。LED光源有利于提高22~28日龄肉鸡的平均日采食量和平均日增质量,分别比荧光灯光源下的肉鸡显著增加了13.13%和13.23%(P0.05)。LED光源有利于提高22~28日龄肉鸡对饲料中粗蛋白,钙和磷的表观消化率,22日龄时,分别比荧光灯鸡舍的肉鸡显著提高了9.45%,14.90%和7.48%(P0.05),28日龄时分别提高了7.30%,6.78%和8.29%(P0.05)。试验期间LED光源鸡舍内肉鸡发病率为2.45%,显著低于荧光灯鸡舍3.57%的发病率(P0.05)。LED光源鸡舍内肉鸡死淘率为1.18%,显著低于荧光灯鸡舍2.36%的死淘率(P0.05)。同时,LED光源鸡舍的25日龄以后的NH3浓度、22日龄以后的粉尘和微生物浓度均显著低于荧光灯光源鸡舍(P0.05)。因此,LED光源有利提高肉鸡的生产性能和改善肉鸡生活空间的空气环境质量,加之LED光源在养殖生产上的应用具有安全、经济、环保的特点,值得进一步推广和应用。  相似文献

Abstract

Fast urbanization and population increase resulted in the demand for ornamental seedling supply in a high efficiency under the culture of continuous artificial lighting. Light emitting diode (LED) can be considered as the replacement of high-pressure sodium (HPS) lamps by providing lights with specific wavelengths for plant growth at low energy cost. In this study, nutrient use efficiency was detected in Podocarpus macrophyllus seedlings under lighting spectra generated by white light-emitting diodes (LEDs) (red (R)/green (G)/blue (B)=8.4:75:16.6) and high-pressure sodium (HPS) lamps (R/G/B/=4.3:69.3:26.5). Compared to seedlings under the HPS spectrum, those under white LED spectrum had better growth, greater biomass accumulation, higher photosynthesis and gas exchange. In contrast, LED-spectrum treated seedlings had greater nitrogen (N) and phosphorus (P) contents than HPS-spectrum treated ones. The LED spectrum promoted nutrient use index for both N and P in seedlings compared to the HPS spectrum. The LED spectrum induced steady-state uptake of P but resulted in the dilution of N. In conclusion, white LED can be considered to replace the usage of HPS lamps for the culture of tree seedlings due to the spectrum effect on N utilization, P uptake and the use efficiencies of N and P.  相似文献

水资源是制约中国农业生产最重要的因素之一,土壤含水率的检测对农业生产实践具有至关重要的指导意义。该文根据土壤水分对不同光谱的吸收和反射特性,以发光二极管作为测量仪器光源,中心波长为1 940 nm的光为测量光,1 800 nm为参考光设计了专用的测量电路和相应的软件程序来测量土壤含水率。系统光源发出的光照射到土壤表面,经反射后光电转换器,再送至放大电路、模数转换器、显示和存储。试验表明:土壤含水率与相对吸收深度之间存在正比例关系,线性回归的确定系数为0.86。为了证明测量仪器的测量进度,将实际土壤含水率与仪器的测量结果进行比较修正,结果表明:烘干法与本仪器测量的土壤含水率的结果的均方根误差均为3.9%。因此,本次设计的测量仪器可满足对土壤含水率的测量要求,研究结果在指导农业生产,水资源合理利用,精细灌溉的实时监测中具有良好的应用前景。  相似文献

This paper investigates the electrocatalytic oxidation of (-)-epigallocatechin gallate (EGCG), the main monomer flavanol found in green tea, with a novel ionic liquid, n-octylpyridinium hexafluorophosphate (OPFP) carbon paste electrode (CPE). Due to the natural viscosity and high conductivity of OPFP, this novel OPFP-CPE exhibited very attractive properties, such as high stability and electrochemical reactivity, low background current, and wide electrochemical window. Therefore, this electrode is a very good alternative to traditional chemically modified electrodes because the electrocatalytic effect can achieved without any further electrode modification. Comparative experiments were carried out using CPE and a glassy carbon electrode (GCE). With OPFP-CPE, highly reproducible and well-defined cyclic voltammograms were obtained for EGCG. Under optimal experimental conditions, the peak current of differential pulse voltammetry (DPV) response increased linearly with EGCG concentration over the range of 5.0 × 10(-7)-1.25 × 10(-5) M. The limit of detection (LOD) and the limit of quantification (LOQ) were 1.32 × 10(-7) and 4.35 × 10(-7) M, respectively. The method was applied to the determination of EGCG in green tea infusion samples, and the recovery of the spiked EGCG to the diluted (10-fold) tea extract was from 87.62 to 99.51%.  相似文献

As a consequence of the adulteration of infant formulas and milk powders with melamine (MEL) in China in 2008, much attention has been devoted to the analysis of MEL [and cyanuric acid (CA)] in dairy products. Several methods based on high-performance liquid chromatography (HPLC), liquid chromatography-tandem mass spectrometry (LC-MS/MS), nuclear magnetic resonance (NMR), or Raman spectroscopy have been described in the literature. However, no method is available for the simultaneous determination of MEL and CA in other raw materials, which are considered as high-risk materials for economically motivated adulteration. The present paper reports the results of an interlaboratory-based performance evaluation conducted with seven laboratories worldwide. The purpose was to demonstrate the ability of a cleanup-free LC-MS/MS method, originally developed for cow's milk and milk-powdered infant formula, to quantify MEL and CA in egg powder and soy protein. Limit of detection (LOD) and limit of quantification (LOQ) were 0.02 and 0.05 mg/kg for MEL in egg powder and soy protein, respectively. For CA, LOD and LOQ were 0.05 and 0.10 mg/kg in egg powder and 1.0 and 1.50 mg/kg in soy protein, respectively. Recoveries ranged within a 97-113% range for both MEL and CA in egg powder and soy protein. Reproducibility values (RSD(R)) from seven laboratories were within a 5.4-11.7% range for both analytes in the considered matrices. Horwitz ratio (HorRat) values between 0.4 and 0.7 indicate acceptable among-laboratory precision for the method described.  相似文献

蛋壳是鸡胚发育过程中与外界环境进行气体和热量交换的重要媒介,其结构、形态等因素均为影响鸡胚发育的重要因素。单色绿光照射可缩短种蛋孵化时间,促进出雏、提高种蛋孵化率。有鉴于此,该研究针对单色绿光调控孵化过程中的种蛋蛋壳色度、蛋壳结构、色素含量及元素含量等因素进行监测,从种蛋蛋壳特性变化的角度探究单色绿光在种蛋孵化过程中促进雏鸡出雏的机制。结果表明,与传统黑暗孵化环境相比较,孵化过程单色绿光形成的光环境促使蛋壳对孵化环境产生适应性响应,蛋壳表面亮度显著降低(P<0.01),抑制了蛋壳中绿色色素-胆绿素的分解(P<0.01)。且单色绿光有助于鸡胚吸收利用蛋壳中的Ca元素,显著促进孵化过程中蛋壳表面孔隙面积的风化增大(P<0.000 1),提高蛋壳的通透性及散热效果,利于出雏期间雏鸡出壳,有助于促进出雏并降低孵化死亡率。该研究阐析了单色绿光孵化过程中种蛋蛋壳特性的变化,为单色绿光促进种蛋出雏及其在孵化产业的应用奠定了一定的理论基础。  相似文献
