Lima bean (Phaseolus lunatus L.) is an important food source in Brazil, especially in the northeast region, where its production and consumption are high. The goals of the present study were to estimate natural outcrossing rates and genetic diversity levels of Lima bean from Brazil, using ten microsatellite loci to obtain information for their conservation and breeding. Fourteen accessions were selected from an experiment in field with open-pollinated and with the presence of pollinating insects. Twelve seeds of each of the 14 selected accessions were grown in screenhouse for tissue harvest and DNA extraction. The multilocus model was used to determine the reproductive system. The outcrossing rate was 38.1 % (tm = 0.381; ts = 0.078), and the results indicated a mixed mating system with a predominance of selfing (1 ? tm = 61.9 %). The biparental inbreeding rate was high (t m  ? t s  = 0.303) and the multilocus correlated paternity was quite high (r p(m) = 0.889), indicating that the progeny was mostly composed of full sibs. The average effective number of pollen donors per maternal plant (N ep ) was low (1.12), and the fixation index for maternal genotypes (F m ) was 0.945, indicating that most genitors resulted from inbreeding. The studied families presented considerable genetic variability: A = 6.10;  %P = 30; H e  = 0.60 and H o  = 0.077. Total diversity was high (H T = 0.596), and a portion was distributed within families (H S = 0.058). In addition, diversity was higher between families (D ST = 0.538), and genetic differentiation was high (G ST = 0.902). The results presented here can be used in the implementation of Lima bean conservation and breeding programs in Brazil.  相似文献

Microbiological activities are essential in the bioremediation of polluted soils. The enzymatic activities of microorganisms are usually used as a biological indicator of soil health. The aim of this work was to observe the catalase, acid phosphatase (AcP), and alkaline phosphatase (AlP) activities in soil that was amended with agro-industrial by-products and macronutrients during the process of total petroleum hydrocarbon (TPH) removal. To this end, microcosm tests were performed with soil and agro-industrial by-products ratios of 100:2:2, for soil:sugarcane bagasse pith:filter cake mud (SSF); 100:2, for both soil:sugarcane bagasse pith (SS); and for soil filter cake mud (SF). The macronutrients—carbon, nitrogen, and phosphorus—in the experimental treatments were adjusted to 100:10:1 with a solution of NH4NO3 and K2HPO4. The best TPH removal (51.4%) was obtained with SSF at 15 days. In addition, a significant correlation was observed between TPH removal and AlP as well as AcP (r = 0.74, p < 0.0001; r = 0.70, p < 0.0107, respectively). Fungi growth was also correlated with both AlP (r = 0.97, p < 0.0001) and AcP (r = 0.95, p < 0.0001) activities. Besides, bacterial and fungi growth showed a correlation with TPH (r = 0.86, p < 0.001; r = 0.77, p < 0.0034, respectively). It could be said that the agro-industrial by-products and macronutrients contributed to pollutant removal from the oil-polluted soil at relatively short amount of time. In addition, the enzymatic activities were increased after the treatment; in this study, the high sensitivity enzyme was AlP, and it could be used as an indirect indicator of oil pollutant removal.  相似文献

The genus Zanthoxylum, belonging to Rutaceae, has a long history of cultivation both for economic and chemical values in China. To effectively conserve and sustainably utilize this genus resource, a study on genetic diversity and relationships of Zanthoxylum germplasms was carried out by employing SRAP markers. We used 16 primer combinations to assess genetic variations and relationships among 175 accessions from eight cultivated provenances, including Shandong, Henan, Shanxi, Shaanxi, Gansu, Sichuan, Guizhou and Yunnan. A total of 145 clear repetitive and intense bands were yielded, and the percentage of polymorphic bands was 100 % for per primer combination, indicating a relatively high diversity among Zanthoxylum germplasms. From a geographic perspective, the highest genetic diversity level was observed within Guizhou provenance (N a  = 1.97, Ne = 1.52, H = 0.31, I = 0.46) while Henan provenance had the lowest genetic diversity (N a  = 1.68, Ne = 1.45, H = 0.25, I = 0.37). Based on AMOVA results, the abundant genetic variation was mainly caused by variation of intra-provenances (84.96 %), rather than among provenances (15.038 %). The results indicated low genetic differentiation (G st  = 0.133) and high gene flow (N m  = 3.2605) among provenances. The neighbor-joining tree revealed that the 175 accessions could be divided into four groups, and groupings indicated a divergence between the cultivated accessions of Zanthoxylum bungeanum Maxim. and Z. armatum DC. Moreover, three accessions of Z. piperitum DC. var. inerme without prickles introduced from Japan gathered one cluster. Cluster IV is composed of accessions of different geographical origin, including 11 wild species and 10 cultivated accessions of Z. bungeanum. The cluster analysis also reflected a relatively close relationship between the geographical origins and the classification of accessions in cluster I. Structure analysis indicated that collected Zanthoxylum accessions could be divided into two major groups. The information obtained from our research would benefit to make use of Zanthoxylum germplasms and assist the management of a Zanthoxylum germplasms collection.  相似文献

We conducted a microcosm experiment with soil being sterilized, reinoculated with native microbial community and subsequently manipulated the bacterivorous nematodes, including three treatments: without (CK) or with introducing one species of the two bacterivores characterized with different body size but similar c-p (colonizer-persister) value (Rhabditis intermedia and Protorhabditis oxyuroides, accounted for 6 and 59% of bacterivores in initially undisturbed soil, respectively). We monitored the N2O and CO2 emissions, soil properties, and especially quantified gross N transformation rates using 15N tracing technique after the 50 days incubation. No significant differences were observed on soil NH4 + and NO3 ? concentrations between the CK and two bacterivores, but this was not the case for gross N transformation rates. In comparison to CK, R. intermedia did not affect soil N transformation rates, while P. oxyuroides significantly increased the rates of mineralization of organic N to NH4 +, oxidation of NH4 + to NO3 ?, immobilization of NO3 ? to organic N and dissimilatory NO3 ? reduction to NH4 +. Furthermore, the mean residence time of NH4 + and NO3 ? pool was greatly lowered by P. oxyuroides, suggesting it stimulated soil N turnover. Such stimulatory effect was unrelated to the changes in abundance of bacteria and ammonia-oxidizing bacteria (AOB). In contrast to CK, only P. oxyuroides significantly promoted soil N2O and CO2 emissions. Noticeably, bacterivores increased the mineralization of recalcitrant organic N but decreased soil δ13C-TOC and δ15N-TN values, in particular for P. oxyuroides. Combining trait-based approach and isotope-based analysis showed high potential in moving forward to a mechanistic understanding of bacterivore-mediated N cycling.  相似文献

Purpose

There is a paucity of data regarding the multiple timescale variations of heterotrophic respiration (R H) and autotrophic respiration (R A) as well as the primary controlling factors. The objective of this study is to find the temporal variations of total soil respiration (R S) and its components, revealing the driving factors at different timescales.

Materials and methods

A trenching method was used to distinguish R S, R H, and R A in a spruce-fir valley forest in northeastern China. We used the closed dynamic chamber method to measure the soil respiration rate. Analyses of R S, R H, and R A in relation to biotic and abiotic factors were conducted to realize the temporal variations at different timescales.

Results and discussion

Only R S and R H showed a distinct diurnal variation and soil temperature (T S) can explain 68 and 59 % of the daily variation, respectively. R S, R H, and R A showed a pronounced, single peak curve seasonally, and T S can explain 11–95 % of the seasonal variation. Soil moisture (W S) maintained at a relatively high level and was not related to R S, R H, or R A on a seasonal scale, and there was no significant relationship between the seasonal R S, R A, and root biomass. However, for 5 years, only the mean R A of the growing season was significantly related to the mean W S, which can explain 39 % of the inter-annual variation of R A. The annual variations of litterfall and the relative growth rate of stems were not related to R S, R H, or R A. The contribution of R H to R S was larger, and the temperature sensitivity was 2.01–3.71 for R S, 1.90–3.08 for R H, and 2.20–5.65 for R A.

Conclusions

R S, R H, and R A show different temporal variations at multiple timescales. When W S is not restricted, T S is the primary driving factor of daily and seasonal variation of R S and R H. In this site, R H accounts for a large proportion of R S and plays a crucial role in determining the magnitude and temporal variation of R S.

Purpose

Re-establishment of soil nitrogen (N) capital is a priority in mine rehabilitation. We aimed to evaluate the effects of biochar addition on improving mine spoil N pools and the influence of elevated CO2 concentration on mine rehabilitation.

Materials and methods

We assessed the effects of pinewood biochar, produced at three temperatures (650, 750 and 850 °C, referred as B650, B750 and B850, respectively), on mine spoil total N concentrations with five different plant species, including a tree species (Eucalyptus crebra), N-fixing shrubs (Acacia floribunda and Allocasuarina littoralis) and C3 and C4 grasses (Austrodanthonia tenuior and Themeda australis) incubated at ambient (400 μL L?1) and elevated (700 μL L?1) atmospheric CO2 concentrations, as well as the effects of elevated CO2 on mine rehabilitation.

Results and discussion

Soil total N significantly improved following biochar incorporation under all plant species (P < 0.05) except for T. Australis. E. crebra had the highest soil total N (0.197%, 0.198% and 0.212% for B650, B750 and B850, respectively). Different from the negligible influence of elevated CO2 on soil properties under the grasses and the N-fixing shrubs, elevated CO2 significantly increased soil water and hot water extractable organic C (WEOC and HWEOC, respectively) and decreased total C under E. crebra, indicating that the nutrient demands were not met.

Conclusions

Biochar addition showed the potential in mine rehabilitation in terms of improving soil N pool, especially with E. crebra. However, it would be more difficulty to rehabilitate mine spoils in future with the rising atmospheric CO2 concentration.

Purpose

This study aimed to assess the effects of biochar on improving nitrogen (N) pools in mine spoil and examine the effects of elevated CO2 on soil carbon (C) storage.

Materials and methods

The experiment consisted of three plant species (Austrostipa ramossissima, Dichelachne micrantha, and Lomandra longifolia) planted in the N-poor mine spoil with application of biochar produced at three temperatures (650, 750, and 850 °C) under both ambient (400 μL L?1) and elevated (700 μL L?1) CO2. We assessed mine spoil total C and N concentrations and stable C and N isotope compositions (δ13C and δ15N), as well as hot water extractable organic C (HWEOC) and total N (HWETN) concentrations.

Results and discussion

Soil total N significantly increased following biochar application across all species. Elevated CO2 induced soil C loss for A. ramossissima and D. micrantha without biochar application and D. micrantha with the application of biochar produced at 750 °C. In contrast, elevated CO2 exhibited no significant effect on soil total C for A. littoralis, D. micrantha, or L. longifolia under any other biochar treatments.

Conclusions

Biochar application is a promising means to improve N retention and thus, reduce environmentally harmful N fluxes in mine spoil. However, elevated CO2 exhibited no significant effects on increasing soil total C, which indicated that mine spoil has limited potential to store rising atmospheric CO2.

The use of plants for ecological remediation is an important method of controlling heavy metals in polluted land. Cotinus coggygria is a landscape plant that is used extensively in landscaping and afforestation. In this study, the cadmium tolerance level of C. coggygria was evaluated using electrical impedance spectroscopy (EIS) to lay a theoretical foundation for broad applications of this species in Cd-polluted areas and provide theoretical support to broaden the application range of the EIS technique. Two-year-old potted seedlings of C. coggygria were placed in a greenhouse to analyse the changes in the growth, water content and EIS parameters of the roots following treatment with different Cd concentrations (50, 100, 200, 500, 1000 and 1500 mg kg?1), and soil without added Cd was used as the control. The roots grew well following Cd treatments of 50 and 100 mg kg?1. The Cd contents increased with the increase in Cd concentration in the soil. However, the lowest root Cd content was found at 4 months of treatment. The extracellular resistance re and the intracellular resistance ri increased first overall and then decreased with the increasing Cd concentration, and both parameters increased with a longer treatment duration. The water content had a significant negative correlation with the Cd content (P?<?0.01) and the re (P?<?0.05). C. coggygria could tolerate a soil Cd concentration of 100 mg kg?1. There was a turning point in the growth, water content and EIS parameters of the C. coggygria roots when the soil Cd concentration reached 200 mg kg?1. The root water content and re could reflect the level of Cd tolerance in C. coggygria.  相似文献

This study evaluated the effect of silicate fertilizer on denitrification and associated gene abundance in a paddy soil. A consecutive trial from 2013 to 2015 was conducted including the following treatments: control (CK), mineral fertilizer (NPK), NPK plus sodium metasilicate (NPK + MSF), and NPK plus slag-based silicate fertilizer (NPK + SSF). Real-time quantitative PCR (qPCR) was used to analyze the abundances of nirS, nirK, and nosZ genes. Potential N2O emissions and ammonium and nitrate concentrations were related to the nirS and nirK gene abundance. Compared with the NPK treatments, the addition of a Si fertilizer decreased N2O emission rates and denitrification potential by 32.4–66.6 and 22.0–59.2%, respectively, which were probably related to increased rice productivity, soil Fe availability, and soil N depletion. The abundances of nirS and nirK genes were decreased by 17.7–35.8% and 21.1–43.5% with addition of silicate fertilizers, respectively. Rates of total N2O and N2O from denitrification (DeN2O) emission were positively correlated with the nirS and nirK gene abundance. Nitrate, exchangeable NH4 +, and Fe concentrations were the main factors regulating the nirS and nirK gene abundance. Silicate fertilization during rice growth may serve as an effective approach to decreasing N2O emissions.  相似文献

The impact of nanoparticles (NPs) in zooplankton is poorly studied, particularly when organisms are exposed through diet. Food, constituted mainly by unicellular algae, can act as an important route of contamination for zooplankton. Since unicellular algae have a high surface area in relation to their volume, NPs can interact with their cell membranes and walls, as well as with exopolysaccharides secreted by them. In the present research, we investigated both the acute effects of waterborne titanium dioxide nanoparticles (TiO2 NPs), and its chronic effects via dietary exposure on the Neotropical freshwater zooplankton Ceriodaphnia silvestrii Daday, 1902 (Crustacea: Cladocera). The observed acute effects served as support for chronic tests, in which we investigated the effects of TiO2 NPs on survival and life history parameters (body length, numbers of eggs, and neonates produced) of cladoceran adult females, using the freshwater cosmopolitan chlorophycean Raphidocelis subcapitata as source of contamination of TiO2 NPs for zooplankton. R. subcapitata cells were exposed to concentrations of 0, 0.01, 1, and 10 mg L?1 of TiO2 NPs for 96 h, and then provided as food for females of C. silvestrii until the third brood was released. Significant toxic effects were observed in body length and total number of neonates and eggs produced by females of C. silvestrii at concentrations of 1 and 10 mg L?1 of TiO2 NPs. Survival was the most sensitive parameter when exposure was given via food. From the concentration of 0.01 mg L?1 of TiO2 NPs, there was a decrease in the survival of C. silvestrii females. The quantification of TiO2 NPs in algae evidenced that they have retained NPs in their cells, being, therefore, an important route of exposure and toxicity of TiO2 NPs to the studied microcrustacean.  相似文献

Purpose

Evaluate the efficiency of Populus alba clone Villafranca in the uptake and translocation of Zn from contaminated soils.

Materials and methods

The effects of 48 days of zinc treatment (Zn t ) on the growth and the photosynthetic activities of P. alba L. clone Villafranca were studied using ZnSO4 (375 ppm per unit of soil dry weight) added in sand and peat moss substrate at the beginning of the treatment (T 0) and again after 30 days (T 1) in order to reach a target Zn concentration of 375 ppm at T 0 and 750 ppm at T 1 per unit of soil dry weight.

Results and discussion

Zn uptake in the different organs was analyzed after 30 (T 1) and 48 days (T 2) from the beginning of treatment, showing the following order: root ? leaves ≥ woody cutting = stem. The leaf area increased by 12 % in comparison to control plants at the end of second treatment (48 days). Cutting radial growth showed a high synchronicity in the growth rate fluctuation among control and Zn t plants, but a higher increase in radial diameter of Zn t cutting was observed starting from day 38 (after 8 days of second Zn t ) reaching after 48 days 38 % higher than control plants.

Conclusions

Although our data of leaf Zn concentration were in the range usually reported as toxic for plants, Villafranca clone in Zn t substrate were unaffected in terms of net CO2 assimilation and stomatal conductance to water vapor.

Purpose

The USLE-MM estimates event normalized plot soil loss, Ae,N, by an erosivity term given by the runoff coefficient, QR, times the single-storm erosion index, EI30, raised to an exponent b1?>?1. This modeling scheme is based on an expected power relationship, with an exponent greater than one, between event sediment concentration, Ce, and the EI30/Pe (Pe = rainfall depth) term. In this investigation, carried out at the three experimental sites of Bagnara, Masse, and Sparacia, in Italy; the soundness of the USLE-MM scheme was tested.

Materials and methods

A total of 1192 (Ae,N, QREI30) data pairs were used to parameterize the model both locally and considering all sites simultaneously. The performances of the fitted models were established by considering all erosive events and also by distinguishing between events of different severity.

Results and discussion

The b1 exponent varied widely among the three sites (1.05–1.44) but using a common exponent (1.18) for these sites was possible. The Ae,N prediction accuracy increased in the passage from the smallest erosion events (Ae,N?≤?1 Mg ha?1, median error =?3.35) to the largest ones (Ae,N?>?10 Mg ha?1, median error =?1.72). The QREI30 term was found to be usable to predict both Ae,N and the expected maximum uncertainty of this prediction. Soil erodibility was found to be mainly controlled by the largest erosion events.

Conclusions

Development of a single USLE-MM model appears possible. Sampling other sites is advisable to develop a single USLE-MM model for a general use.

Purpose

Soil acidification is universal in soybean-growing fields. The aim of our research was to evaluate the effects of soil additives (N fertilizers and biochar) on crop performance and soil quality with specific emphasis on ameliorating soil acidity.

Materials and methods

Four nitrogen treatments were applied as follows: no nitrogen (N0), urea (N1), potassium nitrate (N2), and ammonium sulfate (N3), each providing 30 kg N ha?1. Half plot area of the N1, N2, and N3 treatments was also treated with biochar (19.5 t ha?1) to form N-biochar treatments (N1C, N2C, N3C). Both bulk and rhizosphere soils were sampled separately for the following analyses: pH, exchangeable base cations (EBC), exchangeable acidity (EA), total inorganic N (IN), total N (TN), and microbial phospholipid fatty acids (PLFAs). Soybean biomass and nutrient contents were also determined. Correlation analysis was applied to analyze the relationships between soil chemical properties and soybean plant parameters.

Results and discussion

With N-biochar additions (N1C, N2C, N3C), soil chemical properties changed as follows: pH increased by 0.6–1.2 units, EBC, IN, and TN increased by 175–419, 38.5–54.7, and 136–452 mg kg?1, respectively, and PLFAs increased by 23.6–40.9 nmol g?1 compared to the N0 in the rhizosphere. Microbial PLFAs had positive correlations with soil pH; EBC; exchangeable K, Ca, Na, and Mg; TN; IN; NH4 +; and NO3 ? (r?=?0.66–0.84, p?<?0.01). There were negative correlations between PLFAs and EA or exchangeable Al (r?=??0.64, ?0.66, p?<?0.01), which indicated that the additives increased microbial biomass by providing a suitable environment with less acid stress and more nutrients. The additives increased soil NH4 + and NO3 ? by promoting soil organic N mineralization and reducing NH4 + and NO3 ? leaching. Moreover, the soybean seed biomass and the nutrient contents in seeds increased with N-biochar additions, especially in the N3C treatment.

Conclusions

N-biochar additions were effective in ameliorating soil acidity, which improved the microenvironment for more microbial survival. N-biochars influenced N transformations at the plant–soil interface by increasing organic N mineralization, reducing N leaching, and promoting N uptake by soybeans. The soil additive ammonium and biochar (N3C) were best in promoting soybean growth.

Vitellaria paradoxa C. F. Gaertn., commonly known as shea tree or Vitellaria, is ranked the most important tree species of the savannah regions in the most African countries due to its ecological and economic importance for livelihoods and national economies. However, the savannah regions are the most vulnerable areas to the global climate change. Moreover, the Vitellaria populations on farmlands are threatened by the dominance of old trees with low or lack of regeneration. In this study both morphological and genetic diversity were assessed using several phenotypic traits and 10 microsatellite markers, respectively, to assess the impact of land use and agro-ecozone types on Vitellaria in Ghana. The land use types were forests and farmlands, and the agro-ecozone types included the Transitional, Guinea, and Sudan savannah zones. The mean values of morphological traits, such as diameter at breast height (DBH) and canopy diameter (CD), were statistically different between forest (DBH = 22.20, CD = 5.37) and farmland (DBH = 39.85 CD = 7.49) populations (P < 0.00001). The Sudan savannah zone with mean petiole length of 4.96 cm showed significant difference from the other zones, likely as a result of adaptation to drier climate conditions. Genetic data analysis was based on 10 microsatellite markers and revealed high genetic diversity of Vitellaria in Ghana: mean expected heterozygosity, H e was 0.667, and allelic richness, measured as number of effective alleles A e , was 4.066. Both farmlands and forests were very diverse indicating lack of negative influence of farmer’s selection on genetic diversity. Fixation index was positive for all populations (mean F IS = 0.136) with farmlands recording relatively higher values than forests in all ecological zone types studied, probably indicating less gene flow in the farmlands. Moderate differentiation (F′ ST = 0.113) was comparable to other similar tree species. Both land use and ecological zone types influenced genetic differentiation of Vitellaria at varying levels. The species was spatially structured across three ecozones and following climatic gradient. The forest reserves are used in situ conservation for Vitellaria in Ghana. High diversity observed in the most arid zones provides opportunity to find and use appropriate plant materials for breeding climate change resilient trees.  相似文献

Purpose

Input of N as NH4 + is known to stimulate nitrification and to enhance the risk of N losses through NO3 ? leaching in humid subtropical soils. However, the mechanisms responsible for this stimulation effect have not been fully addressed.

Materials and methods

In this study, an acid subtropical forest soil amended with urea at rates of 0, 20, 50, 100 mg N kg?1 was pre-incubated at 25 °C and 60 % water-holding capacity (WHC) for 60 days. Gross N transformation rates were then measured using a 15N tracing methodology.

Results and discussion

Gross rates of mineralization and nitrification of NH4 +-N increased (P?<?0.05), while gross rate of NO3 ? immobilization significantly decreased with increasing N input rates (P?<?0.001). A significant relationship was established between the gross nitrification rate of NH4 + and the gross mineralization rate (R 2?=?0.991, P?<?0.01), so was between net nitrification rate of NH4 + and the net mineralization rate (R 2?=?0.973, P?<?0.05).

Conclusions

Stimulation effect of N input on the gross rate of nitrification of NH4 +-N in the acid soil, partially, resulted from stimulation effect of N input on organic N mineralization, which provides pH-favorable microsites for the nitrification of NH4 + in acid soils (De Boer et al., Soil Biol Biochem 20:845–850, 1988; Prosser, Advan Microb Physiol 30:125–181, 1989). The stimulated gross nitrification rate with the decreased gross NO3 ? immobilization rate under the elevated N inputs could lead to accumulation of NO3 ? and to enhance the risk of NO3 ? loss from humid forest soils.

New methodological approaches and an algorithm for the quantitative assessment of accumulating a substance by a plant (C p ) with due regard for its concentration in the soil (C n ) are proposed. The first approach is the approximation of concentration curves by the functions C p = f(C n ) and their parameters. The second one is the standardization of the coefficients of biological sorption (K b ) of a substance upon its stable concentration in the soil using the function K b = f(C n ). As compared to the variation-statistical values of the mean and extreme concentrations, or the coefficients K b , the proposed parameters are characterized by a higher accuracy and sensitivity. They may be successfully used for quantitative studies of the mechanism and the intensity of the absorption of substances by plants, for the prediction of the accumulation of substances in the trophic chain, and the assessment of soil self-purification (detoxification) and comparative ratings.  相似文献

C-banded pattern in two accessions of Thinopyrum bessarabicum (Save ex Rayss) A. Löve (2n = 2x = 14, EbEb) and their idiogram was established. C-banding analysis was further used to identify the chromosomes of Tritipyrum amphiploid (2n = 6x = 42, AABBEbEb) and a BC1F2 genotype from wheat and Tritipyrum. Two 18S-26S rDNA loci were detected on Th. bessarabicum chromosomes by in situ hybridization using an 18S-26S rDNA probe. Eb chromosomes in Tritipyrum generally were identified by their distinctive C-banding patterns which reflected heterochromatin regions. C-banding procedure resulted in sharp and distinct bands in one or both ends of Eb chromosomes without interval bands. Observed C-bands in Eb genome mainly reflected the telomeric and subtelomeric sequences which also showed more strong signals in genomic in situ hybridization. Results showed the importance of the C-banding technique as a screening tool in identification of addition and substitution lines in the progenies of wheat and Tritipyrum crosses during segregating generations.  相似文献

Filamentous mat-forming algae are increasingly impairing freshwater resources. To restore water utility, reactive management programs often involve application of copper-based algaecides. Copper algaecide formulations can differ significantly, and this research outlined an advanced approach to evaluate formulation efficiency for controlling filamentous algae. Two common algal species (Lyngbya wollei, Pithophora varia) were used to assess copper internalization and adsorption as well as relation to control among copper formulations. Captain® XTR achieved control (7-day EC85) of L. wollei with internal copper concentrations of 0.78 and 0.76 mg Cu/g based on chlorophyll a content or filament viability, respectively. Cutrine® Ultra achieved control of L. wollei based on filament viability only at 0.85 mg Cu/g. Internalized copper concentrations required for control following Captain XTR exposures were similar for P. varia, 0.81 and 0.95 mg Cu/g, whereas Cutrine Ultra and copper sulfate did not elicit control nor attain the critical internal copper threshold. The relationship between internalized copper and responses, among all formulations, was significant (P?<?0.0001) with R2 values of 0.920 and 0.935 for L. wollei and 0.807 and 0.826 for P. varia based on filament viability and chlorophyll a content, respectively. Formulation efficiency, internalized copper versus total amended, was greatest with Captain XTR (average 0.17), followed by Cutrine Ultra (0.13), and copper sulfate (0.09). By measuring the efficiency of a specific algaecide and the corresponding amount required to achieve control of targeted algal biomass, management objectives can be achieved while decreasing environmental loads of copper, number of treatments, and operational costs.  相似文献

Purpose

This work investigated changes in priming effects and the taxonomy of soil microbial communities after being amended with plant feedstock and its corresponding biochar.

Materials and methods

A soil incubation was conducted for 180 days to monitor the mineralization and evolution of soil-primed C after addition of maize and its biochar pyrolysed at 450 °C. Responses of individual microbial taxa were identified and compared using the next-generation sequencing method.

Results and discussion

Cumulative CO2 showed similar trends but different magnitudes in soil supplied with feedstock and its biochar. Feedstock addition resulted in a positive priming effect of 1999 mg C kg?1 soil (+253.7 %) while biochar gave negative primed C of ?872.1 mg C kg?1 soil (?254.3 %). Linear relationships between mineralized material and mineralized soil C were detected. Most priming occurred in the first 15 days, indicating co-metabolism. Differences in priming may be explained by differences in properties of plant material, especially the water-extractable organic C. Predominant phyla were affiliated to Acidobacteria, Actinobacteria, Chloroflexi, Gemmatimonadetes, Firmicutes, Planctomycetes, Proteobacteria, Verrucomicrobia, Ascomycota, Basidiomycota, Blastocladiomycota, Chytridiomycota, Zygomycota, Euryarchaeota, and Thaumarchaeota during decomposition. Cluster analysis resulted in separate phylogenetic grouping of feedstock and biochar. Bacteria (Acidobacteria, Firmicutes, Gemmatimonadetes, Planctomycetes), fungi (Ascomycota), and archaea (Euryarchaeota) were closely correlated to primed soil C (R 2?=??0.98, ?0.99, 0.84, 0.81, 0.91, and 0.91, respectively).

Conclusions

Quality of plant materials (especially labile C) shifted microbial community (specific microbial taxa) responses, resulting in a distinctive priming intensity, giving a better understanding of the functional role of soil microbial community as an important driver of priming effect.

The nitrous oxide and molecular N emissions from 5-cm length subsamples taken from 20-cm length sample corers containing eutric Cambisol soil fertilised either with urea, ammonium or nitrate for 1 year have been examined using gas chromatography. At the beginning of the incubation, the same N rate (260 kg N/ha) was added to the soil and kept constant during the experiment. The total abundance of the soil Bacteria and Archaea and that of nitrifiers and denitrifiers was estimated by quantitative PCR of the corresponding biotic variables 16S rRNA, amoA and napA, narG, nirK, nirS, norB, nosZI and nosZII genes. The abiotic variables dissolved oxygen, pH, exchangeable NH4+-N and NO3?-N contents and total C and total N were also analysed. None of the three fertilisers affected the total abundance of Bacteria and Archaea and nitrification was the main driver of nitrous oxide production in the 0- to 5-cm and 5- to 10-cm soil layers while denitrification was in the 10- to 15-cm and 15- to 20-cm soil horizons. Parallel to the reduction in the content of dissolved oxygen along the soil profile, there was a decrease in the total and relative abundance of the bacterial and archaeal amoA gene and an increase in the abundances of the denitrification genes, mainly in the 10- to 15-cm and 15- to 20-cm soil layers. A non-metric multidimensional scaling plot comparing the biotic and abiotic variables examined in each of the four 5-cm soil subsamples and the whole 20-cm sample showed a disparate effect of N fertilisation on N gas emissions and abundance of nitrifiers and denitrifiers bacterial and archaeal communities.  相似文献
