The genetic basis of several different components of resistance to Rhynchosporium secalis in barley was investigated in a mapping population derived from a cross between winter and spring barley types. Both the severity of visual disease symptoms and amount of R. secalis DNA in leaf tissues were assessed in field trials in Scotland in the 2007/2008 and 2008/2009 growing seasons. Relative expression of symptoms was defined as the residual values from a linear regression of amount of R. secalis DNA against visual plot disease score at GS 50. Amount of R. secalis DNA and visual disease score were highly correlated traits and identified nearly identical QTL. The genetic control of relative expression of symptoms was less clear. However, a QTL on chromosome 7H was identified as having a significant effect on the expression of visual disease symptoms relative to overall amount of R. secalis colonisation.  相似文献

Three quantitative trait loci (QTL) for scald resistance in barley were identified and mapped in relation to molecular markers using a population of chromosome doubled‐haploid lines produced from the F1 generation of a cross between the spring barley varieties ‘Alexis’ and ‘Regatta’. Two field experiments were conducted in Denmark and two in Norway to assess disease resistance. The percentage leaf area covered with scald (Rhynchosporium secalis) ranged from 0 to 40% in the 189 doubled‐haploid (DH) lines analysed. One quantitative trait locus was localized in the centromeric region of chromosome 3H, Qryn3, using the MAPQTL program. MAPQTL was unable to provide proper localization of the other two resistance genes and so a non‐interval QTL mapping method was used. One was found to be located distally to markers on chromosome 4H (Qryn4) and the other, Qryn6, was located distally to markers on chromosome 6H. The effects of differences between the Qryn3, Qryn4 and Qryn6 alleles in two barley genotypes for the QTL were estimated to be 8.8%, 7.3% and 7.0%, respectively, of leaf covered by scald. No interactions between the QTLs were found.  相似文献

Beet yellows virus (BYV) can dramatically reduce yield from sugar beet crops. A BYV resistant plant from a Beta germplasm accession was crossed with a sugar beet plant to generate the segregating population BYV1. This population was evaluated for BYV resistance and analysed with amplified fragment length polymorphism and single-nucleotide polymorphism markers to identify resistance loci. At 2 weeks after inoculation a number of plants displayed either mosaic or vein-clearing disease symptoms on infected leaves. The locus that largely controlled this disease reaction was mapped to chromosome IV. We refer to this novel genetic locus as Vc1 . Three BYV resistance quantitative trait loci (QTLs) were identified and mapped to chromosomes III, V and VI. QTL mapping results suggested that the chromosome III and V QTLs acted only in plants with mosaic symptoms and that the chromosome VI QTL acted only in plants with the mosaic symptom allele of Vc1 . These findings agreed with further statistical tests using general linear model analysis. There is now the potential to breed for BYV resistance using molecular marker technology.  相似文献

Scald is a serious foliar disease that infects barley (Hordeum vulgare L.) causing reduced yields and adversely affecting quality. A means to combat the disease is to breed cultivars that possess genetic resistance. However, all known resistance alleles have so far originated from within the primary genepool of barley. This reliance on H. vulgare and H. vulgare ssp. spontaneum as resistance sources may encourage virulent forms of the pathogen to become established. To broaden the genetic base of cultivated barley and provide novel resistances to many diseases we have used a species from the secondary genepool of barley, H. bulbosum, in a resistance‐breeding programme. In this study we describe the development and trialling of a scald‐resistant recombinant line derived from a hybrid between H. vulgare and H. bulbosum. The scald resistance is simply inherited and located on the short arm of barley chromosome 4 (4HS).  相似文献

Spot blotch caused by Bipolaris sorokiniana is an important disease in barley worldwide, causing considerable yield losses and reduced grain quality. In order to identify QTL conferring resistance to spot blotch, a highly diverse worldwide barley set comprising 449 accessions was phenotyped for seedling resistance with three isolates (No 31, SH 15 and SB 61) and for adult plant resistance at two locations (Russia and Australia) in two years. Genotyping with the 50 k iSelect barley SNP genotyping chip yielded 33,818 informative markers. Genome-wide association studies (GWAS) using a compressed mixed linear model, including population structure and kinship, revealed 38 significant marker-trait associations (MTA) for spot blotch resistance. The MTA corresponded to two major QTL on chromosomes 1H and 7H and a putative new minor QTL on chromosome 7H explaining between 2.79% and 13.67% of the phenotypic variance. A total of 10 and 14 high-confidence genes were identified in the respective major QTL regions, seven of which have a predicted involvement in pathogen recognition or defence.  相似文献

Changes in alleles frequencies of marker loci linked to yield quantitative trait loci (QTL) were studied in 188 barley entries (landraces, old and modern cultivars) grown in six trials representing low and high yielding conditions in Spain (2004) and Syria (2004, 2005). A genome wise association analysis was performed per trial, using 811 DArT® markers of known map position. At the first stage of analysis, spatially adjusted genotypic means were created per trial by fitting mixed models. At the second stage, single QTL models were fitted with correction for population substructure, using regression models. Finally, multiple QTL models were constructed by backward selection from a regression model containing all significant markers from the single QTL analyses. In addition to the association analyses per trial, genotype by environment interaction was investigated across the six trials. Landraces seemed best adapted to low yielding environments, while old and modern entries adapted better to high yielding environments. The number of QTL and the magnitude of their effects were comparable for low and high input conditions. However, none of the QTL were found within a given bin at any chromosome in more than two of the six trials. Changes in allele frequencies of marker loci close to QTL for grain yield in landraces, old and modern barley cultivars could be attributed to selection exercised in breeding, suggesting that modern breeding may have increased frequencies of marker alleles close to QTL that favour production particularly under high yield potential environments. Moreover, these results also indicate that there may be scope for improving yield under low input systems, as breeding so far has hardly changed allele frequencies at marker loci close to QTL for low yielding conditions.  相似文献

A set of test crosses of diploid potatoes was used to identify QTLs for foliage resistance against Phytophthora infestans and QTLs for foliage maturity type, and to assess their genetic relationship. The most important locus for both traits was found on chromosome 5 near marker GP21: the allele of marker GP21 that is associated with resistance to late blight is also associated with late foliage maturity. An additional QTL with a small effect on foliage maturity type was identified on chromosome 3, and additional QTLs for late blight resistance were found on chromosomes 3 and 10. Another QTL was detected on chromosome 7 when resistance was adjusted for the effect of foliage maturity type. The additional QTLs for resistance against P. infestans on chromosomes 3 and 10 seem to be independent of foliage maturity type and are not affected by epistatic effects of the locus on chromosome 5. The effects of the additional QTLs for resistance are small, but early maturing genotypes that necessarily have the allele for susceptibility for late blight on chromosome 5 may benefit from the resistance that is provided by these QTLs on chromosomes 3 and 10.  相似文献

Net blotch, caused by Pyrenophora teres f. teres, is a damaging foliar disease of barley worldwide. It is important to identify resistance germplasm and study their genetics. 'Chevron', a six-rowed barley used as a parent for the production of a doubled haploid (DH) population for mapping of Fusarium head blight (FHB) resistance, was also found to be resistant to net blotch. To map the resistance genes, the population was evaluated for resistance at the seedling stage in a greenhouse. The resistance data showed a two-peak distribution. Through linkage mapping, one resistance gene, tentatively called Rpt, was located on chromosome 6HS flanked by Xksua3b-Xwg719d, which was also detected by QTL mapping. This QTL explained 64% of the phenotypic variance for the resistance in this DH population. In addition, a minor QTL was found on chromosome 2HS defined by Xcdo786-Xabc156a. 'Chevron' and 'Stander' contributed the resistant alleles of Rpt and the 2HS QTL, respectively. Both QTLs together explained nearly 70% of the phenotypic variance. The markers for these QTLs are useful for marker-assisted selection of net blotch resistance in barley breeding.  相似文献

Pyrenophora graminea is a seed-borne pathogen and is the causal agent of the barley leaf stripe disease. Our aim is to study the genetic basis of barley resistance to leaf stripe. A qualitatively acting resistance factor has been identified in the cultivar ‘Vada’ and the partial resistance of the cultivar ‘Proctor’ to a P. graminea isolate has been demonstrated to be dominated by a major quantitative trait locus (QTL), mapped on barley chromosome 1. Map colinearity between the leaf stripe ‘Proctor’ resistance QTLs,‘Vada’ resistance to leaf stripe, and other disease resistance loci have been investigated in this work using molecular markers. Moreover, since inoculation of barley rootlets by the fungus had been shown to induce the accumulation of several PR (pathogen-related) mRNA families, seven barley PR genes have been mapped as RFLPs, and one assigned to a chromosome arm via ditelosomic analysis to verify possible map associations with resistance QTLs. This work discusses the genetic relationships between the known leaf stripe resistance loci, resistance loci towards other seed-borne pathogens and defence gene loci.  相似文献

Nitrogen (N) is one of the most important plant nutrients, controlling growth and, ultimately, yield of a cultivar. Hordeum vulgare ssp. spontaneum, the wild barley progenitor of cultivated barley, is known to possess genes that can improve tolerance against biotic and abiotic stresses. A quantitative trait locus (QTL) study with two levels of N fertilization was conducted under glasshouse in order to locate wild barley alleles that improve N stress tolerance in the genetic background of an elite barley cultivar. For this, a set of 28 barley introgression lines (S42ILs), which originate from the cross ??Scarlett???×???ISR42-8??, was studied. The S42ILs, containing single or multiple wild barley introgressions, and ??Scarlett?? were evaluated in regard to a total of 15 traits, related to morphological parameters, grain parameters as well as to carbon (C) and N content parameters. A mixed model analysis and a subsequent Dunnett test was conducted to identify S42ILs that significantly deviate from the recurrent parent ??Scarlett??, either tested separately for each N level, or simultaneously across both N levels. In total, 65 QTLs were detected for the S42IL set. Most QTLs were found for chlorophyll content during heading (10 QTLs) and the fewest for C/N ratio of straw (1 QTL). The individual S42ILs possessed different numbers of QTLs. For S42IL-108, a maximum of eight QTLs were found whereas S42IL-145 did not show any significant difference from ??Scarlett??. Wild barley alleles revealed decreasing effects at 32 QTLs and increasing effects at 33 QTLs. Although 25 QTLs exhibited similar effects across both N levels, 18 and 22 QTLs exhibited effects that were only detected under N0 or N1, respectively. We, thus, conclude that it may be worth to select improved barley cultivars for N stress tolerance separately under low N fertilization, rather than extrapolating trait performances from experiments carried out under standard N fertilization conditions. A number of wild barley QTL alleles improved N stress tolerance. For example, a wild barley QTL allele on chromosome 4H, present in the Hsp introgression of S42IL-119, was associated with a 13.0?% increase of thousand grain weight across both N levels and a 20?% increase under low N supply. QTLs detected in the present study were compared with those of previous field studies of the same cross and with other QTL studies in barley and other small grains. Accordance between QTL studies (QTLs showing similar effects at the same map location) is documented and discussed. Based on our study, promising wild barley QTL alleles are available in S42ILs, which can be readily utilized to select for improved N use efficiency in barley breeding.  相似文献

Phytohormones, a group of structurally unrelated small molecules are plant-signalling compounds that trigger induced resistance against certain pathogens and herbivores. The hormones jasmonic acid (JA), ABA, salicilic acid (SA) and ethylene (ET) are known to play major roles in regulating plant defence responses. In order to determine the changes in growth and in the chlorophyll content induced by the exogenous application of these elicitors, a set of DH lines of the Oregon-Wolfe Barley mapping population, previously screened to locate aphid resistant genes, was investigated. The aim of the current research was to map the induced defence genes and to reveal the relationship with aphid resistance. There were highly significant differences between controls and hormone treated plants in the aerial fresh and dry weights (AFW, ADW), the foliar area (FA) and the root dry weight (RDW). More than 15 JA and ET-induced lines exceeded the chlorophyll (Ch) values of their controls. Most of the plant traits were associated with the same genetic windows on chromosomes 3H, 5H and 7H in the controls and hormone treated plants. QTL(s) identified on chromosome 3H and 5H explained most of the variation of AFW, ADW, FA and RDW of controls and treated plants. QTL(s) located on chromosome 5H were associated with the variation of chlorophyll contents on JA-treated plants. The Ch in ET and ABA-treated plants was associated with two different regions on chromosome 7H. One of the latter genetic windows also explained the variation of RDW of ET- and ABA-treated plants. A sequence homology search was performed to derive the putative function of the genes linked to the QTLs. Several QTLs were identified located close to aphid resistance genes previously mapped. This is the first report of genes associated with hormone response in barley that could be involved with insect resistance. Those recombinant lines carrying the appropriate alleles could be useful for breeding barley to enlarge the genetic base of defence against stress.  相似文献

Based on the RAPD marker OP‐C04H910 which is closely linked to the barley mild mosaic virus (BaMMV) resistance gene rym9 derived from the variety ‘Bulgarian 347’ the marker STS‐C04H910 cosegregating with OP‐C04H910 and generating a single additional band on plants carrying the recessive resistance encoding allele has been developed. Furthermore, the simple sequence repeats (SSRs) WMS6 and HVM67 have been integrated into the genetic map of the rym9 region on chromosome 4HL. Because of their close linkage to rym9 and distinct banding pattern STS‐C04H910 and HVM67 are well‐suited for marker‐ assisted selection, enhanced backcrossing procedures and pyramiding of resistance genes.  相似文献

Cereal cyst nematodes ( Heterodera avenae Woll.) are economically damaging barley parasites in most cereal growing areas of the world and the development of resistant cultivars is the best measure against the pathogen. An ISSR (inter-simple sequence repeat) marker identified as closely linked with the H. avenae race 1 and 2 resistance gene ( Ha2 ) has been converted into a codominant sequence characterized amplified region marker ( Ha2S18 ) and mapped in barley 'SW Buddy' × 'SW Cecilia' DH population at 4.3 cM from the Ha2 locus on the long arm of chromosome 2H. The potential usefulness of Ha2S18 in large scale marker assisted selection schemes has been evaluated in a broad genetic background and is an important complement to the bioassay and to other linked DNA-markers for this trait.  相似文献

以96对SSR引物对BC3F2群体进行辅助筛选,构建了一组基本覆盖野生大麦ISR42-8染色体,并导入轮回亲本Scarlett的1H~7H全基因组的外源基因渗入系,为大麦QTL精细定位提供了良好的作图群体。这组渗入系一共66个,目标片段平均长度为27.6 cM,最长的系IL-52片段长度为100.5 cM,最短的系IL-50片段长度为1.5 cM。含单个渗入片段的有33个系。聚类分析结果表明,66个渗入系的遗传背景高度相似,遗传相似系数变幅为0.708~1.000,平均值为0.917。检测到一个位于4H染色体87.5 cM到110.0 cM区间的分蘖相关QTL,长度为22.5 cM,其中包含多态标记MGB396。  相似文献

A doubled haploid barley (Hordeum vulgare L.) population from a cross between the cultivar `Ingrid' and the Ethiopian landrace `Abyssinian' was mapped by AFLP, RFLP, SSR and STS markers and tested for resistance to isolates`4004', `2', `16-6', `17', `22' and `WRS 1872' of Rhynchosporium secalis (Oudem.) J.J. Davis, the causal agent of leaf scald. Resistance tests were conducted on parents, DH-lines, a near-isogenic line of `Abyssinian' (NIL) into `Ingrid', and an F2 population descended from the same F1 plants as the DHs. The DH population segregated for at least two major R. secalis resistance QTL. All isolates tested identified a major QTL on chromosome 3 (3H) associated with R. secalis resistance, in a 4 cM support interval between the co-segregating markers Bmac0209/Falc666 and MWG680. The QTL was linked with the markers Falc666 (2.3 cM), YLM/ylp (0.3 cM), MWG680 (1.7 cM), cttaca2 (2.5 cM) and agtc17 (9.8 cM). The second QTL was located on chromosome 1 (7H).However, this QTL was only detected by one isolate and was located in an interval of 16 cM in the distal part of the chromosome. At this QTL the allele for improved scald resistance originated from the parent `Ingrid'. There were a number of minor QTL on chromosomes 2 (2H), 4 (4H) and 6 (6H) that were not repeatable either across replications or analysis methods. Multi-environment trials represent a highly valuable tool for the identification of the genetic bases of crop yield potential and stress adaptation. A Diversity Array Technology®-based barley map has been developed in the ‘Nure’ × ‘Tremois’ biparental Doubled Haploid population, harbouring the genomic position of a gene set with a putative role in the regulation of flowering time and abiotic stress response in barley. The population has been evaluated in eighteen location-by-year combinations across the Mediterranean basin. QTL mapping identified several genomic regions responsible for barley adaptation to Mediterranean conditions in terms of phenology, grain yield and yield component traits. The most frequently detected yield QTL had the early flowering HvCEN_EPS2 locus (chromosome 2H) as peak marker, showing a positive effect from the early winter parent ‘Nure’ in eight field trials, and explaining up to 45.8 % of the observed variance for grain yield. The HvBM5A_VRN-H1 locus on chromosome 5H and the genomic region possibly corresponding to PPD-H2 on chromosome 1H were significantly associated to grain yield in five and three locations, respectively. Environment-specific QTLs for grain yield, and clusters of yield component QTLs not related to phenology and or developmental genes (e.g. on chromosome 4H, BIN_09) were observed as well. The results of this work provide a valuable source of knowledge and tools for both explaining the genetic bases of barley yield adaptation across the Mediterranean basin, and using QTL-associated markers for MAS pre-breeding and breeding programmes.  相似文献

Quantitative trait loci (QTLs) for resistance against non-parasitic leaf spots (NPLS) were first characterized in a spring barley double haploid population derived from the cross IPZ 24727/Barke (Behn et al., 2004). The aim of the present study was to identify QTLs for NPLS resistance in the half-sibling DH population IPZ 24727/Krona and to compare them with the QTLs of the population IPZ 24727/Barke. An anther culture-derived doubled haploid population of 536 DH lines was developed from the cross IPZ 24727 (resistant)/Krona (susceptible). Field trials were performed over two years in two replications, scoring NPLS and agronomic traits that might interact with NPLS. A molecular linkage map of 1035 cM was constructed based on AFLPs, SSRs and the mlo marker. QTL analyses for NPLS identified three QTLs that accounted for 30% of the phenotypic variation. For comparison of the QTLs from each DH population, a consensus map was generated comprising 277 markers with a length of 1199 cM. In both populations, the QTLs for NPLS mapped to chromosomes 1H, 4H and 7H. A common QTL with a great effect in both populations and over all environments was localized at the mlo locus on chromosome 4H, indicating that the mlo powdery mildew resistance locus has a considerable effect on NPLS susceptibility. The steps necessary to validate the QTLs and to improve the NPLS resistance by breeding were discussed.  相似文献

Phosphorus (P) is an important macronutrient required for plant growth and yield formation. Since decades, breeders aim to optimize P efficiency in crops. We studied a set of 47 wild barley (Hordeum vulgare ssp. spontaneum, Hsp) introgression lines (ILs) in hydroponic culture to identify quantitative trait loci (QTLs) improving growth and nutrient content under P deficiency. Applying a mixed model analysis, a total of 91 independent QTLs were located among 39 ILs, of which 64 QTLs displayed trait‐improving Hsp effects. For example, an unknown Hsp allele on barley chromosome 4H increased shoot dry weight under P deficiency in three overlapping ILs by 25.9%. Likewise, an Hsp allele on barley chromosome 6H increased root dry weight under P deficiency in two overlapping ILs by 27.6%. In total, 31 QTLs confirmed Hsp effects already identified in previous field and glasshouse experiments with the same ILs. We conclude that wild barley contains numerous trait‐improving QTL alleles, which are active under P deficiency. In future, the underlying genes can be subjected to cloning and, simultaneously, used in elite barley breeding.  相似文献

Leaf rust of barley, caused by Puccinia hordei Otth, is an important foliar disease in most temperate regions of the world. Sixteen major leaf rust resistance (Rph) genes have been described from barley, but only a few have been mapped. The leaf rust resistance gene Rph7 was first described from the cultivar ‘Cebada Capa’ and has proven effective in Europe. Previously mapped restriction fragment length polymorphism (RFLP) markers have been used to determine the precise location of this gene in the barley genome. From the genetic analysis of a ‘Bow‐man’/‘Cebada Capa’ cross, Rph7 was mapped to the end of chromosome 3HS, 1.3 recombination units distal to the RFLP marker cMWG691. A codominant cleaved amplified polymorphic site (CAPS) marker was developed by exploiting allele‐specific sequence information of the cMWG691 site and adjacent fragments of genomic DNA. Based on the large amount of polymorphism present in this region, the CAPS marker may be useful for the marker‐assisted selection of Rph7 in most diverse genetic backgrounds.  相似文献

Fusarium crown rot (FCR) is becoming a major disease in many parts of the cereal‐growing regions worldwide. Significant QTL conferring FCR resistance have been reported on 13 of the 21 possible hexaploid wheat chromosomes in wheat and on three of the seven chromosomes in barley. Available results show that host resistance to FCR is not pathogen species‐specific, that resistance QTL have strong additive effect and that both plant height and growth rate affect FCR severity. Further, different loci seem to be responsible for resistances to FCR and Fusarium head blight although both diseases can be caused by the same Fusarium pathogens. Although marker‐assisted selection for FCR resistance has been initiated, the available markers are all derived from QTL mapping, which provides only limited resolution. Further work has to be conducted in developing diagnostic markers before significant progress can be made in deploying marker‐assisted selection as a routine tool to accelerate and improve FCR in breeding programmes.  相似文献
