安徽省猪呼吸道疾病五种病原的分离与鉴定   总被引:1,自引:0,他引:1  
猪呼吸系统疾病目前是困扰我国养猪业主要疾病之一。为了弄清安徽省猪呼吸道疾病主要致病微生物,本研究利用细菌学鉴定技术、RT-PCR技术对2010年-2011年安徽省皖南、皖中、皖北地区采集的180份呼吸障碍性病(死)猪病料进行了猪繁殖与呼吸综合征病毒、圆环病毒2型、链球菌、巴氏杆菌、副猪嗜血杆菌等5种病原进行了分离鉴定。结果显示,猪繁殖与呼吸综合征病毒感染率高达68%,圆环病毒2型感染率高达83.3%;猪繁殖与呼吸综合征病毒与圆环病毒2型混合感染率达27.8%,猪链球菌感染率为22.8%。结果提示,猪繁殖与呼吸综合征病毒缺失株、圆环病毒2型、链球菌的普遍感染是近年安徽省猪呼吸道疾病的重要致病病原,研究结果为安徽省今后猪群呼吸道疫病防控策略的制定提供了参考依据。  相似文献

猪源多杀性巴氏杆菌的生物学鉴定与荚膜PCR分型   总被引:8,自引:1,他引:8  
从表现猪萎缩性鼻炎临床症状的猪群中分离出13株多杀性巴氏杆菌(Pasteurella multocida,Pm),采用Pm种特异性的KMT1-KMT2引物进行PCR扩增,结果与传统的生化反应鉴定完全一致。基于对甘露醇、卫茅醇、山梨醇、海藻糖的发酵能力和产生鸟氨酸脱羧酶的特性,Q_1、Q_3、Q_6、Q_7、Q_(10)、Q_(13)和C_(48-1)鉴定为Pm多杀亚种(Pm subsp.multocida);Q_2、Q_4、Q_5、Q_8、Q_9、Q_(11)、Q_(12)鉴定为Pm败血亚种(Pm subsp.septica)。对13株Pm分离物采用A型、B型和D型引物进行PCR扩增,8株鉴定为A血清型(61.5%);5株鉴定为D血清型(38.5%);没有发现B血清型的菌株。同时对金黄色葡萄球菌抑制试验、中性吖啶黄沉淀试验与荚膜PCR分型的相关性进行了讨论。  相似文献

为了解2017年我国猪源多杀性巴氏杆菌(Pasteurella multocida, Pm)的流行情况及其耐药情况,利用细菌分离培养和PCR方法分离鉴定Pm,对其荚膜血清型、地区分布和感染方式进行调查,并采用纸片法测定分离株对20种抗菌药物的敏感性。结果显示,从全国6288份临床病料中分离并鉴定出236株Pm,总分离率为3.75%。随机选取55株进行血清型分型,其中A型25株,D型27株,F型1株,未定型2株;分离率最高的省份为广东省(4.97%),其次是河南省(3.69%)和湖北省(3.25%);感染模式中,Pm单纯感染占比42.80%,混合感染占比57.2%,最常与链球菌和副猪嗜血杆菌共感染,比例分别为28.81%和12.29%。耐药性试验显示,Pm对强力霉素的耐药率高达83.64%,对卡那霉素、庆大霉素、链霉素和阿米卡星等氨基糖苷类药物的耐药率为43.64%~52.73%,对多粘菌素B和氧氟沙星最敏感(耐药率均为1.82%)。本研究表明我国猪群中Pm流行的主要血清型仍然是A型和D型,临床上Pm与其他细菌发生混合感染的情况普遍存在,并且对多种抗生素产生了耐药性。  相似文献

为了解云南省宣威市某规模化猪场育肥猪发生大量死亡的原因,试验采用临床症状观察、剖检病变、细菌分离培养、ELISA和PCR/RT-PCR方法进行检测,并对所分离的细菌进行形态观察、生化鉴定和药敏试验。结果表明:所分离的细菌形态特征及生化特性与猪胸膜肺炎放线杆菌一致,分离菌对氟苯尼考和卡那霉素高度敏感。RT-PCR法检出猪繁殖与呼吸综合征病毒特异性条带,判为阳性;样品的猪繁殖与呼吸综合征病毒抗体阳性率为61%(11/18)。PCR法检出胸膜肺炎放线杆菌,抗体阳性率为78%(14/18)。说明该猪场猪大量死亡的原因为猪繁殖与呼吸综合征和传染性胸膜肺炎混合感染。  相似文献

在已建立的副猪嗜血杆菌(HPS)、多杀性巴氏杆菌(Pm)、胸膜肺炎放线杆菌(APP)的单重PCR检测方法的基础上,通过对扩增条件的优化,利用一次PCR反应可同时扩增出APP的256 bp、Pm的457 bp、HPS的822 bp的特异性片段,建立了HPS、Pm、APP的多重PCR实验室诊断方法。该复合PCR可检测60 pg的HPS、120 pg的Pm、50 pg的APP。利用该方法检测猪繁殖与呼吸综合征病毒(PRRSV)阳性猪体内分离到的39株细菌,结果显示HPS、Pm、APP分别为12、16、2株,对39份病料的检测与常规细菌分离鉴定结果一致,可见该方法适用于HPS、APP、Pm的临床快速检测。  相似文献

为确诊贵州省都匀市某养殖场的猪死亡病因,采集病死猪病料进行猪繁殖与呼吸综合征病毒、猪瘟病毒、猪伪狂犬病病毒、猪圆环病毒2型核酸检测,细菌分离鉴定及药物敏感试验。结果:猪繁殖与呼吸综合征病毒实时荧光定量RT-PCR检测和猪圆环病毒2型PCR检测核酸均为阳性,其余病毒核酸检测均为阴性。细菌分离培养表明存在细菌感染,分子生物学鉴定为奇异变形杆菌;分离菌对硫酸安普霉素、硫酸粘菌素、盐酸大观霉素+盐酸林可霉素、氟苯尼考较敏感,对多种药物产生耐药。结论:确诊病例为猪繁殖与呼吸综合征病毒、猪圆环病毒2型、奇异变形杆菌混合感染。  相似文献

为确诊贵州省某猪场发病原因,对送检病料进行细菌分离鉴定和病毒核酸检测。结果:猪繁殖与呼吸综合征病毒实时定量荧光RT-PCR核酸检测为阳性;猪圆环病毒2型PCR扩增出大小为325 bp目的基因条带;细菌分离与生化鉴定显示存在链球菌感染,且药敏试验结果表明分离菌对硫酸安普霉素、延胡索酸泰妙菌素高度敏感。实验结果诊断为猪圆环病毒2型、猪繁殖与呼吸综合征病毒、链球菌混合感染。  相似文献

【目的】掌握中国规模化猪场猪呼吸道疾病综合征主要细菌性病原的感染情况。【方法】2021年采集来自全国26个省、市、自治区的647个规模化猪场中有明显呼吸道症状猪的肺脏、气管及分泌物、脾脏和淋巴结等病料2 204份。将病料接种平板后进行纯化培养,可疑菌落通过形态学观察、革兰染色和PCR进行鉴定,对分离的猪链球菌(Streptococcus suis,SS)、副猪嗜血杆菌(Haemophilus parasuis,Hps)、多杀性巴氏杆菌(Pasteurella multocida,Pm)、支气管败血波氏杆菌(Bordetella bronchiseptica,Bb)和胸膜肺炎放线杆菌(Actinobacillus pleuropneumoniae,APP)进行血清型分析、混合感染分析和药敏试验。【结果】从病料中分离鉴定出647株病原菌,病原菌分离率为29.35%(647/2 204),其中包括306株SS(13.88%)、137株Hps(6.22%)、86株Pm(3.90%)、74株Bb(3.35%)和44株APP(2.00%)。对分离到的SS、Hps和APP进行血清型鉴定,其中SS以血...  相似文献

Associations between pathogens in healthy pigs and pigs with pneumonia   总被引:2,自引:0,他引:2  
The aim of this study was to evaluate the associations between different pathogens in the development of pneumonia and bronchopneumonia in pigs. Samples of bronchoalveolar lavage fluid from 100 pigs showing no clinical signs and 239 pigs with clinical signs of respiratory disease were examined for Mycoplasma hyopneumoniae, Mycoplasma hyorhinis, US-type porcine respiratory and reproductive syndrome virus (PRRSV), EU-type PRRSV, porcine circovirus type 2 (pcv-2), influenza virus type A, alpha-haemolytic Streptococcus species, beta-haemolytic Streptococcus species, Pasteurella multocida, Bordetella bronchiseptica, Haemophilus parasuis and Actinobacillus pleuropneumoniae. These potential pathogens were detected more frequently in the pigs with respiratory problems than in the pigs with no clinical signs. pcv-2 and alpha-haemolytic streptococci were the pathogens most frequently detected; A pleuropneumoniae was isolated in only two cases. There were more often associations between the organisms in the pigs with clinical signs than in the healthy pigs. In particular, alpha-haemolytic streptococci and M hyopneumoniae were both associated with the presence of M hyorhinis, EU-type PRRSV, P multocida and B bronchiseptica, and alpha-haemolytic streptococci also occurred more often in pigs that were already infected with other pathogens. P multocida and B bronchiseptica were both significantly associated with M hyopneumoniae, alpha-haemolytic streptococci, EU-type PRRSV and US-type PRRSV.  相似文献

OBJECTIVE: To determine effects of intranasal inoculation with porcine reproductive and respiratory syndrome virus (PRRSV) or Bordetella bronchiseptica on challenge with nontoxigenic Pasteurella multocida in pigs. ANIMALS: Seventy 3-week-old pigs. PROCEDURE: In experiment 1, pigs were not inoculated (n= 10) or were inoculated with PRRSV (10), P. multocida (10), or PRRSV followed by challenge with P. multocida (10). In experiment 2, pigs were not inoculated (n = 10) or were inoculated with B. bronchiseptica (10) or PRRSV and B. bronchiseptica (10); all pigs were challenged with P. multocida. Five pigs from each group were necropsied 14 and 21 days after initial inoculations. RESULTS: Pasteurella multocida was not isolated from tissue specimens of pigs challenged with P. multocida alone or after inoculation with PRRSV. However, in pigs challenged after inoculation with B. bronchiseptica, P. multocida was isolated from specimens of the nasal cavity and tonsil of the soft palate. Number of bacteria isolated increased in pigs challenged after coinoculation with PRRSV and B. bronchiseptica, and all 3 agents were isolated from pneumonic lesions in these pigs. CONCLUSIONS AND CLINICAL RELEVANCE: Infection of pigs with B. bronchiseptica but not PRRSV prior to challenge with P. multocida resulted in colonization of the upper respiratory tract and tonsil of the soft palate with P. multocida. Coinfection with PRRSV and B. bronchiseptica predisposed pigs to infection of the upper respiratory tract and lung with P. multocida. Porcine reproductive and respiratory syndrome virus and B. bronchiseptica may interact to adversely affect respiratory tract defense mechanisms, leaving pigs especially vulnerable to infection with secondary agents such as P. multocida.  相似文献

Objective To provide serological data to support Australia's claim of freedom from porcine reproductive and respiratory syndrome.
Design A national serological survey was designed to provide 99% confidence of detecting at least one infected pig herd in Australia, assuming that at least 5% of herds would have been exposed to porcine reproductive and respiratory syndrome virus and that at least 25% of the 'finisher' pigs in these herds would have antibodies to the virus.
Procedure A two-stage testing regime was used. All samples were tested with a commercially available enzyme-linked immunosorbent assay. If assay reactions were found, all samples from the herd were to be tested using the indirect immunofluorescence antibody assay.
Results Of the 875 samples from 163 herds from all States in Australia, there was some evidence of reactivity in only four samples from four herds on the enzyme-linked immunosorbent assay. Further testing using the indirect immunofluorescence antibody assay according to the study protocol demonstrated that the reactions were not due to the presence of specific porcine reproductive and respiratory syndrome virus antibodies in the sera.
Conclusion The results of this study support the view that Australian pigs are free of porcine reproductive and respiratory syndrome virus.  相似文献

为了了解唐山地区猪蓝耳病的带毒情况,采用RT-PCR方法对2006-2011年采自64个县级屠宰场276份商品猪的肺脏或肺门淋巴结进行猪繁殖与呼吸综合征病毒(PRRSV)病原学检测,结果屠宰场阳性率为51.56%,样品总阳性率为20.29%,其中高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)样品总阳性率为15.94%,经典猪繁殖与呼吸综合征病毒(C-PRRSV)样品总阳性率为4.35%。总之,猪繁殖与呼吸综合征病毒,尤其是高致病性猪繁殖与呼吸综合征病毒在唐山地区猪群中污染面积较大,应值得高度重视。  相似文献

对2006~2009年河北省“猪高热病”的主要病原进行了调查分析。结果表明,猪繁殖与呼吸综合征病毒(PRRSV)、猪圆环病毒Ⅱ型(PCV-2)、猪伪狂犬病毒(PRv)、猪瘟病毒(CSFV)、副猪嗜血杆菌(HPS)、胸膜肺炎放线茵(APP)、链球菌(S.S)、大肠杆菌(E。coli)、沙门氏菌(Sal.)和巴氏杆菌(Pro)的总检出率分别为64.52%、30.27%、11.26%、6.13%、3.10%、2.58%、1.46%、1.11%、1.08%和0.10%。其中高致病性猪繁殖与呼吸综合征病毒(HP—PKP.SV)和经典猪繁殖与呼吸综合征病毒(C—PV.P.SV)总检出率分别为60.92%和3.60%。HP—PRIKSV同其他病原体混合感染比较普遍,感染以二重感染、三重感染为主。总之,河北省“猪高热病”的主要致病病原是HP-PRRSV,并多与PCV-2、PRV、CSFV、HPS、APP、E.cold、S.S和Sal.、Pm等病原体中的-种或几种病原混合感染或继发感染-种“综合症候群”型疫病。  相似文献

Pasteurella multocida is a widespread respiratory pathogen in pigs associated with atrophic rhinitis and contributing to aggravation of the pulmonary lesions. The aims of the present study were to characterize isolates of P. multocida from porcine bronchopneumonia by pulsed-field gel electrophoresis (PFGE), PCR based capsular typing and multilocus sequence typing (MLST) and to compare clonal complexes outlined with the type of histological lung lesions to investigate if a correlation between clonal lineages and lesions might exist. Isolates of P. multocida were obtained from cases of cranioventrally located porcine bronchopneumonia. All lung lesions were described and classified according to histological lesions. A total of 139 isolates, from lung (n=111), pericardial sac (n=21) and kidney (n=7) of 111 pigs were described using PFGE with ApaI as the restriction enzyme. Furthermore, 20 and 29 isolates were characterized by capsular serotyping and multilocus sequence typing, respectively. PFGE demonstrated 15 different clusters showing 50% or more similarity. All selected isolates were of capsular serotype A and only three main sequence types (ST) were detected among the isolates. Associations were not found between histopathology and clonal complexes of P. multocida. In conclusion, PFGE demonstrated a high diversity of genotypes of P. multocida associated with porcine bronchopneumonia. However, isolates obtained mainly belonged to few STs, indicating that isolates of P. multocida associated with porcine bronchopneumonia originates from a limited number of clonal lineages and therefore might have adapted to porcine hosts. No correlation was demonstrated between genotypes and types of lesions, and extra-pulmonary spreading was only rarely demonstrated.  相似文献

Association of porcine circovirus 2 with porcine respiratory disease complex   总被引:31,自引:0,他引:31  
A retrospective study was performed on natural cases of porcine respiratory disease complex (PRDC) to determine the association and prevalence of PRDC with porcine circovirus 2 (PCV2) and other co-existing pathogens in Korea. Histologically, alveolar septa were markedly thickened by infiltrates of mononuclear cells. Moderate to marked multifocal peribronchial and peribronchiolar fibrosis were present and often extended into the airway lamina propria. Among the 105 pigs with PRDC, 85 were positive for PCV2, 66 were positive for porcine reproductive and respiratory syndrome virus (PRRSV), 60 were positive for porcine parvovirus (PPV), and 14 were positive for swine influenza virus (SIV). There were 80 co-infections and 25 single infections. A co-infection of PCV2 with another additional bacterial pathogen is frequently diagnosed in PRDC. The combination of PCV2 and Pasteurella multocida (38 cases) was most prevalent followed by PCV2 and Mycoplasma hyopneumoniae (33 cases). The consistent presence of PCV2, but lower prevalence of other viral and bacterial pathogens in all pigs examined with PRDC, has led us to speculate that PCV2 plays an important role in PRDC.  相似文献

The aim of this study was to evaluate the vasculature in porcine circovirus type 2-infected (PCV2-infected) lungs and to identify the PCV2 subtypes involved in porcine pneumonia. Pulmonary samples from 140 pigs, 2 weeks to 7 months of age, from 36 Hungarian commercial herds with clinical signs of respiratory disease were examined for the presence of respiratory pathogens, with bacterial culture, pathologic evaluation, and immunohistochemistry for PCV2, porcine reproductive respiratory syndrome virus, and swine influenza virus. PCV2 was the most commonly identified pathogen (49 cases) among the 74 of 140 cases (53%) with respiratory pathogens. PCV2 was detected immunohistochemically in the wall of 13% to 100% of pulmonary vessels (mean, 89%) in 38 of 49 cases (78%). Detection of PCV2 antigen was positively correlated with the presence of vascular lesions (P < .001, odds ratio [OR]: 159.54). Other pathogens capable of vascular injury in swine were found in 29 of 49 of the PCV2-positive cases (59%). The probability of detecting vascular lesions in PCV2-infected lung was higher than in infection with porcine reproductive respiratory syndrome virus (P < .002, OR: 14.63), Pasteurella multocida infection (P < .001, OR: 5.75), or Streptococcus spp. infection (not significant, OR: 1.45). Sequence analysis of open reading frame 2 amplicons was possible in 6 PCV2-positive cases, from which 5 cases proved to be PCV2b subtype and 1 case, PCV2a subtype. In conclusion, PCV2 antigen was commonly colocalized with pulmonary vascular lesions in pneumonia in Hungarian swine, and PCV2b was the dominant subtype.  相似文献
