Studies were made on the distribution of lymphoid tissues and immunoglobulin (Ig: IgA, IgG and IgM)-containing cells (cIg: cIgA, cIgG and cIgM) and the localization of immunoglobulins (Igs) in the oviducal walls of laying hens. Lymphocyte accumulations were occasionally observed, located mainly in the middle infundibulum and in the regions from the isthmus to the vagina. The number of cIgG significantly predominated over that of cIgA or cIgM in the mucosal connective tissue of the magnum and the isthmus. In contrast, in the regions other than the magnum and the isthmus, these three types of cIg were fewer in number. Igs were localized in some superficial epithelial cells (SECs) and glandular cells (GlCs) of the oviduct. Many IgG-containing SECs were found in the infundibulum, the isthmus, and the cranial and major uterus. IgA- or IgM-containing SECs were rare throughout the oviduct. Three types of Ig-containing GlCs were numerously found in the magnum, though lymphocyte accumulations were scarce there. In the isthmus, many IgG-containing GlCs were found, while IgA- or IgM-containing GlCs were rarely observed. Ig-containing GlCs in the magnum were considerably decreased in number after the egg passage. The results suggest that the maternal Igs are transferred to the egg mainly through GlCs in the magnum of the chicken oviduct, and that the oviducal lymphoid tissues have little relationship to the passive immunity.  相似文献

The morphological characteristics of the oviduct of 12 sexually mature rheas (Rhea americana) were studied. Only the left oviduct is developed as a long tube with a length of 122 +/- 23.1 cm, and is subdivided into infundibulum (15.2 +/- 4.0 cm), magnum (63.3 +/- 9.4 cm), isthmus (5.6 +/- 3.1 cm), uterus (16.0 +/- 4.2 cm) and vagina (11.5 +/- 1.4 cm). The mucous membrane of the oviduct, as a whole, possesses luminal folds covered by ciliated columnar epithelium with secretory cells. The infundibulum part presents a cranial opening with thin and long fimbriae with few tubular glands in caudal tubular portion. In the magnum, the largest portion of the oviduct, the folds are thicker and are filled with tubular glands. The isthmus is short and presents less bulky folds and a few tubular glands. A bag-shaped uterus in the cranial area shows thin folds, and in the caudal region (shell gland) more ramified folds with few tubular glands. The vagina has long luminal folds and a thick muscular tunic; no glands with sperm-storage characteristics have been observed. In conclusion, the oviduct in sexually mature rhea has morphological similarities with the other species of birds already described; however it presents its own characteristics to produce a big egg.  相似文献

A biometrical study of the reproductive tracts of 173 hens was carried out after slaughter at 53 weeks of age.

Mean weights and total lengths of the reproductive tracts of four strains of actively laying turkey hens were 265 - 279.5 grams and 80.9 - 84.5 centimetres. Mean measurements in centimetres of oviduct anatomical divisions were: infundibulum 10.9 - 11.7, magnum 44.5 - 45.2, isthmus 12.6 - 14.5, uterus 9.6 - 10.8, and vagina 2.6 - 3.5.

This study describes the distribution of vimentin, desmin, smooth muscle actin (SMA) and laminin in the oviduct of the laying domestic fowl. Vimentin immunostaining was localised in the luminal epithelium of the infundibulum, magnum, magnum–isthmus junction and isthmus. The luminal epithelium of the shell gland regions displayed weak vimentin immunostaining. Vimentin immunostaining was demonstrated in the glandular grooves of the tubular infundibular region. In contrast, gland cells in the magnum, isthmus and shell gland regions were vimentin immunonegative. Fibroblasts and vascular endothelial cells in the lamina propria of the oviductal regions studied exhibited vimentin immunostaining. Strong desmin and SMA immunostaining were present in the smooth muscle cells of the tunica muscularis and vascular tunica media. In this study, basement membranes underlying the luminal and glandular epithelia were immunopositive for laminin. In addition, basement membranes associated with smooth muscle cells exhibited laminin immunostaining. The results of the study indicate that the immunolocalisation of desmin, SMA and laminin in the oviduct of the domestic fowl is similar to that in the mammalian uterus. The immunolocalisation of vimentin in the domestic fowl varies depending on the oviductal region.  相似文献

1. Our aim was to determine the presence and numbers of immunocompetent cells in the oviduct of laying and moulting hens. Immunocompetent cells were localised by immunocytochemistry throughout the entire length of the oviduct.

2. In laying birds, MHC class II + cells were observed in the subepithelial and middle part of the stroma of all oviducal segments and the mucosal epithelium of the infundibulum and vagina. CD3 + cells were also localised in subepithelial and middle part of stroma as well as in mucosal epithelium of each oviducal segment. Bu‐lb + and IgG + cells were also observed in the epithelium and subepithelial and middle part of the stroma of all oviducal segments, though stroma of the magnum, isthmus and uterus contained few Bu‐lb + cells. IgA + cells were observed only in the mucosal epithelium of the magnum in small numbers.

3. In moulting hens, there were few numbers of immunocompetent cells in the mucosal epithelium of each oviducal segment, although CD3 + cells were observed in the infundibulum and vagina. In the subepithelial stroma, the populations of MHC class II + cells in the infundibulum, magnum and uterus, CD3 + cells in the infundibulum and vagina, as well as IgG + cells in each oviducal segment except for isthmus were smaller than in laying hens. In contrast, the number of immunocompetent cells in the middle part of stroma of moulting hens were equal to or greater than in laying hens.

4. These results suggest that the oviducal immune function is active in the surface tissues of the mucosa in laying hens, whereas it is reduced in moulting hens.  相似文献

Activities of alkaline (ALP) and acid phosphomonoesterases (ACP) and aspartic (GOT) and alanine (GPT) aminotransferases in the glandular epithelia of the infundibulum, magnum, isthmus and shell gland of the laying quail were measured when the egg was in the shell gland. 2. The activities of all the enzymes were significantly (P less than 0.001) greater in shell-gland than in isthmus and magnum. 3. Three reproductively quiescent female emus (Dromaius novaehollandiae) were embalmed with 10% formalin solution. The reproductive tract was dissected and described. The reproductive tract consists of an ovary and oviduct situated on the left side of the abdominal cavity. The left ovary is dark brown to black in colour with follicles covering the ventral surface. The ovary is located medial to the spleen and closely associated with the ventral surface of the cranial and middle lobes of the left kidney. The oviduct is a relatively straight tube that extends from the level of the cranial extent of the left ilium to the caudal border of the left pubic bone. The oviduct is grossly divided into the infundibulum, magnum, isthmus, uterus and vagina using variations in the mucosal fold pattern.  相似文献

The contents of retinol and retinyl esters as well as retinol-binding protein (RBP) in the plasma, urine, liver and kidneys of dogs, raccoon dogs and silver foxes were investigated. In the plasma and urine of all three species, vitamin A was present as retinol and retinyl esters. Vitamin A levels (1376+/-669 microg x g(-1)) were significantly higher in the livers of dogs than in the kidneys (200+/-217 microg x g(-1), P < 0.001 ). However, vitamin A levels in the kidneys of raccoon dogs (291+/-146 microg x g(-1)) and silver foxes (474+/-200 microg x g(-1)) were significantly higher than in the liver (67+/-58 microg x g(-1) and 4.3+/-2.4 microg x g(-1), respectively, both P < 0.001). RBP was immunologically detected in the blood plasma of all species, but never in the urine. In the liver, immunoreactive RBP was found in hepatocytes. In the kidneys of all species, RBP was observed in the cells of the proximal convoluted tubules. The levels of vitamin A in the livers of raccoon dogs and silver foxes were extremely low, which would be interpreted as a sign of great deficiency in humans. This observation might indicate that the liver status cannot be used as an indicator of vitamin A deficiency in canines. The high levels of vitamin A in the kidneys in all three species may indicate a specific function of the kidney in the vitamin A metabolism of canines.  相似文献

In 13 anaesthetized hens in the peak phase of their first laying year the influence of intravenously injected substance P (SP), 1–10 g/animal, on oviductal pressure, duodenal pressure, blood pressure and heart rate has been studied within 5 h of oviposition. The neuropeptide induced a significant pressure increase in the different segments of the oviduct (infundibulum, magnum, isthmus, uterus and vagina) as well as in the duodenum. Blood pressure revealed a distinct biphasic response: a short period of hypotension accompanied by a tachycardia and a more pronounced and sustained hypertension, inducing a subsequent bradycardia. The complexity of the observed effects demonstrates the overall impact of intravenously administered SP on the anaesthetized hen.  相似文献

In this study we investigated the temporal relationship between ovulation, egg formation, oviposition and the changes in plasma concentrations of progesterone, luteinizing hormone and estradiol-17beta during the egg laying cycle in farmed ostriches. In 10 egg-producing birds, transcutaneous ultrasound scanning was performed at 3h intervals and blood sampling at hourly intervals during a period of at least 48h (one egg laying cycle). In hens (n=8) that ovulated during the observational period, the ovulated egg was first detected 2h after oviposition; thus, ovulation occurred shortly after oviposition in all birds. During the period between two consecutive ovipositions, the developing egg remained for 9h in the proximal part (infundibulum, magnum or isthmus) and for 39h in the distal part of the oviduct (uterus). In ovulating hens, plasma progesterone concentrations showed a characteristic and consistent profile: from basal levels of around 0.1ng/ml concentrations started to increase 12h before oviposition, reached an average maximum of 3.5ng/ml at 3h before oviposition and returned to basal levels 3h and 30min after oviposition. Changes in plasma luteinizing hormone and estradiol-17beta concentrations showed comparable patterns of elevation and decline relative to the timing of oviposition and ovulation. However, variation in their individual basal concentrations was generally larger and peak values were less conspicuous than those of progesterone. In non-ovulating hens (n=2) neither progesterone, nor luteinizing hormone nor estradiol-17beta showed elevations to peak concentrations before oviposition. These data demonstrate that during the egg laying cycle of ostriches, events such as ovulation, egg development and oviposition evolve according to a rather strict time schedule, and that progesterone, luteinizing hormone and estradiol-17beta reach peak concentrations shortly before ovulation. Additionally, our findings also show that on-farm ultrasound scanning is a useful technique to discriminate between ovulating and non-ovulating hens.  相似文献

Fasting dogs do transport vitamin A (VA) in plasma not only as retinol but predominantly as retinyl esters. Contrary to retinol, nothing is known concerning the effects of athletic performance on plasma retinyl ester concentrations. The aim of this study was therefore to examine whether physical stress because of exercise and modification of the oxidative stress by supplementation of alpha-tocopherol influences the concentrations of retinol and retinyl esters in plasma of sled dogs. The study was carried out on 41 trained adult sled dogs, which were randomly assigned into two groups. One group (19 dogs) was daily substituted with 50 mg dl-alpha-tocopheryl acetate per kilogram body weight and the control group (22 dogs) was maintained on a basal diet during 3 months prior to exercise. The plasma concentrations of retinol, retinyl esters, alpha-tocopherol and triglycerides were measured immediately before, directly after and 24 h after exercise. The supplementation of alpha-tocopheryl acetate had no effect on plasma retinol and retinyl ester concentrations at any measurement time point. However, retinyl ester levels doubled in the non-supplemented group immediately after the race (p < 0.001), whereas in the supplemented group similar high levels were observed not until 24 h post-racing (p < 0.001). The high levels of retinyl esters were paralleled to some extent by an increase in plasma triglyceride concentrations, which were significantly higher 24 h post-racing than immediately before (p < 0.001) and after exercise (p < 0.001) in both groups. The increase in retinyl ester concentrations might be indicative of their mobilization from liver and adipose tissue. Whether plasma retinyl esters can be used as an indicator for the extent of nutrient mobilization during and post-exercise in sled dogs remains to be elucidated.  相似文献

OBJECTIVE: To determine plasma and urine concentrations of retinol, retinyl esters, retinol-binding protein (RBP), and Tamm-Horsfall protein (THP) in dogs with chronic renal disease (CRD). ANIMALS: 17 dogs with naturally developing CRD and 21 healthy control dogs. PROCEDURE: A diagnosis of CRD was established on the basis of clinical signs, plasma concentrations of creatinine and urea, and results of urinalysis. Concentrations of retinol and retinyl esters were measured by use of reverse-phase high-performance liquid chromatography. Concentrations of RBP and THP were measured by use of sensitive ELISA systems. RESULTS: Dogs with CRD had higher plasma concentrations of retinol, which were not paralleled by differences in plasma concentrations of RBP. Calculated ratio of urinary total vitamin A (sum of concentrations of retinol and retinyl esters to creatinine concentration) and ratio of the concentration of urinary retinyl esters to creatinine concentration did not differ between groups. However, we detected a significantly higher retinol-to-creatinine ratio in the urine of dogs with CRD, which was paralleled by a higher urinary RBP-to-creatinine ratio. Thus, in dogs with CRD, the estimated fractional clearance of total vitamin A, retinol, and RBP was increased. Furthermore, dogs with CRD had a reduced urinary THP-to-creatinine ratio. CONCLUSIONS AND CLINICAL RELEVANCE: Results of this study documented that CRD affects the concentrations of retinol in plasma and urine of dogs. Analysis of the data indicates that measurement of urinary RBP and urinary THP concentrations provides valuable information that can be helpful in follow-up monitoring of dogs with CRD.  相似文献

Vitamin A (VA) deficiency and Tamm-Horsfall glycoprotein (THP), a protein that binds retinol and retinyl esters in canine urine, might be involved in the pathogenesis of urolithiasis in dogs. In the present study, we assessed levels of retinol, retinyl esters, retinol-binding protein (RBP) and THP in plasma and urine of dogs with a history of urolithiasis (n = 25) compared with clinically healthy controls (n = 18). Plasma retinol concentrations were higher in dogs with uroliths of struvit (P < 0.01), calcium oxalate (P < 0.05), urate (P < 0.01) and cysteine, but there were no differences in the concentrations of plasma RBP and retinyl esters. Excretion of urinary retinol and retinyl esters were tentatively, but not significantly higher in the stone-forming groups, which was accompanied by increased levels of urinary RBP (P < 0.01) and lower excretions in THP (P < 0.01). The results show that VA deficiency may be excluded as a potential cause for canine urolithiasis. However, the occurrence of RBP and a concomitant reduction of THP in urine indicates a disturbed kidney function as cause or consequence of stone formation in dogs.  相似文献

The infundibulum and magnum of the oviduct were examined in hens in full lay which were infected with two Australian strains of infectious bronchitis virus (IBV). The ultramicroscopic changes in the infundibulum and magnum were compared with control hens which had eggs at different positions in the oviduct. The ciliated and granular cells of the surface epithelia and secretory epithelial cells of the tubular glands were the target cells of IBV. No pathological changes were recorded during 2-8 days post-infection (p.i.). Patchy loss of cilia occurred at 10-14 days p.i. Between 16 and 24 days p.i., there was no cilia loss and lymphoid nodules were observed in the muscularis layer of the infundibulum and magnum of some hens from both infected groups. Virus particles were detected mostly in the rough endoplasmic reticulum (RER) and Golgi complex between 10 and 12 days p.i. Cytopathology was noticed in various cell organelles between the 10th and 14th days p.i. There was an increase in RER deposits in infected cells, irrespective of egg position in the oviduct. The magnum was more affected than the infundibulum. Cellular changes were more severe in the infundibulum and magnum of T-infected hens as compared to N1/88-infected hens. Eggs with watery whites which were laid by infected hens could be attributed to cytopathological changes in the granular epithelial cells and tubular gland epithelial cells of the magnum resulting in reduced synthesis of albumen proteins. IBV can cause pathology in parts of the fully functional oviduct which may persist up to the 30th day p.i. However, both the challenge strains of IBV can cause a small number of hens to cease production. Loss of cilia in both the infundibulum and magnum pose a potential threat of secondary bacterial infection and also may affect fertility in breeder hens.  相似文献

1. In the present study the distribution of large granular lymphocytes (LGLs) and influence of oestrogen on these cells was investigated in the oviduct of chickens by the observation of H and E stained sections. 2. The LGLs first appeared in the oviduct at 9 weeks and their frequency of occurrence increased with age and was highest in the infundibulum and vagina from 21 to 32 weeks of age. The frequency of occurrence of LGLs from magnum to uterus (glandular part) was low. These cells were located at the base, middle and apical parts of the lining epithelium as well as in the sperm-storage glandular epithelium. 3. When chickens of 21 weeks were treated with DES, LGLs abundantly infiltrated in the oviducal epithelium in comparison to the control. 4. These results suggested that the development of LGLs in the oviduct of chickens correlates with oestrogen secretion.  相似文献

Vitamin A transport in blood of most species in the order carnivora is fundamentally different from other animals and man. Very high levels of vitamin A in blood plasma of canines and mustelides are due to a high percentage of retinyl esters. These esters (basically retinyl palmitate and stearate) are transported nonspecifically bound to lipoproteins with very-low and low density lipoproteins representing the major fractions (about 70% of total vitamin A esters). A high percentage of retinyl esters could also be found in the families Ursidae, Procyonidae, Viverridae and Felidae in which total vitamin A in blood was low. In man and rat severe signs of vitamin A intoxication can be observed due to a similarly high percentage of nonspecifically bound vitamin A esters. No clinical or clinical-chemical signs of vitamin A intoxication were observed in the animals investigated. Because vitamin A esters are transported by lipoproteins, their blood levels are readily affected by dietary vitamin A supply as it is known to be the case for vitamin E. Furthermore, canines seem to be the only ones that excrete vitamin A as retinol and retinyl esters (basically retinyl palmitate) under physiological conditions with the urine. These fat-soluble metabolites of vitamin A are bound to a protein with a molecular weight between 50,000 and 125,000 Da which shows characteristics of a lipoprotein when subjected to ultracentrifugation or selective precipitation. Higher levels of vitamin A in organs and tissues, as well as the excretion of vitamin A with the urine, might be a consequence of the nonspecific binding of large amounts of retinyl esters to lipoproteins in blood.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The aim of the study was to contribute to the discussion of overdosing vitamin E in laying hens. A total of 45 laying hens, divided into 5 groups were fed diets supplemented with either 0; 100; 1000; 10,000 or 20,000 mg dl-alpha-tocopheryl acetate/kg diet over a period of 10 weeks. Concentrations of vitamins A and E were measured in plasma, various tissues and egg yolk. Furthermore egg yolk colour and some carotinoids were measured in egg yolks. None of the vitamin E doses significantly influenced performance of the hens. As expected, vitamin E concentration in plasma, all tissue samples and egg yolk was significantly increased with increasing tocopherol content in the diet. The egg yolk showed the highest vitamin E concentration, followed by liver and muscles. Feeding 1000 mg alpha-tocopheryl acetate per kg diet resulted in an increase of vitamin A concentration in the liver. Very high doses (10,000 and 20,000 mg/kg diet) significantly decreased retinol concentration in the liver and egg yolk, as well as carotinoid concentration in the egg yolk. The lower carotinoid concentration in egg yolk resulted in a decreased intensity of egg yolk colour. A prooxidative and/or competitive effect of very high doses of vitamin E with other fat soluble substances has been discussed.  相似文献

1. Eighteen Warren SSL hens of 71 weeks of age were forced-moulted by ad libitum feeding of a high-zinc diet (10,000 ppm zinc for 2 days followed by 5,000 ppm zinc-supplement diet for 4 days). From the start of the treatment, eggs were collected and 3 hens were slaughtered on days 0, 2, 3, 4, 5 and 6 of the study. 2. Zinc analyses were carried out on the different components of the eggs and on liver, pancreas, kidney, different yolky follicles of the ovary and various segments of the oviduct. 3. Seven-, six- and threefold increases in zinc concentration were found in pancreas, liver and kidney, respectively. 4. The shell gland and isthmus, but not the magnum, also showed slight but significant increases in Zn content. 5. Zinc accumulation was also high and almost identical in ovarian follicles F1 to F4 but slightly less in F5 and F6 follicles. 6. In the egg, a significant increase in zinc concentration was only observed in the yolk.  相似文献

In vitro protein binding of spiramycin (SP) in the plasma and oviducts of laying hens was studied. The data for SP were compared with those for oxytetracycline (OTC), sulphadimidine (SDD), sulphamonomethoxine (SMM) and sulphaquinoxaline (SQ). The two oviduct segments, magnum (M) and isthmus plus shell gland (IS), were collected. The soluble (cell sap) fractions from the magnum (M-S9) and the isthmus plus shell gland (IS-S9) were used as samples. Plasma protein binding was highest for SQ (81.4%) (P < 0.01), and lowest for SDD (30.9%) (P < 0.01). No M-S9 protein binding of OTC was found. The IS-S9 protein binding of SP (60.4%) was very much higher than those of OTC (0.8%), SDD (4.1%), SMM (4.0%) and SQ (12.3%) (P < 0.01). Biological half-lives of these drugs in egg albumen were directly correlated to the extent of their binding to IS proteins. Of plasma, M-S9 and IS-S9, variation in SP concentration in the ranges from 1 to 20 micrograms/ml did not alter the binding properties of the drug.  相似文献

The objective of this study was to evaluate the effect of feeding laying hens diets containing cashew nut shell liquid (CNSL) as a source of anacardic acid on the blood biochemical parameters as well as the enzymatic activity and lipid peroxidation of liver and tissues of the reproductive system (ovary, magnum, and uterus). A total of 216 Hisex White commercial laying hens were distributed randomly into six treatments, with six replicates of six birds. Treatments consisted of a diet without growth promoter (GP); a diet with GP; and diets without GP, with addition of increasing levels of CNSL (0.25, 0.50, 0.75 and 1.0%). Addition of CNSL to the diet did not affect the blood biochemical parameters (uric acid, creatinine, alanine aminotransferase, aspartate aminotransferase, total cholesterol, high density lipoproteins, low‐density lipoproteins and triglycerides), the enzymatic activity (superoxide dismutase and nonprotein sulphydryl groups) in the organs (liver, ovary, magnum and uterus) or the peroxidation of lipids from the blood serum, liver, magnum and uterus (p > 0.05). However, the addition of 0.75% and 1.00% CNSL provided a lower thiobarbituric acid reactive substances content in the birds' ovary (p < 0.001) compared to birds of other treatments, whereas the treatment without the GP provided a higher value. Addition of up to 1% of the CNSL as a source of anacardic acid in the laying hens' diets does not influence blood biochemical parameters or the endogenous enzymatic activity in the liver, ovary, magnum and uterus, but affects the lipid peroxidation in the ovary, although the problem is reduced from the inclusion of 0.75% CNSL.  相似文献
