The genetics of host-pathogen interactions in the Hordeum vulgare – P. teres f. teres pathosystem was studied in twelve resistant barley accessions, i.e. CI 9825, CI 9819, Diamond, CI 4922, CI 5401, Harbin, c-8755, c-21849, c-8721 c-23874, c-19979, c-15811. F2 analyses of crosses with susceptible genotypes employing various isolates (from Europe, USA, Canada, and Australia) revealed that resistance is mostly isolate-specific and controlled by one or two genes. Segregation in ascospore progeny from two crosses between isolates of different origin revealed that avirulence in P. teres is also determined by one or two genes. An epistatic effect of suppressor genes on avirulence genes is proposed for the genetics of virulence to Diamond, Harbin, CI 5401 and c-8721 in the fungal crosses D (181-6 × A80) and F (H-22 × 92-178/9). Segregation in F2 of crosses of three new sources of resistance (c-23874, c-19979, c-15811) to the susceptible cv. Pirkka was studied in laboratory and greenhouse tests by using seven P. teres isolates, i.e. 181-6, d8-3, d8-4, d9-1, d9-4, F4 and F74. In addition, virulence to these barley accessions of ascospore progeny from crosses of the same isolates was studied. Based on these studies it was concluded that depending on the isolate used, resistance of c-23874 is determined at least by two genes and in c-19979 and c-15811 by three genes. The results of this parallel analyses of genetics of resistance and genetics of virulence allows the postulation of a gene–for–gene interaction in the P. teres – H. vulgare pathosystem.  相似文献

To determine the potential of sexual reproduction among host-adapted populations of Phytophthora infestans sensu lato in Ecuador, 13 A1 isolates belonging to clonal lineages US-1, EC-1 and EC-3, and 11 A2 isolates belonging to the clonal lineage EC-2, were paired on agar plates to induce crossing. In the first experiment, six A1 isolates (three US-1, two EC-1 and one EC-3) were each crossed with three A2 isolates (total = 18 crosses). Matings involving isolates of the EC-1 lineage produced more oospores of healthy appearance than did matings with isolates of US-1 or EC-3. In the second experiment, the oospores of 35 crosses (21 EC-1 × EC-2; 10 US-1 × EC-2; four EC-3 × EC-2) were dispersed on water agar to assess oospore germination. Overall, germination percentages were low. Only one cross produced enough progeny for evaluation. Twenty-three single-oospore offspring were isolated and evaluated for mating type; electrophoretic patterns of glucose-6-phosphate isomerase ( Gpi ) and peptidase ( Pep ) alloenzyme loci; mitochondrial DNA haplotype; and genomic DNA fingerprint. Multilocus genotype data indicated that all 23 isolates resulted from meiotic recombination. Four progeny with homothallic phenotype appeared to be unstable heterokaryons. Markers at several loci segregated according to simple Mendelian expectations for a diploid organism, but the ratios of three RFLP loci and the Pep locus were not consistent with Mendelian expectations. All progeny were nonpathogenic on hosts of the parental genotypes. Reduced mating success and reduced pathogenic fitness of progeny appear to be postmating mechanisms of reproductive isolation in populations of P. infestans sensu lato in Ecuador.  相似文献

BACKGROUND: Tribolium castaneum (red flour beetle) is a serious insect pest of stored products around the world. Current control measures for this species have several limitations: loss of registration of insecticides, insecticide resistance and consumer concerns about chemical residues in food. The objective of this study was to determine whether methoprene affects progeny production of T. castaneum. Late‐instar larvae or young adults were exposed to methoprene‐treated wheat, and progeny production was determined. The pairing of male and female adults was performed as untreated × untreated, treated × untreated or treated × treated, to study sex‐based effects. RESULTS: There were three outcomes to late‐instar larvae held on methoprene‐treated wheat kernels (0.001 and 0.0165 ppm): (1) failure to emerge as an adult; (2) emergence as an adult, and almost no offspring produced; (3) emergence as an adult and normal production of offspring. Male larvae were more susceptible to methoprene than female larvae. In contrast, young adults exposed to methoprene (1.67–66.6 ppm) showed no reduction in offspring production. CONCLUSION: Methoprene concentrations will decline with time following its application. However, this research indicates that methoprene can still reduce populations of T. castaneum by reducing their progeny production, even if adults emerge. Sesamum mulayanum is a wild relative of cultivated sesame, Sesamum indicum, and sometimes grows in sesame crop fields as an associated weed. This species shows deep seed dormancy and is characterized by conspicuous purple pigmentation on the lower lip of the corolla. The present study examined the inheritance mode of seed dormancy by using reciprocal progeny from crosses between the two species. The seeds of S. indicum and F1 (S. indicum×S. mulayanum) showed good germination, but those of S. mulayanum and F1 (S. mulayanum×S. indicum) showed deep dormancy. The F2 seeds from both reciprocal crosses showed deep dormancy. These results, combined with the maternal inheritance of seed‐coat characteristics, indicated that the seed dormancy of S. mulayanum can be attributed to its seed‐coat structure (coat‐enhanced dormancy). The F3 (S. indicum×S. mulayanum) seeds varied in their depth of seed dormancy and those seeds with deep dormancy (<50% germination) and those with no or shallow dormancy (≥50% germination) occurred in the expected ratio of 3:1, indicating that this trait is polygenic but is controlled by a single dominant major gene. The purple pigmentation of the corolla was expressed in both reciprocal F1 plants and the presence and absence of pigmentation was segregated among the F2 plants at the expected ratio of 3:1, indicating that this trait is also controlled by a single dominant gene. The segregation of the major gene controlling seed dormancy and that controlling purple pigmentation was not independent (9:3:3:1), indicating that these genes are linked, providing insights on sesame domestication.  相似文献

A study was carried out to investigate the inheritance of resistance to anthracnose, caused by Colletotrichum sublineolum, in sorghum. Crosses between resistant and susceptible parents and backcrosses between F1 plants and the susceptible parents were carried out under field conditions. The F1 generations and the segregant populations were evaluated under artificial inoculation conditions in the greenhouse. In the F1 generation of all crosses with the respective isolates, all of the plants presented a resistance reaction except for the F1 plants resulting from the BR009 × SC283 cross. In the F2 generation, the frequencies of resistant and susceptible plants conformed to the hypothesis that one gene with two alleles controls host resistance, except in one cross. Out of the eight backcrosses, six presented segregation that corresponded to the hypothesis formulated. For most crosses, resistance was dominant, and the proportions of resistant and susceptible plants in the segregant populations conformed to the frequencies expected under the hypothesis of gene‐for‐gene resistance and dominant gene action.  相似文献

The population of Phytophthora infestans in Brazil consists of two clonal lineages, US-1 associated with tomatoes and BR-1 associated with potatoes. To assess whether host specificity in these lineages resulted from differences in aggressiveness to potato and tomato, six aggressiveness-related epidemiological components – infection frequency (IF), incubation period (IP), latent period (LP), lesion area (LA), lesion expansion rate (LER) and sporulation at several lesion ages (SSLA) – were measured on detached leaflets of late blight-susceptible potato and tomato plants. Infection frequency of US-1 was similar on potato and tomato leaflets, but IF of BR-1 was somewhat reduced on tomato. Incubation period was longer on both hosts with US-1, although this apparent lineage affect was not significant. Overall there was no host effect on IP. On potato, BR-1 had a shorter LP (110·3 h) and a larger LA (6·5 cm2) than US-1 (LP = 162·0 h; LA = 2·8 cm2). The highest LER resulted when isolates of BR-1 (0·121 cm2 h−1) and US-1 (0·053 cm2 h−1) were inoculated on potato and tomato leaflets, respectively. The highest values of the area under the sporulation capacity curve (AUSC) were obtained for isolates of US-1 inoculated on tomato leaflets (6146) and for isolates of BR-1 on potato leaflets (3775). In general, higher values of LA, LER, SSLA and AUSC, and shorter values of LP were measured when isolates of a clonal lineage were inoculated on their original host than with the opposite combinations. There is evidence that there are quantitative differences in aggressiveness components between isolates of US-1 and BR-1 clonal lineages that probably contribute to host specificity of P. infestans populations in Brazil.  相似文献

Limited knowledge is available on Phytophthora infestans populations in Sub‐Saharan Africa (SSA). Therefore, and in response to recent severe late blight epidemics, P. infestans isolates from potato, tomato and Petunia × hybrida from eight SSA countries were characterized. Isolates were characterized with ‘old’ markers, including mating type (176 isolates), mitochondrial DNA haplotype (mtDNA) (281 isolates), glucose‐6‐phosphate isomerase (Gpi) (70 isolates), restriction fragment length polymorphism analysis with probe RG‐57 (49 isolates), and by metalaxyl sensitivity (64 isolates). Most isolates belonged to the US‐1 genotype or its variants (US‐1.10 and US‐1.11). The exceptions were genotype KE‐1 isolates (A1 mating type, mtDNA haplotype Ia, Gpi 90/100 and unique RG‐57 genotype), identified in two fields in Kenya, which are related to genotypes previously identified in Rwanda (RW‐1 and RW‐2), Ecuador and Europe. Metalaxyl‐resistant P. infestans isolates from potato were present in all the countries except Malawi, whereas all the isolates from tomato were sensitive. Genotyping of 176 isolates with seven simple sequence repeat (SSR) markers, including locus D13 that was difficult to score, revealed 79 multilocus genotypes (MLGs) in SSA. When this locus was excluded, 35 MLGs were identified. Genetic differentiation estimates between regional populations from SAA were significant when locus D13 was either excluded (P = 0·05) or included (P = 0·007), but population differentiation was only low to moderate (FST = 0·044 and 0·053, respectively).  相似文献

The effect of RUS, a major quantitative resistance (QR) factor inherited from Populus trichocarpa, was assessed in two Populus deltoides × P. trichocarpa F1 progenies against eight isolates of Melampsora larici‐populina, the causal agent of Eurasian poplar rust. Six isolates were identified on which RUS had no significant effect. The first RUS‐defeating isolate identified suggested a pre‐existing potential to overcome RUS in the pathogen’s populations. The P. deltoides genetic background made no difference either to the ability of a given isolate to overcome RUS or to the relative and absolute aggressiveness of the isolates. This study illustrates how extreme the isolate‐specificity of QR can be. It also yields insights into the relationship between size of uredinia and sporulation rate, discussing the epidemiological significance of spore production per mm2 uredinium.  相似文献

ABSTRACT To test the hypothesis that host-related differences in the genotypic composition of populations of the late blight pathogen Phytophthora infestans can be explained by differential pathogenicity, the aggressiveness of isolates of the pathogen collected in France from potato and tomato was measured on detached leaflets of potato (cv. Bintje) and tomato (cv. Marmande). A preliminary trial with four isolates (two each from potato and tomato) showed that lesion appearance and development were similar for each isolate in detached leaflets and in whole plant tests in growth cabinets. Isolates collected from tomato were more pathogenic to tomato than isolates collected from potato. This was particularly the case for isolates belonging to the A2 mating type. Isolates originating from potato had a higher infection efficiency and a higher sporulation capacity on this host, but they induced lesions that generally spread more slowly than those caused by isolates from tomato. Extensive variation for components of aggressiveness on potato, and to a lesser extent on tomato, was observed in collections of isolates from each of the two hosts. Competition experiments between one potato isolate and one tomato isolate in field plots of the susceptible potato cv. Bintje clearly demonstrated the higher competitive fitness of the potato isolate on its host of origin. Therefore, differential pathogenicity to potato and tomato certainly contributes to the differentiation between P. infestans populations present on potato and tomato in France; however, additional factors, possibly related to survival ability or random genetic drift, are probably also involved and may explain the persistence of weakly pathogenic isolates in these populations.  相似文献

BACKGROUND: The diamondback moth (DBM), Plutella xylostella (L.), is a cosmopolitan pest of cruciferous crops. Fufenozide, a novel non‐steroidal ecdysone agonist, exhibits good efficacy and plays an increasingly important role in the control of Lepidopterous pests in China. A laboratory strain of DBM was selected for resistance to fufenozide, and the genetic basis of resistance was studied. RESULTS: The resistant strain, selected under laboratory conditions, exhibited a higher level of resistance to fufenozide (302.8‐fold based on LC50s) than the laboratory susceptible strain. Mortality data from the testing of F1 progeny of reciprocal crosses of resistant and susceptible DBM indicated that resistance was autosomal and incompletely recessive with a degree of dominance of ?0.664. Chi‐square analysis from responses of a backcross of crossed F1 progeny and the resistant strain and F2 progeny were highly significant, suggesting that the resistance was probably controlled by more than one gene. The estimated realised heritability (h2) of fufenozide resistance was 0.08, indicating that diamondback moth may have a lower chance of developing resistance to fufenozide than other kinds of insecticide. CONCLUSION: The resistance of DBM to fufenozide might be autosomal and incompletely recessive, and the resistance is probably controlled by more than one gene. These results provide the basic information for pest management programmes. Copyright © 2009 Society of Chemical Industry  相似文献

Mefenoxam is one of the most commonly used fungicides for managing diseases caused by Phytophthora spp. on ornamentals. The objectives of this study were to determine whether Phytophthora nicotianae, a destructive pathogen of numerous herbaceous annual and perennial plant species in nurseries, has developed resistance to mefenoxam, and to evaluate the fitness of mefenoxam‐resistant isolates. Ninety‐five isolates of P. nicotianae were screened for sensitivity to mefenoxam on 20% clarified V8 agar at 100 a.i. µg mL?1. Twenty‐five isolates were highly resistant to this compound with EC50 values ranging from 235·2 to 466·3 µg mL?1 and four were intermediately resistant with EC50 values ranging from 1·6 to 2·9 µg mL?1. Sixty‐six isolates were sensitive with EC50 values less than 0·04 µg mL?1. Nine resistant and seven sensitive isolates were tested for mefenoxam sensitivity on Pelargonium × hortorum cv. White Orbit. Mefenoxam provided good protection of pelargonium seedlings from colonization by sensitive isolates, but not by any resistant isolates. Four resistant and four sensitive isolates were compared for fitness components and their relative competitive ability on Lupinus Russell Hybrids in the absence of mefenoxam. Resistant isolates outcompeted sensitive ones within 3 to 6 sporulation cycles on lupin seedlings, regardless of their initial proportions in mixed zoospore inoculum. Resistant isolates exhibited greater infection rate and higher sporulation ability than sensitive ones when they were applied separately onto lupins. These results suggest that fungicide resistance may pose a serious challenge to the continued effectiveness of mefenoxam as a control option for nursery growers.  相似文献

Baseline sensitivity to flumorph, a carboxylic acid amide (CAA) fungicide used to control some oomycetes, was examined using 83 Phytophthora capsici isolates, resulting in a unimodal distribution of effective concentration for 50% inhibition of mycelial growth ranging from 0·716 to 1·363, with a mean of 1·033 ± 0·129 μg mL?1. To assess the potential risk of developing flumorph resistance, 13 flumorph‐resistant mutants of P. capsici were obtained using ultraviolet irradiation. Most of these mutants and their progeny had high levels of fitness, including mycelial growth, sporulation and virulence. The resistance to flumorph changed slightly, either increasing or decreasing, after 10 transfers on agar media. Cross‐resistance was found between flumorph and other CAA fungicides (dimethomorph and iprovalicarb), but not between flumorph and non‐CAA fungicides (cymoxanil, metalaxyl, azoxystrobin and cyazofamid). To investigate the genetics of the flumorph resistance, 619 progeny were obtained by self‐crossing and sexual hybridization. Segregation of sensitivity to fungicide was measured as a ratio of sensitive (S) to resistant (R) isolates. Segregation of the progeny, from self‐crossed isolate PCAS1 (flumorph resistant), was 1:15 in the first generation; and 0:1 or 1:15 in the second generation. In sexual hybridization, segregation of progeny was 0:1 and 1:7 for R × R hybridization; and 1:3 for R × S hybridization. Therefore, the resistance of P. capsici against flumorph was controlled by two dominant genes.  相似文献

Wildtype strains of Fusarium circinatum, the causal agent of pitch canker, were crossed to obtain an F1 generation. Progeny of this cross were tested for virulence by inoculating Pinus radiata seedlings, and were found to induce a wide range of lesion lengths. Two strains from the F1 generation that induced long lesions (= high virulence) were used as parents to produce an F2 generation, followed by a second round of selection for high virulence to obtain an F3 generation. Mean lesion lengths were not significantly different between the three generations ( 0.196). A parallel set of crosses was performed to select for low virulence by using progeny in the F1 and F2 generations that induced short lesions as parents for F2 and F3 generations, respectively. In this case, both rounds of selection resulted in a significant reduction in mean lesion length, from 33.8 ± 0.8 mm in the F1 generation, to 19.7 ± 0.7 and 12.9 ± 0.7 mm in the F2 and F3 generations, respectively. Thus it is apparent that F. circinatum retains the genetic capacity for avirulence to pines, which could reflect a lack of strong selection for virulence in nature. Progeny of a cross between high and low virulence parents manifested nearly continuous variation in lesion lengths, consistent with virulence being a quantitatively inherited trait. Based on this cross, broad‐sense heritability (H2) was determined to be 0.74, which suggests that virulence is under strong genetic control.  相似文献

A laboratory assay was designed to determine the insecticidal efficacy of Beauveria bassiana (Balsamo) Vuillemin (Hyphomycetes: Moniliales) and diatomaceous earth (Diafil 610) against Rhyzopertha dominica (F.) (Coleoptera: Bostrychidae). The fungus B. bassiana was applied at 2.23 × 107, 2.23 × 108 and 2.23 × 109 conidia kg−1 of wheat individually as well as mixed with 200 and 400 ppm of Diafil 610. The conditions for the trials were 30 ± 2oC with 55% r.h. and the counts for mortality were made after 8, 16 and 24 d. All the dead adults were removed after each count and the vials were kept for the next 60 d to assess the emergence of the F1 generation. The findings from these studies proved that the extended exposure interval and the highest combined dose rate of the entomopathogenic fungus and the diatomaceous earth gave the maximum mortality of the beetles. The emergence of the progeny was also highly suppressed where the maximum dose rate of the synergized treatments was applied. The rate of mycosis and sporulation in the cadavers of R. dominica was maximum where the low dose rates of B. bassiana were applied.  相似文献

The pathogenic type (form and race) of Fusarium oxysporum, which generates wilt symptoms on tomato, was rapidly identified with a polymerase chain reaction (PCR)-based technique. We compared the partial nucleotide sequences of endo polygalacturonase (pg1) and exo polygalacturonase (pgx4) genes from isolates of F. oxysporum ff. sp. lycopersici (FOL) and radicis-lycopersici (FORL) from Japan and designed specific primer sets (uni, sp13, sp23, and sprl) based on the nucleotide differences that appeared among the pathogenic types. PCR with the uni primer set amplified a 670∼672-bp fragment from all isolates of FOL and FORL. With the sp13 primer set, an amplicon of 445 bp was obtained only from isolates of FOL race 1 and 3. With the sp23 primer set, a 518-bp fragment was obtained from isolates of FOL race 2 and 3. The sprl primer set yielded a 947-bp fragment from isolates of FORL, but not from FOL. A combination of amplifications with these primer sets effectively differentiated the pathogenic types of F. oxysporum in tomato.  相似文献

Three sexual crosses involving isolates of P. nicotianae with differing sensitivity to mefenoxam were established to study the inheritance of mefenoxam resistance. Mefenoxam sensitivity was determined by measuring the mycelial growth on both mefenoxam-amended clarified V8 agar and non-amended agar and then calculating the relative growth. When both parents had the same phenotype (both were resistant or both were sensitive), all F 1 progeny had the parental phenotype and no segregation for a major effect gene was observed in sensitivity to mefenoxam. However, variations in the mycelial growth of progeny indicated the segregation of minor-effect genes. When the cross involving the mefenoxam-resistant isolate 3A4 and a sensitive parent, the F 1 progeny segregated for mefenoxam resistance in a ratio of 1:1 (resistant: sensitive), indicating that the mefenoxam resistance is controled by a single dominant gene. Mating type was not linked to the mefenoxam-resistance gene locus. One RAPD marker linked in trans to the MEX locus was obtained by bulked segregant analysis using RAPD markers and was converted to a sequence characterized amplified region marker (SCAR). The SCAR maker identified in this study is a limited but useful tool for differentiating homozygous resistant isolates from sensitive isolates of P. nicotianae.  相似文献

Anisopteromalus calandrae (Howard) (Hymenoptera: Pteromalidae) is an important parasitoid of stored-grain insect pests. Partial cDNA sequences of an esterase-like enzyme have been obtained from a malathion-resistant (R) strain and a susceptible (S) strain of this wasp. A single-base substitution in the R strain has been confirmed by using PCR amplification of specific allele (PASA) to amplify genomic DNA extracted from individual resistant and susceptible parents, F1 hybrids from double reciprocal crosses, and progeny from backcrosses. The R allele appeared to be inherited in a strict Mendelian fashion in both diploid female and haploid male progeny. The esterase fragment co-segregated with resistance in these crosses and backcrosses. Female wasps in a mixed population of A calandrae that survived a malathion screen carried the R allele for the esterase-like enzyme, while those wasps that died did not have the R allele. The single base-pair mutation, guanine in the R strain and thymine in the S strain, presumably results in a tryptophan-to-glycine amino acid substitution in the encoded protein. We do not know how these amino acid substitutions may relate to functional differences in the enzyme. However, this esterase gene or another linked esterase gene may encode the resistance-associated malathion detoxifying activity in the R strain. © 1999 Society of Chemical Industry  相似文献

Phytophthora capsici causes root, crown, and fruit rot of vegetable and tropical hosts. Cucumber, zucchini, tomato, and pepper fruits were inoculated using 6-mm-diameter agar plugs of P. capsici, incubated in clear plastic boxes at room temperature (25 ± 2°C and 100% relative humidity), and virulence was estimated by measuring the lesion diameter, pathogen growth diameter, and pathogen sporulation density three (cucumber, zucchini) or four (tomato, pepper) days later. When isolates were grouped by genetic cluster, significant differences in virulence were observed on cucumber and zucchini, with isolates belonging to genetic cluster five causing larger lesions than isolates from genetic cluster six. On tomato, no significant differences were observed for isolates grouped by genetic cluster, but isolates from vegetable crops were generally more virulent than isolates from tropical hosts. Isolates from fabaceous hosts sporulated better on cucumber fruits than isolates from solanaceous hosts. Isolates from vegetable hosts sporulated better on zucchini than isolates from tropical hosts. No significant differences in lesion diameter were noted on pepper when isolates were grouped by host family of origin or genetic cluster, but differences in pathogen sporulation were apparent by host family. Our findings suggest that isolate characteristics such as host family of origin and genetic cluster membership may be used to guide initial isolate selection for cucurbit fruit resistance screening. Final isolate selection should incorporate the phenotypic and genetic diversity of P. capsici, including isolates with differing virulence to the host organ of interest.  相似文献

Screening of 51 promising safflower germplasm lines in Fusarium wilt-infested plots resulted in identification of highly wilt-resistant selections viz., 86-93-36A, 237550, VI-92-4-2 and II-13-2A, with some moderate resistance in HUS-305. Progenies from crosses made using these resistant lines were tested for their reaction to wilt. F1 progenies from 86-93-36A × 237550 and 86-93-36A × II-13-2A recorded zero wilt incidence, while 237550 × 86-93-36A was highly resistant to the Rajendranagar geographical isolate. The reaction for the three progenies showed stability for wilt resistance with no segregation until the F7 generation. Geographical isolates of Fusarium oxysporum f. sp. carthami (Foc) were collected from different safflower growing regions and tested for their pathogenic variability on six host differentials under glasshouse conditions. Based on the reaction of the differentials, the Foc isolates were grouped into four biotypes. The three resistant progenies were tested for their reaction to the four biotypes. The progeny of cross 86-93-36A × 237550 showed an immune reaction to all the biotypes, except for a highly resistant reaction to biotype 3. The progenies of the two other crosses (86-93-36A × II-13-2A and 237550 × 86-93-36A) exhibited immune reactions to biotypes 2,3 and 1,3, respectively, and were highly to moderately resistant to biotypes 1,4 and 2,4, respectively.  相似文献
