Antibiotic use not only selects for resistance in pathogenic bacteria, but also in the commensal flora of exposed individuals. Veterinary surgeons regularly prescribe antibiotics for food animals to treat bacterial infections just as doctors do for human patients. In addition, however, animal feeds contain added antibiotics not for therapy but for economic reasons: to enhance the growth rate of these animals. Several of the antibiotics used as growth promoters are analogues of and fully cross resistant with important antibiotics used in human medicine. As a result of this high exposure to antibiotics, the prevalence of resistant bacteria in the faecal flora of these animals is high. These resistant bacteria can be directly and indirectly, via foods of animal origin, transferred to humans and either colonize the human intestinal tract or exchange their resistance genes with commensal bacteria of humans. As the intestinal flora functions as a reservoir of resistance genes for pathogenic bacteria and because many bacterial species of the intestinal flora are potential pathogens, the efficacy of antibiotic therapy in human medicine may be jeopardized.  相似文献

Risks for the consumer regarding the acquisition of resistant bacteria and/or resistance genes via the consumption of pork are discussed. In general, Salmonella spp. and Escherichia coli that originate from animals do not easily transfer their resistance genes to the resident intestinal flora of humans. The prevalence of resistant E. coli in humans seems more associated with being a vegetarian (odds ratio (OR) 1.89) than with the consumption of meat and meat products. Other risk factors are treatment with antimicrobials (OR 2-5), becoming hospitalized (OR 5.93), or working in a health setting (OR 4.38). In the Netherlands, annually an estimated 45,000 people (0-150,000) become a carrier of resistant E. coli and/or resistance genes that ori ginate from pigs, while an estimated 345,000 persons (175,000-600,000) become a carrier of resistant E. coli and/or resistance genes that originate from hospitals, e.g. other patients. Any problems with resistant Salmonella spp. that stem from pigs are, in fact, an integral part of the total problem of food-borne salmonellosis. Sometimes there are outbreaks of a specific multi-resistant clone of S. typhimurium that causes problems in both farm animals and humans. The probability that in the next 30 years there is no or maximally one outbreak of a specific clone that originates from pig herds is estimated at about 75%. Antimicrobials used as a growth promoter can have a measurable influence on the prevalence of resistant bacteria. The likely chain of events regarding avoparcin and the selection and dissemination of resistance against vancomycin in the enterococci gives the impression that the impact of the use of antimicrobials in animals on the prevalence of resistance in humans is largely determined by whether resistance genes are, or become, located on a self-transferable transposon. Furthermore, consumer health risks of antimicrobials used in slaughter pigs are mainly determined by the selection and dissemination of bacterial resistance and much less by the toxicological properties of any residues in pork. It is also concluded that most of the problems with resistant bacteria in humans are associated with the medical use of antimicrobials, and that the impact of particularly the veterinary use of antimicrobials is limited. However, the impact of antimicrobials used as a feed additive appears to be much greater than that of antimicrobials used for strictly veterinary purposes. The use of antimicrobials as a feed additive should therefore be seriously reconsidered.  相似文献

The term growth promotion applies to the increase in performance or productivity achieved in food producing animals following the addition to their diet of feed antibiotics or growth promoters. Numerous methods are available for the different animal species. Steroidal substances are widely used in cattle whilst in pigs and poultry a wide range of substances are used to influence the gastro-intestinal microflora and thereby bring about a cost-effective improvement in productivity. Although certain antibiotics have been used for growth promotion for over three decades they are still highly effective and appear not to have reduced the value of other widely used antibiotics for the treatment of disease. The mechanisms by which growth promotion is produced are only poorly understood but it is now generally accepted that any improvement in performance is directly related to a variety of direct effects on the gut microbial flora and associated indirect effects on intestinal tissues. The direct effects include inhibition of bacterial growth, interference with bacterial cell wall development, induction of filament formation and interference with the metabolism of intestinal bacteria. The indirect effects include a reduction in the thickness of the intestinal mucosal layer and a decrease in the production of certain mucosal cell enzymes. Other effects seen in the gut of growth promoted animals and birds include a decrease in the prevalence of antibiotic resistance plasmids and a reduced frequency of plasmid transfer in enteric coliforms. It is clear from available data that conventional animals are depressed in performance and this depression can be alleviated by the administration of growth promoting agents in the diet of these animals. The lifting of this depression results in an improvement in performance especially daily live weight gain and feed conversion efficiency.  相似文献

The normal microflora colonizing the digestive tract in livestock species at five to six days after birth can be summarized as following: 400 to 500 different bacterial strains for a total count of 10(14) bacteriae. The role of the gut flora is multifarious: fermentation of feed compounds, induction of anatomical and physiological changes in the intestinal cell wall structure, increase in animal resistance against enteropathogenic bacteriae, etc. In the present review, the numerous hypotheses for explaining the beneficial effects of probiotics on zootechnical performances and also the healthy aspects for men and animals are summarized, and the possible mechanisms of bacterial attachment are enumerated. The different assumptions on mode of action and fixation mechanisms of probiotics in the gut are critically discussed, and the required characteristics of the ideal probiotic are listed. Finally, some zootechnical results obtained with lactic acid bacteria are shown for calves, piglets and broilers.  相似文献

Two different groups of bacteria carrying genes encoding for resistance to antibiotics may be transmitted from animals to humans via food products: a.) obligate infectious agents (enteric pathogens, e.g. Salmonella enterica spp., Campylobacter spp., EHEC) and b) facultative pathogenic species (e.g. E. coli, enterococci). Thus far, it is unknown whether genes encoding for resistance to antibiotics from these bacteria may be transferred to bacteria in normal flora of the host. The transfers of genes encoding for resistance to vancomycin from animal sources to the mucosa of humans has been suggested. Thus, there is a threat that these plasmid-encoded resistance genes may also be transferred to other gram-positive organisms present in the human flora. Vancomycin is the antibiotic in reserve for treatment of infections caused by oxacillin (methicillin) resistant strains of S. aureus and by strains of pneumococcus resistant to penicillin.  相似文献

Resistant bacteria from food-producing animals may compromise the success of antibiotic treatment in animals and in humans. Therefore, the level of antibiotic resistance in bacteria from farm animals and its development over time needs to be monitored. In Switzerland, a monitoring program for antibiotic resistance is currently being developed. Pilot-monitoring programs were conducted in selected animal species in order to obtain current data on antibiotic resistance. The data on the prevalence of bacteria and antibiotic resistance in poultry were used to optimize the sampling plan. The influence of sampling more farms compared to sampling more animals per farm on the prevalence estimate for antibiotic resistance was analyzed by a Monte Carlo simulation model. Accounting for the costs for sample collection, transportation and laboratory analysis of the samples, the number of samples to be taken at the respective step in the production line was optimized. Optimization was defined as maximizing the precision of the prevalence estimate while minimizing the costs. The model will be expanded to other bacterial and animal species in the future.  相似文献

Public pressure to reduce or eliminate antimicrobials as ingredients of feed for poultry and other agricultural animals is mounting, primarily due to the fear of multidrug-resistant bacteria in clinical infections in both animals and humans. Exploration of the occurrence of antibiotic resistance in the gut flora of wildlife avian flocks that presumptively do not receive antimicrobials will determine the rate of resistance in a na?ve population. Fecal samples collected from a healthy population of the yellow-headed blackbirds (YHB) (Xanthocephalus xanthocephalus) in North Dakota were cultured to determine what genera and species of gram-negative facultative anaerobic bacteria these wild birds carry in their intestinal flora and to evaluate the antimicrobial susceptibility profiles. Isolates of Escherichia coli were further characterized for the presence of putative virulence factors and for pathogenic potential using the chicken embryo lethality assay (ELA). The ELA was performed in chicken embryos with challenges at both 12 days and 16 days of incubation to determine whether the 16-day-old embryos were better able to fight the infection and subsequent disease and also to determine whether the ELA could distinguish between primary and secondary avian Escherichia coli pathogens. After screening 33 isolates from the 21 fecal samples, only two E. coli isolates were identified. The predominant genus and species of bacterium identified was Pantoea agglomerans. Collectively, 12 of the 33 isolates (36%) exhibited no resistance to any antimicrobial tested. However, several multidrug-resistant isolates of varying genera were identified. Among the antimicrobial resistances observed, the most common was to ampicillin (60%), followed by cephalothin (33%). Neither E. coli isolate belonged to serogroups that are notorious for causing major outbreaks of colibacillosis in poultry, and only one E. coli isolate retained resistance to any antibiotics; nevertheless, the ELA results indicate that at least one of these E. coli may be a primary pathogen of chickens. This study demonstrates that antibiotic resistance occurs in the gut flora of natural populations of YHB despite the absence of antibiotic pressure. In addition, these results indicate that YHB will harbor E. coli isolates that are potentially pathogenic in poultry. However, these E. coli isolates are not a significant reservoir for multiple antibiotic resistances nor are they widespread in the population of YHB surveyed in North Dakota.  相似文献

The aim of this study was to investigate the effects of two commercially available probiotics, alone and in combination with an antibiotic, on the caecal flora of Japanese quail (Coturnix coturnix japonica) reared under unstressed conditions. Thirty-four 90-day-old Japanese quail were selected for this study. The birds were divided into four groups, two groups of nine birds and two groups of eight birds. The animals in these groups were given feed containing 0.5 kg per tonne of probiotics alone (Protexin or Biosacc), a mixture of probiotic plus antibiotic (Biosacc plus Zinc Bacitracin at 0.5 and 0.0525 ppm, respectively), and a group where no supplement was added to be used as controls. The total count of aerobic bacteria, lactobacilli, enterobacteriaceae, coliforms, enteroccoci, salmonellae, sulphite-reducing anaerobic bacteria (clostridia), and pH values in the caecal content of the birds were examined. No significant differences were detected among the four groups for pH values and bacterial number (p>0.05), except for sulphite-reducing anaerobic bacteria (p<0.001). These results suggest that the use of probiotics alone and/or a mixture of a probiotic plus antibiotic as a feed supplement does not have a major suppressing effect on the majority of bacterial groups in the caecal flora of mature, healthy Japanese quail reared in unstressed conditions.  相似文献

The Health Council of the Netherlands has issued a report on the risk of development of resistance among bacteria as result of the use of antibiotics as growth promotors in livestock farming. The committee appointed by the Health Council conclude that the use of antimicrobial growth promotors contributes to the problem of resistance among human pathogens. The conclusions are based on evidence regarding the development of resistance in livestock as the result of the use of antimicrobial growth promotors, the possibility of colonisation/infection of humans with resistant bacteria from the intestinal flora of productive livestock, and the transfer of resistance genes from livestock bacteria to human pathogenic microorganisms. Effective measures for the limitation of the public health risk should focus on termination of the use of antimicrobial growth promotors that confer resistance to (related) antibiotics currently used (or which will be available) to treat patients suffering from bacterial infections. In addition, the committee advised ending the use of antimicrobial growth promotors in 3 years.  相似文献

Extended-spectrum beta-lactamases (ESBL)-producing Gram-negative bacteria pose a serious threat to Public Health in human medicine as well as increasingly in the veterinary context worldwide. Several studies reported the transmission of zoonotic multidrug resistant bacteria between food-producing animals and humans, whilst the contribution of companion animals to this scenario is rather unknown. Within the last decades a change in the social role of companion animals has taken place, resulting in a very close contact between owners and their pets. As a consequence, humans may obtain antimicrobial resistant bacteria or the corresponding resistance genes not only from food-producing animals but also via close contact to their pets.This may give rise to bacterial infections with limited therapeutic options and an increased risk of treatment failure. As beta-lactams constitute one of the most important groups of antimicrobial agents in veterinary medicine, retaliatory actions in small animal and equine practices are urgently needed. This review addresses the increasing burden of extended-spectrum beta-lactam resistance among Enterobacteriaceae isolated from companion animals. It should emphasize the urgent need for the implementation of antibiotic stewardship as well as surveillance and monitoring programs of multi resistant bacteria in particular in view of new putative infection cycles between humans and their pets.  相似文献

Anaerobic flora plays a key role in preventing intestinal colonization with potential pathogens. Nowadays, the mechanisms involved in the colonization resistance provided by the anaerobic microflora are to be clarified. Numerous factors seem to intervene in the regulation of the intestinal flora.

The purpose of the present study was to correlate the presence or relative absence of Clostridium sp. and Bacteroides with the colonization by C. perfringens, which is involved in lethal infections in an immunologically compromised host.

The intestinal bacterial colonization of 20 newborns delivered by cesarian section was assessed sequentially over the first 14 days of life. C. perifringens is a strongly reducing microorganism and undoubtedly causes a decrease in the oxidoreduction potential of the newborn feces favouring the subsequent colonization by other putrefactive bacteria. C. perfringens seems to be the precursor for installation of putrefactive bacteria, as Bacteroides and other Clostridium sp.  相似文献

The early curative uses of antimicrobial drugs such as fluoroquinolones before the onset of symptoms in veterinary medicine may be regarded as irrational antibiotic consumption. However, it should be stressed that in early curative antimicrobial treatment as in metaphylaxis, the bacterial burden at the infection site is often very low, and so the rapid eradication of the bacterial population could result. We investigated the impact of early versus later curative administrations of 1 or 40 mg/kg of marbofloxacin on the survival of mice, the eradication of the targeted pathogen and the selection of resistant bacteria in a mouse lung infection with Pasteurella multocida. In this model, for a given marbofloxacin dose, the clinical and bacteriological outcomes were better, and the selection of resistance less frequent, for the early rather than for the late treatment. Moreover, the early administration of 1mg/kg led to better clinical and similar bacteriological (eradication and selection of resistance) outcomes than the late administration of 40 mg/kg marbofloxacin. Our results suggest that the optimal doses for the animals' cure could be lower when administered early during the time course of the infection than when administered after the disease outbreak. As the main argument against early treatments such as metaphylaxis is the possible enhancement of resistance at the gut level, further studies should assess if lower doses of antibiotic administered to all the animals of a herd could have less impact on the commensal digestive flora than higher doses only administered to animals showing clinical symptoms.  相似文献

Turtoises are a great puzzle when it comes to their bacterial flora, the composition and structure of which are still unknown in details. Its component which has been best described so far is Salmonella spp., presumably due to the threat of reptile-associated salmonellosis in humans. This investigation tried to assess and characterize intestinal bacterial flora of imported tortoises found dead during quarantine. Most of the animals carried various serovars of Salmonella showing no antimicrobial resistance. Presence of multiresistant Escherichia coli was possibly a result of industrial breeding and high usage of antimicrobials. Thirteen bacterial species or genera like Citrobacter spp., Morganella spp., Pseudomonas spp. were identified. Their commensal character is assumed, although pathogenic potential might be verified. The results indicate global tortoise trade as a source of common and exotic bacteria or antimicrobial resistance mechanisms in new geographical areas. These dangers indicate the need for a systematic survey of exotic pets and establishment of legal requirements for reptile health conditions on breeding, trade premises and in households with such pets.  相似文献

Historically, the use of antibiotics was not well regulated in veterinary medicine. The emergence of antibiotic resistance (ABR) in pathogenic bacteria in human and veterinary medicine has driven the need for greater antibiotic stewardship. The preservation of certain antibiotic classes for use exclusively in humans, especially in cases of multidrug resistance, has highlighted the need for veterinarians to reduce its use and redefine dosage regimens of antibiotics to ensure efficacy and guard against the development of ABR pathogens. The minimum inhibitory concentration (MIC), the lowest concentration of an antibiotic drug that will prevent the growth of a bacterium, is recognised as a method to assist in antibiotic dosage determination. Minimum inhibitory concentrations sometimes fail to deal with first-step mutants in bacterial populations; therefore dosing regimens based solely on MIC can lead to the development of ABR. The mutant prevention concentration (MPC) is the minimum inhibitory antibiotic concentration of the most resistant first-step mutant. Mutant prevention concentration determination as a complementary and sometimes preferable alternative to MIC determination for veterinarians when managing bacterial pathogens. The results of this study focused on livestock pathogens and antibiotics used to treat them, which had a MIC value of 0.25 µg/mL for enrofloxacin against all 27 isolates of Salmonella typhimurium. The MPC values were 0.50 µg/mL, with the exception of five isolates that had MPC values of 4.00 µg/mL. The MPC test yielded 65.52% (18 isolates) Salmonella isolates with florfenicol MICs in the sensitive range, while 11 isolates were in the resistant range. Seventeen isolates (58.62%) of Pasteurella multocida had MIC values in the susceptible range and 41.38% (12 isolates) had an intermediate MIC value. Mutant prevention concentration determinations as done in this study is effective for the antibiotic treatment of bacterial infections and minimising the development of resistance. The MPC method can be used to better control to prevent the development of antibiotic drug resistance used in animals.  相似文献

Giving medical feed to weanling pigs it is possible to produce a vitamin K-deficiency as a side effect, which is caused by destruction of the intestinal flora. In this investigation the effect of different drugs in a vitamin K-deficient diet on blood coagulation of weanling pigs was examined. In a first trial clinical symptoms of a vitamin K-dependent coagulation disorder could be seen in five from six animals after feeding a combination of sulfadimidine, tylosin, furazolidone and arsanilic acid. Animals showed haemorrhages, when they were housed on flatdecks as well as on concrete floor. This indicated that coprophagy plays no role in pigs for supply of vitamin K. Feeding these drugs in other combinations or one of the drugs alone caused no clinical signs in a second trial. Significant differences to untreated control pigs were found only in one group (sulfadimidine/arsanilic acid) for activities of coagulation factors. Other groups demonstrated only prolongation of clotting times in single animals. Pigs, which received sulfadimidine within their ration, were more often affected than other animals. In a third trial suckling pigs were treated over a period of three weeks with a drug combination used in the first trial to allow an adaptation of the intestinal flora. Same treatment after weaning was unable to produce any clinical symptoms, but led to distinct coagulation disorders in pigs treated before as well as in untreated animals. It is supposed that the development of a vitamin K-deficiency depends on faecal bacterial count and on the specific resistance of intestinal flora in individuals.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
