The ontogenesis of main pancreatic and intestinal enzymes was investigated in the recent promising Mediterranean candidate species of meagre, Argyrosomus regius, during larval development until 40 days after hatching (DAH). The green-water technique was carried out for larval rearing. Whole-body homogenates were used for enzymatic analysis in larvae younger than 15 DAH; after this date, older larvae were dissected into two segments as pancreatic and intestinal segment. Trypsin was detected as early as hatching and sharply increased concurrently with age and exogenous feeding 15 DAH, but constant decline was observed until the end of experiment. Amylase was determined at 2 DAH and sharply increased 10 DAH. Then, slight decreases were found between 10 and 15 DAH, and then slow alterations were continued until the end of the experiment. Lipase was firstly measured on day 3; then, sudden decline was observed between 20 and 25 DAH. After this date, slow fluctuations were maintained until the end of the experiment. Pepsin was firstly assayed 15 DAH related to gastric gland secretion and sharply increased 30 DAH. Then, it slowly varied until end of the experiment. Enzymes of brush border membranes, alkaline phosphatase and aminopeptidase N showed similar pattern on specific activities during the first 10 days. Thereafter, while specific activity of alkaline phosphatase slightly decreased 15 and fluctuated until 20 DAH, aminopeptidase N activity slowly increased 20 DAH. Then, activity of alkaline phosphatase and aminopeptidase N constantly increased 30 DAH, indicating maturation of the intestinal digestive process, and also, these activities continued to slowly increase until the end of the experiment. The specific activity of cytosolic peptidase, leucine–alanine peptidase, smoothly increased on day 8, then fluctuated until 15 DAH. After this date, in contrast to enzymes of brush border membranes, it sharply decreased 25 DAH and continued to gradually decline until the end of the experiment. These converse expressions were indicative of a maturation of enterocytes and the transition of an adult mode of digestion.  相似文献

This work describes the ontogeny of the digestive tract in thick lipped grey mullet (Chelon labrosus) larvae reared until day 36 post-hatching with the semi-extensive technology in mesocosms. Diet was constituted by live preys, rotifers, Artemia and wild zooplankton, then compound diet was added from day 20 (p. h.). Linear growth, weight growth and digestive enzymes specific activities were studied during larval ontogeny. Pancreatic enzymes (trypsin and amylase) and intestinal enzymes (leucine-alanine peptidase “Leu-ala”, aminopeptidase N “AN” and alkaline phosphatase “AP”) were assayed in larvae sampled throughout the rearing trial to evaluate gastrointestinal maturation along the development.The trypsin specific activities were very high during the first two weeks and then declined as observed in marine fish species. A following increase in trypsin specific activity from day 20 was attributed notably to ingestion of particle compound diet. In contrast to the pattern generally described in fish larvae, amylase specific activity showed a continuous increase. This could be attributed to the fact that C. labrosus is an omnivorous species and suggests that the fish might be able to use efficiently diets containing higher levels of starch or other carbohydrates since the end of larval development.Relative expression of intestinal brush border membrane enzymes (AP and AN) and cytosolic enzyme (Leu-ala), showed an abrupt increase of both AP/leu-ala and AN/leu-ala ratios at day 8 (p. h.), indicating that maturation of intestinal tract in C. labrosus larvae is particularly precocious. It is assumed that larvae of C. labrosus might support early co-feeding and weaning strategies, which could reasonably be initiated since mouth opening.  相似文献

The major digestive enzymes in Pacific bluefin tuna Thunnus orientalis larvae were characterized, and the physiological characteristics of the enzymes during early ontogeny were clarified using biochemical and molecular approaches. The maximum activity of trypsin (Try), chymotrypsin (Ct) and amylase (Amy) was observed at pH 6–11, 8–11 and 6–9, respectively. Maximum activity of Try, Ct and Amy occurred at 50 °C, that of lipase (Lip) was at 60 °C and that of pepsin (Pep) was at 40–50 °C. These pH and thermal profiles were similar to those for other fish species but differed from those previously reported for adult bluefin tuna. Enzyme activity for all enzymes assayed was found to decrease at high temperatures (Try, Ct, Amy and Pep: 50 °C; Lip: 40 °C), which is similar to findings for other fish species with one marked exception—increased Try activity was observed at 40 °C. Lip activity appeared to be dependent on bile salts under our assay conditions, resulting in a significant increase in activity in the presence of bile salts. Ontogenetic changes in pancreatic digestive enzymes showed similar gene expression patterns to those of other fish species, whereas marked temporal increases in enzyme activities were observed at 10–12 days post hatching (dph), coinciding with previously reported timing of the development of the pyloric caeca in bluefin tuna larvae. However, complete development of digestive function was indicated by the high pep gene expression from 19 dph, which contradicts the profile of Pep activity and previously reported development timing of the gastric gland. The development of a simple and effective long-term storage protocol for harvested fresh Undaria pinnatifida is required to increase the market for U. pinnatifida products. We investigated the quality of U. pinnatifida after the application of various freezing and thawing methods to establish an optimum storage protocol. Four freezing temperatures (?40, ?30, ?20, and ?10?°C), four freezing conditions (in air without seawater and with 50, 75, and 100?% seawater), and four thawing times (6, 12, 18, and 24?h) were employed. Changes in color value, tensile strength, total bacterial count, and most probable number of coliforms were measured. The best method for preserving the overall quality of U. pinnatifida is freezing at ?30?°C with 50?% seawater and thawing in running tap water for 6?h. During retail display, changes in color, odor, tensile strength, and bacterial count of U. pinnatifida that had been freeze-thawed under optimum conditions were also measured, and a sensory evaluation was performed. There was no significant difference compared with the control within two?days at 10?°C. In conclusion, we developed a simple and effective long-term storage protocol that led to no significant loss of quality of fresh U. pinnatifida.  相似文献

研究鲢(Hypophthalmichthys molitrix)在冷冻-解冻过程中的品质变化规律。鲢于–18℃条件下冻藏10 d,流水解冻后再贮藏于–18℃条件下,5 d后解冻,共反复冷冻-解冻5次。在解冻1、3、5次后随机取3条鲢测定其解冻损失、蒸煮损失、硬度、咀嚼性、恢复性、硫代巴比妥酸(TBA)值、L*值、b*值、盐溶性蛋白(SSP)含量、Ca2+-ATP酶活性、总巯基含量、表面疏水性和感官值等指标,评价反复冷冻-解冻对鲢品质的影响。结果表明,在第1次冷冻-解冻时,硬度、咀嚼性、恢复性、盐溶性蛋白(SSP)含量和Ca2+-ATP酶活性与鲢初始值相比显著降低(P<0.05),b*值和表面疏水性显著升高(P<0.05);解冻损失、L*值和TBA值在第3次冷冻-解冻时显著上升(P<0.05);总巯基含量在第5次冷冻-解冻时显著下降(P<0.05);而蒸煮损失在冷冻-解冻过程中没有显著变化(P>0.05);感官值在第5次冷冻-解冻时为47,感官品质已不可接受。各指标变化表明,鲢品质会随着反复冷冻-解冻次数增加而下降,在运输、贮藏和销售过程中要防止温度波动,避免反复冻融对鲢品质造成不利影响。  相似文献

The aim of this study was to investigate the effects of freezing and frozen storage on protein functionality and texture of squid (Loligo vulgaris), octopus (Octopus vulgaris), and cuttlefish (Sepia officinalis) muscles. Squid, octopus, and cuttlefish samples were cut into pieces of 4 × 4 cm. These pieces were packed in polyethylene bags. The bags were frozen in a blast freezer at ?45°C until the thermal center reached ?18°C. Frozen samples were stored in a deep freezer at ?18°C for 30 days. After freezing and during frozen storage, total soluble protein and water holding capacity decreased and total free amino acid and cooking loss increased in all cephalopod muscles. According to instrumental texture analysis results, freezing and frozen storage affected textural characteristics of squid and cuttlefish but not of octopus. Sensory hardness and chewiness values of all cephalopods increased after freezing, but elasticity values did not change. There were no significant differences between storage days in hardness values of squid and octopus. However, significant differences in hardness values of cuttlefish were observed between the 1st day of storage and the last day.  相似文献

The maturation of the digestive functions in sea bass (Dicentrarchus labrax) larvae was evaluated by the enzymatic profile of pancreas and intestine brush border membranes. Sea bass larvae were weaned at day 25 with three simplified diets different by their protein nature: 100% fish meal (FP), 100% casein mixture (CP) and 50% fish meal-50% casein mixture (CFP). The casein mixture contained 35% of hydrolysate. The control group was fed live preys. The specific activity of amylase decreased with age irrespectively of the diets whereas the specific activity of trypsin was enhanced. The casein mixture reduced pancreatic secretion in amylase and trypsin. The CFP group differed from the other groups fed on compound diets, exhibiting as soon as day 32 high activities of brush border enzymes, similar to controls. This sharp increase between day 25 and 32 appeared to be crucial for larval survival. The addition of a protein hydrolysate in a weaning diet seems to facilitate this maturation process.  相似文献

This study aimed to evaluate the contribution of protein oxidation to the changes in the water-holding capacity (WHC) and texture of bighead carp (Aristichthys nobilis) fillets under chilled and partial frozen storage (4°C and ?3°C). The results indicated that less protein oxidation occurred to fillets at ?3°C than at 4°C, which was reflected by the higher value of salt-soluble protein contents (SSP), total sulfhydryl content (SH), Ca2+-ATPase activity, lower water-soluble protein contents (WSP), total disulfide content (SS), and surface hydrophobicity (So-ANS). However, the fillets had better WHC and texture at 4°C, as well as lower drip loss and higher centrifugal loss, hardness, and springiness. A significant linear relationship existed between protein oxidation parameters with WHC and texture characteristics for fillets under both types of storage, but the process of freezing and then thawing, instead of protein oxidation, was the main factor affecting the texture and WHC of fillets at ?3°C.  相似文献

The activities of some digestive enzymes were investigated in Eurasian perch Perca fluviatilis larvae fed live preys during the first month of life. Activities of pancreatic and intestinal enzymes displayed a pattern characteristic of developing animals. Pancreatic enzymes were efficient at the time of hatching. Trypsin and amylase specific activities increased during the first developmental days, then decreased even though segmental activities increased continuously as the larvae grew. The increase of chymotrypsin specific activity was proportional to fish growth. From day 21 after hatching to the end of experiment, the strong increase in the activities of alkaline phosphatase, leucine-aminopeptidase N, maltase and γ-glutamyl transpeptidase reflected the development of brush border membranes of enterocytes. This phenomenon occurred concurrently with a decrease in a cytosolic enzyme, the leucine-alanine peptidase. These opposite patterns indicate a maturation of enterocytes and the acquisition of an adult mode of digestion within the first month of live. Enzyme activity was evaluated in the intestine of juvenile pacu, Piaractus mesopotamicus, fed diets containing 0, 10 or 20 % of lyophilized bovine colostrum (LBC) inclusion for either 30 or 60 days. The enzymes intestinal acid and alkaline phosphatase (ACP and ALP, respectively), nonspecific esterase (NSE), lipase (LIP), dipeptidyl aminopeptidase IV (DAP IV) and leucine aminopeptidase (LAP) were studied using histochemistry in four intestinal segments (S1, S2, S3 and rectum). Moderate activity of the DAP IV was detected in the three last intestinal segments, but no differences among the treatments were detected. Enzymes LAP, NSE and LIP were weakly stained in all intestinal segments and the inclusion of 10 or 20 % of LBC in the diet commanded a moderate reaction to NSE in the S3 segment at day 60. ACP activity was detected only in the brush border of the S1 segment of fish fed 0 % LBC for either 30 or 60 days. The activity of ALP was very strong in the first intestinal segment, but a weak reaction was seen in the last segments. The inclusion of 20 % of LBC changed the pattern of staining to the ALP, eliciting moderate staining in S2 at day 30 and S1 at day 60. The consumption of diets containing LBC by juvenile pacu did not have significant implications in intestinal enzymatic activity, which still was not fully stimulated.  相似文献

The leopard grouper is an endemic species of the Mexican Pacific with an important commercial fishery and good aquaculture potential. In order to assess the digestive capacity of this species during the larval period and aid in the formulation of adequate weaning diets, this study aimed to characterize the ontogeny of digestive enzymes during development of the digestive system. Digestive enzymes trypsin, chymotrypsin, acid protease, leucine–alanine peptidase, alkaline phosphatase, aminopeptidase N, lipase, amylase and maltase were quantified in larvae fed live prey and weaned onto a formulated microdiet at 31 days after hatching (DAH) and compared with fasting larvae. Enzyme activity for trypsin, lipase and amylase were detected before the opening of the mouth and the onset of exogenous feeding, indicating a precocious development of the digestive system that has been described in many fish species. The intracellular enzyme activity of leucine–alanine peptidase was high during the first days of development, with a tendency to decrease as larvae developed, reaching undetectable levels at the end of the experimental period. In contrast, activities of enzymes located in the intestinal brush border (i.e., aminopeptidase and alkaline phosphatase) were low at the start of exogenous feeding but progressively increased with larval development, indicating the gradual maturation of the digestive system. Based on our results, we conclude that leopard grouper larvae possess a functional digestive system at hatching and before the onset of exogenous feeding. The significant increase in the activity of trypsin, lipase, amylase and acid protease between 30 and 40 DAH suggests that larvae of this species can be successfully weaned onto microdiets during this period.  相似文献

As amino acids (AAs) are vital molecules in the metabolism of all living organisms and are the building blocks of enzymes, a 6-week feeding trial was conducted for determining the influence of dietary essential amino acid (EAA) deficiencies on pancreatic, plasma, and hepatic enzyme activities in silvery-black porgy (initial weight 4.7?±?0.01 g) juveniles. Eleven isoproteic (ca. 47%) and isoenergetic (ca. 20.5 MJ kg?1) diets were formulated including a control diet, in which 60% of dietary nitrogen were provided by intact protein (fish meal, gelatin, and wheat meal) and 40% by crystalline AA. The other 10 diets were formulated by 40% reduction in each EAA from the control diet. At the end of the experiment, fish fed with threonine-deficient diet showed the lowest survival rate (P?<?0.05), whereas growth performance decreased in fish fed all EAA-deficient diets, although the reduction in body growth varied depending on the EAA considered. Pancreatic enzymes (trypsin, lipase, α-amylase, and carboxypeptidase A) activities significantly decreased in fish fed the EAA-deficient diets in comparison with fish fed the control diet (P?<?0.05). Fish fed with the arginine-deficient diet had the highest plasma and liver alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase levels (P?<?0.05). Plasma and liver lactate dehydrogenase and superoxide dismutase showed the highest and lowest values, respectively, in fish fed the arginine and lysine-deficient diets (P?<?0.05). Plasma metabolites were significantly affected by dietary EAA deficiencies (P?<?0.05). The results of this study suggesting dietary EAA deficiencies led to reduction in growth performance as well as pancreatic and liver malfunction. Furthermore, arginine and lysine are the most limited EAA for digestive enzyme activities and liver health in silvery-black porgy.  相似文献

Quality changes of vacuum-packed Atlantic mackerel (Scomber scombrus) fillets during 12 months’ frozen storage at ?27°C and 9 days’ chilled storage at +4°C were evaluated. Freezing at ?27°C preserved the long chain n-3 polyunsaturated fatty acids (LC n-3 PUFAs), both in light and dark muscle, vitamin D, and the low molecular weight metabolites (LMW) (studied by high resolution nuclear magnetic resonance spectroscopy, HR NMR). Protein oxidation took place, especially between 1 and 7 months, decreasing water holding capacity and protein extractability. During chilled storage, no lipid or protein oxidation was observed, but lipolysis increased, and several LMW metabolites relevant for sensory and nutritional quality degraded into non-favorable compounds. The content of biogenic amines was high at day 9 (e.g., 18 mg histamine/100 g), jeopardizing safety. Preservation of mackerel fillets by freezing at ?27°C is thus a better option compared to prolonged chilled storage at +4°C; the quality was well preserved for 12 months’ frozen storage.  相似文献

Abstract

Hoki were trawl-caughl under commercial conditions and subjected to one of five treatments before heading and gutting and freezing to ?35°C. Fish were either processed within 1 hour of capture, held at ambient temperatures or in ice until rigor was established or held at ambient temperatures or in ice until rigor was resolved. Fish from each treatment were then stored at ?20°C for periods of up to 56 weeks and tested for gaping, pH and other related physical and sensory properties. Fish frozen post-rigor showed the most gaping and fish frozen pre-rigor had less than those frozen in rigor. The temperature of storage before freezing had little effect on gaping other than extending the time taken to proceed through rigor. Extended time in frozen storage resulted in decreased water-holding capacity but had little effect on gaping. Gaping was not related to pH levels and did not affect the texfural properties of the cooked fish.  相似文献

This study examined the microscopic structure of the small intestine and digestion and absorption in the hybrid tilapia, Oreochromis niloticus×Oreochromis aureus. The small intestine was lined on the luminal face with villi, which were narrower and more dense in the proximal small intestine and became wider and sparser distally. The columnar epithelial cells lining the villi were predominantly enterocytes containing a brush border, interspersed with mucus containing goblet cells approximately every 4–6 cells. These mucus‐secreting cells contained both acid and basic mucins and increased in density in the distal and the terminal sections of the intestine. Proliferation of enterocytes was observed in the intervillus areas and lower third of the villus. Fish were fed diets of either fish meal or fish meal–wheat to which chromic oxide had been added as a non‐absorbed reference standard. Activity of the pancreatic enzymes, lipase, trypsin and amylase were determined along the small intestine relative to the non‐absorbed marker. Activity of these pancreatic enzymes was maximal in the proximal segments and decreased distally. Changes in the activity patterns of trypsin and amylase were observed with the different diets in the proximal small intestine. Net secretion of both N and fatty acids was found in the hepatic loop and absorption commenced distally decreasing in rate with distance from the pylorus. The pattern of N absorption in the proximal small intestine changed with the different diets. The structure and function of the small intestine of the tilapian omnivore thus are generally similar to mammals and digestion and absorption adapt with diet.  相似文献

ABSTRACT

The effects of a short chilled storage period before freezing, frozen storage temperature, and freezing rate on lipid oxidation of bluefin tuna (Thunnus thynnus) meat during frozen storage were investigated. After 12-months storage, all samples had increased in peroxide value though they were less at the lower temperatures (?45 and ?60°C). Peroxide values in all samples stored at ?20°C increased after 3 months storage, particularly those processed and stored 51 h after harvest. The lowest increase in peroxide value occurred in the samples frozen rapidly 3 h after harvest. Vitamin E levels decreased faster during frozen storage at ?20°C. There were no apparent differences in levels of triacylglycerides nor in n-3 fatty acid levels between treatments, storage periods, and storage temperatures. After 12-months storage, headspace oxidative volatiles were highest in samples stored at ?20°C and lowest in those stored at ?60°C. Lipid oxidation in tuna meat stored at ?45°C is similar to that at ?60°C, and rapid freezing rather than slow freezing should be used.  相似文献

Two common live feeds, the Brachionus plicatilis species complex SS-type and L-type were used to assess whether there were any differences in protein hydrolysis and digestive trypsin activity in first feeding Japanese flounder. There were no significant differences in hydrolysis activity at 2, 3 and 7 days after hatching (DAH). At 5 DAH, hydrolysis activity was significantly higher in larvae fed SS-type (p?<?0.05) at 50 kDa in 1.5- and 3-h incubation whereas L-type treatment had not completely hydrolyzed the proteins after 3 h at the same molecular weight. Larvae fed SS-type had significantly higher (p?<?0.05) trypsin activity at 3, 5, 6, 7 DAH. Contribution of live prey to trypsin fraction in larvae showed significantly higher (p?<?0.05) fraction for SS-type at 5 DAH (2.18?±?0.44%) and 6 DAH (2.04?±?0.29%) and the effect of exogenous trypsin from live prey was relatively low when compared to the total trypsin activity in larvae. This study discusses the differences in ability to digest proteins in Japanese flounder when fed different rotifer morphotypes and highlights the adaptability of this species to alternative rotifer morphotypes during its early developmental stages.  相似文献

The present study examined the possibility of long‐term storage, by cryopreservation in liquid nitrogen, of the sperm of filefish (Thamnaconus septentrionalis). Changes in motility, survival rate, ultrastructure and fertilization rate of the sperm after freezing and thawing were tested. For selection of the immobilizing solution, artificial seawater (ASW) of 250, 350 and 450 mOsmol kg?1 were tested. Sperm motility was significantly inhibited in 350 mOsmol kg?1 ASW, and restored entirely after 100% ASW (1200 mOsmol kg?1) was added. Two cryoprotectants, dimethyl sulphoxide and glycerol, were employed. The sperm was diluted at the ratio of 1:6 with the extenders, and frozen at a freezing rate of ?40°C min?1 to ?100°C after equilibration for 10 min at room temperature, followed by plunging into liquid nitrogen. The highest post‐thawed sperm motility and survival rate were obtained with 5% glycerol. Afterwards, the effect of different freezing rates was examined using 5% glycerol as a cryoprotectant, and the rate of ?30°C min?1 to ?100°C showed the best result.  相似文献

The activity of the enzymes located in the digestive tract (pyloric caeca and intestine) in two strains and F1 crosses of the Australian freshwater fish silver perch [Murray River (M) and Cataract Dam (C)] was evaluated. The effect of the fish holding system (cages or ponds) on the activity of alkaline phosphatase (AP) and the brush border enzymes: maltase, leucine amino peptidase (LAP) and γ‐glutamyl transferase (γ‐GT) was examined. The enzymatic activity was tested in three intestinal sections: the pyloric caeca, upper and lower intestine. All the tested enzyme activities were influenced by an interaction between the intestinal section and the strain or cross of silver perch. For fish maintained in ponds, there were two distinct enzymatic activity patterns, one for the pure strains (C × C and M × M) and one for the crosses (C × M and M × C). Significantly higher (P<0.05) enzymatic activities of the crosses compared with the pure strains were particularly noticeable in the pyloric caeca. In fish held in cages, there was a significantly higher (P<0.05) enzymatic activity in the pyloric caeca of the C × C strain. The total enzymatic activities in the fish reared in the ponds were significantly higher (P<0.05) than those in the fish held in cages. The results of the present investigation confirm that the different genetic background of the silver perch in the Murray River and in the Cataract Dam influences the activity of the brush border digestive enzymes. This activity also varies between maintenance systems.  相似文献

The pancreatic digestive enzymes, trypsin, chymotrypsin, lipase and amylase were partially characterized, and changes in their activities were examined during the initial ontogeny of Japanese eel Anguilla japonica larvae from 5 to 34 days post-hatching (dph). The pH optima of the eel larval enzymes were narrower than those other fish species; trypsin activity was highest at pH 9, chymotrypsin and amylase activities were highest at pH 7 and 8, and lipase activity was highest at pH 8 and 9. In an analysis of thermal profiles, the larval pancreatic enzymes had a high optimal temperature and high thermal stability, which are typical of fish from the tropics. At 12 and 13 dph, lipase activity and gene expression levels of trypsin (-a and -b), lipase and amylase decreased markedly, suggesting a marked change in larval metabolism at that time. These data could be useful in the development of artificial larval diets in Japanese eel.  相似文献
