The influence of supplementation of drum-dried flake feed with various levels of Daphnia sp. on the growth and fecundity of broodstock Xiphophorus helleri was evaluated. Weekly supplementation showed no measurable advantage over flake feed only. A direct correlation was demonstrated between fecundity and size of female fish when fed a formulated flake feed only. Fish fed a daily Daphnia supplement had a mean final weight of 5.97 g, feed conversion ratio of 1.33 and mean fecundity of 190 embryos per female, which was significantly higher than the yield from the non-supplemented or weekly supplemented treatments. Despite these differences, the number of young harvested during the experiment did not differ (P > 0.05) between treatments and this discrepancy was ascribed to parental cannibalism of new born juveniles. The importance of long-chain polyunsaturated fatty acids, especially the eicosanoid precursors, is addressed in this paper. It has been generally recognized that eicosapentaenoic (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3) are of significant importance in fish reproduction while arachidonic acid (AA, 20:4n-6) has often been overlooked. The ratio between C20 fatty acids EPA and AA might be important for many physiological functions depending on the species evolution and its requirements. Arctic char (Salvelinus alpinus) has a much more pronounced freshwater history and therefore different fatty acid requirements than the other commonly farmed salmonids such as salmon (Salmo salar), brown trout (Salmo trutta) and rainbow trout (Oncorhynchus mykiss). Therefore there is reason to formulate a feed that is more suitable for farming of this freshwater species. In this study, freshwater wild-origin char eggs were compared to farmed eggs of char. The ratio n-3/n-6 of total phospholipids of eggs was much lower in the wild fish, 3.5 versus 13.5, and the hatching rate of eggs from natural environment was much higher (20–70% vs. >80%). We conclude that feed based on marine raw product does not fulfill the requirements for essential fatty acids for freshwater char and we suggest that AA is supplemented to the broodstock diet and that at least linoleic acid (18:2n-6) is included in the on-growth diet formulas to lower the n-3/n-6 fatty acid ratio.  相似文献

Blue mussel (Mytilus edulis) broodstock collected from the Irish Sea during wintertime (November) was conditioned with three different microalgae diets. Positive flagellates (PF) treatment consisted of Pavlova lutherii, Isochrysis galbana (T-Iso), and Chaetoceros calcitrans (1:1:1). Positive diatoms (PD) treatment consisted of Pavlova lutherii, Chaetoceros calcitrans, and Skeletonema costatum (1:1:1). Broodstock animals in the PF and PD treatments were fed a total of 2.4 × 1011 algae cells per day. Animals in the negative flagellates (NF) treatment received only 1/8th of the total amount of algae of the PF diet. The conditioning diets had an impact on spawning success and broodstock fecundity but not on hatching rate, which was similar in all three groups. The best results were obtained with the PD diet where 84% of the conditioned animals spawned and females released 5.0 × 106 eggs on average. Animals belonging to the PF and NF treatments released, on average, only 3.6 × 106 and 1.6 × 106 eggs, respectively. Although the amounts of algae provided to the broodstock animals had no effect on the hatching rate, the D-larvae resulting from the NF treatment were smaller in size than the larvae from the other treatments. Biochemical analysis of the different broodstock groups at the end of the experiment revealed higher carbohydrate levels in group NF than in PF and PD, supporting the theory that gametogenesis is mainly supported by the energy from the glycogen reserves. As far as we are aware this is the first study describing hatchery broodstock conditioning of blue mussels under fully controlled conditions.  相似文献

The effects of increasing levels of dietaryphospholipids (PL) on the reproductive performance,egg and larval quality, and lipid composition offemales of the freshwater prawn Macrobrachiumrosenbergii were investigated. Three isolipidic dietscontaining similar amounts of highly unsaturated fattyacids but varying levels of PL (0.8, 2.4 and 4.6%)were fed during 180 days to three groups of eightfemales originating from Thai ponds. No significantdifferences were observed for fecundity, egg size andhatchability, starved larvae size, and size, survivaland tolerance to stress of 8 day-old larvae.Similarly, no major differences in the lipidcomposition of the midgut gland, ovaries and muscletissue of females could be detected. Results indicatethe lack of need of dietary supplementation of PL forM. rosenbergii broodstock, as previouslyreported for earlier life stages. It is suggested thatthe basal level of 0.8% dietary PL was sufficient tomeet the dietary demands of the prawn broodstock. ThePL requirements of M. rosenbergii broodstock, ifany, may be satisfied in commercial feeds through theinclusion of ingredients containing some phospholipidsendogenously.  相似文献

鳜是我国名贵的淡水经济鱼类,2019年其养殖产量已达33万t。本文从养殖现状、养殖模式及良种选育等方面介绍鳜的养殖概况。同时,鳜摄食习性独特,终生以活鱼为食。养殖生产中,主要以活饵料鱼直接饲喂,这种以鱼养鱼的方式不仅资源利用率低,对环境和资源的破坏也极大。据此,本文从环境因素和饵料性质等方面概述了影响鳜摄食的外在因素,并从摄食感觉器官和摄食调控因子剖析鳜摄食特性的内在因素,这有助于通过人工干预的方式调整鳜的摄食习性,为其转食配合饲料提供必要的支撑。此外,需加强鳜摄食调控机制及营养生理需求特性的研究,实现鳜配合饲料的突破,促进其养殖业的绿色健康发展。  相似文献

Attempts to commercially farm the Chilean oyster, Tiostrea chilensis in New Zealand are hampered by high levels of shell infestation by the polydorid polychaete, Boccardia acus. Wild stock in the warmer, northern waters of New Zealand typically present a mean of 8.6±,0.7 worms per right upper valve from an early stage of development (~17 mm shell height). Both hypersaline and freshwater treatments were effective in reducing worm numbers, but freshwater treatment for 180–300 min was most effective and resulted in >98% worm mortality without producing any obvious harmful effects on the oyster. Hyposaline treatment is therefore recommended as a simple and cost-effective method for the control of B. acus in farmed T. chilensis.  相似文献

为探究ncc、nkcc基因在花鲈渗透调节中发挥的作用,实验通过全基因组鉴定、多重序列比对、系统进化树构建以及蛋白结构预测对花鲈ncc进行了鉴定及序列分析,利用实时荧光定量PCR (qRT-PCR)检测ncc和nkcc在海水、淡水花鲈鳃组织中的表达水平,利用原位杂交技术确定ncc2和nkcc1a在海水及淡水花鲈鳃中的表达位置。结果显示,从花鲈中鉴定出2个ncc基因,即ncc1和ncc2,其编码序列(CDS)长度分别为2 691和3 120bp,编码896和1 039个氨基酸,在进化上具有保守性。ncc2在淡水花鲈鳃组织中的表达量显著高于海水,而nkcc1a在海水花鲈鳃组织中的表达量显著高于淡水,ncc1、nkcc1b、nkcc2在海淡水中的表达量则无显著差异。淡水适应过程中花鲈鳃组织中的ncc2的表达量逐渐上调,而nkcc1a的表达量逐渐下调;海水适应过程则呈现相反的表达趋势。此外,原位杂交结果显示,ncc2和nkcc1a基因分别位于淡水与海水中鳃组织的相邻鳃小片间的鳃丝上皮。以上结果表明,ncc2和nkcc1a基因分别编码淡水及海水花鲈鳃中重要的Na+及Cl  相似文献

A possible factor behind the successful use of polychaetes for prawn broodstock diet was investigated. Some steroid hormones in polychaetes have been suspected as being a potential component for prawn reproductive system development. Progesterone (P4) and 17-alpha hydroxyprogesterone (17α-OHP4) were extracted from polychaetes (Perinereis sp.) and female Penaeus monodon broodstock at various maturation stages. Concentration of both hormones was determined using RIA or HPLC. Immature natural polychaetes (2 months old) displayed the highest level of P4 and 17α-OHP4 compared to more mature ones (4, 6 and 8 months old polychaetes) (47.48 ± 6.70 ng P4/mg Protein and 143.74 ± 28.70 ng 17α-OHP4/mg Protein) whereas opposite results were received with cultured polychaetes in which a reverse relationship between P4 and its derivative with polychaete maturation was obtained. Diverse concentrations of both hormones may be linked with other factors such as types of polychaetes, habitats, seasons, and feed intakes. Twenty-four hours in vitro incubation of prawn previtellogenic oocytes with P4 or 17α-OHP4 extracted from polychaetes or with synthetic hormones, significantly increased percentages of vitellogenic oocytes and oocytes with cortical rod compared to the control with no hormones. P4 was more effective in enhancing the final maturation of oocytes while 17α-OHP4 had more effects on vitellogenic oocytes. Synthetic steroid hormones at equal hormone concentrations produced similar results to steroid hormones extracted from natural polychaetes.  相似文献

Enrichment of Artemia nauplii with a known probiotic yeast Saccharomyces boulardii (SB) and its role in enhancing resistance against the pathogen Vibrio harveyi was investigated. SB was cultured, then fed to instar II Artemia nauplii in three different treatments; 102 (T1), 103 (T2) and 104 (T3) colony forming units (CFU) per ml in triplicate. The algae Nanochloropsis sp. was used as control diet. Survival and total count of CFU nauplii−1 was observed on different media (Sabouraud, for enumerating yeasts, thiosulphate citrate bile salts sucrose, for enumerating Vibrio and seawater agar, for enumerating total aerobic flora) for each replication. Enhanced survival of nauplii was observed in treatments as compared to control. Results indicated that enrichment of SB in Artemia nauplii proceeded in a linear fashion, and up to 3500 CFU of SB could be detected in one nauplii at 104 CFU ml−1 treatment. No conclusive trend could be observed in the count of Vibrio and total aerobic flora due to treatment. Enriched nauplii were then challenged with the pathogen V. harveyi for 24 and 48 h at a concentration of 6.1 × 106 CFU ml−1. The survival counts at 48 h showed that the resistance of the nauplii was significantly (P < 0.01) improved in those fed with 104 CFU  ml−1 SB (90% survival rate after 48 h of challenge versus less than 40% for the infected control group without SB and treatments T1 and T2). This study shows that SB, which has been used for the first time in an aquatic live feed organism, has a profound beneficial effect on the nauplii by increasing its resistance to a pathogenic Vibrio infection. Great scallop (Pecten maximus L.) broodstock collected at two different seasons, spring (May) and winter (December), were given four different diets during conditioning. The spring group was spawned in late June, and the winter group in February.The highest number of veligers in the summer spawning (30 millions per 10broodstock), was produced using a diet consisting of 80% Tahitian Isochrysis galbana. In the winter spawning, broodstock given a dietconsisting of 80% diatoms (Skeletonema costatum/Chaetocerosgracilis) produced the highest number of veligers (29 millions per 10broodstock). Fecundity and the number of egg-releasing individuals were higher in the early summer spawning (June) than in the winter spawning (February), however, the total number of three days old veligers produced was nearly the same in the two seasons.  相似文献

为获得鱼源益生乳酸菌,本研究从健康鲫肠道内分离鉴定得到38株乳酸菌,并选择其中8株乳酸菌进行体外益生特性分析。结果显示,8株乳酸菌均能在p H=4.5和10%鲫胆汁环境中存活,对嗜水气单胞菌、豚鼠气单胞菌、无乳链球菌和副溶血弧菌均有较强的抑菌能力,但菌体表面疏水性和自凝集能力具有菌株特异性。选择5株疏水性较高(63%~89%)、自凝集能力强(37%~45%)的乳酸乳球菌进行体外黏附能力测定,结果显示,黏附能力在菌株间具有差异性(4.5%~7.9%),但均能显著降低嗜水气单胞菌NJ-35的体外黏附率,黏附抑制率达30%~35%;最后对筛选出的3株高黏附力的乳酸乳球菌进行安全性评价,发现3株菌对鲫均无致病力。鱼源乳酸菌的筛选,为鲫养殖生产中益生菌的实际应用提供了理论依据。  相似文献

通过93 d围隔实验比较了增加摄食配合饲料的鱼(草鱼和银鲫)密度和添加EM菌对三角帆蚌、草鱼、银鲫、鲢和鳙综合养殖系统中浮游植物群落和初级生产力的影响。采用2×2实验设计,设4个处理:LF0(20尾草鱼+10尾银鲫)、LFA(20尾草鱼+10尾银鲫+EM菌)、HF0(40尾草鱼+20尾银鲫)和HFA(40尾草鱼+20尾银鲫+EM菌)。所有处理中三角帆蚌、鲢和鳙密度相同,均为每个围隔内40只蚌、8尾鲢和2尾鳙。实验期间围隔内不换水,每天分2次投喂配合饲料;定期向LFA和HFA围隔内泼洒EM菌。结果显示,围隔内出现浮游植物超过81种,分别隶属7门、32科、73属;实验前期浮游植物优势种为微囊藻和栅藻,后期转为微囊藻、平裂藻和腔球藻;浮游植物生物量平均为3.2×108~8.3×108个/L;摄食配合饲料的鱼密度和EM菌对浮游植物种类组成和多样性无显著影响,但高密度草鱼和银鲫组(HF0和HFA)中浮游植物生物量和群落呼吸强度较高,初级生产力较低;添加EM菌可降低蓝藻在浮游植物生物量中的比例,增加初级生产力。研究表明,在蚌鱼综合养殖中放养摄食配合饲料的鱼密度不宜过高。  相似文献

为研究浙江近海浒苔Ulva spp.(Enteromorpha spp.)外生细菌多样性,采用传统的形态学和16S rDNA测序分析的方法,从舟山朱家尖、宁海国华电厂和奉化南沙3个地区分离到可培养的浒苔外生细菌及其周围海水细菌65株。根据细菌菌落特征和革兰氏染色结果等将分离到的细菌分为26种表型。16S rDNA序列测序比对发现:菌株与不动杆菌属(Acinetobacter)、交替单胞菌属(Alteromonas)、弧菌属(Vibrio)、假交替单胞菌属(Pseudoalteromonas)、微小杆菌属(Exiguobacterium)、芽孢杆菌属(Bacillus)、赤细菌属(Erythrobacter)、微球菌属(Micrococcus)、假单胞菌属(Pseudomonas)、深海杆菌属(Idiomarina)、Phaeobacter、Roseivirgaj和Silicibacter等23个属相应菌株具有较高同源性。对不同地区浒苔外生细菌进行了Shannon-Wiener多样性指数分析。研究表明:(1)宁海国华电厂的浒苔外生细菌多样性最丰富,多样性指数为93.98%;(2)浒苔外生细菌与其生活地区密切相关,其群落组成具有地域差异,其优势类群也不尽相同,但均归属于变形菌门。  相似文献

The virulence of two Vibrio strains, previously isolated from diseased corkwing wrasse Symphodus melops and identified as V. tapetis and V. splendidus, to corkwing and goldsinny wrasse Ctenolabrus rupestris and to Atlantic salmon Salmo salar, was studied under laboratory conditions. Both bacteria were shown to be opportunistically pathogenic to corkwing wrasse, causing significantly higher mortality in the challenged groups than in the controls. Bacterial cultivation of kidney samples and re-isolation of V. tapetis and V. splendidus from most mortalities confirmed the two strains as the probable cause of mortality in the challenged groups. The control group also suffered relatively high mortality, but no specific pathogens that were suspected to be the main cause of death were isolated, other than a mixture of Vibrio spp. and, in the case of one individual, atypical Aeromonas salmonicida. Following injection challenge with both bacterial strains, no mortality was recorded in Atlantic salmon. In bath challenge trials with goldsinny wrasse, only slight mortality was observed in the challenged groups and the unchallenged control group. Bacterial examination showed that atypical Aeromonas salmonicida was the probable cause of death in both bath challenged and control groups of goldsinny wrasse, and no indication of infection by any Vibrio sp. was found.  相似文献

Larval rearing of many marine organisms is dependent on the availability of live food. The aim of this study was to optimize larval first feeding for the mud shrimp Upogebia pusilla, by comparing the effectiveness of the two most commonly used live feeds: Brachionus plicatilis and Artemia sp. nauplii. Survival, larval duration, molt synchronism and megalop size were compared using five feeding treatments: Artemia from zoea I to IV (B0), Brachionus during zoea I and Artemia from zoea II to IV (B1), Brachionus during zoea I and II and Artemia during zoea III and IV (B2), Brachionus from zoea I to III and Artemia during zoea IV (B3) and Brachionus from zoea I to IV (B4). The proportion of larvae that reached the megalop stage was 0.00% in treatment B0, 3.33% in treatment B1, 33.33% in treatment B2, 66.67% in treatment B3 and 76.67% in treatment B4. Larvae fed on rotifers until zoea III or zoea IV stages had a higher survival but no differences were found either in time to reach megalop or in megalop size. This study demonstrates that rotifers are essential for the survival and development of U. pusilla early larval stages but that rotifers can be successfully replaced by Artemia nauplii in the zoea IV stage.  相似文献

The effects of saline-dissolved or Freund's incomplete adjuvant (FIA)-emulsified GnRHa treatment on the induction of ovulation in rainbow trout, Oncorhynchus mykiss, were examined. The FIA-emulsified GnRHa was first diluted in 0.25 ml physiological saline and then mixed with an equal volume of FIA. Fish were selected in the beginning of the spawning season and were allocated into four groups and were treated intraperitoneally with (a) 0.5 ml of emulsified GnRHa (GnRHa–FIA), (b) 0.5 ml saline-dissolved GnRHa in a single injection (GnRHa-1), (c) 0.5 ml saline-dissolved GnRHa in two injections spaced 1 d apart (GnRHa-2) and (d) 0.5 ml of saline (Control). The GnRHa dose in all hormone treatments was 25 µg kg− 1. All fish in the FIA–GnRHa and GnRHa-2 groups ovulated within 10 and 11 d after treatment, respectively. In contrast, only 75% in the Control fish and 60% of the fish in the GnRHa-1 group ovulated within 36 d after treatment. None of the treatments caused any pre- or post-spawning mortality in the broodstock. Fertilization, eyeing and hatching percentages of the produced progeny were normal in all the treatment groups and did not differ significantly among them. In conclusion, FIA-emulsified GnRHa can be effective in advancing the onset of and synchronizing the ovulation of rainbow trout within a two-week period, thus shortening the egg collection period, without affecting broodstock survival and egg quality.  相似文献

The Mekong giant catfish Pangasianodon gigas is endemic to the Mekong River and is a critically endangered species. The genotypes of the microsatellite DNA (msDNA) and mitochondrial DNA (mtDNA) markers (right domain of the control region) were detected to evaluate the present status of genetic divergence of this species from the Mekong River in Thailand and Cambodia. The observed and expected heterozygosity values of Mekong giant catfish in Thailand and Cambodia were relatively low in comparison with those of other nonendangered freshwater fish species. These two populations from Thailand and Cambodia showed similar levels of genetic diversity, as evaluated by the 384 nucleotides of the mtDNA control region with 13 haplotypes. The pairwise F ST value between the two populations based on the genotype frequencies of msDNA and mtDNA markers suggested a close genetic relationship between the populations in Thailand and Cambodia. The results of this study support the conclusion that the Mekong giant catfish is critically endangered. Care should be taken to sustain the genetic diversity of this species, as the level of genetic variability has already decreased in the wild population. This species is a target species for an ongoing stock enhancement program in the Mekong River in Thailand. It is proposed to apply these markers for proper broodstock management, such as for minimal kinship selective breeding in the hatchery.  相似文献

The effect of increasing levels of dietary phospholipids (PL) on the ovarian development and reproductive performance of Chinese mitten crab Eriocheir sinensis were investigated using four semipurified formulated diets supplemented with 0%, 1.2%, 2.4%, and 3.6% PL. Four groups of 40 females, with an average individual body weight of 95–120 g, were fed the experimental diets for a period of 7 months. Male crabs were introduced into the female rearing system in March, and mating, spawning, and egg hatching occurred in the following month. After 10 weeks of feeding, females fed the diet with 2.4% PL had a significantly higher gonadosomatic index (GSI) than females fed the diet with 0% PL, whereas females fed the diet with 1.2% PL had a significantly higher hepatic moisture content and lower hepatic lipid content than the other groups (< 0.05). After mating, the spawning rate was higher among females fed diets with 3.6% and 2.4% PL (95% and 92%, respectively) compared with females fed diets with 0% and 1.2% PL (both 81%). The results showed that egg production (total number of eggs/female) and fecundity increased with increasing dietary PL level, with females fed diets with 0% and 1.2% PL supplementation having significantly lower values than females fed the diet with 3.6% PL (< 0.05). In conclusion, our results suggest that dietary PL supplementation has a positive effect on ovarian development and reproductive performance of E. sinensis broodstock. Further study should aim to investigate the optimal PL level in the broodstock diet of E. sinensis in respect to offspring quality.  相似文献

Infections of gill amoebae that manifest as amoebic gill disease (AGD) occur in Atlantic salmon in Tasmania. The treatment of choice is freshwater bathing; however, the effectiveness of this treatment has declined over time. In this experiment, cage trials of chloramine‐T (Cl‐T) to treat AGD in Atlantic salmon were conducted over 3 months, and involved an initial bath in either freshwater or seawater with Cl‐T, followed by a second bath 6 weeks later. Amoeba densities were reduced to 50–80% of original values for both treatments. Neoparamoeba sp. density was not affected by bathing, and was not significantly different over the course of the experiment. Lesion prevalence was higher for Cl‐T‐treated fish than for freshwater‐treated fish, with overall prevalence levels of 14.30±1.00% and 8.03±0.57% respectively. This was also seen for gross gill scores. In the fortnight after each of the two baths, Cl‐T‐treated fish had significantly higher lesion levels, although this difference was then resolved by 4 weeks post bathing. The use of Cl‐T in seawater is at least as effective as freshwater at reducing amoebae density, and may be a more practical alternative when freshwater is in short supply.  相似文献

用血卵涡鞭虫可溶性抗原免疫BALB/c小鼠,经常规融合、间接ELISA方法筛选,将所得阳性克隆再经3次亚克隆后,共获得3株针对血卵涡鞭虫的单克隆抗体(2B2、3G4、4G7),单克隆抗体亚类鉴定表明,三者为IgG类抗体。用筛选的杂交瘤细胞株制备小鼠腹水抗体,其细胞上清及腹水效价分别为5.12×10-4和8.00×10-4。进一步利用单克隆抗体建立间接荧光抗体方法对单抗特异性进行鉴定,阳性虫体被染上黄绿色荧光,而正常梭子蟹血淋巴则未被染色。用单克隆抗体和多克隆兔抗血清以羊抗鼠HRP-IgG为酶标抗体,建立了检测血卵涡鞭虫的双抗体夹心ELISA方法,该方法对血卵涡鞭虫阳性标本检测符合率为100%。结果表明,制备的单克隆抗体效价高、特异性好,可用于血卵涡鞭虫的早期临床诊断。  相似文献
