动物精子通过多次复杂的细胞分化最终形成精子,此过程受许多因素的调控,其中起着决定性作用的是生精细胞内的激素调控。文章对动物精子发生调控的相关激素,如促卵泡激素、促黄体生成素、抑制素、褪黑素的研究进展进行了综述,并对动物精子发生调控激素的研究进展进行了展望,以期为研究者提供一定参考。  相似文献

为探究促性腺激素对坝上长尾公鸡精液品质的影响,本试验选用28周龄健康、体重一致的坝上长尾公鸡36只,随机分成3组,每组12只,将三个组别分别注射0、10、40IU促卵泡素,每间隔12h重复注射,第4天注射100 IU的人绒毛膜促性腺激素(h CG),在第7天对36只鸡进行人工采精,收集各组数据并进行对比分析。采精后用生理盐水溶液对原精液进行5倍、10倍、15倍稀释,然后进行精液品质检查。在显微镜下观察,测得不同稀释倍数生理盐水溶液在低温条件下对精子活率的影响。结果显示,注射促卵泡素10、40IU与对照组相比差异极显著(P0.01);对照组与注射10IU促卵泡素、注射40IU促卵泡素的各个分组中精子畸形率数据表现为差异极显著(P0.01);稀释处理后的精液在低温保存下结果显示,稀释5倍的精子保存6 h后活率无显著变化(P0.05),稀释10倍和15倍的两组的精子活率则显著降低(P0.05);在低温保存12 h后,稀释10倍和15倍的精子活率均显著低于稀释5倍的精子活率(P0.05)。结果表明,注射促卵泡素可以提高精子活率,降低精子畸形率;精液稀释倍数不同,稀释倍数越高的精子活率越低,稀释倍数越低的精子活率越高;每只最佳注射剂量为10 IU,精液稀释倍数不超过5倍。  相似文献

为研究饲料添加淫羊藿的番鸭睾丸组织相关基因和调控通路变化,本研究将60只公番鸭随机分为2组,每组3个重复,一组饲喂基础日粮(对照组),另一组饲喂基础日粮的基础上添加量为1%的淫羊藿(实验组)。饲喂75 d结束后,测定血清生殖激素和抗氧化指标,并进行睾丸组织形态学观察和高通量转录组测序。结果显示,实验组生精上皮层数多,生精细胞数量多,管腔饱满,成熟精子多。实验组血清生殖激素中睾酮含量、雌二醇含量、促卵泡素含量和促黄体素含量均显著高于对照组,实验组血清抗氧化指标中超氧化物歧化酶含量、谷胱甘肽过氧化物酶含量、总抗氧化水平均显著高于对照组,丙二醛含量显著低于对照组。通过转录组测序分析,本研究最终得到2 414个差异表达基因,这些差异表达基因主要被注释到白细胞激活、受体等过程,主要富集于细胞因子受体相互作用、铁死亡等代谢途径和信号通路。本文研究表明饲料添加淫羊藿激活了番鸭睾丸相关基因或通路表达,对番鸭繁殖力的提高具有重要意义。  相似文献

试验采用两步酶消化法对2月龄公羔睾丸进行处理,得到了总数为7.44×109的生精细胞悬液,经台盼蓝染色,细胞活率在90%以上;支持细胞的贴壁性极好,生精细胞附着在支持细胞上,随着培养时间延长,生精细胞、支持细胞逐渐退化,呈现“集合”的趋势,细胞数量逐渐减少;经BrdU标记、H.E.染色,检测培养的生精细胞到分化到圆形精子细胞阶段,表明成功构建了山羊睾丸生精细胞共培养体系,可为后续深入研究山羊精子体外成熟培养提供一定的实验依据.  相似文献

为研究羔羊睾丸支持细胞发育过程中的形态学变化,本实验以湖羊1-5月龄的羔羊睾丸为材料,采用常规石蜡包埋、组织切片技术,应用Olympus DP71显微镜照相系统和DP Control软件拍照获取图像,观察了睾丸支持细胞、生精细胞的的形态学变化,利用显微测微系统测量了支持细胞大小、数量、生精小管管径以及面积变化。结果表明:性成熟前羔羊睾丸的解剖特征参数总体表现出左侧大于右侧;随着羔羊睾丸的发育,支持细胞的结构更加清晰完整,细胞体积逐渐增大,生精小管管径、睾丸支持细胞和生精细胞的数目都有增加,并没有在生精小管中发现圆形精子。性成熟之前,羔羊睾丸支持细胞的快速增长主要表现在生精小管直径显著性增大,随着羔羊睾丸生长发育,支持细胞的增殖能力增强,并对生精细胞的发育有一定的影响,本研究可为进一步研究不同季节及不同发情时期滩羊睾丸发育特征提供参考。  相似文献

支持细胞对维持精子形成过程中的微环境起决定作用,它可以通过分泌功能、细胞间连接形成的血睾屏障功能以及吞噬功能等来促进精子的形成过程,其发育异常会导致不同程度的雄性生殖缺陷。基于支持细胞在雄性动物生殖过程中的作用,体外培养高纯度支持细胞可成为研究睾丸两大核心功能-精子发生和性激素分泌功能相关调节机制重要的细胞模型。此外,体外培养睾丸支持细胞也可作为生殖毒理学等新兴热点领域的细胞模型,为评估和研究环境因素对雄性生殖的影响提供便利。因此,作者系统地归纳、总结了目前关于动物支持细胞生物功能的研究及常用的体外分离纯化、培养及鉴定方法,以期为利用动物支持细胞开展雄性生殖领域的研究提供参考。  相似文献

Sertoli cells are the only somatic cells in the seminiferous epithelium which directly contact with germ cells. Sertoli cells exhibit polarized alignment at the basal membrane of seminiferous tubules to maintain the microenvironment for growth and development of germ cells, and therefore play a crucial role in spermatogenesis. Androgens exert their action through androgen receptor (AR) and AR signalling in the testis is essential for maintenance of spermatogonial numbers, blood–testis barrier integrity, completion of meiosis, adhesion of spermatids and spermiation. In the present study, we demonstrated that AR gene could promote the proliferation of immature porcine Sertoli cells (ST cells) and the cell cycle procession, and accelerate the transition from G1 phase into S phase in ST cells. Meanwhile, miR-124a could affect the proliferation and cell cycle procession of ST cells by targeting 3′-UTR of AR gene. Furthermore, AR bound to the RNF4 via AR DNA-binding domain (DBD) and we verified that RNF4 was necessary for AR to regulate the growth of ST cells. Above all, this study suggests that AR regulates ST cell growth via binding to RNF4 and miR-124a, which may help us to further understand the function of AR in spermatogenesis.  相似文献

Activins and inhibins are regulatory proteins of the reproductive function. Inhibins antagonise the activin signalling at different levels and are responsible for the negative feedback in the regulation of the release of pituitary follicle stimulating hormone (FSH), which, in turn, is promoted by locally produced activins. In the canine ovary, both peptides are expressed by developing follicles and corpora lutea. Activins may play a stimulatory role in follicular development, promoting the aromatase function; inhibins modulate these processes and suppress the hyperplasic/neoplastic stimuli. Activins are required for ovulation and corpus luteum formation, while inhibins stimulate progesterone synthesis. The exclusive production of alpha‐inhibin by granulosa cells allows the peptide to be used as marker to identify canine ovarian stromal tumours by immunohistochemistry. In the male, activins are powerful morphogenetic factors in the foetal testis. In the adult, they display a modulating action on spermatogenesis and Sertoli cell function. Inhibins, produced mainly by Leydig cells, promote testosterone secretion. Canine testicular tumours, such as Leydig, Sertoli and granulosa cell tumours (GCTs), may express inhibin subunits and produce high circulating levels of these glycoproteins. In the canine prostate, activins inhibit epithelium proliferation, antagonising androgen effects, but they are synthesised under androgenic stimulus.  相似文献

Quantitative morphological methods were used to analyse the histomorphometric changes and variations in the number and size of cells from diverse cellular populations in testes of newly hatched chicks treated with follicle stimulating hormone (FSH) during embryonic development. The tissue was fixed and embedded in Epon and sections were morphometrically measured under light microscopy, using point counting for volume densities and the Floderus equation to determine numerical density. The average volume of the individual cell was determined by dividing the volume density by the numerical density. Results indicate that FSH administration causes an increase in the number of Sertoli cells and spermatogonia, as well as enlargement of the individual Sertoli cells leading consequently to an increase in the diameter and volume density of the testicular seminiferous tubules. Results also reveal an increase in the volume density of the interstitial cords of the Leydig cells, this expansion is due to the enlargement of the individual Leydig cells and not to an increase in their number, which remains constant. We conclude that testes of chick embryos are able to respond to FSH treatment, as revealed by the changes in the number and size of the cells conforming the diverse cellular populations of the testis. FSH treatment during embryonic development induces histomorphometric changes in both the interstitial tissue and seminiferous tubules, accelerating their growth and differentiation.  相似文献

Sertoli cells produce special microenvironment for developing germ cells; therefore it is assumed that they play primary role in the onset and control of spermatogenesis. In this connection we extended our previous study on the ultrastructure of Sertoli cells in different domestic and wild animals with special regard to nucleolus. Sertoli cells of domestic and wild ruminants possess the typical vesicular nucleolus except for fallow deer, in this species no vesicular nucleolus occurs in Sertoli cells even during the rut. In roe.buck, another wild ruminant with seasonal spermatogenesis, cyclic changes were found in the nucleolus of Sertoli cells. If no spermatogenesis is present, the Sertoli cells have a reticular nucleolus. Membranous vesicles appear in the nucleolus of Sertoli cells of roe-buck at the onset of spermatogenesis 1-2 months before rut. In domestic ruminants with continuous spermatogenesis the vesicular nucleolus in Sertoli cells is present permanently. During postnatal development of bull and ram the vesicular nucleolus appears in Sertoli cells just before the onset of spermatogenesis. In experimental cryptorchidism of bulls a vesicular nucleolus is found in the Sertoli cells. Our observations and experiments support a hypothesis that Sertoli cells have primary role at the onset and the maintenance of spermatogenesis.  相似文献

Tight junctions (TJ) in inter-Sertoli junctional areas and epididymal epithelia are important for the formation of blood-testis barrier (BTB) and blood-epididymal barrier (BEB). In this study, the expression of occludin, an integral member of the TJ, was verified in canine testis and epididymis. Both low molecular weight (MW) (25-28 kDa) forms as well as high MW (68-72 kDa) forms of occludin were detected in the testis and epididymis using Western blot. The relative amount of the high MW forms of occludin vs low MW forms was higher in the testis than in the epididymis. Some difference in the composition of different MW forms of occludin was found along the segments of epididymis, suggesting the possible correlation between cellular composition of occludin proteins and paracellular permeability of epithelia along the epididymal tubule. In the testis, intense occludin immunoreactivity was found in the basally located inter-Sertoli junctional area. Diffused immunoreactivity of occludin was also found in the cytoplasm of Sertoli cells. A similar pattern of zonula occludens-1 immunoreactivity was found in the cytoplasm of Sertoli cells, suggesting that occludin was not confined to the inter-Sertoli junctional areas and that subcellular localization of occludin in the Sertoli cells was dynamically regulated during spermatogenesis in canine testis. In the epididymis weak immunoreactivity was found in the apical sides and cytoplasm of epithelial cells.  相似文献

The present study demonstrates the postnatal developmental changes in immunohistochemical localization of alpha-smooth muscle actin (SMA) and vimentin in the bovine testis. In the peritubular myoid cells of seminiferous tubules and the sub-epithelial and stromal cells of straight tubules and the rete testis, alpha-SMA starts appearing at around 4 months of age. Peritubular alpha-SMA attains the continuous mature pattern at around 5 months of age whereas sub-epithelial and stromal alpha-SMA increases with advancing age. Vimentin is localized in the perinuclear zone of Sertoli cells, peritubular and vascular wall cells, a few interstitial cells, and in the basal part of the epithelia of straight and rete tubules. Developmental changes are only evident in the Sertoli cell vimentin, which is basal and weak at birth and increases moderately until 4 months of age. From around 5 to 8 months of age when the Sertoli cells are under morphological transformation, vimentin intensity is considerably increased and the characteristic vimentin extensions connect the Sertoli nuclei to the basal membrane. These extensions get shorter at around 9 month of age as the Sertoli nuclei are positioned basally. The mature Sertoli cell perinuclear vimentin is strong and stable without infranuclear extension. In conclusion, the age of appearance of alpha-SMA coincides with the onset of postnatal division of spermatogonia, and vimentin may play a key role in stabilizing Sertoli cell nuclei during their transformation in bovine.  相似文献

The proliferation and differentiation ability of testicular Sertoli cells directly affects spermatogenesis and male reproductive development. WNT proteins are involved in the regulation of cell proliferation, differentiation and spermatogenesis. Therefore, to study whether lncRNAs, which regulate the expression of WNT proteins during cell proliferation and differentiation, are worthwhile. In this study, testicular tissue from the Dazu black goat (Capra, goat, Chongqing, China) at neonatal time (less than 7 days old), early puberty time (45 days old) and sexual maturity time (90 days old) at three ages was subjected to high-throughput sequencing to predict testicular growth and development associated with WNT lncRNA. The final screening of lncWNT3-IT may be targeted to regulate the expression of WNT3. At the same time, the expression of WNT3 was verified by lncWNT3-IT by paraffin sectioning, fluorescence in situ hybridization, interference, overexpression, cytotoxicity assay, Western blotting and qPCR. The following results were obtained: lncWNT3-IT was expressed in the testicular Sertoli cells and played a role in the Sertoli cell cytoplasm. Fluorescence in situ hybridization localization analysis showed that lncWNT3-IT positively regulated the expression of WNT3, and through cell viability and cell proliferation experiments, it was found that the expression of lncWNT3-IT assisted in Sertoli cell proliferation. In summary, lncWNT3-IT can influence the proliferation of Sertoli cells by positively regulating the expression of WNT3.  相似文献

Calretinin is a calcium-binding protein expressed abundantly in the central and peripheral neural tissues. It has been demonstrated to be a valuable marker in human testicular neoplasia. The immunohistochemical expression of calretinin has been studied in 102 samples of normal (n=25) and three different neoplastic canine testicular tumours (n=77). In normal canine testis, calretinin expression was restricted to Leydig and Sertoli cells of the testis. In tumour tissues, calretinin expression was detected in all tumours investigated (interstitial cell tumours, seminoma, and Sertoli cell tumours), with a cytoplasmic and nuclear pattern of cellular distribution. The present work reports, for the first time, calretinin immunohistochemical expression in normal and neoplastic canine testis.  相似文献
