A study was undertaken in 2004 and 2005 to characterize pathogens associated with damping-off of greenhouse-grown cucumber seedlings in 13 districts in Oman. Identification of Pythium to the species level was based on sequences of the internal transcribed spacer (ITS) of the ribosomal DNA. Of the 98 Pythium isolates collected during the survey, Pythium aphanidermatum , P. spinosum , P. splendens and P. oligandrum accounted for 76%, 22%, 1% and 1%, respectively. Pythium aphanidermatum was isolated from all of the districts, while P. spinosum was isolated from seven districts. Pathogenicity tests showed inter- and intraspecific variation in aggressiveness between Pythium species. Pythium aphanidermatum , P. spinosum and P. splendens were found to be highly aggressive at 25°C. However, the aggressiveness of P. spinosum decreased when the temperature was raised to 30°C, which was found to correspond to the lower frequency of isolation of P. spinosum in the warmer seasons, compared to the cooler time of the year. Pythium aphanidermatum exhibited limited intraspecific variation in the sequences of the ITS region of the rDNA and showed 100% similarity to the corresponding P. aphanidermatum sequences from GenBank. The ITS sequence data, as well as morphological characteristics of P. spinosum isolates, showed a high level of similarity within and between P. spinosum and P. kunmingense , and suggested that the two species were synonymous. This study represents the first report of P. spinosum, P. splendens and P. oligandrum in Oman.  相似文献

Four Pseudomonas strains were evaluated for their intrinsic properties conferring their ability to protect long English cucumber against Pythium aphanidermatum in hydroponic culture. Two of the strains, BTP1 and its siderophore-negative mutant M3, increased plant yield as compared with the non-inoculated control plants. Strain BTP7 was intermediate in its biocontrol activity while strain ATCC 17400 failed to reduce disease development. The role of pyoverdines could not be confirmed since treatment with either BTP1 or its siderophore-negative mutant M3 provided similar suppression of Pythium disease. In addition, no siderophores were detected in the nutrient solution. BTP1 did not inhibit pathogen growth in vitro on several media, suggesting that antibiosis was not a mechanism of suppression. Quantification of root bacterial populations did not indicate differences among the strains. On the other hand, roots treated with either BTP1 or its sid mutant M3 contained more antifungal phenolics than roots from any other treatments including controls. These results suggest that antifungal compounds induced by inoculation of cucumber roots with the fluorescent Pseudomonas strains BTP1 and M3 participate actively in the protection of cucumber plants against P. aphanidermatum  相似文献

A total of 651 isolates of cucumber corynespora leaf spot fungus ( Corynespora cassiicola ) collected from cucumber in Japan, either with (438 isolates) or without (213 isolates) a prior history of boscalid use, were tested for their sensitivity to boscalid by using a mycelial growth inhibition method on YBA agar medium. Additionally, seven isolates of C. cassiicola obtained from tomato, soybean, eggplant (aubergine) and cowpea in different locations in Japan were tested before boscalid registration. Minimum inhibitory concentration (MIC) and 50% effective concentration (EC50) values for 220 isolates from crops without a prior history of boscalid use ranged from 0·5 to 7·5 μg mL−1 and from 0·04 to 0·59 μg mL−1, respectively. Two hundred and fourteen out of 438 isolates collected from ten cucumber greenhouses in Ibaraki Prefecture, Japan, which received boscalid spray applications showed boscalid resistance, with MIC values higher than 30 μg mL−1. Moreover, resistant isolates were divided into two groups: a moderately resistant (MR) group consisting of 189 isolates with EC50 values ranging from 1·1 to 6·3 μg mL−1, and a very highly resistant (VHR) group consisting of 25 isolates with EC50 values higher than 24·8 μg mL−1. MR isolates were detected from all ten greenhouses, but VHR isolates were detected from only three. As a result of fungus inoculation tests which used potted cucumber plants, control failures of boscalid were observed against resistant isolates. Efficacy of boscalid was remarkably low against VHR isolates in particular. This is the first known report on boscalid resistance in Japan.  相似文献

Metalaxyl was used to control Pythium diseases of carrots in experiments on farms with a history of cavity spot. The first experiment compared the method of application (sprayed, banded or broadcast) and rate (0, 1.5, 3 or 6 kg a.i. ha−1) one week after sowing. Three additional experiments compared the rate (0, 0.75, 1.5 or 3 kg a.i. ha−1) and time (sowing, 1- to 2- or 4- to 5-true-leaf stage) of application. In expt 1, the application of metalaxyl, but not the method by which it was applied, increased yield by 20% and significantly reduced the incidence of cavity spot, forking and misshapen carrots. In expts 2, 3 and 4, neither the rate nor time of application affected yield or reduced the incidence of Pythium diseases. Comparison of the sites showed that they differed in past metalaxyl usage. Metalaxyl had not been used on the site of expt 1, but had been used previously at sites 2, 3 and 4. Laboratory experiments were conducted to determine whether these differences in efficacy resulted from reduced sensitivity of Pythium isolates to metalaxyl, or reduced persistence of metalaxyl in soil. ED50 values showed that there was no reduction in metalaxyl sensitivity. The half-life of metalaxyl was 82 days in soil from expt 1, but was 10 days or fewer in soils from expts 2, 3 and 4. Thus the failure of metalaxyl to control Pythium diseases was associated with reduced persistence in soil, not reduced sensitivity of the target fungi.  相似文献

Hydroponically grown cucumber (Cucumis sativus) seedlings were inoculated with zoospores of 1 mycoparasitic (Pythium oligandrum) and 2 pathogenic (Pythium aphanidermatum and Pythium group F) Pythium spp. During the first 2 days after inoculation, all the Pythium spp. caused reduction in the root length. However, roots treated with Pythium oligandrum quickly reached the length of the control and on the 8th day, and for the rest of the experimental period, stimulation of root elongation was noted. Pythium oligandrum was not pathogenic on cucumber and no differences in the fresh weights of control and Pythium oligandrum inoculated plants were observed in the course of the experiment. Pythium group F and Pythium aphanidermatum were pathogenic on cucumber seedlings, but their pathogenicities differed. Thus, while Pythium group F had a constant, negative influence on root length and plant growth, measured as fresh weight, Pythium aphanidermatum caused generalized necroses of the root system, inhibiting consistently root elongation and plant growth and finally causing plant death. Moreover, the zoospores of 2 mycoparasitic species, Pythium oligandrum and Pythium periplocum, were not attracted to roots of cucumber and accumulated on the roots in very low numbers compared to those of the pathogenic species, Pythium aphanidermatum, which were strongly attracted and accumulated in large numbers. Finally, it was also found that Pythium oligandrum colonized the roots very poorly, while Pythium group F and Pythium aphanidermatum were significantly better root colonizers. The significance of these findings is discussed in relation to the ecology of Pythium species and biocontrol.  相似文献

Pseudomonas putida 40RNF applied to seed pellets reduced the occurrence of Pythium damping-off of sugar beet. A density of 6 × 107 40RNF per pellet reduced Pythium damping-off from 70 to 26% when seeds were sown in artificially infested soil (250 propagules Pythium ultimum per g dry soil). The efficacy of 40RNF was dependent on its density in the seed pellet (in the range 2 × 104–6 × 108 per pellet) and on the number of propagules of Pythium in soil. 40RNF declined to or stabilized at approximately 1 × 106 per pellet 3 days after planting, and this was independent of the inoculum density. This indicated that the crucial steps resulting in damping-off of sugar beet caused by Pythium ultimum must occur within 3–4 days of sowing. 40RNF reduced pericarp colonization by P. ultimum by 43% 48 h after planting and caused a 68% decrease in the number of sporangia of P. ultimum in the surrounding soil (0.0–5.0 mm). P. putida 40RNF also reduced pre and post-emergence damping-off (from 69.5 to 37.5%) caused by indigenous populations of Pythium species in an infested soil and this was as effective as the fungicide hymexazol (69.5 to 40%).  相似文献

The effects of Coniothyrium minitans inoculum quality and an 8-week interval between inoculum application and crop planting on sclerotinia ( Sclerotinia sclerotiorum ) disease in three successive lettuce crops were investigated in a glasshouse trial. Spore suspensions of three isolates of C. minitans (Conio, IVT1 and Contans) applied at 108 CFU m−2 and a standard Conio maizemeal–perlite application (06 L m−2, 1011 CFU m−2) were assessed for their ability to control S. sclerotiorum . Only the maizemeal–perlite inoculum (isolate Conio) consistently reduced sclerotinia disease. In the third lettuce crop only, isolates IVT1 and Contans formulated by Prophyta and isolate IVT as an oil–water formulation, all applied as spore suspensions, reduced disease at harvest compared with the untreated control. Recovery, viability and C. minitans infection of sclerotia buried during the 8-week period prior to each of the three lettuce crops, and of sclerotia formed on the crop, were tested. Only the maizemeal–perlite inoculum (isolate Conio) reduced the recovery of sclerotia buried in soil for weeks between inoculum application and crop planting, reducing their viability and increasing infection by C. minitans . Eight weeks was sufficient to enable C. minitans to infect sclerotia of S. sclerotiorum , and may account for disease control. After harvest of the second and third crops, maizemeal–perlite treatment (isolate Conio) reduced the number and viability of sclerotia recovered on the soil surface and increased infection by C. minitans compared with spore-suspension treatments. The effect of inoculum concentration and the influence of soil temperature (varying with time of year) on infection of sclerotia by C. minitans are discussed.  相似文献

Sesbania rostrata ( S. rostrata) Brem. & Oberm., a member of the Fabaceae family, has been used as a promising halophytic plant to ameliorate soil salinity in north-east Thailand. To obtain information regarding the mechanism of salt tolerance, the physiological responses of S. rostrata to NaCl was compared with those of the salt-susceptible species, kidney bean ( Phaseolus vulgaris L. cv. Meal). Seedlings were grown hydroponically with 0, 50, 100 and 150 m m NaCl for 10 days and their effects on growth, chlorophyll content, fluorescence yield ( F v/ F m), inorganic elements and amino acid content were determined. The results showed that tolerance to NaCl was clearly different between the two plants. At the highest concentration (150 m m ), the dry weight of S. rostrata was more than 50% greater than the control, whereas the kidney bean could not survive. Chlorophyll a content drastically reduced only in the kidney bean. The F v/ F m of S. rostrata did not change with increasing concentrations of NaCl, but that of kidney bean decreased. Greater percentages (≥80%) of absorbed Na+ and Cl were translocated and accumulated in the shoots of S. rostrata , but remained largely in the roots of kidney bean. The enhancement of contents of amino acids, including proline, with increasing NaCl was observed in both species. These results strongly suggest that the salt tolerance of S. rostrata is associated with the ability of the plant to translocate and sequester Na+ and Cl in the shoot cells.  相似文献

The liana, hiptage ( Hiptage benghalensis ), is currently invading the wet tropics of northern Queensland and remnant bushland in south-eastern Queensland, Australia. Trials using seven herbicides and three application methods (foliar, basal bark, and cut stump) were undertaken at a site in north Queensland (158 700 hiptage plants ha−1). The foliar-applied herbicides were only effective in controlling the hiptage seedlings. Of the foliar herbicides trialed, dicamba, fluroxypyr, and triclopyr/picloram controlled >75% of the treated seedlings. On the larger plants, the cut stump applications were more effective than the basal bark treatments. Kills of >95% were obtained when the plants were cut close to ground level (5 cm) and treated with herbicides that were mixed with diesel (fluroxypyr and triclopyr/picloram), with water (glyphosate), or were applied neat (picloram). The costings for the cut stump treatment of a hiptage infestation (85 000 plants ha−1), excluding labor, would be $A14 324 ha−1 using picloram and $A5294 ha−1 and $A2676 ha−1, respectively, using glyphosate and fluroxypyr. Foliar application using dicamba for seedling control would cost $A1830 ha−1. The costs range from 2–17 cents per plant depending on the treatment. A lack of hiptage seeds below the soil surface, a high germinability (>98%) of the viable seeds, a low viability (0%) of 2 year old, laboratory-stored fruit, and a seedling density of 0.1 seedlings m−2 12 months after a control program indicate that hiptage might have a short-term seed bank. Protracted recolonization from the seed bank would therefore be unlikely after established seed-producing plants have been controlled.  相似文献

Summary Density:yield loss models rely on fixed coefficients, parameterized from a particular site and season to predict the impact of weeds on crop yields. However, the empiricism of this approach and failure to incorporate environmental effects, has major biological and economic implications. In this study, seasonal variability in wheat yield loss and associated economic costs from Avena spp. were quantitated. A competition experiment at Wagga Wagga, NSW, showed large seasonal differences in wheat yield loss from densities of Avena spp. across 2 years. Gross margins, simulated over a 51-year period, decreased as Avena spp. density increased and were more variable at low crop densities and higher weed densities. For example, at a density of 200 Avena spp. plants m−2, coefficient of variation in crop gross margin ( CV ) was $AUS 47 ha−1 for a crop density of 200 wheat plants m−2 compared with a CV of $AUS 75 ha−1 for a crop density of 50 wheat plants m−2. The value of yield loss predictions will be vastly improved by making parameter values in yield loss models a function of seasonal factors such as rainfall.  相似文献

Application of Pythium oligandrum to a soil-based compost as a mycelial suspension (5 × 102 CFU g−1 of dry compost) and oospore alginate pellets (105 oospores/g of dry compost) controlled pre- and postemergence damping-off of sugar beet caused by Pythium ultimum to a level similar to metalaxyl seed treatment. Oospore seed treatments and aqueous suspensions of oospores applied to compost failed to control disease. Problems in the use of P. oligandrum oospore inocula for the control of damping-off were highlighted. It was shown that treatment of oospores with cellulase (20 g L−1) increased germination approximately three-fold in comparison to untreated spores. Untreated and cellulase pretreated oospores were subsequently evaluated as seed treatments for their ability to control damping-off of sugar beet. The highest rate of pretreated oospores (104 oospores/seed) gave levels of emergence and establishment in infested compost that were not significantly different from the uninfested controls, whereas seed treatment with untreated oospores gave no significant reduction in disease. In a trial carried out in a controlled environment to assess the effect of pH (4.5–8.0), P. oligandrum (104 cellulase pretreated oospores/seed) was shown to control pre- and postemergence damping-off of sugar beet at pH 7.0 and 7.5 only.  相似文献

Pythium spp. that cause damping-off of seedlings also can cause root rot of older plants and lead to yield reductions. This can be especially severe in soilless cultures where the fungus can spread easily with the nutrient solution. 39Pythium isolates obtained from discolored roots were assayed for their ability to cause damping-off on cucumber seedlings in sand-peat and for their pathogenicity in soilless culture of cucumber in rockwool or hydroponic solution. Isolates ofPythium aphanidermatum, P. irregulare, P. sylvaticum andP. ultimum were highly pathogenic in sand-peat, but onlyP. aphanidermatum strains were pathogenic in soilless conditions and led to root decay, plant death in rockwool culture and growth reduction in hydroponic culture. One strain ofP. aphanidermatum significantly reduced the yield of cucumber grown in rockwool under conditions similar to those of commercial cultures.  相似文献

The mutual effects of redroot pigweed ( Amaranthus retroflexus ) on corn ( Zea mays ) were evaluated in an experiment conducted in 2005 at the Iranian Plant Protection Research Institute at Qazvin, considering the different densities of redroot pigweed against four different corn densities. Redroot pigweed, at 0, 35, 50, 65, and 80 plants m−1 row−1, was arranged factorially with corn at four, five, six, and seven plants m−1 row−1 in a randomized complete block design. Crop–weed competition resulted in a reduction in the total dry matter, Leaf Area Index, and crop growth rate of corn. Furthermore, an increasing weed density ≤65 plants m−1 row−1 reduced the corn grain yield and biological yield. Overall, six corn plants m−1 row−1 was suggested as the optimum density of this crop in competition with redroot pigweed.  相似文献

Host–parasite relationships in root-knot disease of spinach caused by Meloidogyne incognita race 1 were studied under glasshouse conditions. Nematode-induced mature galls were large and usually contained one or more females and egg masses with eggs. Feeding sites were characterized by the development of giant cells containing granular cytoplasm and many hypertrophied nuclei. The cytoplasm in these giant cells was aggregated alongside the thickened cell walls. Stelar tissues within galls appeared disorganized. The relationship between initial nematode population density ( P i) in a series from 0–128 eggs and second-stage juveniles per cm3 soil and growth of spinach cv. Symphony F1 seedlings was tested under glasshouse conditions. A Seinhorst model [ y = m  + (1 −  m ) z P–T ] was fitted to fresh top- and total plant-weight data for inoculated and control plants. Tolerance limits ( T ) of spinach cv. Symphony F1 to M. incognita race 1 for fresh top and total plant weights were 0·25 and 0·5 eggs and second-stage juveniles per cm3 soil, respectively. The minimum relative values for fresh top and total plant weights were zero in both cases at P i ≥ 32 eggs and second-stage juveniles per cm3 soil. Root galling was least at low initial population densities and greatest at 16 eggs and second-stage juveniles per cm3 soil. Maximum nematode reproduction rate was 33·1-fold at the lowest P i.  相似文献

In a survey conducted during October 1995, single-lesion isolates of the sugar beet leaf-spot fungus, Cercospora beticola , were tested for sensitivity to the sterol demethylation inhibiting fungicides (DMIs) flutriafol and bitertanol. The isolates were collected from fields in three different areas of northern Greece. Fields at Serres and Imathia had been sprayed with DMIs for about 15 years to control sugar beet leaf-spot. At the third site, Amyndeon, DMI fungicides had not been used. From each area 150 isolates were tested. ED50 values were calculated for individual isolates by regressing the relative inhibition of colony growth against the natural logarithm of the fungicide concentration. The mean ED50 values for flutriafol for the Serres, Imathia and Amyndeon populations were 1·07, 0·73 and 0·5 µg mL−1, respectively (significantly different at P  = 0·05). For bitertanol the mean ED50 values for the Serres and Imathia populations were 0·72 and 0·81 µg mL−1, respectively, which were not significantly different at P  = 0·05. The mean ED50 value of the Amyndeon population was 0·48 µg mL−1, which was significantly lower than those of the other two populations ( P  < 0·05). A cross-resistance relationship was found to exist between the two triazole fungicides tested when log transformed ED50 values of 60 isolates were subjected to a linear regression analysis ( r  = 0·81).  相似文献

Sensitivity to the new carboxylic acid amide (CAA) fungicide mandipropamid (MPD) in Phytophthora infestans was measured for isolates collected between 1989 and 2002 in Israel prior to the commercial use of MPD (baseline sensitivity, 44 isolates), and from MPD-treated (25 isolates) and untreated fields (215 isolates) in nine European countries and Israel between 2001 and 2005. All isolates were sensitive to MPD, with EC50 values ranging between 0·02 and 2·98  µ g mL−1. Plastic-tunnel (UK), shade-house (Israel) and field experiments (Israel) conducted during 2001–05 showed that enforced selection pressure, applied preventively or curatively, imposed by repeated sublethal (5  µ g mL−1) or excessive (500–1000  µ g mL−1) doses of MPD on mixed isolates of P. infestans produced no isolates resistant to the compound. The results of this study indicate that the probability of a buildup of resistant sub-populations of P. infestans to mandipropamid in the field is low.  相似文献

The sensitivity of 127 Phytophthora infestans isolates to flumorph was determined in 2003 and 2004. The isolates originated from two geographical regions and showed similar levels of sensitivity in both years. Baseline sensitivities were distributed as a unimodal curve with EC50 values for growth of mycelia ranging from 0·1016 to 0·3228  µ g mL−1, with a mean of 0·1813 (± 0·0405) µ g mL−1. There was no cross-resistance between flumorph and metalaxyl. Laboratory studies were conducted to evaluate the risk of P. infestans developing resistance to flumorph. Mutants resistant to metalaxyl or flumorph were obtained by treating mycelium of wild-type isolates with ultraviolet radiation. Metalaxyl-resistant mutants were obtained with a high frequency and exhibited resistance factor values (EC50 resistant/EC50 sensitive phenotypes) of more than 100, while flumorph-resistant mutants were obtained at much lower frequencies and had very small resistance factors (1·5–3·2). There was cross-resistance between flumorph and dimethomorph, but not with azoxystrobin or cymoxanil. Most flumorph-resistant mutants showed decreases in hyphal growth in vitro and in sporulation both in vitro and on detached leaf tissues. These studies suggested that the risk of resistance developing was much lower for flumorph than metalaxyl. However, as P. infestans is a high-risk pathogen, appropriate precautions against resistance development should be taken.  相似文献

A study was carried out to demonstrate that Rice yellow mottle virus (RYMV), a virus known to be transmitted by beetles, can spread between rice plants by direct leaf contact caused by wind. Almost all healthy plants surrounding an infected plant became infected when exposed to a fan blowing for 15 min at a distance of 50 cm. Spread of RYMV by plant contact, mediated by wind, was also demonstrated in field experiments, the extent of spread depending on plant density. Infection was almost 10 times higher in plots with a density of 33 plants m−2 than in plots with 16 plants m−2. Less spread was observed in plots protected by 1·5 m high windscreens. It is suggested that wind-mediated spread of RYMV may result from abrasive contact between leaves of plants.  相似文献
