Rice with low glutelin content is suitable as functional food for patients affected by kidney failure. Low glutelin-content gene Lgc1 in rice has a 3.5-kb deletion between two highly similar glutelin genes GluB4 and GluB5, which locates on the short arm of chromosome 2. To improve the selection efficiency in low glutelin-content rice breeding, two molecular markers designated as InDel-Lgc1-1 and InDel-Lgc1-2 were developed to detect the low glutelin-content gene Lgc1. A double PCR detection indicated that combined use of the two markers could easily distinguish the genotypes of Lgc1 from different rice varieties. Therefore, as a simple and low-cost technique, the molecular marker could be widely used to identify different varieties with Lgc1 gene and applied in marker-assisted selection of low glutelin-content rice.  相似文献

小麦高、低分子量麦谷蛋白亚基对品质性状的影响   总被引:2,自引:0,他引:2  
为给小麦品质改良提供理论依据,以6个国家的532个品种(系)为材料,分析了小麦高、低分子量麦谷蛋白亚基与品质性状的关系。结果表明,各位点上不同的等位变异对品质性状的效应存在显著差异,亚基1、2*、17+18、5+10、GluA3f、GluB3b和GluB3g对面团形成时间和稳定时间具有显著的正向效应;亚基N、1、14+15、17+18、7+8、2+12、3+12、4+12、GluB3d、GluB3f、GluB3h和GluB3g对蛋白质和湿面筋含量具有显著正向效应;从亚基对品质性状的综合影响来看,1、17+18、GluA3f、GluB3g和GluB3f可作为优势亚基。具有1、17+18、2+12和1、17+18、5+10高分子量麦谷蛋白亚基组合的品种(系)综合品质性状显著优于含有N、14+15、2+12亚基组合的品种(系);具有低分子量麦谷蛋白亚基组合GluA3c、GluB3g,GluA3d、GluB3g和GluA3f、GluB3b品种(系)的综合品质性状显著优于含有GluA3b、GluB3j亚基组合的品种(系)。  相似文献

The molecular weight (MW) distribution of proteins extracted with different solvents from defatted rice endosperm was examined by size exclusion-high performance liquid chromatography (SE-HPLC) with 2.0% sodium dodecyl sulfate (SDS) (w/v) as mobile phase. The resulting protein peaks were further characterized by SDS-PAGE. Under the experimental conditions, 2.0% SDS extracted 64% of the proteins. Adding 6.0 M urea resulted in a 15% increase in extractability (up to 79%). With using 20–100 mM NaOH, 70–81% of the proteins were extractable. Maximum extractability was reached with 2.0% SDS, 6.0 M urea and 0.5–1.5% dithiothreitol (DTT). Apparent MW profiles of rice endosperm proteins allowed classification into six fractions of decreasing apparent MW. Fraction VI contained the low MW albumin, globulin, and prolamin protein material. Fractions IV and V originated from α and β glutelin subunits, respectively. The polypeptides of fraction III consisted of an α and a β subunit linked by an intermolecular disulfide bond. The polypeptides of fractions I and II were dimers, trimers or more highly polymerized forms of the (α–β) glutelin subunit dimer in fraction III. While the work confirmed that rice glutelin is composed of polymers of α and β subunits, remarkably, higher MW glutelin aggregates (fractions I–III) only partly dissociated on reduction. Low MW protein material (fraction VI) was entrapped in the aggregated protein network and was released on reduction. The rapid and reproducible SE-HPLC method developed for rice protein separation allows a more quantitative approach than SDS-PAGE.  相似文献

应用水稻种子生物反应器开发口服重组胰岛素原具有重要应用前景。通过分子设计保证重组胰岛素原在人体肠道内的自主加工成熟,根据水稻密码子偏爱性人工合成了霍乱毒素β亚基和人胰岛素原的融合基因(cholera toxin B subunit fused with human proinsulin,CTBIN),并在C末端添加内质网滞留信号KDEL。通过PCR技术从粳稻品种日本晴全基因组中克隆谷蛋白启动子及其信号肽序列pGluB1sig(GluB1 promoter and its signal peptide)用于驱动融合基因CTBIN的表达,插入载体pCAMBIA1302,构建了水稻种子蛋白体靶向表达口服重组胰岛素原的载体pCAMBIA1302-pGluB1sig-CTBIN-Nos。采用农杆菌介导法转化日本晴,获得了46株转基因水稻植株,Western杂交检测到CTB-人胰岛素原融合蛋白在水稻种子中表达。  相似文献

1988年选用14个,1989年选用15个蛋白质含量差异较大的主要为江淮地区的大豆品种,进行随机区组试验。大豆种子中球蛋白、清蛋白、醇溶谷蛋白以及谷蛋白的品种间平均分别为28.56%、7.50%、1.81%和5.71%,品种间变幅分别为6.08%、10.25%、1.03%和6.51%。清蛋白含量与全蛋白含量。谷蛋白含量与全蛋白含量之间显著正相关,清蛋白含量对种子全蛋白含量品种间差异的相对贡献最大,而球蛋白含量对品种间全蛋白含量差异的相对贡献最小。球蛋白含量与株高、分枝数和单株粒重存在显著的正相关;但株高和清蛋白含量、单株荚数及单株粒数和谷蛋白含量有显著负相关。  相似文献

The Osborne protein fractions of seeds of 17 Lupinus albus cultivars from different regions in Portugal (obtained from plants grown at the same location) were analysed. The objective of this work was to examine the variability in the seed protein fractions of such a L. albus germplasm sample in order to disclose existing genetic relationships and to obtain information useful for a breeding program. Globulins and, interestingly, the fraction extracted with NaOH (“glutelins”) were the major seed protein constituents. The several protein fractions were separated by electrophoresis (reducing SDS polyacrylamide gel electrophoresis) and the patterns examined by multivariate analysis. Bands of “glutelins” together with those of glycosylated polypeptides were found to have the highest discriminating capacity. Lupin populations were grouped according to their clinal geographical distribution from north (small-seeded, higher plant architecture and later flowering type) to south (large-seeded, shorter and early flowering type). It is also possible that polypeptide patterns could reflect microclimatic specificities related with altitude and temperature. In fact, one cultivar from the south but from high altitude was grouped with the north plant types adapted to colder and wetter weather, whereas two cultivars from a warmer north region were grouped with the southern types. The results show, in addition, that a general correlation exists between cultivar early flowering characteristics and higher amounts of both total seed protein and glutelins, and lower glutelin glycosylation. Since high seed protein content and early flowering are important aims of lupin breeding programs, these findings on the seed protein fractions could stimulate and direct future research in lupin improvement.  相似文献

Seeds of Mucuna pruriens (L.) DC. (Fabaceae) were analyzed for protein composition of protein bodies isolated from cotyledons. Protein bodies were successfully separated by Lympholyte and those of dry seeds, observed by scanning electron microscope, were elliptical or spherical in shape with a diameter of 5–12 μm. Protein content in dry seed protein bodies was 10.6 mg/g dry weight. Globulin was the largest protein fraction isolated (62.5 %), followed by albumin (18.3 %), glutelin (15.8 %) and prolamin (3.4 %). The prolamin fraction and high glutelin content are uncommon in legumes. SDS-PAGE of albumins, globulins, prolamins and glutelins provided different band numbers and molecular weights under reducing and non reducing conditions and suggested that the albumin fraction is rich in disulphide bonds.  相似文献

Six chickpea strains were analysed for their protein content and various protein fractions. The protein content ranged from 20.9–25.27%. Albumin, globulin, prolamin and glutelin contents ranged from 8.39–12.31%; 53.44–60.29%; 3.12–6.89% and 19.38–24.40% respectively. Salt soluble proteins (albumin + globulin) and globulins resolved into 19–23 bands whereas albumin proteins resolved into 30–34 bands. The molecular weights of various polypeptides ranged from 10–91 kD. Amino acid analysis of total proteins revealed that glutamic acid was present in maximum concentration followed by aspartic acid and arginine. Just like other pulse proteins, chick pea proteins were also found deficient in sulphur containing amino acids.  相似文献

 用十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE)方法,对粳稻BT 细胞质雄性不育系六千辛A,保持系六千辛B,恢复系六千辛R、77302-1,以及杂交组合六千辛A/77302-1的F1和F2 种子的胚乳贮藏蛋白进行了分析。结果表明,在谷蛋白α3区域,恢复系有两条带α3a 和α3b,而六千辛A和六千辛B只有一条带α3。 F2代具有α3的种子和具有α3a 加α3b的种子1∶1分离。谷蛋白α4带的移动速率,恢复系比六千辛A快。把较快的α4带记为α4f。 F2代具有α4的种子和具有α4 加α4f的种子也是1∶1分离,与配子体不育类型的F2代花粉育性恢复基因分离比一致。系谱分析表明六千辛R中α3a 和α3b来源于IR8。六千辛A比六千辛B容易提取醇溶蛋白。  相似文献

The effect of 7 days of germination on levels of nitrogenous and other nutrition related parameters, protein fractions and in vitro protein digestibility of fluted pumpkin (Telfairia occidentalis) seed was studied. The non-protein nitrogen gradually increased and the protein nitrogen content decreased during germination. Albumin and globulin fractions were found to be the major seed proteins of fluted pumpkin seeds, constituting about 58.6% of the total protein of the ungerminated (raw) seeds. The protein fractions, albumin and glutelin, were observed to increase by 61.5% and 57.0%, respectively, while a 54.6% decrease was noted in the prolamine fraction. The globulin fraction increased at the beginning of germination but decreased at the end. Germination significantly (p 0.05) increased the crude protein, nitrogen solubility and in vitro protein digestibility but decreased the fat, phytic acid and polyphenol contents of the seeds.  相似文献

Albumin, globulin, glutelin and prolamin fractions were isolated from an Australian rice variety (cv. Langi) and characterised by yield, protein content and molecular weight profile using both capillary electrophoresis (SDS-CE) and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). The influence of pre-extraction enzymatic hydrolysis of starch and heating to 70 °C was also investigated, as was the extraction of the glutelin fraction without prior removal of the albumin and globulin fractions. Pre-extraction treatment affected mainly the albumin fraction, increasing dry matter yield but reducing protein content. SDS-CE was able to separate the protein fractions over a wider molecular weight range than SDS-PAGE, and the peaks from SDS-CE showed slightly higher molecular weight compared to equivalent bands from SDS-PAGE. The glutelin fraction extracted without prior removal of albumin and globulin fractions had different characteristics compared to those obtained by conventional extraction methods. Pre-extraction hydrolysis of starch did not significantly affect the emulsifying, foaming and gelling properties of extracted protein. Although rice glutelin had poor solubility, emulsifying and foaming properties in aqueous systems, it had good gelling properties which could be important for food applications.  相似文献

The percent distributions of protein fractions namely albumin + globulin, prolamine and glutelin were studied in developing grains of NP 113 barley and its high lysine mutant Notch-2. During development the percentage of albumin + globulin fraction decreased in NP 113, while those of prolamine and glutelin remained unchanged. The increase in prolamine was substantial from 24 to 31DAA. In Notch-2 the trend followed by albumin + globulin and prolamine was like that in NP 113, while the glutelin fraction showed an increase as compared to 10 DAA. The percent of albumin + globulin was slightly higher in Notch-2 as compared to NP 113. The absolute amount (mg/grain) of all the protein fractions increased during development in both NP 113 and its mutant Notch-2. During the grain development the prolamine content was substantially lower in the mutant than in the parent NP 113. The albumin + globulin content per endosperm was in general also higher in NP 113 than Notch-2. Amino acid analysis of the protein fractions did not reveal significant changes in lysine between NP 113 and Notch-2. Thus, the improvement in lysine in the mutant is primarily due to reduced synthesis of the prolamine fraction and not due to an increase in lysine in the mutant hordein fraction. Serotonin N-hydroxycinnamoyltransferase (SHT) is a key enzyme in the synthesis of feruloylserotonin (FS) and 4-coumaroylserotonin (CS). These serotonin derivatives show strong antioxidant activity, making them valuable for both nutritional and pharmacological use in humans. Ectopic expression of SHT under the control of the endosperm specific-glutelin and prolamin promoters from rice was produced via Agrobacterium-mediated transformation. SHT expression was confirmed by Southern blot analysis, followed by Northern blotting and SHT enzyme activity analyses using total RNA and protein, respectively, extracted from transgenic seeds. The glutelin A3 (GluA3) promoter produced low SHT mRNA expression in rice seeds, whereas the prolamin promoter expressed high levels of SHT mRNA. In spite of the ectopic expression of SHT in rice seeds, both transgenic genotypes accumulated levels of serotonin derivatives similar to those found in wild-type rice. Furthermore, our data suggest that serotonin, rather than phenylpropanid-CoAs, is the rate-limiting substrate in the biosynthesis of serotonin derivatives in SHT-overexpressing transgenic rice seeds.  相似文献

选用我国江浙地区近期育成的晚粳稻品种(系)29个,对它们在浙江杭州相同栽培条件下籽粒中的植酸、蛋白质含量与其组分差异及其相关性进行了分析。供试29个品种(系)植酸含量的平均值为0.868%,变化幅度为0.699%~1034%,其中秀水系列品种的植酸含量一般较低,而武育粳系列和淮字系列品种(系)的植酸含量则相对较高;供试29个品种材料总蛋白质含量的平均值为8.722%,其含量大致呈正态分布。在4种蛋白质组分中,谷蛋白、球蛋白和清蛋白在品种间的变异系数较大,而醇溶蛋白则相对较小,且在分布频率上也表现出品种类型间的差异特征;除谷蛋白与总蛋白质之间存在着极显著的正相关外,供试29个品种(系)的植酸含量与蛋白质含量及4种蛋白质组分含量间并无密切的关系。  相似文献

The starch and protein properties of rice grain are important factors for sake brewing and these properties are reported to be influenced by temperatures during grain ripening. Amylose content, nitrogen content, protein composition, pasting properties measured by a Rapid Visco Analyser (RVA), and their relationship to temperatures during ripening were investigated in a rice cultivar, Yamadanishiki, which was grown under various conditions in the same experimental field. The average temperature after heading was significantly correlated with amylose content and RVA properties, but not with nitrogen content. Under high temperatures during ripening, a decrease in prolamin, which accumulated in type I protein body (PB-I), and an increase in glutelin, which deposits in type II PB (PB-II), were recognized. The ratio of PB-II/PB-I and RVA pasting temperatures were distinctly increased as the temperature became higher. High temperatures during grain ripening would lead to difficulties in digesting steamed rice grains by Aspergillus oryzae, together with ease in digesting rice protein. The average temperature of 11–20 days after heading showed a higher correlation coefficient than that of 1–10 or 21–30 days, implying that temperatures during the middle stage of grain development would be important in determining the rice component that relates to brewing properties.  相似文献

Seed nitrogen content, protein composition and free amino acid content were determined in rice mutant lines with altered storage protein composition and their parental cultivars. The Lgc1 gene, which causes low glutelin content and high content of 13 kDa prolamin and 26 kDa globulin, did not affect grain weight, nitrogen content, or free amino acid content. The glb1 gene, which lacks the 26 kDa globulin, did not affect grain weight or nitrogen content, but in mutant lines with glb1 gene the content of the major free amino acids was significantly (1.3–1.5 times) higher than those of their parental cultivars. These results suggest that absence of 26 kDa globulin is accompanied by an accumulation of a high level of free amino acids in rice grains.  相似文献

The indigestible remains of rice (Oryza sativaL.) found in faeces of monogastric animals (faecal protein particles) appear as isolated, refractive spheres 1–2 μm in diameter. Still controversial, the prolamin-containing PB-I rice protein body has generally been proposed as the progenitor. We used polyclonal antibodies to purified fractions of rice prolamins and glutelins, to total rice protein, and to enzymatically- and chemically-derived waste particles of rice. Three classes of residual particles were examined immunocytochemically using Protein A-gold procedures to determine the polypeptide types present, and thus their possible origin. All particles contained antigenic determinants for all antibodies to the extracted rice polypeptides studied. There was also evidence of protein(s) peculiar to the residual bodies; the label on intact protein bodies showed a small amount of the unique protein(s), but label increased for all residual particles studied. In contrast, antigenic determinants for total rice protein were much reduced in residual particles. While measurement of profiles of intact PB-I and faecal protein particles shows no significant difference between their diameters, the immunocytochemical evidence linking faecal protein particles exclusively with PB-I rice protein bodies is less clear because intact PB-I and PB-II both show evidence of the unique protein also found in the residual particles.  相似文献

Rice flour and commercial rice protein concentrate (enzymatically destarched rice flour) from mature rice grains (Oryza sativaL., cv. Lemont) were embedded and sectioned for use in electron-microscopical immunocytochemistry. Polyclonal antibodies were raised in rabbits against the following purified rice protein fractions: 10 and 13 kDa prolamins, 23–25 and 38 kDa glutelins, ‘total prolamins’, ‘total glutelins’, and two insoluble particulate waste products derived from protein concentrate. Western blot procedures showed that the antibodies produced were specific for their particular fractions, and that antibodies to particulate waste fractions reacted with bands at positions of approximately 12 and 28 kDa, normally not evident as strong bands in SDS-PAGE gel preparations of rice. Immunogold procedures, using Protein A and electron microscopy, significantly labelled both types of rice protein body (PB-I and PB-II) in the embedded substrates for each antibody. This varies from other reported research when rice proteins have been fractionated on gels, or when antibodies to total glutelins or total prolamins have been tested on developing rice grains. Estimates of labelling efficiencies show that each protein body type contains a substantial number of antigenic determinants for all the species of polypeptide studied.  相似文献
