In vitro Culture of Cucumis sativus   总被引:1,自引:0,他引:1  
The variability of morphological traits in 1800 individuals of R2 generation of Cucumis sativus was studied. R2 generation was obtained from 60 R1 planes which regenerated from leaf explants of a highly inbred Line of cv. Borszczagowski. All the plants of R1 population were phenotypically identical with the initial variety; some (8,3 %) of the R2 progeny, however, exhibited segregations. Variability was manifested in the following traits: 1. type of growth (dwarf and bushy), 2, male sterility, 3, mosaic type leaves, 4, xantha type chlorophyll change. In R3 generation, only one new phenotype (yellow fruit flesh) has been found. The traits mentioned above were frequently accompanied by lower viability and fertility of R2 plants. The latter was connected with disturbances in microsporogenesis and lover pollen germination ability. Genic instability of one trait is described.  相似文献

Sixteen inbred lines and one hybrid of manse were tested for their capability of somatic embryogenesis, and fully developed plants could be regenerated, from ten inbred, lines. The highest frequency of plant regeneration was expressed in the inbred line CHI 31, and when this line was crossed with a recalcitrant, non-regenerating line, the F1 and BC hybrids were regenerable. The results of reciprocal crosses demonstrated that dominant nuclear genes and cytoplasmic factors are primarily responsible for the heritable determination of embryogenic callus proliferation and in vitro regeneration of maize plants. Somaclonal and radiation-induced variability was studied in maize to assess their nature and potential contribution to plant breeding., The inbred line CHI 31 possessing a high in vitro capacity of somatic embryo formation was used as experiments.] material. CHI 31 plants were selfed and twelve-day old zygotic embryos irradiated with 60Co gamma radiation in situ. Mature caryopses were harvested and assigned as M1 material. In another series, immature zygotic embryos (size 1.2—1.5 mm) were cultured in vitro on N-6 medium supplemented with 2,4-D (2.5 μM), and somatic embryos regenerated into plants; these were transplanted into soil and self-pollinated. Regenerants from non-irradiated cultures were grown as R1 generation, while regenerants from irradiated explants were considered as M1R1 generation. The genetic variability was evaluated in the M2, R2 and M2R2 generations, respectively, and compared with a non-treated seed control. Irradiation induced a variety of chlorophyll and morphological variants segregating in the M; generation; however, the frequency of deviant types obtained in the R: generation (somaclonal variation) was significantly exceeding the one derived from the M2 populations. The combination of expert irradiation and in vitro regeneration was most effective for the manifestation of chlorophyll and morphological o if types in the M2R2 generation, and increased drastically the frequency of early flowering variants. Differences in the segregation patterns of mutant phenotypes amonsister somaclones in the R3 and M3R3 generations indicate a different genetic basis, of plants originating from the same explant. This phenomenon suggests a mutational sectoring of the callus during culture. Radiation induced and somaclonal variation exerted a similar spectrum of chlorophyll and morphological deviants.  相似文献

Different wheat genotypes were treated with gamma-rays, sodium azide (SA) and EMS before tissue culture and immature embryos from M1 plants or plants shortly after exposure to gamma-rays were used to initiate callus culture. Thousands of plants were regenerated and used to investigate the effect of mutagenic treatments on the regenerated plants and somaclonal variation in the M3R2 and M2R2 generations. The results showed that mutagen-induced damage in terms of reduction in plant height, fertility and spike length were not outstanding in the regenerated plants as compared with the untreated control. In the M3R2 generation, only SA treatment had significantly higher frequencies of somaclonal variations than the control. Increases in the variation frequencies were observed when explant embryos were irradiated with 2.5 and 5 gy gamma-rays and the highest frequency appeared when embryos were exposed to 5 gy gamma-rays on the 5th day after anthesis. Increased variation spectra also resulted from mutagenic treatments and most of the variants recovered were unsuitable for plant improvement.  相似文献

The inheritance and expression of the reporter gene uidA, encoding β-glucuronidase (GUS), was previously analysed in the T1 generation of 25 independent transformed lines of Lotus corniculatus cv. Leo. In the work reported here, GUS activity in various tissues of seven of these lines was tested in the T2 generation. Four representative lines were chosen for more detailed study in the T3 generation. Lines 25 and 38 had multiple, independently segregating transgene inserts; lines 24 and 39 appeared to transmit one segregating transgene insert to their T1 progeny, although transgene expression was low and was detected in fewer seedlings than expected in line 39. The uidA gene was inherited and expressed in seedlings of T1, T2 and T3 generations of all four lines. In all lines, transgene expression varied between tissues, with more embryos than seedlings having detectable GUS activity. Studies in the T2 generation showed that use of transgenic plants as female or male parents altered the frequency of expression of the transgene in progeny. By contrast, in the T3 generation the use of transgenic plants as female or male parents did not effect either frequency of transmission, or expression of the transgene, in any of the four lines. Transgene inheritance was also similar among individual pods within flower heads and between individual flower heads. The morphological and genetic variations in somaclones of chili pepper (Capsicumannuum L.) derived from tissue culture were evaluated. Cotyledonary node explants of cultivars, Shishitou and Takanotsume, were cultured on Murashige and Skoog (MS) medium supplemented with benzyladenine (BA)5 mg/l for shoots regeneration and regenerated shoots were rooted on MS medium supplemented with naphthalene acetic acid(NAA) 0.1 mg/l and indol-3-butyric acid(IBA) 0.05 mg/l. The regenerated plants(R0) were selfed to obtain seeds for next generation (R1 lines). Qualitative characters were studied in R0generation and both qualitative and quantitative characters were studied in R1 generation. In R0 generation, variations were noticed in plant growth habit, stem color, flower color and color of unripe fruits, and expression of anthocyanin in unripe fruits. Comparison among the R1 lines and their parents were made for morphological and agronomic characters. Significant variation among R1 lines and differences between R1 lines and their parents were observed. Genetic variations among three somaclones were revealed by random amplified polymorphic DNA (RAPD) analysis. Variation, such as early flowering and increase of yield components, is an indication of the response to selection for any specific character. Occurrence of productive variants among somaclones of established cultivars, like Shishitou and Takanotsume, indicates the possibility of their improvement through somaclonal variation. This report describes a study of androgenesis in Dactylis glomerata, where the main aim was to find anther culture-responsive clones. Two types of media and two sugars were compared for their effectiveness in anther culture induction and subsequent plantlet production. Embryo formation from the cultured anthers was obtained from 28 of the 108 cloned genotypes using two different substrates, R2M and FW. Both induction media supported the formation of embryos from the cultured anthers, but around 4.5 times more embryos (0.81 embryos per 100 anthers) were obtained with R2M compared with FW, and R2M also gave 5.5 times more green plants (0.054 green plants per 100 anthers) than the FW substrate. In the investigation of a carbohydrate source, responsive clones from the genotype study were tested using maltose as a substitute for sucrose in R2M. Using maltose instead of sucrose increased embryo formation so that 133 embryos per 100 anthers were obtained compared with 7.1 embryos per 100 anthers obtained with sucrose. The total number of green plants obtained was also improved with maltose compared with sucrose, resulting in 66.3 and 1.9 green plants per 100 cultured anthers, respectively.  相似文献

Summary Multiple shoot formation in cotyledonary callus of Indian mustard (Brassica juncea cv. Prakash) was induced on modified MS media supplemented with high cytokinin (kinetin or zeatin) and low IAA concentrations. Complete plants were obtained on prolonged incubation of shoots on the same medium. 6-Benzyladenine alone or in combination with IAA or NAA did not support plantlet regeneration. A total of 71 plants were transferred to greenhouse. The seed, however, could be collected from 37 plants only. The seed was sown in the field to evaluate the material for somaclonal variation in R1 generation. Data were recorded for yield, plant height, number of primary branches, siliqua number, 1,000 seed weight and oil content. Somaclonal lines showed tremendous amount of variation for all the characters studied. A number of plants in this generation showed significantly higher yield and/or other improved agriculturally important characteristics as compared to the control. A line with dwarf plant type was also identified. A number of plants were selected from this generation and carried forward to R2 generation. Most of these lines bred true in R2 generation. The material seems to be very promising for future breeding programmes.  相似文献

To clarify the effect of the type of in vitro culture on the generation of somaclonal variation, protoplast-derived rice plants were compared with rice plants derived from suspension culture or primary calli (not derived from protoplasts). Regenerated plants showing polyploid-like phenotypes appeared at a higher frequency (33–70%) in plants derived from protoplasts than in those not derived from protoplasts (3–6%). In the first progeny (R1) generation of all regenerated plants, 120 of 368 lines (33%) segregated plants with mutated characters such as albino, dwarf and sterile. In quantitative traits, 62 (21%) and 144 (50%) of 290 Rj lines showed shorter culm and lower seed-fertility, respectively, compared with the control line derived from the selfed seeds of the original cultivar. The frequency of the mutant-possessing R1 lines among lines derived from protoplasts was not significantly different from those not derived from protoplasts. These results indicate that isolation and culture of protoplasts does not enhance genetic changes other than cytogenetical changes.  相似文献

Summary The phenotypes of seed progeny (R2 generation) of Oryza sativa L. cv. Taipei 309, which carried the neomycin phosphotransferase II (npt II) gene, were compared with those of non-transformed, protoplast-derived plants of the same generation and non-transformed, seed-derived plants under field and glasshouse conditions. Under both conditions the transgenic plants were generally smaller, took longer to flower and had reduced fertility. Significant differences were observed between individuals within the group of transgenic plants. The npt II gene was present in most of the transgenic plants, but NPT II activity was only detected in a minority of individuals.  相似文献

Somaclonal variation is a possible source of variation in plant breeding. To apply this approach to eggplant breeding, somaclonal variations were observed among plants regenerated through somatic embryogenesis induced by 1-naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D). Variations including leaf shape, plant height, flower number per cluster, fruit shape, anther number per flower and pollen fertility were compared among ? 300 plants (R0). Although these variations were confirmed among plants regenerated using both auxins, the frequencies of somaclonal variations in leaf shape, plant height, fruit shape and pollen fertility in the NAA experiment were higher than those in the 2,4-D experiment. Variations in flower number and anther number were also confirmed among plants from both experiments, although no significant differences in their frequency were observed. Subsequently, the inheritance of variations (leaf shape, fruit shape, and flower number) observed in R0 was investigated from generation to generation (R1). Variations in leaf shape and fruit shape were inherited while those in flower number were not. From these results we concluded that, although a high frequency of somaclonal variations were observed among plants from both methods, embryogenesis with NAA was more efficient than 2,4-D in eggplant.  相似文献

Data on the attack of the tubers of 2,036 F1 seedlings of 32 different cross combinations of potato varieties were collected. A survey is given of the field resistance of a large number of established varieties and of their parents.From both sets of data it appears that crossing two highly resistant parents results in a high percentage of resistant offspring. When crossing a susceptible with a resistant parent one gets a majority of resistant F1 seedlings and even among the offspring of two susceptible varieties a few resistant seedlings are encountered.The genes R 1 and R 3 for hypersensivity of the leaves exert a strong influence. Preliminary data suggest that the cambial layer just below the skin in this case is a strong barrier to the penetration of the parasite as is stated also with some varieties devoid of R-genes (Bravo, Libertas and others).  相似文献

Summary Inheritance studies on the stem termination in pigeon pea using F1, F2 and F3 generations of two crosses between determinate and indeterminate lines suggested that two dominant genes with epistatic (inhibitory) interaction of one of them control the interminate growth habit. The gene symbols D. idid and ddIdId have been designated to the parental plants with determinate and indeterminate growth habits, respectively. The gene IdId was epistatic (inhibitory) to the gene D giving a ratio of 13 indeterminate: 3 determinate inthe F2's observed. Summary Progenies of crosses of the following genetical types were inoculated with and selected for immunity from the races 1, 2, 4 and 2, 3, 4 of Phytophthora infestans, viz. (dominant symbols only) R1×R3, R1R3×r, R1R3×1, R1R3×R1, R3×R1R2, R3×R1R3, r×R1R3, R1R3×R1R2, R1R3×R1R2 and R1R3S.P. (spontaneously pollinated). One R3×R1R3 progeny, two R1R3×R1R3 progenies and six (out of ten) S.P. progenies segregated with a larger or smaller deficit of immune seedlings as was expected according to the hypotheses that were postulated before and which appeared to be in agreement with the observed segregations in the other progenies. Agrobacterium tumefaciens mediated in planta transformation protocol was developed for castor, Ricinus communis. Two-day-old seedlings were infected with Agrobacterium strain EHA105/pBinBt8 harboring cry1AcF and established in the greenhouse. Screening the T1 generation seedlings on 300 mg L−1 kanamycin identified the putative transformants. Molecular and expression analysis confirmed the transgenic nature and identified high-expressing plants. Western blot analysis confirmed the co-integration of the nptII gene in the selected transgenic plants. Bioassay against Spodoptera litura corroborated with high expression and identified five promising effective lines. Analysis of the T2 generation plants proved the stability of the transgene indicating the feasibility of the method.  相似文献

The aim of this work was to obtain a series of genetically related lines of Eragrostis curvula with different ploidy levels and reproductive modes, which can be used for the discovery of genes associated to diplospory. E. curvula is a widely cultivated forage grass native to Southern Africa that reproduces naturally by obligate diplosporous apomixis (asexual reproduction by seeds) of the Antennaria type. In vitro culture of immature inflorescences of the apomictic cultivar Tanganyika led to the regeneration of a diploid plant with a particular morphology (plant UNST1122). Embryo sac analysis and progeny tests determined that plant UNST1122 reproduces by sexuality. UNST1122 was cloned by dividing its tillers and a R 1 generation was obtained from seeds after open pollination among the diploid clones. A set of 500 seeds from a diploid R 1 plant was treated with colchicine and two plants showing a duplicated chromosome number (2n = 4x = 40) were obtained (plants UNST1112 and UNST1131). Progeny tests using RAPDs indicated that these plants reproduce also by sexuality. The availability of sexual tetraploid genotypes of E. curvula will allow the production of hybrids with new interesting combinations of agronomic traits and the developing of mapping populations segregating for diplospory. Besides, these genetically related plants with different reproduction modes constitutes an excellent model for the identification of gen(es) involved in diplosporous apomixis by mRNA profiling as well as to study the genomic rearrangements and gene expression alterations associated to changes at ploidy levels.  相似文献

Somaclonal variation for plant height, plant spread, leaf shape, leaf size, leaf form, herb yield, essential oil content and 10 important constituents of the essential oil was studied in an Indian cultivar, ‘Bourbon’ of rose‐scented geranium (Pelargonium graveolens). Significantly larger variance was observed among in vitro‐regenerated plants of the SC1 generation (first generation following an in vitro phase) than among parental plants raised from stem cuttings for herb yield, plant height, leaf size, essential oil content, and for the contents of cis‐rose oxide, trans‐rose oxide, isomenthone and 10‐epi‐γ‐eudesmol in the essential oil. Plants of the SC1 generation showing values beyond the higher or lower limit (depending on the trait) of the range observed among parental plants for different traits were developed into clones by their multiplication through stem cuttings, and evaluated again in the SC2 generation (second generation following the in vitro phase). Somaclones selected for plant spread, and for the contents of linalool and trans‐rose oxide in their essential oils did not breed true in the SC2 generation. However, for the remaining traits studied, 13–100% of the selected clones retained their selected traits in the SC2 generation, suggesting that the changes in these somaclones were due to genetic causes.  相似文献

The cytogenetic study was investigated in the intergeneric F1 hybrid, F2and backcross progenies (BC1). The plants used were Brassica juncea(2n=36) and Diplotaxis virgata(2n=18). Three intergeneric F1 hybrids between two species were produced through ovary culture. They showed 36 chromosomes. It might consist one genome of B. juncea and two genomes of D. virgata. The morphology of the leaves resembled that of B. juncea. The color of the petals was yellow that was like in D. virgata. The size of the petal was similar to that of B. juncea. The mean pollen fertility was15.3% and the chromosome associations in the first meiotic division were(0–1)IV+(0–2)III+(8–12)II+(12–20)I. Many F2 and BC1seeds were harvested after open pollination and backcross of the F1 hybrids withB. juncea, respectively. The F2seedlings showed different chromosome constitutions and the range was from 28 to54 chromosomes. Most seedlings had 38chromosomes followed by 36, 40 and 54. The BC1 seedlings also showed different chromosome constitutions and the range was from 29 to 62. Most seedlings had both 40and 54 chromosomes followed by 36, 46 and52. In the first meiotic division of F2 and BC1 plants, a high frequency of bivalent associations was observed in all the various kinds of somatic chromosomes. Many F3 and BC2 seeds were obtained by self-pollination and open pollination of both F2 and BC1 plants, and by backcrossing both F2 and BC1plants with B. juncea, respectively,especially, three type progeny with 36, 40or 54 chromosomes. The somatic chromosomes of the F3 and BC2 plants were further investigated. The bridge plants between B. juncea and D. virgata with 36 chromosomes may be utilized for breeding of other Brassica crops as well as B. juncea. Brassica napus somatic hybrids with low linolenic acid (18:3) content in their seed oil have been produced using fusion partners screened for low 18:3. One somatic hybrid contained only 3.5% 18:3, a level significantly below the mid-parental mean. The low level of 18:3 proved stable in the R1 generation. Oil content of the lowest 18:3 selection increased from the mid-parental mean (29.3%) in the R0 generation to 36% in a R1 field bulk. The effectiveness of early generation selection under low and high plant density was evaluated in faba bean (Vicia faba L.). For this, the faba bean cv. 'Polycarpe' was pollinated by eight higher yielding cultivars of different origin. The F1's and F2's together with 'Polycarpe' and the pollinator with the highest yielding ability (cv. 'Effe') were evaluated under high plant density with adjacent controls and under low plant density in a honeycomb design. It was found that the eight hybrids had a different ranking at low than at high plant density for both generations (F1and F2). Hybrid Pol.x A-58 had the best combined yield performance in the F1and F2under high and hybrid Pol.x A-37 under low plant density. Application of individual plant selection in the F2and F3generation of the two crosses (Pol.x A-58 and Pol.x A-37), under both low plant density according to UNR-O honeycomb design and high plant density using a moving average, resulted in the selection of four groups with 20 selected F3plants each. Within each group, equal numbers of seeds from each plant were mixed to establish four mixtures. These mixtures together with two controls ('Polycarpe' and 'Effe') were evaluated in a randomized complete block design (22 plants/m2) in one location for two years. It was found that the material selected continuously under low plant density had a significantly higher yield than the material selected under high plant density. In addition, the material selected from Pol.x A-37 (H1R2) under high plant density had a lower mean yield than that selected continuously under low plant density (H1H2). Their average yield over years was significantly higher than the yield of the material selected from Pol.x A-58 under both plant densities. Furthermore, the material selected from Pol.x A-58 under low plant density, had a significantly higher mean yield over years than its sibs selected under high plant density. It was concluded that honeycomb selection at low plant density can be used effectively for early generation selection in faba bean.  相似文献

In vitro regenerated sunflower (Helianthus annuus L.) plants (R1) were self-fertilized and the R2 generation was evaluated for qualitative traits. A broad range of phenotypic variation was observed and mutation frequencies were calculated. Some in vitro induced variant phenotypes were similar to known spontaneous or induced mutations in sunflower, while others were new. Chlorophyll and carotenoid deficiencies, chimaerical variegation, fasciated stem and capitulum, abnormal shoot development, and other morphological variations, were noted. Substitution of anthers with petaloid structures in a disk-floret mutant indicates a possible homeotic mutation induced by in vitro tissue culture.  相似文献
