Early‐stage mortality due to surface water tension‐related death and due to sinking to the tank bottom was investigated for yellowfin tuna, Thunnus albacares (YFT), larvae. Different aeration rates and rearing water surface conditions were examined to evaluate the effect on larval survival, swim bladder inflation and growth. The percentage survival of yolk sac larvae was significantly higher when the rearing water surface was covered with fish oil at aeration rates of 0 and 50 mL min?1. The highest mortality occurred at the highest aeration rate of 250 mL min?1 regardless of surface water condition. A second experiment was conducted twice under different water surface conditions: the water surface was covered by fish oil (FO), skimmed of fish oil (SS), and was not treated (NC). The percentage survival was not significantly different between treatments after 7 days of feeding. In contrast to the survival, the proportion of larvae with inflated swim bladders was significantly higher for the NC and SS groups than that of the FO group. Results of these experiments indicate that the addition of oil to the rearing water surface without its removal interferes with the initial swim bladder inflation in YFT larvae. These results also indicate that YFT larvae need to obtain (gulp) air at the water surface for initial swim bladder inflation, and success of initial swim bladder inflation may be crucial for their survival.  相似文献

Success of swim bladder inflation (SBI) is crucial for early survival of Pacific bluefin tuna (PBF) larvae, because it reduces larval sinking death by enhancing buoyancy. In Experiment 1, we examined the effect of photoperiod on SBI and survival in PBF larvae by comparing photoperiods of 9L: 15D (9L), 14L: 10D (14L: natural photoperiod), 19L: 5D (19L) and 24L: 0D (24L) during 2–10 days post hatch (dph). In Experiment 2, the combined effects of photoperiod (24L and 14L) and nighttime aeration rate (enhanced night‐time aeration: ENA of 1300 mL min?1 as a countermeasure for sinking death and 130 mL min?1) on the survival and SBI were also examined during 2–10 dph. Moreover, in Experiment 3 the effect of photoperiod on vertical distribution of larvae in night‐time was examined on 3–5 dph. Photoperiod of 24L in Experiment 1 significantly inhibited SBI compared with 14L and 19L; nevertheless, it significantly improved survival compared with other photoperiods with a dark period. On the other hand, the shortened light period (9L) showed significantly reduced SBI and also survival. In Experiment 2, the countermeasure for sinking death of ENA under 24L did not further improve the survival; rather it tended to reduce the survival. In Experiment 3, larvae distributed less in the bottom layer in 24L than in 14L, suggesting the reducing effect of 24L on sinking death. The results indicate that 24L without ENA is suitable for survival which is the most serious problem in PBF larviculture.  相似文献

In mass culture of Pacific bluefin tuna Thunnus orientalis, a marked growth variation is observed after they start feeding at 6–7 mm in body length (BL) on yolk‐sac larvae of other species, and the growth variation in tuna larvae is a factor leading to the prevalence of cannibalism. To examine the relationship between prey utilization and growth variation, nitrogen stable isotope ratios (δ15N) of individual larvae were analysed. A prey switch experiment was conducted under two different feeding regimes: a group fed rotifers (rotifer fed group), and a group fed yolk‐sac larvae of spangled emperor, Lethrinus nebulosus (fish fed group) from 15 days after hatching (6.87 mm BL). The fish fed group showed significantly higher growth than the rotifer fed group. Changes in the δ15N of the fish fed group were expressed as an exponential model and showed different patterns from those of the rotifer fed group. The δ15N of fast‐growing tuna larvae collected in an actual mass culture tank after the feeding of yolk‐sac larvae was significantly higher than those of the slow‐growing larvae, indicating that slow glowing larvae depended largely on rotifers rather than the yolk‐sac larvae.  相似文献

In mass culture of Pacific bluefin tuna Thunnus orientalis, yolk‐sac larvae of other species are fed as a major prey item to tuna larvae from 7 to 8 mm in total length. Marked growth variations in tuna larvae are frequently observed after feeding of yolk‐sac larvae, and this variation in the growth of tuna larvae is subsequently a factor leading to the prevalence of cannibalistic attacks. To elucidate details of the mortality process of hatchery‐reared tuna larvae after the initiation of yolk‐sac larvae feeding, we compared the nutritional and growth histories of the surviving (live) tuna larvae to those of the dead fish, found dead on the bottom of the tank, as direct evidence of their mortality processes. Cause of mortality of tuna larvae 3 and 5 days after the initiation of feeding of yolk‐sac larvae was assessed from nitrogen stable isotope and otolith microstructure analyses. Stable isotope analysis revealed that the live fish rapidly utilized prey fish larvae, but the dead fish had depended more on rotifers relative to the live fish 3 and 5 days after the initiation of feeding of yolk‐sac larvae. The growth histories based on otolith increments were compared between the live and dead tuna larvae and indicated that the live fish showed significantly faster growth histories than dead fish. Our results suggest that fast‐growing larvae at the onset of piscivory could survive in the mass culture tank of Pacific bluefin tuna and were characterized by growth‐selective mortality.  相似文献

This study investigated the effect of feedings taurine‐enriched rotifers on the growth and development of larval red sea bream (RSB). Rotifers incubated in taurine‐enriched water at a taurine concentration of 800 mg L?1 (T‐800) and 0 mg L?1 (T‐0) were fed to larvae from 3 to 20 days after hatching (DAH). Notochord length, body weight and specific growth rate of T‐800 group were significantly greater than those of T‐0 at 14, 17, 9–11 and 18–20 DAH. Taurine content of larvae in the T‐800 group increased rapidly from 11 DAH and thereafter remained significantly higher than T‐0. Flexion larvae firstly appeared in both groups at 8 DAH, however, at 20 DAH post‐flexion larvae were significantly more abundant in T‐800 than T‐0. While nucleic acid and protein contents (μg mg?1 wet fish) showed remarkable changes, ontogenetic growth in RSB larvae stage was observed to switch from hyperplastic growth to hypertrophic growth with the start of the flexion stage. Although a similar change in nucleic acid contents was observed between the two groups, the protein content (μg fish?1) and protein/DNA ratio of T‐800 remained higher than that of T‐0 during the hypertrophic growth period. These results suggest that dietary taurine accelerates the growth and development in RSB larvae especially during hypertrophic growth (flexion stage) after the early hyperplastic growth.  相似文献

Gustatory neural responses of the Pacific bluefin tuna Thunnus orientalis (Temminck and Schlegel) to extracted compounds of prey organisms, such as amino acids, nucleotide‐related substances, organic acids and organic bases, were electrophysiologically recorded from the facial nerve supplying the anterior palate. Of the 17 amino acids tested, l ‐proline was the most potent amino acid at 10?2 M, and its threshold was the lowest at around 10?6 M. l ‐leucine, l ‐methionine, l ‐alanine, l ‐valine and l ‐isoleucine were also highly stimulatory at 10?2 M; however, the other 11 amino acids examined were not as effective or were ineffective. Thus, the gustatory receptors for amino acids of the Pacific bluefin tuna show a narrowly tuned response profile. Among the seven nucleotide‐related substances tested, uridine‐5′‐monophosphate, inosine‐5′‐monophosphate and adenosine‐5′‐diphosphate were highly stimulatory, and their thresholds were 10?4–10?5 M. Inosine elicited a positive response at 10?2 M but its response magnitude was not so high. Organic acids l ‐lactic and pyruvic acids were effective at 10?2 M, but no response was elicited at 10?3 M. Among organic bases, betaine was highly stimulatory, and its response magnitude at 10?2 M is almost equal to that of l ‐proline at the same concentration. The threshold for betaine was determined to be at around 10?5 M. Trimethylamine oxide and ammonium chloride were ineffective.  相似文献

High levels of mortality occur in large net‐cage culture of Pacific bluefin tuna (PBF) Thunnus orientalis due to poor growth on commencement of sea‐cage culture obstructing the mass production of fingerlings. Therefore, we carried out this study to develop a countermeasure by using small sea cages (square with 13‐m sides). PBF juveniles were directly transferred to a 30‐m‐diameter cage (control) and compared them to fish transferred to small sea cages for 12 days before being merged into another 30‐m‐diameter cage. The survival rate, growth performance and potential factors increasing mortality were examined. The results of our study showed that survival rate in small sea cages was approximately 20% higher than that of the control. Poor growth also occurred in the small sea cages; however, its frequency and daily mortality rate were half those in the control. In addition, growth performance such as specific growth rate and weight gain of PBF juveniles in small sea cages significantly increased compared to the control. These results suggest that using small sea cages could be an effective countermeasure for poor growth, which may mitigate the high‐mortality conditions of current sea‐cage culture systems for PBF.  相似文献

This study was conducted to evaluate the effects of dietary taurine on growth performance and feed utilization of Nile tilapia (Oreochromis niloticus) larvae. Four plant protein‐based, isonitrogenous (400 g kg?1 protein), isoenergetic (19 MJ kg?1) diets supplemented with four taurine concentrations (0.0, 5.0, 10.0 and 15.0 g kg?1; designated as T0, T0.5, T1 and T1.5, respectively) were prepared. The diets were fed to triplicate groups of fish larvae (0.024 g average body weight), to apparent satiation, three times per day for 60 days. Larval growth rates and feed utilization efficiency were significantly improved with increasing supplemental taurine up to 10 g kg?1 and decreased with further taurine supplementation. The quadratic regression analyses indicated that the maximum larval performance occurred at about 9.7 g kg?1 of total dietary taurine. Fish survival was significantly lower at 15 g kg?1 dietary taurine than at other taurine levels. Body protein significantly increased, while body moisture and ash decreased, with increasing dietary taurine up to 10 g kg?1 and decreased with further taurine supplementation to 15 g kg?1. Body lipid was not significantly affected by dietary taurine concentration. A number of body amino acids (tryptophan, arginine, histidine, leucine, isoleucine, valine, alanine, glycine, threonine and taurine) significantly increased with increasing supplemental taurine up to 10 g kg?1 and then decreased with further increase in dietary taurine levels. The rest of body amino acids were not significantly affected by dietary taurine. The present results suggest that about 9.7 g kg?1 dietary taurine is required for optimum performance of Nile tilapia larvae fed soybean meal‐based diets.  相似文献

This study was carried out to investigate the suitability of Artemia enriched with docosahexaenoic acid (DHA) and choline as live food on the growth and survival rate of the Pacific bluefin tuna (PBT; Thunnus orientalis) larvae. The PBT larvae were fed either Artemia enriched with oleic acid (Diet 1), DHA (Diet 2), DHA+choline 1.0 mg L?1 (Diet 3) and DHA+choline 2.0 mg L?1 (Diet 4) or striped knifejaw larvae (Diet 5, reference diet), in duplicate for 12 days. Enrichment of Artemia with DHA significantly increased the DHA levels to 13.9, 13.8 and 12.5 mg g?1 on a dry matter basis in Diets 2, 3 and 4 respectively; however, the levels were significantly lower than the reference diet (26.9 mg g?1 dry matter basis; Diet 5). Although growth and survival rate were significantly improved by the enrichment of Artemia with DHA and choline, the improvement was negligible compared with the enhanced growth and survival rate of the fish larvae‐fed group (P<0.05). The results demonstrated that enriched Artemia does not seem to be the right choice to feed the PBT larvae perhaps because of the difficulties in achieving the correct balance of fatty acid with higher DHA/EPA from Artemia nauplii.  相似文献

Initial swimbladder inflation (ISI) of Pacific bluefin tuna (PBT), Thunnus orientalis, larvae was studied to increase the survival of cultured larvae. Experiment 1 was conducted to explore promotion and inhibition of ISI under different water surface conditions; including the use of surface skimmer to remove autogenous surface substances (SS), covering the water surface with liquid‐paraffin‐layer (LP) and oil film (OF), and a control (non‐treatment, NT). Significantly higher inflation frequency was observed in SS (62.2%) than NT (11.9%), LP (2.7%) and OF (3.9%). This indicates that ISI in PBT larvae can be promoted by removal of surface substances on rearing water which inhibit larval air gulping. Experiment 2 aimed to elucidate proper day of larval age to start skimming for promoting ISI with four different periods of oil film removal: from 3 to 8 (SF3D), 4 to 8 (SF4D), 5 to 8 (SF5D), 6 to 8 (SF6D) days‐post‐hatch (dph). Significant improvement in ISI frequency was observed in SF3D (80.2%) but the frequency was very poor in SF4D, SF5D, and SF6D (17.8–7.5%). This implies the need of oil film removal without missing a narrow window, 1 day of 3 dph, to promote ISI in practical PBT larviculture.  相似文献

Diel and ontogenetic changes in larval body density related to swim bladder volume were investigated in Pacific bluefin tuna, Thunnus orientalis, to determine the causality of larval mortality – adhesion to the water surface and contact with the tank bottom during seedling production. The density of larvae with deflated swim bladders increased with total length and days post hatch. Diel density change was observed after day 2 post hatch; owing to daytime deflation and night‐time inflation of the swim bladder, the density was relatively higher during the daytime. Increased swim bladder volumes clearly reduced larval density during the night‐time after day 9 post hatch. However, the density of larvae with inflated swim bladders was greater than rearing water density (Δρ>0.0099). The small density difference between larvae and rearing water (Δρ=0.0022?0.0100) until day 4 post hatch may have caused larval mortality by adhesion to the water surface because larvae can be easily transported to the water surface by aeration‐driven upwelling in rearing tanks. Density increased noticeably from day 5 to day 9 post hatch. The increased density difference (Δρ=0.0065?0.0209) in larvae and rearing water possibly induced mortality by contact with the tank bottom because larvae sink particularly during the night‐time on ceasing swimming.  相似文献

This study describes the effects of feeding taurine‐supplemented Artemia on the growth, survival, whole body taurine content and jaw malformation rate of larval yellowtail kingfish Seriola lalandi. Larvae were fed rotifers containing no supplemental taurine from 3 to 15 day post hatch (dph) and Artemia co‐enriched with taurine from 12 to 22 dph. Artemia were supplemented at concentrations of either 0, 0.8, 1.6, 2.4, 3.2 or 4.0 g of taurine L?1 during the 18 h HUFA enrichment process. Taurine content in the Artemia increased from 0.76 ± 0.04% DW in those without supplementation to 3.95 ± 0.17% DW in those supplemented at 4.0 g L?1. Survival rates of larval yellowtail kingfish were significantly lower in all taurine‐supplemented treatments compared to the unsupplemented control. Growth was significantly improved in those larvae fed taurine‐supplemented Artemia; however, we cannot attribute this improvement solely to taurine, as improved growth may have been a function of the reduced survival, and therefore increased prey availability, in these treatments. The whole body taurine content of larvae fed unsupplemented Artemia was significantly lower (1.85 ± 0.03% DW) than those fed supplemented Artemia, which did not differ from each other (pooled average 2.48 ± 0.03% DW), suggesting either a functional excretion mechanism is in place or that this represents the saturation value for larvae of this age. Jaw malformation rates were not affected by Artemia taurine content. The results of this research suggest yellowtail kingfish larvae may have a lower requirement and/or a reduced tolerance to excess dietary taurine than juveniles.  相似文献

The genetic population structure of the Pacific bluefin tuna (PBF) Thunnus orientalis and the yellowfin tuna (YFT) T. albacares in the North Pacific Ocean was investigated. The polymorphism of microsatellite (SSR) loci and sequences of mitochondrial DNA control region (mtCR) were analyzed for 71 samples of PBF from Japan and Mexico and 45 samples of YFT from Japan and Panama. In the SSR analyses, both single-locus (?0.010 to 0.008 in PBF and ?0.023 to 0.020 in YFT) and global multilocus (0.003 in PBF and ?0.002 in YFT) F ST values among the geographic populations were low and not significant in these species. In the mtCR analyses, neither the neighbor-joining tree nor the minimum spanning network showed genetic differentiation among the geographic populations in each species. The pairwise F ST values among the geographic populations of them (?0.005 in PBF and ?0.020 to ?0.014 in YFT) were low and not significant. Our SSR and mtCR data suggested that genetic differentiations were not evident among the eastern and western populations in the North Pacific Ocean either in PBF or in YFT. Mismatch distributions, demographic parameters, and neutrality tests suggested that sudden population expansion of PBF and YFT in the North Pacific Ocean occurred 628,000–731,000 and 450,000–525,000 years ago, respectively.  相似文献

The larval rearing performance of California yellowtail (Seriola lalandi) and white seabass (Atractoscion nobilis) was compared between larvae fed taurine‐supplemented rotifers (Brachionus plicatilis) and Artemia (Artemia franciscana) and control groups fed live feeds with no taurine enhancement. Results from the S. lalandi trial demonstrated that when fed taurine‐supplemented rotifers, survival was significantly higher than the control group (20.1% vs. 9.1%, P < 0.01) as was notochord length (5.43 mm vs. 5.13 mm, P < 0.005). No larval performance differences were detected between experimental and control treatments of A. nobilis. Biochemical analyses demonstrated that taurine concentrations were significantly elevated in both S. lalandi (23.7 vs. 2.7 mg g?1, P < 0.001) and A. nobilis (33.0 vs. 21.0 mg g?1, P < 0.005) when live feeds were supplemented with taurine. Results suggest that taurine is a limiting nutrient in the larviculture of S. lalandi but may not be for A. nobilis.  相似文献

We investigated the changes in myoglobin (Mb) content and proximate compositions of the cephalad parts of the dorsal ordinary muscles (Ce‐DOM) of full‐cycle cultured (FC) Pacific bluefin tuna [body length: 42.6–115.4 cm, body weight (BW): 1.7–38.4 kg, killed from July 2004 to October 2005]. The Mb content of Ce‐DOM increased (P<0.01) until a BW of 7.0 kg was reached. However, the Mb content of Ce‐DOM had a wide variety above a BW of 7.0 kg. On the other hand, the moisture, protein and ash contents of Ce‐DOM of FC tuna decreased (P<0.05 and 0.01) with an increase in BW. However, the lipid content of Ce‐DOM increased gradually above about a BW of 20.0 kg. Furthermore, the Mb content in protein of the Ce‐DOM of FC tuna increased with an increase in BW. These results indicated that the increase in Mb content of Ce‐DOM of FC tuna was not related to changes in proximate composition between a BW of 1.7 and 38.4 kg.  相似文献

This study investigated the influence of initial swimbladder inflation (ISI) failure on survival of Pacific bluefin tuna (PBT), Thunnus orientalis, larvae to prevent mass mortality due to sinking death. In Experiment 1, swimbladder inflation frequency and survival within an ISI promoted (PS) group, for which surface film of rearing water was removed, and a group without ISI promotion (NPS) were compared in 20 and 30 kL tanks. The PS group demonstrated significantly higher swimbladder inflation frequency and increased survival than the NPS group within 20 kL tanks at 9 days post hatching (dph). Similar tendencies were observed within 30 kL tanks. In Experiment 2, larval distribution and swimbladder inflation frequency in the night‐time were examined through larval sampling within the upper and middle layers of the water column and tank bottom within 1.6 and 30 kL tanks at 5 dph. Larvae at tank bottom had higher distributional density and significantly lower swimbladder inflation frequency than those distributed in the upper and middle layers within 1.6 kL tanks. Similar tendencies were observed within 30 kL tanks. Results of this study suggest the improvement of larval survival due to prevention of sinking death through ISI promotion in mass‐scale PBT larviculture.  相似文献

This study examined the influence of swimbladder inflation (SBI) failure on mortality (Experiment 1), lordotic deformity (Experiment 2) and growth (Experiment 1, 2) in Pacific bluefin tuna, Thunnus orientalis, postflexion larvae and juveniles by producing larvae lacking inflated swimbladders. Experiment 1 was conducted for postflexion larvae to juveniles (from 18 to 30 days‐post‐hatch; dph). Mortality was not significantly different between the fish with (WIS) and without (WOIS) inflated swimbladders. Standard length (SL) and body weight (BW) were significantly smaller in WOIS than WIS. Moreover, the SBI was found in WOIS after postflexion stage. In Experiment 2, two examination trials were conducted on SBI, vertebral deformity and growth for juvenile stage. Lordotic deformities were found neither in the WOIS nor in the WIS. Although SL and BW were significantly smaller in WOIS than WIS at 22 dph, after 37 dph no significant differences were found between them. The results of Experiment 1 and 2 indicate that SBI failure causes growth retardation until juveniles of 30 dph; however, it does not cause growth retardation in juveniles after 37 dph and mortality of postflexion larvae and juveniles. Moreover, SBI failure did not cause lordotic deformity in PBT differently from many other fish.  相似文献

Eastern little tuna (ELT, Euthynnus affinis) is expected to see use as a novel aquaculture species due to its rapid growth and high economic value. In this study, closing of the complete life cycle of ELT in land‐based tanks was successfully carried out. Seed production of a first generation (F1) derived from wild‐caught ELT broodstock was conducted in 2010, 2011 and 2012. Survival percentages of F1 ELT at 365 days post hatching for the 2010, 2011 and 2012 year classes were 0.9%, 0.4% and 6.3% respectively. On average, F1 ELT were approximately 40 cm in total length and weighed 1500 g by the time they reached 1 year of age. Successful artificial induction of spawning was achieved in 1‐year‐old F1 ELT using a gonadotropin‐releasing hormone analogue (GnRHa). Induction of spawning was successful at the first administration of GnRHa in the 2010, 2011 and 2012 year classes. First spawning was observed at 4 or 5 days post implantation, which indicated that oogenesis and spermatogenesis progressed within 1 year of rearing in land‐based tanks. During spawning periods, the average number of fertilized eggs and the average number of hatched larvae per spawning event in the 2010, 2011 and 2012 year classes were 20,480 eggs day?1 and 177 larvae day?1, 39,423 eggs day?1 and 9347 larvae day?1, and 819,555 eggs day?1 and 674,445 larvae day?1 respectively. Therefore, it was concluded that both male and female ELT reared in land‐based tanks were capable of reproduction at 1 year of age.  相似文献

Usual diets for rearing leptocephalus larvae of Japanese eel Anguilla japonica include eggs of the endangered spiny dogfish Squalus acanthias (SE). We investigated the effects of alternative food materials, hen egg yolk (HEY) and exoskeleton‐free (skinned) Antarctic krill (SAK), on the growth and survival of eel larvae. We found that feed comprising whole krill including exoskeleton (WAK) containing higher levels of fluoride (37.89 mg kg?1) was acutely toxic to eel larvae exposed to this alone. In contrast, extract from SAK containing lower concentrations of fluoride (4.25 mg kg?1) showed no apparent adverse effects. Growth of larvae fed a mixture of SE and SAK in a feed trial of 58 days [mean body weight (BW), 6.0 mg] was about twofold higher than that of larvae fed a mixture of SE and WAK (3.2 mg) (< 0.01). A mixture of HEY and SAK also had some dietary benefits for eel larvae, enabling them to survive for up to 58 days and to grow significantly (mean BW, 2.4 mg), compared with their initial weight (mean BW, 0.2 mg) (< 0.001). Although additional nutritional improvements are needed, the present results suggest that combination diet HEY and SAK may be a good alternative to SE as an effective diet for eel larvae.  相似文献
