The present study was designed to characterize phenotypically and genotypically nine Arcanobacterium abortisuis strains collected from specimen of pigs in a period of nine years. All nine A. abortisuis strains and A. abortisuis reference strain DSM 19515 displayed a synergistic hemolytic reaction with Staphylococcus aureus β-hemolysin, Rhodococcus equi, and Arcanobacterium haemolyticum indicator strains and showed the typical biochemical properties of this species. The species identity could be confirmed by identification and sequencing of the 16S-23S rDNA intergenic spacer region (ISR), which appeared to be a useful target for genotypic characterization of this bacterial species. The A. abortisuis strains of the present study were isolated from specimen of pigs together with various other bacterial species indicating that the pathogenic importance of this newly described species remains to be elucidated.  相似文献

A microorganism isolated from an aborted sow's placenta has been proposed as a novel species, Arcanobacterium abortisuis. The lesions in the placenta were suppurative and necrotic placentitis, whereas lesions of three fetuses were suppurative bronchopneumonia. Gram and Grocott-positive organisms in these lesions were immunolabeled with anti- Arcanobacterium abortisuis antibody. This is the first case report, which showed the association of Arcanobacterium abortisuis with the aborted lesions of a pig.  相似文献

Tests were conducted to assess the efficiency of the roots of Dahlstedtia pentaphylla (Taub.) Burk. (Leguminosae, Papilionoideae, Millettiae) plant against infestations of Boophilus microplus (Canestrini, 1887). These tests were performed on 30 bovine animals in the Paraíba Valley, State of S?o Paulo, Brazil, divided into three groups (control, extract diluted at 1:10 mL and extract diluted at 1:20 mL), after artificial infestation with some 4000 larvae/animal on days -21, -14, -7, -1, 0, 7 and 14. The extract of D. pentaphylla was obtained by dehydration, spraying and extraction in absolute ethanol, at a proportion of one part of root powder to three parts of ethanol, this being taken as standard (100%). This standard extract was then diluted in water at one part of extract to 10 and 20, for spraying on the bovines. The best result obtained (an efficiency rate of 76.10%) was seen 3 days after the application of the extract at a concentration of 1:10 mL. The extract showed no effect in inhibition of the laying or hatching of larvae on engorged females, these being collected from the bovines after treatment, and kept in the laboratory.  相似文献

为了适应现场家畜血吸虫病的筛查需要,设计了简便快速的胶体染料试纸条诊断技术用于检测家畜血清中的抗血吸虫抗体,以该方法检测血吸虫病牛、猪血清83份,其结果为阳性检出率100%,健康牛、猪血清73份的检测结果为阴性符合率分别为95.3%和94.0%,与感染肝片形吸虫的牛、羊血清16份的交叉反应率为18.8%.表明本方法敏感性高,特异性较强.  相似文献

Vaccine potential of a tick vitellin-degrading enzyme (VTDCE)   总被引:2,自引:0,他引:2  
VTDCE (Vitelin-Degrading Cysteine Endopeptidase) is a peptidase with an active role in Rhipicephalus (Boophilus) microplus embryogenesis. VTDCE is found in the tick's eggs and was shown to be the most active protein in vitellin (VT) hydrolysis of the three peptidases already characterized in R. microplus eggs (Boophilus Yolk pro-cathepsin (BYC), Tick Heme Binding Aspartic Proteinase (THAP) and VTDCE). VTDCE activity was assessed in vitro using the natural substrate and a synthetic substrate (N-Cbz-Phe-Arg-MCA). The activity was inhibited by anti-VTDCE antibodies. In the present study, it was shown that VTDCE acts differently from BYC and THAP in VT hydrolysis and that the vaccination of bovines with VTDCE induces a partial protective immune response against R. microplus infestation. Immunized bovines challenged with R. microplus larvae presented an overall protection of 21%, and a reduction in the weight of fertile eggs of 17.6% was observed. The data obtained indicate that VTDCE seems to be important for tick physiology, and that it induces partial protective immune response when inoculated in bovines. This suggests that VTDCE can be useful to improve the protective capacity observed for other antigens.  相似文献

The present study determined the genetic relationships between 41 Staphyloccocus (S.) aureus isolates from bovines, humans, and food using a single enzyme amplified fragment length polymorphism (AFLP) technique. We evaluated the prevalence of staphylococcal enterotoxin (SE) genes and other virulence gene determinants by PCR. The identification of S. aureus was based on culturing and biochemical tests, and by amplifying a specific section of the 23S rRNA gene. PCR amplification of the SE genes (sea, seb, sec, see, seg, seh, and sei) singly or in combination was observed. Most isolates of bovine origin harbored hla (84%) and cap5 (74%), while most isolates from humans harbored hla (73%), cap8 (91%), and fnbA (100%). Strains from food sources were positive for hla (100%), cap5 (100%), and cap8 (64%) unlike isolates from humans or bovines. A single enzyme AFLP analysis revealed a correlation between AFLP clusters of some strains and the source of the isolates The genotypic results of the present study might help to better understand the distribution of prevalent S. aureus clones among humans, bovines, and food and will help control S. aureus infections in Indonesia.  相似文献

Trichinellosis is a foodborne disease caused by the consumption of raw meat and raw meat-derived products from swine, horse and some game animals infected with nematode worms of the genus Trichinella. Between June 2006 and February 2011, 16 million domestic pigs and 0.22 million wild boars (Sus scrofa) were tested for Trichinella sp. in Hungary. Trichinella infection was not found in any pigs slaughtered for public consumption. Nevertheless, Trichinella spiralis was detected in four backyard pigs when trace back was done following a family outbreak. Trichinella infection was demonstrated in 17 wild boars (0.0077%). Larvae from wild boars were identified as Trichinella britovi (64.7%), T. spiralis (29.4%) and Trichinella pseudospiralis (5.9%). Although the prevalence of Trichinella sp. infection in wild boars and domestic pigs is very low, the spatial analysis reveals that the level of risk differs by region in Hungary. Most of the T. britovi infected wild boars (63.6%) were shot in the north-eastern mountain area of Hungary; whereas domestic pigs and wild boars infected with T. spiralis were detected only in the southern counties bordering Croatia and Romania. In the north-western and central counties, the prevalence of Trichinella infection seems to be negligible.  相似文献

Experimental reproduction of necrotising enteritis of sucking pigs was successfully achieved by using both Clostridium perfringens Type C strains, which had been isolated from sucking pigs with necrotising enteritis, and Type C strain 3628 of A.T.C.C. (sub-type C1). The lethal dose for sucking pigs was between 20 X 10(6) and 12 X 10(7) pathogens per animal. The disease could not even be induced by repeated application of no-bacterial toxin of Cl. perfringens Type C nor by administration of Cl. perfringens Type A strains which had been cultured from broilers with necrotising enteritis. Necrotising enteritis was found to develop in two phases in sucking pigs. First, the pathogen will deposit to the villous epithelium and then penetrate the superficial strata of the mocous membrane. In the second phase, the villous structure will be destroyed by the lethal, haemolysing, and necrotising toxins of Cl. perfringens. The role played by individual toxin fractions is discussed together with the importance of humoral and localised infection defence. Sucking pigs may be sufficiently protected against infection based on single or ten-fold lethal infectious dosage by two vaccinations of the mother animal, five and three weeks prior to parturition, using "Enterotoxaemia Vaccine Dessau bivalent". Infection then would not occur unless a hundredfold lethal dose was applied. Characteristics include diarrhoea, apathy, exhaustion, and death.  相似文献

The protozoan parasite Toxoplasma gondii infects almost all warm blooded animal species including humans, and is one of the most prevalent zoonotic parasites worldwide. Post-natal infection in humans is acquired through oral uptake of sporulated T. gondii oocysts or by ingestion of parasite tissue cysts upon consumption of raw or undercooked meat. This study was undertaken to determine the prevalence of oocyst-shedding by cats and to assess the level of infection with T. gondii in meat-producing animals in Switzerland via detection of genomic DNA (gDNA) in muscle samples. In total, 252 cats (44 stray cats, 171 pet cats, 37 cats with gastrointestinal disorders) were analysed coproscopically, and subsequently species-specific identification of T. gondii oocysts was achieved by Polymerase Chain Reaction (PCR). Furthermore, diaphragm samples of 270 domestic pigs (120 adults, 50 finishing, and 100 free-range animals), 150 wild boar, 250 sheep (150 adults and 100 lambs) and 406 cattle (47 calves, 129 heifers, 100 bulls, and 130 adult cows) were investigated by T. gondii-specific real-time PCR. For the first time in Switzerland, PCR-positive samples were subsequently genotyped using nine PCR-restriction fragment length polymorphism (PCR-RFLP) loci (SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1 and Apico) for analysis. Only one of the cats shed T. gondii oocysts, corresponding to a T. gondii prevalence of 0.4% (95% CI: 0.0-2.2%). In meat-producing animals, gDNA prevalence was lowest in wild boar (0.7%; 95% CI: 0.0-3.7%), followed by sheep (2.0%; 95% CI: 0.1-4.6%) and pigs (2.2%; 95% CI: 0.8-4.8%). The highest prevalence was found in cattle (4.7%; 95% CI: 2.8-7.2%), mainly due to the high prevalence of 29.8% in young calves. With regard to housing conditions, conventional fattening pigs and free-range pigs surprisingly exhibited the same prevalence (2.0%; 95% CI: 0.2-7.0%). Genotyping of oocysts shed by the cat showed T. gondii with clonal Type II alleles and the Apico I allele. T. gondii with clonal Type II alleles were also predominantly observed in sheep, while T. gondii with mixed or atypical allele combinations were very rare in sheep. In pigs and cattle however, genotyping of T. gondii was often incomplete. These findings suggested that cattle in Switzerland might be infected with Toxoplasma of the clonal Types I or III, atypical T. gondii or more than one clonal Type.  相似文献

Toll样受体(TLRs)能识别各种微生物成分并诱导免疫反应。作为TLR家族中一员的TLR4是识别革兰氏阴性菌内毒素-脂多糖(LPS)的主要受体,其多态性与动物对相关病原的免疫力有着明显的相关性。本文利用PCR-SSCP并结合PCR产物双向测序的方法,对梅山猪、新淮猪、大白猪、长白猪和杜洛克猪共203个样本TLR4基因外显子3部分片段的单核苷核多态性进行了研究。结果检测到了5个SNP,分别是T611A,G826A,G960A,G962A和C1027A,其中有4个非同义的SNP,有2个SNP的氨基酸性质发生了改变。猪TLR4的SNP出现频率在各猪种中有差异,其中T611A仅在新淮猪和大白猪中检测到,而G826A和C1027A仅分别在新淮猪和大白猪中检测到。  相似文献

The aim of this study was to obtain information that could help to ease the weaning transition in commercial pig production. Before weaning, phytohemagglutinin (PHA) in the form of a crude preparation of red kidney bean lectin was fed by gavage to 24 crossbred [(Swedish Landrace x Yorkshire) x Hampshire] piglets, whereas 24 control piglets were fed alpha-lactalbumin by gavage, to study the effect on growth, occurrence of postweaning diarrhea, feeding behavior, and some anatomical and physiological traits of the gastrointestinal tract. Within the litter, piglets were randomly assigned to PHA treatment or control and remained in the same pen from the beginning (PHA exposure at 7 d before weaning) until the end of the experiment (14 d post-weaning). Weaning took place at the age of 31 to 34 d. Pigs treated with PHA grew faster (P = 0.013) during the first week postweaning and tended to have lower total diarrhea scores (P = 0.10) than did control pigs. On d 5 after weaning, piglets treated with PHA spent more time eating (P = 0.028) than control pigs. No immunostimulating effect of PHA, measured by plasma immunoglobulin G, could be detected. An increase in the intestinal barrier properties before weaning, as a response to PHA treatment, was demonstrated in intestinal absorption studies using Na-fluorescein and BSA as gavage-fed markers. Less uptake (measured as plasma concentrations) of the marker molecule Na-fluorescein occurred during a 24-h study period, and numerically lower levels of BSA were observed compared with studies in control pigs of the same age. A total of 12 pigs (6 control, 6 PHA-treated) were euthanized on the day of weaning for analyses of gastrointestinal properties. The PHA-treated pigs tended to have a longer total small intestinal length (P = 0.063) than that of the control pigs. The enzyme profile of the jejunal epithelium responded to PHA exposure with a decrease in lactase activity and an increase in maltase and sucrase activities, which is similar to changes normally observed after weaning. No differences were found in the size of the pancreas or in its contents of trypsin and amylase. In conclusion, exposing piglets to crude, red kidney bean lectin for 3 d during the week before weaning led to changes in performance and small intestinal functional properties that would be expected to contribute to a more successful weaning.  相似文献

Sera collected from 507 hunter-killed wild pigs (Sus scrofa) between 1993 and 2004 from five geographic regions in northern Spain and seven regions in southern Spain were assayed for antibodies to Toxoplasma gondii by the modified agglutination test (MAT). Antibodies to T. gondii were detected in 185 (38.4%) of 507 pigs with titers of 1:25 in 71, 1:50 in 111 and > or =1:500 in 3; seroprevalence was significantly higher (P<0.05) in pigs from southern regions. Seroprevalence was density dependent; it was higher in pigs from high stocking per hectare and availability of forage. Statistically significant differences were not observed between T. gondii seroprevalence and hunting estates (open versus fenced), sex or age. Serological results indicate a widespread exposure to T. gondii among Spanish wild boars, suggesting that this population could represent a public health risk for persons that handle or consume raw or undercooked infected wild pig meat.  相似文献

From 397 fecal specimens from apparently healthy and from diarrheic pigs, dogs, cats and cattle 59 strains (= 15%) of thermophilic Campylobacter (C.) spp. were isolated by culture. 39 strains were identified as C. coli and 18 as C. jejuni whereas 2 isolates could not be classified. None of the strains was found to be positive for cytotoxic enterotoxin in the GM1-ELISA. In the Vero-cell test 5 isolates showed a cytotoxic effect. The salt aggregation test (SAT) for indicating cell surface hydrophobicity was positive with 24 strains (5 C. jejuni, 19 C. coli). A correlation of isolation results with clinical manifestation could not be observed.  相似文献

A diagnostic project was initiated across the United States in 2006 to improve the understanding of porcine circovirus associated diseases (PCVAD) as well as to identify co-factors in PCVAD-affected farms. A Porcine circovirus-2 (PCV-2) DNA real-time polymerase chain reaction quantitation (qPCR) was established according to a published method and sera from a total of 23pig farms across the United States were examined for viral loads for PCV-2 and analyzed for any possible effects of Porcine reproductive and respiratory syndrome virus (PRRSV) vaccination on this parameter. Vaccination against PRRS resulted in significantly lower viral loads for PCV-2 in animals 13 wk or older compared with nonvaccinated animals, but vaccination of pigs against PRRS had no effect on qPCR results for PCV-2 in 4- to 12-week-old pigs. Interestingly, PRRS vaccinates had significantly lower viral loads when peak wasting disease was observed in the herds. The qPCR method for PCV-2 proved to be an important tool for help in the antemortem diagnosis of PCVAD as well as in the monitoring of infection dynamics.  相似文献

Babesiosis and Theileriosis are tick-borne diseases widespread in tropical and sub-tropical regions with high economic impact worldwide. In Portugal there are at least 4 tick vectors known to be competent for the transmission of Babesia and Theileria sp. identified: Rhipicephalus bursa, Rhipicephalus (Boophilus) annulatus, Ixodes ricinus and Haemaphysalis punctata. All these potential Babesia and Theileria tick vectors are widely distributed in Portugal, although they are predominant in the Southern region. In this study, 1104 cattle blood samples were randomly collected from Central and Southern regions of Portugal and analyzed by PCR-reverse line blotting (RLB) for the detection of Babesia and Theileria sp. Testing indicated that 74.7% of the bovines tested were positive for either Babesia and/or Theileria sp. In addition, five different apicomplexan species, namely, Theileria buffeli, Theileria annulata, Babesia divergens, Babesia bovis, and Babesia bigemina were detected by RLB among the bovines tested. T. buffeli was the most frequently found species, being present in 69.9% of the positive samples either as single infections (52.4%), or as mixed infections (17.5%). The Babesia specie most frequently found was B. divergens, detected in 4.2% of the infected bovines. Overall, infected bovines were found in all regions tested; however the highest number of infected bovines was observed in évora district (96.2%) and in cattle from Limousin breeds (81.7%). The results indicate widespread Babesia and Theileria infections in Portuguese bovines, suggesting the need for improved control of ticks and tick-borne diseases.  相似文献

The pathogenicity of 2 isolates of each of serovars 7, 3, 1 and 2 of Actinobacillus pleuropneumoniae was tested by intranasal inoculation into 60, 6-week-old large white pigs. Four dose rates varying from 0.27 to 560 x 10(6) organisms per pig with 10-fold serial dilutions were used. Surviving pigs were necropsied 7 days after inoculation. The proportion of pigs dying and developing gross lesions following infection was significantly greater for pigs given serotype 1 than for each of the other 3 serotypes, which did not differ significantly from each other. Twelve of 16 pigs given either of the 2 isolates of serovar 1 died after acute illness and 1 of 44 pigs given either of the 2 isolates each of serovars 7, 3 and 2 died. Pigs given serovar 1 showed high temperatures, severe respiratory distress, frothy haemorrhagic nasal discharge and weight loss. Lung lesions were produced in all 16 pigs given serovar 1, in 7 of 14 pigs given serovar 7, 7 of 14 pigs receiving serovar 3 and in 5 of 16 pigs given serovar 2. The lethal infections were characterised by a severe acute fibrinohaemorrhagic necrotising pleuropneumonia, whereas non-lethal cases had lung lesions ranging from necrotising purulent pleuropneumonia to abscessation. Significant differences between isolates in proportions of tissues culture positive for A. pleuropneumoniae for serovars 7 and 2, but not for serovars 3 and 1 suggested that isolates may vary in virulence within serovars, but more detailed studies are needed to clarify this point.  相似文献

Toxoplasma gondii and Neospora caninum are phenotypically and phylogenetically closely related cyst-forming coccidia, both of which may cause abortion in livestock animals. T. gondii exhibits also zoonotic potential by causing diaplacental infections in the human fetus and harmful infections in immunosuppressed individuals. Humans get infected either by consuming inappropriately prepared cyst-containing meat or by ingesting oocysts originating from cat feces. Therefore, in order to assess infection risk we need to have knowledge on the prevalence of the parasite in consumable meat and thus slaughtered animals. So far, no data indicate any zoonotic potential for N. caninum. Due to its high economic impact in the bovine production in Switzerland, we included this parasite in the present study as well. The prevalence of both parasite species were investigated by PCR in muscle and brain samples of slaughtered bovines, sheep, pigs and horses. Comparatively, a serum sample from each animal was simultaneously tested serologically by a Toxoplasma-P30-ELISA and a Neospora-SA-ELISA. The prevalences determined by the T. gondii-PCR were the followings: adult cows 3%, young bulls 2%, young cows prior to gravidity 6%, calves 1%, sheep 6%, horses and pigs each 0%. For N. caninum, the PCR-prevalence was 2% for adult cows and 0% for all other animal groups. Conversely, the seroprevalences were much higher for both parasite species and all animal groups, with the exception of the fattening pigs. However, as T. gondii was principally detectable in bovine (cows and calves) as well as in sheep meat, the consumption of this meat harbours a potential infection risk for humans. In contrast, the lack of any parasite detectability in fattening pig and horse meat allows to consider this infection source as neglectable when compared to bovine and ovine meat.  相似文献

Raw or inadequately cooked pork is an important source of Toxoplasma gondii infection, and the infection rate in animals used as human food, is an important risk predictor. The prevalence of this infection was estimated in 396 sera from 5-month old pigs obtained at abattoirs in S?o Paulo, Brazil (300) and Lima, Peru (96). The seroprevalence was higher in pigs from Peru (32.3%) as compared to Brazil (9.6%), as detected by ELISA and Western blot. Hemagglutination gave poor resolution which was not useful for the diagnosis of T. gondii infection. Specific antibody avidity is correlated with infection time, as shown in experimentally infected piglets. Using an arbitrary cut-off of 50% avidity index, Brazilian pigs were found to be more recently infected than Peruvian pigs. Pork should be considered a significant source of human T. gondii infection both in Brazil and Peru. Avidity assays could help in the detection of the time of T. gondii infection in pigs, allowing preventive management.  相似文献
