(1) Cellulases and xylanases display a modular architecture that comprises a catalytic module linked to one or more non-catalytic carbohydrate-binding modules (CBMs). On the basis of primary structure similarity, CBMs have been classified into more than 30 different families. These non-catalytic modules mediate a prolonged and intimate contact of the enzyme with the target substrate, eliciting efficient hydrolysis of the insoluble polysaccharides. (2) Xylanases are very effective in improving the nutritive value of wheat- or rye-based diets for broiler chicks although the role of non-catalytic CBMs in the function of exogenous modular xylanases in vivo remains to be determined. (3) A study was undertaken to investigate the importance of a family 6 CBM in the function of recombinant derivatives of xylanase 11A (Xyn11A) of Clostridium thermocellum used to supplement cereal-based diets for poultry. (4) The data show that birds fed on a wheat-based diet supplemented with the modular xylanase display an increased final body weight when compared with birds receiving Xyn11A catalytic module or birds receiving the enzyme mixture Roxazyme G. (5) Interestingly, the modular xylanase was truncated and transformed into its single domain counterpart on the duodenum of birds fed on the wheat-based diets, most possibly due to the action of pancreatic proteases. (6) Together the data point to the importance of CBMs in the function of feed xylanases and suggest, that in chicken fed on wheat-based diets, the main sites for exogenous enzymes action might be the gastrointestinal (GI) compartments preceding the duodenum, most probably the crop.  相似文献

1. A total of 1920 Ross broiler chicks were used in a 6 treatment by 8 replicate growth study to investigate the effect of conditioning temperature and exogenous xylanase addition on diet characteristics and bird performance. 2. Increasing conditioning temperature from 80 to 90 degrees C increased the viscosity of the diets by approximately 6 mPa.s and reduced body weight gain of the broiler chickens by 7%. 3. Furthermore, there was a significant interaction between temperature and the addition of exogenous xylanase, with a proportionately greater positive response to added xylanase in the diets that were pelleted at higher temperatures. 4. Addition of exogenous xylanase also increased the proportion of soluble arabinoxylan in the feed and reduced dietary viscosity, which is likely to be related to the degree of polymerisation of the soluble carbohydrate. 5. These results suggest that conditioning temperature should be considered as a factor when attempting to predict the response of broilers to added xylanase, especially when high-viscosity or 'low-quality' wheat is being used. 6. It can be concluded that pelleting wheat-based diets above 80 degrees C can compromise bird performance and that, if higher temperatures are to be employed, the use of exogenous xylanase is critical to maintain productivity.  相似文献

1. The potential for the nutritional improvement of pea-based diets by supplementation with a cocktail of exogenous carbohydrases was investigated using growing broiler chicks. 2. Pea meals (grown in the UK) were included in wheat-based diets at 300 g/kg as a partial replacement for an approximately isonitrogenous mixture of wheat and soybean meal. A wheat/soybean meal diet served as a control and each diet was supplemented with a cocktail of alpha-amylase, pectinase and cellulase. The diets were fed to 1-d-old broiler chicks for a period of 21 d. Weight gain and feed conversion were monitored weekly and excreta were collected during the final week in order to determine nutrient digestibility coefficients and metabolisable energy. On d 21, the gastrointestinal (GI) tract was excised and gross morphology measured. 3. Inclusion of pea meal reduced weight gain, feed conversion, nutrient digestibility and also increased the relative sizes of the distal sections of the GI tract. 4. Enzyme addition partially ameliorated the detrimental effects of pea meal inclusion although similar improvements were also noted for birds fed on the control diet. 5. It is concluded that the nutritive value of pea-meal-based diets can be improved by the addition of carbohydrases, and that some pea cultivars show considerable potential as vegetable protein sources for broiler chicks.

1. The aim of the present study was to investigate the effect of an enzyme cocktail composed of xylanase, amylase and protease on the performance and nutrient utilisation of broiler starters fed on a maize-based diet. A maize/soy-based control diet was formulated to be marginal in apparent metabolisable energy (AME) and amino acids and this diet was fed without or with two concentrations (250 and 500 g/tonne) of the exogenous enzyme cocktail. 2. Growth performance was measured over a 21-d period after which ileal contents were collected for measurement of digestible energy, nitrogen and dry matter. Excreta were collected from d 17 to 20 post-hatch and used to determine the AME, and retention of nitrogen and dry matter. 3. Supplementation of the control diet with 250 and 500 g/tonne of the enzyme cocktail resulted in enhanced performance of the chicks (1.4 and 8.3%, respectively, for weight gain and 1.2 and 2.2%, respectively, for feed per gain), but the improvements were significant (P < 0.05) only at 500 g/tonne. 4. Addition of both 250 and 500 g/tonne improved AME, nitrogen-corrected AME and dry matter retention compared with the control though the improvements were greater with the higher dose. These effects were also reflected in the apparent ileal digestibility coefficients of dry matter, nitrogen and energy, with 500 g/tonne of the enzyme cocktail improving ileal digestible energy by 3.4%. 5. These results demonstrate that a cocktail of exogenous enzymes containing xylanase, amylase and protease is effective in improving the performance of broiler chicks fed on a maize/soybean-meal-based diet. However, these benefits may be dose-dependent and so an economic analysis involving ingredient and enzyme pricing and the relative performance and digestibility enhancements could allow the end user to select the most economical dose to maximise return on investment.  相似文献

1. The aim of the present experiment was to examine the influence of particle size and xylanase supplementation on the performance, energy utilisation, and gross morphological and histological parameters of the digestive tract of broiler starters fed wheat-based diets. The experimental design was a 2 x 2 factorial arrangement of treatments evaluating two wheat particle sizes (medium and coarse) and two levels of xylanase supplementation (without or with 1000 xylanase units/kg diet). The two particle sizes were achieved by grinding the whole wheat in a hammer mill to pass through 3 and 7 mm screens, respectively. Broiler starter diets, based on wheat and soybean meal, were formulated and each diet was fed ad libitum to 6 pens of 8 male broilers each from d 1 to 21 post-hatch. 2. Wheat particle size had no effect on the performance of broilers. Xylanase supplementation had no effect on weight gain and feed intake. However, a significant particle size x xylanase interaction was observed for feed per unit gain with xylanase lowering feed per gain only in the coarse particle size diet. Xylanase supplementation improved the nitrogen-corrected apparent metabolisable energy at both particle sizes. 3. Xylanase supplementation decreased the relative weight and relative length of all gut components. Villus height and crypt depth and total extent of the mucosal layer were unaffected by both wheat particle size and xylanase supplementation. 4. The apparent viscosity of intestinal digesta, determined over the physiological range of shear rates, was not influenced by xylanase addition or particle size. 5. The findings of this study suggest that the effectiveness of exogenous xylanase in broiler diets is influenced by wheat particle size.  相似文献

(1) The aim of the experiment was to re-examine variations in digestibilities of food components in 3-week-old broiler chickens fed on pelleted diets based on wheats differing in lipase activity and hardness. Fourteen wheat (Triticum aestivum) samples, each from a different cultivar, were included at 550 g/kg in 14 different diets given to male Ross broiler chicks from 7 d of age. The other main ingredients consisted of soyabean meal (353 g/kg) and rapeseed oil (55 g/kg). A 15th diet containing durum wheat (Triticum durum) was also tested. (2) Hardness of wheats (Triticum aestivum) varied between 14 (very soft) and 88 (very hard), and lipase activity of wheats varied from 1 to 13.1 (relative scale). No significant correlation was found between in vitro viscosities and other parameters such as hardness, particle size of wheat flours and lipase. Hardness was correlated with the mean particle size of wheat flours and durability of pellets. (3) Individual lipid digestibilities were negatively correlated with in vitro viscosities of wheats. (4) Individual starch digestibilities were negatively correlated with wheat hardness, particle size of wheat flour before pelleting, and pellet durability. The ratio of measured AME(N) to predicted AME(N) was also negatively correlated with wheat hardness. Simple regression calculation showed that a 100-point increase in wheat hardness resulted in a 3% decrease in the AME(N) value of diets. Multiple regression calculation showed the food/gain ratio (d 10 to d 21) to be positively related to wheat hardness and negatively related to pellet durability. (5) Wheat lipase activity was positively correlated with individual starch digestibility, which was the reverse of a result obtained in a previous experiment. Thus, wheat lipase activity did not seem consistent for predicting starch digestibility and AME(N) values. (6) Among all wheat samples, durum wheat showed the highest protein content and the lowest content of water-insoluble cell-wall. Starch digestibility of durum wheat tended to be lower than that of other wheats (0.916 vs 0.936). However, no significant difference in AME(N) was observed between the durum wheat sample and other wheats. (7) Gut morphometric data measured at d 24 did not show significant differences between dietary treatments.  相似文献

1. A commercial enzyme preparation (Quatrazyme HP) containing xylanase and beta-glucanase was examined in two laying hen experiments with wheat/barley- or maize-based diets. The activities of other enzymes were measured also. Starch, cell wall contents and effects of Quatrazyme HP on in vitro viscosity of wheat, barley, maize and soybean meal were determined. 2. In the first experiment, 90 ISA Brown laying hens at 28 weeks of age were given a wheat/barley basal diet with or without 20mg of Quatrazyme HP, which provided 560 and 2,800 IU of xylanase and beta-glucanase/kg diet. In the second experiment, 66 ISA Brown laying hens at 45 weeks of age were given a maize/soybean meal basal diet with or without 20 mg of Quatrazyme HP/kg diet for 9 weeks. Egg production, egg weight, egg mass, feed conversion ratio and change in body weight were recorded as response criteria. 3. There was a significant improvement in feed conversion ratio with enzyme supplementation. Birds given an enzyme-supplemented diet gained 86 g while those fed on the unsupplemented diet lost 103 g of their body weight by the end of the experiment. 4. The enzyme preparation did not affect either egg production, egg weight or egg mass of birds fed on the maize/soybean meal diet. However, a significant improvement in feed conversion ratio was detected. Birds on either the supplemented or unsupplemented diet exhibited an increase in their body weight at the end of the experiment. 5. Addition of xylanase and beta-glucanase decreased in vitro viscosity of wheat, barley, maize and soybean meal. This effect was greater for wheat and barley than for maize and soybean meal. 6. It was concluded that the beneficial effect of using an enzyme preparation containing xylanase and beta-glucanase is not limited to wheat/barley-based diets but also occurs with maize/soybean meal-based diets.  相似文献

1. Three experiments were conducted to determine growth of broiler chicks fed on test diets formulated to be deficient or adequate in tryptophan (Trp) using gelatin by-product as a means of generating a Trp deficiency. Growth response estimates of broiler chicks to graduations of Trp were determined by dose-response criteria and regression analyses. Experiments were conducted using broiler chicks from 1 to 20 d of age. 2. Broiler chicks fed Trp-deficient diets had poor body weight gain, feed intake and feed conversion. Recommended total Trp needs were 2.0, 2.1 and 2.2 g/kg for feed intake, body weight gain and feed conversion, respectively. 3. Blood plasma Trp exhibited a sigmoidal trend while blood plasma glucose increased in a linear manner to supplemental Trp. Physiological stress variables measured were unaffected by dietary Trp.  相似文献

Rice bran, a by-product of the rice industry, is available for animal feeds. However, it has not been a common part of poultry rations in Argentina. Hydrolytic and oxidative rancidity development, phytate content, enzyme inhibitor, and high fiber content are the most important antinutritive factors cited as limiting factors for its use. An experiment utilizing 1-d-old male broiler chicks was conducted to determine the responses of zootechnical and bone mineralization parameters to diets with different concentrations of rice bran. The feed conversion and tibia ash were more sensitive than weight gain to detect antinutritive factors in rice bran. High concentrations of rice bran (in excess of 20%) produced a significant reduction in body weight. Furthermore, feed conversion and bone mineralization variables were impaired by feeding 10% rice bran diets. The adverse effects of rice bran on weight gain, feed conversion, and mineralization in the current studies suggest that rice bran should be included in broiler diets at a level between 10 and 20% if strategies are not used to decrease the antinutritive activity.  相似文献

This study investigated the influence of pre‐pelleting inclusion of whole wheat (WW) and exogenous enzyme supplementation on growth performance, coefficient of apparent ileal nutrient digestibility (CAID) and apparent metabolizable energy (AME) in broilers fed wheat‐based pelleted diets. A 2 × 3 factorial arrangement of treatments was used with two methods of wheat inclusion [622 g/kg ground wheat (GW) and 250 g/kg WW replaced GW (wt/wt) pre‐pelleting (PWW)] and three enzymes (xylanase, phytase and xylanase plus phytase). A total of 288, one‐day‐old male broilers (Ross 308) were individually weighed and allocated to 36 cages (8 broilers/cage), and the cages were randomly assigned to the six dietary treatments. Birds fed PWW diets gained more (p < 0.05) weight than those fed GW diets. There was no effect (p > 0.05) of WW inclusion on feed intake (FI). Phytase alone increased (p < 0.05) FI compared to xylanase or the combination. Whole wheat inclusion increased (p < 0.05) the gain‐to‐feed ratio (G:F). Feeding xylanase plus phytase and phytase‐added diets resulted in the greatest and lowest G:F, respectively, with xylanase supplemented diets being intermediate. Birds fed PWW diets had greater (p < 0.05) relative gizzard weights than those fed GW diets. There was no effect (p > 0.05) of WW inclusion on the CAID of nitrogen (N), starch and fat. Combination of xylanase and phytase resulted in greater (p < 0.05) digestibility of N, starch and fat than that of individual additions. Feeding PWW diets resulted in greater (p < 0.05) AME values than GW diets. Combination of xylanase and phytase increased (p < 0.05) the AME compared to the diets with individual additions of xylanase or phytase. The current results suggest that the influence of pre‐pelleting WW inclusion and exogenous enzymes on nutrient digestibility and broiler performance is not additive.  相似文献

Faba beans (Vicia faba) are an alternative protein source that likely can be used to a higher extent in broiler diets. White-flowered faba beans contain antinutritional substances (ANS) such as non-starch polysaccharides (NSP), trypsin inhibitors, and lectins, which might limit its inclusion level. Lectins and trypsin inhibitors are heat labile and previous studies have shown that steam-pelleting and enzyme treatment improves the nutritional value of faba beans. However, alternative to pelleting would facilitate for farmers to add faba beans on-farm. Currently, there are machines available for toasting faba beans on-farm, which might be used for broiler mash diets. The objective of this study was to investigate the effect of inclusion level (0, 10, 20 and 30%), toasting (140°C 5.5 min) and different enzymes (xylanase + phytase vs. xylanase, phytase, amylase, protease) of faba bean diets on growth performance and organ parameters in broilers. To test this, 2 experiments 34 and 35 days, using a total of 480 chickens were performed. Feed intake, body weight (BW) and feed conversion ratio were registered weekly, in addition, organ and carcass weights were registered at slaughter. The results showed that inclusion of 20% faba beans is possible in a pelleted diet with maintained broiler growth performance. When 20% was included in a mash diet, feed intake and BW decreased compared to chickens fed pelleted diets, irrespectively of pre-toasting of the beans. It can be concluded that toasting cannot replace pelleting. Supplementation of protease and amylase in addition to xylanase and phytase did not improve the nutritional value of faba beans.

The efficiency of a mixture of xylanase and beta-glucanase enzymes was evaluated in two separate experiments on growing turkeys offered diets based on wheat or wheat and barley. 2. In the first experiment, the addition of 560 and 2800 IU of xylanase and beta-glucanase, respectively, per kg of diet significantly improved feed efficiency in turkeys fed on wheat- and barley-based diet throughout the entire experiment (42 d). 3. In the second experiment, the enzyme mixture significantly increased N-corrected apparent metabolisable energy (AMEN) by approximately 5%. 4. Furthermore, the combination of xylanase and beta-glucanase significantly improved body weight gain and feed efficiency. Feed efficiency was increased by 2.94, 2.47 and 5.91% in diets based on 500 then 540 g of wheat/kg of diet, 394 then 384 g of wheat and 100 then 150 g of barley/kg of diet and 180 g of wheat and 300 then 350 g of barley/kg of diet, respectively. 5. This enzyme mixture decreased in vitro viscosity of wheat, barley and soybean meal, the effect being larger for wheat and barley than for soybean meal. 6. In conclusion, the supplementation of diets based on wheat and barley with xylanase and beta-glucanase significantly improved body weight gain and feed efficiency. Therefore, wheat and barley could be used more efficiently by turkeys in the presence of an appropriate enzyme preparation.  相似文献

Pathology and histopathology of gossypol toxicity in broiler chicks   总被引:1,自引:0,他引:1
Two experiments were conducted to determine the toxicity, pathology, and histopathology of purified gossypol in broiler chicks. Gossypol was added to broiler feed at 0, 100, 200, and 400 mg/kg of feed in Experiment 1 and at 0, 800, and 1600 mg/kg of feed in Experiment 2. Day-old broiler chicks were fed these diets from 1 to 21 days in Experiment 1 and from 1 to 23 days in Experiment 2. In Experiment 1, body weight and feed intake at 21 days were not significantly affected by dietary gossypol. However, chicks fed gossypol at 400 mg/kg of feed had poor feed conversion ratio compared with the other treatment. Feed conversion ratios were 1.493, 1.564, 1.471, and 1.60 for chicks fed gossypol at 0, 100, 200, and 400 mg/kg of feed, respectively (Experiment 1). Chicks fed 400 mg/kg gossypol also had mild perivascular lymphoid aggregate formations and bilary hyperplasia in the liver. In Experiment 2, gossypol at 1600 mg/kg resulted in 28.1% mortality. Gossypol at 800 and 1600 mg/kg feed resulted in significant decreases in body weight and feed intake of chicks. The average body weights of 23-day-old chicks in Experiment 2 were 676, 224, and 111 g for 0, 800, and 1600 mg/kg gossypol, respectively. Feed conversion ratios of chicks fed 800 and 1600 mg/kg gossypol were significantly higher than those of chicks fed control diets (1.383 vs. 1.564 vs. 1.745 for 0, 800, and 1600 mg/kg gossypol, respectively). Plasma iron and hematocrit values were significantly reduced by gossypol at 800 and 1600 mg/kg of feed. Enlarged gallbladder was the only gross pathology symptom associated with gossypol levels. Severe cases of perivascular lymphoid aggregate formation, biliary hyperplasia, and hepatic cholestasis were observed in chicks fed 800 and 1600 mg/kg of gossypol in feed. No gossypol-related changes were observed in kidney tissues of chicks. These results show that gossypol is toxic to broiler chicks at high levels. This study also shows that histopathologic changes in liver due to gossypol also occur at levels lower than the levels that affect body weight.  相似文献

The objective of the present study was to determine the effect of broiler breeder egg weight on egg quality, chick quality, and broiler performance using Hubbard Classic broiler breeder flock. Hatching eggs from a commercial broiler breeder flock (Hubbard classic strain) were obtained at the age of 60 wk and divided into 3 egg-weight categories, namely small, medium, and large. For this purpose, 930 eggs having weights 63.09 ± 0.21, 68.85 ± 0.23, and 74.81 ± 0.11g were divided into 3 egg categories labeled small, medium, and large respectively. We further divided these eggs into 2 categories, eggs for quality parameters and eggs for incubation and posthatch performance parameters. Eggs from the small egg category had greater proportion of the shell weight and more shell strength. Egg weight did not significantly affect the albumen and yolk weight percentage. Egg weight affected the shape index and specific gravity for which lower values were observed for large eggs. Egg weight positively affected the chick weight, chick yield, and chick length (P < 0.05) of both male and female chicks. Results of the body weight gain showed that effect of egg weight on the posthatch performance of chick diminishes with the age of broilers. Egg weight significantly affected the body weight gain of male and female chicks up to 3 wk of age. Egg weights did not significantly affect the feed intake and mortality of broiler. Different egg-weight categories significantly affected the feed conversion ratio of female chicks at wk 2 and 3 of age and did not affect the feed conversion of male and female chicks at the end of wk 5. In conclusion, egg weight positively affected the chick characteristics (e.g., chick weight, chick length) and did not affect the final live body weight, feed conversion ratio, feed intake, and mortality in broilers.  相似文献

Two experiments were conducted with broiler chicks to determine the nutritive value of high-oil sunflower meal (HO-SFM), a sunflower oil extraction by-product obtained through screw-press extraction and expanding processes with a proximate composition of 32% CP, 12% crude fiber, and 19% ether extract. In Experiment 1, the effects of a high level (46.4%) of HO-SFM on chick performance and gastrointestinal organs were tested. The objective of Experiment 2 was to determine if pelleting the feed could overcome the bulkiness resulting from inclusion of HO-SFM at a high level. The results of Experiment 1 indicated that addition of 46.4% HO-SFM to broiler starter diets significantly (P < 0.05) depressed body weight, feed intake, and gain but not feed conversion. Fat pad and liver lipid were again significantly (P < 0.05) decreased in the HO-SFM treatment. Impaired performance might have been due to the difference of the density of HO-SFM diet (608 g/L) compared with the soybean meal control (723 g/L). When bulkiness was overcome by pelleting in Experiment 2, it was found that pelleting the feed significantly enhanced growth of broiler chicks compared with SFM or soybean meal mash diets. Liver weights and lipid content were again decreased in HO-SFM diets. The results of this study suggest that HO-SFM can be used up to 28% without adverse effects on broiler chicks. Further improvement was observed with pelleting. Liver weight and lipid content were consistently reduced by feeding HO-SFM.  相似文献

Four experiments were conducted with a total of 2288 pekin ducks. Day-old ducklings were group-penned on straw bedding and were fed complete, pelleted diets ad libitum for up to 49 days depending on experiment. In each experiment, starter diets (until day 21) and grower diets (from day 22) were used adequate in ME content and nutrient content. The sum of wheat, rye, and triticale amounted to at least 57% (starter diet) and 63% (grower diet), respectively. The inclusion level of wheat, rye, and triticale was different between experiments, with a maximum rye inclusion of 45%. Five different enzyme preparations all having, 1,4-beta-xylanase as the main activity were considered in this study with either one (2 preparations) or three (3 preparations) levels of supplementation. The effect of enzyme supplementation on ileal digesta viscosity was studied at the end of two experiments comprising 4 enzyme preparations. A significant reduction in digesta viscosity was determined for all preparations. The viscosity of digesta was higher in birds that were fed 45% rye in their diet as compared to those fed a diet based on triticale and wheat, even with enzyme supplementation. Differences in digesta viscosity were not reflected in growth or feed conversion data. In one experiment, the body weight of ducks on day 21 was significantly improved by enzyme supplementation. This effect disappeared with progress in experiment. In another experiment, feed intake was significantly improved with enzyme supplementation. Apart from this, no statistically significant improvement in performance could be detected. On overall average, the final BW of ducks fed an enzyme was (as compared to the unsupplemented control = 100), 100, and the feed conversion ratio was 101. There is no indication from the growth and feed conversion data that an enzyme effect becomes more pronounced with increasing inclusion rate of soluble NSP by rye. It is concluded that supplementary xylanases are efficient in reducing digesta viscosity in ducks fed diets with high inclusion of wheat, rye and triticale. No consistent effect of enzyme supplementation, however, can be expected on growth and feed conversion.  相似文献
