Coupling of immunomagnetic separation and polymerase chain reaction was used after preenrichment for the rapid detection of salmonellae in minced meat and chicken giblets. The primers used are specific to the origin of DNA replication (oriC) on the Salmonella chromosome and produce a 163 bp DNA fragment. The method allowed a screening of Salmonella free samples within 24 hours. It detected 5 serovars when inoculated in minced meat at a contamination level of 1 to 4 cfu/g. Compared with the method L 00.00-20 of the Official Collection of Methods of Analysis according to section 35 of the German Law on Food and Commodities it showed a sensitivity of 94% and a specificity of 87.5% for the examination of chicken giblets (n = 45) and a specificity of 100% for the examination of minced meat (n = 63).  相似文献

The prevalence of Salmonella in fresh pork cuttings in Denmark in the years 2002 and 2006 was investigated at retail and compared with the retail supply pattern. A total of 1025 and 3473 samples were taken in 2002 from butcher's shops and supermarkets, respectively. The corresponding numbers in 2006 were 259 from butchers' shops and 628 from supermarkets. In 2002, 1.2% of all samples were positive for Salmonella; butchers' shops and supermarkets had 1.8% and 1.0% positive samples, respectively. The overall prevalence in 2006 was 4.2%, with prevalence of 8.1% and 2.6% for butchers' shops and supermarkets, respectively. Hence, increases around 3- to 5-fold were found. There was neither observed any parallel increase in Salmonella positive carcasses in Danish slaughterhouses during the study period, nor were any changes in supply routes towards slaughterhouses with higher prevalence observed, which could explain the apparent increase. We hypothesize that hygiene levels and ability to avoid cross-contamination and prevent growth of the organism, in the meat processing chain after slaughter were the most likely responsible factors. Results from this study indicate that the hygiene performance, particularly at retail, has a significant impact on the occurrence of Salmonella. This implies that there is no direct link between slaughterhouse Salmonella surveillance data and the level of Salmonella contamination at retail. To improve risk assessment of Salmonella in fresh pork meat, this study underlines the need for comprehensive retail data.  相似文献

A total of 444 samples of raw chicken meat (thighs, breasts, wings, livers, gizzards, hearts and ovaries) that retailed at 145 different supermarkets in 47 prefectures in Japan were examined for contamination with Staphylococcus aureus in association with its enterotoxigenicity. S. aureus was isolated from 292 (65.8%) of the samples, and from 131 of the 145 supermarkets. There was no significant difference in the detection rate of S. aureus according to the type of meat examined. About 80% of 714 isolates belonged to the poultry (57.1%) and human biotypes (22.1%). Seventy-eight (21.7%) of 360 isolates were enterotoxigenic and isolated from 78 samples in 53 supermarkets in 31 prefectures. Staphylococcal enterotoxins (SEs) produced were SEB (50 isolates), SEA (14), SEC (8), SED (2), SEA+SEB (2), and SEA+SEC (2). Most of the enterotoxigenic isolates belonged to the human and poultry biotypes, coagulase type VII, VIII or IV, and were lysed by phages of group III. Identical SE types, biotypes, coagulase types and pulsed-field gel electrophoresis (PFGE) patterns were shown in isolates from different types of meat at the same supermarket and from samples taken from different supermarkets in the same prefectures or in isolates from samples obtained from several different prefectures. Among the 50 SEB-producing isolates, 27 yielded three similar PFGE patterns that differed by only a few fragments, suggesting that they were closely related genetically. The three patterns were found in isolates of samples that retailed at 17 supermarkets in 11 prefectures, indicating that they may be disseminated among raw chicken meat in Japan.  相似文献

A total of 200 poultry giblets, 50 each of chickens, ducks, squab and turkeys, were examined for the presence of Campylobacter jejuni. In chicken giblets, C. jejuni was isolated from gizzards, hearts, livers and spleens with incidences of 28%, 10%, 40% and 16% respectively while 24%, 6%, 36% and 10% of duck gizzards, hearts, livers and spleens were positive for the organism, respectively. C. jejuni was detected in 6% of squab gizzards, in 10% of squab livers but failed to be detected in squab hearts & spleens. In turkey giblets, 16% of gizzards, 4% of hearts, 30% of livers and 8% of spleens were positive for the organism. C. jejuni was more frequently isolated from liver samples than gizzard, spleen and heart samples, each constituting of 29%, 18.5%, 8.5% and 5%, respectively. High incidence of C. jejuni was recorded among chicken giblets (23.5%), followed by duck giblets (19%), then turkey giblets (14.5%) and finally squab giblets (4%).  相似文献

Human salmonellosis is a major public health problem worldwide. Infections can pass to humans by contact with contaminated substances in the food chain. This study aimed to determine the prevalence and contamination levels of Salmonella isolated from pork, chicken and beef sold in different types of retail stores in Chiang Mai and Lamphun provinces and to investigate the genetic relatedness among Salmonella isolates in food chains in that area. A total of 360 meat samples from supermarkets, mini‐grocery stores and fresh markets were obtained. Salmonella Rissen and S. Weltevreden were found in all meat sample types and in human cases. The overall prevalence of Salmonella in the chicken, pork and beef samples was 34.17%, 32.50% and 3.33%, respectively. Quantitatively, Salmonella contamination was highest in pork (1.24 log10MPN/g), followed by chicken (1.08 log10MPN/g), and beef (0.75 log10MPN/g). The highest frequency of Salmonella contamination was found at the fresh markets (85.71%), whereas the highest quantity of contamination level was from mini‐grocery stores (1.27 log10MPN/g). The rep‐PCR analysis results revealed that some of the Salmonella from meat samples and human cases were identical clones.  相似文献

Animal feed-additive samples (n = 300) were examined for the presence of salmonellae, using the selenite-F broth-enrichment method followed by subculturing on Salmonella-Shigella and brilliant green agar with sulfadiazine selective agar plates. Samples consisted of a variety of feed additives: 119 bone meal samples, 77 meat meal samples, 40 fish meal samples, and 64 miscellaneous meal samples. Results of examination found 49 (41.2%) of the bone meal samples, 6 (7.8%) of the meat meal samples and 2 (5%) of the fish meal samples contained salmonellae. Of 57 isolates representing 24 serotypes, 4 most frequently isolated serotypes were Salmonella meleagridis (35.1%), Salmonella tennessee (7%), Salmonella chester (5.2%), and Salmonella senftenberg (5.2%). This study shows a high Salmonella-contamination rate of bone meal compared with meat meal and fish meal samples. Of 12 known positive bone meal samples that were examined, 100% of 25-g samples, compared with 70% to 100% of 2.5-g samples and 30% to 90% of 0.25-g samples and 30% to 90% of 0.25-g samples, were positive for salmonellae.  相似文献

To estimate the prevalence and distribution of Salmonella in the chain from cattle to the consumer, faeces, mesenteric lymphnodes and beef cuts from 235 cattle, stool samples from 300 workers of the same Addis Ababa abattoir, and 330 minced beef samples from supermarkets in Addis Ababa were analyzed. Isolated Salmonella strains were serotyped and tested for antibiotic susceptibility. Low prevalence in faeces and lymphnodes, and higher contamination rates of beef cuts (diaphragm, abdominal muscles) indicate severe cross-contamination during slaughter. Animals of poor health status were far more frequently carriers of salmonellae, which stresses the need of intensive ante-mortem inspection on slaughter animals. During transport from slaughterhouse to the supermarkets, production and selling of minced beef, the prevalence of Salmonella did not increase.  相似文献

In March 1989, the USDA Food Safety and Inspection Service sampled raw chicken carcasses and giblets at a federally inspected slaughter establishment in Puerto Rico to determine the effects of adding chlorine to carcass and giblet chill water on bacterial contents of raw poultry products. Over four 8-hour workdays, 200 carcass rinse samples were collected at 3 sites in the establishment; 39 giblet rinse samples were collected at 1 site. Analyses of the carcass rinse samples indicated that carcasses had average aerobe plate counts of log10 3.20 before chilling and 2.51 after chilling; Enterobacteriaceae counts of log10 2.57 before chilling and 1.75 after chilling; and Escherichia coli counts of log10 2.04 before chilling and 1.20 after chilling. Salmonellae were found on 43% of the carcasses before chilling and on 46% after chilling. Analyses of the giblet and neck rinse samples indicated that raw giblets and necks after chilling had average aerobe plate count of log10 3.49, Enterobacteriaceae count of log10 2.57, and E coli count of log10 1.06. Salmonellae were found on 12% of the giblets and necks sampled. Results compared favorably with giblet and neck rinse sample results obtained during a baseline sampling study in November and December 1987. The baseline results indicated aerobe plate count of log10 3.72; Enterobacteriaceae count of log10 2.90; E coli count of log10 1.14; and salmonellae on 69% of the giblets and necks sampled. Placing raw chicken carcasses in chlorinated chill water reduced aerobe, Enterobacteriaceae, and E coli plate counts. The aim of the present study was to evaluate the bacterial quality of chicken (n = 96) and turkey (n = 96) marketed in Casablanca, Morocco. Poultry samples (n = 192) were collected randomly from traditional shops and supermarkets during 2 sampling periods (hot and cold seasons). The samples were analyzed for the presence and counts of various bacteria. Results indicated that aerobic plate counts and fecal coliforms were particularly high in all the samples analyzed. Escherichia coli, coagulase-positive Staphylococcus, Clostridium perfringens, Salmonella, and Listeria monocytogenes were detected, respectively, in 48.4, 10.4, 7.2, 1.6, and 0.5% of the poultry meat samples. The chicken and turkey samples contained, respectively, 25 and 33.3% of bacteria above the maximum limits established by the Moroccan regulatory standards. The highest bacterial counts in poultry meat product samples were recorded with the traditional slaughtering process during the hot season (P < 0.05). These high levels of microbial contamination and occurrence of pathogenic bacteria reflect the poor hygienic quality of poultry meat under these conditions.  相似文献

From 10 egg production poultry farms 1516 samples were collected and examined for the presence of salmonella. The samples were: 201 chicken, 36 sparrows, 35 rats, 35 pools of 20 flies each, 450 eggs, 60 mattresses, 188 feces, 425 feedstuffs and 86 water samples. Salmonellae were isolated only from 163 (10.8%) samples. From the 146 (89.6%) of these S. gallinarum was isolated. From the rest 17 (10.4%) the following mobile salmonella strains were isolated: two strains of S. virchow and Salmonella of subgroup II, four strains of S. typhimurium var. Copenhagen, seven strains of S. Livingstone, one S. enteritidis and one S. infantis The S. gallinarum was isolated from dead or sick chicken (46%), eggs (10.4%), rats Rattus norvegicus (14.3%) and mattresses 1.6%. The mobile salmonellae were isolated from feedstuffs (2%), flies (14.3%), rats (2.8%), feces (1%). From the present study, it seems that rats, chicken and eggs are important for the salmonella dissemination.  相似文献

Campylobacter was isolated from retail meat samples collected during the fiscal year 2009 in Japan. The higher percentages of contamination of chicken products were observed from June (39.3%) to November (83.3%). However, the highest number of human campylobacteriosis cases was reported in June in the Infectious Agents Surveillance Report. The chicken isolates with distinct clusters IVb and I, based on the restriction fragment length polymorphism of the flaA gene, were predominantly obtained during the periods between April and November 2009 and between February and March 2010, respectively. Extensive monitoring of Campylobacter contamination in chickens produced in various places is needed to analyse the seasonal variations between contamination of the meat products and the number of human cases with campylobacteriosis.  相似文献

A total of 39 Salmonella cultures isolated from raw minced beef and chicken (gizzard, liver, and heart) samples in Addis Ababa were examined for susceptibility to a group of 10 selected antimicrobials. 34 isolates (87.2%) were resistant to one or more antibiotics. The antibiotics to which isolated Salmonella strains were most often fully resistant included nitrofurantoin (48.7%), furazolidone (48.7%) and streptomycin (46.2%). Only 4 antimicrobials (gentamycin, kanamycin, rifampicin and sulphamethoxazole-trimethoprim) were effective against all Salmonella isolates with the exception of 2 which were intermediate in resistance to kanamycin (1) and sulphamethoxazole-trimethoprim (1). 77.8% of the S. Enteritidis strains showed multiple resistance to up to four antibiotics followed by S. Typhimurium (60.0%) and S. Dublin (33.3%). The high level of antibiotic resistance of foodborne Salmonella isolates in the study area is an indication of indiscriminate and continuous use of subtherapeutic doses of antibiotics in animals.  相似文献

Cultural monitoring was used to determine the incidence and sources of salmonellae in a 4160-bird broiler flock raised on litter in 32 pens. Twenty-five of the pens remained apparently free of salmonellae during the 49-day growing period. Salmonella johannesburg, first detected in the meat meal component of the starter ration, was recovered from the litter of seven pens and from the intestines of dead or culled chicks from two pens. Salmonella alachua was also recovered from two of these pens. Culture of swabs collected from the plastic crates used to transport this flock for processing showed that 97/112 (86.6%) were contaminated with salmonellae (15 serovars) before the birds were loaded. The crate washer at the plant did not remove salmonellae from these crates: 97/132 (73.5%) crates sampled after washing yielded salmonellae. Eleven serovar were recovered, including S. johannesburg and S. alachua introduced by the infected flock. Twelve of 31 chickens (38.7%) collected when the birds were unloaded at the processing plant were intestinal carriers of S. johannesburg and/or S. alachua and 29 (93.5%) were external carriers. Salmonella johannesburg, S. alachua and four other serovars were isolated from the feathers of these birds. Eleven of 25 (44%) carcasses tested from this flock yielded salmonellae. Salmonella johannesburg or S. alachua, first isolated from the infected flock, were recovered from five carcasses and S. haardt and S. Typhimurium, first isolated from the transport crates were recovered from six carcasses.  相似文献

Cultural monitoring was used to study the incidence and sources of salmonellae in a 4160 bird broiler flock during the growing period, transport and processing in a commercial plant. No salmonellae were isolated from any of 132 litter samples of 189 chickens cultured during the seven-week growing period, even though nest litter samples from four of the eight parent flocks yielded salmonellae and Salmonella worthington was isolated from the meat meal component of the grower ration. On arrival at the plant, 2/23 birds sampled carried S. infantis on their feathers, although intestinal cultures failed to yield salmonellae. Three of 18 processed carcasses samples yielded salmonellae (S. infantis, S. heidelberg, S. typhimurium var copenhagen). The most likely source of these salmonellae was the plastic transport crates, since 15/107 sampled before the birds were loaded yielded salmonellae (S. infantis, S. typhimurium). The crate washer at the plant did not reduce the incidence of Salmonella-contaminated crates, since 16/116 sampled after washing yielded salmonellae (S. infantis, S. typhimurium, S. heidelberg, S. schwarzengrund, S. albany).  相似文献

We studied serovars of Salmonella strains isolated from chicken and humans in Lithuania over the period from 2000 to 2004. Salmonella strains were isolated and identified according to the techniques recommended by International Organisation for Standardization (Microbiology of Food and Animal Feeding Stuff--Horizontal Method for the Detection of Salmonella, 1998, ISO, Geneva). The per cent of infected flocks with Salmonella in separate years was between 1.01% and 3.2% during the period of investigation. The contamination rate of broiler legs and breasts was higher (2.36% and 4.25%) than that of wings (0.82%). Eighteen serovars of Salmonella were identified from the total 300 isolated samples. The most prevalent serovars in chicken were Salmonella Enteritidis, Salmonella Infantis and Salmonella Typhimurium. Other serovars such as Salmonella Montevideo, Salmonella Djugu, Salmonella Isangi, Salmonella Bovismorbificans, Salmonella Mbankada, Salmonella Hadar were detected only in one to two samples. In general, similar serovars of Salmonella were found in humans and chicken (S. Enteritidis and S. Typhimurium), although distinct serovars were found only in humans or only in chicken. Analysis of the distribution of Salmonella serovars in humans during the seasons of the year indicated that the highest incidence of Salmonella was in Summer and in the beginning of Autumn. Analysis of the distribution of serovars during the study period indicated that there is a shift over time in both humans and chicken.  相似文献

Three flocks raised for broiler or roaster performance tests were studied to determine the incidence and sources of salmonellae during the growing period, transport and processing and to relate these to contamination of processed carcasses. Day old chicks in two of the tests, (tests IV and V), were treated with a culture of intestinal anaerobes derived from mature chickens. The incidence of salmonellae during the growing period was too low to permit any conclusions about the efficacy of this culture in preventing Salmonella infection, but it had no adverse effect on flock performance. Carcasses from all three flocks were contaminated with salmonellae. Although the test IV flock was raised free of salmonellae, 46% of the carcasses tested from this flock were contaminated. The apparent source was the transport crates, 99% of which yielded salmonellae before the flock was loaded. In test V, 92% of the carcasses tested yielded salmonellae. The apparent sources were: flock infection (apparently originating from the parent flock), contaminated crates, spread during transport, and plant contamination. The flock of test VI was infected with Salmonella albany, and 54% of the carcasses tested were contaminated with this serovar. Carcasses of chicks infected early in life were more likely to be contaminated than those of chickens which contacted salmonellae later in the growing period.  相似文献

为了了解食源性病原沙门菌在肉鸡产业链中的流行情况及其耐药性现状。本研究从上海某肉鸡屠宰加工企业分别采集待宰肉鸡、屠宰胴体和市售鸡肉样品388、200、127份,并参照GB/T4789.4-2010和PCR法进行沙门菌的分离鉴定。结果显示,共获得沙门菌73株。待宰肉鸡、屠宰胴体和市售鸡肉分别获得沙门菌9、48和16株,分离率依次为2.32%、17.5%和8.66%。采用微量肉汤稀释法测定分离菌株对12种抗菌药物的敏感性。药敏结果显示,所有菌株至少耐两种药物。其中对复方新诺明和多西环素的耐药率最高,分别为100%和95.89%;头孢唑啉、阿莫西林/克拉维酸、氯霉素、氨苄西林、萘啶酸的耐药率也均超过了40%,而对头孢噻呋、头孢曲松、庆大霉素、美罗培南的耐药率相对较低。结果表明,该地区肉鸡源沙门菌的耐药情况较严重,亟需加强养禽业抗菌药物的合理应用;同时沙门菌流行情况也对改进肉鸡屠宰加工工艺,降低沙门菌的污染提出了要求。  相似文献

OBJECTIVE: To compare the efficacy of a Salmonella bacterin and a modified live Salmonella ser. Choleraesuis vaccine on a commercial dairy. ANIMALS: 450 cows in late gestation and 80 calves. PROCEDURE: Group-1 cows (n = 150) were vaccinated once with a modified live S. Choleraesuis (serogroup C1) strain 54 (SC54) vaccine, group-2 cows (150) were vaccinated on enrollment and 30 days later with a Salmonella ser. Montevideo (serogroup C1) bacterin, and group-3 cows (150) served as unvaccinated controls. One gallon of colostrum harvested from the first 80 cows to calve was fed to each calf. Outcome assessments included fecal shedding of Salmonella spp for the first 10 days after parturition (cows) or birth (calves), milk production, involuntary culling rate, mastitis incidence, antimicrobial use, and mortality rate. RESULTS: Salmonellae were isolated from 306 of 309 (99%) cows and 64 of 74 (86.5%) calves. Shedding frequency was less in SC54-vaccinated cows and calves that received colostrum from those cows, compared with the other groups, and vaccination was specifically associated with less shedding of serogroup C1 salmonellae. Production data were similar among groups. CONCLUSIONS AND CLINICAL RELEVANCE: Vaccination of pregnant cows with an autogenous Salmonella bacterin had no effect on fecal shedding of salmonellae, whereas vaccination with a modified live S. Choleraesuis vaccine reduced the frequency of fecal shedding of serogroup C1 salmonellae during the peripartum period. A commercial S. Choleraesuis vaccine licensed for use in swine may be more efficacious than autogenous Salmonella bacterins on dairies infected with serogroup C1 salmonellae.  相似文献

OBJECTIVE: To evaluate bacterial and protozoal contamination of commercially available raw meat diets for dogs. DESIGN: Prospective longitudinal study. SAMPLE POPULATION: 240 samples from 20 raw meat diets for dogs (containing beef, lamb, chicken, or turkey), 24 samples from 2 dry dog foods, and 24 samples from 2 canned dog foods. PROCEDURE: Each product was purchased commercially on 4 dates approximately 2 months apart. Three samples from each product at each sampling period were evaluated via bacterial culture for non-type-specific Escherichia coli (NTSEC), Salmonella enterica, and Campylobacter spp. Antimicrobial susceptibility testing was performed on selected isolates. Polymerase chain reaction assays were used to detect DNA from Cryptosporidium spp, Neospora spp, and Toxoplasma spp in samples obtained in the third and fourth sampling periods. RESULTS: One hundred fifty-three of 288 (53%) samples were contaminated with NTSEC. Both raw and prepared foods contained NTSEC during at least 1 culture period. Salmonella enterica was recovered from 17 (5.9%) samples, all of which were raw meat products. Campylobacter spp was not isolated from any samples. In 91 of 288 (31.6%) samples, there was no gram-negative bacterial growth before enrichment and in 48 of 288 (16.7%) samples, there was no aerobic bacterial growth before enrichment. Susceptibility phenotypes were variable. Cryptosporidium spp DNA was detected in 3 samples. CONCLUSIONS AND CLINICAL RELEVANCE: Bacterial contamination is common in commercially available raw meat diets, suggesting that there is a risk of foodborne illness in dogs fed these diets as well possible risk for humans associated with the dogs or their environments.  相似文献

Two hundred faecal samples, collected from pigs at a Budapest abattoir, were examined for the presence of salmonellae. Ninety-six isolates, belonging to 7 different serotypes, were obtained. Salmonella derby (70.8%) was the most prevalent serotype. The other serotypes isolated were S. typhimurium (8.33%), S. bredeney (4.16%), S. agona (2.08%), S. infantis (9.22%), S. london (3.12%), and S. panama (2.08%). A modified rapid presumptive test to detect salmonellae in food and food ingredients was described by Hoben et al. (1973). This test medium was evaluated to determine its efficacy in detecting salmonellae in pig faecal samples. The test gave an efficiency of 88.67%. However, the incidence of false positive results was rather high (9.43%). The public health importance of the isolates obtained and the application of the rapid presumptive test at the farm level are discussed.  相似文献
