An in vivo experiment was conducted to monitor the changes in fermentation end products in the feces of weaning piglets due to the inclusion of selected fermentable carbohydrates in the diet. The experiment involved 3 groups of 16 piglets each. Specially raised piglets (neither antibiotics nor creep feeding) were weaned abruptly at 4 wk of age. The piglets were offered 1 of 2 dietary treatments [a control diet (CON), or a fermentable carbohydrate-enriched diet (CHO)] and were subjected to 1 of the 2 fasting treatments (fasting for 2 d at the beginning of the experimental period or nonfasting). Fecal samples were collected per rectum every day during the experimental period. Piglets were slaughtered at the end of the 10-d experimental period, and digesta samples were collected from different parts of the gastrointestinal tract (GIT): the first half of the small intestine, the second half of the small intestine, the cecum, and colon. The DM, VFA profile, and ammonia concentrations were analyzed from the fecal and digesta samples. Daily feed intake was also recorded. There was no difference in concentrations of VFA in feces between the treatment groups. Ammonia concentration was lower (P < 0.05) in piglets fed the CHO diet compared with those fed the CON diet in both feces and digesta from different parts of GIT. Fasting had no effect on fermentation end products in feces. This study demonstrated that the inclusion of fermentable carbohydrates in weanling diets reduces protein fermentation along the GIT and also reduced the fecal concentration of ammonia.  相似文献

For evaluation of physiologically significant organic anions in the colonic environment, 87 samples of normal feces were collected from the rectum of 15 calves less than 60 days old. The calves were fed milk replacer with free access to starter diet and hay. After fecal extraction with water, pH, D- and L-lactate, succinate, and volatile fatty acid (VFA) concentrations were determined. There was wide variation in fecal pH (4.3 to 7.7). Higher lactate concentrations were observed in feces samples with lower pH, and most of these samples were collected during the first 4 weeks of life. Elevated lactate concentrations included both the D- and L-isomers, and the D-isomer comprised approximately 30-50% of total lactate. Elevated succinate concentrations were observed in only 8 fecal samples, while other samples had lower or trace amounts of succinate. Elevated fecal succinate showed no relationship with fecal pH or VFA concentrations. Fecal VFA concentrations were lower in samples collected in the early postnatal stage, but fecal VFA concentrations were not necessarily related to age. We confirmed that fecal D- and L-lactate concentrations increased with a concomitant decrease of VFA in the acidic lumen of the colon, although acidic feces were not necessarily accompanied by elevated concentrations of lactate. OBJECTIVES: To describe the effects of an abrupt increase of concentrates in the diet of dairy cows on myoelectric activity of the spiral colon and on fermentation patterns in the rumen and large intestine. ANIMALS: 6 healthy lactating Simmental x Red-Holstein cows. PROCEDURES: The diet of 6 cows implanted with bipolar electrodes in the spiral colon was changed from hay only to a ration of 50% hay:50% starch-rich concentrates during a period of 60 hours. Myoelectric activity of the spiral colon, concentrations of absolute and undissociated volatile fatty acids (VFA), and pH of ruminal and large intestinal contents were monitored before, during, and after the dietary change. RESULTS: Significant changes in patterns of myoelectric activity of the spiral colon were restricted to phases III and IV of the bovine migrating myoelectric complex and to propagation velocity. Significant alterations were not observed in pH or VFA concentrations in ruminal fluid, but pH decreased and VFA concentrations increased significantly in fecal specimens after the change of diet. CONCLUSIONS AND CLINICAL RELEVANCE: Although rumen fluid is of limited value for measurement of certain indicators of fermentation, fecal samples can be used for measurement of pH and VFA concentrations, which serve as indicators of fermentation patterns in the large intestine. Increased concentrations of VFA and low pH in large intestinal digesta have a minimal influence on myoelectric activity of the spiral colon. Increased luminal VFA concentrations are unlikely to play an important role in the etiopathogenesis of motility disorders of the large intestine in cattle.  相似文献

To clarify the significance of fecal organic anions in neonatal diarrhea, a total of 252 fecal samples (91 diarrheic, 161 normal) were collected from 136 dairy calves (including three crossbreds) less than 4 weeks old. Fecal pH, D‐ and L‐lactate, succinate and volatile fatty acid (VFA) were analyzed. In normal feces, lactate was highest and VFA was lowest at week 1 of age, and lactate progressively decreased and VFA progressively increased with advancing age. In diarrheic samples, although higher pH and lower lactate levels were confirmed at week 1, samples at weeks 3–4 showed lower pH and VFA accompanied by higher lactate of D and L‐isomers. In diarrhea, fecal butyrate was significantly lower at all stages, but succinate levels did not differ significantly. The proportion of lactate to organic anions (sum of lactate, succinate and VFA) in diarrheic feces was lower at week 1, and higher in weeks 2–4, while that of VFA to organic anions showed the opposite pattern. Strong relationships were observed between fecal pH and lactate, and VFA proportions in organic anions, though the relationship was weak in diarrhea. Most of the elevated lactate was observed in fecal samples with lower VFA. However, succinate had no relationship with VFA or lactate levels.  相似文献

Normal fecal samples were taken from lactating cows fed either a total mixed ration (TMR; n = 30) or pasture‐based diet (20) and from dry cows fed mainly on hay (15). Diarrheic fecal samples (n = 51) were collected from 21 sick dairy cows. Fecal analyses of ammonia, urea, lactate and volatile fatty acid (VFA) levels were used to evaluate colonic fermentation. Most normal feces had reasonably neutral pH, however, alkaline feces were observed in diarrheic cows. Although fecal lactate is higher in cows on grazing pasture, lactate levels were generally lower in the cows in the present study. Fecal VFA levels were higher in lactating cows than in dry cows. Elevated fecal urea was observed in diarrheic cows, however, many fecal samples in normal and diarrheic cows contained no urea. Fecal VFA levels in diarrheic cows were lower than in normal lactating cows, but were approximately equivalent to those in dry cows. Grazing or dry cows showed higher acetate and lower n‐butyrate proportions compared with TMR‐fed or diarrheic cows. Higher proportions of branched chain VFAs were observed in diarrheic cows, and the lowest level was observed in grazing cows. The present results indicate that intracolonic nitrogen equilibrium and proteolytic fermentation are altered by diarrheic status.  相似文献

Three beef cattle diets were assessed for their potential to produce odorous compounds from cattle feces excreted during the growing and finishing periods. Eight pens containing 51 steers of varying proportions of Brahman and MARC III genotypes were fed either a chopped bromegrass hay diet or a corn silage diet for a 119-d growing period. After the growing period, all steers were switched to the same high-corn finishing diet (high corn) and fed to a target weight of 560 kg (finishing period). Fecal slurries were prepared from a composite of fresh fecal pats collected in each pen during both periods and incubated anaerobically. In additional flasks, starch, protein, or cellulose was added to the composite fecal subsamples to determine the preferred substrates for fermentation and odorous compound production. The content and composition of the fermentation products varied both initially and during the incubation, depending on the diet fed to the steers. The corn silage and high corn feces had the greater initial content of VFA (381.0 and 524.4 micromol/g of DM, respectively) compared with the bromegrass feces (139.3 micromol/g of DM) and accumulated more VFA than the bromegrass feces during the incubation. l-Lactic acid and VFA accumulation in the high corn and corn silage feces was at the expense of starch, based on starch loss and the production of straight-chain VFA. In the bromegrass feces, accumulation of branched-chain VFA and aromatic compounds and the low starch availability indicated that the protein in the feces was the primary source for odorous compound production. Substrate additions confirmed these conclusions. We conclude that starch availability was the primary factor determining accumulation and composition of malodorous fermentation products, and when starch was unavailable, fecal microorganisms utilized protein.  相似文献

Necropsy of 4 neonatal calves with a peracute syndrome of abdominal distention, diarrhea, dehydration, shock, and death revealed abomasal tympany, marked edema, hemorrhage, and emphysema of ruminal and abomasal walls and histopathologic lesions characteristic of forestomach acidosis. The presence of acid damage and the production of substantial quantities of gas strongly suggested that the pathogenesis of the syndrome involved exuberant fermentation of intragastric substrate. This supposition led to attempts to experimentally induce the syndrome by dosing neonatal calves with an alimentary tract flora, followed by milk replacer supplemented with excessive fermentable carbohydrate (D-glucose and cornstarch). Two of the 5 calves thus treated developed a syndrome very similar to that observed in naturally affected calves. Literature suggests involvement of several Clostridial species, Sarcina spp., and possibly other microbes or a combination thereof to be involved in very similar syndromes. Therefore it is suggested that this complex of lesions and clinical signs is not dependent on a single etiologic agent. Rather, it is proposed that the natural occurrence of disease requires a quantity of highly fermentable substrate (starch, glucose, lactose, etc.) and a bacterial flora that is capable of rapidly fermenting that particular substrate with a resultant production of gas and acid.  相似文献

To clarify colonic fermentation during the perinatal period, 22 dairy calves less than 6 weeks old were used. They were given a milk replacer following colostrum feeding. A total 100 samples of normal feces including meconium were collected from the rectum of the calves. Fecal pH, alcohols, lactate and volatile fatty acids (VFAs) were analyzed. Higher ethanol and n -propanol concentrations were found in many fecal samples particularly in the first 2 weeks after birth, but these metabolites showed consistently lower concentrations thereafter. By contrast, higher concentrations of methanol were observed in some samples for all ages examined. Fecal VFA increased abruptly within a few days of birth, and mainly consisted of acetate and n -butyrate. During the first 2 weeks, the proportion of n -butyrate in VFAs decreased and that of propionate increased gradually. Proportions of VFAs were almost stable at 3–6 weeks of age (acetate, propionate and n -butyrate in increasing order). Higher concentrations of lactate and lower pHs were observed in the fecal samples during the first 2 weeks, and concentrations decreased thereafter. Accelerated colonic production of ethanol and n -propanol was confirmed during the early 2 weeks, in addition to organic acid fermentation as reported previously.  相似文献

Fructooligosaccharides (FOS) and inulin may modulate hindgut fermentation. It was tested if digesta batch cultures taken from horses adapted to FOS and inulin show different fermentation compared with such taken from nonsupplemented horses. Six horses received 0.15 g FOS and inulin/kg body weight/d via Jerusalem artichoke meal (JAM) upon a hay-based diet; six horses received corncob meal without grains (CMG) as placebo. The horses were euthanized after 20 days. Digesta samples were taken from stomach, cecum, ventral colon ascendens (VCA), and colon transversum (CT). Digesta batch cultures were incubated 48 hours to measure in vitro gas production as well as pre- and post-incubation pH and oxidation-reduction potential (ORP). A distinct fermentation of the surplus of fructans present in the inoculum was found with JAM-adapted batch cultures. Gas production was accelerated in inoculated gastric contents of horses adapted to JAM compared with CMG adapted ones (7.8 vs. 16.4 hours to achieve half of the 48 hours gas quantity, respectively; P > .05). Although buffered, pH decreased during fermentation. Postincubation pH was lower with JAM than CMG-adapted batch cultures (P > .05). Preinoculation ORP was lower with stomach batch cultures adapted to CMG than with such adapted to JAM. The ORP increased twofold from pre- to post-incubation with the latter. Asymptotic maximal gas production decreased gradually using cecum, VCA, or CT digesta. Parts of FOS and inulin of digesta are fermented in the stomach, which reduce possible effects on hindgut fermentation. Elevated fermentation may considerably impact stomach health.  相似文献

The effect of hay type on the microbiome of the equine gastrointestinal tract is relatively unexplored. Our objective was to characterize the cecal and fecal microbiome of mature horses consuming alfalfa or Smooth Bromegrass (brome) hay. Six cecally cannulated horses were used in a split-plot design run as a crossover in two periods. The whole plot treatment was ad libitum access to brome or alfalfa hay fed over two 21-d acclimation periods with subplots of sampling location (cecum and rectum) and sampling hour. Each acclimation period was followed by a 24-h collection period where cecal and fecal samples were collected every 3 h for analysis of pH and volatile fatty acids (VFA). Fecal and cecal samples were pooled and sent to a commercial lab (MR DNA, Shallowater, TX) for the amplification of the V4 region of the 16S rRNA gene and sequenced using Illumina HiSeq. The main effects of hay on VFA, pH, and taxonomic abundances were analyzed using the MIXED procedure of SAS 9.4 with fixed effects of hay, hour, location, period, and all possible interactions and random effect of horse. Alpha and beta diversities were analyzed using the R Dame package. Horses fed alfalfa had greater fecal than cecal pH (P ≤ 0.05), whereas horses fed brome had greater cecal than fecal pH (P ≤ 0.05). Regardless of hay type, total VFA concentrations were greater (P ≤ 0.05) in the cecum than in feces, and alfalfa resulted in greater (P ≤ 0.05) VFA concentrations than brome in both sampling locations. Alpha diversity was greater (P ≤ 0.05) in fecal compared with cecal samples. Microbial community structure within each sampling location and hay type differed from one another (P ≤ 0.05). Bacteroidetes were greater (P ≤ 0.05) in the cecum compared with the rectum, regardless of hay type. Firmicutes and Firmicutes:Bacteroidetes were greater (P ≤ 0.05) in the feces compared with cecal samples of alfalfa-fed horses. In all, fermentation parameters and bacterial abundances were impacted by hay type and sampling location in the hindgut.  相似文献

Two experiments were conducted to study the effects of alterations in ruminal pH and volatile fatty acid (VFA) concentrations on utilization of Mg and other minerals. In Exp. 1, two metabolism trials were conducted with 12 ruminally cannulated crossbred wethers fed 800 g/d of orchard-grass (Dactylis glomerata, L.) hay. After each feeding, wethers were ruminally infused with 500 ml (4.2 ml/min) or either 1) deionized water, 2) 40% (w/v) glucose solution, 3) .26 M propionic and .17 M butyric acid solution or 4) .35 M HCl. The pH of the VFA solution was adjusted to 6.8 with 10N NaOH. In Exp. 2, a metabolism trial was conducted with 12 ruminally cannulated crossbred wethers fed 600 g of orchard-grass hay and infused with a buffered VFA solution prepared as in Exp. 1 or with an unbuffered solution. In both experiments each trial consisted of a 5-d adaption period followed by four 5-d collections of feed, feces and urine. Compared with the glucose treatment, infusion of the buffered VFA solution produced similar acetic and propionic and higher (P less than .05) butyric acid concentrations (Exp. 1). The HCl solution produced changes in ruminal and pH values similar to those of the glucose infusion. In Exp. 1, apparent absorption of Mg was increased over twofold by the glucose infusion (P less than .05), but the other infusions had no effect. Apparent absorption of P was decreased (P less than .05) by HCl infusion, and K absorption was decreased by HCl and glucose infusions. In Exp. 2, infusion of the unbuffered VFA solution decreased apparent Mg absorption by 15.7%, compared with infusion of the buffered solution. These experiments suggest that the increased Mg absorption observed with carbohydrate supplementation is not due to alterations in ruminal pH or VFA levels.  相似文献

Pasture-induced laminitis in the horse is associated with the overconsumption of fermentable carbohydrate, in the form of simple sugars, fructans, or starch. The fermentation of carbohydrate in the cecum and large intestine results in the production of lactic acid and other toxins or "laminitis trigger factors." Vasoactive amines have been suggested as possible initiating factors. The aim of this study was to feed a commercially available form of fructan carbohydrate (inulin, 3 g/kg of BW per day) to normal ponies and to ponies predisposed to laminitis, to mimic a change from a basal hay diet to lush spring-summer pasture. Five normal and 6 laminitis-prone, native-breed ponies were acclimated to a basal hay diet before the inclusion of inulin and chopped dried grass. Blood samples, fecal samples, and foot temperature measurements were taken throughout the study. Amines were measured in the feces and plasma by HPLC and liquid chromatography-mass spectrometry, respectively. The pH of the fecal samples decreased from 6.89 +/- 0.11 on the hay diet to a minimum of 6.18 +/- 0.11 with the addition of inulin (P <0.05). An increase was observed in the fecal concentrations of a number of amines, including tryptamine (2.5-fold increase, P <0.05) and tyramine (2-fold increase, P <0.05). No changes were noted in plasma amine concentrations or plasma D- or L-lactate, indicating that there may be a threshold of hindgut pH change before mucosal damage can result in the release of these factors into the circulation. No differences in pH or any of the measured compounds were observed between the group of normal ponies and those predisposed to laminitis. This indicates that differences in the intestinal microflora do not account for this predisposition. However, the results from this study indicate that moderate increases in dietary fructan carbohydrate can produce increases in bacterial fermentation products and other compounds in the large intestine, which may be relevant to the pathogenesis of acute laminitis in ponies on pasture.  相似文献

ABSTRACT

1. This study determined the effects of three protein sources (PS), each at two digestibility crude protein (DCP) levels, on performance, gut morphology and fermentation characteristics in the hindgut of broilers.

2. It was hypothesised that broilers fed ingredients high in indigestible CP, i.e. rapeseed meal (RSM) or maize gluten (MG), could potentially cause reduced growth, impaired gut health, and more protein fermentation products in caecal digesta. Increasing the DCP level in each of the indigestible CP diets may compensate for these detrimental effects.

3. In total, 288 one-d-old male Ross 308 broilers were used in a completely randomised 3 × 2 factorial design, with six replicate pens per treatment. Three PS: soybean meal (SBM), rapeseed meal (RSM) or maize gluten (MG), and two DCP levels: 15.8 and 17.2% were used.

4. Broilers fed SBM had increased feed intake and BWG and improved FCR compared with those fed RSM and MG diets. Broilers fed high DCP had better performance compared with those on low DCP. No significant effects of PS or DCP level were found on gastrointestinal tract development, caecal ammonia or volatile fatty acid concentrations.

5. Broilers fed SBM had longer villi, smaller crypts and increased villus height to crypt depth ratio compared with those fed RSM and MG diets. Broilers fed RSM diet had a lower caecal pH, and had 16.5% and 14.9% more branched chain fatty acid contents in caecal digesta compared with those fed SBM and MG diets, respectively, indicating more proteolytic fermentation.

6. Replacing SBM by RSM and MG negatively affected growth performance and gut morphology. Hindgut protein fermentation was substantially increased in RSM fed birds.

7. To a certain extent, retarded growth performance in RSM and MG fed birds could be counterbalanced by increasing the dietary level of digestible CP.  相似文献

The objective of this study was to evaluate dietary galactooligosaccharide (Gal OS) addition on swine nutrient digestibility, ileal and fecal bacterial populations, and ileal short-chain fatty acid (SCFA) production, and to determine their impact on ileal fermentative characteristics in vitro. Twelve T-cannulated pigs (BW = 25 kg) were fed a diet free of Gal OS for 21 d. On d 22, ileal digesta samples were collected for an in vitro fermentation experiment (Exp. 1). Substrates included: raffinose/stachyose combination (R + S), soy solubles (SS), and transgalactooligosaccharides (TOS). Also included were the non-OS components of SS and TOS. Nine pigs (three donors per treatment) served as ileal effluent donors. Each substrate was fermented in vitro for 6 h, and pH and SCFA and gas production were determined. Pigs then were allotted to three treatments: a Gal OS-free control diet and the control diet with either 3.5% added Gal OS from SS or TOS. Diets, feces, and digesta samples collected weekly for 6 wk on d 6 (feces) and 7 (digesta) were analyzed for DM, OM, CP, and chromic oxide concentrations. Feces and ileal digesta were analyzed for bifidobacteria and lactobacilli populations. Ileal digesta samples were analyzed for SCFA. On d 64, a second in vitro fermentation experiment (Exp. 2) was conducted using ileal effluent from three pigs per treatment and the same substrates used in Exp. 1. In vivo results showed that ileal and total tract DM and OM digestion were decreased (P < 0.05) by addition of both SS and TOS to the diet. Ileal and total-tract N digestibilities were decreased (P < 0.05) by dietary addition of SS. Fecal bifidobacteria and lactobacilli were increased (P < 0.05) by addition of SS and TOS to the diet. Ileal propionate and butyrate concentrations were greater (P < 0.05) for pigs fed diets containing both sources of Gal OS. In vitro results showed that fermentation data were not affected by donor animal adaptation to treatment. For both in vitro experiments, gas and SCFA production were higher (P < 0.05) for R + S than for SS or TOS. Fermentation of R + S resulted in a higher pH (P < 0.05) than did SS or TOS. Fermentation of non-OS components of SS and TOS resulted in more (P < 0.05) gas and SCFA production, and pH values that did not differ (P > 0.05) compared to SS and TOS. The Gal OS used in this study were prebiotics, increasing beneficial bacteria in vivo and SCFA concentrations both in vivo and in vitro.  相似文献

Two experiments with growing pigs were conducted to determine the effects of dietary P and Ca level, phytase supplementation, and ileal pectin infusion on ileal and fecal P and Ca balance, chemical composition of fecal mixed bacterial mass (MBM), and bacterial metabolic activity. Pigs (initial BW = 30 kg) were fitted with simple T-cannulas at the distal ileum. They were fed a low-P corn-soybean meal control diet (3 g of P/kg) or the control diet supplemented with monocalcium phosphate (MCP; 7 g of P/kg; Exp. 1) or 1,000 FTU phytase/kg (Exp. 2). The daily infusion treatments consisted of 60 g of pectin dissolved in 1.8 L of demineralized water or 1.8 L of demineralized water as the control infusion, infused via the ileal cannula. In each experiment, 8 barrows were assigned to 4 dietary treatments according to a double, incomplete 4 x 2 Latin square. The dietary treatments in Exp. 1 were the control (Con-) diet with water infusion; the control (Con+) diet with pectin infusion; the MCP diet with water infusion; and the MCP diet with pectin infusion. In Exp. 2, the pigs received the same Con- and Con+ treatments as in Exp. 1 and, in addition, the phytase-supplemented diet in combination with water or pectin infusion. In this study, the effect of feeding different types and amounts of dietary fiber (DF) on luminal environment and morphology in the small and large intestine of sows was studied. Three diets, a low-fiber diet (LF) and 2 high-fiber diets (high fiber 1, HF1, and high fiber 2, HF2) were used. Diet LF (DF, 17%; soluble DF 4.6%) was based on wheat and barley, whereas the 2 high-fiber diets (HF1: DF, 43%; soluble DF, 11.0%; and HF2: DF, 45%; soluble DF, 7.6%) were based on wheat and barley supplemented with different coproducts from the vegetable food and agroindustry (HF1 and HF2: sugar beet pulp, potato pulp, and pectin residue; HF2: brewers spent grain, seed residue, and pea hull). The diets were fed for a 4-wk period to 12 sows (4 receiving each diet). Thereafter, the sows were killed 4 h postfeeding, and digesta and tissue samples were collected from various parts of the small and large intestine. The carbohydrates in the LF diet were well digested in the small intestine, resulting in less digesta in all segments of the intestinal tract. The fermentation of nonstarch polysaccharides in the large intestine was affected by the chemical composition and physicochemical properties. The digesta from pigs fed the LF diet provided low levels of fermentable carbohydrates that were depleted in proximal colon, whereas for pigs fed the 2 high-DF diets, the digesta was depleted of fermentable carbohydrates at more distal locations of the colon. The consequence was an increased retention time, greater DM percentage, decreased amount of material, and a decreased tissue weight after feeding the LF diet compared with the HF diets. The concentration of short-chain fatty acids was consistent with the fermentability of carbohydrates in the large intestine, but there was no effect of the dietary composition on the molar short-chain fatty acid proportions. It was further shown that feeding the diet providing the greatest amount of fermentable carbohydrates (diet HF1, which was high in soluble DF) resulted in significant morphological changes in the colon compared with the LF diet.  相似文献

Management of the performance horse often incorporates meal feeding of highly digestible starches and reduced access to high-fiber forage. Such regimens are associated with equine gastric ulceration syndrome (EGUS) and can alter hindgut homeostasis. In-feed buffering of gastric contents and promotion of energy derivation from high-fiber forage in the hindgut are therefore desirable properties of a nutritional supplement. A marine-derived, multimineral supplement with known buffering properties containing calcium, magnesium, and short-chain fructo-oligosaccharides (scFOS) was tested under in vitro simulations of equine stomach and hindgut conditions. Six fiber:concentrate diets were incubated for 4 hours with or without the supplement at 37°C in pepsin HCl solution adjusted to pH 4.1 and 2.6. pH was measured at 1, 2, and 4 hours postincubation. Highest overall pH values were observed with the high cereal feeds; however, the supplement significantly increased (P < .001) the pH across all feeds by 0.17 and 0.19 for feeds incubated at pH 4.1 and 2.6, respectively. A gas production technique was used to measure the fermentation of four fiber:concentrate diets with and without additional supplement, using equine feces as the microbial inoculum. Addition of the supplement decreased (P < .05) the lag time and increased the initial fermentation rate, although as the incubation continued, this effect was reduced. These results demonstrate that the supplement had a significant buffering action for 4-6 hours under simulated in vitro stomach digestion conditions and also stimulated in vitro hindgut fermentation activities.  相似文献

Short-chain fructooligosaccharides (FOS) were supplemented to the diets of nine quarter horses ranging in age from 489 to 539 d with initial BW averaging 400.6 +/- 21.2 kg. The objectives of this study were to determine the effects of dietary FOS on the fecal responses in terms of pH, the microbial population, and VFA concentrations. The horses were used in a 3 x 3 replicated Latin square design, fed according to NRC requirements, and their individual diets were supplemented with no FOS (CON), 8 g of FOS/d (LOW), or 24 g of FOS/d (HIGH) over three 10-d feeding periods. On the last 3 d of each 10-d feeding period, a single fecal sample was collected between 0730 and 0930. Fecal pH decreased linearly (P = 0.01) from 6.48 with the CON diet to 6.38 with the HIGH diet, but there was no change (P = 0.19 for linear effect) in fecal consistency among treatments. A quadratic effect (P < 0.01) was observed for fecal Escherichia coli population, but no difference (P = 0.88 for linear effect) was found in fecal Lactobacilli enumeration among treatments. The presence of fecal Bifidobacteria was unable to be confirmed and was therefore not reported. Fecal acetate concentrations increased linearly (P = 0.03), with means of 2.13, 2.18, and 2.52 mg/g of wet feces for CON, LOW, and HIGH treatments, respectively. Similarly, fecal propionate concentrations increased linearly (P = 0.01), with means of 0.58, 0.64, and 0.73 mg/g for CON, LOW, and HIGH treatments, respectively. Fecal butyrate concentrations also increased linearly (P = 0.02), with means of 0.40, 0.46, and 0.54 mg/g for CON, LOW, and HIGH treatments, respectively. Total VFA (P = 0.01) and lactate (P = 0.02) concentrations increased linearly, with total VFA means of 3.47, 3.69, and 4.25 mg/g for CON, LOW, and HIGH treatments, respectively, and lactate means of 0.36, 0.41, and 0.47 mg/g for CON, LOW, and HIGH treatments, respectively. Supplementing FOS in diets fed to yearling horses altered fecal microbial populations, fecal VFA concentrations, and pH.  相似文献

The effects of two types of nondigestible oligosaccharides (NDO), fructooligosaccharides (FOS), and transgalactooligosaccharides (TOS) were studied on growing and weanling pigs' nutrient digestion. Dietary NDO were included at the expense of purified cellulose. Twenty-five 57-d-old growing pigs, averaging 15.9+/-.6 kg on d 0 of the experiment, were fed a corn-based control diet or the control with 6.8 or 13.5 g of FOS/kg or 4.0 or 8.0 g of TOS/kg (five pigs per diet). Feces were collected on d 28 to 32, and small-intestinal digesta were collected (slaughter technique) on d 42 to 47 of the experiment. Feeds, feces, and digesta were analyzed for DM, inorganic matter, CP, ether extract, and crude fiber. Dietary NDO did not significantly affect apparent fecal and small intestinal digestion of nutrients in growing pigs. After being fed a NDO-free diet through d 10 after weaning, 38-d-old weanling pigs (n = 20), averaging 10.4+/-.8 kg on d 0 of the experiment, were fed a control diet (based on cornstarch, casein, and oat husk meal) or the control with 10 or 40 g of FOS or TOS/kg (four pigs per diet). Feces and urine were collected on d 13 to 17, and ileal digesta were collected via a postvalve T-cecum cannula on d 33 to 37 of the experiment. Feeds, feces, and digesta were analyzed for DM, inorganic matter, CP, ether extract, starch, NDF, ADF, ADL, Ca, P, Mg, Fe, Cu, and Zn. Nonstarch neutral-detergent soluble carbohydrates (NNSC) completed the mass balance for the carbohydrates. Urine was analyzed for N and minerals. The apparent fecal digestion of NNSC increased in the NDO-supplemented diets. The TOS-fed pigs tended (P<.10) to have a higher apparent fecal digestion of CP than the FOS-fed and control pigs but excreted more N via the urine (P<.01). Nitrogen and mineral balances were not affected. The FOS was nearly completely degraded prececally. Mean fiber digestion was lower at the fecal compared with the ileal level, as was the extent of NDO effects. This indicates that fiber digestion requires more than 2 wk to adapt to dietary NDO. Apparent ileal digestion of hemicellulose increased for the NDO-supplemented diets (P<.05), but that of NNSC decreased (P<.001). Thus, under the well-controlled conditions of this experiment, dietary NDO hardly affected nutrient digestion in well-kept growing and weanling pigs. However, digestion of dietary nonstarch carbohydrates may be affected.  相似文献

1. A standard methodology was developed for the determination of the viscosity of digesta in broiler chickens. 2. The best variables considered for use (those which gave the most consistent results) were: digesta obtained from the foregut area (defined as the region from the pancreas to Meckel's diverticulum) using a minimum of 6 replicates and offering experimental diets for 5 d before slaughter at 25 d of age. 3. There was evidence that the viscosity of digesta obtained from the hindgut (defined as the region between Meckel's diverticulum and the ileo-caecal junction) was reduced the longer the birds were fed on one diet, suggesting possible dietary acclimatisation. Hindgut digesta viscosity from birds fed on a barley-based diet over a longer period was 10% to 30% less than that of birds fed for 5 d. 4. There was an apparent reduction in viscosity with age of bird up to 45 d of age, with hindgut digesta viscosity from birds fed on barley-based diets falling from 20.6 cps at 25 d to 9.8 cps at 45 d. 5. In an assessment of the procedure developed, the following digesta viscosities were obtained at 25 d: 3.1 cps (wheat-based diet, foregut), 19.8 cps (barley-based diet, foregut), 5.7 cps (wheat-based diet, hindgut), 22.8 cps (barley-based diet, hindgut).  相似文献
