The effect of acepromazine maleate, xylazine and thiopentone on the packed cell volume, plasma protein content, factor VIII activity and von Willebrand factor antigen concentration of blood was studied in normal dogs. The same variables were measured in dogs with haemophilia A given acepromazine maleate and thiopentone. Both the packed cell volume and plasma protein content decreased after the administration of either acepromazine maleate or xylazine. Values were not changed further after administration of thiopentone. Changes in the haemostatic variables measured were generally small. Consequently, blood samples collected from dogs under the influence of premedicant doses of acepromazine maleate or xylazine, and when subsequently anaesthetised with thiopentone, are adequate for the assay of factor VIII activity and von Willebrand factor antigen concentration for establishing an animal's haemophilia A and von Willebrand's disease status.  相似文献

To evaluate indices of renal function in healthy, growing Beagle puppies from 9 to 27 weeks of age and to determine whether indices change with age during this period. Animals-6 healthy Beagle puppies. PROCEDURE: Urine collections were performed at 2-week intervals in puppies 9 to 27 weeks old. Daily excretion of urinary creatinine, protein, sodium, potassium, chloride, phosphorus, and calcium were determined, as were quantitative urinalyses including endogenous creatinine clearance, urine protein-to-creatinine ratios (UPr/C), and fractional clearances of sodium (FNa), potassium (FK), chloride (FCI), calcium (FCa), and phosphorus (FP). RESULTS: Significant differences among age groups were detected for endogenous creatinine clearance, and daily urinary protein, potassium, calcium, and phosphorus excretion. Significant differences also existed among age groups for UPr/C, FNa, FK, FCI and FP. Age-related effects fit a linear regression model for FNa, UPr/C, daily phosphorus excretion, and daily protein excretion. Quadratic regression models were judged most appropriate for endogenous creatinine clearance, FK, daily chloride excretion, and daily potassium excretion. Endogenous creatinine clearance measurements higher than adult reference ranges were observed from 9 to 21 weeks of age. The FNa, FK, FCI, FCa, and FP were slightly higher than those reported for adult dogs. CONCLUSIONS AND CLINICAL RELEVANCE: Selected results of quantitative urinalyses in healthy 9- to 27-week-old Beagle puppies differ with age and differ from those measured in adult dogs. Diagnostic measurements performed in puppies of this age range should be compared with age-matched results when possible.  相似文献

Sera from 154 West Highland White Terrier puppies between 6 and 12 weeks of age were assayed for total IgE using a sandwich ELISA method. Development of clinical signs of atopic dermatitis in these dogs was monitored by use of an annual owner questionnaire, until the dog reached 3 years of age. Of 114 evaluated dogs, skin disease severe enough to warrant veterinary examination was reported in 52 (46%) during the three study years. A diagnosis of atopic dermatitis was made by the attending veterinarian in 28 dogs (25%). Certain litters had an especially high prevalence of apparently atopic dogs, consistent with the genetic predisposition towards atopy in this breed, but clear evidence of consistent heritability was not present. The median IgE concentration in 154 puppies at 6–12 weeks of age was 0.9 units mL−1, with a skewed distribution. Significant ( P < 0.01) variation in serum IgE concentrations was observed between litters, with median serum IgE concentrations for a litter ranging from 0 to 27.7 units mL−1. The median serum IgE concentration in puppies that later developed clinical signs of atopic dermatitis was not significantly different from that of puppies that remained healthy. There were no apparent correlations or significant differences found between serum IgE concentration as a puppy, parental history of skin disease, and subsequent emergence of clinical signs of atopic dermatitis. We conclude that early total serum IgE determinations seem to have little usefulness in predicting the later onset of atopic dermatitis in this breed.  相似文献

OBJECTIVE--To determine the long-term effects of juvenile pubic symphysiodesis (JPS) in dysplastic puppies. STUDY DESIGN--Prospective, randomized, clinical trial. ANIMALS--Seven dysplastic Chesapeake Bay retrievers and 2 beagle-crosses (BX1 and 2). METHODS--Five puppies had JPS performed with electrocauterization at 12, 16, 20, 22, and 24 weeks of age, respectively. Two puppies served as controls. BX1 and BX2 were used to obtain biopsies of the symphysis. Hips were evaluated for: pelvic development (transverse computerized tomography for acetabular angle [AA] and dorsal acetabular rim angle [DARA]); laxity [hip extended and stress radiography [distraction index (DI)]); Ortolani maneuver with reduction angles; acetabular coverage (Norberg angles); and function (coxofemoral range of motion, hip pain, and gait analysis by force-plate technique at 44 and 137 weeks of age). RESULTS--The pubis fused prematurely in every puppy that was operated on with the JPS technique. Greater acetabular responses were related to younger ages at surgery. The final mean AA in dogs that had JPS was 25 degrees greater than preoperative values; 40% increased over control. The DARA final mean was 10 degrees, 52% less than preoperative values and 46% less than control. The final mean DI in dogs having JPS was 0.28, 47% improved over preoperative values and 58% better than control. Mean pelvic dimensions in dogs that had JPS were 18% less than control. Gait analyses were normal for all dogs at 137 weeks. No urinary or bowel complications occurred. CONCLUSIONS--Significant ventrolateral acetabular rotation, increased hip coverage, diminished hip laxity, normal pain-free gait, and insignificantly reduced pelvic size occurred after JPS. CLINICAL SIGNIFICANCE--Dysplastic hips in young dogs were significantly improved by JPS.  相似文献

OBJECTIVE: To optimize methods used to measure coagulation factor activities in canine plasma, define reference ranges in dogs, and compare activities between canine and human plasma. SAMPLE POPULATION: Human plasma samples (n = 5) and plasma from healthy dogs (140) and dogs with low factor V activity (7), high factor V activity (7), and low factor VIII:C activity (6). PROCEDURE: Coagulometric tests incorporated human plasma deficient in a single coagulation factor (human deficient plasma). Standard curves were generated with pooled plasma from 100 healthy dogs. Effect of sample dilution was evaluated, using plasma from dogs with high or low factor V activity and low factor VIII:C activity. Reference ranges for healthy dogs were established. Activities in human plasma were determined by comparison with standard curves obtained with canine plasma. RESULTS: Activities of factors V and VIII:C in samples diluted < or = 1:20 influenced results of tests for other coagulation factors. Activities of factors V and VIII:C in human plasma were significantly less than in canine plasma. For the other coagulation factors, significant differences in human plasma-to-canine plasma activity ratios were detected among different sample dilutions. CONCLUSIONS AND CLINICAL RELEVANCE: Accurate measurement of coagulation factor activities in canine plasma, using human deficient plasma, requires higher sample dilutions (ie, > 1:20) than typically used for human plasma. Differences in activities between human and canine plasma and nonparallelism of the standard curves emphasize the necessity for use of species-specific standard curves for accurate determination of coagulation factor activity.  相似文献

Objective: To determine a reference range for venous blood lactate concentrations in healthy neonatal dogs. Design: A prospective cohort study. Setting: All work was conducted at the College of Veterinary Medicine, Texas A&M University. Animals: Clinically healthy dogs: 68 puppies and 30 adults. Measurements and main results: A blood sample was collected from each puppy into lithium heparin via jugular venipuncture at 4, 10, 16, 28, 70, and 80 days of age. A single venous sample was collected from each adult dog. Lactate concentration in each sample was measured immediately using an automated analyzer. Two hundred seventy‐seven blood samples were analyzed. Blood lactate concentrations of adult dogs were 1.80±0.84 mmol/L (mean±SD). Mean blood lactate concentrations of puppies were significantly higher at 4, 10, 16, and 28 days of age compared with those of adult dogs. The reference range for lactate concentration for puppies at 4 days of age was 1.07–6.59, and for the puppies from 10 to 28 days of age was 0.80–4.60. Conclusions: Assessment of perfusion can be challenging in neonates due to normal physiologic variation and small size. Measurement of lactate is rapid, minimally invasive, and has potential to be a useful marker of perfusion in neonatal dogs. However, lactate concentrations of neonatal dogs in this study were significantly higher than those of adult dogs. Reference ranges for venous lactate concentrations in adult dogs should not be used for puppies younger than 70 days of age.  相似文献

Two litters of German shepherd dogs were evaluated for a haemorrhagic tendency that was characterised by excessive bleeding from the umbilicus at birth, haemorrhage and haematoma formation at vaccination, excessive bruising, and lameness due to haemarthrosis. Platelet counts, clotting times and Von Willebrand's factor (VWF) assays were assessed in all dogs. Factor VIII determination was performed in 1 puppy and its parents. Based on the clotting times and VWF assay, 6 puppies (4 male and 2 female) showed type I Von Willebrand's disease (VWD), 5 (4 male and 1 female) possible type II VWD, and 4 were unaffected. One puppy with possible type II VWD had very low factor VIII activity; its sire had a normal factor activity, whereas the dam was in the low-normal range. This article reports type I and possible type II VWD in 2 related litters of German shepherd dogs, the latter being rare in German shepherd dogs.  相似文献

Chemistry tests were conducted on serum from young Beagle dogs in order to deter mine the effect of age on these parameters. Blood was collected at regular intervals from 25 normal Beagle puppies (13 males and 12 females) at ages ranging from 2 weeks to 12 months. Serum chemistry profiles, protein electrophoresis and radioimmunoassays for thyroxine and triiodothyronine were included. Rapidly changing age related differences were observed in several parameters. Urea nitrogen, cholesterol, triglycerides, lactate dehydrogenase, thyroxine, glucose, gamma glutamyl transpeptidase, and total bilirubin values were elevated early in life, and decreased during the first 6 to 8 weeks, while alanine aminotransferase activity was low initially and increased during this period. Lactate dehydrogenase, thyroxine, gamma glutamyl transpeptidase, total bilirubin and alanine aminotransferase attained stability by 3 months, but the remaining parameters showed slight changes subsequently, gradually approaching adult values. More gradual age related changes were observed in other parameters. These included alkaline phosphatase, inorganic phosphorus and calcium values, which were higher in younger dogs, and creatinine, aspartate aminotransferase and total protein values, which were lower in younger dogs. Creatinine and aspartate aminotransferase values were stable by approximately 6 months; alkaline phosphatase, inorganic phosphorus, calcium and total protein values continued to change gradually up to 1 year.  相似文献

Numerous investigations and reports have covered the potential negative effects of an excess of calcium (Ca) in the skeletal development of young dogs. However, it still remains unclear why not all puppies, particularly of larger breeds, develop clinical signs of skeletal disorders after an overexposure to Ca. This trial with the small Beagle breed and a larger Foxhound-crossbred dog (with an adult weight of around 35 kg) investigated the effects of Ca-excess on parameters of bone growth. The employed small breed is known for its chondrodystrophic predispositions, no such data are reported for the slender hound-type Foxhound-crossbred dogs. The Ca-supply had no influence on weight development and general health of the dogs. The measurements of bone lengths and widths in X-rays of the forearm of Beagles and Foxhound-crossbred dogs at 6 weeks of age, and again after a period of overexposure to Ca at about 27 weeks of age, revealed a growth-reducing influence only in Beagles, without influence on clinical parameters of skeletal health.  相似文献

Myosin heavy chain (MHC) isoform expression was established in the masseter, axial and limb skeletal muscles in 18 young dogs from late prenatal to 6 months of age. Results obtained by immunohistochemical procedure using 11 monoclonal antibodies, SDS-PAGE electrophoresis, Western-blotting and enzyme-histochemical reactions were compared to those found in skeletal muscles of adult dogs. In the skeletal muscles of both fetuses in the last stages of development and neonatal dogs, the developmental isoforms MHC-emb and MHC-neo were prevalent. During the first 10 days after birth, the level of embryonic isoform expression declined, as did that of the neonatal isoform during the second and third weeks. Correspondingly, the expression of MHC-IIa, and later, of MHC-I increased. By 6 weeks of age the presence of MHC-IIx became obvious. This transitional pattern was similar in all muscles except in the rhomboideus, where secondary slow fibres were present before birth. In the other muscles, only one primary, centrally located, slow fibre was present in a fascicle and the secondary slow fibres evolved from developmentally fast fibres between the second and third weeks of age. Our results indicate that timing of muscle maturation depends on its activity right after birth. The fastest developing muscles were the masseter muscle and the diaphragm. They were followed by others, postural and locomotory muscles. A pronounced swap from developmental to adult isoforms was noted between the 4th and 6th week of age. At this age puppies are weaned and begin playing intensively and investigating their surroundings. The results of our study indicate that dog skeletal muscles exhibit an immature morphology at birth and develop relatively slowly compared to those of other domestic animals.  相似文献

BACKGROUND: Idiopathic childhood epilepsies with benign outcomes are well recognized in human medicine, but are not reported in veterinary literature. We recognized such a neurologic syndrome in Lagotto Romagnolo dogs. ANIMALS: Twenty-five Lagotto Romagnolo puppies from 9 different litters examined because of simple or complex focal seizures and 3 adult Lagotto Romagnolo dogs exhibiting similar clinical signs were used. METHODS: Clinical and diagnostic evaluations of affected dogs were conducted, including electromyography, electroencephalography, and other testing. RESULTS: Seizures in puppies began at 5 to 9 weeks of age and usually resolved spontaneously by 8 to 13 weeks. Those with the most severe seizures also had signs of neurologic disease between these seizures, including generalized ataxia and hypermetria. There were no abnormalities in routine laboratory screenings of blood, urine, and cerebrospinal fluid. Electromyography, brainstem auditory-evoked potentials, and magnetic resonance imaging revealed no specific and consistent abnormalities. Fourteen of 16 (87.5%) affected puppies and 2 of 3 (67%) adult dogs revealed epileptiform activity in the electroencephalogram. Histopathologic examination in 1 puppy and 1 adult dog revealed lesions of Purkinje cell inclusions and vacuolation of their axons restricted to the cerebellum. Pedigree analysis suggests an autosomal recessive mode of inheritance. CONCLUSIONS AND CLINICAL IMPORTANCE: This disorder, with simple or complex focal seizures and cerebellar lesions, represents a newly recognized epileptic syndrome in dogs.  相似文献

In the present study, six commercial reagents for the determination of the activated partial thromboplastin time (APTT) were compared with respect to their factor VIII:C and factor IX sensitivity for measurements of canine plasma. For this purpose, plasma with different levels of factor VIII:C or factor XI activity (100, 80, 70, 60, 50, 40, 30, 20, 15, and 10% [factor VIII:C: additionally 5%]) was prepared by mixing pool plasma with plasma of dogs with haemophilia A or B. Double measurements of three different sample mixtures were carried out for each activity level. The sensitivity of the reagents was measured first based on the ratios of the coagulation time to the 100% values. In addition, the single factor activity (F VIII:C/IX(X0.975)), whose accompanying APTT corresponded to the upper limit of the reference range (97.5%-quantile, n = 50) of the respective reagent, was determined graphically. The APTT reflected a decrease of factor IX activity generally more sensitive than a reduction of factor IX activity of an identical degree. Based on ratios distant differences respecting factor VIII:C and factor IX sensitivity were found between different reagents using two way analysis of variance (p < 0.05). Significant differences between various reagents were also found with respect to the F VIII:C(X0.975) and the F IX(X0.975). These corresponded to values between 27 and 50% or 32 and 64%, respectively, dependent on the reagent. As a result, the more sensitive reagents fulfilled the demands on the sensitivity of APTT in humans. Based on the latter criterion the highest sensitivity for both factors was found for the same reagent (Pathromtin) consisting of kaolin as a contact activator and human placental phospholipid. Respecting all proofed reagents, however, no relation was found between contact activator and single factor sensitivity.  相似文献

OBJECTIVE: To determine relative heart size in clinically normal puppies and assess whether relative heart size changes with growth. DESIGN: Prospective radiographic study. ANIMALS: 11 puppies without evidence of disease. PROCEDURE: Standardized measurements of the long and short axes of the heart, midthoracic vertebrae, and other structures were made at 3, 6, 12, and 36 months of age. Measurements were recorded in millimeters and number of thoracic vertebral lengths spanned by each dimension, measured caudally from T4 on lateral radiographic views. The long and short axis measurements of the heart, expressed in vertebral lengths, were added to yield vertebral heart size. RESULTS: Mean +/- SD vertebral heart sizes on lateral radiographic views at 3, 6, 12, and 36 months of age were 10.0 +/- 0.5, 9.8 +/- 0.4, 9.9 +/- 0.6, and 10.3 +/- 0.6 vertebrae, respectively. Significant differences were not detected. CONCLUSIONS AND CLINICAL RELEVANCE: Vertebral heart size measurements in puppies are within the reference range for adult dogs (9.7 +/- 0.5 vertebrae) and do not change significantly with growth to 3 years of age. Standards for determining cardiac enlargement are similar in puppies and adult dogs.  相似文献

The results of eye examinations of 741 rough collies for collie eye anomaly (CEA) are described. Of the examined dogs, 40-8 per cent had CEA, including, in most cases, chorioretinal dysplasia (CRD). 7-7 per cent had coloboma of the optic disc while 3–5 per cent had complications of CEA, either retinal detachment or intraocular bleeding. There was no sex difference nor was the frequency of the disease influenced by coat colour. After estimating the total frequencies of the different manifestations of the disease, the dogs were divided into two groups according to the age at first examination. In the group of dogs examined between seven weeks and three months old, 48-9 per cent had CEA, compared to a frequency of 25-6 per cent in the group of dogs examined after three months of age. All the puppies with coloboma also showed signs of CRD, while among the older dogs 11 of the 66 CEA cases reported showed signs of coloboma without CRD. Twenty-two dogs which had been examined as puppies and given the diagnosis CEA with minor chorioretinal changes were re-examined at an older age. Of these dogs, 15 had ophthalmoscopi-cally normal fundi at re-examination while seven were unchanged from the first examination.  相似文献

The aim of this study was to investigate the prevalence of wobbler disease within a Dobermann pinscher population from three geographical locations in New Zealand. The study population consisted of 138 adults (aged 1-13 years) and 32 puppies (aged 6 weeks to 11 months). Data collected for each dog included age, sex, geographic location, if a choker chain was used or not and, in adults, the following body measurements: dimensions of head length, head circumference, width between shoulders, neck length, height at withers and withers to rump length. In addition, lateral radiographs were taken of the caudal cervical vertebrae of each dog and the radiological abnormalities associated with wobbler disease scored, so that each dog could be assigned to one of three radiological groupings. Based upon a neurological examination, each animal was also placed into one of three neurological groupings. The relationship between radiological and neurological groupings and the independent variables was initially compared using a univariate and subsequently a multivariate analysis. It was found that 48.8% of the dogs investigated had some abnormal radiological sign associated with wobbler disease, and 32.0% of them showed neurological signs. Dogs with radiological signs of the disease were 5.56 times more likely to have neurological signs. Statistical analysis of the data indicated that more severe radiological and neurological abnormalities occurred in the older dogs. In addition, dogs located in Hawke's Bay region had less chance of showing radiological changes than dogs from the other two regions, Hamilton and Wellington. Twelve of the 32 puppies were examined for radiological and neurological changes over the first year of their life. No abnormalities were detected in puppies under 12 weeks of age, but 28% (n=9) of the 32 puppies over 3 months of age did show some radiological changes. Only 9% (n=3) of puppies showed any neurological signs. Although several pedigree lines were investigated, the lineage data were incomplete, and therefore there was no conclusive evidence that wobbler disease was an inherited trait. This study showed that, although the radiological signs of wobbler disease were present throughout a wide age range, the associated neurological changes tended to appear at a later age. In both instances, the severity of these changes increased with age.  相似文献

Multifocal retinopathy of Great Pyrenees dogs   总被引:1,自引:0,他引:1  
Forty-four related Great Pyrenees dogs were examined ophthalmoscopically. Focal retinal elevations, multiple gray-tan-pink subretinal patches, and discrete areas of tapetal hyper-reflectivity were seen in 19 dogs, ranging from 13 weeks to 10 years of age. These lesions varied in size from focal spots that were barely visible with the indirect ophthalmoscope to areas that were larger than the optic disc. Complete blood cell counts, serum biochemical profiles, urinalyses, and blood pressure measurements were completed on four affected dogs and all were within normal reference ranges. Photopic and scotopic electroretinography was completed and the a-wave and b-wave amplitudes and latencies were similar for affected and age-matched nonaffected Great Pyrenees and other normal dogs. Electroretinograms that were examined twice during a 3-year period on three affected adult dogs did not reveal significant progressive deterioration of the a or b-wave parameters. Fluorescein angiography was completed on four affected dogs of ages 1 (n = 2), 5, and 6 years. These angiograms were repeated in three of these dogs 1 year later. The blood ocular barrier was intact in these dogs but there was blocked choroidal fluorescence. Postmortem examination, light microscopy, scanning and transmission electron microscopy were performed on three affected puppies and two affected adult dogs. These examinations revealed that the lesions in the puppies were limited to bilateral multiple areas of retinal pigment epithelial vacuolation, hypertrophy, and apparent separation from Bruch's membrane, and multiple serous retinal detachments. The affected adult dogs had focal retinal degeneration and retinal pigment epithelial hypertrophy, hyperplasia and pigmentation. Pedigree analysis and test mating confirm that this condition is inherited, probably as an autosomal recessive trait. This condition develops at approximately 13 weeks of age and the focal areas of retinal detachment and retinal pigment epithelial vacuolation progress to permanent and stable focal areas of retinal degeneration, and retinal pigment epithelial hypertrophy and pigmentation.  相似文献

The aim of this paper is to report serum trypsin-like immunoreactivity (TLI) concentrations in the progeny of a pair of English setters. These dogs had a history of producing puppies with a high prevalence of exocrine pancreatic insufficiency (EPI) secondary to either congenital deficiency or early onset juvenile atrophy of pancreatic acinar cells. Serum TLI concentrations in the clinically healthy parents of the puppies with EPI were normal, as were serum TLI concentrations in the unaffected puppies. However, serum TLI concentrations were extremely low (less than 1 μg/litre) in three puppies at eight to 12 weeks of age when clinical signs of EPI were present. In addition, one of the three puppies, with low TLI concentrations at eight weeks of age, had serum TLI concentrations of 0.7 μg/litre at two weeks of age even though no clinical signs were evident at that time. These results suggest that serum TLI assay may be a useful diagnostic aid in identifying puppies at a very early age with this unusual form of EPI.  相似文献

In humans, obesity is closely associated with insulin resistance (IR) and dyslipidaemia. The purpose of this study was to explore the effect of age on metabolic disturbances related to obesity in dogs ( n  = 25). Three age-groups of dogs (puppies, young adults and mature adults) were overfed to induce obesity, and body composition, insulin sensitivity index (IIS) (euglycaemic–hyperinsulinaemic glucose clamp) and plasma lipids were measured. Fat mass was similar in the three obese groups (30 ± 1% in puppies, 34 ± 1% in young adults and 39 ± 1% in mature adults). In mature adults, body weight (BW) increased (+45%, p < 0.001) and IIS decreased (−60%, p < 0.001) over 22 weeks. In young adults, BW gain was similar but slower (60 weeks) and IIS decreased to a lesser extent (−49%, p < 0.001). Overfed puppies weighed 30% more (p < 0.01) than normally-fed control puppies, but there was no change in IIS. Unlike young and mature adults, obese puppies did not exhibit significant changes in triglycerides (TG) and free fatty acid concentrations. In conclusion, as in humans, obese dogs develop IR that is associated with high TG levels; however, younger animals may be better able to balance energy needs with energy consumption.  相似文献

Objective To measure intraocular pressure (IOP) in a group of dogs as puppies and young adults to determine if there is any change during maturation. Animals studied Thirty‐two healthy Labrador Retriever dogs. Procedures Intraocular pressure was measured using a Tonopen XL initially at approximately 6 weeks of age (T1), then again approximately 1 year later (T2). Exact ages were known based on whelp date. Results The dogs had marginally higher IOP OU at T2 (mean = 14.9 mmHg) compared to T1 (mean = 13.4 mmHg). However, the difference was not statistically significant. No differences were seen based on sex and litter. Intraocular pressure OD was statistically greater than OS at T1 but not at T2. Conclusions Normal values for intraocular pressure are the same in puppies and adults. The results of this study do not support the previously suggested theory that younger dogs have sustained increased IOP as a requirement to drive growth of the globe. However, it does not rule out the possibility that a dynamic relationship between intraocular pressure and expansion of the globe may exist.  相似文献
