Vaccines composed of either virulent or attenuated Eimeria spp. oocysts have been developed as an alternative to medication of feed with ionophore drugs or synthetic chemicals. The purpose of this study was to evaluate the use of gel-beads containing a mixture of Eimeria acervulina, Eimeria maxima, and Eimeria tenella oocysts as a vaccine against coccidiosis. Newly hatched chicks (Gallus gallus domesticus) were either sprayed with an aqueous suspension of Eimeria oocysts or were allowed to ingest feed containing Eimeria oocysts-incorporated gel-beads. Control day-old chicks were given an equivalent number of Eimeria oocysts (10(4) total) by oral gavage. After 3 days, chicks were randomly assigned to individual cages, and feces were collected between days 5 and 8 postinfection. All samples were processed for total Eimeria oocysts. At 4 wk of age, all chickens and a control nonimmunized group received a high-dose E acervulina, E maxima, and E. tenella challenge infection. Oocyst excretion by chicks fed gel-beads or inoculated by oral gavage was 10- to 100-fold greater than that of chicks spray-vaccinated with the Eimeria oocysts mixture (log 6.3-6.6 vs. log 4.8). Subsequent protection against challenge as measured by weight gain and feed conversion efficiency was significantly greater (P < 0.05) in gel-bead and oral gavage groups compared with spray-vaccinated or nonimmunized groups. Also, gel-bead and oral gavage groups showed no significant difference (P > 0.05) in weight gain and feed conversion efficiency compared with nonchallenged controls. These findings indicate that incorporation of Eimeria spp. oocysts in gel-beads may represent an effective way to deliver live oocyst vaccines to day-old chicks for preventing subsequent outbreaks of coccidiosis in the field.  相似文献

The present study was conducted to evaluate the adverse effects of an interaction between low levels of dietary aflatoxins (AF) and Eimeria tenella infection on broiler chicks. A set of 1-day-old chicks were raised for 35 days in the following groups: a control group, a group fed AF, a group fed AF and inoculated with E. tenella (AF + E.ten), and a group inoculated with E. tenella alone. AF in the contaminated diet were given at 200 ppb starting from the seventh day after hatching while E. tenella was inoculated at a dose of 5 × 104 sporulated oocysts per chick at the 14th day after hatching. Worsened performance traits and high mortality were all observed in the treated birds, particularly the AF + E.ten group. Lesion scores and oocyst outputs were not different within groups. Chickens fed with AF had significantly increased serum ALT and ALP activities as well as decreased albumin content. They also showed hepatomegaly, hepatocytic vacuolation and necrosis, an atrophied bursa of Fabricius, and a thymus with tissue depletion. E. tenella-infected broilers displayed a significant reduction in packed cell volume, hemoglobin content and lymphocyte percentage, and showed hemorrhagic typhlitis. The deficits in hepatic function and hematologic parameters as well as the gross pathological, and histopathological changes, were more common and more severe in the group that was exposed to both aflatoxicosis and coccidiosis than in the groups exposed to either treatment alone. Thus, the combination of aflatoxicosis and E. tenella infection may influence the course of coccidial infection due to additive effects.  相似文献

Broiler chicks were fed on a high energy, commercial‐type ration with 20 g oxycarotenoids/ton1 for 5 weeks. They were then transferred to two finisher rations: one with 20 g oxycarotenoids from the ingredients plus 3 g canthaxanthin/ton; the other containing 26 g oxycarotenoids/ton derived solely from the ingredients. Three replicated groups fed on each finisher ration were inoculated daily for 5 d with either 6000 sporulated oocysts of E. maxima, 30,000 sporulated oocysts of E. acervulina or no coccidial oocysts. The resultant mild infections caused reduced growth and impaired food conversion irrespective of the ration fed. Pigmentation, based on shank visual scores, plasma and toe‐web total carotenoid concentrations, was adversely affected in all infected groups. Groups fed canthaxanthin had higher visual scores than comparable groups not so supplemented; however, the presence of coccidiosis depressed flesh pigmentation proportionately in both groups judged by several pigmentation parameters. E. acervulina infection was as deleterious to the broiler as E. maxima, based on these criteria.  相似文献

Immune responses in chickens to Eimeria tenella using oral and subcutaneous routes of infection were investigated. The results obtained indicated that sporulated oocysts inoculated subcutaneously in doses up to 50 000 oocysts per bird were not fatal to 21-day-old chicks. Subcutaneous inoculation of oocysts was found to be less immunogenic than oral administration. The dynamics of the antibody responses were different for the two routes of infection. Orally administered oocysts stimulated a dramatic primary increase in the serum antibody titre with a tendency towards a decrease in the titre 14 days post infection irrespective of second infections at that time. However, a third oral dose of oocysts stimulated a slight increase in antibody titre. Two doses of oocysts injected subcutaneously induced only a slight increase in serum antibody titre. Such a low titre was dramatically increased following a subsequent oral dose of oocysts. Antibodies specific to E. tenella are IgM and IgG immunoglobulins. IgA immunoglobulin was not investigated.  相似文献

The development of antibodies as measured by the indirect fluorescent antibody (IFA)_ test was studied in groups of birds infected with Eimeria tenella or E. maxima and fed diets containing methylzoquate (MBQ) and/or meticlorpindol (MCP). The drugs suppressed both the E. tenella and E. maxima infections, the extent depending on the concentrations in the feed. The IFA test titres in both the E. tenella and E. maxima infections were related to the oocyst production. Effective suppression of the infection resulted in on or minimal IFA response. Since both drugs used inhibit the sporozoites, it is concluded that development of the parasite is necessary to stimulate the host to produce circulating fluorescent antibodies.  相似文献

The progression of coccidiosis and the resultant mortality were followed in chicks fed a OTA-contaminated diet. More complex and rapid progress of coccidiosis occurred in OTA-treated chicks than in chicks fed a OTA-free diet. The concentration of total protein in the serum was significantly decreased in the chicks in the OTA-treated group, whereas this was significantly increased in chicks infected with Eimeria tenella, irrespective of additional treatment with OTA. The serum glucose concentration was significantly increased in all the chicks exposed to OTA and/or suffering from coccidiosis, as was serum retention of uric acid in all groups, most notably in those consuming OTA. OTA induced degenerative changes in, and an increase in the weight of the kidneys, liver, heart and ventriculum; there was depletion of lymphoid tissue and a decrease in the lymphoid organs' weight and body weight. Coccidiosis induced only a slight growth depression and a slight increase in the relative weight of the kidneys and liver. The intensity of the clinical signs, the impairment of kidney function, macroscopic and histopathological changes, deviations in the weight of some organs and general depression in growth were greater when chicks infected with E. tenella were also given OTA.  相似文献

Artemisinin has received much attention in the treatment of malaria in recent years, and it is now considered as a potential candidate to reduce coccidial infection in chickens. It is a sesquiterpene compound which has been isolated from Aretemisia annua for the first time. The present study aimed to investigate the occurrence of artemisinin in A. sieberi (AS) and to test the anticoccidial effects of plant extract in broiler chickens. The aerial parts of the plant were collected during different seasons from Yazd Province, in the centre of Iran. The artemisinin content of the AS was extracted with petrol ether and analysed by high-performance liquid chromatography using UV detection. Anticoccidial effects of the plant extract were tested on chicks challenged with various species of Eimeria. The infected chickens were treated with doses of 1 or 2.5 mg/kg per day artemisinin via oral administration of plant extract. The analytical results showed that the level of artemisinin in AS was 0.2% and 0.14% of dried weight (DW) of plant materials in summer and autumn, respectively. Treatment of experimentally infected chickens with AS extracts showed that artemisinin was able to reduce the severity of coccidial infection induced by Eimeria tenella and E. acervulina, but not E. maxima. The anticoccidial effects of artemisinin were shown by significant decrease in output of number of oocysts per gram of faeces in chickens challenged with different species of Eimeria. This study showed that the levels of artemisinin in AS were comparable with those in other species including A. annua, and that the extract of this plant can reduce coccidial infection in broiler chickens.  相似文献

A pen trial with 720 Ross 308 male chicks evaluated effects of Paracox-5 vaccination at d 1, dietary mannan oligosaccharide (MOS; Bio-Mos at 2, 1, and 0.5 kg/tonne in starter, grower, and finisher, respectively), and d-15 Eimeria challenge on d-22 lesions and 1 to 42-d live performance. Available floor area in each of 24 floor pens was 2.1 m2, and 30 chicks were placed per pen initially. A 3-phase feeding program was used: starter (0 to 15 d), grower (16 to 22 d), and finisher (23 to 42 d). The 4 dietary and vaccine treatments were as follows: 1) without vaccination, dietary MOS, or coccidiosis challenge; 2) without vaccination and dietary MOS but with a 3-strain pathogenic Eimeria challenge; 3) with Paracox-5 vaccination and Eimeria challenge but no dietary MOS; and 4) with Paracox vaccination, dietary MOS, and Eimeria challenge. The pathogenic Eimeria sporulated oocysts mixture from the French National Institute for Agricultural Research (Paris, France), standardized and well-defined, was given via feed at d 15 (100,000 Eimeria acervulina, 10,000 Eimeria maxima, and 15,000 Eimeria tenella per bird). Coccidiosis lesion scores were recorded at 22 d of age (scored as 0, 1, 2, or 3 with increasing severity). Overall mean lesion scores (P < 0.001) were higher in challenged birds. Paracox-5 alone improved 15-d BW and 1 to 15- and 15 to 22-d average daily gain of Eimeria-challenged broilers (P < 0.05). Dietary MOS improved (P < 0.05) 15 to 42- and 22 to 42-d FCR of Paracox-5 vaccinated, Eimeria- challenged broilers. Mortality was 4.4 to 5.8% by treatment, with no significant differences. It was concluded that dietary MOS improved the feed conversion ratio of Paracox-5-vaccinated (d 1), challenged (d 15) broilers from 15 to 42 d of age.  相似文献

Anticoccidial effects of green tea (GT)-based diets were evaluated in chickens following oral infection with Eimeria maxima an ubiquitous intestinal parasite of poultry that impairs the growth and feed efficiency of infected birds. Five-week-old chickens were assigned to four groups (GT 0.5%, GT 2.0%, untreated/infected and non-infected control) and each group consisted of 15 chickens. Chickens were fed a standard diet supplemented with ground green tea for 2 weeks prior to infection with E. maxima (10,000 sporulated oocysts per bird). The effects of green tea on E. maxima infection were assessed by two parameters, fecal oocyst shedding and body weight gain. The green tea-fed chickens produced significantly reduced fecal oocysts (P<0.05) when compared to the E. maxima-infected group fed standard diet. The green tea-based diet, however, did not improve body weight loss caused by E. maxima infection. This study is the first to demonstrate anticoccidial effect of green tea on Eimeria parasites.  相似文献

Coccidiosis is a disease of almost universal importance in Poultry production. The subclinical form of coccidiosis has most significant economic impact due to impaired growth rate and feed conversion. Some factors such as: age, size of flock, season, etc., may effect on severity of this disease. In this study, the relationships between some major risk factors and prevalence of subclinical coccidiosis were investigated. This study was done in 120 broiler farms in five different cities of Mazandaran province, north of Iran. Five chicks (3–8 weeks of ages) were taken randomly from every 120 farms and post-mortem and parasitological examinations were performed. Five Eimeria spp. were recognized: Eimeria tenella, Eimeria maxima, Eimeria acervulina, Eimeria brunetti, and Eimeria necatrix. The prevalence rate of subclinical coccidiosis among them is 75% (90 farms out of 120). E. acervulina was the most prevalent species (65.5%) followed by E. maxima (17.7%), E. tenella (15.5%), E. brunetti (10%), and E. necatrix (5.5%). According to what the results approve, the occurrence of subclinical coccidiosis is significantly related to the age and size of flock, whereas the other factors such as the season of year, industrial strains, chicken’s keeping system, and anticoccidial drugs do not affect this phenomenon remarkably.  相似文献

Anticoccidial effects of Aloe secundiflora crude leaf extract was tested in broiler chickens following oral infection with Eimeria tenella. Sixty 22-day-old birds were divided into six groups of ten birds each. Three treatment groups A, B, and C were fed with the extract (100, 250, and 500 mg/day, respectively) mixed in feed for 10 days, and three control groups: group D (drug control) administered 300 mg/l of sulfachloropyrazine sodium soluble powder in drinking water for 5 days, group E (infected/non-medicated positive control), and group F (uninfected/non-medicated negative control). Except for group F, all groups were orally inoculated with 75,000 sporulated oocysts of E. tenella. The effects of the extract on E. tenella infection were evaluated by severity of bloody diarrhea, body weight (BW) gain, oocyst output, and lesion score. No bird in the treated groups died of coccidiosis, and severity of bloody diarrhea was milder than in the positive control group. BW gains in the treated groups were significantly higher than in group E (p < 0.05). The lesion scores of the treated groups were significantly lower than that of group E. Oocyst output in groups A, B, and C were 11.23, 8.24, and 6.82 × 106, respectively. As compared with the negative control group (12.84 × 106), the reductions in oocyst production were 12.54, 35.83, and 46.88%, respectively. Oocyst output significantly reduced with an increase in Aloe dosage. The findings of this study suggest that Aloe secundiflora extract presents an alternative anticoccidial agent for the control of avian coccidiosis.  相似文献

Sporulated oocysts ofEimeria acervulina were administered orally to cage-housed broilers at a dose of 3.5×105 resulted in mild subclinical coccidiosis.Clostridium perfringens incorporated in feed at a level of 2.5×108 organisms/g. produced lesions characteristic of necrotic enteritis. Mortality of 8% (7/80) occurred in birds fed a ration inoculated withCl. perfringens alone. Mortality of 35% (28/80) was observed in birds which received an oral dose ofE. acervulina and which were fed simultaneously with a ration containingCl. perfringens. Birds which were fed an inoculated ration two days after an oral dose ofE. acervulina showed 41% (33/80) mortality. Birds which received an inoculated ration for two days before administration of an oral dose ofE. acervulina demonstrated 18% mortality (15/80). Birds which were fed an inoculated ration four days after an oral dose ofE. acervulina showed 10% mortality. Infection withE. acervulina reduced the pH of intestinal contents with a simultaneous depression in serum protein. A 39% increase in intestinal passage time from 178 to 248 minutes occurred on the fifth day after infection withE. acervulina. These experiments suggest that necrotic enteritis, attributed to proliferation of a toxigenic strain ofCl. perfringens, followed intestinal stasis and minimal lesions induced by mild intestinal coccidiosis.  相似文献

Plant extracts(PE)are gaining increased attention as potential alternatives to in-feed antimicrobials(AM)due to their known antimicrobial activities.This study was conducted to examine the potential of PE,a microencapsulated product composed of eugenol and garlic tincture as an alternative to AM-agent on performance and intestinal health in broilers under necrotic enteritis(NE)challenge.A total of 960 dayold mixed-sex Cobb 500 chicks were randomly distributed to 48-floor pens with 6 treatments replicated 8 times with 20 birds each.The 6 treatments were as follows:UC,unchallenged control;CC,challenged control;PE,challenged group plus PE;AM,challenged group plus AM;FAP,challenged group plus a full dose of AM with PE;HAP,challenged group plus a half dose of AM with PE in starter,grower and finisher phases.Birds in the challenged groups were inoculated with Eimeria spp.on d 9 and Clostridium perfringens on d 14.The body weight gain(BWG),feed intake(FI),feed conversion ratio(FCR),and livability of birds were compromised,and intestinal lesions and mortality were increased(P<0.05)by NE challenge,illustrating a successful clinical NE challenge.Birds fed AM had higher BWG and FI,and lower FCR,mortality,and intestinal lesions compared to the CC group(P<0.05).Birds fed PE had improved FCR(P<0.05)and livability(5.8%)in an overall period compared to the CC group.On d 16,PE supplementation reduced ileal lesion scores in only male birds(P<0.05).Birds fed PE had decreased Eimeria maxima and Eimeria acervulina oocyst counts in caecal content(P<0.05).Birds fed PE had decreased Eimeria brunetti and total oocyst counts in caecal content,and E.acervulina oocyst counts in ileal content in only female birds(P<0.05).On d 35,PE supplementation reduced variation of BW in both male and female birds and increased yellowness(b*value,14.4%)in the thigh.These findings suggest the potential of PE supplementation in diets to improve the performance and intestinal health of birds under clinical NE as indicated by improved FCR,livability,uniformity,reduced ileal lesions,oocyst counts and increased skin yellowness.However,the protective effect of PE may not be apparent in the presence of AM in the feed.  相似文献

A study was carried out to examine the effect of dietary supplementation of oregano essential oil on performance of broiler chickens experimentally infected with Eimeria tenella at 14 days of age. A total of 120 day-old Cobb-500 chicks separated into 4 equal groups with three replicates each, were used in this study. Two groups, one infected with 5 x 10(4) sporulated oocysts of E. tenella and the other not, were given a basal diet and served as controls. The other two groups also infected with E. tenella were administered diets supplemented with oregano essential oil at a level of 300 mg/kg, or with the anticoccidial lasalocid at 75 mg/kg. Following this infection, survival rate, bloody diarrhoea and oocysts excretion as well as lesion score were determined. Throughout the experimental period of 42 days, body weight gain and feed intake were recorded weekly, and feed conversion ratios were calculated. Two weeks after the infection with E. tenella supplementation with dietary oregano oil resulted in body weight gains and feed conversion ratios not differing from the non-infected group, but higher than those of the infected control group and lower than those of the lasalocid group. These parameters correspond with the extent of bloody diarrhoea, survival rate, lesion score and oocyst numbers and indicated that oregano essential oil exerted an anticoccidial effect against E. tenella, which was, however, lower than that exhibited by lasalocid.  相似文献

Donor chickens given feed medicated with one or two levels of decoquinate or given non-medicated feed were infected with oocysts of Eimeria tenella or E. maxima per os. Twelve hours after inoculation with oocysts liver, mid-intestine or ceca homogenates were fed to previously uninfected recipient chickens. The results showed that continuous medication with decoquinate was effective in preventing the transfer of sporozoites from the intestine to the liver. Oocysts were detected in the feces of all recipients of tissue from non-medicated donors, showing that some sporozoites of E. maxima and E. tenella are normally transferred to liver. Young broiler chickens were immunized by oral inoculation of E. maxima oocysts. The immune status of similar chickens inoculated with sporozoites of the same species directly into the liver or spleen were assessed. During the experimental period half of the chicks were provided with non-medicated food and the remainder were given feed supplemented with decoquinate; decoquinate was effective in arresting the development of the sporozoites. Two weeks after initial infection the birds were challenged with oocysts of E. maxima per os. Injection of sporozoites into the spleen did not protect against challenge. Birds inoculated with sporozoites into the liver were unable to develop a significant level of immunity. When the drug pressure was removed from these birds, parasitism of the intestine occurred and immunity developed.  相似文献

Guar meal contains relatively high levels of saponins, which are known to have antiprotozoal activity and may be effective against coccidiosis. A 2x2 factorial experiment investigated the impact of guar meal (0 or 5%) corn-soy-based starter broiler diets on chicks unchallenged or challenged with Eimeria tenella. At 1 day of age, 120 unsexed RossxRoss broiler chicks were randomly distributed among four treatment groups. Chicks were challenged with 5x10(3) sporulated oocysts of E. tenella in 0.5ml at 10 days of age by oral gavage. Weekly body weight, body weight gains, feed conversion ratio and mortality rate were recorded for chicks fed from 0 to 21 days of age. Oocysts shed per gram feces were recorded from 6 to 10 days post-challenge. Results showed that challenged chicks fed 0% guar meal had significantly higher oocysts per gram shed in feces than the other groups. No significant differences among treatment groups in mortality rate were observed. Body weights of unchallenged and challenged chicks fed 0% guar meal were significantly higher than those fed 5% guar meal at 2 weeks of age. Results indicated that including 5% guar meal in the diet of chicks challenged with E. tenella decreased oocysts shed per gram feces and prevented bloody diarrhea, but without affects on body weight and feed conversion ratio at 11 days post-challenge.  相似文献

The effect of Eimeria praecox on concurrent Eimeria maxima infection was studied in susceptible chickens. Clinical signs of coccidiosis were assessed in single E. praecox or E. maxima infections and compared to dual infection with both Eimeria species. Groups infected solely with 10(4)E. maxima oocysts displayed weight gains that were 48% of weight gain in uninfected controls. Weight gain in chickens infected only with 10(4)E. praecox oocysts was 90% of uninfected controls. Average weight gain in chickens infected with both E. maxima and E. praecox was 79% of controls, and showed no significant difference (P>0.05) from weight gain in E. praecox-infected chickens. Feed utilization (feed conversion ratio, FCR) in chickens infected with both species showed no significant difference (P>0.05) from FCR in non-infected controls or chickens infected with E. praecox alone; all showing a significant difference (P<0.05) from FCR in chickens infected solely with E. maxima. Although E. praecox did not appear to have a negative effect on weight gain and FCR, it did cause a significant decrease in serum carotenoids. Analysis of oocysts excreted by chickens during dual infection showed little effect of E. praecox on E. maxima oocyst production.  相似文献

Background: Besides the anticoccidial drug resistance problem, increasing consumer concerns about food safety and residues have propelled the quest for alternative prevention and control strategies amongst which phytotherapy has gained appeal due to a renewed interest in natural medicine.Objective: The objective was in vivo screening of four phytochemicals/extracts and a fungal immunomodulatory protein (FIP) against an Eimeria acervulina infection in broilers.Animals and methods: Four phytochemicals/extracts (extract from Echinacea purpurea, betaine (Betain?), curcumin, carvacrol (two different doses)), and a recombinant FIP from Ganoderma lucidum cloned and expressed in Escherichia coli were investigated for their anticoccidial potential. The experiment was conducted in a battery cage trial with 54 cages of eight birds each. Broilers infected with E. acervulina (a low and high infection dose of 104 and 105 sporulated oocysts, respectively) and treated with the phytochemicals/extracts or the FIP were compared with broilers treated with the anticoccidial salinomycin sodium (Sacox®) and with an untreated uninfected and an untreated infected control group. Coccidiosis lesion scores, body weight gains and oocyst shedding were used as parameters.Results: The results showed a coccidiosis infection dose effect on the mean coccidiosis lesion scores. The phytochemicals/extracts and the FIP failed to reduce coccidiosis lesion scores and oocyst shedding, while salinomycin efficiently controlled the E. acervulina infection and enabled significantly higher body weight gains.Conclusion: In conclusion, the selected phytochemicals/extracts and the FIP did not reduce the lesions of an experimentally induced E. acervulina infection.  相似文献

Oocysts were found in all four birds each infected with one of the four sporocysts of a single oocyst of Eimeria acervulina and in three of the four birds infected with single sporocysts from two other oocysts. The virulence of a field strain of the chicken coccidian parasite Eimeria acervulina (Boreham I), dually resistant to the chemically unrelated anticoccidial agents decoquinate and clopidol, was compared with that of a drug-sensitive laboratory strain (Ongar) of the same species. Following a single heavy infection (prevented from recycling), both strains exhibited pathogenic effects typical of their species, viz., pathognomonic lesions, adverse effects on body weight gain and feed conversion ratio (FCR), but no mortality. One week after infection, chicks infected with either strain had a statistically significantly worse weight gain than the uninfected control; the Boreham I strain produced more oocysts, and caused slightly more severe duodenal lesions and poorer FCRs than the Ongar strain (all those effects being non-significant). After 3 weeks, there were no significant differences between any cumulative effects of either strain, nor any differences from the uninfected control. However, from 2 to 3 weeks after infection, chicks infected with either strain had a greater feed consumption and growth rate than uninfected chicks. When chicks reared on solid floors were given lighter infections of either strain, which were allowed subsequently to recycle naturally, there were no consistent reductions in weight gains, but feed consumption was higher than that of uninfected chicks. Whatever, the mode of infection, there were no significant differences between the weights of infected and uninfected chicks after 3 weeks, but the FCR of infected chicks was usually poorer than that of uninfected chicks. The difference between the virulences of the Boreham I and Ongar strains was not greater than that between various drug-resistant strains or between various sensitive strains of several Eimeria species recorded in the literature. It is therefore concluded that there was no difference between the virulences of the two strains of E. acervulina that could be attributed to the drug-resistance of one of them.  相似文献
