Zeaxanthin epoxidase (Zep) is one of at least two genes important for the incidence and amount of carotenoids in yellow-fleshed potato. The recessive allele of the gene encoding zeaxanthin epoxidase (Zep1) has previously been shown to inhibit the conversion of the xanthophyll zeaxanthin to other, more polar carotenoids. We examined the effect of the dosage of Zep1 on total carotenoids in yellow-fleshed tetraploid potato germplasm. The dosage of heterozygous individuals was determined using high-resolution DNA melting. We also surveyed a wide range of germplasm available in the USDA-ARS potato breeding program in Washington for the presence and dosage of Zep1. Genotypes with zero, one, or two copies of Zep1 had statistically similar levels of total tuber carotenoids. Triplex individuals showed a small but significant increase in total carotenoids over the previous three classes. In turn, individuals that were homozygous for Zep1 had much higher levels of total carotenoids than all other dosage classes. This suggests that a slight dosage effect is present, but that the effect of Zep1 is indeed largely recessive. A significant amount of variation in total carotenoids was observed within all Zep1 dosage classes, reinforcing the evidence that additional loci are important for high total carotenoids in potato. Our survey of breeding germplasm supported earlier research that Zep1 is uncommon in tetraploid potato germplasm. The use of high-resolution DNA melting again allowed us to identify the dosage of Zep1 in heterozygous individuals. This is important for breeding high-carotenoid potatoes at the tetraploid level, since recovery of homozygous Zep1 individuals is very rare. Identification of agronomically desirable parents with two or more copies of Zep1 would allow for higher recovery of homozygous Zep1 progeny, permitting additional selection for other desirable traits.  相似文献

Potato has a defined complement of metabolites that contribute to the human diet. Among these are the carotenoids and anthocyanins. Carotenoids are found in all potatoes in the flesh. White-fleshed varieties have 50 to 100 μg per 100 g fresh weight (FW), while moderately yellow-fleshed varieties will generally possess from 100 to 350 μg per 100 g FW. The more intensely yellow-fleshed genotypes, which may look orange, at the higher extremes are at levels above 1,000 μg per 100 g FW. The highest level published is 2,600 μg per 100 g FW in diploid germplasm derived from South American Papa Amarilla cultivars. Potato generally has predominantly lutein, a xanthophyll which is also found in the human retina, and must be obtained in the diet. The genotypes with extremely high levels of total carotenoids have zeaxanthin, an isomer of lutein, which is also present in the human retina. Anthocyanins are present in red- or purple-skinned and fleshed varieties. Total anthocyanins range from 1.5 mg to 48 mg per 100 g FW in a solidly pigmented purple-skinned, purple-fleshed breeding line. The degree of pigmentation in the flesh appears to be under polygenic control. Anthocyanins are potent antioxidants and anti-inflammatory substances. The level of total anthocyanins is correlated with antioxidant level (r?=?0.94, P?<?0.001). Several methods of cooking interacted with genotypes in the antioxidant level remaining after cooking compared to raw potatoes. No method of cooking completely eliminated antioxidant activity, while boiling appeared to increase it compared to raw potato in the case of the most highly pigmented clone.  相似文献

Diploid potato clones, interspecific hybrids ofSolanum species, having in their originS. tuberosum,S. chacoense, S. yungasense, S. phureja, S. gourlayi, andS. demissum, with resistance to soft rot, were crossed to tetraploid potato clones in 4x-2x crosses. The 24 tetraploid families obtained in a North Carolina II design were examined for tuber resistance to soft rot in a laboratory test and for basic agronomic traits in field trials conducted for two consecutive years. In addition, one family originating from a 4x-2x cross of two susceptible parents was tested. Statistical analysis of the data revealed significant general (GCA) and specific combining ability (SCA) effects, year, GCA (female) × year, GCA (male) × year, and SCA × year upon the inheritance of resistance to soft rot. About 35% of the progeny was selected as resistant to tuber soft rot, and of these 11% showed high resistance combined with good tuber yield, tuber weight, and tuber appearance. The relationships between resistance to soft rot and chosen agronomic traits were not noted or were weakly significant and sporadic. The resistance to tuber soft rot found in diploid potato hybrids can be transferred to the cultivated tetraploid pool through 4x-2x crosses, and a high frequency of offspring posses resistance.  相似文献

This paper describes a new source of host-plant resistance to the Colorado Potato Beetle (CPB;Leptinotarsa decemlineata (Say)) in a tetraploid potato (Solanum tuberosum L.) selection, ND2858-1. This hybrid was recorded as a cross betweenS. fendleri ssp.fendleri (A. Gray) and NDSU selection ND1215-1. Field defoliation of this and progeny clones from a subsequent cross to an cultivar was reduced by 50% to 90% in open-choice field screening plots. Progeny of ND2858-1 are vigorous, with high yield potential, and some clones have tuber glycoalkaloid content equivalent to commercial cultivars. These clones are male and female fertile and may be useful to other potato breeding programs. Further characterization of this resistance source is in progress.  相似文献

The aim of this work is the evaluation of tetraploid potato clones with germplasm fromS. chacoense andS. phureja for chipping and resistance to tuber soft rot (Erwinia carotovora subsp.carotovora). Tetraploid potato clones were obtained via 2n eggs through 2x–4x crosses. Cytological mechanisms were investigated in the diploid hybrid 2n-egg producer used as parent in the unilateral sexual polyploidization scheme; the omission of the second meiotic division and cytokinesis failure at the end of meiosis were identified with a genetic consequence equivalent to second division restitution (SDR). Tuber marketable yield, specific gravity and chip-processing ability evaluated at harvest and after cold storage with and without reconditioning were higher in some clones as compared to theS. tuberosum tetraploid parent. Tuber soft rot resistance, contributed byS. chacoense, was successfully transferred to the4x level. After inoculation withErwinia carotovora subsp.carotovora and estimation of the rotted area, one tetraploid clone proved to be resistant and three partially resistant. The genetic value of exotic germplasm and sexual polyploidization strategy are discussed in relation to potato improvement.  相似文献

A three-year study was conducted from 1991 through 1993 to evaluate the field performance of potato clones that had been selected for resistance to wilt symptom development and stem colonization byVerticillium dahliae. The total yield, size distribution, and specific-gravity of these highly resistant clones were compared with standard cultivars and two parent clones with high Verticillium resistance and high yield, A66107-51 and A68113-4. Two groups of Verticillium resistant germplasm were selected in 1991. One group was the highly resistant progeny from a cross between A66107-51 and A68113-4. The second group consisted of hybrids between wild species accessions with high Verticillium resistance and cultivated diploid and tetraploid germplasm. Twenty-one out of 125 progeny clones from the A66107-51 × A68113-4 cross were highly resistant to Verticillium infection and were tested in yield trials for two years. Another eight selections had high Verticillium resistance but such low yield of seed tubers that they were tested only one year for yield. Eleven of the original 29 highly resistant selections were significantly lower in total yield than Russet Burbank. Only one clone was significantly higher in yield than RB, and none outyielded either of the resistant parents. Reddale and Century Russet had moderate to high Verticillium resistance, respectively. Both outyielded Russet Burbank and were comparable with the high yielding resistant parents. Of 15 interspecific hybrids tested in 1991, only four had sufficient yield to produce seed for yield trials and good wilt resistance after three successive seasons of evaluation. We concluded from this study that selecting at early stages of variety development strictly for Verticillium resistance is likely to be inefficient. Rather, selection should be for yield and other agronomic criteria in Verticillium infested fields.  相似文献

Freshly harvested potato tubers contain up to 50 mg/100g ascorbic acid (AA), but levels decline rapidly during cold storage. Genetic alterations to boost the AA content of stored tubers would contribute to human nutrition and might improve tuber resistance to oxidative damage during chilling. While studying the breeding potential ofSolanum phureja germplasm, we identified a 24-chromosome Phureja-haploid Tuberosum hybrid (clone I, USW5295.7) that retained a twofold higher content of tuber AA than neighboring clones after storage at 5 C. Clone I produces 2n-pollen through a mechanism genetically equivalent to first-division restitution (FDR), which transmits much of the nonadditive genetic variance for tuber yield. We now report a survey of clone I progeny showing significant family and ploidy effects on tuber AA levels during cold storage, consistent with the transmission of information for higher AA by FDR 2n-pollen. These results encourage further study of 2n-gametes and wild species germplasm to breed for increased tuber AA.  相似文献

Summary Offspring were produced from a cross between two long-day-adaptedSolanum phureja clones which carried resistance to tuber soft rot (Erwinia carotovora subsp.atroseptica). In tests carried out on the produce of field-grown plants raised from tubers, over fifty per cent of the 173 offspring were found to be highly resistant. Assessments were also carried out of tuber yield, mean tuber weight, tuber number, shape, regularity, flesh colour, texture of the steamed flesh, fry colour, after-cooking blackening, sprout length after storage and overall dormancy. There were statistically significant differences between clones for all characters (P<0.001). Twelve of the clones were selected on the basis of high resistance, yield, tuber weight, regularity of shape and absence of after-cooking blackening. The value of resistant long-day-adapted diploid material for commercial breeding is discussed.  相似文献

Sentinel (FV8957-10) is a new yellow-fleshed potato cultivar that produces a high yield of attractive round to oval tubers at early harvest. Sentinel is suitable for fresh market use and chip processing. It was selected at the Lethbridge Research Centre of Agriculture and Agri-Food Canada, AB from a cross made at the Potato Research Centre of Agriculture and Agri-Food Canada, NB between breeding clones F72218 and F47024. Sentinel has erect medium-height vines and produces uniform tubers of medium-size with a light buff skin, light yellow flesh and shallow eyes. In multi-date-harvest trials, Sentinel produced similar or higher marketable yield compared to Atlantic and NorValley in southern Alberta. When harvested at 80 days after planting, Sentinel produced similar or greater yield than Superior and Conestoga at test sites across western Canada. Specific gravity of Sentinel was less than Atlantic and Conestoga but higher than NorValley and Superior, whereas chip colour was similar to Atlantic and lighter than NorValley or Superior. Sentinel showed little tendency to slough or to darken after cooking. It showed clear foliar and tuber symptoms when infected with bacterial ring rot. Sentinel is moderately susceptible to verticillium wilt, early blight, fusarium wilt and common scab and susceptible to late blight.  相似文献

Studies of the breeding behavior of clones containing high levels of anthocyanins were conducted. Red-fleshed clones appeared in proportions suggesting multigenic control of degree of pigmentation. Red-fleshed and pur-ple-fleshed clones were always accompanied by red and purple skin, respectively. Red flesh ranged from partial pigmentation to complete pigmentation represented by pigment present in all tuber tissues. Percentage of com-pletely red-fleshed progeny was 14.5% and 4.1% in red x red crosses vs red × white (or the reciprocal), respectively. Purple-fleshed progeny were obtained from red x white crosses where the white-fleshed parent harbored theP pigment gene in juxtaposition with the nulliplex recessive state of theI gene (i.e.,iiii), which suppressed expression. Total anthocyanin ranged from 6.9 to 35 mg per 100 g fresh weight in the red-fleshed and 5.5 to 17.1 in the purple-fleshed clones. Red-fleshed clones contained predominantly acylated glycosides of pelargonidin while the purple-fleshed clones contained predominantly acylated glycosides of petunidin and peonidin. Oxygen Radical Absorbance Capacity and Ferrous Reducing Ability of Plasma revealed that the antioxidant levels in the red or purple-fleshed potatoes were two to three times higher than white-fleshed potato.  相似文献

Potato seedlings grown from true seed were screened for resistance to leaf roll virus. Infected seedlings were discarded, and the screening process was repeated for four subsequent tuber generations. Forty-one of an original 717 clones (six crosses and three selfed lines) remained free from the virus. Challenge inoculations were made with aphids,Myzus persicae (Sulz.), that had been colonized on infected excised leaves ofPhysalis floridana Rydb. Recovery tests were attempted with non-viru-liferous aphids that were tested on healthyP. floridana. In comparative tests, more clones remained free from the virus among progeny of the cross F5569×F4896 and of a self of the Katahdin variety, than among any other progeny tested.  相似文献

Summary Experiments were carried out withSolanum phureja germplasm which had been selected for tuberization response under W. European growing conditions. Selected clones and seedling (true potato seed; TPS) families ofS. phureja were raised in the field and tubers assessed for yield, tuber number, skin colour, size, shape and cooking characteristics. Some families had low within-family variances for cooked flesh hardness and some appeared as uniform for tuber size and shape as the selectedS. phureja clones. Skin colours of the tubers within TPS families were often assorted. The commercial potential ofS. phureja as a possible TPS-derived crop is discussed, as its self-incompatibility makes possible the cheap production of true potato seed.  相似文献

In a genetic study with cultivated diploid potatoes the distribution of anthocyanin to the tuber flesh was found to be controlled by a single dominant allele. The gene symbolPf (pigmented flesh) is proposed. The geneI, which controls the distribution of pigment to the tuber skin, is necessary for the expression ofPf and is closely linked to thePf locus. ThePf gene is a potentially useful genetic marker which does not appear to be associated with any deleterious traits and can be screened for without laboratory equipment.  相似文献

The potato tuberworm, Phthorimaea operculella (Zeller), is an important insect pest of potato, Solanum tuberosum L., both in storage and in the field. In this research, tubers of eight commercial potato cultivars and four Iranian selections with equal weight and dormancy were exposed to 10 pairs of adult P. operculella in a climate chamber set at 25?±?1 °C, 65?±?5 % RH and total darkness. In a free-choice situation, oviposition was lower on 397082–2, Khavaran and Morene compared to the other germplasm that were exposed to adults of P. operculella. Number of mines per tuber, length of mines per tuber, time of development of larvae, number of pre-pupae produced per tuber, weight of pre-pupae and number of eggs developed in ovaries per female were counted and/or measured on each commercial cultivar and selection. There were fewer and shorter mines on tubers of 397082–2, Khavaran and Morene compared to the other potato germplasm. The number of pre-pupae produced per tuber and the weight of pre-pupae were lower when P. operculella was reared on tubers of 397082–2, Khavaran and Morene. Also the development, survival and fecundity were lower when P. operculella was reared on those same germplasm. Flesh firmness was negatively correlated with larval survival (r 2?=?0.87); in addition, the percentage of starch and macronutrient composition was low on these three germplasm. Thus, tuber flesh firmness of these germplasm could delay larval penetration and lower establishment 397082–2, Khavaran and Morene showed promising traits that can be integral component of potato breeding for resistance to P. operculella and pest management programs.  相似文献

Breeding potato (Solanum tuberosum L.) with high protein content is important for food value and for the processing sector to extract protein from potatoes. Due to the narrow genetic base of the common potato, a new germplasm has to be explored for breeding high-protein potatoes. The objectives of this research were to quantify the crude protein content of diploid hybrid Solanum phurejaSolanum stenotomum (PHU-STN) genotypes, determine the correlation of crude protein content with dry matter and other important agronomic traits, and estimate broad sense heritability for crude protein content. In 2008 and 2009, 70 diploid clones were grown in replicated field plots in Jiagedaqi, Heilongjiang. Also, two tetraploid potato cultivars, Kexin 2 (table potato) and Atlantic (high dry matter), were included in this experiment as controls. The crude protein content on a fresh weight basis (FW) over replications and years for PHU-STN clones ranged from 2.85% to 5.01% with a grand mean of 3.72%, and most of these clones were significantly or highly significantly higher in crude protein content than the two tetraploid controls. Correlations between crude protein content and agronomic traits were all negative, but the magnitudes were generally weak or moderate. Therefore it should be possible to select diploid clones with high crude protein content and acceptable agronomic traits. The estimate of broad sense heritability for crude protein content (FW) on a clonal mean basis was 0.66 with a 95% confidence interval of 0.46 to 0.79, whereas for crude protein content on a dry weight basis the estimate of broad sense heritability was 0.65 with a 95% confidence interval of 0.43 to 0.78. These results suggest that these diploid hybrid PHU-STN clones would be a good germplasm source for a high-protein-breeding effort, and identification of tetraploid progenies with high protein content and acceptable agronomic traits from 4x–2x cross should be possible.  相似文献

Potato breeding selections A66107-51 and A68113-4 (Solanum tuberosum gptuberosum) are being released as germplasm that is highly resistant to Verticillium wilt (Verticillium dahliae Kleb.) These selections have been extensively evaluated for resistance to Verticillium as measured by symptom severity and degree of colonization byV. dahliae. They combine outstanding wilt resistance with good tuber maturity characteristics, high yield, and french fry processing tuber type. A66107-51 has long-oblong, russeted tubers, with high yield and variable processing quality. A68113-4 has long-oblong, partially russeted tubers with high yield and good processing quality. Both selections are fertile as female parents, and A66107-51 is occasionally pollen fertile. The Verticillium resistance of these two clones has been transmitted to progeny. These selections were developed by USDA-ARS in cooperation with the Idaho Agricultural Experiment Station. Seed is available from USDA-ARS, Aberdeen, Idaho.  相似文献

Late blight, caused by Phytophthora infestans, is a devastating disease on potato worldwide and new lineages of the pathogen continue to develop in the U.S. Breeding for resistance is important for economic and environmental purposes. The Solanaceae Coordinated Agricultural Project (SolCAP) focuses on linking allelic variation in genes to valuable traits in elite cultivated potato germplasm. This research assessed the SolCAP diversity panel (206 clones in Washington and 213 clones in Wisconsin) for tuber resistance to the US-24 clonal lineage of P. infestans after potatoes were harvested from fields in Washington and Wisconsin in 2011. This is the first time this germplasm has been evaluated for tuber resistance to P. infestans using a non-intrusive zoospore inoculation technique. Clones with a percent incidence of 30% or less were considered resistant and only eight clones (Palisade Russet, AWN86514–2, MSL268-D, MSM171-A, MSM182–1, MSN230-1RY, Patagonia and Yukon Gem) were characterized as resistant at both locations. These clones have previously demonstrated high to moderate partial foliar resistance to isolates of P.infestans and therefore represent germplasm with both foliar and tuber resistance. Nine clones (AWN86514–2, F66041, MN 18747, MSM 182–1, MSN230-1RY, Modoc, Ama-Rosa, Patagonia and Purple Majesty), were characterized as slow-rotting at both locations with a mean percent internal rot of 75% or less after 33 days of storage. Two clones, MN 18747 and Modoc, are considered to have the highest risk of being a carrier for P. infestans of all the clones evaluated in the SolCAP collection. Not a single clone demonstrated complete tuber resistance to the US-24 strain at both locations.  相似文献

Early blight, caused byAlternaria solani Sorauer, is a serious disease of potatoes that occurs in most potatogrowing regions in the world. There is little resistance to early blight among commercial potato cultivars. However, resistance to early blight in diploid (Solanum phureja-S. stenotomum) potatoes has been identified, and was found to be highly heritable and readily transferred to the tetraploid level via- crosses. The purposes of this study were to identify good levels of early blight resistance in open-pollinated4×-2× (Solanum tuberosum×S. phureja-S. stenotomum) hybrids selected for horticultural characteristics, to estimate broad-sense heritability for early blight resistance in these hybrids, and to investigate the general and specific combining ability for resistance to early blight from some of these early blight resistant tetraploid selections. Four early blight resistant clones were crossed as female parents with four different male parents in a design II mating scheme to generate 16 families. Approximately 20 randomly chosen offspring per family were visually evaluated for early blight resistance in 1995 and 1996 in a randomized complete block design in Pennsylvania. Broad-sense heritability for early blight resistance among the open-pollinated- hybrids which were originally selected for horticultural characteristics was 0.91 with a 95% confidence interval of 0.80, 0.94. There were significant differences among female and male parents for area under the disease progress curve, indicating the importance of general combining ability for early blight resistance in this germplasm. The female x male source of variation was not significant, indicating that specific combining ability was not important. The greatest number of resistant progeny were observed in families where both parents were derived from the early blight resistant population; however, at least one highly resistant progeny was produced in all families. These results suggest that the early blight resistance in these clones can be readily incorporated into the commercial tetraploid breeding population.  相似文献

Progenies from a group of tetraploid parental clones from the USDA potato breeding program were used to investigate variation in resistance to the potato leafhopper,Empoasca fabae (Harris), and the potato flea beetle,Epitrix cucumeris (Harris). The study utilized two mating designs: i) selfing and testing both the parents and the S1 progeny; and ii) nine clones, used as males, were each crossed to three different clones, and progenies from the resulting families were tested. Statistically significant differences between families were measured in each test for both insects. Non-additive genetic variance was larger than additive genetic variance in progeny reaction to leafhopper infestation and hopperburin, but was smaller in progeny reaction to flea beetles. Environmental variation contributed heavily to the total variation of plant reaction to both insect species. Selecting individuals was indicated to be slightly more effective than selecting males on half-sib progeny performance but not as effective as selecting clones on S1 progeny performance. Because of the large environmental variance and small additive variance for both the leafhopper and flea beetle, slow progress in increasing the level of resistance to these two species in this sample population was predicted. Resistance to leafhopper infestation was genetically quite highly correlated (positive) with resistance to hopperburn, but phenotypically the correlation was considerably smaller. Negative genotypic, phenotypic, and environmental correlations between leafhopper infestation and flea beetle infestation suggest that selecting for resistance to one of these species, in the population sampled for these tests, would tend to increase susceptibility to the other.  相似文献
