Ginkgo biloba extract (GBE) has many remarkable pharmacological and clinical effects, and it is the most frequently used product as a phytomedicine in many countries. The combination of primary extraction with 70% ethanol followed by extraction using supercritical carbon dioxide provides an efficient and economical means for obtaining flavonoids and terpenoids from Ginkgo biloba leaves. The supercritical fluid extraction (SFE) is affected by pressure, temperature, and the concentration of modifier in the extractant. At the most favorable experimental conditions of 300 MPa, 60 degrees C, and carbon dioxide containing 5% ethanol as modifier, the yield of GBE powder is 2.1% (based on the air-dry weight of Ginkgo biloba leaves) compared to a yield of only 1.8% by conventional solvent extraction. The contents of flavonoids and terpenoids in SFE products are 35.9% and 7.3%, respectively, which are significantly higher than the general standards of 24% and 6%, respectively.  相似文献

沙棘叶中富含黄酮类化合物等多种活性成分,为提高其提取率和利用率,本研究采用常规溶剂萃取、超声辅助、微波辅助、超声波微波协同提取4种方法对沙棘叶黄酮进行提取,测定沙棘叶黄酮得率并观察沙棘叶的微观组织结构,比较筛选沙棘叶黄酮的最佳提取方法。并采用响应面法对最佳提取方法进行工艺优化,同时测定沙棘叶黄酮组成和体外抗氧化活性。结果表明,超声波微波协同提取法是提取沙棘叶黄酮的最佳方法,黄酮得率较常规溶剂萃取法提高了42.54%(P<0.05),沙棘叶细胞损伤最严重。超声波微波协同提取沙棘叶黄酮的最佳工艺为乙醇体积分数61%、提取时间18 min、微波功率446 W,此时黄酮得率为42.09 mg·g-1。沙棘叶黄酮提取液中共鉴定出6种黄酮类成分,分别为儿茶素、丁香酸、山萘酚、槲皮素、异鼠李素、杨梅素,其中儿茶素含量最高,为1.474 8 mg·g-1,其余5种的含量均在0.1~0.3 mg·g-1之间,山萘酚最低,为0.125 2 mg·g-1;沙棘叶黄酮提取液具有较强的还原力及较高的1,1-二苯基-2-三硝基苯肼(DPPH)、2,2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)和羟基自由基清除率,抗氧化活性较高。本研究为沙棘叶黄酮的工业化生产提供了科学依据。  相似文献

Subcritical water extraction of antioxidant compounds from rosemary plants   总被引:7,自引:0,他引:7  
Subcritical water extraction at several temperatures ranging from 25 to 200 degrees C has been studied to selectively extract antioxidant compounds from rosemary leaves. An exhaustive characterization of the fractions obtained using subcritical water at different temperatures has been carried out by LC-MS, and the antioxidant activities of the extracts have been measured by a free radical method (DPPH). Results indicate high selectivity of the subcritical water toward the most active compounds of rosemary such as carnosol, rosmanol, carnosic acid, methyl carnosate, and some flavonoids such as cirsimaritin and genkwanin. The antioxidant activity of the fractions obtained by extraction at different water temperatures was very high, with values around 11.3 microg/mL, comparable to those achieved by SFE of rosemary leaves. A study of the effect of the temperature on the extraction efficiency of the most typical rosemary antioxidant compounds has been performed.  相似文献

Supercritical fluid extraction (SFE) has been utilized by the food industry in many applications to extract, fractionate, and recover compounds from various food matrices. However, little research has been conducted using SFE as an alternative process for producing reduced-fat cheese. Lipids in cheeses may be selectively extracted due to the nonpolar properties of supercritical carbon dioxide (SC-CO2), without leaving residual chemicals as is the case in solvent extraction. The objective of this study was to evaluate the influence on the extraction process due to cheese variety and protein breakdown by age. A Latin square design was utilized to test the extractability of lipids from Parmesan and Cheddar cheeses, aged young (9-10 months) or old (24 months). Extraction took place in a 500 mL SFE vessel using 100 g of grated cheese samples. The SFE parameters of the extraction were 350 bar, 35 degrees C, and supercritical carbon dioxide at a flow rate of 20 g/min for 55 min. Compositional analysis measured all treated samples and controls of total lipids, lipid profiling, total protein, protein/peptide analysis, moisture, ash, and pH. Cheese type was a major variable in fat extraction. The extraction in Cheddar showed an average fat reduction of 53.56% for young cheese, whereas that in old Cheddar was 47.90%. However, young Parmesan was reduced an average of 55.07%, but old Parmesan was reduced at 68.11%, measured on a dry basis. SFE extracted triglycerides and cholesterol, but did not remove phospholipids. This investigation introduces the observations of the effect of Cheddar and Parmesan varieties on SFE, offering data on the important parameters to consider in the design of SFE processes to reduce fat in cheese.  相似文献

Four techniques, Soxhlet extraction (SOX), ultrasonic extraction (USE), fluidized-bed extraction (FBE) and accelerated solvent extraction (ASE) with different solvents (methanol, hexane/acetone and acetonitrile) were used for the extraction of DDT analogues in sediments. Results revealed that the four extraction techniques had high recoveries (〉 86.0%) with low standard deviations (〈 12.0%) for most of DDT analogues, meaning that they could all successfully extract DDT analogues in sediments. Accelerated solvent extraction using methanol and hexane/acetone (1:1), fluidized-bed extraction using hexane/acetone (1:1) and the ultrasonic extraction using hexane/acetone (1:1) were comparable or better than Soxhlet extraction using hexane/acetone (1:1). Considering solvent- and time-consumption, level of automation, and environmental risk, accelerated solvent extraction with hexane/acetone (1:1) was better than the other extraction techniques.  相似文献

Several rapid extraction methods were evaluated for use with a monoclonal antibody-based competitive inhibition ELISA (cELISA) to detect sulfadimethoxine (SDM) in chicken liver tissue. These methods included extraction of the samples with (1) aqueous buffer with or without ultrafiltration, (2) acetonitrile/water, (3) methanol/water, or (4) acetone. The organic extraction methods were evaluated with or without solvent evaporation prior to dilution into assay buffer for the cELISA. The aqueous-based extraction methods were compatible with the cELISA. However, of the organic extraction methods, only the acetone liver extract with solvent evaporation prior to analysis was compatible with the cELISA. The cELISA method coupled to aqueous- or acetone-based sample extraction as well as an HPLC method was evaluated for the analysis of chicken liver tissues fortified with SDM at levels from 0.2 to 0.025 ppm. Mean SDM recoveries for the HPLC method and for the cELISA method using samples prepared by aqueous extraction, aqueous extraction and ultrafiltration, or acetone extraction, evaporation, and reconstitution were 68.9, 95.7, 60.1, and 52.5%, respectively. For the analysis of samples obtained from an SDM incurred residue study, HPLC and cELISA analysis of the same organic extract gave results that were highly correlated (R(2) = 0.976; p < 0.0001). However, results obtained from the analysis of aqueous extracts by cELISA did not correlate well with those obtained by HPLC (R(2) = 0.61, p > 0. 0006). This was attributed to the coextraction of cross-reactive SDM-related residues that were not quantified by the HPLC method. The presence of these residues should be considered during data interpretation when ELISA methods coupled with rapid aqueous extraction of samples are used in SDM residue monitoring programs.  相似文献

This research focuses on the overall extraction process of alkylresorcinols (ARs) from uncooked grains and baked products that have been processed with wheat, corn, rice, and white flour. Previously established extraction methods developed by Ross and colleagues, as well as a semiautomated method involving accelerated solvent extraction (ASE), were applied to extract ARs within freshly ground samples. For extraction of alkylresorcinols, nonpolar solvents such as ethyl acetate have been recommended for the extraction of uncooked foods, and polar solvents such as 1-propanol:water (3:1 v/v) have been recommended for the extraction of baked foods that contain rye, wheat, or other starch-rich grains. A comparison of AR extraction methods has been investigated with the application of gas chromatography and a flame ionization detector (GC-FID) to quantify the AR content. The goal of this research was to compare the rapid accelerated solvent extraction of the alkylresorcinols (ASE-AR) method to the previous manual AR extraction methods. Results for this study as well as the investigation of the overall efficiency of ASE-AR extraction with the use of a spiking study indicated that it can be comparable to current extraction methods but with less time required. Furthermore, the extraction time for ASE (approximately 40 min) is much more convenient and less tedious and time-consuming than previously established methods, which range from 5 h for processed foods to 24 h for raw grains.  相似文献

The surface lipid content (SLC) of rice is often used to objectively measure the degree to which bran has been removed from rice kernels, commonly known as degree of milling (DOM). This study was conducted to evaluate new, rapid extraction technology for potential timesaving measurements of SLC of milled rice. The SLC of two long‐grain rice cultivars, Cypress and Drew, were determined using three extraction systems: Soxtec, accelerated solvent extraction (ASE), and supercritical fluid extraction (SFE). Before milling, rough rice was separated into three thickness fractions (<1.84, 1.84–1.98, and >1.98 mm) and samples from each thickness fraction were milled for durations of 10, 20, and 30 sec. Head rice collected from each milling duration was extracted using each of the three methods. Results showed that regardless of the extraction method, thinner kernels had lower SLC measurements than thicker fractions. In most cases, both the ASE and Soxtec produced SLC greater than that of the SFE. The ASE also showed SLC measurements at least as great as those from Soxtec extraction, suggesting that the ASE is as thorough in extracting lipids as commonly used methods.  相似文献

Supercritical fluid extraction of organochlorine pesticides in eggs   总被引:2,自引:0,他引:2  
The efficacy of supercritical fluid extraction (SFE) for the recovery of 16 common organochlorine pesticides (OCPs) from liquid whole eggs was investigated by employing supercritical carbon dioxide (SC-CO(2)) without the use of a solvent modifier to minimize interfering coextractives. The OCPs tested included aldrin; alpha-, beta-, delta-, and gamma-BHCs; p,p'-DDD, -DDE, and -DDT; dieldrin; endosulfans I, II, and sulfate; endrin; endrin aldehyde; heptachlor; and heptachlor epoxide. The SFE conditions were as follows: 10000 psi (680 bar), 40 degrees C, SC-CO(2) flow rate of 3.0 L/min with an extraction time of 40 min for a total of 120 L of CO(2). The OCPs were trapped off-line in an SPE cartridge containing Florisil and then eluted by an acetone/hexane mixture and analyzed by gas chromatography-electron capture detection (GC-ECD). Recovery studies were carried out on homogenized eggs fortified at the 0.05, 0.10, and 0.20 ppm levels. At the lowest level, 0.05 ppm, recoveries ranged from 81.8 to 108.3%, with CVs < 9.8%. All recoveries were significantly higher than those obtained by an AOAC/FDA solvent extraction method. Eggs containing incurred endosulfan I were also effectively extracted by SFE. This study suggests that the application of SFE for the extraction of OCPs from eggs will result in significant savings in analysis time and lower solvent use and disposal costs compared to conventional solvent extraction procedures.  相似文献

Bergamot peel is an underutilized byproduct of the essential oil and juice-processing industry. As with other Citrus peels, it still contains exploitable components, such as pectins and flavonoids. Commercial glycoside hydrolases, specifically a combination of pectolytic and cellulolytic enzymes, solubilized a high percentage of the material (81.94%). The flavonoid profile of the peel consisted of characteristic Citrus species flavanone rutinosides and neohesperosides derived from naringenin, eriodictyol, and hesperetin. In addition, a number of minor flavanone and flavone glycosides, not found in orange and lemon peels, were identified. The majority of flavonoids were extracted in the two 70% v/v EtOH extractions. Processing this material clearly has economic potential leading to low environmental impact.  相似文献

Renewed interest in plant-derived drugs has led to an increased need for efficient extraction methods. Hypericum perforatum L. contains several groups of bioactive compounds with noteworthy pharmacological activities. Direct sonication of H. perforatum was investigated and compared with conventional maceration, indirect sonication, Soxhlet extraction, and accelerated solvent extraction (ASE). Highly selective liquid chromatography/tandem mass spectrometry analysis showed that the content of six investigated active compounds (hypericin, pseudohypericin, hyperoside, rutin, quercitrin, and hyperforin) in extracts obtained by direct sonication was significantly higher than in extracts obtained by the other methods. The active compound contents increased on increasing the ultrasonic power from 40 to 60 W when using direct sonication. Conventional maceration gave the lowest amount of analyzed active compounds. Soxhlet extraction gave better results than ASE or indirect sonication.  相似文献

Antioxidants from orange juice are isolated by the use of countercurrent supercritical fluid extraction (CC-SFE) and characterized by reversed-phase liquid chromatography (RPLC) coupled to mass spectrometry (MS) and diode-array detection (DAD). A pilot-scale SFE plant equipped with a packed column has been employed for countercurrent extraction and fractionation of raw orange juice with carbon dioxide. Several experiments have been performed in order to study the effect of the countercurrent conditions on the content of antioxidative compounds. In this study, the main variable that has been considered is the solvent-to-feed ratio (S/F) because it plays an essential role in the extraction efficiency. The values tested covered a wide range of sample and solvent (CO(2)) flow rates. In each experimental run, two different extracted fractions and the residual nonextracted juice were obtained and characterized. Different flavonoids have been identified in the fractions obtained after CC-SFE. The possibility of using this process for antioxidant compounds enrichment is discussed.  相似文献

Large amounts of contaminated process dust remain from the procedure of pesticide treatments applied to seed pellets. A pilot study in analytical-scale supercritical fluid extraction (SFE) was performed to determine the possibility of using supercritical carbon dioxide for the extraction of the nonpolar insecticide carbosulfan and the more polar insecticide imidacloprid present in contaminated dust waste, at concentrations of up to 20% (w/w). The effects of various experimental conditions, such as temperature, flow rate, and addition of modifier, on the recovery of the analytes were evaluated by extracting the pesticides both from spiked support material and from real dust samples. It was found that carbosulfan could easily be extracted from the dust waste within 30 min at 138 bar and 40 degrees C with a recovery of 98.9% (RSD = 2.3%, n = 10), compared to values obtained with a validated liquid extraction method. A sufficient removal of the more polar substance imidacloprid required the addition of a modifier, and the results showed a strong dependence of the extraction efficiency on the choice of modifier. Extractions at 276 bar and 80 degrees C with a solvent consisting of supercritical carbon dioxide modified with methanol (5%) gave a recovery of 97.0% (RSD = 3.6%, n = 10) using a 40 min extraction time. The results indicate that it seems to be possible to use process-scale SFE for the decontamination of pesticides from dust waste. The conditions outlined also permit analytical determinations of the two insecticides based on a combination of SFE and liquid chromatography.  相似文献

This paper describes a new in-cell method for pursuing accelerated solvent extraction (ASE) prior to lipid analysis from food samples. It is difficult to pursue direct ASE with acid- or base-hydrolyzed samples due to the corrosive nature of the reagents and material limitations. In this study ion exchange based materials were used to remove acid or base reagents in-cell without compromising the recovery of lipids. The performance data are presented here for the new methods for lipid extraction for a variety of food samples and compared to the Mojonnier method. NIST Standard Reference Materials (SRM-1546 and SRM-1849) were used to validate the ASE methods. Excellent fat recoveries were obtained for the ASE methods. The new methods presented here enhance the utility of ASE and eliminate labor intensive protocols.  相似文献

In this paper, accelerated solvent extraction (ASE) for the analysis of ochratoxin A (OTA) is applied for the first time. Optimization of the method is given for the extraction of OTA from rice samples. Several parameters such as type of solvent, temperature, pressure, static time, and cell size were investigated thoroughly to find the optimal extraction conditions. The optimum ASE operating conditions were methanol as extraction solvent, 1500 psi, 40 degrees C, 5 min of static time, 50% flush volume, 60 s of purge, 1 cycle, and 11 mL cell size. The total extraction time was approximately 15 min. OTA was determined by liquid chromatography coupled with a fluorescence detector and confirmed by methyl ester derivatization. The analytical performance of the method was monitored with quality control parameters. Finally, the optimized method was used to evaluate 12 rice samples, 1 of which was positive with an OTA content of 4.17 ng/g. The proposed method offers the possibility of a fast and simple process to obtain a quantitative extraction of OTA.  相似文献

One way of investigating the function of soil is via the pool of low molecular weight organic compounds in the soil microbial biomass. This is because low molecular weight organic compounds have key roles in metabolism of soil microbes, can function in osmotic adjustment and other stress responses, and are intermediates in the breakdown of polymers to inorganic nutrients. Methods for measuring low molecular weight microbial metabolites in soil rely upon extracting total metabolites and then subtracting the contribution from metabolites in the soil extracellular matrix (i.e. microbial = total − extracellular). Recent studies have tested methods for extracting organic N monomers from the extracellular matrix of soil, but there has not been similar testing of methods for extracting total organic N monomers. The aims of this study were to examine methods for extracting total organic N monomers by a) contrasting chloroform gas fumigation with chloroform direct extraction, and b) examining whether it is possible to extract soil with two methods that combine quenching of metabolic activity with extraction, namely cold methanol/chloroform/water and hot aqueous ethanol. To evaluate methods, organic N compounds were extracted from soil and then capillary electrophoresis–mass spectrometry identified and quantified 42 organic N monomers including amino acids, quaternary ammonium compounds, nucleobases and nucleosides, amines and polyamines. Absolute concentrations of 32 out of the 42 quantified organic N monomers were significantly different between soil extracted by chloroform gas fumigation and chloroform direct extraction. These differences were probably a function of gains and losses of compounds due to oxidation, hydrolysis and deamidation during the two-day chloroform gas fumigation. Cold methanol/chloroform/water yielded large amounts of the extremely labile compound ergothioneine, probably because the extraction method rapidly quenched metabolic activity. The primary limitation of extraction with methanol/chloroform/water is that it was ineffective at extracting strongly cationic compounds (e.g. polyamines). Extraction with hot aqueous ethanol was unsuccessful with soil presumably because soil microbes are difficult to lyse. It is recommended that future studies examining organic N monomers in soil microbial biomass use chloroform direct extraction or cold methanol/chloroform/water rather than chloroform gas fumigation.  相似文献

This paper describes a new method for the rapid extraction and unequivocal confirmation of 13 sulfonamides (SAs) in raw meat and infant foods. The highly automated extraction procedure is based on accelerated solvent extraction followed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) as a confirmatory analysis. After 1 g of food matrix was blended with 2 g of C18 as a solid support material, the mixture was packed into the extraction cell and the SAs were extracted with 10 mL of hot water at 160 degrees C and 100 atm; 100 microL of the extract was directly injected into the LC-MS system. The analytes were ionized in an electrospray interface operating in the positive ion mode and were identified by selecting two multireaction monitoring transitions, which guaranteed method specificity. Typical recoveries from crude meat and baby food samples ranged from 70 to 101% at a fortification level of 100 ppb, corresponding to the maximum residue limits established by the European Union and the U.S. Food and Drug Administration. The interday method precision was less than 8.5%, and the limits of detection were below 2.6 ppb. This study has taken matrix-induced suppression of ionization into account, by comparing standard and matrix-matched calibration curves. Four of the 13 monitored SAs have been detected in some baby foods and raw meat samples, bought from Roman supermarkets and butchers' shops, using the described methodology.  相似文献

Supercritical fluid carbon dioxide (SC-CO2), when used with an extraction enhancer, comprises a supercritical fluid extraction (SFE) system for extraction of pesticides and matrix components from fatty and nonfatty foods. After being mixed with the enhancer, samples ranging from 95% water to pure lipophilic oils can be extracted efficiently with SC-CO2. This extraction technique yields analyte recoveries in excess of 85% for over 30 types of pesticides at incurred levels ranging from 0.005 to 2 ppm in such diverse matrixes as carrots, lettuce, peanut butter, hamburger, and fortified butter fat and fortified potatoes. SC-CO2 provides a solvent medium that is nontoxic, nonflammable, and inexpensive while also eliminating the use and disposal of potentially carcinogenic organic solvents.  相似文献

黑苦荞米黄酮提取工艺优化及其降血糖活性研究   总被引:2,自引:2,他引:0  
为了获得合适的黑苦荞米黄酮提取工艺,该文采用单因素试验和响应面设计,选择乙醇体积分数、料液比、提取时间、提取温度4个因素,优化黑苦荞米黄酮提取工艺。试验结果表明,黑苦荞米黄酮的最佳提取条件是:乙醇体积分数54%,料液比1:24 g/m L,提取时间62 min,提取温度71℃。在此条件下,理论黄酮得率为2.21%,实际黄酮得率为2.20%,相对误差为0.45%。在此基础上进一步研究了黑苦荞米黄酮的α-淀粉酶抑制活性,结果表明7.5 mg/m L的黑苦荞米黄酮对α-淀粉酶活性的抑制率为54.05%,与二甲双胍(5 mg/m L)效果相当;此外,与空白对照相比,50μg/m L黑苦荞米黄酮能显著(P0.05)提高肝脏细胞Hep G2的葡萄糖消耗量(48.73%),并促进肝脏细胞糖原的合成。研究结果表明,黑苦荞米黄酮具有较好的辅助降血糖功效。  相似文献
