This study was designed to evaluate the effectiveness of recombinant Ovalbumin-LHRL (OL) immunization on changes in testicular size, histological appearance and testosterone production in buck kids. Thirty native buck kids at 18 weeks of age were divided into three groups, control (n = 10), immunization (n = 10) and castration (n = 10) groups. Immunized animals received OL protein generated by recombinant DNA technology. Ultrasonographic and histological examinations of the testes were performed. Animals were slaughtered at 44 weeks of age. Semen and epididymides were evaluated for the presence of sperm cells. Immunized animals generated anti-LHRH antibodies. Testosterone production, testicular and accessory glands development and sperm production were suppressed in the immunized animals (p < 0.01). Semineferous tubule diameters decreased (p < 0.01), basal membrane of the tubule was thickened and hyalinized in immunized kids. Immunization affected ultrasonographic appearance of the testes drastically. While testes of control animals gained their normal ultrasonographic appearance as the age increased, immunized animals had uniform hypoechogenic testicular structure as observed at 18 weeks of age until slaughter. Simultaneous histological and ultrasonographic evaluations indicated that the changes in testicular histology could partly be monitored via ultrasonographic imaging; nevertheless, it is difficult to claim that ultrasonographic image reflects the exact changes in such instances. In conclusion, these results indicate that recombinant OL fusion protein is effective in immunocastration in buck kids and has a potential to be used as an alternative to physical castration. Further researches should be conducted to help assessing reproductive status of testes from ultrasound images.  相似文献

Two recombinant luteinizing hormone releasing hormone (LHRH) fusion proteins were evaluated for their effectiveness in suppression of testicular development and histology by injecting together. Recombinant fusion proteins, ovalbumin-LHRH-7 and thioredoxin-LHRH-7, were generated using recombinant DNA technology and expressed in E. Coli. Eleven ram lambs ranked by age and body weight were randomly assigned to receive either ovalbumin and thioredoxin recombinant protein mixture (control group, n = 5) or ovalbumin-LHRH-7 and thioredoxin-LHRH-7 recombinant fusion protein mixture, anti-LHRH vaccine, (immunization group, n = 6). Animals in each group were weaned at 17 wk of age and were injected (primary immunization) with either mixture at 18 wk of age. Both groups received a booster immunization 8 wks later (26 wk of age). Scrotal circumference, scrotal length, testicular diameter and testicular length were measured in both groups every other week. All animals were slaughtered at 36 wk of age. Immediately after slaughter, a small testicular tissue was taken and processed for histological examination. In the ram lambs in immunization group scrotal circumference and testicular diameter increased steadily until second booster and then remained as a plateau until the end of the experiment. The differences in scrotal circumferences and testicular diameter were significant between the two groups during the last three weeks of the study (p < 0.05). There were no differences in testicular and scrotal length throughout the study (p > 0.05). Seminiferous tubules lost their regular shape and were decreased in diameter in immunization group. Although a few spermatozoa were seen in some tubules, in general, there were atrophy of the seminiferous tubules and loss of spermatogenesis, nevertheless, it seemed that animals in this group were potentially fertile.  相似文献

The objective of this study was to evaluate the effect of a GnRH-vaccine in the ram lamb. Experiments were performed using 20 male lambs, randomly divided into a test (GnRH-immunization) and control group (physiological NaCl-solution). At a body weight of 20 kg (age 2-3 months) and three weeks later, all animals of the test group received 2 ml of Improvac (CSL Limited, Parkville, Victoria, Australia). The body weight as well as the blood testosterone concentration were measured weekly for 16 weeks. Thereafter, blood samples for testosterone analysis were taken monthly in immunized lams only. After the booster injection testicular growth was suppressed and plasma testosterone remained at low values < 0.1 ng/ml for at least 12 weeks. The mean corresponding testosterone concentrations for the control lambs ranged between 0.1 and 0.9 ng/ml plasma. An increase of testosterone was observed in 8 of 10 immunized animals between 3 to 7 months after the booster dose. The control lambs showed a tendency for better growth rate than vaccinated animals, but the difference was not significant. Our results demonstrate that in prepubertal ram lambs two immunizations with Improvac, three weeks apart, can suppress testosterone secretion and testicular growth at least for three months after the booster injection. For a suppression of reproductive function longer than three months after the second vaccination, a third immunization is needed at this time or when testicular growth is beginning.  相似文献

Spermatogonial stem cell transplantation is a technique that has potential in livestock to enhance genetic gain and generate transgenic offspring through the male germ line. A means for depletion of endogenous germ cells in a recipient's seminiferous tubules is necessary for this technology to be applied. The objectives of this study were to evaluate several methods for depletion of endogenous germ cells in the testes of adult rams and to evaluate ultrasound-guided injections into the rete testes as a means for infusing a suspension into the seminiferous tubules. Sixteen adult rams were randomly divided into 4 treatment groups (n = 4 per group). Treatments consisted of active immunization against LHRH (IMM), localized testicular irradiation (IR), LHRH immunization + irradiation (IMM+IR), and untreated control. Serial bleedings were conducted pretreatment and monthly after treatment for 4 mo, at which time all rams were castrated. Both IMM and IMM+IR rams received exogenous gonadotropin in the form of Perganol weekly for 8 wk before castration to bypass the immunization. All rams also received an ultrasound-guided injection of PBS containing 0.4% trypan blue into the rete testis of one testicle before castration. Rams receiving IMM and IMM+IR treatments had higher (P < 0.05) average percentages of seminiferous tubule cross sections with depleted germ cells compared with controls. Serum testosterone was decreased (P < 0.05) in IMM and IMM+IR rams 1 mo after treatment and throughout the remainder of the study compared with controls and IR rams, which were not different from each other. Serum inhibin concentration was unchanged in all rams following treatment indicating that Sertoli cell function was unaltered. A greater (P < 0.05) average percentage of the total testicular area could be filled with the trypan blue solution by rete testis injection in IMM and IMM+IR rams. These data demonstrate the depletion of endogenous germ cells in adult ram testes without alteration of Sertoli cell viability and function that have potential as methods for preparing recipient animals for germ cell transplantation.  相似文献

The gonadal hormone inhibin regulates daily sperm production (DSP) indirectly through negative feedback control of FSH secretion and may also affect DSP via direct actions within the testis. Studies attempting to increase DSP through the immunization against inhibin have yielded equivocal results. The current study compared 2 inhibin antigens for effects on DSP and hormone secretion. Hampshire ram lambs (BW = 42 +/- 2 kg; age = 113 +/- 3 d) were assigned randomly to 3 groups: 1) control (n = 4); 2) alpha-peptide conjugate (PTC, n = 6); and 3) alpha-subunit (SUB, n = 6). Antigen PTC consisted of an alpha-inhibin, N-terminal, 25-amino acid peptide conjugated to ovalbumin. Antigen SUB was the complete inhibin alpha-subunit. Lambs were immunized on d 0 (June 19, 2006), 18, 38, and 63. Body weight was recorded on immunization days and scrotal circumference on d 63. Blood samples were collected on d 0, 7, 14, 18, 28, 35, 38, 49, 56, 63, and 70. Rams were slaughtered on d 71. Testes were weighed, and parenchyma was obtained for DSP determination. Plasma alpha-inhibin antibody titer and LH, FSH, and testosterone concentrations were measured. alpha-Inhibin antibody titer was first detectable on d 14 in both PTC- and SUB-immunized ram lambs and generally increased thereafter. Mean DSP per gram of testis (DSP/g) was increased (P < 0.01) 26% in PTC- and SUB-immunized ram lambs over that in control ram lambs. Total DSP per ram lamb and testes weight did not differ among the 3 treatment groups. Variation in DSP per ram lamb and testes weight were greater (P = 0.05) in PTC- and SUB-immunized ram lambs than in control ram lambs. Plasma FSH concentrations were similar in PTC- and SUB-immunized ram lambs. Immunization against either alpha-inhibin antigen did not alter LH, testosterone, BW, or scrotal circumference. Findings indicate that 1) the 2 alpha-inhibin antigens increase DSP/g to similar extents; 2) alpha-inhibin antibody may act at least in part through an intratesticular mechanism because DSP/g was increased in some animals without concomitant increases in FSH; and 3) immunization against alpha-inhibin may affect testes weight by actions independent of those that regulate DSP/g.  相似文献

The effect of active immunization against GnRH on production, carcass, and behavioral traits was examined in ram lambs fed to a uniform slaughter weight. Lambs (initial BW = 32.6+/-1 kg) were stratified by BW and assigned at random to one of four treatment groups (n = 12 lambs/group). Lambs were untreated, castrated, or actively immunized against GnRH using a GnRH-keyhole limpet hemocyanin conjugate (1 mg) emulsified with either Freund's complete adjuvant (FCA) or another oil-based adjuvant (ISA). Animals were housed individually and slaughtered at 58 kg BW. Immunoneutralization of GnRH reduced (P < .05) testes weight and the concentration of testosterone in serum at slaughter. Suppression of testicular size and function was most clearly evident in animals immunized using FCA. Final anti-GnRH titer was also highest in lambs immunized using FCA. Several measures of sexual behavior (frequency of mounts and ejaculations) were also reduced (P < .05) in animals immunized using FCA. The duration of the feeding period was greater (P < .05) for castrated lambs than for untreated lambs, and intermediate feeding periods were required for FCA and ISA lambs. Average daily gain was greater (P < .05) in untreated than in castrated, FCA, or ISA lambs. Similarly, feed efficiency for untreated lambs was greater (P < .05) than for castrated, FCA, or ISA lambs, but feed efficiency did not differ among castrated, FCA, or ISA lambs. Longissimus muscle area, lean and bone maturity, overall quality, muscling score, flank streaking, and color of fat did not differ among treatments. Intact, FCA, and ISA lambs had more (P < .05) desirable yield grades, less (P < .05) backfat, and less (P < .05) marbling than castrated lambs. In summary, immunization against GnRH decreased testicular weight and reduced (P < .05) feedlot performance and sexual behavior to levels comparable to those of castrated males. Partitioning of nutrients for growth and deposition of fat, however, seems to differ among immunologically castrated and physically castrated lambs. This difference in nutrient partitioning may be due to residual testicular activity in immunized lambs.  相似文献

The objectives of this study were 1) to evaluate the ability of trenbolone acetate (TBA) administered in tandem with LHRH immunization to suppress reproductive function in bulls and 2) to examine the effects of LHRH and androgen (TBA) signaling on pituitary gland function. Forty-four Angus × Hereford crossbred calves (BW=225 ± 2 kg; age=187 ± 6 d) received castration, LHRH immunization, or TBA administration in a 2 × 2 × 2 factorial design. Treatment groups receiving LHRH immunization contained 6 animals, whereas other treatment groups contained 5 animals. Animals immunized against LHRH received a primary injection and 2 booster injections of ovalbumin-LHRH-7 fusion protein on d 0, 42, and 196, respectively. Animals treated with TBA were implanted on d 224. Serum LHRH antibodies increased (P<0.05) after each booster for immunized animals, but were negligible in nonimmunized animals throughout the experiment. Serum testosterone concentration (P<0.001) and scrotal circumference (P<0.05) were depressed in LHRH-immunized bulls compared with nonimmunized bulls by d 84 and 168 of the experiment, respectively. Treatment with TBA tended (P=0.08) to decrease serum testosterone concentrations of nonimmunized bulls. Weights of testes at slaughter were decreased (P<0.001) for LHRH-immunized (232 ± 41 g) compared with nonimmunized (752 ± 45 g) bulls, but did not differ (P=0.80) between TBA-implanted (500 ± 49 g) and nonimplanted bulls (484 ± 36 g). Both LHRH immunization and castration decreased pituitary gland stores of LH and FSH (P<0. 001). There was no effect (P>0.10) of TBA on pituitary gland FSH content and only a tendency (P=0.09) to increase pituitary gland LH content. Immunization against LHRH decreased expression of LH β-subunit and common α-subunit genes (P<0.001). Castration increased expression of LH β-subunit and common α-subunit genes (P=0.02). Treatment with TBA further suppressed (P=0.04) α-subunit mRNA expression in LHRH-immunized steers. In summary, LHRH immunization decreased synthesis and storage of LH and decreased storage, but not synthesis of FSH in bulls. The increased synthesis of LH and FSH in nonimmunized, but not LHRH-immunized steers suggests that castration removes the negative feedback on gonadotropin synthesis but that LHRH is still needed for release of these hormones. Androgen replacement with TBA did not restore the negative feedback control of gonadotropin synthesis.  相似文献

Spermatogonial transplantation will provide a new way to study spermatogenesis in domestic animals, disseminate male genetics and produce transgenic animals, if efficiency can be improved. We evaluated a 'surgical' method for transplanting donor cells into testes of ram lambs, where the head of the epididymis is reflected, and a catheter introduced into the extra-testicular rete testis. We also tested transduction of ram spermatogonia with a lentiviral (LV) vector as a means to identify permanent colonization, and introduce genes into donor cells. Eight ram lambs, 11- to 13-week olds, were the recipients: in five, spermatogonia were injected into one testis, and the contralateral testis was an un-manipulated control: in two, spermatogonia were injected into one testis and the contralateral was sham-injected: in one, both testes were injected. Six lambs received spermatogonia labelled with a cell-tracking dye and these were collected 1 or 2 weeks after transplantation; three lambs received spermatogonia transduced with a LV vector driving the expression of enhanced Green Fluorescence Protein and these were collected after 2 months. Donor cells were detected by immunohistochemistry in tubules of seven of nine recipient testes. Approximately 22% of tubule cross-sections contained donor cells immediately after transplantation, and 0.2% contained virally transduced cells 2 months after transplantation. The onset of spermatogenesis was delayed, and there were lesions in both injected and sham-injected testes. Despite the effects of the surgery, elongated spermatids were present in one recipient testis 2 months after surgery. The results suggest that, after modifying the surgical and transduction techniques, this approach will be a means to produce good colonization by donor spermatogonia in sheep testes.  相似文献

Twenty-five ram lambs were immunized against α-inhibin peptide emulsified in Freund's adjuvant (FRA), Emulsigen (EML) containing an oligodeoxynucleotide as an immunostimulant, or adjuvant without α-inhibin antigen (control). Four immunizations were administered during an 85-d period, after which testes were obtained for determination of daily sperm production (DSP) and histological evaluation. α-Inhibin antibody (Ab) titers were 70-fold greater in lambs treated with FRA than in EML-treated ram lambs. α-Inhibin immunization had no effect on testes weight or on plasma concentrations of follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone. Mean DSP/g tended (P = 0.1) to be greater in α-inhibin–immunized (EML = 17.6 × 106; FRA = 15.8 × 106) ram lambs than in control animals (14.4 × 106). One of the 8 control ram lambs had an elevated DSP/g, which was a statistical outlier. Without data from this lamb, DSP/g was increased (P < 0.01) in α-inhibin–immunized ram lambs by 28% over controls. No association was found between the titer of α-inhibin Ab developed and DSP/g. Histologically, the percentage of testicular area occupied by seminiferous tubules differed (P = 0.01) by treatment and was greatest (82%) in EML-treated ram α-inhibin–immunized lambs and lowest (74%) in control animals. Percentage tubular area and DSP/g were correlated (r = 0.57, P = 0.003). Findings show that (1) the extent of the increase in DSP/g is not dependent on the titer of α-inhibin Ab; (2) the increase in DSP/g is achieved through an increase in the mass of seminiferous tubules; and (3) FRA elicits a greater α-inhibin Ab titer than EML containing an oligodeoxynucleotide.  相似文献

The relationship between systemic luteinizing hormone (LH) and testicular development was investigated in Finn and Suffolk ram lambs treated at 2 wk of age with sc progesterone implants for 0, 4, 8 or 12 wk. Serial samples of blood (30-min intervals for 3 h) were collected from five rams X breed-1 X treatment-1 at 6, 8, 10, 12, 14, 18 and 22 wk of age plus 1 and 2 wk after implant removal. Scrotal circumference was measured at 10, 14, 18 and 22 wk of age. A testicular biopsy was obtained at 14, 18 and 22 wk for microscopic evaluation of testicular development and spermatogenesis. Pulsatile LH releases occurred 60 to 180 min apart in control lambs between 6 and 22 wk of age; LH secretion was not affected by age except for a decrease (P less than .01) in mean LH at 22 wk. Transient increases in testosterone were found subsequent to LH pulses. Systemic testosterone increased (P less than .01) progressively with age, was higher (P less than .05) in Finn than in Suffolk rams at 18 and 22 wk and was correlated positively (P less than .01) within breeds with seminiferous tubule diameter at 14 wk and with scrotal circumference at 10 and 14 wk. Tubule diameter was larger (P less than .01) but testes size and weight were smaller (P less than .01) in Finn than in Suffolk rams. Progesterone implants decreased (P less than .01) LH secretion at 4, 6, 8, 10 and 12, but not at 14 wk of age; decreased systemic testosterone between 4 and 14 wk; decreased (P less than .01) seminiferous tubule diameter at 14 wk, which was related inversely to duration of progesterone treatment; and delayed puberty from 18 to 22 or more wk in rams implanted from 2 to 10 or 14 wk of age. Both control and treated rams with elongated spermatids at 18 vs 22 or more wk had increased LH secretion, higher systemic testosterone and larger tubule diameter and testes size at a younger age. These results suggest that rate of sexual maturation in ram lambs is related to level of postnatal LH stimulation and to the prepubertal age when increased LH stimulation occurs.  相似文献

Two field trials were conducted in Brazil to evaluate LHRH immunocastration of Bos indicus bulls (d 0 = 2 yr of age). In Study I, 72 bulls were assigned randomly to one of three treatment groups: LHRH0-immunized, castrated, and intact. Immunized animals (n = 25) received a primary and two booster injections of ovalbumin-LHRH-7 and thioredoxin-LHRH-7 fusion proteins on d 0, 141, and 287. Twenty-three bulls were surgically castrated on d 141, and 24 served as intact controls. All animals were slaughtered on d 385, at approximately 3 yr of age. In Study II, 216 bulls were assigned randomly to the same three treatments as in Study I; however, because of a drought in the area, bulls were kept on pasture an additional year, and a fourth treatment was added, in which one-half the LHRH-immunized bulls received an additional booster on d 639 (fourth immunization). All animals in Study II were slaughtered on d 741 (4 yr of age). Luteinizing hormone-releasing hormone antibodies increased following each immunization for immunized bulls, but they were not detectable in castrate or intact animals in either study. Consequently, scrotal circumference was suppressed in immunized bulls compared with intact controls in both studies. By d 287, serum concentrations of testosterone in LHRH-immunized bulls were decreased compared with intact controls (P < 0.01). In both studies, testes and epididymal weights at slaughter were greater (P < 0.01) for intact (500 +/- 17 and 60 +/- 2 g, respectively) than for immunized bulls (173 +/- 22 and 26 +/- 2 g, respectively) and fourth immunization bulls (78 +/- 23 and 20 +/- 2 g, respectively; Study II). At the end of each study, BW was greater (P < 0.01) for intact bulls than for castrated and LHRH-immunized animals. In these two studies, the efficacy of the LHRH fusion proteins to induce an effect similar to that of surgical castration was considered 92 and 93%, respectively. These data support the concept that immunocastration of bulls at 2 yr of age was successful and that it has practical application as a tool for producing grass-fattened bulls in Brazil.  相似文献

We have investigated, under the normal conditions of local Chinese pig farming, castration of young male pigs by vaccination with a newly developed vaccine against gonadotrophin releasing hormone (GnRH). Because of the very early onset of puberty, long fattening period and relatively harsh circumstances in Chinese pig production, an investigation of the endocrine response of Chinese breeds to this type of vaccination was of particular interest. Fifteen crossbred boars (Yorkshire × Yanan) from three different litters were randomly assigned to three groups of five animals each. The first group was immunized at 13 weeks of age with a GnRH tandem dimer OVA‐conjugate in Specol and received a booster immunization 8 weeks later. The second group was injected with Specol alone and served as untreated controls. The remaining group was surgically castrated at the time of weaning (at 6 weeks of age). Pigs were fed ad libitum from weaning onwards. All animals were slaughtered at 31 weeks of age. Immunized boars had undetectable or low serum testosterone (0.09 ± 0.12 ng/ml), low fat androstenone (0.05 ± 0.01 μg/g) levels and very low testes weights (19.1 ± 4.3 g). Intact controls had much higher serum levels of testosterone (9.76 ± 4.81 ng/ml), fat androstenone levels (2.26 ± 0.87 μg/g) and testes weights (114.3 ± 29.41 g) at slaughter. Both the immunized and castrated group grew significantly faster than intact boars (p < 0.01). Average daily gains in immunized, castrated and intact animals were 0.69 ± 0.08, 0.63 ± 0.05 and 0.42 ± 0.07 kg (mean ± SD), respectively. The present data demonstrate for the first time that the newly developed anti‐GnRH vaccine works very well under practical Chinese pig farming conditions, and can be an attractive alternative to surgical castration.  相似文献

Objective: To describe laparoscopic‐assisted colopexy and sterilization, and evaluate outcome and effects in healthy male dogs. Study Design: Experimental study. Animals: Male Beagle dogs (n=7). Methods: A laparoscopic‐assisted, extracorporeally sutured colopexy, and sterilization by ligation and section of the testicular vessels and ductus deferens were evaluated 11 weeks after surgery. Ex vivo tensile tests were performed on the colopexy sites and loss of testicular function was assessed by monitoring serum testosterone, and by ultrasonographic and histologic examinations of the testes. Systemic inflammation and potential iatrogenic colonic functional disorders were investigated by monitoring serum C‐reactive protein (CRP) in the perioperative period and from a sulfapyridine (SP) kinetic profile obtained before and 10 weeks after surgery. Results: No intraoperative complications were recorded and clinical outcome was considered fair in all dogs. A mean tensile force of 42 N was required to disrupt colopexies. No relevant postoperative increase in CRP concentrations or changes in SP kinetics were observed. Testicular function was lost. Conclusions: Laparoscopic‐assisted colopexy achieved adhesion of the colon to the abdominal wall and testicular endocrine function and spermatogenesis were eliminated by laparoscopic castration.  相似文献

This study evaluated the effectiveness of a LHRH fusion protein vaccine on endocrine changes, feedlot performance, and carcass quality of bulls compared with steers and hormone-implanted steers. Crossbred bulls (n = 30; mean weight, 179 +/- 4 kg; mean age, 130 +/- 2 d) were randomly assigned to three treatment groups: 1) castrated (castrated; n = 10); 2) castrated-implanted with trenbolone acetate (implanted; n = 10); and 3) immunized against a cocktail of recombinant fusion proteins, ovalbumin-LHRH-7 and thioredoxin-LHRH-7 (immunized bulls; n = 10). Blood was collected every 2 wk to evaluate antibody and hormone concentrations. Serum LHRH antibodies (P < 0.001) were detected in animals of the immunized group, which had reduced serum LH concentrations (P < 0.001) compared with the castrated groups and serum FSH concentrations, which did not decrease but were significantly different when compared with castrated and implanted animals. Serum testosterone concentrations in the immunized bulls were not different from the two castrated groups (P > 0.05) by d 60 after primary immunization. Initial mean scrotal circumference of the immunized bulls was 18.0 +/- 0.6 cm on d 0 and increased to 22.6 +/- 1.3 cm by d 310. No differences (P > 0.05) in ADG were observed among treatment groups. Immunized animals had an intermediate BW gain (P > 0.05) when compared with the castrates, whereas the castrated groups differed (P < 0.05) from each other. Carcass characteristics were similar (P < 0.05) among the three groups. Vaccinating bulls against a LHRH fusion protein cocktail suppressed LH and testosterone, which led to reduced testicular development and no bullock carcasses. Growth and carcass characteristics of the immunized animals were similar to the steers.  相似文献

A 19‐year‐old Hackney pony was referred to a veterinary teaching hospital for further investigation of left‐sided scrotal enlargement. Both testes were normally positioned within the scrotum and there was no pain on palpation. Ultrasonography revealed multiple masses of soft tissue echogenicity, located adjacent to the testicular parenchyma. They were hyperechoic when compared to the testicular parenchyma and affected both testes. Surgical excision was performed by closed castration. Gross examination revealed firm, white, nodular masses of varying size on the surface of both testes. The cut surface of individual masses had a dense, whorling, fibrous and somewhat shiny appearance. The histopathological diagnosis was bilateral, multicentric, well differentiated leiomyoma of the tunica albuginea. The smooth muscle origin was confirmed by immunohistochemical staining with an anti‐human monoclonal antibody reactive to smooth muscle actin. ObjectivesTo characterize changes in the cerebro‐cortical response of lambs to rubber‐ring castration during the first 6 weeks of postnatal life.AnimalsCoopworth‐Texel cross ram lambs between 3 hours and 44 days of age.MethodsThe electroencephalogram (EEG) and heart rate responses to rubber ring castration were compared. Anaesthesia was induced via a face mask and maintained with halothane in oxygen (e′Hal = 1.2%). Once a stable plane of anaesthesia had been achieved, data collection of EEG and electrocardiogram (ECG) commenced, and the lambs were castrated 15 minutes later, using rubber rings. Heart rate was derived from the ECG and the median frequency (F50), spectral edge frequency (F95) and total power (ptot) were derived from the EEG.ResultsCastration‐induced changes in F50 increased from 2 ± 1.8% in the youngest lambs to a maximum of 33 ± 8.9% in the 36 ± 0.5 day‐old lambs. Changes in F95 increased from ?7 ± 3% in the youngest lambs to a maximum of 16 ± 11% in the 36 ± 0.5 day‐old lambs. Linear regression analysis in lambs up to 10 days of age demonstrated a change in the response with age that was significantly different from zero for F50 (r2 = 0.28, p = 0.007) and F95 (r2 = 0.38, p = 0.001), but not for ptot or heart rate.ConclusionThis study identified significant changes in the responsiveness of the lambs’ cerebral cortex to the noxious stimulation of castration over the first 7–10 days of postnatal life. The results suggest that mechanisms that suppress responses of the fetus to noxious stimulation may still be active in the first few days after birth.Clinical relevanceThis study documents changes in central nociceptive processing which may reflect the ability of neonatal animals to perceive pain.  相似文献

The objective of the present study was to determine if postpubertal boars (12-13 months of age; 156 +/- 8 kg) with large testes had altered hypothalamic control of secretion of luteinizing hormone (LH). Seven boars with the highest estimated 150 d, paired testis weights from a line selected for large testes (769 +/- 60 g = mean weight of excised testes) and 8 boars from a control group (control, 544 +/- 20 g) were tethered in stalls and fitted with indwelling jugular catheters. Males were bled when they were intact, 14 days after castration and during administration of sodium pentobarbital anesthetic (subsequent to castration) to block secretion of endogenous LH-releasing hormone (LHRH). Blood samples were collected at 12-min intervals for 6 hr before and 1 hr after intravenous injection of LHRH in intact and castrated males. During anesthesia, LHRH was administered 4 times at 1-hr intervals and blood samples were collected every 6 min. All samples were analyzed for concentrations of LH and pooled samples were analyzed for concentrations of 17-beta estradiol (E2) and testosterone (T). In intact and castrated males, mean concentrations of LH, frequency and amplitude of pulses of LH, and concentrations of E2 and T were not different between boars of the two groups (P greater than .10). Response to exogenous LHRH was less (P less than .05) in intact males with large testes than in corresponding males from the control group (P less than .05). Objective To investigate the effects of age at castration on the subsequent behavioural response to tail docking. Study design Randomised prospective blinded experimental study. Animals Forty‐five male lambs were admitted to the study at birth. Methods The lambs were allocated into two groups for castration by rubber ring at 1 (1D, n = 20) or 10 (10D, n = 25) days of age. Following castration both groups of lambs were tail‐docked by rubber ring at between 26 and 34 days of age. Their behavioural responses to tail docking were recorded by video for 30 minutes before (n = 11) and 30 minutes after (n = 45) tail docking. Statistical differences were tested using anova with Mann‐Whitney post‐hoc testing. Results Two data sets were removed resulting in n = 19 and = 24 at 1D and 10D, respectively. A significant increase in the incidence of active and abnormal postural behaviour was seen in both groups after tail docking. Three behaviours were significantly different (p < 0.05) between the two groups. These behaviours were rolling, standing unsteadily and the total proportion of time standing with abnormal posture. These behaviours all had greater incidence in the lambs castrated at 1D of age. Conclusions Lambs castrated at 1D appeared to perceive a greater intensity of pain after subsequent tail docking than lambs castrated at 10D. Clinical relevance The timing of a single noxious stimulus in young animals can affect subsequent pain perception.  相似文献

A comparative study has been made in lambs 3 to 6 weeks of age of the behavioural responses and changes in plasma immunoreactive beta-endorphin (ir beta-endorphin) and cortisol after docking or docking plus castration by the application of rubber rings or by surgery. The use of rubber rings on lambs at this age was characterised by very agitated behaviour indicative of considerable distress for a period of up to 1 h. In contrast, surgery was accompanied by some post-operative shock exhibited by reduced exploratory and social behaviour, at least in the lambs exposed to docking plus castration. In the latter group there were highly significant increases in both plasma ir beta-endorphin and cortisol concentrations that may be consistent with the induction of stress-induced analgesia. We conclude that surgery caused less distress than the rubber rings, and is therefore preferable for docking and castration of lambs 3 to 6 weeks of age.  相似文献

Prepubertal Quarter horse colts were immunized at 6 mo of age with either estrone-17-oxime-bovine serum albumin (n = 4; treated) or with albumin only (n = 5; controls). All colts received booster injections of the appropriate antigen at 8, 10, 12, 16 and 20 mo of age. Blood samples were drawn every 20 d from 6 to 26 mo of age; body weights were determined monthly. Immunization against estrone-albumin resulted in increased binding of [3H]-estradiol in serum within 40 d that was maintained through 24 mo of age. Antisera from treated colts crossreacted equally well with estrone and estradiol and moderately with other estrogens; androgens, progesterone and glucocorticoids all cross-reacted less than .005%. Body weights were not affected by treatment. Concentrations of testosterone were generally higher (P less than .05) in estrogen-immunized colts compared with controls after immunization. Concentrations of luteinizing hormone were not affected by treatment, whereas concentrations of follicle-stimulating hormone were initially increased (P less than .05) in treated colts after immunization. At castration at 27 mo of age, estrogen-immunized colts had greater (P less than .05) testicular and parenchymal weights and produced more (P approximately equal to .055) spermatozoa per horse than did control colts. Seminal characteristics immediately before castration were not affected by treatment. It appears that estrogens are involved in the regulation of several reproductive traits in the colt. Moreover, active immunization against estrogen in the prepubertal colt may be a useful method of increasing testicular size and sperm production rates in the stallion after puberty.  相似文献
