We previously reported that growth hormone (GH) pulses were negatively associated with neuropeptide Y (NPY) profiles in cerebrospinal fluid (CSF) of the third ventricle of Shiba goats. In addition, while most GH pulses were coincident with GH-releasing hormone (GHRH) pulses, there was no correlation between GH and somatostatin (SRIF) levels. The present study was performed to elucidate the relationship between GH pulses and these neuropeptide levels in CSF when estradiol (1.0 mg/head) was subcutaneously administered to ovariectomized goats. CSF and plasma samples were collected every 15 min for 18 h (from 6 h before to 12 h after injection). GH levels in peripheral blood and GHRH, SRIF and NPY levels in CSF were measured by radioimmunoassay. Pulse/trough characteristics and correlations were assessed by the ULTRA algorithm and cross-correlation analysis. Before estradiol was injected, significant coincidence was found between GHRH pulses and GH pulses, and negative coincidence was found between NPY troughs and GH pulses. Six to 12 h after estradiol injection, the amplitude and area under the curve (AUC) of the GH pulses were markedly increased. The duration and AUC of the GHRH pulses in the CSF were also increased, and stronger synchrony of GHRH with GH was observed. In contrast, the baseline of NPY was significantly decreased, and the negative correlation between the GH pulses and NPY troughs disappeared. The parameters of SRIF troughs were not clearly changed. These observations suggest that estrogen enhances the pattern of secretion of GH in the goat via enhancement of GHRH pulses and decrease of NPY levels.  相似文献

Bovine growth hormone (bGH) gene polymorphism of leucine (Leu)-threonine (Thr) (allele A), valine (Val)-Thr (allele B), and Val-methionine (Met) (allele C) at codons 127 and 172 was shown to relate with carcass trait variations in Japanese Black cattle. In this study, 10-mo-old Japanese Black heifers with growth hormone (GH) genotypes AA, AB, BB, AC, BC, and CC (N = 141) were compared for basal GH, insulin-like growth factor-1 (IGF-1), insulin, ghrelin, glucose, and nonesterified fatty acid (NEFA) concentrations. Growth hormone release was also measured as response to growth hormone–releasing hormone (GHRH) (0.4 μg/kg body weight [BW]) using 18 heifers with GH genotypes AA, BB, and CC (n = 6 for each group). The genotype AA heifers showed the greatest BW among genotypes (P < 0.05). Genotype AC, BC, and CC heifers showed greater GH concentrations than genotype AA, AB, or BB heifers, in which genotype CC heifers had the highest concentrations (P < 0.05). However, IGF-1 concentrations did not significantly differ. The genotype AA and BB heifers had a greater GH release at 60 min following GHRH injection than did the genotype CC heifers. The area under the curve (AUC; P < 0.07) and incremental area (IA; P < 0.08) of GH responses to the GHRH challenge tended to be the highest in the genotype AA heifers and the lowest in the genotype CC heifers. In conclusion, GH gene polymorphism altered GH, which may have contributed to differences in BW and carcass traits among genotypes.  相似文献

Growth hormone (GH) is secreted in a pulsatile manner, but the underlying mechanisms of GH pulse generation remain to be resolved. In the present study, we investigated the relationship between GH pulses in the peripheral circulation and GH-releasing hormone (GHRH) and somatostatin (SRIF) profiles in the cerebrospinal fluid (CSF) of male goats. The effects of an intracerebroventricular (icv) injection of neuropeptide Y (NPY), galanin and ghrelin were also analyzed. Blood and CSF samples were collected every 15 min for 8 hr from the jugular vein and third ventricle, respectively. GH pulsatility in the goat was found to consist of distinct large pulses of 5 hr periodicity and small pulses of 1 hr periodicity. GHRH and SRIF in the CSF fluctuated in a pulsatile manner with 1 hr periodicity, and most of the descending phase of SRIF pulses were associated with the initiation of GH pulses. Icv injections of NPY, galanin and ghrelin stimulated GHRH release without affecting SRIF release. In addition, NPY suppressed, and galanin and ghrelin induced large GH pulses, although ghrelin was much more effective than galanin. These results suggest that an hourly fall in SRIF is involved in generating intrinsic circhoral rhythm of GH pulsatility. The mechanisms underlying the generation of large GH pulses of 5 hr periodicity remain unknown, while direct action of NPY and/or ghrelin on the pituitary might be involved.  相似文献

We tested the hypothesis that recombinant ovine leptin would attenuate the acute effects of neuropeptide Y (NPY) on secretion of GH and gonadotropins (LH and FSH) in cows. Ovariectomized cows (n=6) fitted with third ventricle guide cannulas were assigned randomly to each of three groups in a Latin square arrangement: (1) control; saline treatment only, (2) NPY; saline followed by NPY, and (3) L-NPY; leptin pretreatment followed by NPY. Treatments were: s.c. injection of saline or leptin (30 microg/kg BW) at time 0, i.v. injection of saline or leptin (30 microg/kg BW) at 70 min, and intracerebroventricular (i.c.v.) injection of saline or NPY (500 microg) at 90 min. Plasma leptin was elevated (P<0.01) at least four-fold throughout the experiment in the L-NPY group. Mean plasma concentrations of LH declined within 1 h and were lower (P<0.03) than controls in both the NPY and L-NPY groups beginning 2 h after NPY injection. An acute increase in plasma concentrations of GH was observed within 1 h after NPY in the NPY group and mean values were greater (P<0.01) than controls. However, in the L-NPY group, leptin pretreatment attenuated the NPY effect on GH. Treatments had no effect on FSH secretion. Results confirm suppressive and stimulatory effects of NPY on LH and GH secretion, respectively, and indicate that leptin can attenuate the acute effects of NPY on GH secretion in cattle.  相似文献

Estradiol increases basal growth hormone (GH) concentrations in sheep and cattle. This study sought to determine the effects of estradiol on GH-releasing hormone (GRH)-stimulated GH release in sheep. Growth hormone secretory characteristics, the GH response to GRH, and steady-state GH mRNA concentrations were determined in castrated male lambs treated with 2 different doses of estradiol 17-β for a 28-d experimental period. Although no differences between treatments in mean GH, basal GH, or GH pulse number were observed after 28 d of estradiol treatment, GH pulse amplitude was greater (P < 0.05) in the 2.00-cm implant-treated animals than in the control and 0.75-cm implant group. The effect of estradiol treatment on GRH-stimulated GH release revealed differences between the control and estradiol-treated animals (P < 0.05). The 15-min GH responses to 0.075 μg/kg hGRH in the control, 0.75-cm, and 2.00-cm implant groups, respectively, were 76 ± 10, 22.6 ± 2.1, and 43.6 ± 15.0 ng/mL. Growth hormone mRNA content was determined for pituitary glands from the different treatment groups, and no differences in steady-state GH mRNA levels were observed. There were no differences in the mean plasma concentrations of IGF-I, cortisol, T3, or T4 from weekly samples. Growth hormone release from cultured ovine pituitary cells from control sheep was not affected by estradiol after 72 h or in a subsequent 3-h incubation with estradiol combined with GRH. These data suggest that estradiol has differing actions on basal and GRH-stimulated GH concentrations in plasma, but the increase in pulse amplitude does not represent an increased pituitary sensitivity to GRH.  相似文献

The present study was undertaken to examine whether beta-adrenergic blockade with propranolol might influence and make less variable the growth hormone (GH) response to exogenous GH releasing hormone (GHRH) 1-44 in the dog. On four separate occasions eight healthy beagles, one to two years old, randomly received either propranolol (40 micrograms kg-1 intravenously) or an equivalent volume of saline, 30 minutes before either GHRH 1-44 (1 microgram kg-1 intravenously) or vehicle was injected. After propranolol alone, GH secretion did not differ from saline (area under the curve [AUC]: 649.5 +/- 128.3 v 633.2 +/- 87.7 ng min ml-1, respectively). GHRH alone elicited a significant increase in GH secretion (AUC: 1230.5 +/- 210.5 ng min ml-1) with a peak concentration of 16.7 +/- 4.8 ng ml-1. When GHRH was injected after propranolol the mean peak (59.1 +/- 14.7 ng ml-1) and secretory area (AUC: 2631.0 +/- 474.4 ng min ml-1) were greater than those observed after GHRH alone. However, from a clinical point of view propranolol pretreatment does not modify the great individual variability of the GH response to GHRH.  相似文献

This study was designed to examine the effects of the proportion of concentrate in the diet on the secretion of growth hormone (GH), insulin and insulin‐like growth factor‐I (IGF‐I) secretion and the GH‐releasing hormone (GHRH)‐induced GH response in adult sheep fed once daily. Dietary treatments were roughage and concentrate at ratios of 100:0 (0% concentrate diet), 60:40 (40% concentrate diet), and 20:80 (80% concentrate diet) on a dry matter basis. Mean plasma concentrations of GH before daily feeding (10.00–14.00 hours) were 11.4 ± 0.4, 10.1 ± 0.5 and 7.5 ± 0.3 ng/mL on the 0, 40 and 80% concentrate diet treatments, respectively. A significant decrease in plasma GH concentration was observed after daily feeding of any of the dietary treatments and these decreased levels were maintained for 8 h (0%), 12 h (40%) and 12 h (80%), respectively (P < 0.05). Plasma IGF‐I concentrations were significantly decreased 8–12 h and 4–16 h after the end of feeding compared with the prefeeding level in the 40 and 80% concentrate diet treatments, respectively (P < 0.05). GHRH injection brought an abrupt increase in the plasma GH concentrations, reaching a peak 10 min after each injection, but, after the meal, the peak plasma GH values for animals fed 40% (P < 0.05) and 80% (P < 0.01) concentrate diet were lower than that for roughage fed animals. The concentrate content of a diet affects the anterior pituitary function of sheep resulting in reduced baseline concentrations of GH and prolonged GH reduction after feeding once daily.  相似文献

The objective of this study was to determine the effects of dietary supplementation with chitooligosaccharide (COS) on growth performance, nutrient digestibility, blood characteristics and immune response in lipopolysaccharide-challenged weanling pigs. A total of 90 crossbred weanling pigs (5.44 ± 0.50 kg BW) were employed in Exp. 1. The three dietary treatments were basal diets supplemented with 0, 2.5, and 5 g COS/kg, and fed for 28 d. Each treatment had 6 replications with 5 pigs per pen. Increasing the level of supplemental COS tended to linearly (P < 0.10) improve ADG and ADFI during phase 2 and overall period, while there were no differences in G:F. The linear improvement in the apparent DM (P < 0.05) and N (P < 0.10) digestibility in pigs fed COS supplemented diets was noticed. The tested blood characteristics were not influenced under non-challenge conditions. In Exp. 2, a total of 20 pigs (5.22 ± 0.31 kg BW) were initially assigned to two dietary treatments and fed basal diets supplemented with 0 or 0.5 g COS/kg for 28 d. At the end of d 28, half of the pigs in each treatment (n = 5) were injected i.p. with Escherichia coli lipopolysaccharide at a concentration of 100 μg/kg of BW. The other half of the pigs in each treatment were injected with sterile saline solution at a concentration of 100 μg/kg of BW. This arrangement resulted in a 2 × 2 factorial design with diet and LPS challenge as the main effects. Blood sample and rectal temperature data were collected at 0, 2, 4 and 12 h post-challenge. Rectal temperatures increased as the result of LPS injection at 4 and 12 h post-challenge (P < 0.05). Serum cortisol, IGF-1, and TNF-α concentration were also increased as the result of LPS challenge (P < 0.05). The COS treatments resulted in lower cortisol concentrations at 2 h and higher IGF-1 concentrations at 4 h post-challenge (P < 0.05). COS and LPS interactions were also observed on cortisol and IGF-1 when the COS effects were presented (P < 0.05). Haptoglobin concentrations remained unaffected throughout the challenge period. White blood cell counts were increased in the LPS-treated pigs at 2 and 4 h post-challenge (P < 0.01). Lymphocyte count was elevated at 2 h and reduced at 12 h post-challenge as the result of LPS challenge (P < 0.05). However, there were no COS main effects observed on lymphocyte count throughout the challenge period. The comparison between two LPS challenged treatments also indicated that COS treatment has beneficial effects on rectal temperature, cortisol and IGF-1 concentrations. In conclusion, dietary supplementation with COS had little effect on nutrient digestibility and inflammatory stress markers in weanling pigs.  相似文献

The aim of the present study was to clarify the effects of hypothalamic dopamine (DA) on the secretion of growth hormone (GH) in goats. The GH‐releasing response to an intravenous (i.v.) injection of GH‐releasing hormone (GHRH, 0.25 μg/kg body weight (BW)) was examined after treatments to augment central DA using carbidopa (carbi, 1 mg/kg BW) and L‐dopa (1 mg/kg BW) in male and female goats under a 16‐h photoperiod (16 h light, 8 h dark) condition. GHRH significantly and rapidly stimulated the release of GH after its i.v. administration to goats (P < 0.05). The carbi and L‐dopa treatments completely suppressed GH‐releasing responses to GHRH in both male and female goats (P < 0.05). The prolactin (PRL)‐releasing response to an i.v. injection of thyrotropin‐releasing hormone (TRH, 1 μg/kg BW) was additionally examined in male goats in this study to confirm modifications to central DA concentrations. The treatments with carbi and L‐dopa significantly reduced TRH‐induced PRL release in goats (P < 0.05). These results demonstrated that hypothalamic DA was involved in the regulatory mechanisms of GH, as well as PRL secretion in goats.  相似文献

The aim of the present study was to clarify the effect of photoperiod on the secretion of growth hormone (GH) in goats. Adult female goats were kept at 20°C with an 8‐h or 16‐h photoperiod, and secretory patterns of GH for 4 h (12.00 to 16.00 hours) were compared. In addition, the goats were kept under a 16‐h photoperiod and orally administered saline (controls) or melatonin, and the effects of melatonin on the secretion of GH were examined. GH was secreted in a pulsatile manner. There were no significant differences in pulse frequency between the 8‐ and 16‐h photoperiods; however, GH pulse amplitude tended to be greater in the group with the 16‐h photoperiod (P = 0.1), and mean GH concentrations were significantly greater in the 16‐h photoperiod (P < 0.05). The GH‐releasing response to GH‐releasing hormone (GHRH) was also significantly greater for the 16‐h photoperiod (P < 0.05). There were no significant differences in GH pulse frequency between the saline‐ and melatonin‐treated groups. However, GH pulse amplitude and mean GH concentrations were significantly greater in the saline‐treated group (P < 0.05). The present results show that a long photoperiod enhances the secretion of GH, and melatonin modifies GH secretion in female goats.  相似文献

This study investigated the effects of dietary δ-aminolevulinic acid (ALA) on growth performance, nutrient digestibility, blood parameters and whether ALA improved the immune response of weanling pigs challenged with Escherichia coli lipopolysaccharide (LPS). Eighty pigs (body weight = 7.21 ± 0.51 kg) were allotted to four dietary treatments, with four pens per treatment and five pigs per pen. Basal diets were supplemented with 0, 5, 10, and 15 mg/kg ALA (as-fed basis) and fed for 35 days. At the end of the feeding period, 10 pigs were selected from both the 0- and 10-mg/kg ALA treatment groups; five were injected i.p. with LPS (50 μg/kg BW) and the other five pigs with an equivalent amount of sterile saline, resulting a 2 × 2 factorial arrangement. Blood sample and rectal temperature data were collected at 0, 2, 4 and 12 h after challenge. Growth performance was not affected by dietary treatments over the total experimental period. However, dry matter (DM) and nitrogen (N) digestibility was improved in the 15-mg/kg ALA treatment group at day 35 (P < 0.05). Serum hemoglobin (Hb) and iron levels were also increased, with the 10-mg/kg ALA treatment showing the highest concentration (P < 0.05). On day 35, red (RBC) and white blood cell (WBC) counts were elevated, with the 5- and 10-mg/kg ALA treatments having the highest counts (P < 0.05). During challenge, LPS injection elevated rectal temperature at 2 and 4 h postchallenge (P < 0.05). Plasma cortisol concentration was also increased by LPS injection at 2 and 4 h postchallenge and an ALA-alleviating effect was evident at 2 h postchallenge (P < 0.01). Concentration of plasma insulin-like growth factor-I (IGF-I) was increased in the ALA-supplemented treatments at 2 h postchallenge (P < 0.05). LPS injection increased plasma tumor necrosis factor-α (TNF-α) concentrations at 2, 4 and 12 h (P < 0.01), while an ALA-alleviating effect was observed at 2 and 4 h postchallenge (P < 0.05 and P < 0.10, respectively). Challenge with LPS decreased WBC counts at 2 and 4 h postchallenge (P < 0.01). At 12 h postchallenge, RBC, WBC and lymphocyte counts were affected by LPS challenge, while an ALA effect was only observed on WBC count (P < 0.05). In conclusion, dietary supplementation of ALA in weanling pigs can improve DM and N digestibilities, and iron status and have a beneficial effect on the immune response during inflammatory challenge.  相似文献

This study investigated the physical state and metabolic status of lambs of different birth ranks soon after birth. The aim was to identify the factors that result in poorer survival rates in triplet than single or twin lambs. Six hundred and twenty one Romney ewes were mated over a 4-day period and 46 of them were observed continually from day 143 of pregnancy until they finished lambing. Blood samples, taken from all lambs within 5 min of birth, were placed on ice, centrifuged and the plasma harvested and subsequently analysed for glucose, fructose, lactate, thyroxine (T4) and gamma-glutamyl transferase (GGT). The rectal temperature of each lamb was recorded at birth and at 1, 2, 3 and 6 h of life. Lambs were weighed and measured at 3 h of age.At birth, triplet lambs were significantly lighter (P < 0.05) and had lower (P < 0.05) plasma fructose and thyroxine than twin and single lambs, higher (P < 0.05) lactate concentrations and a lower rectal temperature (P < 0.05) than twin lambs. These observations suggest that they are subject to placental insufficiency. The packed cell volume was significantly (P < 0.001) higher in twin than triplet lambs. The order at birth of twin or triplet lambs had no effect on birthweight or plasma metabolite concentrations. The smallest triplet was significantly lighter (P < 0.05) and had higher plasma lactate (P < 0.05) concentration than the largest triplet.These physiological limitations found in smaller triplet lambs at birth leave them prone to perinatal mortality. Management techniques, to increase triplet lamb birthweight and thereby improve their survival outcome need further investigation.  相似文献

Sixty-eight multiparous grazing dairy cows were randomly allocated to two precalving pasture allowances to achieve differential dry matter intakes (DMI) for 29 ± 7.7 d precalving (Low or High DMI; 4.8 or 11.9 kg DM). At calving, cows within each precalving treatment were randomly allocated to one of two levels of feeding (Low or High DMI; 8.6 or 13.5 kg DM) for 35 d postcalving in a completely randomized design and a 2 × 2 factorial arrangement of treatments. Following treatments all cows grazed together and were fed pasture and pasture silage. Daily milk yields were recorded, and fat, protein and lactose concentrations determined weekly for 15 weeks. Blood was sampled weekly pre- and postcalving and analysed for indicators of energy status, growth hormone (GH) and insulin-like growth factor-1 (IGF-1). Body condition score (BCS) at calving was reduced by precalving restriction (P < 0.001; 2.8 and 3.0 for precalving Low and High treatment groups, respectively). Precalving restriction reduced milk fat production by 8.4% during the first 5 weeks postcalving, but differences were not significant subsequently. Postcalving feed restriction reduced yield of fat corrected milk (FCM), fat and protein by 23, 21 and 28%, respectively, during the first 5 weeks postcalving. Decreased (P < 0.05) yields of FCM, fat and protein (11, 10 and 9%, respectively) were also evident for 10 weeks after the feed restriction finished. There was a tendency (P < 0.1) for a precalving × postcalving DMI interaction in milk protein yield during the first 5 weeks of lactation with no effect of precalving level of feeding in cows that were restricted postcalving. The plasma concentrations of non-esterified fatty acids (NEFA), β-hydroxybutyrate (BOH) and GH were elevated in restricted cows precalving, and IGF-1 concentration was reduced. Plasma NEFA and BOH concentrations were elevated postcalving in restricted cows, but no effect of postcalving DMI on GH or IGF-1 concentration was evident. Results suggest that under grazing systems milk production responses to precalving DMI are small, but energy restrictions in early lactations result in significant carryover effects in milk production.  相似文献

The aim of this study was to examine whether increased frequency of luteinizing hormone (LH) pulses influences luteal progesterone (P4) secretion by measuring progesterone concentrations at the secreted (caudal vena cava) and circulating levels (jugular vein) in lactating dairy cows. Cows received six intravenous administrations of 2.5 μg of GnRH (gonadorelin acetate, n=4) or 2 ml saline (n=3) at 1-h intervals on 12.4 ± 0.4 (mean ± SE) days after ovulation. Blood samples were collected from the caudal vena cava and jugular vein every 12 min for 12 h (6 h before and after treatment). During the 6 h after treatment, frequency of LH pulses (5.3 ± 0.3 and 3.0 ± 0.0 pulses/6 h) and mean LH concentration (0.50 ± 0.06 and 0.38 ± 0.05 ng/ml) were greater (P<0.05) in GnRH-treated cows than in saline-treated cows. Mean P4 concentration and amplitude of P4 pulses in the caudal vena cava during the 6 h after treatment were greater (P<0.05) in GnRH-treated cows than in saline-treated cows, but the frequency of P4 pulses was not different between the groups. Mean P4 concentration in the jugular vein during the 6 h after treatment was also higher (P<0.05) in GnRH-treated cows than in saline-treated cows (7.0 ± 1.3 and 5.4 ± 0.9 ng/ml). These results indicate that the increased frequency of LH pulses stimulates progesterone secretion from the functional corpus luteum and brings about higher P4 concentrations in the circulating blood in lactating dairy cows.  相似文献

Serum hormones and metabolites of lean and obese gilts were determined. Fifteen Iberian and fifteen Landrace gilts, of approximately 20 kg body weight, were fed isoenergetic (13.6 kJ ME/g DM, semisynthetic diets either with equilibrated amino acid profile at two crude protein (CP) levels (12% (0.81% lysine (Lys)) and 16% CP (1.08% Lys) as-fed basis) or lysine deficient (12% (0.30% Lys) or 16% CP (0.36% Lys) as-fed basis for Iberian and Landrace, respectively). Lysine deficient diets were offered only at the optimal protein level for maximum protein accretion for each breed. Each dietary treatment was assayed in 5 animals. Gilts were allocated in metabolic cages at 21 ± 1.5 °C with free access to water and fed four times daily at 90% ad libitum during 10 days. On day 11th blood samples were taken 5–5.5 h postprandial and serum obtained and frozen at − 20 °C. Growth hormone (GH), insulin like growth factor I (IGF-1), insulin, leptin, glucose, urea, creatinine, cholesterol and triglycerides were determined. Landrace gilts had higher serum glucose (P < 0.05) and creatinine (P < 0.001) than Iberian while no differences (P = 0.122–0.494) were found for the rest of the biochemical variables. Higher serum levels of insulin, IGF-1 and leptin (60, P < 0.05; 79, P < 0.05 and 189%, P < 0.001, respectively) and no differences in GH (P = 0.512) were encountered in Iberian gilts compared to Landrace. Dietary crude protein did not alter the serum hormonal profile (P = 0.110–0.454). Serum cholesterol (total, P < 0.01; HDL, P < 0.05; LDL, P < 0.05) decreased and triglycerides and urea increased (P < 0.05 and P < 0.001, respectively) as dietary crude protein increased. When lysine deficient diets were used, serum glucose decreased (P < 0.05) and urea increased (P < 0.001) compared to equilibrated diets. Serum insulin (P = 0.052), IGF-1 (P < 0.05) and leptin (P = 0.051) concentration decreased when lysine deficient diets were fed to the animals while GH remained unaffected (P = 0.214).These data suggest that Iberian and Landrace growing gilts have distinct hormone and metabolite serum profiles which are altered by a severe dietary lysine deficiency.  相似文献

Insulin-like growth factor-I (IGF-I), growth hormone (GH), and prolactin (PRL) play important roles in milk protein synthesis, and their plasma concentrations were reported to be affected by dietary protein intake. To investigate the relationship between circulating amino acid (AA) and concentrations of these hormones, 18 Wistar rats aged 14 wk were assigned to a low (LP; 9% protein), standard (SP; 21% protein), or high-protein (HP; 35% protein) diet from parturition through day 15 of lactation. Plasma, liver, pituitary gland, skeletal muscle, and mammary gland samples were collected at the end of treatment. Circulating and hepatic IGF-I concentrations increased linearly with elevated dietary protein concentrations (P < 0.0001). Rats receiving the HP diet had higher circulating GH (P < 0.01) and pituitary PRL concentrations (P < 0.0001) but lower pituitary GH concentration (P < 0.0001) relative to those in rats receiving the LP and SP diets. Pearson correlation test performed on composed data across treatments showed that several circulating AAs were correlated with circulating and tissue concentrations of IGF-I, GH, and PRL. Multiple linear regression analyses identified Leu, Gln, Ala, Gly, and Arg as the main AAs associated with hormone responses (R2 = 0.37 ~ 0.80; P < 0.05). Rats fed the LP and HP diets had greater Igf1 and Ghr gene expression in skeletal muscle than those fed the SP diets (P < 0.01). However, LP treatment decreased Prlr mRNA abundance in mammary glands as compared with the SP and HP treatments (P < 0.05). The HP diets increased AA transporter expression (P < 0.01) but decreased mammalian target of rapamycin (P < 0.05) and 70 kDa ribosomal protein S6 kinase 1 (P < 0.01) phosphorylation in mammary glands as compared with the LP and SP diets. The results of the present study suggested that several circulating AAs mediated the effects of dietary protein supply on concentrations of IGF-I, GH, and PRL, which in turn altered the metabolism status in peripheral tissues including the lactating mammary glands.  相似文献

Eighty male pigs from 20 litters were used to evaluate dietary addition of 250 mg/kg of Zn from zinc amino acid complex (ZnAA) to sows during the last trimester of gestation and gastric intubation of 40 mg Zn from soluble zinc methionine (ZnMet) to suckling pigs at birth and on day 7 and 14 (weaning) on small intestinal morphology, Zn status and bacterial translocation in early-weaned pigs. At weaning, pigs were challenged with an intramuscular injection of saline without or with 120 μg/kg BW of lipopolysaccharide (LPS; from Escherichia coli O26:B6) and were euthanized 24 h later prior to collection of intestinal lymph nodes and small intestinal sections. Zinc concentration in serum 7 days after birth and at weaning were higher in pigs from ZnAA-supplemented sows and those receiving gastric intubation with ZnMet (P = 0.05 and P < 0.0001, respectively). Post-weaning liver tissue concentrations for Zn (P < 0.0001) and Fe (P = 0.04) were higher and for Cu lower (P < 0.0001) in pigs intubated with ZnMet. Pigs from ZnAA-supplemented compared with control-fed sows tended (P < 0.1) to have increased villi height and villus:crypt ratio in the jejunum and higher (P = 0.1) goblet cell counts in the ileum. Goblet cell counts of ZnMet-intubated (P = 0.03) and LPS-challenged pigs (P = 0.05) were also higher in the jejunum. Supplementation of ZnAA to gestating sows increased (P = 0.04) E. coli colony forming unit counts in the small intestinal mesenteric lymph nodes of early-weaned pigs. In conclusion, dietary addition of ZnAA to gestating sows and gastric intubation of ZnMet improved Zn status of suckling pigs.  相似文献

The objective of this study was to determine the effects of dietary levels of crude protein (CP) and levels of fermentable carbohydrates (FC) and their interaction on odour emission, odour intensity, odour hedonic tone, and ammonia emission from pig manure, and manure characteristics. An experiment was conducted with finishing pigs (n = 36) in a 2 × 3 factorial randomized complete block arrangement with 6 treatment combinations in 6 blocks. There were 2 dietary CP levels (low 12%; high 18%) and 3 digestible FC levels: (low 95.5; medium 145.5; and high 195.5 g/kg feed, as-fed basis). Pigs with an initial body weight (BW) of 57.7 ± 2.5 kg were penned individually in partly slatted floor pens. Faeces and urine of each pig accumulated in separate manure pits under the slatted floor. In the 6th week of the collection period air samples were collected directly above the manure in each pit. Manure samples were taken for manure characteristics. Air samples were analyzed for odour concentration and for hedonic tone and odour intensity. Manure samples were analyzed for volatile fatty acids (VFA), indoles, phenoles, sulphurous compounds, ammonium, and total N concentrations. Dietary CP level and FC level did not affect odour emission, odour intensity and hedonic tone but their interaction affected odour emission at P = 0.06. At a high dietary CP level, increased FC level decreased odour emission, while at a low CP level, increased FC level increased odour emission from pig manure. Total N and ammonium concentrations, and ammonia emission from pig manure were reduced at low dietary CP level (P < 0.001). High FC level led to low ammonia emission from pig manure (P = 0.01). Manure pH increased at high dietary CP level (P < 0.001) and decreased when FC level increased (P < 0.05). Total VFA concentration increased at high dietary CP level (P < 0.001) and when FC level increased (P < 0.001). Enhanced dietary CP increased the manure concentrations of phenol (P < 0.001), cresols (P = 0.01), indole (P < 0.001), 4-ethylphenol (P < 0.001) and carbon disulfide (P < 0.001), but FC did not affect concentrations of these compounds (P > 0.05) in the manure. We conclude that the interaction between dietary CP and FC plays a role in odour production and emission. Ammonia emission from pig manure can be reduced substantially by decreasing dietary CP and by increasing FC.  相似文献

Four growing pigs (initial liveweight 25.9 ± 0.54 kg, final liveweight 43.0 ± 1.06 kg) were used to study the effect of dietary lysine level on nutrient digestibility, whole-body protein turnover, plasma insulin-like growth factor-I (IGF-I), growth hormone (GH), insulin, glucose, and urea nitrogen (PUN). Four diets, containing 7.0 g (L1), 9.5 g (L2), 12.0 g (L3) and 14.5 g (L4) lysine per kg diet respectively, were formulated as experimental treatments. The animals and diets were allocated in a 4 × 4 Latin square design. Nitrogen (N) metabolism and whole-body protein turnover were measured by classical method and single-dose 15N end-product method, respectively. The blood samples were taken at the end of each experimental period. Results showed that N retention (NR) and N biological value (NBV) were significantly increased from L1 to L4 (P < 0.05). However, differences in NR and NBV between L2, L3 and L4 were not significant (P > 0.05). There was no significant difference on dry matter (DM) digestibility, organic matter (OM) digestibility and N digestibility between different treatments (P > 0.05). Whole-body protein synthesis, protein degradation and protein accretion increased markedly from L1 to L2 (P < 0.05), but did not increase further from L2 to L4. Whole-body protein accretion (y, g/kg W0.75/d) increased with dietary lysine (x, g/kg) in a quadratic manner: y = − 0.09x2 + 2.12x − 5.14 (r2 = 0.96, n = 4, P < 0.05).The results also showed that differences in plasma IGF-I, GH, glucose and PUN concentration between different treatments were not significant (P > 0.05). Plasma insulin concentration (y, μIU/ml) was increased with dietary lysine (x, g/kg) in a quadratic manner: y = 0.23x2 − 4.10x + 32.25 (r2 = 0.99, n = 4, P < 0.05), but it was not found that plasma insulin concentration was related to NR. A significant correlation was found between NR (y, g/d) and plasma IGF-I (x, ng/ml): y = − 3.1 × 10− 3x2 + 1.31x − 122.28 (r2 = 0.99, n = 4, P < 0.05).It was concluded that dietary lysine level had a significant influence on NR and whole-body protein turnover but not on plasma IGF-I and GH concentration. Plasma IGF-I may be an important factor controlling N metabolism of growing pigs. Further research was needed to study the mechanism.  相似文献

The goal of this study was to assess if oxidative stress, as measured by alterations in the concentrations of antioxidant enzymes in the liver and erythrocytes of cattle, could be induced following dl-ethionine administration. Whole blood, serum and liver biopsy samples were collected 0, 4, 7 and 10 days after intra-peritoneal ethionine administration to five cows. The activities of the antioxidant enzymes copper zinc superoxide dismutase (Cu, Zn SOD) and catalase were assessed in the liver biopsies which were also examined histopathologically.Significant increases in hepatic Cu, Zn SOD concentrations (P < 0.01) were noted on days 7 and 10 post-treatment. Hepatic catalase activity decreased significantly (P < 0.01) on days 4, 7 and 10 post-treatment and erythrocyte Cu, Zn SOD activity was significantly increased on day 10. Serum biochemical analysis revealed a significant increase (P < 0.01) in non-esterified fatty acid concentrations on day 4 and significant decreases in total cholesterol and phospholipid levels on days 4 (P < 0.05), 7 (P < 0.01) and 10 (P < 0.01). In this model system, dl-ethionine administration was effective in inducing oxidative stress particularly reflected in the liver.  相似文献
