本研究探讨了Aldh3a2a在斑马鱼色素细胞发育中的意义及其对醛代谢和肝脏健康的影响。利用CRISPR/Cas9技术,我们成功敲除斑马鱼体内的aldh3a2a,导致黑色素细胞、黄色素细胞和虹彩细胞合成数量减少。转录组分析揭示了基因表达谱的变化,特别是与钾通道和类固醇生物合成途径相关的基因表达谱。我们的研究结果表明,aldh3a2a和aldh1a2之间存在调控关系,对视黄酸受体信号传导有影响。GO和KEGG富集分析强调了对色素细胞发育以外的细胞过程的更广泛影响。此外,aldh3a2a基因敲除会导致醛积累,导致肝脏异常,类似于Sj?gren-Larsson综合征,本研究揭示了aldh3a2a在斑马鱼中的多方面作用及其与人类遗传疾病的相关性。 相似文献

Size determination represents a fundamental requirement for multicomponent biological structures. Some pathogenic bacteria possess a weapon derived from the flagellum. Like the flagellum, this type-III secretion apparatus, called the injectisome, has a transmembrane basal body, but the external component is a needle-like structure instead of a hook and a filament. A batch of eggs was exposed to a patterned sound continuously from day 12 to day 18 of incubation, while a control group was hatched in the quiet. In a postnatal test all chicks tended to creep toward a stationary sound source, but the experimental group showed a preference for the sound heard during incubation. 使用一种新的屈曲路径跟踪策略,对拱在平面内的弹塑性极值点屈曲和二次分岔屈曲全过程进行跟踪分析,得到全跨均布荷载作用下材料、截面相同而矢跨比不同的拱的弹塑性极值点屈曲荷载和二次分岔屈曲荷载,以及半跨均布荷载作用下其极值点屈曲荷载.研究结果表明：对于弹塑性拱结构,在全跨均布荷载作用下,二次分岔屈曲总是最危险的屈曲形式,其发生必定先于极值点屈曲.对于材料、截面相同,矢跨比0.1～0.5的拱,半跨均布荷载作用下矢跨比0.23的拱极值点屈曲极限承载力最大;全跨均布荷载作用下,矢跨比0.1的拱极值点屈曲和二次分岔屈曲极限承载力均大于其他拱.将得到的全跨和半跨均布荷载作用下不同长细比、不同矢跨比拱的弹塑性极限承载力计算结果归纳总结,得到极限承载力简化计算公式,可以直接查用,便于工程设计中使用. 相似文献

Lipoprotein(a) [Lp(a)] is a macromolecular complex found in human plasma that combines structural elements from the lipoprotein and blood clotting systems and that is associated with premature coronary heart disease and stroke. It is assembled from low-density lipoprotein (LDL) and a large hydrophilic glycoprotein called apolipoprotein(a) [apo(a)], which is homologous to the protease zymogen plasminogen. Plasma Lp(a) concentrations vary 1000-fold between individuals and represent a continuous quantitative genetic trait with a skewed distribution in Caucasian populations. Variation in the hypervariable apo(a) gene on chromosome 6q2.6-q2.7 and interaction of apo(a) alleles with defective LDL-receptor genes explain a large fraction of the variability of plasma Lp(a) concentrations. Though of high theoretical and practical interest, many aspects of the metabolism, function, evolution, and regulation of plasma concentrations of Lp(a) are presently unknown, controversial, or mysterious. 相似文献

The complement cleavage product C5a is a potent stimulant of inflammatory processes; thus, inhibition of C5a activity is of therapeutic interest. The three-dimensional structure of the major portion of C5a was modeled from the homologous C3a crystal structure by comparative modeling techniques. The model shows that core residues of C5a are completely conserved, while external residues differ from C3a. Even though the amino-terminal 12 residues of C3a are disordered in the crystal, this sequence in C5a may form an amphipathic helix. The distribution of species sequence differences in the complete C5a structure suggests a possible receptor binding site. 相似文献

beta-Glucuronidase catalyzes the hydrolysis of benzo[a]pyrene-3-glucuronide to 3-hydroxybenzo[a]pyrene. During the enzymatic hydrolysis, a benzo[a]pyrene derivative is formed which binds to DNA to a far greater extent than either the 3-hydroxybenzo[a]pyrene or its glucuronide. These results suggest that conjugates of benzo(a)pyrene may be converted by beta-glucuronidase at intracellular and organ sites distal to the initial sites of oxygenation and conjugation of benzo(a)pyrene to activated intermediates that are possibly carcinogenic. 相似文献

为全面了解杨树-农作物间作系统的固碳效能,研究了沙质平原区3、9、13、17a杨农(杨树与农作物)间作系统土壤有机碳库特征。结果表明,杨农间作系统土壤有机碳含量随着土层深度增加而逐步下降,随着间作年限增加而增加;0～60cm土壤有机碳密度随着间作年限的增加而增加,间作3、9、13、17a分别达到11.25、11.19、13.71、16.17t/hm2;土壤碳密度随着土层深度增加而下降,但随着间作年限增加,40～60cm土壤碳密度所占比例逐步增加,到间作17a时,已达到28.76%,说明随着间作年限增加可以增加深层土壤固碳能力;土壤有机碳固存速率随着间作年限增加而增加,间作3～9a为0.145 0t/(hm2·a),间作9～13a为0.397 5t/(hm2·a),间作13～17a达到最高,为0.615 0t/(hm2·a),平均为0.351 4t/(hm2·a),说明豫东平原杨农间作系统要达到一定年限,才能充分发挥其土壤固碳效能。 相似文献
