A study was conducted with 20 weaned barrows (14 d, 4.98 +/- 0.21 kg) to determine the effect of feeding spray-dried plasma (SDP) after weaning on the pig's stress response to a lipopolysaccharide (LPS) challenge. After weaning, pigs were fed a diet containing 0 or 7% SDP for 7 d. On d 6 after weaning, all pigs were nonsurgically fitted with a jugular catheter. On d 7 after weaning, the pigs were given i.p. injections of either saline or LPS (150 microg/kg BW) followed by serial blood collection every 15 min for a 3-h period. Following the 3-h blood collection, all pigs were killed and tissue was collected for mRNA analysis. Pig weight on d 7 after weaning was not affected by dietary treatment (P > 0.21). Pigs fed the diet with SDP had lower (P < 0.05) levels of hypothalamic corticotropin-releasing hormone (CRH) mRNA, pituitary gland CRH receptor mRNA, and adrenal gland adrenocorticotropin-releasing hormone (ACTH) receptor mRNA. Dietary treatment did not affect pituitary gland proopiomelanocortin (POMC) mRNA. No effect of LPS treatment was observed in any of the mRNA levels examined. For both serum ACTH and cortisol, there was a significant diet x LPS treatment interaction (P < 0.01) such that both the ACTH and cortisol responses to the LPS challenge were greater in the pigs fed the diet with SDP than in the pigs fed the diet without SDP. For pigs given the saline injection, diet did not affect basal serum cortisol concentration; however, basal serum ACTH concentration was lower in those pigs fed the diet with SDP (P < 0.0001). A diet x LPS treatment interaction (P < 0.024) was observed for adrenal gland mRNA expression for steroidogenic acute regulatory (StAR) protein such that the LPS-induced increase in StAR mRNA was greater in the pigs fed SDP than in pigs fed the diet without SDP. These results demonstrate that pigs fed a diet with SDP have an increased activation of the pituitary-adrenal axis following an LPS challenge compared to pigs fed a diet without SDP.  相似文献

Forty barrows (TR4 x C22) were weaned at 17 d of age (BW = 6.27 +/- 0.30 kg), housed (two pigs/pen) in a thermal-neutral environment (TN; constant 26.7 degrees C), and fed diets with or without 7% (as-fed basis) spray-dried plasma (SDP). On d 7, one pig/ pen was moved into a cold environment (CE; constant 15.6 degrees C). Pigs were fitted with jugular catheters on d 11. On d 12, 16 pigs per environment (eight pigs per dietary treatment) were challenged i.v. with 75 microg of lipopolysaccharide (LPS)/kg of BW. Blood samples were collected over a 4.5-h period. Pigs were then killed and tissue samples were harvested for messenger RNA (mRNA) analysis. From d 0 to 7, pigs fed SDP diets had a lower gain:feed ratio (G/F) than pigs fed no SDP (533 +/- 14 vs. 585 +/- 17 g/kg; P < 0.03). Pigs housed in the CE consumed more feed and had a lower G/F than pigs housed in TN from d 7 to 11 (P < 0.001). There were no environment x diet interactions from d 7 to 11 (P > 0.78). Baseline concentrations of serum ACTH and cortisol were lower in the TN pigs than in the CE pigs (P < 0.001). Pigs fed diets without SDP had lower serum cortisol concentrations over the 4.5-h period than pigs fed SDP (time x diet, P < 0.001). Serum concentrations of tumor necrosis factor-alpha (TNF-alpha) were highest for pigs consuming SDP in the CE, whereas there were no differences among the other treatments (time x diet x environment, P < 0.02). Pigs housed in the CE had higher serum interleukin-1beta (IL-1beta) (P < 0.001) and interleukin-6 (IL-6; P < 0.001) than TN pigs. Pigs fed SDP also had slightly higher serum IL-1beta concentrations (P < 0.10) and higher (P < 0.001) IL-6 concentrations than pigs fed no SDP. Pigs fed SDP had 9% lower liver and 13% lower thymus mRNA expression of tumor necrosis factor-alpha (TNF-alpha) than pigs that consumed no SDP (P < 0.06). Liver IL-1beta, IL-6, and LPS-binding protein mRNA were higher in the CE than in the TN (P < 0.03, P < 0.001, and P < 0.05; respectively). In addition, spleen TNF-alpha (P < 0.03) and IL-6 (P < 0.01) mRNA levels were higher in the CE than in the TN. Pigs consuming SDP and challenged with LPS responded with elevated serum concentrations of cortisol and cytokines compared with pigs fed diets with no SDP. Housing pigs in a CE increased the baseline concentrations of ACTH and cortisol, and when coupled with an LPS challenge, resulted in elevated serum and tissue mRNA levels of cytokines. Cold stress and feeding SDP during a LPS challenge may result in increased stress and immune responses in young pigs.  相似文献

A total of 180 weanling pigs (21 ± 3 d of age; 5.98 ± 0.04 kg) were used to investigate the effect of chito-oligosaccharide (COS) on growth performance, intestinal barrier function, intestinal morphology, and cecal microflora. Based on initial BW, gender and litter, the pigs were given 5 treatments during a 14-d feeding experiment, including a basal diet (control), 3 diets with COS supplementation (200, 400, or 600 mg/kg), and a diet with colistin sulfate (CSE) supplementation (20 mg/kg). Six randomly selected pigs from each treatment were used to collect serum, duodenal, jejunal, ileal, and cecal samples on d 7 and 14 postweaning. From d 1 to 7 postweaning, pigs fed COS or CSE had greater ADG and ADFI compared with the control pigs. From d 1 to 14, diets with either 400 or 600 mg/kg COS, or 20 mg/kg CSE increased (P < 0.05) ADG and G:F compared with the control diet. No significant differences were observed in ADG, ADFI, and G:F between the pigs fed COS and CSE. Pigs fed either 400 or 600 mg/kg COS, or 20 mg/kg CSE had less (P < 0.05) diamine oxidase (DAO) in the serum, but greater concentration of (P < 0.05) DAO in jejunal mucosa, than the control pigs on d 7 postweaning. Treatments did not affect villous height and crypt depth of the duodenum, jejunum, or ileum. Pigs fed COS at 400 mg/kg had greater (P < 0.05) concentration of Bifidobacteria and Lactobacilli in the cecum than pigs fed the control diet and CSE diet on d 7 postweaning. Supplementation of COS or CSE decreased (P < 0.05) the population of cecal Staphylococcus aureus compared with the control diet on d 7 postweaning. The number of cecal Bifidobacteria in pigs fed 600 mg/kg COS was greater (P < 0.05) than that of pigs fed the control diet or CSE diet on d 14 postweaning. No significant differences were observed in Escherichia coli counts in the cecum among treatments. The present results indicate that dietary supplementation of COS at 400 or 600 mg/kg promotes growth performance and improves gut barrier function, increases the population of Bifidobacteria and Lactobacilli, and decreases S. aureus in the cecum of weanling pigs.  相似文献

The objective of this growth trial was to determine the interrelationship between immunological criteria, gut morphology, and performance of starter pigs fed soybean proteins processed by different methods. One hundred twenty-five pigs were orally infused with 6 g/d of either dried skim milk, soybean meal (48% CP), soy protein concentrate, extruded soy protein concentrate, or experimental soy protein concentrate from 7 to 11 d of age and then fed a diet containing the same protein sources from weaning (d 21) to 35 d of age. All pigs were fed a corn-soybean meal diet containing 10% dried whey, 1.25% lysine, and 3% soybean oil for the remaining 21 d of the experiment. Xylose absorption and anti-soy immunoglobulin G (IgG) titers were measured on d 6 postweaning, and skin-fold thickness after intradermal injection of protein extracts was measured on d 7 postweaning. A total of 25 pigs (five pigs/treatment) was euthanatized on d 7 postweaning. Villus height and crypt depth from duodenum samples were measured. These measurements were obtained to elucidate a relationship between the hypersensitivity responses to soybean products and growth performance of baby pigs. Pigs fed diets containing soybean meal had a lower (P less than .05) rate of gain (d 0 to 14) and villus height, higher (P less than .01) serum anti-soy IgG titers, and increased skin-fold thickness (d 6 and 7 postweaning) after intradermal injection compared with those fed dried skim milk. Pigs fed other soy proteins also had lower ADG from d 0 to 14 postweaning; however, pigs fed moist extruded soy protein concentrate tended (P less than .09) to have higher ADG and improved feed utilization when compared with pigs fed soybean meal (d 0 to 14).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Seventy-two crossbred pigs (7.58 +/- 0.30 kg BW) weaned at 28 +/- 3 d of age were used to investigate the effects of fish oil supplementation on pig performance and on immunological, adrenal, and somatotropic responses following an Escherichia coli lipopolysaccharide (LPS) challenge in a 2 x 2 factorial design. The main factors consisted of diet (7% corn oil [CO] or 7% fish oil [FO]) and immunological challenge (LPS or saline). On d 14 and 21, pigs were injected intraperitoneally with either 200 microg/kg BW of LPS or an equivalent amount of sterile saline. Blood samples were collected 3 h after injection for analysis of interleukin-1beta (IL-1beta), prostaglandin E2 (PGE2), cortisol, growth hormone (GH), and insulin-like growth factor (IGF)-I. On d 2 after LPS challenge, peripheral blood lymphocyte proliferation (PBLP) was determined. Lipopolysaccharide challenge decreased ADG (487 vs. 586 g; P < 0.05) and ADFI (as-fed, 776 vs. 920 g; P < 0.05) from d 14 to 21 and ADG (587 vs. 652 g; P < 0.10) from d 21 to 28. Fish oil improved ADG (554 vs. 520 g; P < 0.10) and ADFI (891 vs. 805 g; P < 0.10) from d 14 to 21. On d 14, LPS challenge x diet interactions were observed for IL-1beta (P < 0.10), PGE2 (P < 0.001), and cortisol (P < 0.05) such that these measurements responded to the LPS challenge to a lesser extent (IL-1beta: 93 vs. 114 pg/mL, P < 0.05; PGE2: 536 vs. 1,285 pg/mL, P < 0.001; cortisol: 143 vs. 206 ng/mL, P < 0.05) in pigs receiving the FO diet than in pigs fed the CO diet. In contrast, among LPS-treated pigs, pigs fed the FO diet had higher IGF-I (155 vs. 101 ng/mL; P < 0.10) than those fed the CO diet. On d 21 among LPS-treated pigs, pigs fed FO had lower IL-1beta (70 vs. 84 pg/mL; P < 0.10) and cortisol (153 vs. 205 ng/mL; P < 0.05) than those fed CO. Pigs fed FO had lower PGE2 (331 vs. 444 pg/mL; P < 0.05) and higher IGF-I (202 vs. 171 ng/mL; P < 0.10) compared with those fed CO. Lipopolysaccharide challenge decreased GH (0.27 vs. 0.33 ng/mL; P < 0.05) on d 14, whereas it had no effect on GH on d 21. During both LPS challenge periods, the challenge increased PBLP when these cells were incubated with 8 (1.46 vs. 1.32; P < 0.10) or 16 microg/mL (1.46 vs. 1.30; P < 0.05) of concanavalin A. Fish oil had no effect on PBLP. These results suggest that FO alters the release of proinflammatory cytokines, which might lead to improved pig performance during an immunological challenge.  相似文献

The aim of the study was to investigate the effects of dietary linseed (rich in n-3 PUFA) on expression of inflammation-related genes and on growth performance of growing-finishing barrows. Two isoenergetic and isonitrogenous diets were formulated, one as the basal diet and the other containing 10% linseed. Twenty-four Landrace x Yorkshire barrows weighing 35 +/- 3.7 kg were randomly assigned to 1 of 4 treatment groups, with 6 pigs per group. During the entire experimental period of 90 d, these 4 groups of pigs were first fed the basal diet and then fed the linseed diet for 0, 30, 60, and 90 d before slaughter, respectively. Pig growth; messenger RNA (mRNA) expression of peroxisome proliferator-activated receptor-gamma (PPARgamma), IL-1beta0, IL-6, and tumor necrosis factor-alpha (TNF-alpha); and plasma concentrations of the 3 proinflammatory cytokines were measured and analyzed. Average daily feed intake did not differ among treatment groups (P > 0.05), but ADG (P < 0.05) and G:F (P < 0.01) responded quadratically to the duration of linseed diet feeding, and pigs in the 60-d treatment group had the greatest ADG and G:F. The mRNA expression of PPARgamma in loin muscle and spleen increased linearly (P < 0.01) with the duration of linseed diet feeding, whereas its expression in adipose tissue was not affected (P = 0.095). Tumor necrosis factor-alpha and IL-6 mRNA expression in muscle, adipose, and spleen, as well as serum concentration of TNF-alpha, decreased linearly (P < 0.01) with the duration of linseed diet feeding. Peroxisome proliferator-activated receptor-gamma mRNA abundance was negatively correlated with IL-1beta, IL-6, and TNF-alpha mRNA abundance both in muscle (R(2) = 0.63, P < 0.001) and in spleen (R(2) = 0.69, P < 0.001), and PPARgamma mRNA expression in spleen (R(2) = 0.59, P < 0.01) and muscle (R(2) = 0.52, P < 0.05) was negatively correlated with serum TNF-alpha concentration. There were also significant quadratic relations between ADG and expression of PPARgamma (P < 0.05) and splenic TNF-alpha (P < 0.05). These data suggest that intake of n-3 PUFA from the linseed diet led to significant decreases in the expression of proinflammatory cytokine genes, which may stimulate growth in growing-finishing barrows, at least in part, through a PPARgamma-dependent mechanism.  相似文献

To test possible dietary immune modulators, 32 crossbred male pigs were given 1 of 4 dietary treatments (8 pigs/treatment): control, Saccharomyces cerevisiae with beta-glucan (Energy Plus, Natural Chem Industries LTD, Houston, TX; 0.312 g/kg of BW, 2.5% of diet), vitamin C (Stay C 35, DSM Nutritional Products Inc., Prisippany, NJ; 75 ppm), or beta-glucan plus vitamin C together (combination; 0.312 g/kg of BW and 75 ppm, respectively). Supplements were given in whole milk within 36 h of birth and then daily for 2 wk until weaning, when the supplement was given in feed for an additional 2 wk. Growth was recorded during the 4 wk of supplement delivery. An i.v. lipopolysaccharide challenge (LPS; 150 microg/kg) was given 14 d postweaning at 0900. Behavior was observed, and blood samples were collected every 30 min for 4 h via a jugular catheter from -1 (0800) to 3 (1200) h relative to challenge (-60, -30, 0, 30, 60, 90, 120, 150, and 180 min), and tissues were collected after exsanguination. Beta-glucan (glucan and combination) increased (P < 0.05) BW and ADG compared with vitamin C and control. Cortisol concentrations showed an interaction (P < 0.05) of the beta-glucan and vitamin C. Intestinal expression of tumor-necrosis factor (TNF)-alpha mRNA was greatest for vitamin C and beta-glucan compared with control and combination, and liver TNF-alpha mRNA expression showed a main effect (P < 0.01) of beta-glucan. Lung expression of TNF-alpha mRNA exhibited a vitamin C effect (P < 0.01). In contrast, spleen had greater (P < 0.01) relative abundance of TNF-alpha mRNA in beta-glucan pigs. Intestinal expression of IL-1Ra mRNA was greater (P < 0.05) for vitamin C and beta-glucan treatments compared with the control and combination pigs. Liver expression of IL-1 receptor antagonist mRNA exhibited a vitamin C effect (P < 0.01). Lying and sleeping behaviors differed (P < 0.05) among treatments early in the observations (0700 to 0720), then sporadically until 50 min after the LPS injection. The vitamin C group slept less (P < 0.05) on those occasions. The time spent lying was least (P < 0.05) for the glucan and combination pigs immediately after the injection. These results show a complex interaction between vitamin C and this yeast product after LPS challenge, with differential expression in tissues by 2 h after LPS injections. The combination enhanced postweaning growth and reduced TNF-alpha expression of the intestinal and liver tissues, suggesting an important immunomodulatory role of the combination treatment.  相似文献

Four experiments were conducted to evaluate the nutrient contributions and physiological health benefits of spray-dried egg (SDE) containing only unfertilized eggs as a protein source in nursery pig diets. In all experiments, all diets were formulated to the same ME and Lys content, and each pen within a block (by BW) housed the same number of barrows and gilts. In Exp. 1 and 2 (168 and 140 pigs, respectively; 5 kg BW; 16 d old; 14 replicates/experiment), conducted at a university farm, treatments were with or without 5% SDE in a nursery control diet, which included antibiotics and zinc oxide. Pigs were fed for 10 d after weaning to measure ADG, ADFI, and G:F. The SDE increased (P < 0.05) ADG (Exp. 1: 243 vs. 204 g/d; Exp. 2: 204 vs. 181 g/d) and ADFI (Exp. 1: 236 vs. 204 g/d; Exp. 2: 263 vs. 253 g/d) compared with the control diet but did not affect G:F. In Exp. 3 (1,008 pigs; 5.2 kg BW; 20 d old; 12 replicates/treatment), conducted at a commercial farm, treatments were in a factorial arrangement of with or without SDE and high or low spray-dried plasma (SDP) in nursery diets, which included antibiotics and zinc oxide. Pigs were fed for 6 wk using a 4-phase feeding program (phases of 1, 1, 2, and 2 wk, respectively) with declining diet complexity to measure ADG, ADFI, G:F, removal rate (mortality plus morbidity), and frequency of medical treatments per pen and day (MED). The diets with the SDE increased (P < 0.05) ADFI during phase 1 only (180 vs. 164 g/d) compared with the diets without the SDE but did not affect growth performance during any other phases. The diets with SDE reduced MED during phase 1 (0.75% vs. 1.35%; P < 0.05) and the overall period (0.84% vs. 1.01%; P = 0.062) compared with the diets without the SDE but did not affect removal rate. In Exp. 4 (160 pigs; 6.7 kg BW; 21 d old; 10 replicates/treatment), conducted at a university farm to determine whether SDE can replace SDP, treatments were in a factorial arrangement of with or without SDP or SDE in nursery diets, which excluded antibiotics and zinc oxide. Pigs were fed for 6 wk using the same schedule used in Exp. 3 to measure ADG, ADFI, and G:F. The diets with SDE increased (P < 0.05) ADFI during phase 1 only (195 vs. 161 g/d) compared with the diets without SDE but did not affect growth performance during any other periods. In conclusion, SDE can be an efficacious protein and energy source in nursery pig diets and improves health and, in some instances, increases growth rate.  相似文献

Two experiments were conducted to determine the effects of dietary sodium butyrate on growth performance and response to Escherichia coli lipopolysaccharide (LPS) in weanling pigs. In a 28-d experiment, 180 pigs (initial BW 6.3 kg) were fed 0, 0.05, 0.1, 0.2, or 0.4% sodium butyrate, or 110 mg/kg of dietary tylosin. There was no effect of dietary sodium butyrate or tylosin on overall G:F, but there was a linear trend (P < 0.07) toward decreased ADFI and ADG as levels of sodium butyrate increased. In a second 28-d experiment, 108 pigs (initial BW 6.3 kg) were assigned to 1 of 3 dietary treatments: 1) no antibiotics, 2) 0.2% sodium butyrate, or 3) 55 mg/kg of carbadox. On d 14, a subset of pigs from the no-antibiotic and butyrate treatment groups was challenged with E. coli LPS or injected with sterile saline in a 2 x 2 factorial arrangement (+/-LPS challenge; +/-dietary butyrate; n = 6 pigs/treatment group). Four hours after LPS challenge, blood samples were obtained, and samples of LM, liver, and ileum were collected for gene expression analysis. Serum samples were analyzed for IL-6, tumor necrosis factor alpha (TNFalpha), alpha(1)-acid glycoprotein, cortisol, IGF-I, insulin, and metabolites. The relative abundance of tissue cytokine and IGF-I mRNA was measured by real-time PCR. Feeding diets containing sodium butyrate or carbadox did not alter ADG or ADFI compared with pigs fed the control diet. From d 0 to 14, pigs fed diets containing 0.2% sodium butyrate had decreased (P < 0.05) ADG and tended (P < 0.06) to have decreased G:F compared with animals fed diets containing carbadox. Challenge with LPS increased (P < 0.05) serum cytokines and cortisol and decreased (P < 0.05) serum glucose and triglycerides. Injection with LPS increased (P < 0.05) the relative abundance of hepatic IL-6 and TNFalpha mRNA, increased (P < 0.05) LM TNFalpha mRNA content, and decreased (P < 0.05) IGF-I mRNA in LM. For serum cortisol, there was an interaction (P < 0.05) between dietary butyrate and LPS. The increase in serum cortisol attributable to LPS was greater (P < 0.05) in pigs fed butyrate than in pigs fed the control diet. There tended (P < 0.10) to be an interaction between LPS and diet and for butyrate to increase the relative abundance of IL-6 mRNA in LM. Carbadox did not alter cytokine or IGF-I mRNA or serum metabolites, but did decrease (P < 0.05) serum TNFalpha. These data indicate that dietary sodium butyrate does not enhance growth performance, but may regulate the response to inflammatory stimuli in weanling pigs.  相似文献

Feed intake was investigated in early weaned pigs housed in two environments. In the first experiment, pigs in an unfamiliar environment (removed from the sow and placed in nursery pens) were offered a dry diet either ad libitum or at different meal intervals (2, 4 and 6 h). Regardless of meal interval (ad libitum or hourly intervals), early weaned (21 d) pigs failed to consume sufficient feed for maintenance during the first 3 d postweaning. Pigs provided feed ad libitum consumed more (P less than .001) feed (142 vs 84 g/d) and gained more (P less than .05) weight (57 vs 3 g/d) than meal-fed pigs over the 7-d period. Pigs fed at 2-h intervals consumed more (P less than .001) feed than pigs fed at 4-h or 6-h intervals. Compared with preweaning levels (307 mu eq/liter), plasma free fatty acid (FFA) levels increased approximately fourfold by d 1 postweaning (1,372 mu eq/liter), then decreased through d 7 to levels below preweaning (142 mu eq/liter). The FFA levels were not affected (P greater than .1) by meal interval. In a second experiment, feed intake was investigated in weaned pigs that were allowed to consume food by a familiar method and in a familiar environment (suckling the sow). When allowed to nurse following a 24-h weaning period, weaned pigs consumed (24.3 +/- 2.8 g/suckling) the same amount (P greater than .1) as non-weaned littermates (28.7 +/- 1.8 g/suckling).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This investigation addressed the hypothesis that, as a marker of adipocyte differentiation, stearoyl-coenzyme A desaturase (SCD) gene expression would be greater during growth in obese pigs than in crossbred, contemporary pigs. Suckled pigs from a single litter were removed from the sow for sampling at 0, 3, 10, and 17 d. The number of litters at 0, 3, 10, and 17 d of age was zero, two, three, and three (obese sows) and four, two, three, and three (crossbred sows), respectively. Postweaning pigs were removed from the sow at 14 d of age. One set of postweaning pigs was fed a high-fat, milk-based diet from d 28 to 49; pigs were killed on d 28 and 49 for sampling. The grain-fed pigs were switched to a pelleted, grain-based grower diet at d 28, and samples were obtained at 31, 35, or 49 d of age. Adipose tissue from all pigs in a litter for preweaning and postweaning pigs was pooled for the measurement of cellularity and SCD mRNA. There were significant genetic and age effects for adipocyte diameter and volume; overall, adipocytes from obese pigs were larger than those from crossbred pigs. Stearoyl-coenzyme A desaturase mRNA was barely detectable at 0 d of age and increased (P < .01) by 20-fold by 49 d of age. There was a significant genetic x age interaction (P = .026); there was more SCD mRNA in adipose tissue from obese pigs than in that from crossbred pigs during the suckling period, whereas crossbred pigs exhibited greater SCD gene expression than obese pigs during the postweaning period. The lesser SCD gene expression in postweaning obese pigs was caused by a strong depression in SCD gene expression in the grain-fed obese pigs. The data indicate that SCD gene expression provides a marker for terminal differentiation, especially in preweaning pigs. Furthermore, these results provide additional evidence that SCD gene expression is up-regulated by diets high in saturated fatty acids.  相似文献

Four trials were conducted to characterize the consumption of creep feed by nursing pigs and the effects of creep feeding (from 10 d to weaning at 28 d) on the immune response, scouring index and subsequent performance of weanling pigs. Pigs were fed a ground 20% CP corn-soybean meal-whey diet with 1.0% chromic oxide (control, 9 litters), this diet with 2.7% ovalbumin added as a dietary antigen (ovalbumin, 14 litters), or no creep feed (unexposed, 11 litters). At weaning, pigs within a litter were fed a 20% CP corn-soybean meal diet either with or without 2.7% ovalbumin. Creep-fed litters began eating at 11 d of age and disappearance of creep feed increased linearly until weaning (P less than .01). However, based on the chronic oxide coloring of the feces, total creep feed consumption was quite variable from pig to pig (13 to 194 g) and from litter to litter (107 to 1,550 g). Preweaning daily gain was similar between creep-fed and noncreep-fed litters; larger litters generally had lower daily gains (P less than .09) and less feed disappearance per pig (P less than .02). Weekly blood sampling showed that pigs fed the antigen diet had a higher (P less than .001) antibody titer to ovalbumin at 14, 21 and 28 d of age than did pigs fed the control diet or pigs unexposed to creep feed. At 56 and 63 d of age, all pigs given an ovalbumin injection at 49 d (1 ml containing 3 mg of ovalbumin) had responded (P less than .001) to injection, with the lowest titers for pigs fed the control creep diet and the highest titers for pigs fed the ovalbumin creep diet; titers were intermediate for pigs not fed creep. Regardless of preweaning or postweaning treatment, most pigs began scouring 4 to 5 d postweaning; scouring peaked at d 10 and returned to normal after d 15. Although the magnitude of difference was small, creep-fed pigs tended to scour more than pigs not fed creep (P less than .01). Postweaning performance was not influenced by preweaning treatments.  相似文献

Two experiments were conducted to determine the effect of nutritional status, age at weaning and room temperature on growth and immune response of pigs. In the first experiment, 72 pigs were weaned at either 2 or 3 wk and fed either a complex, simple-adequate or simple-inadequate diet for a 24-d period. Total antibody (TAb) titers in response to injections of human red blood cells (RBC) on either 0, 4, or 16 d postweaning were not affected by age at weaning or dietary treatments even though the nutritional status was compromised to such an extent that pigs fed the simple-adequate diet gained at one-half the rate (.172 vs .349 kg/d) of pigs fed the complex or simple-adequate diets (P less than .01). In Exp. 2, the immunological response of 144 pigs weaned at 3 wk were evaluated at 4 and 8 d postweaning. Pigs were weaned into rooms that averaged either 25 or 18 C, and were fed either a complex or simplex-adequate diet at either ad libitum (AL) or restricted (Res) levels for a 24-d period. Restriction of either the complex or simple diet for the first 16 d of the trial drastically reduced (P less than .01) the growth rates of Res pigs compared with AL pigs (.042 vs .236 kg/d). Two experiments were conducted to determine the effect of soybean meal in postweaning diets on villus height (VH), villus shape and lamina propria depth (LPD) in the small intestine. Intestinal samples were taken at 25, 50 and 75% of the small intestine's length. The first experiment determined the normal effects of age on these structures in nursing pigs from 9 to 36 d of age. Villus height decreased (P less than .01 at the 50 and 75% locations) and LPD increased (P less than .01 at the 25% location) in a quadratic fashion with increasing age. In the second experiment, pigs were weaned at 21 d of age and fed one of three 20% protein diets: 1) protein from hydrolyzed casein (CAS), 2) soybean meal (SBM) or 3) a conventional corn-soybean meal diet (CSBM). Intestinal samples were taken 0, 3, 6, 9, 12 and 15 d post-weaning. Villus height decreased dramatically the first 3 d postweaning, with a continuing decline until 12 d post-weaning. Averaged over the d 3 to 15 interval, VH were generally greater for pigs fed CAS than for pigs fed CSBM or SBM. Pigs fed SBM or CSBM had deformed villi, and pigs fed SBM had greater (P less than .05) LPD at all intestinal locations, suggesting less mature enterocytes on the villi. These data suggest that feeding high concentrations of soybean meal to the pig post-weaning has a detrimental effect on the small intestine, but this effect was minimized when soybean meal was fed as part of a corn-based diet.  相似文献

We investigated whether spray-dried plasma (SDP) improved growth and health of piglets challenged with enterotoxigenic Escherichia coli K88 (ETEC). Forty-eight pigs weaned at 21 d (BW = 4.88 +/- 0.43 kg) received one of four diets containing 6% SDP or fish proteins (as-fed basis) either nonmedicated (SDP-NM and FP-NM diets) or medicated with 0 or 250 mg/kg of colistine + 500 mg/kg of amoxycycline (SDP-M and FP-M diets), for 15 d. On d 4, pigs were orally challenged with ETEC. On d 15, eight pigs per dietary group were killed, blood and saliva were collected for analysis of K88 fimbriae-specific immunoglobulin (Ig)-A, and jejunum was removed for villi preparation, histological analysis, and cytokine expression. The presence or absence of K88 receptors (K88+ and K88- pigs respectively) was determined by villous adhesion assay. Effects of protein source on ADG (P = 0.04) and ADFI (P < 0.01), as well of medication on ADFI (P < 0.02), of all pigs were observed. In sacrified pigs, there was an effect of protein source on ADG (P = 0.03) and ADFI (P < 0.001), as well an interaction between medication and presence of K88 receptor (P = 0.02) for feed:gain ratio. Plasma K88 specific IgA were low in all K88 pigs and higher in K88+ pigs fed FP-NM compared with all the other groups (P < 0.05), except SDP-M. An interaction was found among protein source, medication, and presence of K88 receptors (P = 0.04). Saliva IgA concentrations were high in all pigs fed FP-NM and low in all other pigs. Jejunum of pigs fed FP-NM showed some ulcerations, edema, and mild inflammatory cell infiltration (ICI). In pigs fed FP-M, edema was reduced. Conversely, only a mild ICI was observed in pigs fed SDP-NM and SDP-M. Crypt depth was increased in K88+ pigs fed SDP-NM and an interaction between protein source and presence of K88 receptors was observed (P < 0.05). Expressions of tumor necrosis factor-alpha and interleukin (IL)-8 were lower in pigs fed SDP-NM and SDP-M than in those fed FP-NM and FP-M, either K88- or K88+ (P < 0.01). In pigs fed FP diets, expression of IL-8 tended to increase (P = 0.08) in K88+ compared with K88- subjects. Expression of interferon-gamma increased in K88 and K88+ pigs fed FP-M as compared with other pigs (P < 0.01). These results indicate that feeding with SDP improved growth performance and protected against E. coli-induced inflammatory status, and suggest that use of SDP-NM can be considered a valid antibiotic alternative.  相似文献

Peroxisome proliferator-activated receptor gamma (PPARgamma), a member of the nuclear hormone receptor superfamily, has been implicated in regulation of immunity and inflammation in rodents and humans. The objective of the current study was to investigate whether the expression of PPARgamma was altered in the immune system of weaned pigs after Escherichia coli lipopolysaccharide (LPS) injection. PPARgamma expression was investigated in the thymus, spleen, mesenteric lymph node and peripheral white blood cells of weaned pigs (8.54+/-0.24 kg BW) after LPS injection (100 microg/kg BW, n=6) and controls (sterile saline, n=6), by using real-time polymerase chain reaction, Western blot analysis, and immunohistochemistry. Plasma pro-inflammatory cytokines and hormones were also assessed. LPS triggered PPARgamma mRNA and protein expression in the thymus (P<0.05, 4.24-fold; P<0.10, 1.46-fold), spleen (P<0.10, 2.75-fold; P<0.05, 1.84-fold), mesenteric lymph node (P<0.05, 4.32-fold; P<0.05, 1.96-fold) and peripheral white blood cells (P<0.001, 24.44-fold; P<0.001, 1.58-fold). The LPS-injected pigs showed an increase in PPARgamma staining in splenic corpuscle and periarterial lymphatic sheath of white pulp (P<0.05) and red pulp (P<0.001) of spleen, and in medullas of thymus lobule of thymus (P<0.05), and in thymus-dependent area of mesenteric lymph node (P<0.05) compared to the control pigs. Concurrent with up-regulation of PPARgamma expression, LPS induced increases in plasma interleukin-6 (P<0.001), tumor necrosis factor-alpha (P<0.001), cortisol (P<0.001), prostaglandin E(2) (P<0.01) and 15-deoxy-Delta(12,14)-prostaglandin J(2) (15 d-PGJ(2)) (P<0.05), and decreases in plasma insulin (P<0.10) and insulin-like growth factor-1 (P<0.001). These results suggest that induction of PPARgamma expression in immune system may be associated with the release of the natural PPARgamma activating ligand 15 d-PGJ(2), and play an important role in host response to immunological stress. Additionally, it is possible that PPARgamma would be a new therapeutic target in treatment of immunological stress of livestock.  相似文献

The objective of this study was to evaluate the effects of heat stress and dietary bacitracin methylene disalicylate (BMD) on growth performance, carcass characteristics, and immunological responses in finishing pigs. Four groups of 32 finishing pigs (n = 128) with initial BW between 80 to 90 kg were used. Pigs were fed a corn-soybean meal-distillers grains-based control or BMD (31.5 mg/kg) diet for a 14-d adaptation period at the thermal neutral temperature (23°C), and continued to be fed their respective diets when exposed to a constant temperature (23°C) or a cyclical heat stress environment (37°C from 1000 to 1900 h and 27°C from 1900 to 1000 h) for a 28-d experimental period. Each group of pigs was housed in 4 rooms, with 2 pens/room and 4 pigs/pen. Saliva samples from each pig were collected on d -1 (initial baseline), 1, 13, and 27 for cortisol analysis. Concentrations of haptoglobin, IL-1β, and tumor necrosis factor-α were determined in serum samples on d -1, 1, 13, and 27. Pigs exposed to heat stress had 31% less ADG (P < 0.001), 23% less ADFI (P < 0.001), 9% less G:F (P < 0.001), and 34% greater average daily water intake (P = 0.03) than those in the non-heat-stress conditions. Dietary BMD tended to reduce ADG (P < 0.07) compared with the control (0.66 vs. 0.73 kg/d, respectively). Heat stress increased (P < 0.05) saliva cortisol on d 1, but no effects were observed on subsequent days. Serum haptoglobin concentrations were greater (P < 0.05) in heat-stressed pigs on d 1, and concentrations tended to remain greater (P < 0.1) on d 13. Pigs fed the BMD diet tended to have a longer villus height (P = 0.07) in the duodenum and greater crypt depths in the duodenum (P = 0.09) and jejunum (P = 0.07). Heat-stressed pigs tended to have a decreased proportion of propionate (P = 0.08), greater acetate:propionate (P = 0.08), and a reduced proportion of valerate (P = 0.02) in the cecum. These results indicate that BMD did not counteract the negative effects of heat stress on growth performance, but BMD appears to increase villus height and crypt depth in the duodenum. Furthermore, heat stress appears to alter VFA production in finishing pigs.  相似文献

The most common Salmonella serovars causing clinical disease in pigs are Salmonella enterica serovars Typhimurium (Typhimurium) and Choleraesuis. Given that the swine host-adapted serovar Choleraesuis has been reported to cause systemic disease, a different disease outcome from that of Typhimurium, our working hypothesis was that this serovar would likely engage systemic immune-inflammatory mechanisms, resulting in elevated systemic cytokine secretion. Forty-eight weaned pigs were blocked by BW and sex, and randomly allotted to 1 of 3 treatments in a 14-d study. Each treatment had 8 replicates (pens), with 2 pigs/pen. The treatments consisted of a negative control and pigs repeatedly fed 10(8) cfu of Typhimurium or Choleraesuis. On d 0, the pigs were fed Choleraesuis or Typhimurium in dough balls, and the bacteria were refed twice weekly throughout the experiment. Control pigs received dough balls without bacteria. All pigs were housed in temperature-controlled rooms under constant lighting and were fed a standard corn-soybean meal-based nursery diet. Pig BW and feed disappearance were used to determine ADG, ADFI, and G:F. Rectal temperatures were obtained daily from 1 pig/pen beginning 2 d before the first bacterial feeding through d 7 using rapid-response digital thermometers. Serum was collected on d 0, 7, and 14 from a single pig/pen for analysis of IGF-I, tumor necrosis factor-alpha , and IL-1beta. There was no change in the rectal temperature of the control or the Typhimurium-challenged pigs (compared with d 0) or when comparing Typhimurium-challenged pigs with control animals. In contrast, pigs fed Choleraesuis had increased rectal temperatures beginning on d 2 and continuing through d 7 (P < 0.05), with the greatest elevation on d 3 (P < 0.001) compared with the control pigs. Average daily gain and ADFI of pigs challenged with Typhimurium did not differ from those of the control animals. Pigs fed Choleraesuis had a 25% reduction in ADG (P < 0.0001) and ADFI (P < 0.002) compared with the control pigs. On d 7, pigs fed Choleraesuis had reduced serum IGF-I compared with control (P < 0.01) or Typhimurium-challenged pigs (P = 0.01). Bacterial feeding did not affect serum tumor necrosis factor-alpha or IL-1beta compared with control pigs at any time throughout the experiment. We conclude that repeated exposure of weaned pigs to Choleraesuis reduced growth performance in the absence of changes in systemic inflammatory cytokines.  相似文献

Full term crossbred sows were selected to study the interaction of the immune system, hypothalamus-pituitary-adrenal axis, and growth in pigs born by Cesarean section (c-section; n=4 sows) or vaginal birth (n=4 sows). Gestation length and birth weight did not differ between vaginal birth and c-section pigs (P=0.34 and 0.62, respectively). Blood and tissue samples were collected from 44 pigs at birth. Forty-five pigs were weaned at 13 d. On d 14, pigs received an i.p. injection of lipopolysaccaride (LPS; 150 microg/kg) or saline at min 0, and blood samples were collected at -20, -10, 0, 5, 10, 20, 40, 60, 90, and 120 min. Vaginal birth pigs had 21% greater average daily gain than c-section pigs on d 14 (P<0.01). Basal serum concentrations of adrenocorticotrophin (ACTH) and cortisol were greater in c-section than vaginal birth pigs at birth (P<0.01) but were not different at 14 d (P=0.99 and 0.80, respectively). LPS increased serum concentrations of ACTH, cortisol, interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha; P<0.01) but the response was not different between c-section and vaginal birth (P>0.22). Basal serum concentrations of TNF-alpha tended to be greater in c-section vs vaginal birth pigs at 14 d (P=0.0967); however, basal serum concentrations of IFN-gamma tended to be lower in c-section pigs vs vaginal birth pigs at 14 d (P=0.0787). Expression of interleukin (IL)-6, IL-6 receptor, IL-1beta, and TNF-alpha mRNA did not differ between vaginal birth and c-section pigs but changed in an age and tissue dependent manner. Thus, reduced growth rate of c-section pigs is associated with altered immune system function.  相似文献
