Tilapia lake virus disease (TiLVD) is an emerging viral disease in tilapia with worldwide distribution. Although the horizontal transmission of TiLV has been demonstrated through the cohabitation of infected fish with susceptible fish, no direct experiment showed the potential of vertical transmission from broodstock to progeny. In this study, natural outbreaks of TiLV in broodstock and fry in two tilapia hatcheries were confirmed. The TiLV genomic RNA was detected in liver and reproductive organs of infected broodstock, while infective virus was isolated in susceptible cell line. In situ hybridization assay confirmed the presence of TiLV in the ovary and testis of naturally infected fish and experimentally challenged fish. Moreover, early detection of TiLV in 2‐day‐old fry and the presence of TiLV genomic RNA and viable virus in the testis and ovary suggested the possible transfer of this virus from infected broodstock to progenies. As infective virus was present in gonads and fry in natural outbreak and experimental fish, the importance of biosecurity and prevention of the virus to establish in the hatchery should be emphasized. Hence, the development of TiLV‐free broodstock and the maintenance of high biosecurity standards in the hatcheries are essential for any attempt of virus eradication.  相似文献

Tilapia lake virus (TiLV) is an emerging virus associated with high fish mortality and economic losses. This study investigates the virucidal effects of the following disinfectants (active ingredients) on TiLV: 2.5 ppm iodine, 10 ppm sodium hypochlorite (NaOCl), 300 ppm hydrogen peroxide (H2O2), 80 ppm formalin and 5,000 ppm (0.5%) Virkon®. Factors that affect the disinfectants’ efficacy, including temperature, contact time and soiling (organic matter) interference, were examined under conditions mimicking natural aquaculture practices. TiLV inactivation of higher than 5 log10 TCID50 ml?1 was achieved after 10 min and at 28°C for all disinfectants except formalin; similar inactivation levels were reached by NaOCl and Virkon® at 10 min and 4°C. Extended exposure to formalin from 10 to 60 min at 28°C rendered more than 5 log10 inactivation. Increasing synthetic organic matter in the water to mimic soiling interference reduced the efficacy of NaOCl, iodine and H2O2 when tested at 10 min and 28°C; however, Virkon® still achieved more than 5 log10 inactivation. This study demonstrates that most common disinfectants effectively reduced viral loads to minimum levels. To limit the spread of TiLV in aquaculture farms and related facilities, the appropriate use of such disinfectants should therefore be promoted and implemented.  相似文献

Tilapia lake virus disease (TiLVD) has emerged to be an important viral disease of farmed Nile tilapia (Oreochromis niloticus) having the potential to impede expansion of aquaculture production. There is a need for rapid diagnostic tools to identify infected fish to limit the spread in individual farms. We report the first detection of TiLV infection by PCR in farmed and wild Nile tilapia from Lake Victoria. There was no difference in prevalence between farmed and wild fish samples (p = .65), and of the 442 samples examined from 191 fish, 28 were positive for TiLV by PCR. In terms of tissue distribution, the head kidney (7.69%, = 65) and spleen (10.99%, = 191), samples had the highest prevalence (p < .0028) followed by heart samples (3.45%, = 29). Conversely, the prevalence was low in the liver (0.71%, = 140) and absent in brain samples (0.0%, = 17), which have previously been shown to be target organs during acute infections. Phylogenetic analysis showed homology between our sequences and those from recent outbreaks in Israel and Thailand. Given that these findings were based on nucleic acid detection by PCR, future studies should seek to isolate the virus from fish in Lake Victoria and show its ability to cause disease and virulence in susceptible fish.  相似文献

Tilapia lake virus (TiLV) is a highly contagious pathogen that has detrimental effects on tilapia farming. This virus was discovered in 2014 and has received tremendous global attention from the aquaculture sector due to its association with high fish mortalities and its strong economic impact on the tilapia aquaculture industry. Currently, TiLV has been reported in 16 countries, and this number is continuing to rise due to improved diagnostic assays and surveillance activities around the world. In this review, we summarize the up-to-date knowledge of TiLV with regard to TiLV host species, the clinical signs of a TiLV infection, the affected tissues, pathogenesis and potential disease risk factors. We also describe the reported information concerning the virus itself: its morphology, genetic make-up and transmission pathways. We review the current methods for virus detection and potential control measures. We close the review of the TiLV story so far, by offering a commentary on the major TiLV research gaps, why these are delaying future TiLV research and why the TiLV field needs to come together and proceed as a more collaborative scientific community if there is any hope limiting the impact of this serious virus.  相似文献

Tilapia lake virus (TiLV) is an emerging pathogen in aquaculture, reportedly affecting farmed tilapia in 16 countries across multiple continents. Following an early warning in 2017 that TiLV might be widespread, we executed a surveillance programme on tilapia grow-out farms and hatcheries from 10 districts of Bangladesh in 2017 and 2019. Among farms experiencing unusual mortality, eight out of 11 farms tested positive for TiLV in 2017, and two out of seven tested positive in 2019. Investigation of asymptomatic broodstock collected from 16 tilapia hatcheries revealed that six hatcheries tested positive for TiLV. Representative samples subjected to histopathology confirmed pathognomonic lesions of syncytial hepatitis. We recovered three complete genomes of TiLV from infected fish, one from 2017 and two from 2019. Phylogenetic analyses based on both the concatenated coding sequences of 10 segments and only segment 1 consistently revealed that Bangladeshi TiLV isolates formed a unique cluster within Thai clade, suggesting a close genetic relation. In summary, this study revealed the circulation of TiLV in 10 farms and six hatcheries located in eight districts of Bangladesh. We recommend continuing TiLV-targeted surveillance efforts to identify contaminated sources to minimize the countrywide spread and severity of TiLV infection.  相似文献

Tilapia aquaculture is one of the fastest‐growing segments of fish production in Brazil. Nile tilapia (Oreochromis niloticus) is largely cultivated in the state of Parana, where Streptococcus agalactiae is the cause of severe disease outbreaks. The objective of this paper was to evaluate an inactivated S. agalactiae vaccine in tilapia for the control of streptococcal disease outbreaks. Tilapia, weighing approximately 20 g each, were intraperitoneally (i.p.) inoculated with 0.1 mL of the vaccine at a dose of 2.0 × 108 colony‐forming unit (CFU) mL?1. One group of tilapia (treatment 1) received one vaccine dose, and the other group of tilapia (treatment 2) received two doses, with an interval of 21 days. The control group was i.p. inoculated with 0.1 mL tryptic soy broth fish?1. Immunized and control tilapia were i.p. challenged with 0.1 mL of 3.0 × 107 CFU mL?1 at 30 days post vaccination. The fish were monitored daily for disease signs and for mortality for 16 days post challenge. A statistically significant difference (P=0.0045) was found between the mortality of treatments 1 and 2. The value of relative per cent of survival of 83.6% and 96.4%, respectively, indicate that this vaccine was efficient in Nile tilapia.  相似文献

Francisella noatunensis subsp. orientalis (Fno) (syn. F. asiatica) is an emergent Gram‐negative facultative intracellular bacterium. Although it is considered one of the most pathogenic bacteria in fish, there are no commercially available treatments or vaccines. The objective of this project was to determine the most efficacious concentration of florfenicol (FFC) [10, 15 or 20 mg FFC kg?1 body weight (bw) per days for 10 days] administered in feed to control experimentally induced infections of Fno in Nile tilapia, Oreochromis niloticus (L.), reared in a recirculating aquaculture system. The cumulative mortality of fish that received 0, 10, 15 or 20 mg FFC kg?1 bw per day was 60, 37, 14 and 16%, respectively. Francisella noatunensis subsp. orientalis genome equivalents were detected in water from all challenged groups with slight reduction in the concentration in the florfenicol‐treated groups 4 days after treatment. The mean LOG of CFU Fno mg?1 spleen was 3–5 and was present in all challenged groups at necropsy 11 days after treatment (21 days after challenge). Results show that florfenicol administered at doses of 15 and 20 mg FFC kg?1 bw per days for 10 days significantly reduced mortality associated with francisellosis in Nile tilapia.  相似文献

Cage‐pond integration system is a new model for enhancing productivity of pond aquaculture system. A field trial was conducted using African catfish (Clarias gariepinus) and Nile tilapia (Oreochromis niloticus) in cages and carps in earthen ponds. There were four treatments replicated five times: (1) carps in ponds without cage, (2) tilapia at 30 fish m?3 in cage and carps in open pond, (3) catfish at 100 fish m?3 in cage and carps in open pond, (4) tilapia and catfish at 30 and 100 fish m?3, respectively, in separate cages and carps in open pond. The carps were stocked at 1 fish m?2. The cage occupied about 3% of the pond area. The caged tilapia and catfish were fed and the control ponds were fertilized. Results showed that the combined extrapolated net yield was significantly higher (P < 0.05) in the catfish, tilapia and carps integration system (9.4 ± 1.6 t ha?1 year?1) than in the carp polyculture (3.3 ± 0.7 t ha?1 year?1). The net return from the tilapia and carps (6860 US$ ha?1 year?1) and catfish, tilapia and carps integration systems (6668 US$ ha?1 year?1) was significantly higher than in the carp polyculture (1709 US$ ha?1 year?1) (P < 0.05). This experiment demonstrated that the cage‐pond integration of African catfish and Nile tilapia with carps is the best technology to increase production; whereas integration of tilapia and carp for profitability.  相似文献

Sixteen countries, including Bangladesh, have reported the presence of tilapia lake virus (TiLV), an emerging tilapia pathogen. Fish polyculture is a common farming practice in Bangladesh. Some unusual mortalities reported in species co-cultivated with TiLV-infected tilapia led us to investigate whether any of the co-cultivated species would also test positive for TiLV and whether they were susceptible to TiLV infection under controlled laboratory experiments. Using 183 samples obtained from 15 farms in six districts across Bangladesh, we determined that 20% of the farms tested positive for TiLV in tilapia, while 15 co-cultivated fish species and seven other invertebrates (e.g. insects and crustaceans) considered potential carriers all tested negative. Of the six representative fish species experimentally infected with TiLV, only Nile tilapia showed the typical clinical signs of the disease, with 70% mortality within 12 days. By contrast, four carp species and one catfish species challenged with TiLV showed no signs of TiLV infection. Challenged tilapia were confirmed as TiLV-positive by RT-qPCR, while challenged carp and walking catfish all tested negative. Overall, our field and laboratory findings indicate that species used in polycultures are not susceptible to TiLV. Although current evidence suggests that TiLV is likely host-specific to tilapia, targeted surveillance for TiLV in other fish species in polyculture systems should continue, in order to prepare for a possible future scenario where TiLV mutates and/or adapts to new host(s).  相似文献

Aquaflor® [50% w w?1 florfenicol (FFC)], is approved for use in freshwater‐reared warmwater finfish which include tilapia Oreochromis spp. in the United States to control mortality from Streptococcus iniae. The depletion of florfenicol amine (FFA), the marker residue of FFC, was evaluated after feeding FFC‐medicated feed to deliver a nominal 20 mg FFC kg?1 BW d?1 dose (1.33× the label use of 15 mg FFC kg?1 BW d?1) to Nile tilapia O. niloticus and hybrid tilapia O. niloticus × O. aureus held in a recirculating aquaculture system (RAS) at production‐scale holding densities. Florfenicol amine concentrations were determined in fillets taken from 10 fish before dosing and from 20 fish at nine time points after dosing (from 1 to 240 h post‐dosing). Water samples were assayed for FFC before, during and after the dosing period. Parameters monitored included daily feed consumption and biofilter function (levels of ammonia, nitrite and nitrate). Mean fillet FFA concentration decreased from 13.77 μg g?1 at 1‐h post dosing to 0.39 μg g?1 at 240‐h post dosing. Water FFC concentration decreased from a maximum of 1400 ng mL?1 at 1 day post‐dosing to 847 ng mL?1 at 240 h post‐dosing. There were no adverse effects noted on fish, feed consumption or biofilter function associated with FFC‐medicated feed administration to tilapia.  相似文献

Detection of tilapia lake virus (TiLV) in tilapines is mainly from visceral organs of killed fish. However, lethal sampling might not be viable to broodstock and economically important ornamental cichlids. To contribute towards screening of the virus in asymptomatic infected fish, a subclinically infected population of Nile tilapia adults obtained from a local farm was preliminarily tested to compare different non‐lethal sampling methods, for example liver biopsy, gill biopsy, fin clip, mucus, faeces and blood for detection of TiLV. Only liver and blood samples gave positive results by PCR. Since blood sampling is relatively simpler, it was further used for five naturally co‐cultured juvenile fish species from above‐mentioned farm including 40 red tilapia broodstock and 20 Nile tilapia adults from two other different farms. The results showed that from the tested fish, 4 of 5 Nile tilapia, 2 of 5 hybrid red tilapia and 3 of 5 giant gourami blood samples tested positive, while 38 of 40 blood samples of red tilapia tested positive for TiLV in second‐step PCR. Sequencing representative PCR amplicons of positive samples confirmed sequence identity to TiLV. In conclusion, both blood and liver biopsy are practical non‐destructive sampling platforms for TiLV screening in cichlids with blood being more convenient, especially for tilapia broodstock.  相似文献

An 8‐week feeding trial was conducted to evaluate the effects of dietary leucine on growth performance, feed utilization, body composition and non‐specific immune responses of juvenile Nile tilapia. Five isonitrogenous and isoenergetic diets were formulated to contain graded levels of L‐leucine (5.3, 8.1, 10.9, 13.2, 15.6 and 18.1 g kg?1 diet, respectively) from dietary ingredients and crystalline L‐leucine. Each diet was randomly assigned to triplicate groups of 20 juvenile fish (1.94 ± 0.01 g) three times daily to apparent satiation. Results showed that the weight gain (WG) and specific growth rate (SGR) increased as dietary leucine concentrations increased from 5.3 to 13.2 g kg?1 and then decreased slightly with further increase in dietary leucine concentrations. Quadratic regression analysis (y = ?522.6x2 + 1304.x + 132.6, R² = 0.684) on weight gain against dietary leucine levels indicated that the optimal dietary leucine requirement was estimated to be 12.5 g kg?1 diet (corresponding to 43.1 g kg?1 of dietary protein). Leucine supplementation had no impact on the survival and body composition of tilapia. Serum lysozyme activity of fish fed diet containing 13.2 g kg?1 leucine significantly increased compared to fish fed diet containing 5.3 g kg?1. Serum superoxide dismutase activity and immunoglobulin M (IgM) concentration were not significantly affected by dietary leucine supplementation.  相似文献

This study determined the growth performance and acquired resistance of Nile tilapia, Oreochromis niloticus (L.) that survived Streptococcus iniae infection. Tilapia were challenged with three doses of S. iniae (8.8 × 103, 8.8 × 104 and 8.8 × 105 CFU fish?1 for low, medium and high challenges respectively). Groups of non‐injected and tryptic soy broth‐injected fish were maintained as controls. Significantly (P<0.05) higher mortality (45.0%) occurred in the high challenge treatment than in the low challenge treatment group (29.6%). The medium challenge group had mortality (36.3%) that did not differ significantly from the high or low treatment. Few fish died in the non‐injected and broth‐injected treatments (3.4% and 0.8% respectively). The tilapia that survived S. iniae infection used to assess growth performance were selected from survivors without gross clinical signs of disease. These fish were randomly stocked at a rate of 30 fish into each 57 L aquarium in triplicate and fed to apparent satiation for 8 weeks. No significant differences were detected in weight gain, feed intake, feed efficiency ratio or survival between S. iniae‐survived tilapia and the control treatments following the 8‐week growth performance trial. Following the 8‐week feeding study, tilapia were challenged with 1 × 106 CFU fish?1 of S. iniae to assess acquired immunity. Mean cumulative mortality was significantly higher (P<0.05) in the control treatments (41.7% for the non‐injected and 43.3% for the broth‐injected fish) than in the low, medium and high challenge treatments (7.4%, 3.3% and 8.3% respectively). Serum protein was significantly (P<0.05) elevated in the S. iniae‐survived tilapia that were subsequently challenged when compared with controls challenged for the first time. Agglutinating antibody titre was significantly higher in the fish in the medium and high challenge treatments, compared with the control fish challenged for the first time. The results suggest tilapia that survive S. iniae challenge without showing overt disease signs performed as well as non‐infected tilapia. Further, the S. iniae‐survived tilapia challenged following the 8‐week growth performance trial gained acquired resistance to homologous S. iniae challenge.  相似文献

Pollution by the heavy metal lead (Pb) has become a threat to both aquaculture and food safety. In this study, the protective effects of a selected probiotic with good Pb binding capacity (Lactobacillus plantarumCCFM8661) against waterborne Pb exposure were evaluated in Nile tilapia (Oreochromis niloticus). One hundred and eighty fish were divided into four groups as control, Pb‐only, Pb‐plus‐CCFM8661 and CCFM8661‐only. Fish were exposed to a dose of waterborne Pb level at 1 mg L?1 for 4 weeks, and the probiotic was administered at 108 CFU g?1 in fish diet twice daily. The results showed that dietary supplementation of CCFM8661 ameliorated the growth performance and prevented the death of Pb‐exposed fish. Cotreatment with Pb and CCFM8661 also effectively decreased Pb accumulation in kidney, liver, gonad, brain, gills and muscle. Moreover, the administration of this probiotic alleviated Pb‐induced oxidative stress, recovered digestive enzyme activities and blood δ‐aminolevulinic acid dehydratase activity, reversed alterations in innate immune status and decreased the frequencies of the nuclear abnormalities in peripheral blood erythrocytes of fish. These results suggested that CCFM8661 may be a novel dietary supplement against Pb toxicity at least in tilapia.  相似文献

Two floating diets and one sinking diet were prepared for tilapia with local ingredients by extrusion or steam‐pelleting methods. Indoor and outdoor feeding trials were conducted to evaluate their effects on growth performance and quality of tilapia products. Local ingredients included defatted Haematococcus and Spirulina by‐products from Hawaii. The three diets, plus a commercial feed, were each assigned to three replicate tanks, and each tank (120 L) was stocked with 12 juvenile tilapia in an 8‐week indoor trial. The results showed that (1) the floating diet had significantly (P < 0.05) higher weight growth than the sinking diet; (2) increased inclusion of local microalgae by‐product (defatted Haematococcus) in floating diet from 12% to 24% significantly (P < 0.05) decreased tilapia weight growth; and (3) the three microalgae‐added diets obtained higher (P < 0.05) weight growth than the commercial feed, although they contained lower protein (36.9% to 37.7%) and lipid (7.1% to 8.1%) than the commercial feed (42.3%; 8.6%). The floating diet (containing 12% microalgae) and the commercial feed were also tested in outdoor green water tanks, simulating the commercial production conditions of local tilapia farms. Each diet had three replicate tanks (1200 L) stocked with 100 juvenile tilapia. The 12‐week trial showed that the local diet had significantly (P < 0.05) lower growth (12.0 g week?1) than the commercial feed (14.8 g week?1). All tilapia meat products were measured with low n‐3 fatty acid contents. These results provide important information for feed formulation and production using local ingredients for tilapia.  相似文献
