Detection of tilapia lake virus (TiLV) in tilapines is mainly from visceral organs of killed fish. However, lethal sampling might not be viable to broodstock and economically important ornamental cichlids. To contribute towards screening of the virus in asymptomatic infected fish, a subclinically infected population of Nile tilapia adults obtained from a local farm was preliminarily tested to compare different non‐lethal sampling methods, for example liver biopsy, gill biopsy, fin clip, mucus, faeces and blood for detection of TiLV. Only liver and blood samples gave positive results by PCR. Since blood sampling is relatively simpler, it was further used for five naturally co‐cultured juvenile fish species from above‐mentioned farm including 40 red tilapia broodstock and 20 Nile tilapia adults from two other different farms. The results showed that from the tested fish, 4 of 5 Nile tilapia, 2 of 5 hybrid red tilapia and 3 of 5 giant gourami blood samples tested positive, while 38 of 40 blood samples of red tilapia tested positive for TiLV in second‐step PCR. Sequencing representative PCR amplicons of positive samples confirmed sequence identity to TiLV. In conclusion, both blood and liver biopsy are practical non‐destructive sampling platforms for TiLV screening in cichlids with blood being more convenient, especially for tilapia broodstock.  相似文献

Sixteen countries, including Bangladesh, have reported the presence of tilapia lake virus (TiLV), an emerging tilapia pathogen. Fish polyculture is a common farming practice in Bangladesh. Some unusual mortalities reported in species co-cultivated with TiLV-infected tilapia led us to investigate whether any of the co-cultivated species would also test positive for TiLV and whether they were susceptible to TiLV infection under controlled laboratory experiments. Using 183 samples obtained from 15 farms in six districts across Bangladesh, we determined that 20% of the farms tested positive for TiLV in tilapia, while 15 co-cultivated fish species and seven other invertebrates (e.g. insects and crustaceans) considered potential carriers all tested negative. Of the six representative fish species experimentally infected with TiLV, only Nile tilapia showed the typical clinical signs of the disease, with 70% mortality within 12 days. By contrast, four carp species and one catfish species challenged with TiLV showed no signs of TiLV infection. Challenged tilapia were confirmed as TiLV-positive by RT-qPCR, while challenged carp and walking catfish all tested negative. Overall, our field and laboratory findings indicate that species used in polycultures are not susceptible to TiLV. Although current evidence suggests that TiLV is likely host-specific to tilapia, targeted surveillance for TiLV in other fish species in polyculture systems should continue, in order to prepare for a possible future scenario where TiLV mutates and/or adapts to new host(s).  相似文献

Francisella orientalis is a highly virulent, emerging bacterium that causes mass mortalities in tilapia. This pathogen also affects numerous other warm-water fish species, including three-line grunt, hybrid striped bass and various ornamental fish. This study sheds light on two new species of fish that are susceptible to F. orientalis. Asian seabass and largemouth bass showed variable levels of susceptibility in a bacterial challenge experiment. After intraperitoneally injected with a dose of 106 CFU/fish, a total of 64.28% and 21.42% mortalities were obtained in Asian seabass and largemouth bass, respectively. Meanwhile, Nile tilapia showed acute mortality of 100%. All fish showed typical lesions of francisellosis, including multifocal granulomas in the spleen and head kidney. Immunohistochemical analysis revealed strong positive signals inside the granulomas of all fish. The bacterial recovery in solid media from infected fish was highest in Nile tilapia (85.71%), followed by Asian seabass (35.71%) and largemouth bass (21.42%). PCR results tested 100% positive for Nile tilapia, and 78.57% and 21.42% for Asian seabass and largemouth bass, respectively. In conclusion, Asian seabass and largemouth bass are susceptible to this pathogen, which warrants new management strategies when employing predation polyculture systems of these species with tilapia.  相似文献

This study investigated the effects of dietary inulin and Jerusalem artichoke (JA) on intestinal microbiota and morphometry of Nile tilapia fingerlings. Five treatment diets were designed to supplement inulin at 0 (basal diet), 2.5 and 5.0 g/kg, and JA at 5.0 and 10.0 g/kg. Nile tilapia larvae were fed experimental diets from the first feeding through the fingerling stage (84 days). The cultivation‐dependent technique showed that dietary inulin at 5.0 g/kg and JA (at both levels) increased lactic acid bacteria and Bifidobacterium spp., but decreased Vibrio spp. (p < .05). PCR‐DGGE targeting 16S ribosomal RNA gene revealed that dietary inulin and JA generated different profiles of microbial community compared with fish fed a basal diet. Compared with fish fed the basal diet, a greater intestinal villi height was observed in fish fed 5.0 g/kg inulin and JA at both levels (p < .05). A larger relative goblet cell number were observed in the anterior intestine of fish fed 5.0 g/kg inulin or JA (p < .05). Overall, dietary inulin (5.0 g/kg) and JA (5 and 10.0 g/kg) since the first feeding had effects on modulating the intestinal microbiota and morphology of Nile tilapia fingerlings.  相似文献

A novel virus, tilapia lake virus (TiLV), has been identified as a key pathogen responsible for disease outbreak and mass mortality of farmed tilapia. We used a deterministic susceptible‐infectious‐mortality (SIM) model to derive key disease information appraised with published TiLV‐induced cumulative mortality data. The relationship between tilapia mortality and TiLV exposure dosages was described by the Hill model. Furthermore, a disease control model was proposed to determine the status of controlled TiLV infection using a parsimonious control reproduction number (RC)‐control line criterion. Results showed that the key disease determinants of transmission rate and basic reproduction number (R0) could be derived. The median R0 estimate was 2.59 in a cohabitation setting with 2.6 × 105 TCID50 fish?1 TiLV. The present RC‐control model can be employed to determine whether TiLV containment is feasible in an outbreak farm by quantifying the current level of transmission. The SIM model can then be applied to predict what additional control is required to manage RC < 1. We offer valuable tools for aquaculture engineers and public health scientists the mechanistic‐based assessment that allows a more rigorous evaluation of different control strategies to reduce waterborne diseases in aquaculture farming systems.  相似文献

Tilapia lake virus (TiLV) is an emerging pathogen in aquaculture, reportedly affecting farmed tilapia in 16 countries across multiple continents. Following an early warning in 2017 that TiLV might be widespread, we executed a surveillance programme on tilapia grow-out farms and hatcheries from 10 districts of Bangladesh in 2017 and 2019. Among farms experiencing unusual mortality, eight out of 11 farms tested positive for TiLV in 2017, and two out of seven tested positive in 2019. Investigation of asymptomatic broodstock collected from 16 tilapia hatcheries revealed that six hatcheries tested positive for TiLV. Representative samples subjected to histopathology confirmed pathognomonic lesions of syncytial hepatitis. We recovered three complete genomes of TiLV from infected fish, one from 2017 and two from 2019. Phylogenetic analyses based on both the concatenated coding sequences of 10 segments and only segment 1 consistently revealed that Bangladeshi TiLV isolates formed a unique cluster within Thai clade, suggesting a close genetic relation. In summary, this study revealed the circulation of TiLV in 10 farms and six hatcheries located in eight districts of Bangladesh. We recommend continuing TiLV-targeted surveillance efforts to identify contaminated sources to minimize the countrywide spread and severity of TiLV infection.  相似文献

This study examined the antimicrobial susceptibility and mutation(s) in quinolone resistance‐determining regions (QRDRs) in streptococcal pathogens isolated from farmed Nile tilapia Oreochromis niloticus in Thailand. Surveillance of antimicrobial susceptibility in tilapia streptococcal pathogens reveals that Streptococcus agalactiae (= 97) and Streptococcus iniae (= 3) from diseased tilapia were susceptible to amoxicillin, florfenicol, sulfamethoxazole/trimethoprim and sulfadimethoxine/ormetoprim, however, only 78 isolates were susceptible to enrofloxacin. Twenty‐two enrofloxacin‐resistant S. agalactiae isolates were further examined for mutations in the QRDRs of gyrA, gyrB, parC and parE genes. Twenty isolates had single base pair changed in the gyrA sequence, C‐242‐T. Point mutations in gyrB, GC‐1135, 1136‐AA and T‐1466‐G, were identified in one isolate. All resistant isolates harboured a mutation in the parC gene, C‐236‐A, while no mutations were observed in the parE gene. The study represented mutations of gyrA and parC genes as marked modification of the enrofloxacin‐resistant S. agalactiae from farmed tilapia. This study is a primary report of the QRDRs mutations associated with fluoroquinolone resistance from streptococcal pathogen in the cultured fish. The phenotypic and genotypic characterization of enrofloxacin resistance S. agalactiae evident in this study has led to an improved regulation of antimicrobial use in Thai aquaculture.  相似文献

Francisella noatunensis subsp. orientalis is a pathogen of tilapia and other warm‐water fish for which no vaccines are commercially available. In this study, a whole cell formalin‐inactivated vaccine was developed for the first time using the highly virulent isolate STIR‐GUS‐F2f7 and the oil‐based adjuvant Montanide? ISA 763A VG. The efficacy of the vaccine was assessed in red Nile tilapia via intraperitoneal (i.p.) injection using homologous experimental infection and correlates of protection such as seral antibody production and bacterial loads in the spleen. For immunization, fish were i.p. injected with 0.1 ml of the vaccine, the adjuvant alone or PBS. At 840 degree days post‐vaccination, all fish were i.p. injected with 4.0 × 103 CFU/fish of pathogenic bacteria. The RPS at the end of the trial was 100% in the vaccinated group with significantly higher survival than in the adjuvant and control groups. The RPS in the adjuvant group was 42%, and no significant difference was seen in survival between this and the PBS group. Moreover, significantly higher antibody titres in the serum and significantly lower bacterial loads in the spleen were detected in the vaccinated fish by ELISA and qPCR, respectively. These findings highlight the potential of autogenous vaccines for controlling francisellosis in tilapia.  相似文献

Several studies investigated the usage of spirulina (Arthrospira platensis) in aquaculture as a functional dietary additive or plant proteins replacers; however, more efforts still needed to test its nano form inclusion in aquafeed. In this study, Nile tilapia (Oreochromis niloticus) fed dietary spirulina (Aplatensis) nanoparticles (SNP) at 0 (control), 0.1, 1 and 10 g/kg diet while the fifth diet was mixed with 10 g/kg of the normal form of Aplatensis as a positive control. The final weight (FW), weight gain (WG) and feed conversion ratio of fish fed SNP at 10 g/kg displayed improved values comparing with the other groups (p < 0.05). Further, fish fed 1 g SNP had higher FW and WG than the control and 0.1 g SNP and 10 g normal Aplatensis. The incorporation of SNP at 1 and 10 g/kg in Nile tilapia diets significantly (p < 0.05) enhanced total protein, albumin, globulin, lowered the values of serum glucose and activities of ALT and AST compared with control and spirulina groups. The SOD, CAT, GPx, lysozyme and respiratory burst activities were meaningfully promoted (p < 0.05) in fish received 1 and 10 g/kg diet concerning the control and spirulina groups. Nile tilapia challenged with Aeromonas hydrophila showed congestion of the kidney, liver and spleen tissues while SNP alleviated the inflammation induced by Ahydrophila. Different levels of SNP-incorporated group showed lower cumulative mortality than the other groups. In conclusion, the obtained results illustrated that spirulina in nano form is recommended to enhance Nile tilapia well-being.  相似文献

The optimum water temperature required for the normal growth of Nile tilapia is 25–30°C. In this study, tilapia was reared under suboptimal temperature (21.50 ± 1.50°C) and fed four diets with fish oil (FO), corn oil (CO), sunflower oil (SFO) and linseed oil (LnO) for 8 weeks. The results revealed improved final weight, average daily gain and intestinal amylase activity in the LnO group compared to FO and SFO groups (p < .05). The feed intake was increased significantly in FO and LnO groups compared to CO and SFO groups, while the feed conversion ratio was increased in the FO group (p < .05). The CO, SFO and LnO diets resulted in higher carcass lipids than fish fed FO, while CO decreased the ash content (p < .05). The growth hormone was significantly lowered by LnO, followed by SFO, while CO improved the serum alkaline phosphatase activity (p < .05). Glutathione peroxidase enhanced in fish fed SFO, while the lowest activities were recorded in fish fed FO (p < .05). Total superoxide dismutase was significantly elevated by CO and LnO when compared with fish fed FO and SFO (p < .05). Substituting FO with vegetable oils had normal intestinal and liver histological appearance. It could be concluded that substituting FO with either CO or LnO for Nile tilapia could maintain the normal growth performance and feed utilization with enhanced antioxidant capacity under suboptimal temperature.  相似文献

Mass mortality has occurred among cultured Nile tilapia, Oreochromis niloticus, on fish farms in Manzala, Dakahlia province, Egypt, in the summer season, 2019. Moribund fish were reported with deep ulcers, septicaemic lesions and sampled for bacterial isolation. In this study, most isolates were subjected to bacteriological examination, antibiotic sensitivity test, 16S rRNA gene sequencing and histopathological examination. Following isolate identification, intraperitoneal challenge of Nile tilapia with a bacterial suspension 2 × 106 CFU/ml was performed. Samples from liver, spleen and kidney were collected for histological and biochemical analysis. The results showed a high similarity (99%) to Photobacterium damselae strains using phylogenetic analysis of 16S rRNA. P. damselae exhibited resistance to amoxicillin and erythromycin, as well it was highly sensitive to chloramphenicol and doxycycline. Moreover, haemorrhage, oedema, hemosiderosis and melanomacrophage activation in the liver and head kidney of infected fish were detected by light and electron microscopy. Also, significant higher levels of CAT and SOD in the spleen and head kidney, as well as the serum levels of NO were observed in experimentally challenged O. niloticus, compared to the control fish. Our data identified P. damselae for the first time from infected Nile tilapia, describing its sensitivity to a variety of antibiotics, histopathological alterations and oxidative stress impact, and it could be useful indicators for understanding P. damselae pathogenesis, which might provide a preventive efficacy for P. damselae.  相似文献

A cross‐sectional study was conducted from November 2017 to March 2018 at Lake Hawassa to determine the prevalence of larval Contracaecum infestation in Nile tilapia, (Oreochromis niloticus), African Catfish (Clarias gariepinus) and Barbus species (Barbus intermedius). Fifty‐two point six per cent (52.6%) previous prevalence and 5% precision were used to estimate the sample size. Accordingly, a total of 383 randomly sampled fish species comprising of 163 (42.6%) C. gariepinus, 159 (41.5%) O. niloticus and 61(15.9%) B. intermedius were examined. The length and weight of each sampled fish were recorded. All the sampled fish were examined for the evidence of Contracaecum parasites in Hawassa University Veterinary Parasitology and Pathology Laboratory. Chi‐square values and comparison of proportions were used to analyse the data. The overall prevalence of Contracaecum parasites of fish population was 31.6%. The distribution of parasite was significantly affected by fish species (p = .000), sexes (p = .018), length (p = .003) and weight classes of fish (p = .026). As a hygienic problem and gutting activity conducted at the sides of the Lake Hawassa and distribution of discarded fish wastes for surrounding piscivorous birds by butchers and other people, the life cycle of Contracaecum parasite was perpetuated. Thus, the parasite is of zoonotic significance. Therefore, awareness creation activities for societies and control of fish parasites should be conducted in the study area.  相似文献

通过胃排空试验与养殖试验研究了不同投喂频率对尼罗系吉富罗非鱼幼鱼的胃排空、生长性能以及体组成的影响.在试验开始时,观测尼罗系吉富罗非鱼幼鱼的胃内饲料排空情况,胃排空试验结果表明,胃排空率的最佳描述为平方根函数,胃内饲料在饱食投喂后15 h左右完全排空,达到投喂前水平,80%胃排空为9h,也就是投喂后大约9h恢复食欲.360尾试验鱼(初始体质量3.72 g)以不同的投喂频率(1d4次、1d3次、1d2次、2d4次、2d3次、2d2次)分组,每组设立3个平行组,随机养殖于18个网箱中,每箱养殖20尾鱼,按饱食量投饲膨化饲料.养殖期为6周.尼罗系吉富罗非鱼幼鱼在投喂频率为1d4次、1d3次和1d2次时特定生长率和饲料效率显著高于投喂频率为2d4次、2d3次和2d2次时(P<0.05);投喂频率为1d2次、2d4次、2d3次和2d2次时其摄食量显著低于1d4次和1d3次时(P<0.05).随着投喂频率降低,鱼体水分含量逐步上升,脂肪和蛋白质含量逐步下降,其中1d4次、1d3次组鱼与2d2次组鱼有显著性差异(P<0.05).各投喂频率组间的肝体指数无显著性差异(P>0.05).3.7~48.0 g尼罗系吉富罗非鱼幼鱼的适宜投喂频率为1d2次,较2d2次、2d3次和2d4次时明显提高了生长速度和饲料效率,较1d3次、1d4次摄食量显著降低.2种试验结果较为一致.  相似文献

This study examined the potential of using the freshwater shrimp, Caridina nilotica (CNM), and mung beans, Vigna radiata (VRM), to replace Rastrineobola argentea fishmeal (RAF) as the primary protein source in fish feeds in East Africa. Six diets with varying proportions of RAF, CNM, VRM and full fat soybean meal were tested on Nile tilapia (initial body mass ± SD: 3.30 ± 0.27 g). The growth performance was best in groups fed diets containing either R. argentea or C. nilotica or a combination of the two. Growth was poorest in the fish fed a combination of C. nilotica and V. radiata, and intermediate in the groups fed the commercial formulation and a diet containing a combination of R. argentea and V. radiata. The cost of feed per kg of fish produced decreased with increasing inclusion of C. nilotica. The price of feed per kg fish produced was comparatively high in feeds containing V. radiata due to poor feed conversion ratio. Of all the diets tested, the price of feed per kg of fish produced was highest in the commercial formulation. Caridina nilotica is a good candidate to substitute R. argentea fishmeal in practical diets for Nile tilapia.  相似文献

The objective of this study was to evaluate growth, body chemical composition and lipid profile of Nile tilapia juvenile fed with Schizochytrium sp. Two hundred and forty Nile tilapia (Oreochromis niloticus) juvenile (1.33 ± 0.11 g) were distributed in 20 aquariums, at the density of 12 fish per aquarium. The juvenile were fed with five levels of Schizochytrium sp.: Control (0 g/kg of Schizochytrium sp. in feed); AS10 (10 g/kg of Schizochytrium sp. in feed); AS20 (20 g/kg of Schizochytrium sp. in feed); AS30 (30 g/kg of Schizochytrium sp. in feed) and AS40 (40 g/kg of Schizochytrium sp. in feed). The inclusion of Schizochytrium sp. increase the body weight, weight gain, final biomass and biomass gain of tilapia juvenile. The body crude protein of tilapia was increased after addition of microalgae. Juvenile lipid profile also was influenced when fed with 40 g of Schizochytrium sp. per kg and have it levels of omega‐3 and docosahexaenoic acid (DHA) increased, and omega 6:3 ratio decreased. Tilapia juvenile fed with Schizochytrium sp. per kg have better growth, omega‐3 and DHA levels rising in the body, being a considerable source of fatty acids for human nutrition. Levels above 20 g of Schizochytrium sp. per kg in the feed favour the increase of protein in Nile tilapia body.  相似文献

Two experiments were conducted to investigate the effects of feed colour on the performance of Nile tilapia (Oreochromis niloticus) larvae and fingerlings. In the first experiment, triplicate groups of newly hatched larvae (0.01 g fish−1) were stocked in 40 L glass aquaria at a density of 2 fish L−1. The fish were fed a test diet (400 g kg−1 crude protein) with six different colours (dark blue, dark green, red, dark brown, yellow and light brown) for 60 days. The best performance and survival were achieved in fish fed on dark‐coloured diets, while light‐coloured diets (yellow and light brown) resulted in inferior performance. Dark diets also produced higher body protein than light diets. Body water, lipids and ash showed irregular trends. In the second experiment, triplicate groups of Nile tilapia fingerlings (5.30 g fish−1) were stocked in 140‐L aquaria, in a recirculating indoor system. The fish were fed a test diet (350 g kg−1 crude protein) with the same colours used in the larval trial, for 60 days. Growth rates, feed efficiency, survival and body composition were not significantly affected by feed colours. These results suggest that Nile tilapia larvae are visual feeders, and they prefer dark‐coloured diets to light‐coloured diets, while fingerling fish showed no preference to diet colours.  相似文献
